Abstract:Fluoro-solid phase extraction (fluoro-SPE) has been well established to selectively extract targets after they have been fluoro-tagged. Here we report that this technique can also be used to selectively extract fluorosurfactants of PFAS (per- and poly-fluoroakyl substances), which have fluoro-carbon skeletons, from other anionic surfactants that have no fluoro-carbon skeletons. We advance the fluoro-SPE protocol to enable the extraction of fluorosurfactants using 4-5 times (in volume) more solvent for fluorophilic eluting in effort to overcome the strong interaction between the fluorosurfactants and the fluoro-Gel. Based on a smartphone-based app we developed recently to assess the colouration of fluorosurfactants with a dye (ethyl violet) that is similar to MBAS (methylene blue active substances), plus an SPE for pre-concentration and fluoro-SPE for pre-selection, we successfully achieve a detection limit of 1 ppb for the PFAS including perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS) and 6:2 fluorotelomer sulfonate (6:2FTS), which is encouraging and can be conducted in a general laboratory.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are significant environmental and food pollutants that can cause cancer. In this work, a specific monoclonal antibody (mAb) to identify pyrene (PYR) and benzo [a]pyrene (BaP) was prepared, and an indirect competitive enzyme-linked immunoassay (ic-ELISA) was established to detect PYR and BaP residues in living aquatic products for the first time. The effects of complete antigens with different coupling ratios on the production of high-sensitivity mAb was explored. Under the optimal conditions, the IC50 value was 3.73 ± 0.43 μg/L (n = 5). The limits of detection (LODs) for PYR and BaP in fish, shrimp, and crab ranged from 0.43 to 0.98 μg/L. The average recoveries of the spiked samples ranged from 81.5–101.9%, and the coefficient of variation (CV) was less than 11.7%. The validation of the HPLC-FLD method indicated that the ELISA method set up in this experiment provided a trustworthy tool for PAHs residues detection in aquatic products.

Abstract:Pairs of genetically marked bacteriophage lambda DNAs have been injected into Xenopus laevis oocyte nuclei. After suitable incubation, DNA was recovered and packaged into phage particles in vitro. When these were plated onto a selective host, phage recombinant for parental markers were observed. Recombination was dependent on both parents being present in the same oocyte nucleus and was roughly proportional to the physical separation of the markers. Thus, the oocytes appear to contain the machinery necessary for performing typical genetic recombination. This system offers a great deal of scope and flexibility for future studies of recombination mechanisms at the molecular level in vertebrates.

Abstract:In the last years, surface-enhanced Raman scattering based immunochromatographic assay (SERS-ICA) has been getting great attention and been applied for the detection of different target analytes. In this study, a novel SERS-ICA using Ag-MBA@SiO2-Ab as an immunoprobe has been developed for rapid, quantitative and ultrasensitive detection of a pharmaceutical diclofenac (DCF) residue in water samples. Ag, Ag@SiO2 and Ag-MBA@SiO2 nanoparticles (NPs) were synthesized and characterized. The immunoprobe was prepared by sandwiching the Raman reporter mercaptobenzoic acid (MBA) between the core-shell layers and immobilizing the monoclonal antibody (mAb) against DCF on the surface of the Ag@SiO2 NPs. Under optimal conditions, the sensitivity and limit of detection (LOD) of the SERS-ICA for DCF were found to be 9 pg mL(-1) and 0.07 pg mL(-1), respectively. The detection of the SERS-ICA for DCF was completed within 15 min. Ag-MBA@SiO2-Ab was stable within four months without significant loss of SERS intensity. The key to success of the SERS-ICA was the employment of the Ag@SiO2 NPs as the SERS substrate, which possessed gorgeous SERS enhancement capability, predominant biocompatibility and excellent stability. This SERS-ICA is potential as an ultrasensitive analytical method in point-of-care testing, clinical diagnosis, food inspecting and environmental monitoring.

Abstract:The detection of molecules from highly diluted solutions with a limited amount is vital for precancer diagnosis, food safety, and forensic analysis. The sensitivity and convenience of detection techniques are the primary concerns. In this study, a hybrid superhydrophobic/-philic (SH/SHL) microporous platform is designed and fabricated by a femtosecond laser to improve surface-enhanced Raman scattering (SERS) performances. Relying on the micropores fabricated at the center of SHL patterns, sediments distributed at the central regions are avoided, leading to the further enrichment of the target molecules. The engineered micropores with high identification further improve the speed of Raman tests, and the fabricated SERS substrate shows an advantage in outdoor handheld detection and automated inspection applications. The optimized SERS sensor is sufficient for attomolar-level detection (10-17 M) of rhodamine 6G using analyte volumes of just 5 μL, corresponding to an enhancement factor of 5.19 × 1013. Meanwhile, a relative standard deviation of 7.48% at 10-10 M shows the excellent uniformity of this proposed SERS platform. This work further pushes forward the practical applications of SERS technology in ultratrace molecular detections.

