Abstract:Water vitrifies if cooled at rates above $3 \cdot 10^5$ K/s. Surprisingly, this process cannot simply be reversed by heating the resulting amorphous ice at a similar rate. Instead, we have recently shown that the sample transiently crystallizes even if the heating rate is more than one order of magnitude higher. This may present an issue for microsecond time-resolved cryo-electron microscopy experiments, in which vitreous ice samples are briefly flash melted with a laser pulse, since transient crystallization could potentially alter the dynamics of the embedded proteins. Here, we demonstrate how shaped microsecond laser pulses can be used to increase the heating rate and outrun crystallization during flash melting of amorphous solid water (ASW) samples. We use time-resolved electron diffraction experiments to determine that the critical heating rate is about $10^8$ K/s, more than two orders of magnitude higher than the critical cooling rate. Our experiments add to the toolbox of the emerging field of microsecond time-resolved cryo-electron microscopy by demonstrating a straightforward approach for avoiding crystallization during laser melting and for achieving significantly higher heating rates, which paves the way for nanosecond time-resolved experiments.

Abstract:Cryopreservation, one of the most effective preservation methods, is essential for maintaining the safety and quality of food. However, there is no denying the fact that the quality of muscle food deteriorates as a result of the unavoidable production of ice. Advancements in cryoregulatory materials and techniques have effectively mitigated the adverse impacts of ice, thereby enhancing the standard of freezing preservation. The first part of this overview explains how ice forms, including the theoretical foundations of nucleation, growth, and recrystallization as well as the key influencing factors that affect each process. Subsequently, the impact of ice formation on the eating quality and nutritional value of muscle food is delineated. A systematic explanation of cutting-edge strategies based on nucleation intervention, growth control, and recrystallization inhibition is offered. These methods include antifreeze proteins, ice-nucleating proteins, antifreeze peptides, natural deep eutectic solvents, polysaccharides, amino acids, and their derivatives. Furthermore, advanced physical techniques such as electrostatic fields, magnetic fields, acoustic fields, liquid nitrogen, and supercooling preservation techniques are expounded upon, which effectively hinder the formation of ice crystals during cryopreservation. The paper outlines the difficulties and potential directions in ice inhibition for effective cryopreservation.

Here, we propose a novel approach for microsecond time-resolved cryo-electron microscopy that involves melting a cryo specimen <em>in situ</em> with a laser beam. The sample remains liquid for the duration of the laser pulse, offering a tunable time window in which the dynamics of embedded particles can be induced in a liquid environment. After the laser pulse, the sample vitrifies, trapping particles in their transient configurations. As a proof-of-principle, we study the disassembly of particles after they incur structural damage.</p>

Abstract:Sample preparation remains a bottleneck for protein structure determination by cryo-electron microscopy. A frequently encountered issue is that proteins adsorb to the air-water interface of the sample in a limited number of orientations. This makes it challenging to obtain high-resolution reconstructions or may even cause projects to fail altogether. We have previously observed that laser flash melting and revitrification of cryo samples reduces preferred orientation for large, symmetric particles. Here, we demonstrate that our method can in fact be used to scramble the orientation of proteins of a range of sizes and symmetries. The effect can be enhanced for some proteins by increasing the heating rate during flash melting or by depositing amorphous ice onto the sample prior to revitrification. This also allows us to shed light onto the underlying mechanism. Our experiments establish a set of tools for overcoming preferred orientation that can be easily integrated into existing workflows.

