Abstract:A novel AgNPs and PDAs‐based nano platform (Ag/PDAs) offering broad‐spectrum antibacterial activity and biofilm disruption under 808 nm laser irradiation. Ag/PDAs modulate inflammation by affecting macrophage polarization, promoting tissue healing in biofilm‐infected wounds in mice. Due to the presence of spatial barriers, persistent bacteria, and excessive inflammation in bacteria biofilm‐infected wounds, current nanoplatforms cannot effectively address these issues simultaneously during the therapeutic process. Herein, a novel biomimetic photothermal nanoplatform integrating silver and polydopamine nanoparticles (Ag/PDAs) that can damage biofilms, kill bacterial persisters, and reduce inflammation for wound treatment is presented. These findings reveal that Ag/PDAs exhibit a broad‐spectrum antimicrobial activity through direct damage to the bacterial membrane structure. Additionally, Ag/PDAs demonstrate a potent photothermal conversion efficiency. When combined with near‐infrared (NIR) irradiation, Ag/PDAs effectively disrupt the spatial structure of biofilms and synergistically eradicate the resident bacteria. Furthermore, Ag/PDAs show remarkable anti‐inflammatory properties in counteracting bacterium‐induced macrophage polarization. The in vivo results confirm that the topical application of Ag/PDAs significantly suppress Staphylococcus aureus biofilm‐infected wounds in murine models, concurrently facilitating wound healing. This research provides a promising avenue for the eradication of bacterial biofilms and the treatment of biofilm‐infected wounds.

Abstract:Bacterial-associated wound infection and antibiotic resistance have posed a major burden on patients and health care systems. Thus, developing a novel multifunctional antibiotic-free wound dressing that cannot only effectively prevent wound infection, but also facilitate wound healing is urgently desired. Herein, a series of multifunctional nanocomposite hydrogels with remarkable antibacterial, antioxidant, and anti-inflammatory capabilities, based on bacterial cellulose (BC), gelatin (Gel), and selenium nanoparticles (SeNPs), are constructed for wound healing application. The BC/Gel/SeNPs nanocomposite hydrogels exhibit excellent mechanical properties, good swelling ability, flexibility and biodegradability, and favorable biocompatibility, as well as slow and sustainable release profiles of SeNPs. The decoration of SeNPs endows the hydrogels with superior antioxidant and anti-inflammatory capability, and outstanding antibacterial activity against both common bacteria (E. coli and S. aureus) and their multidrug-resistant counterparts. Furthermore, the BC/Gel/SeNPs hydrogels show an excellent skin wound healing performance in a rat full-thickness defect model, as evidenced by the significantly reduced inflammation, and the notably enhanced wound closure, granulation tissue formation, collagen deposition, angiogenesis, and fibroblast activation and differentiation. This study suggests that the developed multifunctional BC/Gel/SeNPs nanocomposite hydrogel holds a great promise as a wound dressing for preventing wound infection and accelerating skin regeneration in clinic.

Abstract:The severe bacterial infection and chronic wound healing caused by the abuse of antibiotics threaten the public health, which calls the need for the development of novel antibacterial agents and alternative therapeutic strategies. Herein, magnetic carboxymethyl cellulose-ε-polylysine hybrids (FCE) were synthesized via a facile one-pot coprecipitation method and further used as matrix to anchor silver nanoparticles (Ag NPs). The as-resulted Ag/FCE hybrids were employed to inactivate pathogenic bacteria and accelerate bacteria-infected wound healing with the assistance of H2O2. In vitro investigation revealed the combination of hydroxyl radical (·OH) originated from low concentration of H2O2 catalyzed by Ag/FCE and the antimicrobial activity of Ag NPs endowed effective antibacterial performance to the hybrids against both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). Investigation on antibacterial mechanism indicated antibacterial activity of the synergetic strategy was achieved by destroying bacterial cell integrity, arresting metabolic, producing intracellular ROS, and oxidizing GSH. Additionally, in vivo assay exhibited Ag/FCE possessed satisfactory biocompatibility and effectively accelerated S. aureus-infected wound healing with the presence of low concentration of H2O2. Altogether, the presented results supported the great potential application of the synergistic antibacterial strategy for the therapy of bacterial-infected wound healing.

