Abstract:Magnesium ions (Mg2+) are bioactive and proven to promote bone tissue regeneration, in which the enhancement efficiency is closely related to Mg2+ concentrations. Currently, there are no well-established bone tissue engineering scaffolds that can precisely control Mg2+ release, although this capability could have a marked impact in bone regeneration. Leveraging the power of biodegradable microspheres to control the release of bioactive factors, we developed lactone-based biodegradable microspheres that served as both injectable scaffolds and Mg2+ release system for bone regeneration. The biodegradable microsphere (PMg) was prepared from poly(lactide-co-glycolide) (PLGA) micro-spheres co-embedded with MgO and MgCO3 at a fixed total loading amount (20 wt%) with different weight ratios (1:0; 3:1; 1:1; 1:3; 0:1). The PMg microspheres demonstrated controlled release of Mg2+ by tuning the MgO/MgCO3 ratios. Specifically, faster release with higher initial concentrations of Mg2+ were detected at higher MgO fractions, while long-term sustained release with lower concentrations of Mg2+ was obtained at higher MgCO3 fractions. All prepared PMg microspheres were non-cytotoxic. Furthermore, they promoted attachment, proliferation, osteogenic differentiation, especially, cell migration of bone marrow mesenchymal stromal cells (BMSCs). Among these microspheres, PMg-III microspheres (MgO/MgCO3 in 1:1) exhibited the strongest promotion of mineral depositions and osteogenic differentiation of BMSCs. PMg-Ill microspheres were injected into the critical-sized calvarial defect of a rat model, resulting in significant bone regeneration when compared to the control group filled with PLGA microspheres. In the PMg-Ill group, the new bone volume fraction (BV/TV) and bone mineral density (BMD) reached 32.9 +/- 5.6% and 325.7 +/- 20.2 mg/cm(3), respectively, which were much higher than the values 8.1 +/- 2.5% (BV/TV) and 124 +/- 35.8 mg/cm(3) (BMD) in the PLGA group. These findings indicated that bioresorbable microspheres possessing controlled Mg2+ release features were efficient in treating bone defects and promising for future in vivo applications. Statement of Significance Magnesium plays pivotal roles in regulating osteogenesis, which exhibits concentration-dependent behaviors. However, no generally accepted controlled-release system is reported to correlate Mg2+ concentration with efficient bone regeneration. Biodegradable microspheres with injectability are excellent cell carriers for tissue engineering, moreover, good delivery systems for bioactive factors. By co-embedding magnesium compounds (MgO, MgCO3) with different dissolution rates in various ratios, tunable release of Mg2+ from the microspheres was readily achieved. Accordingly, significant promotion in bone defect regeneration is achieved with microspheres displaying proper sustained release of Mg2+. The developed strategy may serve as valuable guidelines for bone tissue engineering scaffold design, which allows precise control on the release of bioactive metal ions like Mg2+ toward potential clinical translation. Abstract:Imitating the microenvironment of bone formation is crucial for the design of bone defect repair materials. In this study, cell mono-culture and co-culture experiments were carried out using rat aorta endothelial cells (rAECs) and rat bone marrow mesenchymal stem cells (rBMMSCs) under the addition of increasing concentrations of magnesium chloride. The optimal magnesium ion (Mg 2+ ) concentration for promoting in vitro vascularized osteogenesis was found to be 5 mM, which was possibly caused by the promotion of the PI3K-AKT signaling pathway. Accordingly, we designed a Mg 2+ -enriched Alginate photocrosslinked hydrogel (MAH) which could steadily release Mg 2+ . And, this modified Mg 2+ -enriched MAH possessed excellent. It shows excellent ability to promote osteogenesis in the rat calvarial defect models.

Abstract:The main goal of this study was to develop a novel poly (lactic-co-glycolic acid) (PLGA) composite biodegradable material with magnesium (Mg) metal to overcome the acidic degradation of PLGA and to investigate the cytocompatibility and osteogenesis of the novel material. PLGA composites with 5 and 10 wt% Mg were prepared. The samples were initially cut into 10 mm × 10 mm films, which were used to detect the pH value to evaluate the self-neutralized ability. Murine embryo osteoblast precursor (MC3T3-E1) cells were used for in vitro experiments to evaluate the cytotoxicity, apoptosis, adhesion, and osteogenic differentiation effect of the composite biodegradable material. pH monitoring showed that the average value of PLGA with 10 wt% Mg group was closer to the normal physiological environment than that of other groups. Cell proliferation and adhesion assays indicated no significant difference between the groups, and all the samples showed no toxicity to cells. As for cell apoptosis detection, the rate of early apoptotic cells was proportional to the ratio of Mg. However, the ratios of the experimental groups were lower than those of the control group. Alkaline phosphatase activity staining demonstrated that PLGA with 10 wt% Mg could effectively improve the osteogenic differentiation of MC3T3-E1 cells. In summary, PLGA with 10 wt% Mg possessed effective osteogenic properties and cytocompatibility and therefore could provide a wide range of applications in bone defect repair and scaffold-based tissue engineering in clinical practice.

