Abstract:Alginate (Alg) hydrogels have demonstrated great potential in drug delivery, wound dressings, and tissue engineering. However, the practical applications of conventional Alg hydrogels suffer from rapid degradation, insufficient controlled drug release, and poor adhesion to tissues. In this paper, the physicochemical properties of Alg hydrogels were comprehensively modulated through chemical modification and the incorporation of various polymers. The results showed that introducing neutral polymers stabilized Alg hydrogels at room temperature. However, this stabilizing effect did not persist at 37 °C. The cationic chitosan (CS) can bind to Alg through electrostatic interaction. The incorporation of CS substantially improved the stability of Alg hydrogels and remarkably prolonged the release of BSA from hydrogels to 14 days at 37 °C. Meanwhile, introducing dopamine (DA)-modified Alg enhanced the adhesion of hydrogel to porcine skin. The optimized hydrogels were utilized to encapsulate platelet-rich plasma (PRP). The PRP-loaded composite hydrogel (2 wt% Alg, with a Ca2+/carboxyl group molar ratio of 0.2, a DA/carboxyl group molar ratio of 0.2, and a CS content of 30 % of Alg) exhibited an excellent therapeutic effect in the healing of rabbit shoulder cuff injury. These findings provide valuable insights into the modulation of physicochemical properties of hydrogels for biomedical applications.
Hybrid 3D printing and electrodeposition approach for controllable 3D alginate hydrogel formation

Abstract:Calcium alginate hydrogels are widely used as biocompatible materials in a substantial number of biomedical applications. This paper reports on a hybrid 3D printing and electrodeposition approach for forming 3D calcium alginate hydrogels in a controllable manner. Firstly, a specific 3D hydrogel printing system is developed by integrating a customized ejection syringe with a conventional 3D printer. Then, a mixed solution of sodium alginate and CaCO3 nanoparticles is filled into the syringe and can be continuously ejected out of the syringe nozzle onto a conductive substrate. When applying a DC voltage (∼5 V) between the substrate (anode) and the nozzle (cathode), the Ca2+ released from the CaCO3 particles can crosslink the alginate to form calcium alginate hydrogel on the substrate. To elucidate the gel formation mechanism and better control the gel growth, we can further establish and verify a gel growth model by considering several key parameters, i.e., applied voltage and deposition time. The experimental results indicate that the alginate hydrogel of various 3D structures can be formed by controlling the movement of the 3D printer. A cell viability test is conducted and shows that the encapsulated cells in the gel can maintain a high survival rate (∼99% right after gel formation). This research establishes a reliable method for the controllable formation of 3D calcium alginate hydrogel, exhibiting great potential for use in basic biology and applied biomedical engineering.

Abstract:Numerous efforts have been made to prepare hydrogels with injectability, adhesivity and osteogenic activity for bone regeneration. However, current hydrogels with these characteristics have limited application in clinical translation due to their complex chemical compositions, which causes difficulty in batch production. In this study, a multifunctional hydrogel with a binary component is prepared for bone regeneration. Dopamine (DA) is first grafted to alginate (Alg) by amidation reaction to give Alg-DA. Then an injectable hydrogel is facilely prepared by mixing strontium ions with the Alg-DA aqueous solution, utilizing the dynamic ionic bonds between the strontium ions and carboxyl groups of Alg-DA. Under the simulated oxidation conditions in vivo, the injectable hydrogel can form stable chemical crosslinkages owing to the presence of catechol groups on Alg-DA. In addition, the catechol groups endow the hydrogel with significant tissue adhesivity. More importantly, the strontium ions endow the hydrogel with osteogenic activity. As indicated by animal experiments, the strontium containing hydrogel scaffolds help to treat rat bone defects within 8 weeks compared to hydrogels without strontium and control groups. This simple hydrogel constructed by using a binary component has achieved several necessary functions, and is expected to be used as a scaffold for bone tissue engineering in clinical applications.

