Abstract:Objective To investigate the efficacy of nano-silver bionic wound dressing.Methods Sprague Dawley rats(SD rats) were randomly divided into three groups:nano-silver dressing group(n=14),sulfadiazine silver(SD-Ag) group(n=14) and chitosan film group(n=14),and then deep partial-thickness wound involving 10～13% total body surface area(TBSA) were reproduced in the three groups.Blood samples on post-operation day 2,4,5,7,13 and 16 and liver,brain and kidney on post-operation day 45 were harvested for the determination of trace silver.Results The wound healing rate was higher in nano-silver dressing group than that in other two groups(P0.01).The silver level in tissues and blood was lower than SD-Ag group(P0.01).The blood silver content in nano-silver group returning to normal level was faster than that in SD-Ag group.Conclusion The nano-silver wound dressing can promote the healing of wound,and it can reduce the possibility of silver poisoning.Its good efficacy and safety shows a prosperous future in clinical setting.
An evaluation of the effects of caffeic acid phenethyl ester and Ankaferd blood stopper on secondary wound healing of oral mucosal tissue

Abstract:Background/aim: Caffeic acid phenethyl ester (CAPE) and Ankaferd Blood Stopper (ABS) are considered to contribute to wound healing. The purpose of this study was to investigate the effect of ABS and CAPE on secondary wound healing of oral mucosal tissue. Materials and methods: In total, 63 male Sprague-Dawley rats were used in this study. The animals were randomly divided into three groups and anaesthetized with ketamine (8 mg/100 g, intraperitoneally): a control group, CAPE group, and ABS group. A full-thickness excisional wound was created using a 4 mm punch biopsy tool. Topical ABS and CAPE were then applied in each group for 7, 14, and 21 days (n = 7 in each group). The animals in each group were sacrificed after 7, 14, and 21 days. Palatal specimens were stained with haematoxylin-eosin. Vascular endothelial growth factor (VEGF) and tumour necrosis factor-inducible gene 6 (TSG-6) protein expressions were determined using the Western blot method. Results: Inflammation, vessel dilatation, and haemorrhages were significantly lower in the CAPE group as compared with these parameters in the other groups (P < 0.05). Fibrosis was significantly higher in the ABS group as compared with that in the other groups (P < 0.05). VEGF protein levels were elevated in the 21-day CAPE group and 7-day ABS group. The expression of TSG-6 increased in the 7-day CAPE group and 21-day ABS group. Conclusion: Based on our findings, ABS and CAPE had positive effects on the oral wound healing process.

Abstract:Dressings are necessary during the process of wound healing. Since the early 1980s, several types of wound dressings have been produced, but they cannot always take into account some effects include antibacterial effect, wound healing promotion, and other properties. In this study, we would like to develop an effective dressing with the above properties, especially accelerating wound healing effect. A chitosan‐calcium alginate dressing (CCAD) was developed by coating mixture of chitosan with high‐low molecular weight on calcium alginate dressing (CAD). We investigated the structural characteristics of CCAD with Fourier‐transform infrared spectroscopy (FTIR) and electron microscopy. The cytotoxicity and antibacterial property were evaluated in vitro using CCK‐8 and inhibition zone method. Moisture retention was tested on the skin of Sprague‐Dawley (SD) rats, and wound healing studies were performed on a full‐thickness skin wound model in SD rats. CCAD showed good moisturizing and antibacterial properties with no cytotoxicity. CCAD could inhibit inflammation by decreasing IL‐6, and it could also promote angiogenesis by increasing VEGF, resulting in better wound healing than CAD. CCAD is a better choice in wound care due to its antibacterial property, biocompatibility, moisture retention, healing promotion, and non‐cytotoxicity characteristics.

Abstract:Aims: Whether ankaferd blood stopper (ABS) has a negative or positive effect on bone union during fracture healing is unknown. The purpose of this study was to evaluate the serum changes of oxidative stress markers, Tumor Necrosis Factor-α (TNF-α), Interleukin-1β (IL-1β), and Interleukin-10 (IL-10) during fracture healing process with or without ABS application to bone fracture. Material and methods: Eight rats were used as a control group (1) that was not subject to fracture. The remaining 48 rats were divided into six groups, 8 rats in each. The femoral shaft fracture was produced by cutting with bone-scissors. One ml of ABS was applied on the fracture region in groups 3 (7th day), 5 (21st day), and 7 (45th day) or saline instead of ABS on the fracture regions in groups 2 (7th day), 4 (21st day), and 6 (45th day). Radiographs and above parameters were examined on post-fracture days 7, 21, and 45. Results: Malondialdehyde and protein carbonyls were measured in high levels in the groups 2 and 4 with respect to control. Their levels did not change statistically in the experimental groups after ABS application. The values of TNF-α and IL-1β were elevated on 7th post-fracture day according to control, but were lower (by 11.86 % and 44.48 %) in the group 3 treated with ABS comparing to group 2. Radiographic examination indicated a low callus formation on fracture union in the femoral fractures of groups 3 and 5 treated with ABS. Conclusion: The present findings may suggest that ABS application seems to be ineffective on fracture union in early fracture healing period, except for bleeding control (Tab. 2, Fig. 3, Ref. 26).

