Abstract:Bacterial cellulose (BC) is an ideal candidate for wound dressings due to its natural origin, exceptional water-holding capacity, pliability, biocompatibility, and high absorption capability. However, the lack of essential antimicrobial activity limits its biomedical applications. This study reported BC-based wound dressings containing silk fibroin protein (SF), offering the potential for biomimetic properties, and (-)-epigallocatechin-3-gallate (EGCG) for polyphenol-assisted surface modification to promote infectious wound healing. Glycerol was used as the carbon source to promote the formation of an adhesive layer by facilitating the β-sheet folding of SF, and different concentrations of EGCG were employed to interact with SF through strong hydrogen bonding facilitated by the polyphenolic groups. The functionalized membrane exhibited outstanding water-holding capacity, swelling ratio, and degradation properties, along with enhanced hydrophilicity, adhesiveness, and a remarkable free radical scavenging ability. Both in vitro and in vivo experiments confirmed its potent bacteriostatic activity. The composite membrane displayed excellent biocompatibility, including cellular and hemocompatibility. Importantly, it effectively promoted wound healing in murine back infections. These findings suggest the significant feasibility of the innovative modification approach, and that functionalized membranes have great potential as wound-dressing materials for infection management in future clinical applications.

Abstract:Bacterial cellulose is a bacterially derived polymer with great potential for application in wound healing due to its innate properties such as high biocompatibility and biodegradability. In addition to this, it is naturally biosynthesized by bacteria as a hydrogel, which makes it an optimal substrate for the treatment of dry wounds, where additional moisture is required to facilitate the healing process. However, this polymer lacks antibacterial properties. As bacterial infections are becoming increasingly common and difficult to treat due to antimicrobial resistance, it is of crucial importance to develop strategies for the modification of cellulose to ensure protection against bacterial contamination. In this study, a green-chemistry approach was proposed for the functionalization of cellulose to introduce antibacterial functional groups. Two different active agents, namely glycidyl trimethylammonium chloride and glycidyl hexadecyl ether, were used for the covalent derivatization of the hydroxyl groups of glucose through a heterogeneous reaction in basic aqueous conditions. The modified material was chemically and mechanically characterized by solid-state techniques and rheological measurements. A biological assessment was then carried out both using bacterial cells and human keratinocytes. It was observed that the functionalization performed induced a reduction of approximately half of the bacterial population within 24 h of direct contact withStaphylococcus aureussubsp. aureus Rosenbach 6538P(TM)andEscherichia coli(Migula) Castellani and Chalmers ATCC(R)8739(TM)(respectively, a reduction of 53% and 43% in the cell number was registered for the two strains). In parallel, cytotoxicity studies performed on keratinocytes (HaCaT cell line) showed cell viability in the range of 90 to 100% for up to 6 days of direct contact with both unmodified and modified samples. The morphology of the cells was also visually evaluated, and no significant difference was noted as compared to the control. Finally, thein vitroscratch assay evidenced good wound closure rates in the presence of the samples, with complete coverage of the scratched area after 5 days for both the modified cellulose and the positive control (i.e., keratinocytes growth medium). Overall, the modified hydrogel showed promising features, confirming its potential as an alternative substrate to develop a sustainable, antibacterial and biocompatible wound dressing.
Fabrication of Bacterial Cellulose-Based Dressings for Promoting Infected Wound Healing