Abstract:Abstract Cryoprotective agents (CPAs) are routinely used to vitrify, attain an amorphous glass state void of crystallization, and thereby cryopreserve biomaterials. Two vital characteristics of a CPA-loaded system are the critical cooling and warming rates (CCR and CWR), the temperature rates needed to achieve and return from a vitrified state, respectively. Due to the toxicity associated with CPAs, it is often desirable to use the lowest concentrations possible, driving up CWR and making it increasingly difficult to measure. This paper describes a novel method for assessing CWR between the 0.4 × 105 and 107 °C/min in microliter CPA-loaded droplet systems with a new ultrarapid laser calorimetric approach. Cooling was achieved by direct quenching in liquid nitrogen, while warming was achieved by the irradiation of plasmonic gold nanoparticle-loaded vitrified droplets by a high-power 1064 nm millisecond pulsed laser. We assume “apparent” vitrification is achieved provided ice is not visually apparent (i.e., opacity) upon imaging with a camera (CCR) during cooling or highspeed camera (CWR) during warming. Using this approach, we were able to investigate CWRs in single CPA systems such as propylene glycol (PG), glycerol, and Trehalose in water, as well as mixtures of glycerol-trehalose-water and propylene glycol-trehalose-water CPA at low concentrations (20–40 wt %). Further, a phenomenological model for determining the CCRs and CWRs of CPAs was developed which allowed for predictions of CCR or CWR of single component CPA and mixtures (within and outside of the regime their constituents were measured in), providing an avenue for optimizing CCR and CWR and perhaps future CPA cocktail discovery.

Abstract:Time-resolved structural investigations of crystallization of water in lipid/protein/salt mesophases at cryogenic temperatures are significant for comprehension of ice nanocrystal nucleation kinetics in lipid membranous systems and can lead to a better understanding of how to experimentally retard the ice formation that obstructs the protein crystal structure determination. Here, we present a time-resolved synchrotron microfocus X-ray diffraction (TR-XRD) study based on ∼40,000 frames that revealed the dynamics of water-to-ice crystallization in a lipid/protein/salt mesophase subjected to cryostream cooling at 100 K. The monoolein/hemoglobin/salt/water system was chosen as a model composition related to protein-loaded lipid cubic phases (LCP) broadly used for the crystallization of proteins. Under confinement in the nanoscale geometry, metastable short-living cubic ice (Ic) rapidly crystallized well before the formation of hexagonal ice (Ih). The detected early nanocrystalline states of water-to-ice transformation in multicomponent systems are relevant to a broad spectrum of technologies and understanding of natural phenomena, including crystallization, physics of water nanoconfinement, and rational design of anti-freezing and cryopreservation systems.

Soft Liquid Metal Nanoparticles As Superior Ice-Growth Inhibitors Achieving Reduced Crystal Nucleation and Ultrarapid Rewarming for Cryopreservation

Abstract:High warming rates during cryopreservation are crucial and essential for successful vitrification. However, realizing faster warming rate in low concentration cryoprotective agents appears to be challenging for conventional warming process via convective heat transfer. Here a liquid metal nanosystem which can act as a spatial source to significantly enhance the warming rates with near-infrared laser irradiation during warming process is developed. It demonstrates that the synthetic Pluronic F127-liquid metal nanoparticles (PLM NPs) displayed multiple performances with uniform particle size, superior photothermal conversion efficiency (52%), repeatable photothermal stability, and low cytotoxicity. Specifically, liquid PLM NPs with less surface free energy are more difficult to form crystal nucleation compared with other solid NPs such as gold and Fe3O4, which is beneficial for the cooling process during cryopreservation. The viability of human bone marrow derived mesenchymal stem cells post-cryopreservation reached 77.9±3.4% which is 3-fold higher than that obtained by conventional warming method (25.3±6.43%). Additionally, the cells post-cryopreservation maintained their normal attachment, proliferation, surface marker expression and intact multilineage differentiation properties. Moreover, the results of mouse tails cryopreservation showed relatively improved intact structure when using PLM NPs rewarming compared with convective warming. The new liquid metal nanosystem provides a universal platform for cryopreservation that is expected to have widespread applications, especially the potential in bioengineering, cell-based medicine and clinical translation.
Interplay of vitrification and ice formation in a cryoprotectant aqueous solution at low temperature

