Abstract:A novel electrochemical device for the sensitive determination of dopamine was developed based on a carbon paste electrode with polymeric ferric sulfate doped in the carbon paste and a carboxyl-functionalized carbon nanotube thin film on the surface. The modified electrode was characterized by scanning electron microscopy, electrochemical impedance spectroscopy, and cyclic voltammetry. The conditions for the preparation of electrode were optimized. The carbon nanotubes were shown to be stable on the surface of carbon paste electrode. The novel electrochemical device provided excellent activity toward dopamine. Amperometry and differential pulse voltammetry were used for the determination of dopamine in pH 7.0 phosphate buffer with a long linear range from 0.8 to 261 mu M and a detection limit of 0.2 mu M. The modified electrode showed excellent repeatability, good stability, and satisfactory reproducibility, thus demonstrating potential for practical applications.

Abstract:We fabricated an acoustic composite structure consisting of a periodic array of interspaced membranes and side holes. Experimental data on the transmission, effective density, and phase velocity are presented. The system exhibits two critical frequencies, omega{SH} and omega{c}. Our metamaterial is double negative and transparent for frequencies lower than omega{SH}. For the frequencies omega{SH}<omega<omega{c}, the medium is opaque and only the density is negative. For the frequencies above omega{c}, the system is double positive and transparent. The present medium exhibits a very wide double negative spectral range that opens the possibility of the application of metamaterials for "white lights."

Abstract:Electrochemical sensors can be used to detect neurotransmitters. A miniature potentiometric all-solid-state coated wire dopamine sensor was investigated in this work. Conducting polymer poly (3,4- ethylenedioxythiophene) doped with poly (styrenesulfonate) as the solid contact was electropolymerized on one end of a gold wire (diameter 0.25 mm). The conducting polymer was covered with a dopamine-selective membrane containing 12-crown-4-tetraphenylborate as a neutral carrier, 2-nitrophenyloctyl ether as a plasticizer and poly (vinyl chloride) as the membrane matrix. The dopamine sensor covered a wide linear range from 10(-5) M to 10(-1) M with a slope of 53.85 +/- 0.93 mV/decade, while the detection limit was 5.80 +/- 0.31 mu M. The other characteristics of the dopamine sensor were also evaluated. The experimental results showed that the sensor operated properly within the pH range of 5.5-7.5 and displayed good selectivity, fast dynamic response, high reproducibility and long stability. In addition, the dopamine sensor was successfully applied for the detection of dopamine in artificial cerebrospinal fluid by using the standard addition method, indicating that this sensor is promising for applications in dopamine determination in neuroscientific research.

Abstract:Serum bilirubin determinations were performed on 264 term infants who were consecutively delivered via the vaginal route. Forty-one infants (15.5%) had serum bilirubin concentrations greater than 12 mg/dL. No cause for this was found, initially, in 23 (56%) of these infants. On the third hospital day, the mean (+/- SD) serum bilirubin level was 6.9 +/- 3.6 mg/dL in breast-fed infants and 6.5 +/- 3.2 mg/dL in bottle-fed infants. Of the 23 infants without obvious cause for hyperbilirubinemia, eight (four bottle-fed and four breast-fed infants) had serum bilirubin concentrations greater than 12 mg/dL on the third hospital day, whereas in 15 (14 breast-fed infants and one bottle-fed infant), the elevated serum bilirubin level occurred on day 4 or 5. Breast-feeding does not seem to affect the total serum bilirubin level in the first three days of life but may be associated with an increased incidence of hyperbilirubinemia subsequently. In a normal full-term population, routine investigations do not disclose a cause for hyperbilirubinemia in about half of the patients.