Abstract:Bisphenol A (BPA) is an endocrine disruptor which is widely present in fish under the influence of environmental pollution. It is essential to establish a rapid detection method for BPA. Zeolitic imidazolate framework (ZIF-8) is a typical metal-organic framework material (MOFs) with a strong adsorption capacity, which can effectively adsorb harmful substances in food. Combining MOFs and surface-enhanced Raman spectroscopy (SERS) can achieve rapid and accurate screening of toxic substances. In this study, a rapid detection method for BPA was established by preparing a new reinforced substrate Au@ZIF-8. The SERS detection method was optimized by combining SERS technology with ZIF-8. The Raman peak at 1172 cm−1 was used as the characteristic quantitative peak, and the lowest detection concentration of BPA was as low as 0.1 mg/L. In the concentration range of 0.1~10 mg/L, the linear relationship between SERS peak intensity and the concentration of BPA was good, and R2 was 0.9954. This novel SERS substrate was proven to have great potential in rapidly detecting BPA in food.

Abstract:In recent years, the rapid detection of chloramphenicol (CAP) has become a market demand due to its high toxicity. In this study, for the first time, a portable surface-enhanced Raman scattering (SERS) aptasensor for the rapid and on-site detection of chloramphenicol (CAP) residues in fish was developed. Fe 3 O 4 @Au nanoflowers combined with sulfhydryl (SH)-CAP aptamer complementary DNA acted as capture probes. SH-CAP aptamer modified nanoparticles ( NPs) embedded with 4-mercaptobenzoic acid (4-MBA) were served as reporter probes. The strongest Raman intensity was produced due to the coupling of Fe 3 O 4 @Au nanoflowers (Fe 3 O 4 @Au NFs) and NPs. For CAP detection, a wide linear range from 0.001 to 1000 μg/L, with an R 2 of 0.9805, was obtained. The limit of detection was determined to be 0.87 ng/L. The SERS aptasensor showed excellent performance for analytical applications for real fish samples. Compared with the conventional HPLC method, the developed SERS aptasensor coupled with a handheld Raman spectrometer had flexible application and avoided the limitations of complex operating conditions. It should be a promising portable analytical tool for analysis of drug residues in the field.

Abstract:This study established novel and high-throughput strategies for the simultaneous analysis of 30 fluorinated emerging pollutants in different matrices from the shrimp aquaculture system in eastern China using UHPLC-MS/MS. The parameters of SPE for analysis of water samples and of QuEChERS methods for sediment and shrimp samples were optimized to allow the simultaneous detection and quantitation of 17 per- and polyfluoroalkyl substances (PFASs) and 13 fluoroquinolones (FQs). Under the optimal conditions, the limits of detection of 30 pollutants for water, sediment, and shrimp samples were 0.01–0.30 ng/L, 0.01–0.22 μg/kg, and 0.01–0.23 μg/kg, respectively, while the limits of quantification were 0.04–1.00 ng/L, 0.03–0.73 μg/kg, and 0.03–0.76 μg/kg, with satisfactory recoveries and intra-day precision. The developed methods were successfully applied to the analysis of multiple samples collected from aquaculture ponds in eastern China. PFASs were detected in all samples with concentration ranges of 0.18–0.77 μg/L in water, 0.13–1.41 μg/kg (dry weight) in sediment, and 0.09–0.96 μg/kg (wet weight) in shrimp, respectively. Only two FQs, ciprofloxacin and enrofloxacin, were found in the sediment and shrimp. In general, this study provides valuable insights into the prevalence of fluorinated emerging contaminants, assisting in the monitoring and control of emerging contaminants in aquatic foods.