Abstract:Bacterial cellulose (BC) holds several unique properties such as high water retention capability, flexibility, biocompatibility, and high absorption capacity. All these features make it a potential material for wound healing applications. However, it lacks antibacterial properties, which hampers its applications for infectious wound healings. This study reported BC-based dressings containing ε-polylysine (ε-PL), cross-linked by a biocompatible and mussel-inspired polydopamine (PDA) for promoting infectious wound healing. BC membranes were coated with PDA by a simple self-polymerization process, followed by treating with different contents of ε-PL. The resulted membranes showed strong antibacterial properties against tested bacteria by both in vitro and in vivo evaluations. The membranes also exhibited hemocompatibility and cytocompatibility by in vitro investigations. Moreover, the functionalized membranes promoted infected wound healing using Sprague–Dawley rats as a model animal. A complete wound healing was observed in the group treated with functionalized membranes, while wounds were still open for control and pure BC groups in the same duration. Histological investigations indicated that the thickness of newborn skin was greater and smoother in the groups treated with modified membranes in comparison to neat BC or control groups. Abstract:Wound dressing is commonly used for skin injuries. The design of wound dressing typically stems from the principles of open-wound management such as infection prevention, moisture balance and healing response. A new wound dressing comprising polyvinylpyrrolidone (PVP)–berberine hydrochloride (BHC)/PVP–cellulose acetate (CA)–BHC/CA-aloin tri-layer Janus fiber was successfully fabricated using trifluid side-by-side electrospinning for antibacterial and wound healing functions. The fibrous membrane can hold 5.090 ± 0.276 times of its own water mass and achieve 6 h of microenvironment moisture retention. In vitro antibacterial results show that BHC exhibited selective resistance to S. aureus . The nanofibers were noncytotoxic and showed good cell adhesion properties and enhanced cell proliferation effect compared with BHC or aloin only tri-layer nanofibers. Owing to the specially designed tri-layer Janus structure, the in vitro drug-release profile of drugs-loaded fibrous membrane showed a combination of two-stage release for BHC (76.093 % ± 1.813 % release in first hour and 98.526 % ± 0.604 % release in 6 h) and sustained release for aloin (88.376 % ± 5.282 % release in 6 h). Therefore, the tri-layer Janus structure can be a potential choice for wound dressing application.

Abstract:Electrospun nanofibers made from chitosan are promising materials for surgical wound dressings due to their non-toxicity and biocompatibility. However, the antibacterial activity of chitosan is limited by its poor water solubility under physiological conditions. This study addresses this issue by producing electrospun nanofibers mainly from natural compounds, including chitosan and quaternized chitosan, which enhance both its solubility for electrospinning and the antibacterial activity of the resulting electrospun nanofibers. Additionally, antimicrobial agents like caffeic acid or berberine chloride were incorporated. The glutaraldehyde-treated nanofibers showed improved mechanical properties, with an average tensile strength exceeding 2.7 MPa, comparable to other chitosan-based wound dressings. They also demonstrated enhanced water stability, retaining over 50% of their original weight after one week in phosphate-buffered saline (PBS) at 37 °C. The morphology and performance of these nanofibers were thoroughly examined and discussed. Furthermore, these membranes displayed rapid drug release, indicating potential for inhibiting bacterial growth. Antibacterial assays revealed that S2-CX nanofibers containing caffeic acid were most effective against E. coli and S. aureus , reducing their survival rates to nearly 0%. Similarly, berberine chloride-containing S4-BX nanofibers reduced the survival rates of E. coli and S. aureus to 19.82% and 0%, respectively. These findings suggest that electrospun membranes incorporating chitosan and caffeic acid hold significant potential for use in antibacterial wound dressings and drug delivery applications.

Highly hygroscopicity and antioxidant nanofibrous dressing base on alginate for accelerating wound healing

Abstract:The removal of bacterium and free radicals is important for wound healing. Therefore, it is necessary to prepare biological dressings with antibacterial and antioxidant properties. In this study, high-performance calcium alginate/carbon polymer dots/forsythin composite nanofibrous membrane (CA/CPDs/FT) was explored under the influence of carbon polymer dots and forsythin. The addition of carbon polymer dots improved the nanofiber morphology and therefore enhanced the mechanical strength of the composite membrane. Moreover, CA/CPDs/FT membranes displayed satisfactory antibacterial and antioxidant properties because of the natural properties of forsythin. Meanwhile, outstanding hygroscopicity over 700% was also obtained for the composite membrane. In vitro and in vivo experiments showed that the CA/CPDs/FT nanofibrous membrane could prevent the invasion of bacteria, scavenge free radicals, and promote wound healing. Moreover, its good hygroscopicity and antioxidation characteristics were friendly for the clinical application of high-exudate wounds.
Antibacterial and wound healing properties of cellulose acetate electrospun nanofibers loaded with bioactive glass nanoparticles; in-vivo study