Abstract:Dopamine is an important neurotransmitter for organisms and affects their function, so monitoring and detecting dopamine levels is essential. Sensitivity and selectivity of electrochemical sensors for the detection of dopamine usually relies on hybrid nanocomposites. In this context, we designed a micro-regionally restricted hybrid nanocomposite by modulating the micro-nano structure. Using functional groups on nitrogen-doped reduced graphene oxide (N-rGO) to restrict the random growth of tungsten disulfide nanosheets, we obtained a 2D layered material with bidirectional structural support to achieve high specific surface and high activity for application in robust and novel dopamine electrochemical sensors. A facile electrochemical sensor for dopamine detection was fabricated by modifying a screen-printed carbon electrode with the nanocomposite. The sensor exhibits excellent performance of dopamine detection in the wide range of 1 μmol/L100 μmol/L with a detection limit of 0.2 μmol/L and a high sensitivity of 0.56 μA/μmol/L. The optimized sensor shows a good selectivity, satisfactory long-term stability and reproducibility towards DA and a separation of peak potential of 142 mV with uric acid. In addition, the prepared sensor provides favorable recovery in human serum and artificial cerebrospinal solution (aSCF), indicating great potentials in electrochemical detection and biological sensing applications.

Abstract:Dopamine (DA) signaling is critically important in striatal function, and this metabolically demanding process is fueled largely by glucose. However, DA and glucose are typically studied independently and, as such, the precise relationship between DA release and glucose availability remains unclear. Fast-scan cyclic voltammetry (FSCV) is commonly coupled with carbon-fiber microelectrodes to study DA transients. These microelectrodes can be modified with glucose oxidase (GOx) to generate microbiosensors capable of simultaneously quantifying real-time and physiologically relevant fluctuations of glucose, a nonelectrochemically active substrate, and DA, which is readily oxidized and reduced at the electrode surface. A chitosan hydrogel can be electrodeposited to entrap the oxidase enzyme on the sensor surface for stable, sensitive, and selective codetection of glucose and DA using FSCV. This strategy can also be used to entrap lactate oxidase on the carbon-fiber surface for codetection of lactate and DA. However, these custom probes are individually fabricated by hand, and performance is variable. This study characterizes the physical nature of the hydrogel and its effects on the acquired electrochemical data in the detection of glucose (2.6 mM) and DA (1 μM). The results demonstrate that the electrodeposition of the hydrogel membrane is improved using a linear potential sweep rather than a direct step to the target potential. Electrochemical impedance spectroscopy data relate information on the physical nature of the electrode/solution interface to the electrochemical performance of bare and enzyme-modified carbon-fiber microelectrodes. The electrodeposition waveform and scan rate were characterized for optimal membrane formation and performance. Finally, codetection of both DA/glucose and DA/lactate was demonstrated in intact rat striatum using probes fabricated according to the optimized protocol. Overall, this work improves the reliable fabrication of carbon-fiber microbiosensors for codetection of DA and important energetic substrates that are locally delivered to the recording site to meet metabolic demand.

Abstract:Using 3-dimensional (3D) printing to create a fast and accurate electrochemical sensor has attracted the attention of analytical scientists. This study utilized a carbon black/PLA composite filament as the raw material for the construction of 3D electrodes (3DEs). Au particles were modified onto the activated surface of the carbon black nanonetwork 3DE. The physical and chemical properties of the 3DEs were evaluated through scanning electron microscopy (SEM) and X-ray spectroscopy (XPS). Additionally, a range of electrochemical techniques were employed to investigate the electrochemical performance of the 3DEs. The sensor is capable of detecting DA in the concentration range of 0.01–20 μM with the limit of detection (LOD) of 0.084 μM. The approach is also capable of being applied to the actual sample detection of DA with excellent selectivity. Hence, the method of combining noble metal particles and carbon black nanonetwork for constructing 3DE presents novel prospects towards the creation of electrochemical sensors with exceptional sensitivity.

