Abstract:Wheat is a widely grown cereal crop that provides >20% daily protein intake and calories worldwide. Drought stress drastically affects the crucial physiological (relative water content; RWC), biochemical (proline content), and molecular attributes in wheat plants from germination to maturity stage and ultimately reduces the yield and quality traits. Given this, the present work was conducted to know the effect of salt seed priming on wheat crop. For this, we have used two wheat variety HUW-468 (V1), and HUW-510 (V2) primed with water (T2), Mg(NO3)2 (T3), ZnSO4 (T4), and combinations of Mg(NO3)2 and ZnSO4 salt (T5) with non-primed seed (T1) under water stress (W1) and control (W0) conditions. We have observed significant differences in plant height (84 to 90 cm), leaf number, total leaf area (431.82 to 505.46 cm2 plant−1), relative water content, chlorophyll content (4.78 to 5.86 mg g−1fresh weight), membrane stability index (MSI) (56.25 to 61.42%), superoxide dismutase (SOD) activity (133.47 to 150.10), MDA content, proline content (101.64 to 100.46), nitrate reductase activity (NRA), and nutrient (N, Zn, and Fe) contents under control and stress conditions. These results concluded that seed priming with a combination of Mg(NO3)2 and ZnSO4 salt is best for improved wheat crop traits under drought stress compared to individual salt and non-priming treatments. Overall, this analysis elucidated that seed priming strengthens the antioxidant capacity under drought conditions by enhancing antioxidant enzymes and alleviating oxidative stress.

Abstract:Abstract Background Drought has become a dangerous threat to reduce crop productivity throughout the world. Exogenous applications of regulators, micronutrients, and/or osmoprotectants for inducing drought-tolerance in field crops have been effectively adopted. A controlled pot study was performed to investigate the relative efficacy of salicylic acid (SA), zinc (Zn), and glycine betaine (GB) as foliar applications on the growth, tissues pigments content, relative water content (RWC), leaf gas-exchange, antioxidant enzymes activity, reactive oxygen species (ROS) accumulation, osmolytes contents, and the yield parameters of wheat plants subjected to two soil water conditions (85% field capacity: well-watered, 50% field capacity: water-deficient) during reproductive growth stages. Results Water deficient conditions significantly decreased the growth, yield parameters, RWC, photosynthesis pigment, and gas-exchange attributes except for intercellular CO 2 concentration. However, foliar applications remarkably improved the growth and yield parameters under water deficit conditions. Under drought condition, exogenous applications of SA, Zn, and GB increased the grain yield pot − 1 by 27.99, 15.23 and 37.36%, respectively, as compared to the control treatment. Drought stress statistically increased the contents of hydrogen peroxide (H 2 O 2 ), superoxide anion radical (O 2 •− ), and malonaldehyde (MDA), and elevated the harmful oxidation to cell lipids in plants, however, they were considerably reduced by foliar applications. Foliar applications of SA, Zn, and GB decreased MDA content by 29.09, 16.64 and 26.51% under drought stress, respectively, as compared to the control treatment. Activities of all antioxidant enzymes, proline content, and soluble sugar were increased in response to foliar applications under water deficit conditions. Conclusions Overall, foliar application of GB, SA, and Zn compounds improved the drought-tolerance in wheat by decreasing the ROS accumulation, promoting enzymatic antioxidants, and increasing osmolytes accumulation. Finally, GB treatment was most effective in thoroughly assessed parameters of wheat followed by SA and Zn applications to alleviate the adverse effects of drought stress.