Abstract:Bioactive glasses (BGs) have gained great attention owing to their versatile biological properties. Combining BG nanoparticles (BGNPs) with polymeric nanofibers produced nanocomposites of great performance in various biomedical applications especially in regenerative medicine. In this study, a novel nanocomposite nanofibrous system was developed and optimized from cellulose acetate (CA) electrospun nanofibers containing different concentrations of BGNPs. Morphology, IR and elemental analysis of the prepared electrospun nanofibers were determined using SEM, FT-IR and EDX respectively. Electrical conductivity and viscosity were also studied. Antibacterial properties were then investigated using agar well diffusion method. Moreover, biological wound healing capabilities for the prepared nanofiber dressing were assessed using in-vivo diabetic rat model with induced wounds. The fully characterized CA electrospun uniform nanofiber (100–200 nm) with incorporated BGNPs exhibited broad range of antimicrobial activity against gram negative and positive bacteria. The BGNP loaded CA nanofiber accelerated wound closure efficiently by the 10th day. The remaining wound areas for treated rats were 95.7 ± 1.8, 36.4 ± 3.2, 6.3 ± 1.5 and 0.8 ± 0.9 on 1st, 5th, 10th and 15th days respectively. Therefore, the newly prepared BGNP CA nanocomposite nanofiber could be used as a promising antibacterial and wound healing dressing for rapid and efficient recovery.

Abstract:The present study aimed to evaluate the efficacy of cellulose acetate/gelatin/nanohydroxyapatite (CA/Gel/nHA) nanocomposite mats as the wound dressing. The dressings were prepared with electrospinning of CA/Gel solutions containing 12.5, 25 and 50 mg nHA. The dressings were evaluated regarding their water uptake capacity, morphology, tensile strength, water vapour transmission rate, wettability and cellular response with L929 cell line. The results showed that the concentration of nHA had a direct correlation with porosity, water contact angle, water uptake, water vapor transmission rate and proliferation. In vivo studies showed that all dressings had higher wound closure percent than the sterile gauze, as the control. The highest wound closure value was achieved in the CA/Gel +25 mg nHA group, which showed 93.5 ± 1.6%. The histological and the histomorphometric examinations of the wounds revealed that the CA/Gel +25 mg nHA dressing had the greatest collagen synthesis, re-epithelialization, neovascularization and also the best cosmetic appearance. Based on our finding, it could be concluded the applicability of electrospun nanofibrous CA/Gel/nHA dressings for successful wound treatment.

Abstract:The skin, as the largest organ, serves as a protective barrier against external stimuli. However, when the skin is injured, wound healing becomes a complex process influenced by physiological conditions, bacterial infections, and inflammation. To improve the process of wound healing, a variety of wound dressings with antibacterial qualities have been created. Electrospun nanofibers have gained significant attention in wound dressing research due to their large specific surface area and unique structure. One interesting method for creating Janus-structured nanofibers is side-by-side electrospinning. This work used side-by-side electrospinning to make cellulose acetate/gelatin Janus nanofibers. Curcumin and zinc oxide nanoparticles were added to these nanofibers. We studied Janus nanofibers' physicochemical characteristics and abilities to regulate small-molecule medication release. Janus nanofibers coated with zinc oxide nanoparticles and curcumin were also tested for antibacterial activity. The Janus nanofibers with specified physicochemical characteristics were successfully fabricated. Nanofibers released small-molecule medicines in a controlled manner. Additionally, the Janus nanofibers loaded with curcumin exhibited excellent antibacterial capabilities. This research contributes to the development of advanced wound dressings for promoting wound healing and combating bacterial infections.