Abstract:We report a dual ionic cross-linking approach for the preparation of double-network hydrogels with robustness, high strength, and toughness, sodium alginate/poly(acrylamide-co-acrylic acid)/Fe3+ (SA/P(AAm-co-AAc)/Fe3+), in a facile "one-step" dual ionic cross-linking method. We take advantage of the abundant carboxyl groups on alginate molecules and the copolymer chains and their high coordination capacity with multivalent metal ions to obtain hydrogels with high strength and toughness. The optimal SA/P(AAm-co-AAc)/Fe3+ (SA 2 wt % and AAc S mol %) hydrogels showed a remarkable mechanical performance with 3.24 MPa tensile strength and 1228% strain, both of which remained stable with 76% water content and were highly swelling resistant in an aqueous environment. The hydrogels possessed high fatigue resistance, self recovery, pH-triggered healing capability, shape memory, and reversible gel sol transition facilitated by pH regulation. Moreover, they show three-dimensional (3D) printing processability by properly adjusting the solution viscosity. The approach may provide a convenient way of obtaining hydrogels having high strength and toughness with a number of desirable properties for a broad range of biomedical applications.

Abstract:Biomaterials composed of food polysaccharides are of great interest for future biomedical applications due to their great biocompatibility, tunable mechanical properties, and complex architectural designs that play a crucial role in the modulation of cell adhesion and proliferation. In this work, a facile approach was designed to obtain novel 3D alginate-CaCO 3 hybrid hydrogel particles in situ. Controlling the gel concentration from 3 to 20 mg·mL −1 allows us to control the alginate-CaCO 3 hydrogel particles' size and density (size variation from 1.86 to 2.34 mm and density from 1.22 to 1.29 mg/mm 3 ). This variable also has a considerable influence on the mineralization process resulting in CaCO 3 particles with varied sizes and amounts within the hydrogel beads. The measurements of Young's modulus showed that the inclusion of CaCO 3 particles into the alginate hydrogel improved its mechanical properties, and Young's modulus of these hybrid hydrogel particles had a linear relationship with alginate content and hydrogel particle size. Cell experiments indicated that alginate-CaCO 3 hybrid hydrogel particles can support osteoblastic cell proliferation and growth. In particular, the amount of hydroxyapatite deposition on the cell membrane significantly increased after the treatment of cells with hybrid hydrogel particles, up to 20-fold. This work offers a strategy for constructing inorganic particle-doped polysaccharide hybrid hydrogel scaffolds that provide the potential to support cell growth.

Abstract:Hydrogels are increasingly used in medicine due to their potential to be delivered into the body in a minimally invasive manner and to be gelated at the site of introduction subsequently. The aim of this study was to develop a novel injectable and in situ-forming gel composite (GC) comprised of calcium alginate hydrogel and nano-hydroxyapatite/collagen (nHAC), assess its rheological, mechanical and in vitro degradable properties, and discuss the gelation mechanism. Injectable property test showed that the injectability of GC was tunable. Rheological results indicated that three phases of pre-gel, sol–gel phase transformation and post-gel could be found in the process of gelation. The compressive elastic modulus ( E ) and shear modulus ( G ) are in the range of 17.0–56.0 kPa and 24.7–55.0 kPa, respectively. During the in vitro degradation, the wet weight increased in the first week, then declined in the following 3 weeks, but the dry weight lost continuously during whole study. Meanwhile, the surface changed greatly after 2 weeks, but samples did not break down up to 28 days. These data indicate that GC exhibits controllable initial setting time and final setting time, tunable injectability, which provides a possible injectable material for bone repair and bone tissue engineering.
Preparation and Physicochemical Properties of an Injectable Alginate-Based Hydrogel by the Regulated Release of Divalent Ions Via the Hydrolysis of D-Glucono-δ-lactone

