Abstract:Here we synthesize current understanding of the magnitudes and methods for assessing human and wildlife exposures to poly‐ and perfluoroalkyl substances (PFAS). Most human exposure assessments have focused on two to five legacy PFAS and wildlife assessments are typically limited to targeted PFAS (up to ~30 substances). However, shifts in chemical production are occurring rapidly and targeted methods for detecting PFAS have not kept pace with these changes. Total fluorine (TF) measurements complemented by suspect screening using high resolution mass spectrometry are thus emerging as essential tools for PFAS exposure assessment. Such methods enable researchers to better understand contributions from precursor compounds that degrade into terminal perfluoroalkyl acids (PFAA). Available data suggest that diet is the major human exposure pathway for some PFAS but there is large variability across populations and PFAS compounds. Additional data on TF in exposure media and the fraction of unidentified organofluorine are needed. Drinking water has been established as the major exposure source in contaminated communities. As water supplies are remediated, and for the general population, exposures from dust, personal care products, indoor environments and other sources may be more important. A major challenge for exposure assessments is the lack of statistically representative population surveys. For wildlife, bioaccumulation processes differ substantially between PFAS and neutral lipophilic organic compounds, prompting a revaluation of traditional bioaccumulation metrics. There is evidence that both phospholipids and proteins are important for the tissue partitioning and accumulation of PFAS. New mechanistic models for PFAS bioaccumulation are being developed that will assist in wildlife risk evaluations.This article is protected by copyright. All rights reserved.

Abstract:Per- and poly-fluoroalkyl substances (PFAS) are chemicals of concern—they are ubiquitous, persistent, with known and suspected health impacts. Well studied, primary sources of exposure to PFAS are drinking water and food. The presence of PFAS in human tissue of general populations suggests other important exposure sources/pathways. House dust measurements suggest widespread presence of PFAS in residences. Limited studies report paired analyses of PFAS occurrence in indoor media and PFAS concentrations in serum. While paired samples of house dust and blood serum are currently rare, the National Children's Study (NCS) contains paired samples, as well as sociodemographic information, from pregnant people that participated in the study. These archived NCS data and specimens for 104 participants collected between 2009 and 2014 were leveraged and analyzed for 16 commonly measured PFAS. We evaluated PFAS levels in the home, and the relationships between PFAS in dust and serum, and sociodemographic or housing variables. In addition, mechanistic exposure models, and then steady-state serum level models with simple parameters were used to estimate dust contributions of PFAS to serum. The geometric means for the most commonly found PFAS (full names in table 1) in serum were: 4.1 ng/mL for PFOS, 1.1 ng/mL for PFOA, 0.87 ng/mL for PFHxS, 0.16 ng/mL for PFDA. The geometric means of PFAS in dust were: 17 μg/kg for PFOS, 16 μg/kg for PFOA, 9.6 μg/kg for PFDS, 4.5 μg/kg for PFHpA, 4.4 μg/kg for PFNA, 3.9 μg/kg for PFHxS, 3.5 μg/kg for PFDA, 2.3 μg/kg for PFDoA, 2.1 μg/kg for PFUdA. PFOA was significantly correlated in serum and dust as was the sum of all PFAS detected in > 50 % of serum and dust. PFAS in serum was significantly associated with: Higher income, recent renovations, years lived in the home, and educational attainment. PFAS in dust was significantly associated with: Higher participant age, type of home, amount of carpet, educational attainment, higher income, recent renovation, and membership in the military. For some PFAS, 25 % of the overall exposure, on average, is from dust, but for others, 3–4 % is attributed to dust..We were able to identify important associations in PFAS exposure in the homes of pregnant people based on paired serum and dust samples. This built a clearer picture of which PFAS and at what quantities they exist in these homes, how they relate to each other, and how they are tied to sociodemographic and housing factors. Our results demonstrate that exposure to PFAS via house dust may contribute up to 25% of total exposure for adults, highlighting the importance of understanding what drives residential exposures.