Abstract:Wound healing requires careful, directed, and effective therapies to prevent infections and accelerate tissue regeneration. In light of these demands, active biomolecules with antibacterial properties and/or healing capacities have been functionalized onto nanostructured polymeric dressings and their synergistic effect examined. In this work, various antibiotics, nanoparticles, and natural extract-derived products that were used in association with electrospun nanocomposites containing cellulose, cellulose acetate and different types of nanocellulose (cellulose nanocrystals, cellulose nanofibrils, and bacterial cellulose) have been reviewed. Renewable, natural-origin compounds are gaining more relevance each day as potential alternatives to synthetic materials, since the former undesirable footprints in biomedicine, the environment, and the ecosystems are reaching concerning levels. Therefore, cellulose and its derivatives have been the object of numerous biomedical studies, in which their biocompatibility, biodegradability, and, most importantly, sustainability and abundance, have been determinant. A complete overview of the recently produced cellulose-containing nanofibrous meshes for wound healing applications was provided. Moreover, the current challenges that are faced by cellulose acetate- and nanocellulose-containing wound dressing formulations, processed by electrospinning, were also enumerated.
Synthesis and characterization of cellulose nanofibers/chitosan/cinnamon extract wound dressing with significant antibacterial and wound healing properties

Abstract:Recently, bacterial cellulose-based wound dressings have gained lots of attention, and many studies have tried to improve the efficacy of these membranes with incorporation of different natural materials to fabricate an all-natural wound dressing with significant antibacterial and wound healing properties. In the present study, bacterial cellulose (BC) is produced from Gluconacetobacter xylinus to fabricate BC-based membranes. Then, chitosan (CS) and cinnamon extract (CE) are incorporated into the BC membranes to prepare an all-natural wound dressing. The obtained composite membrane (BC/CS/CE) is characterized according to chemical properties, surface morphology, mechanical properties, water absorptivity, water retention, CE release, antibacterial activity, biocompatibility, in vivo wound healing activity, and compared with BC and BC/CS membranes. The BC/CS/CE membrane is able to maintain appropriate moisture content for an acceptable period of time. Although the tensile strength and elongation at break values of the BC/CS/CE are slightly lower than the BC membrane, they are still in ideal ranges. The BC/CS/CE membrane exhibits significantly more antibacterial effects against Staphylococcus aureus and Escherichia coli in comparison with the BC and BC/CS which can be related to the sustained released profile of CE. In addition, BC/CS/CE is more biocompatible with L929 normal skin fibroblast cells in comparison with the BC and BC/CS membranes. Also, the BC/CS/CE membrane can meaningfully promote the wound healing progress compared with other membranes at the Wistar rats full-thickness skin wound model. The BC/CS/CE membrane-treated wounds have more developed dermis layer in comparison with other groups according to histopathological analyses. Taking together, the CS and CE enriched BC membranes can be a promising wound dressing with desirable properties.

Abstract:Abstract Bacterial cellulose is an extracellular polysaccharide produced by microorganisms, offering advantages such as high water‐holding capacity, flexibility, and biocompatibility. However, its lack of bactericidal activity hampers its wide application. Usnic acid, a secondary metabolite derived from lichens of the Usnea genus, is recognized for its antibacterial and anti‐biofilm efficiency, coupled with anti‐inflammatory properties. Its water insolubility presents challenges for wide utilization and stable release. Sanxan gel, a novel polysaccharide, exhibits exceptional freeze‐thaw stability, suspension properties, and high elasticity, rendering it effective as a suspending agent to improve the bioavailability of water‐insoluble drugs. In this study, a hydrogel membrane was designed by combining bacterial cellulose and usnic acid suspended in sanxan gel through a simple in situ microorganism fermentation. The obtained membranes demonstrated excellent ability for sustained drug release, strong eradication capability against tested bacteria in both in vitro and in vivo experiments, effective inhibition of biofilm formation, and excellent hemocompatibility and cytocompatibility. Additionally, the composite membranes promoted wound healing with reduced inflammation and bacterial infection in a full‐thickness wound infection model in mice. Gelatin-based nanofiber membranes loaded with curcumin and borneol as a sustainable wound dressing

Abstract:Infection is a huge obstacle to wound healing. Thus, to enhance the healing of infected wounds, wound dressings that permit the dual delivery of antimicrobials and antioxidants are highly desirable. In this study, a series of gelatin-based nanofiber membranes with different curcumin contents were fabricated via solution electrospinning. The obtained membranes were characterized in terms of their morphologies, in addition to their physical, mechanical, and in vitro properties. The results showed that the membranes maintained an integrated morphology, excellent water absorption capability, satisfactory mechanical properties, and a high dissolution rate of curcumin. The addition of curcumin and borneol conferred the membranes the ability to inhibit Staphylococcus aureus and eliminate free radicals. Furthermore, cytocompatibility testing using the L929 cell line confirmed the excellent biocompatibility of the membranes. These gelatin-based nanofiber membranes loaded with curcumin and borneol can therefore be considered as promising materials for dressing wounds. Moreover, the use of biodegradable polymers and environmentally sustainable production techniques in this system render it suitable for the commercial manufacture of composite membranes.