Abstract:Microelectrodes are useful electrochemical sensors that can provide spatial biological monitoring. Carbon fiber has been by far the most widely used microelectrode; however, a vast number of different materials and modification strategies have been developed to broaden the scope of microelectrodes. Carbon composite electrodes provide a simple approach to making microelectrodes with a wide range of materials, but manufacturing strategies are complex. 3D printing can provide the ability to make microelectrodes with high precision. We used fused filament fabrication to print single strands of carbon black/polylactic acid (CB/PLA) and multiwall carbon nanotube/polylactic acid (MWCNT/PLA), which were then made into microelectrodes. Microelectrodes ranged from 70 μm in diameter to 400 μm in diameter and were assessed using standard redox probes. MWCNT/PLA electrodes exhibited greater sensitivity, a lower limit of detection, and stability for the measurement of serotonin (5-HT). Both CB/PLA and MWCNT/PLA microelectrodes were able to monitor 5-HT overflow from the ex vivo ileum tissue. MWCNT/PLA microelectrodes were utilized to show differences in 5-HT overflow from ex vivo ileum and colon following exposure to odorants present in spices. These findings highlight that any conductive thermoplastic material can be fabricated into a microelectrode. This simple strategy can utilize a wide range of materials to make 3D-printed microelectrodes for a diverse range of applications.

Abstract:Brain‐on‐Chip devices, which facilitate on‐chip cultures of neurons to simulate brain functions, are receiving tremendous attention from both fundamental and clinical research. Consequently, microsensors are being developed to accomplish real‐time monitoring of neurotransmitters, which are the benchmarks for neuron network operation. Among these, electrochemical sensors have emerged as promising candidates for detecting a critical neurotransmitter, dopamine. However, current state‐of‐the‐art electrochemical dopamine sensors are suffering from issues like limited sensitivity and cumbersome fabrication. Here, we report a novel route in monolithically microfabricating vertically aligned carbon nanofiber electrochemical dopamine microsensors with an anti‐blistering slow cooling process. Thanks to the microfabrication process, we created microsensors with complete insulation and large surface areas. The champion device shows extremely high sensitivity of 4.52× 104μA/μM·cm2, which is two‐orders‐of‐magnitude higher than current devices, and a highly competitive limit of detection of 0.243 nM. Abstract:Dopamine (DA), a catecholamine neurotransmitter , is crucial in brain signal transmission . Monitoring cytoplasmic DA levels can reflect changes in metabolic factors and provide valuable information for researching the mechanisms involved in neurodegenerative diseases. However, the in-situ detection of intracellular DA is constrained by its low contents in small-sized single cells. In this work, we report that noble metal (Au, Pt)-modified carbon fiber micro-nanoelectrodes are capable of real-time detection of DA in single cells with excellent sensitivity, selectivity, and anti-contamination capabilities. Notably, noble metals can be modified on the electrode surface through electrochemical deposition to enhance the conductivity of the electrode and the oxidation current of DA by 50 %. The nanosensors can work stably and continuously in rat adrenal pheochromocytoma cells (PC12) to monitor changes in DA levels upon K + stimulation. The functionalized carbon fibers based nanosensors will provide excellent prospects for DA analysis in the brains of living animals.

Abstract:Neurological disorders can occur in the human body as a result of nano-level variations in the neurotransmitter levels. Patients affected by neuropsychiatric disorders, that are chronic require continuous monitoring of these neurotransmitter levels for effective disease management. The current work focus on developing a highly sensitive and personalized sensor for continuous monitoring of dopamine. Here we propose a wearable microneedle-based electrochemical sensor, to continuously monitor dopamine in interstitial fluid (ISF). A chitosan-protected hybrid nanomaterial Fe 3 O 4 –GO composite has been used as a chemical recognition element protected by Nafion antifouling coating layer. The morphological and physiochemical characterizations of the nanocomposite were carried out with XRD, XPS, FESEM, EDAX and FT-IR. The principle of the developed sensor relies on orthogonal detection of dopamine with square wave voltammetry and chronoamperometric techniques. The microneedle sensor array exhibited an attractive analytical performance toward detecting dopamine in phosphate buffer and artificial ISF. The limit of detection (LOD) of the developed sensor was observed to be low, 90 nM in square wave voltammetry and 0.6 μM in chronoamperometric analysis. The practical applicability of the microneedle sensor array has been demonstrated on a skin-mimicking phantom gel model. The microneedle sensor also exhibited good long-term storage stability, reproducibility, and sensitivity. All of these promising results suggest that the proposed microneedle sensor array could be reliable for the continuous monitoring of dopamine.