Abstract:Drought is a severe environmental challenge of climate change instigating huge losses to forage production and quality. The biochemical and physiological responses of exogenous chitosan (Ct) to confer drought tolerance in alfalfa, is still unclear. Ct seed priming effects on underlying mechanisms were investigated in alfalfa to improve its forage yield and quality under drought stress. Initially, new promising exotic alfalfa genotypes (44) were tested under drought stress (80%, 60%, and 40% FC, field capacity) for their drought tolerance potential. Various Ct doses (0, 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2%) as seed priming were evaluated in second experiment under drought stress of 80% and 40% FC in terms of improved biomass. The identified PI502452 (drought-tolerant) and PI172982 (drought-sensitive) genotypes including Sgd. Lucerne (local variety) were primed to optimized Ct dose (0.6%), and further tested under same drought levels to understand the mechanisms of drought tolerance. Drought exposure remarkably inhibited the chlorophyll pigments and water status, thereby, reduced the biomass production of all tested alfalfas. Nevertheless, drought ameliorative effects of Ct seed priming were more evident in PI502452 and Sgd. Lucerne than PI172982 genotypes in terms of enhanced chlorophyll pigments, water relations, and upregulated antioxidative capacity. A marked effect of Ct seed priming was also exhibited on improved quality traits viz. crude protein, ether extract, ash, and nitrogen free extract of drought-stressed alfalfas. Ct seed priming is considered effective to modulate the physiological and biochemical processes for improving forage yield and quality of alfalfa.

Abstract:The protective effects against drought stress of the endophytic bacterium Bacillus subtilis 10-4 were measured by studying the priming response in two wheat (Triticum aestivum L.)—Ekada70 (E70) and Salavat Yulaev (SY)—lines, tolerant and susceptible to drought, respectively. B. subtilis 10-4 improved germination and growth parameters under normal conditions in both cultivars with the most pronounced effect observed in cv. E70. Under drought conditions, B. subtilis 10-4 significantly ameliorated the negative impact of stress on germination and growth of cv. E70, but had no protective effect on cv. SY. B. subtilis 10-4 induced an increase in the levels of photosynthetic chlorophyll (Chl) a, Chl b, and carotenoids (Car) in the leaves of cv. E70, both under normal and drought conditions. In cv. SY plants, bacterial inoculation decreased the contents of Chl a, Chl b, and Car under normal conditions, but pigment content were almost recovered under drought stress. B. subtilis 10-4 increased water holding capacity (WHC) of cv. E70 (but did not affect this parameter in cv. SY) and prevented the stress-induced decline in WHC in both cultivars. Notably, B. subtilis 10-4 increased endogenous salicylic acid (SA) concentration in both cultivars, especially in cv. E70. Moreover, B. subtilis 10-4 reduced drought-induced endogenous SA accumulation, which was correlated with the influence of endophyte on growth, indicating a possible involvement of endogenous SA in the implementation of B. subtilis-mediated effects in both cultivars. Overall, B. subtilis 10-4 inoculation was found to increase drought tolerance in seedlings of both cultivars, as evidenced by decreased lipid peroxidation, proline content, and electrolyte leakage from tissues of wheat seedlings primed with B. subtilis 10-4 under drought conditions.

Abstract:The germination, seedling vigor, crop establishment and yield of agronomically important crops is negatively affected by soil salinity. The current study aimed to investigate the ability of exogenous fertigation by sodium nitroprusside (SNP) to induce salt tolerance in four high yielding wheat cultivars (Sahar-06, Punjab-11, Millat-11 and Galaxy-13) that differ in their response to salt stress in terms of biomass production, oxidative defense mechanisms and grain yield. Three levels of SNP (0, 0.1 and 0.2 mM) were used for seed soaking. During soaking the seeds were kept in the dark. After soaking for 12 h the seeds were air-dried for 5 h before sowing. Salinity caused a significant reduction in biomass and grain yield, while it increased proline (Pro), ascorbic acid (AsA), hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents. Cultivar Sahar-06 and Galaxy-13 were found more tolerant to salinity based on shoot length root fresh and dry wights, 100 grain weight, decreased MDA and H2O2 accumulation, phenolic and ascorbic acid (AsA) contents, accumulation of proline, activities of SOD, POD and CAT as compared to the other cultivars. Seed priming with SNP was effective in reducing the adverse effects of salt stress induced oxidative stress on plant biomass and grain yield in all the studied wheat cultivars, but maximum amelioration of salt stress tolerance by SNP treatment was found in cv. Sahar-06. The increased salt tolerance in wheat plants by SNP seed priming might be due to the role of NO in improving seed vigor and germination and early establishment of seedlings with better growth. 0.1 mM SNP was found the most effective in improving salt tolerance, as compared to other SNP concentations. Exogenous SNP fertigation increased the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) and the contents of AsA, Pro and total phenolics content (TPC) in the salt stressed wheat plants. Our data indicate that SNP-priming induced salt tolerance by up-regulating the antioxidative defense mechanisms resulting in better biomass production and grain yield.