Abstract:In this study, an injectable dopamine-modified alginate hydrogel combined with nano-hydroxyapatite or strontium hydroxyapatite was cross-linked via the in situ slow release of divalent ions from the hydrolysis of D-glucono-delta-lactone (GDL). The cross-linking mechanism involved the chelation of an alginate with divalent ions and the hydrogen-bonding interaction between alginate and catechol groups. As the GDL concentration at 0.5% (w/v) regulated the release of the divalent ions, which enhanced the chelation effect, it was investigated, and inorganic nanoparticles were homogeneously distributed into the hydrogel system to improve the mechanical properties and stabilize the hydrogel network. Furthermore, the effects of dopamine on the improvement of the mechanical properties were investigated. Results demonstrated that the compressive strength of this injectable hydrogel is enhanced by 4.6 times compared with that of the hydrogel without the addition of GDL, and the compressive strength is enhanced by 4.0 times compared with that of the unmodified alginate hydrogel at strain values ranging from 10% to 50%. The hydrogels combined with strontium hydroxyapatite exhibited a more compact, suitable pore size, with a three-dimensional network structure, and the particle size was 50-100 mu m; this range of values is considered as optimal for the growth of bone tissues. In addition, the gelation time can be adjusted from a few minutes to a few seconds by adjusting experiment variables. Our study provides a potential method for improving the mechanical properties of bone tissue engineering materials. Abstract:We report the ultrastiff and tough poly(acrylamide-co-acrylic acid)/Na-alginate/Fe3+ (P(AM-co-AA)/Na-alginate/Fe3+) hydrogel via the formation of hybrid ionic-hydrogen bond cross-linking networks. The optimal P(AM-co-AA)/Na-alginate/Fe3+ hydrogel possessed super high elastic modulus (similar to 24.6 MPa), tensile strength (similar to 10.4 MPa), compression strength (similar to 44 MPa), and toughness (similar to 4800 J/m(2)). The P(AM-co-AA)/Na-alginate/Fe3+ hydrogel was highly stable and maintained its superior mechanical properties in 0.5-2 M NaCl solution, aqueous solution with pH ranging from 4 to 10. The ionic cross-linking networks of the P(AM-co-AA)/Na-alginate/Fe3+ hydrogels can be locally and selectively dissociated by treating with aqueous NaOH solution with pH of 13 for 1 min and reformed by locally adding the additional Fe3+ solutions, making the hydrogels healable and cohesive. The healed hydrogels from the cutting surfaces can bear a tensile strength of up to 7.1 MPa. Various complex hydrogel structures can be constructed by using the P(AM-co-AA)/Na-alginate/Fe3+ hydrogels as building blocks via the adhesion of as-prepared hydrogels.

Abstract:Dual-crosslinked alginate/carboxymethyl chitosan (CMC) based hydrogels containing in situ synthesized calcium phosphate (CaP) particles were developed by combining two physical crosslinking, the ionic crosslinking of alginate with divalent cation (Ca2+), and the electrostatic interaction between cation of amino groups on CMC and anion of carboxyl groups on alginate. The calcium phosphate particles were in situ synthesized by using the excess Ca2+ of ionic crosslinking. The developed hydrogels were investigated the rheological properties, swelling behavior, degradation, and drug released performance in both water-soluble and insoluble drug models. The results showed that the viscoelasticity and swelling capacity of dual-crosslinked hydrogels were enhanced comparing the single-crosslinked alginate-based hydrogel. The in situ gelation of the developed hydrogel provided the injectable ability and drug-encapsulated capacity. Beside, calcium phosphate, including hydroxyapatite (HAp) was in situ synthesized during the gelation. Biocompatibility of this injectable hydrogel was studied by encapsulation behavior of human adipose-derived stem cells (ASCs). With these qualities, the developed hydrogel may have potential for medical providers such as hydrogel injections and drug providers.