Abstract:Skin is the largest organ in the human body, and it is also the most important natural barrier. However, some accidents can cause skin damage. Bacterial infections and inflammatory reactions can hinder wound healing. Therefore, eliminating bacterial infections and regulating oxidative stress are essential. The use of antibiotics is no longer sufficient because of bacterial resistance. The development of new nanomaterials provides another way of thinking about bacterial drug resistance. In this study, bismuth selenide is modified with polyethylpyrrolidone to obtain a 2D nanomaterial with negligible toxicity and then added to a sodium polyacrylate hydrogel, which is nontoxic and has strong tissue adhesion and a weak antibacterial effect. To further enhance antibacterial performance, photothermal therapy is a good strategy. Under near-infrared light, Bi2Se3/PAAS shows a strong bactericidal effect. Bi2Se3/PAAS hydrogels also have certain antioxidant effects and are used to remove excess free radicals from wound infections. The effective therapeutic effect of Bi2Se3/PAAS/NIR on methicillin-resistant Staphylococcus aureus (MRSA) infection is further verified in animal models. Transcriptome analysis reveals that the Bi2Se3/PAAS hydrogel improves the function of vascular endothelial cells, regulates glucose and lipid metabolism, and promotes the healing of infected wounds.

Silver Nanoparticles Modified with Polygonatum sibiricum Polysaccharide Improve Biocompatibility and Infected Wound Bacteriostasis

Abstract:Silver nanoparticles (AgNPs) exhibit strong antibacterial activity and do not easily induce drug resistance; however, the poor stability and biocompatibility in solution limit their widespread application. In this study, AgNPs were modified with Polygonatum sibiricum Polysaccharide (PSP) to synthesize PSP@AgNPs with good stability, biocompatibility, and antibacterial activity. When PSP@AgNP synthesis was performed under a reaction time of 70 min, a reaction temperature of 35 °C, and an AgNO 3 -to-PSP volume ratio of 1:1, the synthesized PSP@AgNPs were more regular and uniform than AgNPs, and their particle size was around 10 nm. PSP@AgNPs exhibited lower cytotoxicity and hemolysis, and stronger bacteriostatic activity. PSP@AgNPs damage the integrity and internal structure of cells, resulting in the leakage of intracellular nucleic acids and proteins. The rate of cell membrane damage in Escherichia coli and Staphylococcus aureus treated with PSP@AgNPs increased by 38.52% and 43.75%, respectively, compared with that of AgNPs. PSP@AgNPs inhibit the activities of key enzymes related to antioxidant, energy and substance metabolism in cells. The inhibitory effects on the activities of superoxide dismutase (SOD), catalase (CAT), adenosine triphosphate enzyme (ATPase), malate dehydrogenase (MDH), and succinate dehydrogenase (SDH) in E. coli and S. aureus cells were significantly higher than those of AgNPs. In addition, compared with AgNPs, PSP@AgNPs promote faster healing of infected wounds. Therefore, PSP@AgNPs represent potential antibacterial agents against wound infections.