Abstract:The proneness of water to crystallize is a major obstacle to understanding its putative exotic behavior in the supercooled state. It also represents a strong practical limitation to cryopreservation of biological systems. Adding some concentration of glycerol, which has a cryoprotective effect preventing to some degree water crystallization, has been proposed as a possible way out, provided the concentration is small enough for water to retain some of its bulk character and/or for limiting the damage caused by glycerol on living organisms. Contrary to previous expectations, we show that in the ``marginal'' glycerol molar concentration $\approx18\%$, at which vitrification is possible with no crystallization on rapid cooling, water crystallizes upon isothermal annealing even below the calorimetric glass transition of the solution. Through a time-resolved polarized neutron scattering investigation, we extract key parameters, size and shape of the ice crystallites, fraction of water that crystallizes, crystallization time, which are important for cryoprotection, as a function of the annealing temperature. We also characterize the nature of the out-of-equilibrium liquid phases that are present at low temperature, providing more arguments against the presence of an iso-compositional liquid-liquid transition. Finally, we propose a rule-of-thumb to estimate the lower temperature limit below which water crystallization does not occur in aqueous solutions.

Abstract:Studying protein interactions at low temperatures has important implications for optimizing cryostorage processes of biological tissue, food, and protein-based drugs. One of the major challenges is related to the formation of ice nanocrystals which can occur even in the presence of cryoprotectants and can lead to protein denaturation. Here, using a combination of small- and wide-angle X-ray scattering (SAXS and WAXS), we investigate the structural evolution of concentrated Lysozyme solutions in a cryoprotected glycerol-water mixture upon cooling from room temperature (T=300 K) down to cryogenic temperatures (T=195 K). Upon cooling, we observe a transition near the melting temperature of the solution (T~245 K), which manifests both in the temperature dependence of the scattering intensity peak position reflecting protein-protein length scales (SAXS) and the interatomic distances within the solvent (WAXS). Upon thermal cycling, a hysteresis is observed in the scattering intensity, which is attributed to the formation of nanocrystallites in the order of 10 nm. The experimental data is well described by the two-Yukawa model, which indicates temperature-dependent changes in the short-range attraction of the protein-protein interaction potential. Our results demonstrate that the nanocrystal growth yields effectively stronger protein-protein attraction and influences the protein pair distribution function beyond the first coordination shell.

Dissolved Gases and Ice Fracturing During the Freezing of a Multicellular Organism: Lessons from Tardigrades

Abstract:Three issues are critical for successful cryopreservation of multicellular material: gases dissolved in liquid, thermal conductivity of the tissue, and localization of microstructures. Here we show that heat distribution is controlled by the gas amount dissolved in liquids and that when changing the liquid into solid, the dissolved gases either form bubbles due to the absence of space in the lattice of solids and/or are migrated toward the concentrated salt and sugar solution at the cost of amount of heat required to be removed to complete a solid-state transition. These factors affect the heat distribution in the organs to be cryopreserved. We show that the gas concentration issue controls fracturing of ice when freezing. There are volumetric changes not only when changing the liquid into solid (volume increases) but also reduction of the volume when reaching lower temperatures (volume decreases). We discuss these issues parallel with observations of the cryosurvivability of multicellular organisms, tardigrades, and discuss their analogy for cryopreservation of large organs.
Investigation on recrystallization of cryoprotectant solutions during warming under alternating electric fields

Abstract:In the field of cryopreservation, recrystallization during rewarming is a significant factor leading to cryo-injury, causing ice crystal-induced mechanical injury to biological samples. Currently, methods, such as adding ice recrystallization inhibiting agents and increasing warming rates, have been adopted to reduce recrystallization injury. These methods also greatly increase the potential toxicity risk and engineering difficulty of cryopreservation. Accumulating research has progressively demonstrated that application of an electromagnetic field during cryopreservation can influence the preservation efficacy. However, the mechanism of action of the electric field has remained a subject of ongoing debate. We observed the recrystallization phenomenon of cryoprotectant solutions under a polarized light microscope and discovered promising experimental results. The electric field strength did not exhibit a discernible impact on the ice crystal size, while electric field frequency exerted a more significant impact. We found that at specific frequencies, the ice crystal size and growing rate were significantly affected during annealing. Compared to no electric field, an alternating electric field with a voltage of 10 V and a frequency of 5 × 104 Hz increased the recrystallization areas of a 15% (w/v) propylene glycol solution by 57%. And an alternating electric field with a voltage of 3 V and a frequency of 5 × 108 Hz decreased the recrystallization area of a 0.9% (w/v) NaCl aqueous solution by 9.9%. Different types of cryoprotectants exhibit varying responses to alternating electric fields frequency. These findings prompt us to reevaluate the role and mechanism of alternating electric fields in the cryopreservation of biological materials.