Abstract:Background/Aim: Nowadays, many deaths are related to vessel injury-induced blood loss. Failure to control bleeding also increases the risk of death. This study aimed to investigate the hemostatic effects of the Algan Hemostatic Agent (AHA) foam application in a rat model in which severe femoral artery bleeding was induced. Methods: Fourteen rats were randomly assigned to two groups: (1) control (physiological saline) (n = 7) and (2) AHA foam (n = 7). The left femoral artery of the rats was incised and when the bleeding started, and the area was pressed with another sponge for 10 s in all rats. Afterwards, physiological saline solution impregnated gauze or AHA foam was placed over same area. A chronometer was started and area was checked after 2 min. If no bleeding occurred during the first 2 min of application, it was recorded as “successful”. If bleeding occurred, the same procedure was repeated up to three times. If hemostasis could not be achieved even after the third application, it was considered a failure, and “failed” was recorded. All animals were sacrificed under high anesthesia for least 10 min after the experiment. Results: Application of AHA resulted in complete (100%) control of bleeding in all rats within the first 2 min. In control group, hemostasis was achieved in 1 out of 7 (14.3%) rats by the third application. Failure was recorded for the remaining six rats. The hemostatic success rate of the AHA foam was significantly higher than the rates of control group (P = 0.005). Conclusion: AHA foam is a very effective hemostatic agent and can be applied easily on vascular trauma models. Further studies are needed to elucidate hemostatic features of AHA.

Abstract:Journal of Bioactive and Compatible Polymers, Ahead of Print. Gelatin is a collagen-derived biopolymer with high biocompatibility, commonly used in the production of wound dressing materials. Gelatin-based wound dressings, obtained through different processing methods, come with their own advantages and disadvantages depending on their physical structure. In this study, the effects of gelatin wound dressing materials with two different morphologies on wound healing are investigated in detail. The study investigated that the preparation of gelatin-based wound dressing materials using two different methods, followed by their chemical characterization and examination of their effects on biocompatibility and wound healing. To comprehend the impact of nanofiber structure and porous sponge structure on the wound healing process, the chemical structure of the prepared wound dressings was analyzed using Fourier Transform Infrared Spectrophotometer (FTIR). The morphological structure was visualized through Scanning Electron Microscope (SEM), the thermal properties were determined using Thermogravimetric Analyzer (TGA) and Differential Scanning Calorimeter (DSC). Additionally, the in vivo wound healing effects of gelatin-based wound dressings with different structures were evaluated through macroscopic observation and histological analysis in a dorsal region wound-healing rat model, monitored for 21 days. It was observed that wound healing materials with desired nanofiber and porous sponge structures were successfully obtained based on the SEM analysis. The gelatin nanofiber dressing showed a randomly oriented, bead-free, and smooth morphology with an average fiber diameter of approximately 1250 ± 0.35 nm, while the crosslinked gelatin sponge dressing had pore sizes ranging from 20 to 160 μm, and interconnected pore structures. The chemical structure remained unaltered according to FTIR graphs and these results were further supported by DSC and TGA graphs. Results from in vitro biocompatibility studies indicated that the wound dressing materials were biologically compatible, non-toxic, and did not cause irritation or sensitivity to the skin. Regarding in vivo analysis, it was determined that gelatin dressing materials were functional in the wound healing process, without the use of any auxiliary materials.