Abstract:In this study, we propose a novel surface-enhanced Raman scattering (SERS) method for quantifying aflatoxin B1 (AFB1). This method relies on the target-triggered release of a SERS reporter from aptamer-sealed aminated mesoporous silica nanoparticles (MSNs). These MSNs were synthesized to accommodate 4-mercaptophenylboronic acid (4-MPBA) within their well-defined micropores, which were subsequently sealed with AFB1 aptamers. Upon specific binding of AFB1 to its aptamer, the conformational change in the aptamer is regulated by the presence of the target. Consequently, a positive linear relationship between the AFB1 concentration and the 4-MPBA SERS signal was observed. Under optimal conditions, the method exhibited a good linear relationship over the range of 0.1 to 5 ng/mL AFB1, with a limit of detection (LOD) of 0.03 ng/mL. This strategy was validated using wheat samples, yielding results comparable to high performance liquid chromatography-fluorescence detector (P > 0.05), confirming its reliability for detecting AFB1 in complex food matrices.

Abstract:A simple and sensitive method combining solid-phase extraction (SPE) and high-performance liquid chromatography-ultraviolet detection (HPLC-UV) was developed for the determination of benzo[a]pyrene (BaP) in fish. Loofah sponge (LS) was carbonized and used as SPE adsorbent. Carbonization decreased the polarity of LS, and enhanced its aromaticity. Carbonized loofah sponge (CLS) could capture BaP better through π-π interaction. The carbonization temperature and the SPE conditions were optimized. The linear range of the developed method was within 10-1000 ng/g with a satisfactory correlation coefficient (R2) of 0.9999. The limit of detection (LOD) was 2.0 ng/g, which was below the maximum residue limit (5 μg/kg) in meat set by the European Union. The method showed good intra-day and inter-day precision with relative standard deviations (RSDs) ranging from 0.4% to 1.7%. Finally, the developed method was applied to the determination of BaP in fish samples. This method is low-cost and environmentally friendly with natural and renewable LS as raw material and it provides an alternative approach for the efficient and simple determination of BaP in aquatic products.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) present in oily sludge generated by the petroleum and petrochemical industries have emerged as a prominent concern within the realm of environmental conservation. The precise determination of PAHs holds immense significance in both petroleum geochemistry and environmental protection. In this study, a combination of surface-enhanced Raman spectroscopy (SERS) and solid–liquid extraction was employed for the screening of PAHs in oily sludge. Methanol was utilized as the extraction solvent for PAHs, while nanosilver-silicon coupling substrates were employed for their detection. The SERS spectrum was acquired using a portable Raman spectrometer. The nano silver-silicon coupling substrate exhibits excellent uniformity, with relative standard deviations (RSDs) of Phenanthrene, Fluoranthrene, Fluorene and Naphthalene (Phe, Flt, Flu and Nap) being 2.8%, 1.08%, 1.41%, and 5.44% respectively. Moreover, the limits of detection (LODs) achieved remarkable values of 0.542 μg/g, 0.342 μg/g, 0.541 μg/g, and 5.132 μg/g. The quantitative analysis of PAHs in oily sludge was investigated using SERS technology combined with partial least squares (PLS). The optimal PLS calibration model was optimized by combining spectral preprocessing methods and using the SiPLS (Synergy interval partial least squares)-VIP (Variable Importance in Projection) hybrid variable selection strategy. The prediction performance of the D1st (First derivative)-WT (Wavelet transform)-SiPLS-VIP-PLS model was deemed satisfactory, as evidenced by high R 2 P values of 0.9851, 0.9917, and 0.9925 for Phe, Flt, and Flu respectively; additionally, the corresponding MREP values were found to be 0.0580, 0.0668, and 0.0669 respectively. However, for Nap analysis, the D1st-WT-PLS model proved to be a better calibration model with an R 2 P value of 0.9864 and an MREP (Mean relative error of prediction) value of 0.0713. In summary, SERS technology combined with PLS based on different spectral pretreatment methods and mixed variable selection strategies is a promising method for quantitative analysis of PAHs in oily sludge, which will provide new ideas and methods for the quantitative analysis of PAHs in oily sludge.

Abstract:Constructing advanced substrates with excellent features is promising for sensitive surface-enhanced Raman spectroscopy (SERS) detection. Here a novel capillary monolithic 3D structural-substrate SERS platform with Au@cDNA@Ag@Cyanine 3-aptamer nanoparticles (Au@cDNA@Ag@Cy3-Apt NPs) was fabricated for rapid, highly specific profiling of ultra-trace Bisphenol A (BPA). The proposed SERS platform combined both in-capillary SERS and aptamer-affinity recognition strategies, in which the superior SERS properties of Au–Ag NPs, aptamer selectivity, and the advantages of capillary monolith were integrated. A 3D hierarchically porous network was constructed in the monolithic column, which was endowed with rich hotspots for SERS, rapid sample permeation, and better analysis efficiency than most plane-shaped SERS modes. By varying the amount of Ag + precursor, the Ag-shell thickness on SERS was finely tuned to guarantee Cy3 label in proximity to the plasmonic surface. Based on the biorecognition of aptamer, the selective identification of BPA occurred and exhibited a significant change in SERS intensity without obvious interference. As a result, the monolithic SERS platform featured facile operation, excellent specificity, and rapid analysis (10 min, much less than the solution-based or planar substrate SERS modes). Ultra-high sensitivity and robust reproducibility for BPA analysis was achieved with a low limit of detection (LOD) at 9.12 × 10 - 4 ng/L. The feasibility of this SERS platform for monitoring BPA in water and milk samples was also validated. This work lights a new access to capillary monolithic SERS-sensing platform for ultrasensitive and specific analysis of BPA.