Abstract:Hydrogel scaffolds are promising and widely applicable platforms for various therapeutic agents to facilitate bone tissue regeneration due to their biocompatibility and low immunogenicity. Nevertheless, the improvement of local administration efficiency and on-demand release of drugs from a hydrogel system is still an obstacle. In this work, we reported that a novel injectable hydrogel system was fabricated based on coordination of multiarm thiolated polyethylene glycol (PEG-SH) and magnesium ions for bone marrow-derived mesenchymal stem cell (BMSC) proliferation and differentiation. The dynamic nature coordination bond of Mg–S and the dynamic disulfide bond of S–S provide hydrogels with good mechanical performance and typical rheological behavior and thus endow the hydrogels with a satisfactory swelling rate and degradation property. Mg2+ was incorporated in the system not only to act as an effective cross-linker to enhance the hydrogel network structure but also to mediate the sustained release of Mg2+. Due to the controlled release of Mg2+, the PEG-SH/Mg2+ hydrogel can effectively improve BMSC proliferation and osteoblastic activity via the PI3K/Akt/GSK3β/β-catenin signal pathway in vitro. These findings indicated that the novel hydrogel controlled release of a Mg2+ ion is viewed as a promising and flexible platform for bone regeneration clinically.This article has not yet been cited by other publications.

Magnesium Oxide Nanoparticles Reinforced Electrospun Alginate-Based Nanofibrous Scaffolds with Improved Physical Properties

Abstract:Mechanically robust alginate-based nanofibrous scaffolds were successfully fabricated by electrospinning method to mimic the natural extracellular matrix structure which benefits development and regeneration of tissues. Alginate-based nanofibres were electrospun from an alginate/poly(vinyl alcohol) (PVA) polyelectrolyte complex. SEM images revealed the spinnability of the complex composite nanofibrous scaffolds, showing randomly oriented, ultrafine, and virtually defects-free alginate-based/MgO nanofibrous scaffolds. Here, it is shown that an alginate/PVA complex scaffold, blended with near-spherical MgO nanoparticles (⌀ 45 nm) at a predetermined concentration (10% (w/w)), is electrospinnable to produce a complex composite nanofibrous scaffold with enhanced mechanical stability. For the comparison purpose, chemically cross-linked electrospun alginate-based scaffolds were also fabricated. Tensile test to rupture revealed the significant differences in the tensile strength and elastic modulus among the alginate scaffolds, alginate/MgO scaffolds, and cross-linked alginate scaffolds (P<0.05). In contrast to cross-linked alginate scaffolds, alginate/MgO scaffolds yielded the highest tensile strength and elastic modulus while preserving the interfibre porosity of the scaffolds. According to the thermogravimetric analysis, MgO reinforced alginate nanofibrous scaffolds exhibited improved thermal stability. These novel alginate-based/MgO scaffolds are economical and versatile and may be further optimised for use as extracellular matrix substitutes for repair and regeneration of tissues.
Three-Dimensional Printed Poly (Lactic-co-Glycolic Acid)-Magnesium Composite Scaffolds for the Promotion of Osteogenesis Through Immunoregulation