Abstract:The chronic wounds and burn wounds are considered as one of the most important medical conditions that must be properly managed due to the psychological and physical stress that cause for patients, as well as creating a huge financial burden on patients and global health care systems. Nowadays, there is a growing interest in developing nanofiber mats loaded with different types of plant extracts to meet the urgent need for advanced wound dressings. In this study, polyvinyl alcohol (PVA) nanofiber mats loaded with Cynara scolymus L. extract (CSE) were developed in three concentrations (10 wt%, 20 wt% and 40 wt%) by electrospinning. The morphology and chemical properties of the resulting nanofibers were identified using SEM and FTIR analysis. The results demonstrated the presence of evenly distributed, and bead-free nanofibers and the successful loading of the extract into the PVA fibers. The results proved the antimicrobial activity of PVA/CSE nanofiber mats against the most important microbial species that attributed to wound infections. Additionally, the hydrophilicity and the antioxidant activity of these nanofibers were determined by contact angle test and DPPH assay, respectively. The results demonstrated a hydrophilic nature and a high antioxidant activity for the nanofiber mats. Mechanical testing showed that increasing the percentage of CSE in nanofiber mats resulted in an improvement in tensile strength. The MTT test was performed to detect the toxicity of PVA/CSE nanofiber mats on L929 fibroblast cells and no cytotoxicity was found. Also, LC-MS/MS evaluation of CSE identified the significant amounts of phytochemicals which could enhance wound healing. All these findings enhance the opportunity of using these nanofibers as a promising bioactive wound dressing material.

Abstract:Chronic wounds represent a significant socio-economic problem, and the improvement of their healing is therefore an essential issue. This paper describes the preparation and biological properties of a novel functionalized nanofiber wound dressing consisting of a polycaprolactone nanofiber carrier modified by a drug delivery system, based on the lipid particles formed by 1-tetradecanol and encapsulated gentamicin and tocopherol acetate. The cytotoxicity of extracts was tested using a metabolic activity assay, and the antibacterial properties of the extracts were tested in vitro on the bacterial strains Staphylococcus aureus and Pseudomonas aeruginosa. The effect of the wound dressing on chronic wound healing was subsequently tested using a mouse model. Fourteen days after surgery, the groups treated by the examined wound cover showed a lower granulation, reepithelization, and inflammation score compared to both the uninfected groups, a lower dermis organization compared to the control, a higher scar thickness compared to the other groups, and a higher thickness of hypodermis and bacteria score compared to both the uninfected groups. This work demonstrates the basic parameters of the safety (biocompatibility) and performance (effect on healing) of the dressing as a medical device and indicates the feasibility of the concept of its preparation in outpatient conditions using a suitable functionalization device.

Abstract:OBJECTIVE: To evaluate effects of titanium dioxide /gelatin nanocomposite on wound healing in mice as a model study.METHODS: Fifty male rats were randomized into five groups of ten animals each. In group I, 0.1 mL sterile saline 0.9% solution was added to the wounds with no infection. In group II, the wounds were infected with MRSA and only treated with 0.1 mL the sterile saline 0.9% solution. In group III, infected wounds were treated with gelatin. In group IV, animals with infected wounds were treated with 0.1 mL titanium dioxide nanoparticles. In group V, animals with infected wounds were treated with titanium dioxide /gelatin nanocomposite. Wound size was measured on 2, 6, 10, 14, 18 and 20 days after surgery.RESULTS: Reduction in wound area indicated that there was significant difference between group IV and other groups (p<0.05). Quantitative histological and morphometric studies and mean rank of the qualitative studies demonstrated that there was significant difference between group IV and other groups (P<0.05).CONCLUSION: Titatnium dioxide nanoparticles/gelatin composite offered potential advantages in wound healing acceleration and fibroblast proliferation on early days of healing phases. Acceleration in wound repair could be associated with earlier wound contraction and stability of damaged area by rearrangement of granulation tissue and collagen fibers.
Acceleration of Wound Healing in Diabetic Rats through a Novel 3D Organo-Hydrogel Nanocomposite of Polydopamine/TiO2 Nanoparticles and Cu (PDA-TiO2@Cu)

Abstract:Background:: Diabetic wound represents a serious issue with a substantial impact and an exceptionally complex pathology affecting patients’ mental health and quality of life. So, we have developed a novel 3D organo-hydrogel nanocomposite of polydopamine/TiO2 nanoparticles and cu (PDA-TiO2@Cu) and examined its efficacy in diabetic wound healing. Methods:: Forty-five adult male albino rats were divided into normal control rats (non-diabetic rats with non-treated skin wounds), diabetic control rats (diabetic rats with non-treated skin wounds), and organo-hydrogel-treated rats (diabetic wounds treated with topically applied organo- hydrogel once daily). Macroscopic changes of the wound were observed on days 0, 3, 5, 7, and 10 to measure wound diameters. Skin specimens from the wound tissue were taken on days 3, 7, and 10, respectively, and examined histologically and immunohistochemically. Also, the gene expressions of collagen I, Matrix Metalloproteinase-9 (MMP-9), and Epidermal Growth Factor (EGF), and levels of Interleukin 6 (IL-6) and Superoxide Dismutase (SOD) were assessed. Results:: Our observed results indicated that the developed patch significantly accelerated the healing time compared to the normal control and diabetic control groups. Moreover, the patchloaded group revealed complete re-epithelization and a highly significant increase in the mean area % of CD31 immunostaining on day 7. The organo-hydrogel-loaded group displayed a significant decrease in gene expression of MMP-9 and a significant increase in gene expression of EGF and collagen I. Additionally, the organo-hydrogel-loaded group exhibited a significant decrease in levels of IL-6 and a significant increase in levels of SOD, compared to the normal diabetic control groups. Conclusion:: The organo-hydrogel can be used for treating and decreasing the healing period of diabetic wounds.