Abstract:The alarming increase in drug-resistant bacteria in fish resulting from the misuse of antibiotics poses a significant threat to ecosystems and human health. Therefore, the development of a reliable approach for detecting antibiotic residues in fish is crucial. In this study, a rapid and simple method for detecting chloramphenicol (CAP) residue in tilapia was developed using surface-enhanced Raman scattering (SERS) combined with chemometric algorithms. Silver and gold core-shell nanoparticles (Ag@Au CSNPs) were used as SERS nanosensors to achieve strong signal amplification with an enhancement factor of 2.67 × 106. The results demonstrated that the variable combination population analysis-partial least square (VCPA-PLS) model combined with the standard normal variable transformation pretreatment method exhibited the best predictive performance with a detection limit of 1 × 10-5 µg/mL. Thus, an SERS technique was established based on Ag@Au CSNPs combined with VCPA-PLS to rapidly detect CAP in tilapia.
Super-hydrophilic SERS sensor with both ultrahigh activity and exceptional 3D spatial uniformity for sensitive detection of toxic pollutants

Abstract:The powerful surface-enhanced Raman scattering spectroscopy (SERS) practical technique is not only related to signal enhancement but also heavily dependent on spatial uniformity. Different from the routine hydrophobic substrates, an extraordinary super-hydrophilic SERS sensor is established by loading plasmonic Au nanoparticles (NPs) on two-dimensional (2D) hexagonal boron nitride (h-BN) then the hybrids uniformly are grafted into 3D bacterial nanocelluloses (BNCs). The Au /BNCs exhibit a remarkably high SERS activity with a limit of detection (LOD) of dyes at 0.1 femtomole (fM) level (вҲј10 -16 M). It is attributed to the strong synergistic coupling effect between highly dense Au NPs and modified h-BN as well as multiple light-scattering effects in 3D porous supports. More importantly, the distinctive advantage is further highlighted by the exceptional 3D spatial SERS uniformity with relative standard deviation (RSD) less than 3.4% along both horizontal and vertical directions. The unique feature is due to the 3D homogeneous distribution of the injected colloidal analytes via ultra-rapid liquid diffusion throughout the entire body with superior wettability. Besides, the flexible SERS substrates with excellent easy-tailorable mechanical properties can be robust enough to withstand the portable operations in real-world scenarios. These competitive merits are particularly beneficial for reliable SERS quantitative surveillance.

Abstract:Surface-enhanced Raman scattering (SERS) has been proven to be an extremely efficient, reliable, and influential technique to monitor or determine various pollutants in food. It has various advantages, including simplicity, sensitivity, speed, and the ability to create distinguishable analyte fingerprints with minimal interference from water. The goal of this review was to evaluate the SERS-based sensor's progress in detecting and monitoring chemical pollutants in food. Over the last decade, significant progress has been made in the analysis of chemical contaminants, particularly pesticides, heavy metals, and mycotoxins. Several factors leading to improved SERS performance, such as SERS substrate, Raman reporter molecules, antibody, aptamer, lab-on-a-chip, molecularly imprinted polymers, as well as SERS tag techniques such as immunochromatographic assay, colorimetry, and imaging, are briefly discussed. The paper also summarized the theory and application of SERS-based label-free sensors and also emphasized the progress in applying mathematical models to SERS analysis of chemical contaminants.