Abstract:Scaffolds play an important role in bone tissue engineering. The ideal engineered scaffold needs to be biocompatible, bioactive, and able to regulate immune cells to enhance bone regeneration. In this study, magnesium (Mg)-contained poly(lactic-co-glycolic acid) (PLGA) scaffolds (hereinafter, referred to as PLGA-2Mg) were fabricated by 3-dimensional printing using a mixture of PLGA and MgSO 4 powder. Poly(lactic-co-glycolic acid) scaffolds (hereinafter, referred to as PLGA) were also fabricated by 3-dimensional printing and were used as control. The biocompatibility, immunoregulatory ability, and osteogenic properties of PLGA-2Mg were analyzed and compared with those of PLGA. The results indicate that the incorporation of Mg increased the Young modulus and surface roughness of the scaffold, but did not affect its degradation. The PLGA-2Mg further promoted the adhesion and proliferation of MC3T3-E1 cells compared with PLGA, which indicates its improved biocompatibility and bioactivity. In addition, PLGA-2Mg inhibited the polarization of RAW 264.7 cells toward the M1 phenotype by down-regulating the IL-1β , IL-6 , and iNOs gene expression when challenged with lipopolysaccharide stimulation. In contrast, it promoted the polarization of RAW 264.7 cells toward the M2 phenotype by up-regulating the TGF-β , IL-10 , and Arg-1 gene expression without lipopolysaccharide stimulation. Finally, MC3T3-E1 cells were cocultured with RAW 264.7 cells and scaffolds using a transwell system. It was found that the expression level of osteogenic-related genes ( ALP , COL-1 , BMP2 , and BSP ) was significantly upregulated in the PLGA-2Mg group compared with that in the PLGA group. Consequently, PLGA-2Mg with increased biocompatibility and bioactivity can promote osteogenesis through immunoregulation and has the potential to be used as a novel scaffold in bone tissue engineering.
A simple hydrogel scaffold with injectability, adhesivity and osteogenic activity for bone regeneration

Abstract:Numerous efforts have been made to prepare hydrogels with injectability, adhesivity and osteogenic activity for bone regeneration. However, current hydrogels with these characteristics have limited application in clinical translation due to their complex chemical compositions, which causes difficulty in batch production. In this study, a multifunctional hydrogel with a binary component is prepared for bone regeneration. Dopamine (DA) is first grafted to alginate (Alg) by amidation reaction to give Alg-DA. Then an injectable hydrogel is facilely prepared by mixing strontium ions with the Alg-DA aqueous solution, utilizing the dynamic ionic bonds between the strontium ions and carboxyl groups of Alg-DA. Under the simulated oxidation conditions in vivo, the injectable hydrogel can form stable chemical crosslinkages owing to the presence of catechol groups on Alg-DA. In addition, the catechol groups endow the hydrogel with significant tissue adhesivity. More importantly, the strontium ions endow the hydrogel with osteogenic activity. As indicated by animal experiments, the strontium containing hydrogel scaffolds help to treat rat bone defects within 8 weeks compared to hydrogels without strontium and control groups. This simple hydrogel constructed by using a binary component has achieved several necessary functions, and is expected to be used as a scaffold for bone tissue engineering in clinical applications.
Preparation and Physicochemical Properties of an Injectable Alginate-Based Hydrogel by the Regulated Release of Divalent Ions Via the Hydrolysis of D-Glucono-δ-lactone

Abstract:In this study, an injectable dopamine-modified alginate hydrogel combined with nano-hydroxyapatite or strontium hydroxyapatite was cross-linked via the in situ slow release of divalent ions from the hydrolysis of D-glucono-delta-lactone (GDL). The cross-linking mechanism involved the chelation of an alginate with divalent ions and the hydrogen-bonding interaction between alginate and catechol groups. As the GDL concentration at 0.5% (w/v) regulated the release of the divalent ions, which enhanced the chelation effect, it was investigated, and inorganic nanoparticles were homogeneously distributed into the hydrogel system to improve the mechanical properties and stabilize the hydrogel network. Furthermore, the effects of dopamine on the improvement of the mechanical properties were investigated. Results demonstrated that the compressive strength of this injectable hydrogel is enhanced by 4.6 times compared with that of the hydrogel without the addition of GDL, and the compressive strength is enhanced by 4.0 times compared with that of the unmodified alginate hydrogel at strain values ranging from 10% to 50%. The hydrogels combined with strontium hydroxyapatite exhibited a more compact, suitable pore size, with a three-dimensional network structure, and the particle size was 50-100 mu m; this range of values is considered as optimal for the growth of bone tissues. In addition, the gelation time can be adjusted from a few minutes to a few seconds by adjusting experiment variables. Our study provides a potential method for improving the mechanical properties of bone tissue engineering materials. Magnesium Oxide Nanoparticle Coordinated Phosphate-Functionalized Chitosan Injectable Hydrogel for Osteogenesis and Angiogenesis in Bone Regeneration