Abstract:Global warming and salinization are inducing adverse effects on crop yield. Drought priming has been proved to improve drought tolerance of plants at later growth stages, however, whether and how drought priming at early growth stage alleviating salinity stress at later growth stage and improving water use efficiency (WUE) of plants remains unknown. Therefore, two wheat cultivars were subjected to drought priming at the 4th and 6th leaf stage and subsequent moderate salinity stress at 100 mmol NaCl applied at the later jointing growth stage. The growth, physiological responses, ABA signaling and WUE were investigated to unravel the regulating mechanisms of drought priming on subsequent salinity stress. The results showed that drought priming imposed at the early growth stage improved the leaf and root water potential while attenuated the ABA concentration in the leaves ([ABA]leaf) for the primed plants, which increased the stomatal conductance (gs) and photosynthesis (Pn). Consequently, the biomass under the salinity stress was significantly increased due to earlier drought priming. Moreover, drought priming improved the specific leaf N content due to the facilitated root growth and morphology, and this could benefit high leaf photosynthetic capacity during the salinity stress period, improving the Pn and water uptake for the primed plants. Drought priming significantly improved plant level WUE (WUEp) due to considerably enhanced dry biomass compared with non-primed plants under subsequent salinity stress. The significantly increased leaf δ13C under drought priming further demonstrated that the improved leaf δ13C and WUEp was mainly ascribed to the improvement of Pn. Drought primed plants significantly improved K+ concentration and maintained the K+/Na+ ratio compared with non-primed plants under subsequent salinity stress, which could mitigate the adverse effects of excess Na+ and minimize salt-induced ionic toxicity by improving salt tolerance for primed plants. Therefore, drought priming at early growth stage could be considered as a promising strategy for salt-prone areas to optimize agricultural sustainability and food security under changing climatic conditions.

Abstract:Wheat plants are frequently exposed to combined herbicide and drought stress (HDS) which induces complex responses negatively, affects productivity, and is becoming more exacerbated with current climate change. In this work, we studied the influence of seed priming with endophytic bacteria Bacillus subtilis (strains 104 and 26D) on growth and tolerance of two wheat (Triticum aestivum L.) varieties (E70-drought tolerant; SY-drought susceptible) exposed to soil drought after application of selective herbicide Sekator® Turbo in pot experiments under controlled conditions; 17-day-old plants sprayed with herbicide and after 3 days were subjected to soil drought by stopping irrigating the plants for 7 days with subsequent resumption of normal irrigation (recovery). Additionally, the growth of tested strains (104, 26D) in the presence of different concentrations of herbicide Sekator® Turbo and drought (PEG-6000) were evaluated. It was established that both strains are herbicide and drought tolerant and capable to improve seed germination and early seedlings' growth under different herbicide and drought stress degrees. The results of pot experiments showed that HDS exposure declined growth (plant length, biomass), photosynthetic pigments (chlorophyll a and b), leaf area, and increased lipid peroxidation (LPO) and proline accumulation in plants, demonstrating higher damaging effects for SY variety. Strains 104 and 26D mitigated (in different levels) such negative impacts of HDS on growth of both varieties by increasing length of roots and shoots, biomass, photosynthetic pigments (chlorophyll a and b), and leaf area, reducing stress-caused LPO (i.e., malondialdehyde), and regulating proline biosynthesis, as well as contributing to a faster recovery of growth, photosynthetic pigments, and redox-status of plants in post-stress period in comparison with non-primed plants. These ultimately manifested in forming a better grain yield of both varieties primed with 104, 26D, and exposed to HDS. Thus, both strains 104 and 26D (which are herbicide and drought tolerant) may be used as seed priming agents to improve wheat HDS tolerance and grain yield; however, strain 104 more effectively protected plants of E70, while strain 26D-plants of SY. Further research should be focused on understanding the mechanisms that determine the strain and variety-specificity of endophytic symbiosis and the role of bacteria in the modulation of physiological states of primed plants under stress conditions, including HDS.