Abstract:Background: Methicillin-resistant Staphylococcus aureus (MRSA) biofilm-associated bacterial infection is the primary cause of nosocomial infection and has long been an ongoing threat to public health. MRSA biofilms are often resistant to multiple antimicrobial strategies, mainly due to the existence of a compact protective barrier; thus, protecting themselves from the innate immune system and antibiotic treatment via limited drug penetration. Results: A hierarchically structured hydrogen sulfide (H2S)-releasing nano-disinfectant was presented, which was composed of a zinc sulfide (ZnS) core as a H2S generator and indocyanine green (ICG) as a photosensitizer. This nano-disinfectant (ICG-ZnS NPs) sensitively responded to the biofilm microenvironment and demonstrated efficient eradication of MRSA biofilms via a synergistic effect of Zn2+, gas molecule-mediated therapy, and hyperthermia. Physically boosted by released H2S and a near-infrared spectroscopy-induced hyperthermia effect, ICG-ZnS NPs destroyed the compactness of MRSA biofilms showing remarkable deep-penetration capability. Moreover, on-site generation of H2S gas adequately ameliorated excessive inflammation, suppressed secretion of inflammatory cytokines, and expedited angiogenesis, therefore markedly accelerating the in vivo healing process of cutaneous wounds infected with MRSA biofilms. Conclusion: ICG-ZnS NPs combined with NIR laser irradiation exhibited significant anti-biofilm activity in MRSA biofilms, can accelerate the healing process through deep-penetration and anti-inflammatory effectuation. The proposed strategy has great potential as an alternative to antibiotic treatment when combating multidrug-resistant bacterial biofilms.

Abstract:Abstract Serious infection caused by drug-resistant gram-negative bacteria and their secreted toxins ( e.g. , lipopolysaccharide) is a serious threat to human health. Thus, treatment strategies that efficiently kill bacteria and reducing the impact of their toxins simultaneously are urgently required. Herein, a novel antibacterial platform composed of a mesoporous copper silicate microsphere (CSO) core and a platelet membrane (PM) shell was prepared (CSO@PM). CSO@PM specifically targets bacteria owing to formyl peptide receptors on the PM and, combined with photothermal therapy (PTT), exhibits highly effective bacter icidal activity. Importantly, CSO@PM can adsorb lipopolysaccharide secreted by gram-negative bacteria, resulting in inflammation reduction. Thus, CSO@PM stimulates re-epithelialization and granulation-tissue formation, promoting wound healing. Moreover, this antibacterial platform exhibits no obvious toxicity at all the test concentrations in vitro and in vivo. Thus, CSO@PM exhibits a robust antibacterial effect and a strong toxin-adsorption capacity, facilitating the clinical treatment of many bacterial infections and the development of next-generation antibacterial nanoagents. Efficient Antibacterial Polyphosphazene Material with Potential to Prominent Wound Healing

Abstract:Reducing the toxicity of silver-based antibacterial agents used in gels and improving their biocompatibility are of great significance in the development of wound dressings. Thus, a highly crosslinked organic–inorganic hybrid porous polyphosphazene nanospheres (PRV-HMSs) were prepared via precipitation polymerization. The structures and properties were investigated. The results indicated that PRV-HMSs have uniform particle size about 500 nm, excellent thermal stability, high silver loading (26.5 wt.%) and remarkable sustained-release properties after loading AgNO3 (up to 2 weeks). The antibacterial effects were investigated with zone of inhibition testing, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), Live/Dead bacterial staining assay and bacterial growth curve assays. The silver-loaded nanospheres have excellent bactericidal effects for E. coli or S. aureus and an average bactericidal rate of 94.02% and 94.67% in the 120 hour long-acting antibacterial test. The cytotoxicity and laser confocal immunofluorescence staining experiments indicate that the simple combination of nanospheres and agar not only give low cytotoxicity, but also significantly promote the activation of macrophages, which showing potential application in the wound dressings based on hydrogel.

Abstract:Bacterial infection, inflammatory disorder, and poor angiogenesis of tissue in chronic wounds are the main reasons why wounds are difficult to heal. In this study, a novel MSN-PEG@AS/BP nano-spray was designed to solve these issues. Astragaloside IV (AS) was loaded in mesoporous silica nanoparticles (MSN) to enhance angiogenesis and regulate inflammation, and the two-dimensional (2D) nanosheet black phosphorus (BP) was used to kill bacteria through a photothermal effect. Under thermal decomposition, the covalent bond of polyethylene glycol (PEG) was broken, releasing AS to promote the proliferation of fibroblasts, the formation of blood vessels, and the resolution of inflammation. AS can promote the polarization of the anti-inflammatory (M2) macrophage phenotype to enhance the deposition of extracellular matrix and the formation of blood vessels. Besides, BP showed a significant photothermal effect and nearly 99.58% of Escherichia coli and 99.13% of Staphylococcus aureus were killed in an antibacterial study. This nano-spray would be a novel therapeutic agent for infected wound treatment.