Qualitative/Chemical Analyses of Ankaferd Hemostat and Its Antioxidant Content in Synthetic Gastric Fluids

Abstract:Introduction. Ankaferd hemostat (ABS) is the first topical haemostatic agent involving the red blood cell-fibrinogen interactions. The antihemorrhagic efficacy of ABS has been tested in controlled clinical trials. The drug induces the formation of an encapsulated complex protein web with vital erythroid aggregation. The aim of this study is to detect the essential toxicity profile and the antioxidant molecules inside ABS. Methods. The pesticides were analyzed by GC-MS and LC-MS. The determination by ICP-MS after pressure digestion was performed for the heavy metals. HPLC was used for the detection of mycotoxins. Dioxin Response Chemically Activated Luciferase Gene Expression method was used for the dioxin evaluation. TOF-MS and spectra data were evaluated to detect the antioxidants and other molecules. Results. TOF-MS spectra revealed the presence of several antioxidant molecules (including tocotrienols, vitamin E, tryptophan, estriol, galangin, apigenin, oenin, 3,4-divanillyltetrahydrofuran, TBHQ, thymol, BHA, BHT, lycopene, glycyrrhetinic acid, and tomatine), which may have clinical implications in the pharmacobiological actions of ABS. Conclusion. The safety of ABS regarding the presence of heavy metals, pesticides, mycotoxins, GMO and dioxins, and PCBs was demonstrated. Thus the present toxicological results indicated the safety of ABS. The antioxidant content of ABS should be investigated in future studies.

Abstract:<h3>Abstract</h3><p>In order to improve healthcare of injured people, deacetylated acemannan extracted from <em>Aloe vera leaves</em>, having high inhibitory properties, was used as an antimicrobial finish on traditional cotton items. Response surface methodology was employed to define quadratic relationships between the polysaccharide grafting degree and the treatment process properties. An optimized modification process, offering the highest funtionalization degree, is obtained. The cellulosic fiber morphology and roughness modifications induced by polymer grafting are revealed using Atomic Force Microscopy and Scanning Electron Microscopy. Infra Red spectroscopy was used to confirm the grafting effectiveness. Thermogravimetric Analysis and Differential Scanning Calorimeter were further employed to confirm chemical modification. Considering the potential use of this new biomaterial, original properties were also studied. Finishing treatment seems to preserve mechanical properties, and hydrophilicity of the cellulosic substrate. MTT assay were done in HepG2 cells to ensure that the obtained dressings are non-toxic. The biomaterial showed high biocompatibility and promoted cell viability. Antimicrobial studies showed that grafting treatment conserved polymer antibacterial activity. Optimized cotton dressings exhibited a significant inhibitory effect against <em>Staphylococcus aureus</em> and <em>Escherichia coli</em> bacteria, killed respectively at 70.2% and 72.4%.</p>

Abstract:Purpose . Using the classical Ankaferd Blood Stopper (ABS) solution to create active hemostasis during partial nephrectomy (PN) may not be so effective due to insufficient contact surface between the ABS hemostatic liquid agent and the bleeding area. In order to broaden the contact surface, we generated a chimeric hemostatic agent, ABS nanohemostat, via combining a self-assembling peptide amphiphile molecule with the traditional Ankaferd hemostat. Materials and Methods . In order to generate ABS nanohemostat, a positively charged Peptide Amphiphile (PA) molecule was synthesized by using solid phase peptide synthesis. For animal experiments, 24 Wistar rats were divided into the following 4 groups: Group 1: control; Group 2: conventional PN with only 0.5 ml Ankaferd hemostat; Group 3: conventional PN with ABS + peptide gel; Group 4: conventional PN with only 0.5 ml peptide solution. Results . Mean warm ischemia times (WITs) were 232.8 ± 56.3 , 65.6 ± 11.4 , 75.5 ± 17.2 , and 58.1 ± 17.6 seconds in Group 1 to Group 4, respectively. Fibrosis was not different among the groups, while inflammation was detected to be significantly different in G3 and G4. Conclusions . ABS nanohemostat has comparable hemostatic efficacy to the traditional Ankaferd hemostat in the partial nephrectomy experimental model. Elucidation of the cellular and tissue effects of this chimeric compound may establish a catalytic spark and open new avenues for novel experimental and clinical studies in the battlefield of hemostasis.