Abstract:Natural polysaccharide (NPH)-based injectable hydrogels have shown great potential for critical-sized bone defect repair. However, their osteogenic, angiogenic, and mechanical properties are insufficient. Here, MgO nanoparticles (NPs) were incorporated into a newly synthesized water-soluble phosphocreatine-functionalized chitosan (CSMP) water solution to form an injectable hydrogel (CSMP-MgO) via supramolecular combination between phosphate groups in CSMP and magnesium in MgO NPs to circumvent these drawbacks of chitosan-based injectable hydrogels. Water-soluble chitosan deviate CSMP was first synthesized by grafting methacrylic anhydride and phosphocreatine into a chitosan chain in a one-step lyophilization process. The phosphocreatine in this hydrogel not only provides sites to combine with MgO NPs to form supramolecular binding but also serves as the reservoir to control Mg2+ release. As a result, the lyophilized CSMP-MgO hydrogels presented a porous structure with some small holes in the pore wall, and the pore diameters ranged from 50 to 100 μm. The CSMP-MgO injectable hydrogels were restricted from swelling in DI water (lowest swelling ratio was 16.0 ± 1.1 g/g) and presented no brittle failure during compression even at a strain above 85% (maximum compressive strength was 195.0 kPa) versus the control groups (28.0 and 41.3 kPa for CSMP and CSMP-MgO (0.5) hydrogels), with regulated Mg2+ release in a stable and sustained manner. The CSMP-MgO injectable hydrogels promoted in vitro calcium phosphate (hydroxyapatite (HA) and tetracalcium phosphate (TTCP)) deposition in supersaturated calcium phosphate solution and presented no cytotoxicity to MC3T3-E1 cells; the CSMP-MgO hydrogel promoted MC3T3-E1 cell osteogenic differentiation with upregulation of BSP, OPN, and Osterix osteogenic gene expression and mineralization and HUVEC tube formation. Among them, CSMP-MgO (5) presented most of these properties. Moreover, this hydrogel (CSMP-MgO (5)) showed an excellent ability to promote new bone formation in critical-sized calvarial defects in rats. Thus, the CSMP-MgO injectable hydrogel shows great promise for bone regeneration.

Magnesium-Organic Framework-Loaded Bisphosphonate-Functionalized Gel Scaffolds for Enhanced Bone Regeneration

Abstract:The development of magnesium-derived biomaterials is one of the most promising research in bone tissue engineering, and related strategies have been extensively used for tendon, skull, cartilage, and bone regeneration. Also, alendronate, a well-recognized drug for osteoporosis treatment, has recently attracted a great deal of attention for bone repair. However, rapid corrosion in vivo of Mg2+ and low systemic bioavailability of alendronate are the main limitations hampering their full exploitation. In this work, by means of physical and chemical cross-linking conjugating magnesium-metal-organic frameworks (Mg-MOFs) and bone-targeting alendronate to biocompatible gelatin scaffolds, a facile method is developed for the preparation of organic/inorganic nanocomposite gel scaffolds. The results affirmed that the nanocomposite gel scaffolds possessed excellent biocompatibility, continuous slow release of Mg2+ and alendronate, strong bone affinity, and bone regeneration. It is noteworthy that the continuous slow release of Mg2+ and alendronate could induce the macrophage switch to the M2 phenotype and promote osteogenic differentiation in the early stage, resulting in improved bone regeneration during implanting the scaffolds into the distal femoral. In summary, Mg-MOFs-loaded alendronate-modified gelatin gel scaffolds have been developed, exhibiting great potential for bone regenerative.
Precisely controlled delivery of magnesium ions thru sponge-like monodisperse PLGA/nano-MgO-alginate core-shell microsphere device to enable in-situ bone regeneration

Abstract:A range of magnesium ions (Mg2+) used has demonstrated osteogenic tendency in vitro. Hence, we propose to actualize this concept by designing a new system to precisely control the Mg2+ delivery at a particular concentration in vivo in order to effectively stimulate in-situ bone regeneration. To achieve this objective, a monodisperse core-shell microsphere delivery system comprising of poly (lactic-co-glycolic acid) (PLGA) biopolymer, alginate hydrogel, and magnesium oxide nano-particles has been designed by using customized microfluidic capillary device. The PLGA-MgO sponge-like spherical core works as a reservoir of Mg2+ while the alginate shell serves as physical barrier to control the outflow of Mg2+ at ∼50 ppm accurately for 2 weeks via its adjustable surface micro-porous network. With the aid of controlled release of Mg2+, the new core-shell microsphere system can effectively enhance osteoblastic activity in vitro and stimulate in-situ bone regeneration in vivo in terms of total bone volume, bone mineral density (BMD), and trabecular thickness after operation. Interestingly, the Young's moduli of formed bone on the core-shell microsphere group have been restored to ∼96% of that of the surrounding matured bone. These findings indicate that the concept of precisely controlled release of Mg2+ may potentially apply for in-situ bone regeneration clinically.
Magnesium Hydroxide as a Versatile Nanofiller for 3D-Printed PLA Bone Scaffolds