Abstract:Drought stress is a limiting factor for wheat production and food security. Drought priming has been shown to increase drought tolerance in wheat. However, the underlying mechanisms are unknown. In the present study, the genes encoding the biosynthesis and metabolism of abscisic acid (ABA) and jasmonic acid (JA), as well as genes involved in the ABA and JA signaling pathways were up-regulated by drought priming. Endogenous concentrations of JA and ABA increased following drought priming. The interplay between JA and ABA in plant responses to drought priming was further investigated using inhibitors of ABA and JA biosynthesis. Application of fluridone (FLU) or nordihydroguaiaretic acid (NDGA) to primed plants resulted in lower chlorophyll-fluorescence parameters and activities of superoxide dismutase and glutathione reductase, and higher cell membrane damage, compared to primed plants (PD) under drought stress. NDGA + ABA, but not FLU + JA, restored priming-induced tolerance, as indicated by a finding of no significant difference from PD under drought stress. Under drought priming, NDGA induced the suppression of ABA accumulation, while FLU did not affect JA accumulation. These results were consistent with the expression of genes involved in the biosynthesis of ABA and JA. They suggest that ABA and JA are required for priming-induced drought tolerance in wheat, with JA acting upstream of ABA.
Priming of seeds with cyanobacteria improved tolerance in wheat (Triticum aestivum L.) during post-germinative drought stress

Abstract:Seed priming is beneficial in improving the germination and subsequent growth performance of seeds under normal as well as stressed conditions. Mostly, natural and chemical substances have been used for priming. Microbes are also used in priming to counter the negative effects of stress and to improve productivity. In this context, the potential of cyanobacteria to secrete growth-promoting substances makes them ideal candidates for seed priming. Therefore, in the present study, seed priming was carried out using cyanobacteria to investigate its role in imparting drought stress tolerance during post-germination stages in the drought-sensitive cultivar (HD-2967) of wheat. For this, the wheat seeds were subjected to priming with cyanobacteria and the plants germinated were subjected to drought stress during tillering stage. Plants exposed to drought stress showed significant reduction in growth, pigment content and photosynthetic activity. However, plants developed from seeds primed with cyanobacteria, upon drought stress maintained better growth rate, relative water content, chlorophyll, carotenoid and photosynthetic activity. Furthermore, the plants developed from primed seeds showed less accumulation of superoxides, peroxides and lipid peroxidation products and higher activity of ascorbate peroxidise, catalase and glutathione reductase. On the other hand, plants exposed to stress without priming showed enhanced accumulation of free radicals and lipid peroxidation products and lower activity of antioxidant enzymes. The results show that seed priming with cyanobacteria improved growth during post germination stages in wheat plants exposed to drought stress through modulation of physiological and biochemical responses.

Abstract:Abstract Background The effectiveness of two PGPB; Azospirillum brasilense NO40 and Stenotrophomonas maltophilia B11 was investigated in enhancing the drought tolerance of wheat ( Triticum aestivum L.) seedlings cultivar Gemiza9. The inoculated or uninoculated grains were sown in unsterilized sandy soil and watered normally untill the 8 th day. Drought stress was initiated by completely withholding water for 7 days (until wilting). Samples were collected after 15 days from sowing to evaluate some growth criteria, damage and defense indicators and to analyze the roots’ protein pattern. Results The results showed that inoculating wheat seedlings with these strains significantly diminished the inhibitory effects of drought stress on the relative water content of roots, shoots and leaves; area of leaves; contents of pigments (chlorophyll a and b) and ascorbic acid; and on the protein patterns of roots. Moreover, the bacterial inoculation notably reduced the drought-induced damage indicated by lower leakage of electrolytes and less accumulation of Malondialdehyde and hydrogen peroxide, surprisingly with less enhanced production of proline and activities of catalase and peroxidase than their uninoculated counterparts. Under normal conditions, inoculating wheat plants with these PGPB resulted in significantly promoted growth and elevated contents of pigments and altered protein patterns of roots. Conclusion Overall, we can say that both Azospirillum brasilense NO40 and Stenotrophomonas maltophilia B11 were able to deactivate the growth inhibition in wheat seedlings to some extent, while maintaining a certain level of efficient protection against damage under drought stress.
Β-Aminobutyric Acid Increases Abscisic Acid Accumulation and Desiccation Tolerance and Decreases Water Use but Fails to Improve Grain Yield in Two Spring Wheat Cultivars under Soil Drying.