Abstract:The detection of the neurotransmitter dopamine in medicinal and human biological samples has attracted the huge interest of researchers in recent years. Carbon nanofiber is a potential nanocarbon material for the detection of dopamine (DA) due to its good electrochemical properties. In this work, carbon nanofiber (CNF) supported nickel nanoparticles () were prepared by wet impregnation method and characterized by TEM, EDS, XRD and FTIR analysis. The modified screen printed electrode gave superior electrochemical response. Moreover, the /CNF shows a broad linear detection range (0.1-10 μM), a low detection limit (0.03 μM) by using DPV analysis and excellent selectivity. The developed electrode shows good reproducibility, stability, and repeatability. The /SPCE has been also utilized for the detection of DA in pharmaceutical, human blood serum and urine samples with substantial recovery outcomes. Therefore, the prepared /SPCE will be a capable platform for the electrochemical detection of DA in biological or medicinal samples.

Abstract:One big challenge of fabricating nanosensors for spatially resolved electrochemical detection of neurochemicals, such as dopamine (DA), is the difficulty to assembly nanometer-scale patternable and integrated sensors. In this work we develop a novel approach to precisely manufacture nano-Au-electrode (NAE) using lithographic fabrication technique, and characterize the NAE for DA detection. A negative photoresist, SU-8, is used as a substrate and protection layer for the 127-nm Au active sensing layer. The cross surface morphology and thickness of the Au layer are imaged by scanning electron microscopy and an interference microscopy. This NAE could be precisely controlled, repeatedly fabricated and conveniently renewed for several times. The electrochemical sensitivity and selectivity of the NAE towards DA detection are significantly higher than those of a standard Au thin-film electrode. This work demonstrates that the NAE could be used as an attractive means for electrochemically sensing and recording DA. (c) 2012 Elsevier B.V. All rights reserved.
Villous 3D nanoconfined flexible carbon fibers-based electrode toward dopamine electrochemical detection

Abstract:Flexible sensing interfaces for the analysis of biological samples are crucial to realize human dopamine health detection; thus, flexible sensors for biomolecular analysis have attracted considerable attention. Herein, we report a villous three-dimensional (3D) nanoconfined carbon fibers-based flexible sensor for the electrochemical analysis of dopamine (DA). Carbon nanofibers-modified carbon fibers (CNFs/CFs) with abundant nanochannels were obtained by chemical vapor deposition, and the sensing platform achieved ultra-sensitive analysis owing to the construction of nanoconfined spaces. The combination of a 3D network structure and surface N doping (N-CNFs/CFs) significantly enhanced the local enrichment of DA, thus providing a basis for ultrasensitive detection of DA. Furthermore, modified Au nanoparticles (Au/N-CNFs/CFs) exhibited s enhanced the electrocatalytic ability and electron transfer efficiency. The detection limit for DA reached 0.087 μM (S/N = 3) in the linear range of 1–800 μM. Under optimal conditions, the designed electrode material shows good performance in complex biological environments, including human serum, urine, and artificial cerebrospinal fluid). The developed Au/N-CNFs/CFs therefore provide a portable and efficient sensor for the clinical detection of biomolecules.

(Invited) Tunable CNT Fiber and Yarn Microelectrodes for Measurements of Different Neurochemicals

Abstract:Traditionally, in vivo analysis of neurotransmitters has concentrated on the molecule dopamine and much of the surface characterization of new electrodes has been focused on the kinetics of dopamine detection. However, the field of in vivo electrochemistry is expanding to many new molecules and electrode design should follow suit to examine the extent to which it is possible to design new electrodes that have surface chemistry that is advantageous for different neurochemicals. Beyond dopamine, other small neurotransmitters that are often targeted include serotonin, norepinephrine and epinephrine. Serotonin and it’s metabolite 5-HIAA are known to foul the electrode surface of carbon fibers, but recently developed carbon nanotube-polyethyleneimine (PEI/CNT) fiber microelectrodes show reduced susceptibility for fouling. PEI/CNT fiber microelectrodes are more selective to DOPAC, and CNT yarn microelectrodes show significantly higher selectivity to serotonin than other fibers. Chlorosulfonice acid (CA)/CNT fiber microelectrodes have the highest selectivity of dopamine to ascorbic acid and uric acid, because of the negative charge carried by CA. Our lab has also investigated CNT yarn microelectrodes for adenosine detection and found that CNT yarns may be advantageous for detecting all three oxidation peaks for adenosine. In addition, we have explored surface treatments of microelectrodes, including laser treatment, oxygen plasma treatment, and antistatic gun treatment. These treatments increase the sensitivity and change the selectivity of the sensors, allowing the properties to be tuned. Thus, electrode materials, design, and surface materials are important considerations for designing in vivo microelectrodes that are sensitive to a variety of different analytes.
Carbon composite microelectrodes fabricated by electrophoretic deposition.