The results show that SAFe exhibits a strong catalytic effect on the HAuCl4-EG nanoreaction, which could generate purple gold nanoparticles (AuNPs) with resonance Rayleigh scattering (RRS) signals and surface-enhanced Raman scattering (SERS) effects. After the addition of BPA aptamer (Apt), it could encapsulate SAFe through intermolecular interaction, thus inhibiting its catalytic action, resulting in the reduction of AuNPs generated and the decrease of RRS and SERS signals of the system. With the addition of BPA, Apt was specifically combined with BPA, and SAFe was re-released to restore the catalytic ability; the generated AuNPs increased. As a result of this RRS and SERS signals of the system recovered, and their increment was linear with the concentration of BPA. Thus, the quantification of 0.1–4.0 nM (RRS) and 0.1–12.0 nM (SERS) BPA was realized, and the detection limits were 0.08 nM and 0.03 nM, respectively. At the same time, we used molecular spectroscopy and electron microscopy to study the SAFe-HAuCl4-ethylene glycol indicator reaction, and proposed a reasonable SAFe catalytic reaction mechanism.

Abstract:Surface-enhanced Raman spectroscopy offers great potential for rapid and highly sensitive detection of pharmaceuticals from environmental sources. Herein, we investigated the feasibility of label-free sensing of antibiotic residues from wastewater effluents with high specificity by combining with multivariate analysis . Highly ordered silver nanoarrays with ∼34 nm roughness have been fabricated using a cost-effective electroless deposition technique. As-fabricated Ag arrays showed superior LSPR effects with an enhancement factor of 8 × 10 7 . Excellent reproducibility has also been noticed with RSD values within 11%, whilst the sensor showed good stability and reusability characteristics for being used as a low-cost and reusable sensor. SERS studies demonstrated that antibiotics-spiked wastewater effluents can be detected with high efficiency in a label-free method. The molecular fingerprint bands of antibiotics such as sulfamethoxazole, sulfadiazine, and ciprofloxacin were well analyzed in effluent, tap, and deionized water. It has been found that antibiotics can be detected near picomolar levels; meanwhile, liquid chromatography-mass spectrometry (LC-MS) exhibited a detection limit within nanomolar concentrations only. Furthermore, the specificity of SERS sensing has been further analyzed using a multivariate analysis method, principal component analysis followed by linear discriminant analysis (PCA-LDA); which showed prominent discrimination to distinguish each antibiotic residue from wastewater effluents. The current study presented the potential of Ag nanoarray sensors for rapid, highly specific, and cost-effective analysis of pharmaceutical products for environmental remediation applications.

Abstract:4-Carboxyphenylboronic acid was used as the single precursor to facilely prepare fluorescent carbon quantum dots by one-step solvothermal method. The as-obtained carbon dots (CDs) exhibited highly selective and sensitive for benzo[a]pyrene (BaP), and may be a splendid sensor for sensing BaP. The principle was that the as-prepared CDs could form a complex with BaP through hydrophobic interaction which causes the decrease of fluorescence intensity of CDs by static quenching principle. The constructed fluorescent sensor exhibited excellent linearity ranged from 0.002 to 0.06 μg mL−1 and provided a low limit of detection of 0.16 ng mL−1. The experimental results showed that this fluorescent sensor resulted in simplicity, rapidness, low cost, short analytical time, and high sensitivity and stability. Validation with real water samples endowed the sensor high reliability and feasibility for BaP determination in practical application in various samples.

Abstract:Given the prevalence of food safety, online monitoring of food quality is essential. Surface-enhanced Raman scattering (SERS) has excellent sensitivity and molecular fingerprinting capabilities in analytical fields, but its accuracy in food safety monitoring is severely constrained, particularly for gaseous molecules. To further develop the SERS technique in food sensing, in this work, a slippery liquid-infused porous surface (SLIPS) platform was developed for the real-time monitoring of the change in gaseous molecules in shrimp spoilage processes. In order to monitor the change in pH and gaseous biogenic amine molecules (BAs), 4-mercaptopyridine (4-Mpy) and 4-mercaptobenzaldehyde (4-MBA)-functionalized ZIF-8-encapsulated gold nanostars (AuNS@ZIF-8) were utilized as response probes, respectively. Due to the superior gaseous molecule trapping ability of ZIF-8 and the excellent enrichment effect of SLIPS substrates, the use of 4-Mpy and 4-MBA-functionalized AuNS@ZIF-8-SLIPS substrates exhibited excellent online SERS sensing performance for pH and gaseous putrescine molecules. The detection ranges for pH and gaseous BAs were 4.0-9.0 and 10-7-10-3 (v/v) with RSDs of 4.1% and 4.2%, respectively. Furthermore, the SERS monitoring platform was used to monitor shrimp spoilage at 25 °C and 4 °C in real time. Hence, the AuNS@ZIF-8-SLIPS membrane strategy can serve as a promising alternative to ensure accurate, real-time, and non-destructive monitoring of gaseous molecules for food freshness.