Abstract:Polylactic acid (PLA) has attracted much attention in bone tissue engineering due to its good biocompatibility and processability, but it still faces problems such as a slow degradation rate, acidic degradation product, weak biomineralization ability, and poor cell response, which limits its wider application in developing bone scaffolds. In this study, Mg(OH)2 nanoparticles were employed as a versatile nanofiller for developing PLA/Mg(OH)2 composite bone scaffolds using fused deposition modeling (FDM) 3D printing technology, and its mechanical, degradation, and biological properties were evaluated. The mechanical tests revealed that a 5 wt% addition of Mg(OH)2 improved the tensile and compressive strengths of the PLA scaffold by 20.50% and 63.97%, respectively. The soaking experiment in phosphate buffered solution (PBS) revealed that the alkaline degradation products of Mg(OH)2 neutralized the acidic degradation products of PLA, thus accelerating the degradation of PLA. The weight loss rate of the PLA/20Mg(OH)2 scaffold (15.40%) was significantly higher than that of PLA (0.15%) on day 28. Meanwhile, the composite scaffolds showed long-term Mg2+ release for more than 28 days. The simulated body fluid (SBF) immersion experiment indicated that Mg(OH)2 promoted the deposition of apatite and improved the biomineralization of PLA scaffolds. The cell culture of bone marrow mesenchymal stem cells (BMSCs) indicated that adding 5 wt% Mg(OH)2 effectively improved cell responses, including adhesion, proliferation, and osteogenic differentiation, due to the release of Mg2+. This study suggests that Mg(OH)2 can simultaneously address various issues related to polymer scaffolds, including degradation, mechanical properties, and cell interaction, having promising applications in tissue engineering.

Abstract:A new prototype of hybrid silk fibroin and sodium alginate (SF-SA) based osteogenic hydrogel scaffold with a concentration of 2.5% magnesium phosphate (MgP) based gel was prepared with the assistance of an extrusion-based three-dimensional (3D) printing machine in this study. To determine the optimum ratio of MgP-based gel in the hydrogel, a series of physical and biochemical experiments were performed to determine the proper concentration of MgP in two-dimensional hydrogel films, as well as the cell compatibility with these materials in sequence. The SF-SA hydrogel with 2.5wt% magnesium phosphate (SF-SA/MgP) stood out and then was used to fabricate 3D hydrogel scaffolds according to the consequences of the experiments, with SF-SA hydrogel as a control. Then the morphology and osteogenic activity of the scaffolds were further characterized by field emission scanning electron microscope (SEM), calcium mineralization staining, and reverse transcription-polymerase chain reaction (rt-PCR). The SF-SA/MgP hydrogel scaffold promoted the adhesion of rat mesenchymal stem cells with higher degrees of efficiency under dynamic culture conditions. After co-culturing in an osteogenic differentiation medium, cells seeded on SF-SA/MgP hydrogel scaffold were shown to have better performance on osteogenesis in the early stage than the control group. This work illustrates that the 3D structures of hybrid SF-SA/MgP hydrogel are promising headstones for osteogenic tissue engineering.