Abstract:The micro- and nano-scale freezing has been a focus in many fields of scientific research and engineering practices, especially in those involved with cryobiology physics. Although a series of theoretical models and experimental techniques have been available to tackle the tough issues that associate with the freezing phase change process, the topic to quantify the ice crystal formation and its subsequent effects on the biological materials were not sufficiently addressed at all, especially when facing the newly emerging area of nano-cryobiology technology. Not only the interactions between the ice front and the cell, but also the heat and solute balance during freezing produce impact on the output of a cryosurgery or cryopreservation. There is currently a strong lack of the generalized methods which are indispensable to grasp the physical pictures from micro or nanoscale aspect and thus help achieve an intensive understanding of the mechanisms of cell damage during process of ice crystallization. This review article is dedicated to present an overview on the theoretical and experimental strategies potentially appropriate for quantifying the ice crystal formation in cryobiology area. Attentions will be focused on several major theoretical directions such as the thermodynamic theory, the kinetic theory, and the density functional theory etc., concerning the micro or nano phase change and the ice nucleation activities. Interpretation of the effects of external conditions such as freezing rate, interfacial effect, size effects, electric field and restricted space etc. on the ice growth will be given. Numerical simulation method and experimental techniques will also be outlined. The review does not intend to give an exhaustive knowledge of all the themes, but rather to provide a most basic background for further probing the emerging nano-cryobiology field and thus promote innovation in future research and practices of cryobiology.

Abstract:Freezing interface interacting with soft cells is a core issue in cryopreservation. However, most of existing cryopreservation approaches face challenges such as complex processing and poor controllability. Herein we report a new, nanotechnology-based method to modulate interfacial interactions between ice and cells via the premelting theory. Through the interfacial design by controllably modifying biocompatible nanoshells on the surface of cells, the effective Hamaker constant between cells and ice can be modified. The thickness of premelted films between cells and ice is further regulated, and directional migration of coated cells occurs in a thermal gradient to achieve directional thawing of coated cells. The decreased mortality of freezing coated cells suggests the premelting modulation in freezing and thawing can effectively protect cells from mechanical damage of ice formation during freezing and ice recrystallization during thawing.

Flash Melting Amorphous Ice

Abstract:Water can be vitrified if it is cooled at rates exceeding $3*10^5$ K/s. This makes it possible to outrun crystallization in so-called no man's land, a range of deeply supercooled temperatures where water crystallizes rapidly. One would naively assume that the process can simply be reversed by heating the resulting amorphous ice at a similar rate. We demonstrate that this is not the case. When amorphous ice samples are flash melted with a microsecond laser pulse, time-resolved electron diffraction reveals that the sample transiently crystallizes despite a heating rate of more than $5*10^6$ K/s, demonstrating that the critical heating rate for outrunning crystallization is significantly higher than the critical cooling rate during vitrification. Moreover, we observe different crystallization kinetics for amorphous solid water (ASW) and hyperquenched glassy water (HGW), which suggests that the supercooled liquids formed during laser heating transiently retain distinct non-equilibrium structures that are associated with different nucleation rates. These experiments open up new avenues for elucidating the crystallization mechanism of water and studying its dynamics in no man's land. They also add important mechanistic details to the laser melting and revitrification process that is integral to the emerging field of microsecond time-resolved cryo-electron microscopy.