Abstract:The controllable crosslinking between the constituting building blocks plays a key role in endowing the hydrogel with injectability through the formation of a uniform 3D interconnected network. Herein, a uniform-unsaturated crosslinking strategy has been devised to quickly construct injectable sodium alginate (SA) hydrogels. Under vigorous stirring, a moderate amount of metal ions can uniformly coordinate with the guluronate moieties of SA molecules, avoiding the locally excessive crosslinking and the loss of injectability caused by traditional dropping and soaking methods. The injectability of SA hydrogels can be regulated by easily adjusting the concentration of metal ions, and 0.2% (w/v) is the optimal concentration of CaCl 2 for the preparation of injectable SA-Ca hydrogel. Meanwhile, multiple metal ions mediated crosslinking also has been achieved conveniently, expanding the functions. Importantly, SA hydrogels can function as the general platform of composites made of various molecules and materials, for targeting drug delivery, tissue repair, wound infection treatment, and so on. Further, injectable SA-0.2%Cu hydrogel as a model to treat wound infections can promote the healing of full-thickness skin. This study provides a super facile and universal strategy to prepare various SA injectable hydrogels with low cost, which hold great potential in biomedical applications and clinical transformation.

Abstract:Simultaneously enhancing the strength and toughness of physically crosslinked hydrogels is imperative as structural materials, especially for those from natural sources. Herein, inspired by the principle of synergistic effect, a facile yet effective strategy is developed to construct the two-dimensional planar decorated 3D network (2D-3D) in ionic-crosslinked alginate-based hydrogels, generating the strong synergistic mechanics. The strategy is based on a novel hybrid crosslinker (trivalent/divalent cations, e.g. Fe3+/Ca2+) with very low percentage of Fe3+ (1%), which surprisingly imparts multivalent cations with a balanced complexation competition in the hydrogels and thus co-joints loose 2D and rigid 3D structure units in a tightest matching manner. Using the approach in popular Ca-alginate/polyacrylamide system, the complementary architecture induces a 6-fold increase in yield strength accompanied with improved toughness, breaking its mechanical limitations. By varying the molar percentage of each cation in the primary crosslinker, their association abilities with the hydrogels are precisely tuned, giving architectures and mechanics over a wide range. Additionally, the diverse categories of hybrid crosslinkers become the ideal candidates for the development of programmable stimuli-responsive hydrogels, e.g., interesting reverse and unidirectional shape transformations. The method offers a new path in designing metal–ligand coordinated materials, which will certainly become popular in fabricating new generation of alginate-based hydrogels.

Abstract:Hydrogels from natural polysaccharides are of great interest for tissue engineering. This study aims (1) to prepare hydroxyapatite‐loaded macroporous calcium alginate hydrogels by novel one‐step technique using internal gelation in water‐frozen solutions; (2) to evaluate their physicochemical properties; (3) to estimate their ability to support cell growth and proliferation in vitro. The structure of the hydrogel samples in a swollen state was studied by confocal laser scanning microscopy and was shown to represent a system of interconnected macropores with sizes of tens micron. The swelling behavior of the hydrogels, their mechanical properties (Young's moduli) in function of a hydroxyapatite content (5–30 mass%) were studied. All hydrogel samples loaded with hydroxyapatite were found to support growth and proliferation of mouse fibroblasts (L929) at long‐term cultivation for 7 days. The obtained macroporous composite Ca‐Alg‐HA hydrogels could be promising for tissue engineering.

Abstract:Natural-derived hydrogels are expected as promising structural biomaterials, but the soft character severely limits their applications. Here, a facile yet effective strategy was developed to fabricate super-strong and tough alginate composite hydrogels via a self-reinforcing method. The strategy was based on the incorporation of alginate materials with distinctive anisotropic features (fibers, fabrics and aerogels) into the precursor solution of congeneric hydrogels, followed by the in situ ionic-crosslinking. Interestingly, triggered by the concentration difference, the cations-Ca2+ in reinforcing phase could diffuse into the interface and simultaneously chelate with alginate chains of both reinforcing phase and hydrogel matrix, acting as self-generating interfacial binders. Contributed by the intimate interface, the load was effectively transferred into the rigid reinforcing phase, and the hydrogels integrated them into a mechanical network. This research offers a new path to design the interface of polysaccharide composites without extra coupling agents.