Abstract:The rising prevalence of bone injuries has increased the demand for minimally invasive treatments. Microbead hydrogels, renowned for cell encapsulation, provide a versatile substrate for bone tissue regeneration. They deliver bioactive agents, support cell growth, and promote osteogenesis, aiding bone repair and regeneration. In this study, we synthesized superparamagnetic iron oxide nanoparticles (Sp) coated with a calcium phosphate layer (m-Sp), achieving a distinctive flower-like micro-cluster morphology. Subsequently, sodium alginate (SA) microbead hydrogels containing m-Sp (McSa@m-Sp) were fabricated using a dropping gelation strategy. McSa@m-Sp is magnetically targetable, enhance cross-linking, control degradation rates, and provide strong antibacterial activity. Encapsulation studies with MC3T3-E1 cells revealed enhanced viability and proliferation. These studies also indicated significantly elevated alkaline phosphatase (ALP) activity and mineralization in MC3T3-E1 cells, as confirmed by Alizarin Red S (ARS) and Von Kossa staining, along with increased collagen production within the McSa@m-Sp microbead hydrogels. Immunocytochemistry (ICC) and gene expression studies supported the osteoinductive potential of McSa@m-Sp, showing increased expression of osteogenic markers including RUNX-2, collagen-I, osteopontin, and osteocalcin. Thus, McSa@m-Sp microbead hydrogels offer a promising strategy for multifunctional scaffolds in bone tissue engineering.
Fabrication and characteristics of anti-inflammatory magnesium hydroxide incorporated PLGA scaffolds formed with various porogen materials

Abstract:Poly(D,L-lactic-co-glycolic acid) (PLGA) has been widely used as a biodegradable polymer in the fabrication of porous polymer scaffolds, but it is hydrolyzed into acidic by-products such as glycolic acid and lactic acid in the human body. Magnesium hydroxide nanoparticles (Mg-NPs) were incorporated into a PLGA scaffold in order to neutralize the acidic environment caused by the hydrolysis of PLGA, thereby reducing the cytotoxicity and inflammatory response. In this study, three-dimensional porous scaffolds blended with 30% Mg-NP were fabricated using gas foaming (PLGA/Mg/NaHCO3), salt leaching (PLGA/Mg/NaCl), and freeze drying (PLGA/Mg/Ice), and their structures, morphologies, pH change, thermal properties, and mechanical properties were analyzed by Fourier transform infrared spectroscopy, scanning electron microscopy, pH meter, thermogravimetric analysis, and a universal testing machine. The porosity of the PLGA/Mg/Ice scaffold was higher at about 10-13 wt% than those of the PLGA/Mg/NaCl or PLGA/Mg/NaHCO3 scaffolds. The Mg-NP content of the PLGA/Mg/NaHCO3 scaffold remained lower than those of the other scaffolds at about 63%. As a result of this loss of Mg-NP, the PLGA/Mg/NaHCO3 scaffold was confirmed to have lower cell viability (about 70%) than the PLGA/Mg/Ice scaffold (about 100%), owing to the reduced neutralizing effect. Although the PLGA/Mg/Ice and PLGA/Mg/NaCl scaffolds showed similar cell viability, the NaCl of the PLGA/Mg/NaCl scaffold exhibited slight toxicity in the body. The expression level of interleukin-6 (IL-6) was significantly decreased in the PLGA/Mg/Ice scaffold than in the PLGA/Ice scaffold, but the PLGA/Mg/Ice scaffold exhibited an IL-6 expression level that was about 10% lower than that of the PLGA/Mg/ NaCl scaffold. Consequently, the addition of Mg-NP/Ice could conceivably reduce the expression level of IL-6 in PLGA scaffolds. This anti-inflammatory PLGA/Mg/Ice scaffold is therefore expected to show great promise when used as a template in tissue engineering.

Abstract:Bone defects caused by disease or trauma are major challenges in clinic. Designing scaffolds capable of releasing bioactive ions and having osteogenic properties hold promise for tissue engineering. In this study, an innovative scaffold was constructed by incorporating magnesium-organic framework (MgGA) and dicalcium phosphate dihydrate (DCPD) into poly (lactic acid- co -glycolic acid) (PLGA) through an electrospinning process. The composite scaffold (P/DCPD/MgGA) with organic–inorganic components and combined with unique porous structure, enabling a long-term and steady supply of bioactive ions. In addition, in vitro assays indicated that composite scaffold show excellent biocompatibility to human bone mesenchymal stem cells (hBMSCs), promote extracellular matrix mineralization and substantially upregulate the expression of osteogenic-related proteins (ALP, Runx2 and OCN). In addition, the P/DCPD/MgGA scaffold facilitated the osteogenic function by activating the Wnt/ β -catenin signaling pathway. Overall, this composite scaffold carrying therapeutic magnesium-organic framework provided a new idea for bone tissue engineering.