Abstract:The sustained release of antimicrobial therapeutics for wound dressing has become an attractive design strategy for prolonging the timespan of wound dressings and for reducing the risk of chronic wound infection. Recently, cellulose-based membrane has become a preferred option of wound dressings for the treatment of burn wounds and skin ulcers. In this work, novel cellulose membrane incorporated with mesoporous silica particles (SBA-15) was developed as an antimicrobial wound dressing with desirable sustained release functionality for targeting persistent bacterial pathogens. Attributed to a coated layer of calcium carbonate (CaCO3), SBA-15 particles were free from corrosion in alkaline condition during the preparation of cellulose-based composite membranes. SEM, TEM and BET results showed that the morphology, specific surface area, pore size and pore volume of pristine SBA-15 were preserved after the incorporation of CaCO3-coated SBA-15 into the cellulose matrix, while the mesoporous structure of SBA-15 was significantly disrupted without the use of CaCO3 coating. The resultant composite membranes containing 30 wt% SBA-15 (denoted as CM-Ca2-SBA(30%)) achieved 3.6 wt% of antimicrobial drug loading. Interestingly, CM-Ca2-SBA(30%) demonstrated the sustained release property of chloramphenicol for 270 h, driven by a two-stage drug release processes of SBA-15/cellulose. The water vapor permeability (WVTR) and swelling properties of composite membranes were shown to have complied with the primary requirements of wound dressing. Antibacterial assays revealed that strong antibacterial activities (144 h) of the composite membranes against Staphylococcus aureus and Eschericia coli were achieved. All results displayed that the strategy of coating silica with CaCO3 helps to obtain cellulose–silica composite membranes with desirable sustained release profiles and strong antibacterial activities. The antibacterial SBA-15/cellulose composite membranes show potential for the application of wound dressing.

Bilayer nanofibrous wound dressing prepared by electrospinning containing gallic acid and quercetin with improved biocompatibility, antibacterial, and antioxidant effects

Abstract:The purpose of this study was to prepare an antibacterial, antioxidant, and biocompatible bilayer nanofibrous wound dressing by using electrospinning. The micromorphology and bilayer structure characteristics of the GA-Qe-PVP-PCL nanofibers were analyzed by SEM. The physicochemical characteristics were analyzed by XRD and FTIR. The uptake, mechanical properties, water contact angle, water vapor transmission and in vitro drug release were evaluated. In addition, the effect of antibacterial, antioxidant and biocompatability of the nanofibers were evaluated, respectively. The SEM results showed that the GA-Qe-PVP-PCL nanofibers had a smooth surface, no beading phenomenon, and a prominent bilayer structure. The diameter and porosity of the drug-loading layer and waterproof support layer of the nanofibers were 842 ± 302 nm, 242 ± 50 nm, and 88.56 ± 1.67%, 94.49 ± 1.57%, respectively. Moreover, the water uptake, mechanical properties, water contact angle, and water vapor transmission showed ideal performance. The results of in vitro drug release indicated that GA and Qe were both released rapidly, which was conducive to accelerating wound healing. The GA-Qe-PVP-PCL nanofibers exhibited antibacterial effects against both bacteria as well as high antioxidant activity. Additionally, the GA-Qe-PVP-PCL nanofibers possessed good compatibility, could promote the proliferation, adhesion, and migration of L929 fibroblast cells. The nanofibers we developed met the requirements of ideal materials for wound dressing, which makes the nanofibers the potential to be a wound dressing for wound care. Graphical