Abstract:Alginate (Alg) hydrogels have attracted extensive attention in the biomedical field due to their biocompatibility. However, single Alg hydrogels exhibit weak mechanical strength, poor stability and cell adhesion, which severely restricts their biomedical application. For this reason, we designed alginate/gelatin/cellulose nanocrystals (Alg/G/CNCs) composite hydrogels by combining interpenetrating network (IPN) technology, cellulose nanocrystals (CNCs) reinforcement and in situ cross-linking method to improve the functional defects of Alg hydrogels. The structure and properties of the resultant Alg/G/CNCs composite hydrogels were comprehensively evaluated by FT-IR, TGA, XRD, swelling and degradability measurements, and cytocompatibility experiments. Alg/G/CNCs composite hydrogels with regular three-dimensional porous network (3D) structures was successfully fabricated through the ionic cross-linking of alginate and the covalent cross-linking of gelatin, followed by the reinforcement of colloidal cellulose nanocrystals (CNCs) that were prepared by sulfuric acid hydrolysis of microcrystalline cellulose (MCC). The addition of CNCs could generate interaction force with the polymer in the IPN matrix, which was able to regulate the physicochemical properties of the composite hydrogel to a certain extent. Moreover, with the increase of gelatin (G) content, the compressive strength of Alg/G/CNCs composite hydrogels gradually increased, while the swelling property decreased gradually. Meanwhile, Alg/G/CNCs composite hydrogels exhibited good cell adhesion, proliferation and differentiation properties. In particular, Alg/0.5G/CNCs composite hydrogels displayed the best cell proliferation effect, while Alg/2G/CNCs composite hydrogels revealedthe most significant cell differentiation effect. Therefore, Alg/G/CNCs composite hydrogels could exhibit good mechanical properties and biocompatibility, which possessed great application potential in the field of tissue engineering.

Abstract:Injectable hydrogels are of great interest in tissue engineering, and those incorporating hydroxyapatite (HA) are especially acclaimed in the application of bone repair. Synthetic micro-HA were generally used for this purpose and in some cases, surface modification of HA was further applied to improve the interfacial compatibility of rigid inorganic HA with soft organic matrix. In this study, the injectable hydrogels based on oxidized alginate hybrid HA nanoparticles and carboxymethyl chitosan were achieved via Schiff base reaction. Physicochemical characterization confirmed that oxidized HA/Alg hybrids (OHAH) were successfully prepared. Rheological measurements verified the formation of hydrogels based on the dynamic imine bonding, and the gelation time showed a negative correlation to the concentration and oxidation time of OHAH, while the storage moduli exhibited a positive correlation. The self-healing property of these hydrogels was validated by the splicing experiments and rheological experiments. The lyophilized hydrogels showed porous structures with numerous HA nanoparticles distributed on the surface of pore wall. MTT assays and live/dead staining of cell experiments confirmed the cytocompatibility of these hydrogels. Transforming the Stability, Encapsulation, and Sustained Release Properties of Calcium Alginate Beads through Gel-Confined Coacervation

Abstract:Calcium alginate (Ca2+/alginate) gel beads find use in diverse applications, ranging from drug delivery and tissue engineering to bioprocessing, food formulation, and agriculture. Unless modified, however, these gels have limited stability in alkaline media (including phosphate buffers), and their high solute permeability limits their ability to efficiently encapsulate and slowly release water-soluble small molecules. Here, we show how these limitations can be addressed by mixing the alginate solutions used in the bead preparation with the nontoxic anionic polymer polyphosphate (PP). Upon complexing Ca2+ ions, PP undergoes complex coacervation (i.e., liquid/liquid phase separation into a Ca2+/PP-rich coacervate phase and a dilute supernatant phase). At lower PP concentrations, the Ca2+/PP coacervate appears to simply remain dispersed within the beads. Though its presence makes the beads more stable in alkaline media (phosphate-buffered saline and seawater), it has little impact on the bead stiffness, morphology, and (at least in the absence of substantial payload/coacervate association) encapsulation and release properties. When the PP concentrations exceed a critical value, however, Ca2+/PP coacervation within the gelling Ca2+/alginate beads collapses the resulting beads into more compact, interpenetrating polymer networks. Besides their enhanced stability to alkaline environments, these hybrid beads exhibit irregular morphologies with wrinkled and dimpled surface structures and macroscopic (closed) internal pores, and their collapse into these polymer-rich networks also makes them significantly stiffer than their PP-free counterparts. Crucially, these beads also exhibit a much lower solute permeability, which enables highly efficient encapsulation and multiday release of water-soluble small molecules (with the beads encapsulating >90% of the added model payload and sustaining its release over 3-5 d). Collectively, these findings provide a mild and simple (single-step) pathway to generating ionically cross-linked alginate beads with significantly enhanced stability, encapsulation efficiency, and sustained release.