Abstract:Single-walled carbon nanotubes were deposited on one end of the etched carbon fiber electrodes by an electrophoretic method. The carbon nanotube bundles formed a dense network on the carbon fiber surface. The electrochemical properties of the composite carbon electrodes were studied in the buffered neutral solutions. The results in cyclic voltammetry's characteristic indicate that the electrons on the electrodes transfer very fast. Furthermore, the redox reactions of dopamine (DA) on the composite electrodes show good sensitivity. When the DA concentration was 0.02 mM, the peak current in differential pulse technique reached 1.33 mu A after performing the background subtraction. In addition, the simultaneous detection of DA and ascorbic acid (AA) showed that the interference effect was not observed. It was suggested that the carbon composite microelectrodes have potential applications as electrochemical sensors inside a single cell.

Abstract:We report two 3D printed devices that can be used for electrochemical detection. In both cases, the electrode is housed in commercially available, polymer-based fittings so that the various electrode materials (platinum, platinum black, carbon, gold, silver) can be easily added to a threaded receiving port printed on the device; this enables a module-like approach to the experimental design, where the electrodes are removable and can be easily repolished for reuse after exposure to biological samples. The first printed device represents a microfluidic platform with a 500 × 500 μm channel and a threaded receiving port to allow integration of either polyetheretherketone (PEEK) nut-encased glassy carbon or platinum black (Pt-black) electrodes for dopamine and nitric oxide (NO) detection, respectively. The embedded 1 mm glassy carbon electrode had a limit of detection (LOD) of 500 nM for dopamine and a linear response (R(2) = 0.99) for concentrations between 25-500 μM. When the glassy carbon electrode was coated with 0.05% Nafion, significant exclusion of nitrite was observed when compared to signal obtained from equimolar injections of dopamine. When using flow injection analysis with a Pt/Pt-black electrode and standards derived from NO gas, a linear correlation (R(2) = 0.99) over a wide range of concentrations (7.6-190 μM) was obtained, with the LOD for NO being 1 μM. The second application showcases a 3D printed fluidic device that allows collection of the biologically relevant analyte adenosine triphosphate (ATP) while simultaneously measuring the release stimulus (reduced oxygen concentration). The hypoxic sample (4.8 ± 0.5 ppm oxygen) released 2.4 ± 0.4 times more ATP than the normoxic sample (8.4 ± 0.6 ppm oxygen). Importantly, the results reported here verify the reproducible and transferable nature of using 3D printing as a fabrication technique, as devices and electrodes were moved between labs multiple times during completion of the study.

Abstract:Dopamine (DA) neurotransmitter is a prominent component of the central nervous system and its deficiency contributes to several diseases, such as Parkinson's disease and schizophrenia, which significantly influence the quality of life of affected individuals. This paper introduces a circular interdigitated ring electrode, the three-dimensional carbon interdigitated ring array nanofibers (3D C-IDRA NFs), for the detection of DA using amperometric detection. The study utilized cyclic voltammetry and chronoamperometry to investigate DA. The integration of carbon nanofibers (CNFs) with 3D C-IDRA demonstrated enhanced surface area, increased current peak, and superior performance compared to the standalone 3D C-IDRA electrode, as evidenced by a redox amplification factor of 2.94 and a collection efficiency of 81.1%. The results highlight that integrating CNFs with 3D C-IDRA improves the electrochemical performance in detecting DA neurotransmitter. Thus, this discovery elucidates the advantageous potential of the 3D C-IDRA NF electrode in a wide range of applications.