Abstract:To reduce the bleeding time and to shorten the surgery time are vital to patients’ prog-nosis, therefore, in this study, high moisture absorption nonwoven composites are proposed to attain hemostasis in time. Polyacrylate fiber and Tencel® fibers at different blending ratios (10:90, 20:80, 30:70, 40:60, and 50:50) are used to form PT composite nonwoven. Next, composed of a 50:50 ratio, PT composite nonwoven exhibits the maximal vertical wicking height of 4.4 cm along the cross direction. Additionally, the UV-Vis absorption spectra analysis shows that at absorption waves of 413–415 nm, the occurring of distinct peaks suggests the presence of nanoparticles. The XRD patterns indicate the presence of silver nanoparticles with corresponding crystal planes of characteristic peaks at (111), (200), and (220). Polyacrylate/Tencel® nonwoven composites exhibit comparable adsorption capacity of blood and water molecules. In particular, 30PT composite nonwoven outperforms the control group, exhibiting 3.8 times and 4.7 times greater the water absorption and blood absorption, respectively. Moreover, a great number of red blood cells with a size of 4–6 μm agglomerate among fibers as observed in SEM images, while 6hr-PT composite dressing demonstrates the optimal antibacterial efficacy against Escherichia coli and Staphylococcus aureus, proven by the zone of inhibition being 1.9 mm and 0.8 mm separately. When in contact with plasma, hemostasis composites have plasma hemostasis prothrombin time of 97.9%, and activated partial thromboplastin time of 96.7%. As for animal hemostasis model, the arteria over the rats’ thigh bones is cut open perpendicularly, generating mass arteria hemorrhage. To attain hemostasis, it takes 46.5% shorter time when using composite dressings (experimental group) than the control group.

In Vivo Evaluation of Wound Healing Efficacy of Gel-Based Dressings Loaded with PycnogenolTM and Ceratothoa oestroides Extracts

Abstract:Ceratothoa oestroides and French maritime pine bark (PycnogenolTM) extracts are considered promising therapeutic agents in wound healing. This study explores the healing efficacy of composite dressings containing these extracts, aiming to enhance their stability and effectiveness, utilizing a low-temperature vacuum method for producing Sodium Alginate—Maltodextrin gel dressings. Surgical wounds were inflicted on SKH-hr2 hairless mice. Dressings were loaded with PycnogenolTM and/or C. oestroides extracts and assessed for their efficacy. Wound healing was primarily evaluated by clinical and histopathological evaluation and secondarily by Antera 3D camera and biophysical measurements. Dressings were stable and did not compromise the therapeutic properties of C. oestroides extract. All interventions were compared to the C. oestroides ointment as a reference product. Most of the wounds treated with the reference formulation and the C. oestrodes dressing had already closed by the 15th day, with histological scores of 7 and 6.5, respectively. In contrast, wounds treated with PycnogenolTM, either alone or in combination with C. oestroides, did not close by the end of the experiment (16th day), with histological scores reaching 15 in both cases. Furthermore, treatment with 5% PycnogenolTM dressing appeared to induce skin thickening and increase body temperature. The study underscores the wound healing potential of C. oestroides extracts and highlights the need for further research to optimize PycnogenolTM dosing in topical applications.

Abstract:Due to the difficulty of healing chronic wound, in the process of changing dressing, secondary damage on the tissue caused by adhesion should be prevented. In this study, the new dressing of particle hydrogels synthesized with poly-methyl methacrylate and poly-vinyl alcohol precursors were proposed. In addition, cell safety tests, animal’s allergic stimulation, and animal’s wound healing experiments were conducted for particle hydrogels. On one hand, in L929 cell experiment, the results of particle hydrogels extract 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide tests and lactate dehydrogenase test trial show that there are no safety concerns over particle hydrogels. On the other hand, New Zealand white rabbits were chosen for skin sensitization tests in animal trials, which show the consistent results. At last, wound healing tests used diabetes induction with 10-week-old rats and three-month-old Landrace pigs, with the tissue histology. In short, through this experiment, it is found that in the early phase of the diabetic rats and pigs’ wound healing, using particle hydrogels can enhance collagen formation, and achieve the goal of faster wound healing.