Abstract:Per- and polyfluoroalkyl substances (PFASs) are widely used in food packaging, non-stick pots, and surfactants. However, their persistence in the environment, hazardous nature, and potential for bioaccumulation and longrange transport have alarmed an increasing number of scholars and research institutions. Although several literature reviews on PFASs research exist, only a few of them have considered bibliometric indices. In this study, 3,373 PFASs-related articles published between 2001 and 2018 were analyzed using a bibliometric analysis method based on the Science Citation Index (SCI) Expanded. The software tools for mapping knowledge domain (MKD) (VOSviewer and Science of Science (Sci2)) were used to analyze the performance of contributors and PFASs research topics, hotspots, and trends. Our results reveal that the number of PFASs-related articles published annually has increased significantly, with most originating from the United States (followed by those from China). The Chinese Academy of Sciences has published the most articles. A comprehensive analysis of title, keywords, and keywords plus showed that PFASs research hotspots include humans, precursors, and detection methods, with the main focuses being environmental science, toxicology, and environmental engineering. The four main research topics of PFASs were identified, and a literature review was carried out for each one. Overall, this study can supply researchers with a deeper understanding of the development of PFASs studies and provide a comprehensive data reference for researchers to further grasp the research direction in this field.
Method development and evaluation for the determination of perfluoroalkyl and polyfluoroalkyl substances in multiple food matrices

Abstract:A method for the determination of 21 perfluorinated and 10 polyfluorinated alkyl substances (PFAS) was developed for application in different food matrices. Acetonitrile was used as the extraction solvent with solid phase extraction weak anion-exchange (SPE-WAX) clean up, with LC-MS/MS analysis using both surrogate and performance standards to correct for losses during sample preparation and matrix effects. The method has been evaluated in four different matrices (fish, pizza, chicken nuggets and spinach). Originally, the focus was to develop a method for foods commonly thought to be a source of PFASs (e.g. fish). It was expanded to include foods where PFAS exposure would be possible through their presence in grease-proof food packaging (e.g. pizza, chicken nuggets). Vegetables (lettuce) and fruit (tomato) have recently been considered as part of proficiency testing programmes, so the inclusion of some testing in a vegetable matrix (i.e. spinach) was also added to the testing. Limits of quantification ranged from 0.018 ng g-1 (L-PFDS) to 5.28 ng g-1 (FHEA), although method quantification limits for PFBA (12.4 ng g-1), 6:2 PAP (8.96 ng g-1) and 8:2 PAP (3.49 ng g-1) were elevated above instrumental limits owing to their consistent detection in reagent blank samples. PFAS analyses were strongly impacted by matrix, therefore the use of isotopically labelled internal standards was critical to the development of accurate results. The accuracy of the method using numerous proficiency testing schemes or interlaboratory comparison studies has shown the developed method to be successful with z-scores for all concerned analytes in all test matrices remaining within ±2.0, with the exception of PFBA in wheat flour which was -2.4.
PFAS Exposures and the Human Metabolome: A Systematic Review of Epidemiological Studies

Abstract:Purpose of review: There is a growing interest in understanding the health effects of exposure to per- and polyfluoroalkyl substances (PFAS) through the study of the human metabolome. In this systematic review, we aimed to identify consistent findings between PFAS and metabolomic signatures. We conducted a search matching specific keywords that was independently reviewed by two authors on two databases (EMBASE and PubMed) from their inception through July 19, 2022 following PRISMA guidelines. Recent findings: We identified a total of 28 eligible observational studies that evaluated the associations between 31 different PFAS exposures and metabolomics in humans. The most common exposure evaluated was legacy long-chain PFAS. Population sample sizes ranged from 40 to 1,105 participants at different stages across the lifespan. A total of 19 studies used a non-targeted metabolomics approach, 7 used targeted approaches, and 2 included both. The majority of studies were cross-sectional (n = 25), including four with prospective analyses of PFAS measured prior to metabolomics. Summary: Most frequently reported associations across studies were observed between PFAS and amino acids, fatty acids, glycerophospholipids, glycerolipids, phosphosphingolipids, bile acids, ceramides, purines, and acylcarnitines. Corresponding metabolic pathways were also altered, including lipid, amino acid, carbohydrate, nucleotide, energy metabolism, glycan biosynthesis and metabolism, and metabolism of cofactors and vitamins. We found consistent evidence across studies indicating PFAS-induced alterations in lipid and amino acid metabolites, which may be involved in energy and cell membrane disruption.
The role of angiotensin II in the control of aldosterone in man.