Highly biocompatible Ag nanocluster-reinforced wound dressing with long-term and synergistic bactericidal activity

Abstract:Clinical application of antibiotic-free agents like silver nanoparticle-derived materials remains a critical challenge due to their limited long-term antibacterial activity and potential system toxicity. Herein, a highly biocompatible Ag nanocluster-reinforced hydrogel with enhanced synergistic antibacterial ability has been developed. Specifically, bioactive curcumin was incorporated into lysozyme-protected ultrasmall Ag nanoclusters (LC-AgNCs) and further integrated with sodium alginate (Sa) hydrogel (LC-AgNCs@Sa) through multiple interaction forces. Due to the synergistic antibacterial activity, LC-AgNCs could effectively kill both S. aureus and E. coli bacteria with a concentration down to 2.5 μg mL-1. In-depth mechanism investigations revealed that the bactericidal effect of LC-AgNCs lies in their bacterial membrane destruction, reactive oxygen species (ROS) production, glutathione depletion and prooxidant-antioxidant system disruption ability. Curcumin can mediate the intracellular ROS balance to protect NIH 3T3 cells from oxidative stress and improve the biocompatibility of LC-AgNCs@Sa. LC-AgNCs@Sa with long-term antibacterial ability can effectively protect the wound from bacterial invasion in vivo, and significantly accelerate the wound healing process due to their distinctive functions of inhibiting inflammatory factor (TNF-α) production, promoting collagen deposit and facilitating re-epithelization. This study provides a new, versatile strategy for the design of high-performance antibacterial dressing for broad infectious disease therapy.

Abstract:C.d. spectra of crops and mother liquors obtained during fractional crystallisation of diastereoisomeric alkaloidal salts of N-thiobenzoyl-DL-α-amino-acids indicate directly the degree of resolution and, in principle, the absolute configuration of the predominant N-substituted amino-acid component in each fraction. Improvements resulting from c.d. monitoring of the resolution of a DL-amino-acid as its N-thiobenzoyl derivative include the ability to operate on a scale smaller than that possible by polarimetric methods, as illustrated for the resolution of 2-amino-3,3-dimethylbutyric acid (for which a new synthesis from ethyl acetoacetate is described).

Abstract:Chronic wounding treatment based on bacterially infected diabetes suffers an essential limitation in persistent skin injuries due to the resistance of progressive antibiotics, which inhibits the process of healing with wound tissue. Therefore, biologically friendly and nontoxic bio-based mats without antibiotics are taken for granted as a versatile platform for biomedical dressing, but urgently necessitates further functional diversification. Herein, a novel tannic acid (TA)/silver (Ag)-modified poly(lactic acid) (PLA)/Polyurethane (PU) antibacterial hybrid nanofibers were successfully constructed by electrospinning technology. Layer-by-layer (LBL) self-assembly technique was utilized to produce membranes via deposited biocompatible quaternized chitin (QC) and TA. The mats are enabled with outstanding flexibility, antibacterial activity, great hemocompatibility, and good ROS-scavenger in a wounding environment. Consequently, the basis of morphology and structure of electrospun membranes was verified by SEM and FT-IR. Besides, the LBL-structured surface was proved to impart improved wettability and hydrophilic via the test of water contact angle. Additionally, antimicrobial experiments demonstrate the effective broad-spectrum antibacterial ability of as-prepared hybrids, inhibiting infection of gram-positive microbial (S. aureus) as well as gram-negative microbial. Finally, the anti-oxidation performance holds great promise in conducive to the formation favorable physiological environment for wound healing. In conclusion, this work establishes a feasible but effective pathway to construct a multifunctional antibacterial dressing for the skin infection.