Abstract:Highlights • A novel injectable porous hydrogel was developed by doping 1 mg/mL of MPs into CS-GP. SEM, compression, and rheological analyses confirmed that the Mg 2+ -containing hydrogel has a porous internal structure and suitable physical and chemical properties. Experiments verified that this hydrogel has good biocompatibility as the Mg ions can promote cellular osteogenic gene expression in vitro. Using a periodontitis rat model with alveolar bone defect, we demonstrated that this hydrogel can effectively reduce inflammatory cell infiltration, promoting the healing of periodontal soft tissue and connective epithelium. Additionally, the hydrogel facilitated the repair of alveolar bone height and density in vivo. Therefore, this injectable porous hydrogel is not only morphologically adaptable and moldable in situ , but it also possesses both osteogenic and anti-inflammatory effects, rendering it valuable for minimally invasive periodontal treatment. Periodontitis is associated with several systemic diseases, and advanced periodontitis is often linked to an extensive inflammatory microenvironment and irregularly shaped alveolar bone defects. However, eliminating periodontal inflammation in a minimally invasive manner while repairing irregularly shaped bone defects is clinically challenging. In comparison to traditional bone grafts, a thermo-sensitive hydrogel can be injected into deep periodontal pockets, forming and filling the alveolar bone defects in situ . In this study, porous injectable thermo-sensitive hydrogels containing magnesium ions were prepared by adding magnesium particles (MPs) to a glycerophosphate solution and combining this mixture with a chitosan solution. The incorporation of MPs created interconnected pores in the hydrogel, exhibiting high cytocompatibility and maintaining cell viability, proliferation, spreading, and osteogenesis in vitro . Evaluation on an experimental periodontitis rat model, using micro-computed tomography and histological analyses, demonstrated that this Mg 2+ -containing hydrogel effectively reduced periodontal inflammation, inhibited osteoclast activity, and partially repaired inflammation-induced alveolar bone loss. These results suggest that Mg 2+ -containing thermo-sensitive porous hydrogels might be promising candidates for treating periodontitis. Abstract:The osteogenic and angiogenic characteristics are crucial for the selection of bone substitute materials and structural design. This study proposed a porous PLGA/n-HA/MgP composited scaffold by printing of the poly (lactide-coglycolide) (PLGA), nano-hydroxyapatite (n-HA), and magnesium phosphate (MgP). The physicochemical properties, osteogenic activities, and pro-angiogenic impacts of porous 3D printed PLGA/n-HA scaffolds with variable MgP concentration were investigated. The results indicated that the presence of MgP in the composite promoted scaffold degradation and buffered the acidic environment induced by PLGA degradation. The highest pro-osteogenic activity has been observed in PLGA/n-HA/10% MgP (PH10M) group, which up-regulated BMP2 and RUNX2 factors in vitro and in vivo. Nevertheless, at a maximal ion release of 7.2 mmol/L, PLGA/n-HA/30% MgP (PH30M) marginally reduced the osteogenic differentiation of osteoblasts through the RANKL/OPG pathway. In vitro, PLGA/n-HA/20% MgP (PH20M) demonstrated superior migration, pro-angiogenic factor expression, and angiogenesis compared to other groups, and the histological investigation corroborated the improved angiogenesis in PH10M and PH20M. In conclusion, suitable MgP ratio shows the potential to improve the osteogenic and angiogenic properties of PLGA/n-HA-based composites in orthopedic applications.

Effect of contact lens removal or displacement on intraocular pressure.

Abstract:We conducted a controlled, prospective study involving 102 healthy volunteers to determine the effects of contact lens removal and contact lens displacement on subsequent intraocular pressure measurements. Intraocular pressure was measured at baseline, immediately after contact lens removal or displacement, and 5 minutes thereafter. The method of lens removal was either sliding displacement (51 eyes) or plucking (51 eyes). Right eyes were used for the test, and left eyes served as controls. Compared with baseline measurements, mean intraocular pressure immediately after plucking the lens was 0.8 mm Hg lower (P less than .01) and after sliding the lens was 0.5 mm Hg higher (P less than .05). Intraocular pressure changes of 4.0 mm Hg or more occurred in eight (15.6%) of 51 eyes in the pluck group vs four (7.8%) of 51 control eyes and in two (3.9%) of 51 eyes in the slide group vs zero of 51 control eyes. Compared with baseline, significant differences (P less than .05) were not found 5 minutes later. Clinicians who wish to minimize the influence of contact lens removal on tonometry readings should wait several minutes after contact lens removal before measuring intraocular pressure.