Silver nanoparticles loaded triple-layered cellulose-acetate based multifunctional dressing for wound healing

Abstract:Chronic wounds present considerable challenges which delay their effective healing. Currently, there are several biomaterial-based wound dressings available for healing diverse wound types. However, most of commercial wound dressings are too expensive to be affordable to the patients belonging to the middle and lower socioeconomic strata of the society. Thus, in this investigation affordable triple layered nanofibrous bandages were fabricated using the layer-by-layer approach. Here, the topmost layer comprised of a hydrophilic poly vinyl alcohol layer, cross-linked with citric acid. The middle layer comprising of cellulose acetate was loaded with silver nanoparticles as an antibacterial agent, while the lowermost layer was fabricated using hydrophobic polycaprolactone. The triple-layered nanofibrous bandages having a nano-topography, exhibited a smooth, uniform and bead-free morphology, with the nanofiber diameter ranging between 200 and 300 nm. The nanofibers demonstrated excellent wettability, slow in vitro degradation, controlled release of nano‑silver and potent antibacterial activity against Gram-negative (E.coli) and Gram-positive (S. aureus) bacteria. The fabricated bandages had excellent mechanical strength upto 12.72 ± 0.790 M. Pa, which was suitable for biomedical and tissue engineering applications. The bandage demonstrated excellent in vitro hemocompatibility and biocompatibility. In vivo excisional wound contraction, along with H and E and Masson's Trichrome staining further confirmed the potential of the nanofibrous bandage for full-thickness wound healing. Pre-clinical investigations thus indicated the possibility of further evaluating the triple-layered nanofibrous dressing in clinical settings.

Abstract:Bacterial-associated wound infection and antibiotic resistance have posed a major burden on patients and health care systems. Thus, developing a novel multifunctional antibiotic-free wound dressing that cannot only effectively prevent wound infection, but also facilitate wound healing is urgently desired. Herein, a series of multifunctional nanocomposite hydrogels with remarkable antibacterial, antioxidant, and anti-inflammatory capabilities, based on bacterial cellulose (BC), gelatin (Gel), and selenium nanoparticles (SeNPs), are constructed for wound healing application. The BC/Gel/SeNPs nanocomposite hydrogels exhibit excellent mechanical properties, good swelling ability, flexibility and biodegradability, and favorable biocompatibility, as well as slow and sustainable release profiles of SeNPs. The decoration of SeNPs endows the hydrogels with superior antioxidant and anti-inflammatory capability, and outstanding antibacterial activity against both common bacteria (E. coli and S. aureus) and their multidrug-resistant counterparts. Furthermore, the BC/Gel/SeNPs hydrogels show an excellent skin wound healing performance in a rat full-thickness defect model, as evidenced by the significantly reduced inflammation, and the notably enhanced wound closure, granulation tissue formation, collagen deposition, angiogenesis, and fibroblast activation and differentiation. This study suggests that the developed multifunctional BC/Gel/SeNPs nanocomposite hydrogel holds a great promise as a wound dressing for preventing wound infection and accelerating skin regeneration in clinic.