Self-pressurised rapid freezing at arbitrary cryoprotectant concentrations

Abstract:Self-pressurised rapid freezing (SPRF) has been proposed as a simple alternative to traditional high-pressure freezing (HPF) protocols for vitrification of biological samples in electron microscopy and cryopreservation applications. Both methods exploit the circumstance that the melting point of ice reaches a minimum when subjected to pressure of around 210 MPa, however, in SPRF its precise quantity depends on sample properties and hence, is generally unknown. In particular, cryoprotective agents (CPAs) are expected to be a factor; though eschewed by many SPRF experiments, vitrification of larger samples notably cannot be envisaged without them. Thus, in this study, we address the question of how CPA concentration affects pressure inside sealed capillaries, and how to design SPRF experiments accordingly. By embedding a fibre-optic probe in samples and performing Raman spectroscopy after freezing, we first present a direct assessment of pressure build-up during SPRF, enabled by the large pressure sensitivity of the Raman shift of hexagonal ice. Choosing dimethyl sulphoxide (DMSO) as a model CPA, this approach allows us to demonstrate that average pressure drops to zero when DMSO concentrations of 15 wt% are exceeded. Since a trade-off between pressure and DMSO concentration represents an impasse with regard to vitrification of larger samples, we introduce a sample architecture with two chambers, separated by a partition that allows for equilibration of pressure but not DMSO concentrations. We show that pressure and concentration in the fibre-facing chamber can be tuned independently, and present differential scanning calorimetry (DSC) data supporting the improved vitrification performance of two-chamber designs. Lay version of abstract for 'Self-pressurised rapid freezing at arbitrary cryoprotectant concentrations' Anyone is familiar with pipes bursting in winter because the volume of ice is greater than that of liquid water. Less well known is the fact that inside a thick-walled container, sealed and devoid of air bubbles, this pressure build-up will allow a fraction of water to remain unfrozen if the sample is also cooled sufficiently rapidly far below the freezing point. This phenomenon has already been harnessed for specimen preparation in microscopy, where low temperatures are useful to immobilise the sample, but harmful if ice formation occurs. However, specimen preparation cannot always rely on this pressure-based effect alone, but sometimes requires addition of chemicals to inhibit ice formation. Not enough is known directly about how these chemicals affect pressure build-up: Indeed, rapid cooling below the freezing point is only possible for small sample volumes, typically placed inside sealed capillaries, so that space is generally insufficient to accommodate a pressure sensor. By means of a compact sensor, based on an optical fibre, laser and spectrometer, we present the first direct assessment of pressure inside sealed capillaries. We show that addition of chemicals reduces pressure build-up and present a two-chambered capillary to circumvent the resulting trade-off. Also, we present evidence showing that the two-chambered capillary design can avoid ice formation more readily than a single-chambered one.
In situ counter‐diffusion crystallization and long‐term crystal preservation in microfluidic fixed targets for serial crystallography

Abstract:User‐friendly in situ counter‐diffusion crystallization and long‐term storage setups in microfluidic fixed targets provide a reliable approach to forming large crystals and maintaining their hydration for weeks, allowing ample time to grow, select and preserve the best crystal batches before X‐ray beam time.Compared with batch and vapor diffusion methods, counter diffusion can generate larger and higher‐quality protein crystals yielding improved diffraction data and higher‐resolution structures. Typically, counter‐diffusion experiments are conducted in elongated chambers, such as glass capillaries, and the crystals are either directly measured in the capillary or extracted and mounted at the X‐ray beamline. Despite the advantages of counter‐diffusion protein crystallization, there are few fixed‐target devices that utilize counter diffusion for crystallization. In this article, different designs of user‐friendly counter‐diffusion chambers are presented which can be used to grow large protein crystals in a 2D polymer microfluidic fixed‐target chip. Methods for rapid chip fabrication using commercially available thin‐film materials such as Mylar, propylene and Kapton are also detailed. Rules of thumb are provided to tune the nucleation and crystal growth to meet users' needs while minimizing sample consumption. These designs provide a reliable approach to forming large crystals and maintaining their hydration for weeks and even months. This allows ample time to grow, select and preserve the best crystal batches before X‐ray beam time. Importantly, the fixed‐target microfluidic chip has a low background scatter and can be directly used at beamlines without any crystal handling, enabling crystal quality to be preserved. The approach is demonstrated with serial diffraction of photoactive yellow protein, yielding 1.32 Å resolution at room temperature. Fabrication of this standard microfluidic chip with commercially available thin films greatly simplifies fabrication and provides enhanced stability under vacuum. These advances will further broaden microfluidic fixed‐target utilization by crystallographers.