Taken together, this study strongly demonstrates the promising potential of AMPC@siTNF-α as a synergistic therapeutic agent for clinical wound infections.

In the in vivo wound model, the Escherichia coli (E. coli)-infected wound in mice almost completely disappeared after treatment with AMPC@siTNF-α, and no suppuration symptom was observed during the course of the treatment. Importantly, this nanoplatform had negligible side effects both in vitro and in vivo. Taken together, this study strongly demonstrates the promising potential of AMPC@siTNF-α as a synergistic therapeutic agent for clinical wound infections.
Surface Modification of Electrospun Polyethylenimine/Polyvinyl Alcohol Nanofibers Immobilized with Silver Nanoparticles for Potential Antibacterial Applications

Abstract:Objective: In order to investigate the potential biomedical applications of silver nanoparticle (Ag NP)-immobilized electrospun nanofibers with different surface functionalities. Methods: Silver nanoparticles were immobilized within water-stable electrospun polyethylenimine (PEI)/polyvinyl alcohol (PVA) nanofibers by an in-situ reduction method after complexing Ag + . ions with the free PEI amine groups. The obtained Ag NP-incorporated PEI/PVA nanofibers were then hydroxylated, carboxylated, and acetylated to generate different surface functionalities. Different techniques were employed to characterize the Ag NP-containing nanofibers with different surface functionalities. Results: In vitro antibacterial activity tests show that Ag NP-containing nanofibrous mats have high antibacterial activity and are capable of inhibiting the growth of both S. aureus and E. coli bacteria. Cell viability assay data show that the Ag NP-containing nanofibers are cytocompatible, and those treated by hydroxylation and acetylation display better cytocompatibility than those treated by carboxylation and the pristine non-modified fibers to promote cell adhesion and proliferation. Conclusion: Therefore, the hydroxylated or acetylated Ag NP-containing PEI/PVA nanofibers have a great potential for wound dressing, biological protection and tissue engineering applications.

Abstract:The exploration of safe and efficient antibiotic-free antimicrobial agents, harboring the capacities of concurrent antibacterial and wound healing, are highly desired to combat the drug-resistance bacteria. In this report, a nanord-like crown ether-based porous organic polymer (CEP-POP) with intrinsic antibacterial activity containing porphyrin unites, was facilely prepared, which was implemented as a carrier to deposit silver nitroprusside nanoparticles (abbreviated as AgNNPs), denoted as CEP-POP-AgNNP. CEP-POP-AgNNP could effectively address the issues of Ag-based bactericide, which not only significantly enhance the stability of Ag-based bactericide, but also greatly reduce the toxicity to normal tissues. Bacteriostatic study demonstrated CEP-POP-AgNNP could function as a broad-spectrum antibacterial agent with superb synergistic bactericidal activity, whose bactericide rate reached 97.7 ± 0.5 % and 90.7 ± 0.5 %, towards E. coli and S. aureus , respectively, at an extremely low inhibitory concentration (5 μg mL −1 ), far exceeding the as-reported POP-based antimicrobials. Meanwhile, in vivo experiment on mice validated CEP-POP-AgNNP could further enhance the wound healing in synergy with the antibacterial effect. Hence, this novel but simple Ag immobilization strategy paves a new avenue for the rational design of antibiotic-free antimicrobial agents.