Abstract:Recently, the design of polymer nanofibers using the electrospinning process has attracted much interest. Particularly the use of natural polymers has promoted many advantages in their biomedical applications. However, the combination of multiple natural polymers remains a great challenge in terms of electrospun production and applied performance. From this perspective, the current investigation highlights the study of the preparation of electrospun nanomaterial scaffolds based on combined natural polymers for improved wound healing performance. First, we have synthesized a crosslinked polymer by reacting microcrystalline cellulose (MC) and chitosan (CS) biopolymer via the intermediate of citric acid as a crosslinking agent. Then a natural propolis biomolecule was incorporated into the polymer network. Different MC/CS blend ratios of 90/10 and 70/30 were then used and various machine parameters were optimized to obtain nanofiber scaffolds with excellent strength and structures. SEM, IR, physicochemical, mechanical, and morpho-logical characterization were then performed. SEM evaluation revealed homogeneous and bead-free nanofibrous structures, with well-defined morphology and a random deposition that could accurately mimic the extracellular matrix of native skin. The calculated average nanofiber diameters for the MC/CS blend ratios at 90/10 and 70/30 were 431.4 and 441.2 nm, respectively. The results showed that when the chitosan amount increased, larger nanofibers with narrow diameter distribution appeared. The prepared nanomaterials had a significant and close water vapor permeability of about 1735.12 and 1698.52 g per m per day for the two blend ratios of 90/10 and 70/30, respectively. The examination of swelling behavior revealed a noteworthy enhancement in hydrophilicity, a necessary attribute for improved healing efficacy. FT-IR analysis confirmed the success and the stability of the chemical crosslinking reaction between the two biopolymers before nanofiber conception. Excellent mechanical properties were acquired, based on the chitosan content. Both developed nanofiber scaffolds exhibited high tensile strength and Young's modulus values. The incorporation of 30% chitosan versus 10% results in an increase in tensile strength of 11% and 14% in Young's modulus. Therefore, we could adjust the different mechanical properties simply by varying the mixing rate of the electrospun polymers. Using epithelial HepG2 cells, viability and kinetic cell adhesion assays were assessed to obtain biological evaluation. No cytotoxicity was observed and good cytocompatibility was confirmed. Functionalized nanofiber biomaterials with different MC/CS ratios substantiated significant bactericidal effectiveness against Gram-positive and Gram-negative bacterial culture strains. The novel functional electrospun wound dressing scaffold demonstrated effective and promising biomedical performance, healing both acute and chronic wounds.

Abstract:In this paper, a multifunctional wound dressing with sustained-release and antibacterial properties was prepared using cellulose acetate (CA) as the matrix and Dimethyloxallyl Glycine (DMOG) and silver nanoparticles (Ag-NPs) as the drug loading component. The scanning electron microscopy shows that the microstructure of the nanofibers is uniform and droplet free, and the diameters of CA and CA/β-CD/DMOG nanofibers are mainly concentrated in the range of 40–120 nm, and the diameters of CA/DMOG/Ag-NPs nanofibers are mainly concentrated in the range of 120–200 nm. In vitro release test confirms that the CA/DMOG/Ag-NPs nanofiber can slowly release DMOG within about 84 h, which abides by a typical diffusion, and the main driving force is the concentration gradient between the drug-carrying nanofibers and the releasing medium. The antibacterial performance test demonstrates that the material exhibits obvious antibacterial performance against both E. coli and B.subtilis bacteria with little adverse effects on cell viability proved by cell compatibility test. Taken together, CA/DMOG/Ag-NPs can be a wound dressing material with good application prospects to promote the healing of diabetic wounds.

Abstract:As the incidence of chronic wounds increases, the requirements for wound dressings are rising. The specific aim of this study is to propose a novel gallic acid (GA) functionalized silk fibroin (SF) and gelatin (Gel) composite wound dressing in which GA is used as an antibacterial and wound healing substance. Via electrospinning, SF, Gel, and GA mixed solutions could be conveniently fabricated into a composite nanofiber mat (SF-Gel-GA), consisting of uniform fibers with an average diameter around 134.57 ± 84 nm. The internal mesh structure of SF-Gel-GA provides sufficient drug loading capacity, proper moisture permeability, and proper degradation rate. SF-Gel-GA presents excellent biocompatibility. NIH-3T3 fibroblast cells could adhere and spread stably on the SF-Gel-GA surface with slightly promoted proliferation. In the presence of SF-Gel-GA, the growth of both Gram-positive and Gram-negative bacteria, includingStaphylococcus aureusandPseudomonas aeruginosa, is significantly inhibited in both plate and suspension cultures. A cutaneous excisional mouse wound model proves the efficient ability of SF-Gel-GA to promote wound healing. Compared with pure SF dressing and commercial Tegaderm Hydrocolloid3Mdressing, the wound closure rate with SF-Gel-GA treatment is significantly improved. The histological assessments further demonstrate SF-Gel-GA could facilitate collagen deposition, neovascularization, and epithelialization at wound sites to promote wound healing. In conclusion, a novel SF-Gel-GA composite wound dressing with efficient wound healing activities have been developed for chronic wound treatment with broad healing potential.