Abstract:A pot experiment was conducted to investigate the effect of the non-protein amino acid, β-aminobutyric acid (BABA), on the homeostasis between reactive oxygen species (ROS) and antioxidant defence during progressive soil drying, and its relationship with the accumulation of abscisic acid (ABA), water use, grain yield, and desiccation tolerance in two spring wheat (Triticum aestivum L.) cultivars released in different decades and with different yields under drought. Drenching the soil with 100 µM BABA increased drought-induced ABA production, leading to a decrease in the lethal leaf water potential (Ψ) used to measure desiccation tolerance, decreased water use, and increased water use efficiency for grain (WUEG) under moderate water stress. In addition, at severe water stress levels, drenching the soil with BABA reduced ROS production, increased antioxidant enzyme activity, and reduced the oxidative damage to lipid membranes. The data suggest that the addition of BABA triggers ABA accumulation that acts as a non-hydraulic root signal, thereby closing stomata, and reducing water use at moderate stress levels, and also reduces the production of ROS and increases the antioxidant defence enzymes at severe stress levels, thus increasing the desiccation tolerance. However, BABA treatment had no effect on grain yield of wheat when water availability was limited. The results suggest that there are ways of effectively priming the pre-existing defence pathways, in addition to genetic means, to improve the desiccation tolerance and WUEG of wheat.

Abstract:Wheat is an important staple crop not only in Pakistan but all over the globe. Although the area dedicated to wheat cultivation expands annually, the quantity of wheat harvested is declining due to various biotic and abiotic factors. Global wheat production and output have suffered as a result of the drought, which is largely driven by a lack of water and environmental factors. Organic fertilizers have been shown to reduce the severity of drought. The current research was conducted in semi-arid climates to mitigate the negative effects of drought on wheat during its critical tillering (DTS), flowering (DFS), and grain filling (DGFS) stages through the application of three different abscisic acid treatments: ABA0 (0 mgL-1) control, ABA1 (100 mgL-1) and ABA2 (200 mgL-1). Wheat growth and yield characteristics were severely harmed by drought stress across all critical development stages, with the DGFS stage being particularly vulnerable and leading to a considerable loss in yield. Plant height was increased by 24.25%, the number of fertile tillers by 25.66%, spike length by 17.24%, the number of spikelets per spike by 16.68%, grain count per spike by 11.98%, thousand-grain weight by 14.34%, grain yield by 26.93% and biological yield by 14.55% when abscisic acid (ABA) was applied instead of the control treatment. Moreover, ABA2 increased the more physiological indices (water use efficiency (36.12%), stomatal conductance (44.23%), chlorophyll a (24.5%), chlorophyll b (29.8%), transpiration rate (23.03%), photosynthetic rate (24.84%), electrolyte leakage (- 38.76%) hydrogen peroxide (- 18.09%) superoxide dismutase (15.3%), catalase (20.8%), peroxidase (- 18.09%), and malondialdehyde (- 13.7%)) of drought-stressed wheat as compared to other treatments. In the case of N, P, and K contents in grain were maximally improved with the application of ABA2. Through the use of principal component analysis, we were able to correlate our results across scales and provide an explanation for the observed effects of ABA on wheat growth and production under arid conditions. Overall, ABA application at a rate of 200 mgL-1 is an effective technique to boost wheat grain output by mitigating the negative effects of drought stress.