Abstract:Weak performance of hydrogel fabricated by a sole component limits their applications. In this study, we proposed a facile preparation of hydrogels by using three natural polymers, carboxymethyl chitosan, alginate and agarose. The fabricated carboxymethyl chitosan/sodium alginate/agarose hydrogels were further treated by divalent calcium ions to form a double crosslinked network inside. These double crosslinked hydrogels showed tunable mechanical property and the compressive strength ranged from 176 Pa to 685 Pa by varying the crosslinking degree. They also exhibited excellent swelling behavior (more than 1500%), good degradability and good biocompatibility, indicating their potential for application in wound healing, drug delivery and bone tissue engineering.

Abstract:Alginate hydrogels have the most extensively explored injection system among natural molecular. The injectable alginate hydrogel has special physical and biological properties without toxicity, therefore, it has been served as minimally invasive treatments to treat various irregular defects. However, the inconvenient truth is low formability and poor mechanical properties of injectable alginate hydrogels; besides, there are little relevant studies about controlling, measurement and analysis of the sol-gel transition and degradation process. We synthesized an injectable nanocomposite alginate hydrogel, used specific methods and equipment to regulate and evaluate the sol-gel transition of the hydrogel, moreover, we researched the degradation behaviour using scanning electron microscope, nano-indenter and titration analysis. The results showed that gelation time and mechanical properties could be regulated and effectively described. Compression properties of the hydrogels increased to 150%–300% compared with common alginate hydrogels, in addition, the degradation behaviour with ions release could be regulated by the formula. The results will be propitious to solve problems in quality control of hydrogel manufacturing engineering and the nanocomposite hydrogels with good biocompatibility will have great potential for multifunctional biological scaffolds and tissue engineering.

Abstract:In situ injectable hydrogels are effectively employed to fill irregular cavitary bone defects with initiating bone growth in targeted areas. Herein, an injectable composited hydrogel composed of collagen and alginate cross-linked in situ using different concentrations of calcium sulfate (0.15, 0.3 and 0.6%, wt./v) was synthesized. Recently, CaSO4 is frequently supported as a bone graft material for bone regeneration, owing to its biocompatibility and osteoconductive properties. Moreover, hydroxyapatite (Hap) after salinization-step by (3-Aminopropyl) triethoxysilane (APTES) was incorporated for further enhancing the osteoconductive property of injected hydrogels. All fabricated hydrogels were characterized by SEM, FTIR and XRD analyses. While physiochemical characteristics of hydrogels were assessed through swelling index, hydrolytic degradability and thermal stability measurements. In vitro bio-assessments, e.g., antimicrobial activity, cytotoxicity and cell adhesion tests using osteoblast-like cells (MG-63) were investigated. Results showed that addition of Hap offered better control of gelation time and formed uniform hydrogels, additionally improved significantly thermal stability, which leads to hindering of swelling index, prolonging hydrolytic degradability rates and significantly enhanced the antimicrobial activity of hydrogel; compared to hydrogel free-Hap. Hap-loaded Col–Alg–CaSO4 hydrogel with the highest concentration of CaSO4 recorded an enrichment of cell viability among all hydrogel samples. Notably, In vitro cell adhesion test showed that MG-63 cells adhered adequately with all hydrogels. The results support the approach of using an injectable Hap-loaded Col/Alg hydrogel cross-linked with CaSO4 as an alter and novel technique to enhance bone tissue regeneration, host–implant integration, quick/simple technique and easier for clinical handling.