Abstract:Current treatment for chronic infectious wounds is limited due to severe drug resistance in certain bacteria. Therefore, the development of new composite hydrogels with nonantibiotic antibacterial and pro-wound repair is important. Here, we present a photothermal antibacterial composite hydrogel fabricated with a coating of Fe2+ cross-linked carboxymethyl chitosan (FeCMCS) following the incorporation of melanin nanoparticles (MNPs) and the CyRL-QN15 peptide. Various physical and photothermal properties of the hydrogel were characterized. Cell proliferation, migration, cycle, and free-radical scavenging activity were assessed, and the antimicrobial properties of the hydrogel were probed by photothermal therapy. The effects of the hydrogel were validated in a model of methicillin-resistant Staphylococcus aureus (MRSA) infection with full-thickness injury. This effect was further confirmed by changes in cytokines associated with inflammation, re-epithelialization, and angiogenesis on the seventh day after wound formation. The MNPs demonstrated robust photothermal conversion capabilities. The composite hydrogel (MNPs/CyRL-QN15/FeCMCS) promoted keratinocyte and fibroblast proliferation and migration while exhibiting high antibacterial efficacy, effectively killing more than 95% of Gram-positive and Gram-negative bacteria. In vivo study using an MRSA-infected full-thickness injury model demonstrated good therapeutic efficacy of the hydrogel in promoting regeneration and remodeling of chronically infected wounds by alleviating inflammatory response and accelerating re-epithelialization and collagen deposition. The MNPs/CyRL-QN15/FeCMCS hydrogel showed excellent antibacterial and prohealing effects on infected wounds, indicating potential as a promising candidate for wound healing promotion.

Abstract:A novel natural source‐based composite microspheres with dual antibacterial mechanisms and bioadhesive features is prepared by microfluidics electrospray for infected wound healing. The microspheres played the important role in wound‐healing‐related angiogenesis. Based on these strategies, the composite microspheres exhibit excellent anti‐infection and wound healing performances in a rat wound model. Skin damage and infection pose a severe challenge to human health. Construction of a novel versatile dressing with good anti‐infection and healing‐promoting abilities is greatly expected. In this paper, nature‐source‐based composite microspheres with dual antibacterial mechanisms and bioadhesive features by microfluidics electrospray for infected wound healing is developed. The microspheres enable sustained release of copper ions, which not only show long‐term antibacterial properties, but also play important role in wound‐healing‐related angiogenesis. Additionally, the microspheres are coated with polydopamine via self‐polymerization, which renders the microspheres adhesive to the wound surface, and further enhance the antibacterial ability through photothermal energy conversion. Based on the dual antibacterial strategies provided by copper ions and polydopamine as well as the bioadhesive property, the composite microspheres exhibit excellent anti‐infection and wound healing performances in a rat wound model. These results, along with the nature‐source‐based composition and biocompatibility, indicate the great potential of the microspheres in clinical wound repair.

Abstract:An SA/SS/GI/PSP@AgNP composite film was developed, and its physicochemical properties, cytocompatibility, antioxidant activity, and antibacterial properties were investigated. The composite film released PSP@AgNPs in a sustained way, and the film demonstrated favorable mechanical properties, excellent cytocompatibility, and high antioxidant and antibacterial activities in both in vitro and in vivo models. As a result, we conclude that this composite film has potential for application as a medical material. A sodium alginate/silk sericin (SA/SS) film was modified with glycerin (GI) and Polygonatum sibiricum polysaccharide@silver nanoparticles (PSP@AgNPs) to create a SA/SS/GI/PSP@AgNP composite film with antibacterial and antioxidant properties. The physicochemical properties of the SA/SS/GI/PSP@AgNP composite film were analyzed, and its cytocompatibility, antioxidant activity, and antibacterial properties were investigated. The optimum conditions for the preparation of the SA/SS/GI/PSP@AgNP composite film were 2 % SA, 2 % SS, 20 % GI, and 40 μg/mL of PSP@AgNPs. The addition of GI improved the surface smoothness of the composite films, while the presence of PSP@AgNPs did not significantly alter it. Spectroscopic characterization demonstrated that GI and PSP@AgNPs were physically bound to SA/SS. Approximately 53 % of the PSP@AgNP was released from the SA/SS/GI/PSP@AgNP composite film over 24 h of soaking in aqueous solution. The resulting leaching solution did not cause hemolysis or cytotoxicity, indicating its biocompatibility. Additionally, the SA/SS/GI/PSP@AgNP composite film exhibited strong antioxidant and antibacterial properties, and it promoted healing of mouse wounds infected with Staphylococcus aureus.