Abstract:Knowledge of short-term physiological adaption of wheat to soil water deficit is well understood, but little is known about seed ethephon priming effect on stress memory against drought at the tillering stage of wheat. The 42 leading and modern cultivars released between 1991 and 2012 were subjected to drought (45% Field water-holding capacity), and well-watered (75% Field water-holding capacity) for 3 weeks to evaluate the genetic progress of wheat against drought stress. Seeds of the drought-sensitive cultivar (CM104) primed with ethephon were subjected to soil water deficit and well-watered to uncover ethephon-induced stress responses at organic, cellular, and molecular levels. Soil water deficit (45% FC) occurred at the tillering stage resulted in 3.2 to 67.4% yield loss, and genetic improvement of wheat against drought resulted in the yield loss decreased at a rate of 0.70% per year, which was determined mainly by fertile spike. Seed ethephon priming maintained leaf water under soil water deficit at the tillering stage by improving root volume and dry weight, which maintained leaf water and improved the drought avoidance capability of wheat. Seed ethephon priming decreased leaf malondialdehyde content by regulating root ERF, root to leaf ABA signaling, leaf ROS-scavenging capability, and leaf osmotic regulation, which improved the drought tolerance capability of dryland wheat. Comparative root tips transcriptome analysis revealed that differentially expressed genes and pathways involved in ethylene-induced drought tolerance are associated with carbon metabolism, glutathione metabolism, and phenylpropanoid biosynthesis. At maturity, seed ethephon priming increased fertile spikes and reduced spikelets abortion, which provided extra 0.3 t ha−1 of grain yield. These results suggested that seed ethephon priming allowed the recall of long-lasting stress defensive memory, increasing fertile spikes and grain yield by both drought avoidance and drought tolerance.

Abstract:Drought is one of the major environmental constraints, affecting agricultural productivity worldwide. Allelopathic hormesis, low dose stimulatory effect of allelochemicals, offers a pragmatic solution in alleviating the adverse effects of drought stress in plants. This study, therefore, was conducted to evaluate the potential of brassica water extract in enhancing drought tolerance in wheat. The experiment was comprised on three factors viz. drought with three levels (100%, 60% and 30% field capacity; FC), different concentrations of brassica water extract (control, water priming, 0.5%, 1.0%, 1.5%, 2.0%, 2.5% and 3.0%) and two wheat cultivars, Ihsan-2016 (drought-tolerant) and Galaxy-2013 (drought-sensitive). Drought stress, particularly at 30% FC decreased the morpho-physiological attributes of both wheat cultivars; nevertheless, the seed priming with brassica water extract at 1.5% effectively enhanced tolerance against drought stress. Compared with control, seed priming with 1.5% brassica water extract increased the morphological attributes such as seedling length, their fresh and dry weights of both wheat cultivars in the range of 29–142% under 30% field capacity. Besides, 1.5% brassica water extract triggered the activities of antioxidant enzymes including superoxide dismutase, catalase and peroxidase (28–150%), decreased the hydrogen peroxide contents (5–71%) and enhanced chlorophyll a, b, and carotenoid contents (57–201%) as compared to control in both wheat cultivars under 30% field capacity. Vigorous growth and higher drought tolerance in wheat-cultivars under brassica water extract priming were related to improved chlorophyll contents, physiological attributes, better antioxidant defense system, and reduced H 2 O 2 based damaging effect.

Abstract:The effect of soil drenching with 10µM abscisic acid (ABA) on the physiological responses of two spring wheat (Triticum aestivum L.) cultivars released in different decades was evaluated when subjected to a water deficit at jointing or at booting. Exogenous ABA application increased the ABA concentration in the leaves, reduced the stomatal conductance (gs), slowed the rate of water use, decreased the lethal leaf water potential (ψ) used to measure desiccation tolerance and lowered the soil water content (SWC) at which leaf relative water content (RWC) began to decrease and wilting was observed. Exogenous ABA application also reduced reactive oxygen species (ROS) formation and increased antioxidant enzyme activity, leading to a reduction in the oxidative damage to lipid membranes in both cultivars exposed to water stress at jointing and booting. The decrease in leaf RWC and wilting occurred at lower values of SWC in the recently-released cultivar than in the earlier-released cultivar. The recently-released cultivar also had higher grain yield than the earlier-released cultivar at moderate water stress, but the grain yield in both cultivars was reduced by water stress and by the exogenous ABA treatment. However, exogenous ABA treatment increased transpiration efficiency for grain (TEG) of both cultivars under moderate water stress. These results indicate that ABA played an important role in slowing water use and enhancing the antioxidant defence during soil drying, but this did not result in increased yields under drought stress.

Abstract:Various studies showed that green leaf volatiles play vital roles in biotic and abiotic stresses. However, their role in drought resistance has never been studied. In order to investigate the possible role of (Z)-3-hexeny-1-yl acetate (Z-3-HAC, a type of green leaf volatile), various physiology experiments were performed to understand the physiological mechanisms of Z-3-HAC in alleviating wheat drought stress. Drought-resistant wheat seedlings were primed with 100 μM Z-3-HAC at the four-leaf stage before drought treatment. Morphological analysis showed that the primed seedlings grew better and showed higher biomass accumulation in both shoots and roots under drought stress. Additionally, exogenous Z-3-HAC significantly increased the total root length, total root surface area, and total root volume of the seedlings under drought stress. In primed seedlings, physiological measurements showed that the relative water content (RWC), net photosynthetic rate ( P n ), stomatal conductance ( G s ), transpiration rate ( T r ), maximal photochemical efficiency of photosystem II (PSII) ( F v / F m ), photochemical activity of PSII ( F v ′/ F m ′), total chlorophyll content, activities of antioxidant enzymes, and osmolyte accumulation were higher than the control under drought conditions. Furthermore, relative electrolyte conductivity (REC), intercellular CO 2 concentration ( C i ), reactive oxygen species (ROS) accumulation, and malondialdehyde (MDA) content were significantly lower than the non-primed seedlings. Lastly, principal component analysis (PCA) indicated that Z-3-HAC protected wheat seedlings against damage from drought stress mainly through antioxidant and osmoregulatory systems. This study opens a new field of Z-3-HAC application, and provides a new opportunity to improve wheat drought resistance.

Abstract:Abstract Plants of spring wheat (Triticum aestivum L. cv. Vinjett) were exposed to moderate water deficit at the vegetative growth stages six-leaf and/or stem elongation to investigate drought priming effects on tolerance to drought and heat stress events occurring during the grain filling stage. Compared with the non-primed plants, drought priming could alleviate photo-inhibition in flag leaves caused by drought and heat stress episodes during grain filling. In the primed plants, drought stress inhibited photosynthesis mainly through decrease of maximum photosynthetic electron transport rate, while decrease of the carboxylation efficiency limited photosynthesis under heat stress. The higher saturated net photosynthetic rate of flag leaves coincided with the lowered non-photochemical quenching rates in the twice-primed plants under drought stress and in the primed plants during stem elongation under heat stress. Compared to the non-priming treatment, drought priming either applied once or twice alleviated the grain yield reduction by drought stress during grain filling, and priming during the stem elongation stage alleviated yield loss by heat stress at grain filling. The higher concentration of abscisic acid in primed plants under drought stress could contribute to higher grain yield compared to the non-primed plants. Taken together, the results indicate that drought priming during vegetative stages improved tolerance to both drought and heat stress events occurring during grain filling in wheat.

Abstract:A comparative analysis was conducted to evaluate the effects of seed priming with endophytic bacterium Bacillus subtilis 10-4 (BS) on the hormonal system and cell wall tolerance (lipid peroxidation (LPO), electrolyte leakage (EL), and root lignin deposition) of two Triticum aestivum L. (wheat) varieties with contrasting drought sensitivities (Ekada 70—drought-tolerant (DT); Salavat Yulaev—drought-sensitive (DS)) under normal conditions and 12% polyethylene glycol-6000 (PEG)-induced osmotic stress. The results showed that under normal conditions, the growth stimulation in wheat plants by BS was attributed to changes in the hormonal balance, particularly an increase in endogenous indole-3-acetic acid (IAA) accumulation. However, under stress, a significant hormonal imbalance was observed in wheat seedlings, characterized by a pronounced accumulation of abscisic acid (ABA) and a decrease in the levels of IAA and cytokinins (CK). These effects were reflected in the inhibition of plant growth. BS exhibited a protective effect on stressed plants, as evidenced by a significantly lower amplitude of stress-induced changes in the hormonal system: maintaining the content of IAA at a level close to the control, reducing stress-induced ABA accumulation, and preventing CK depletion. These effects were further reflected in the normalization of growth parameters in dehydrated seedlings, as well as a decrease in leaf chlorophyll degradation, LPO, and EL, along with an increase in lignin deposition in the basal part of the roots in both genotypes. Overall, the findings demonstrate that BS, producing phytohormones, specifically IAA and ABA, had a more pronounced protective effect on DT plants, as evidenced by a smaller amplitude of stress-induced hormonal changes, higher leaf chlorophyll content, root lignin deposition, and lower cell membrane damage (LPO) and permeability (EL) compared to DS plants.