Abstract:1. The relationship between circulating plasma angiotensin II and plasma aldosterone in man during sodium deficiency has been investigated in normal male subjects by comparing the amounts of the two hormones: (a) after 5 days of sodium restriction with an injection of frusemide on the third day, (b) during intravenous infusions of angiotensin II given in normal sodium balance. 2. The results show that aldosterone and angiotensin II plasma concentrations were both increased by sodium deficiency, but the increase in aldosterone was much greater than was found when angiotensin II was infused during normal sodium balance so as to equal or even exceed the plasma angiotensin II concentrations of sodium deficiency. 3. Other factors capable of increasing aldosterone secretion (plasma Na+, K+ and ACTH) seemed unlikely to account for the discrepancy, nor was there evidence of sufficient increase in adrenal sensitivity during Na+ deficiency. 4. It is suggested that some other unknown factor or factors are necessary to account for the full aldosterone response to this stimulus in man.

Idiopathic ventricular premature contractions originating from the postero-lateral tricuspid annulus leading to left ventricular disfunction.

Abstract:A 19-year-old patient with premature ventricular complexes (PVCs) more than 30% on 24 hours was addressed for catheter ablation. Echocardiography showed left ventricular (LV) dilation and systolic dysfunction with 37% of ejection fraction. The patient underwent investigations including cardiac magnetic resonance (MRI) with no other cause of cardiomyopathy being found. Successful ablation of the postero-lateral tricuspid annulus ectopic focus using conventional radiological guiding resulted in normalization of the LV size and contractility. No PVCs were found at follow-up. To our knowledge, this is the first report of PVCs originating in the postero-lateral tricuspid annulus resulting in LV dysfunction.

Abstract:The National Academies of Sciences, Engineering, and Medicine recommends per- and polyfluoroalkyl substance (PFAS) blood testing for patients with risk of elevated exposure, and the Agency for Toxic Substances and Disease Registry (ATSDR) suggests PFAS blood testing based on exposure. Barriers to PFAS blood testing include cost, access to labs, and evolving laboratory methods. We quantify water and serum PFAS levels among a highly-exposed cohort in an area with groundwater contaminated by historical agricultural biosolid application. We compare the gold standard PFAS serum test with a commercial test and results from a one-compartment toxicokinetic model. Participants were adults (n = 30) whose household (n = 19) water had levels of the sum of six PFAS > 500 ng/L. Serum PFAS were measured using liquid chromatography-tandem mass spectrometry. Demographic and water consumption data were collected via telephone. Serum PFAS results from the commercial test were accessed via medical record. Statistical analysis included descriptive statistics and bivariate plots of serum levels. Perfluorohexanoic acid, perfluoroheptanoic acid (PFHpA), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorobutanesulfonic acid, perfluorohexanesulfonic acid (PFHxS), and perfluorooctanesulfonic acid (PFOS) were detected in 19 wells, and PFHpA, PFOA, PFNA, perfluorodecanoic acid, perfluoroundecanoic acid, PFHxS, and PFOS were detected in at least 19 participants' serum. In well water, PFOA and PFOS levels had geometric means (GMs) of 1749 ng/L (geometric standard deviation [GSD] 2.4) and 887 ng/L (GSD 19.7), respectively. In serum, PFOA and PFOS had GMs of 116.2 μg/L (GSD 13.5) and 58.3 μg/L (GSD 13.8), respectively. Our results are comparable with and had a wider mix of PFAS than other high-exposure cohorts. There was good agreement between the commercial and gold standard tests for PFOA, PFNA, and PFHxS and mixed agreement between the gold standard test and modeled predictions, suggesting water-based toxicokinetic models of serum PFAS may be inadequate for assessing exposure in this population.

Abstract:Extensive use of per- and polyfluoroalkyl substances (PFAS) has resulted in their ubiquitous presence in human blood. PFAS exposures have been associated with multiple adverse human health effects including increased risk of liver damage, elevated serum lipids, impaired vaccine response, adverse birth outcomes and cancer. Biomonitoring studies have focused on measuring long-chain PFAS, but these are being replaced by shorter chain PFAS and PFAS with alternative structures, resulting in incomplete understanding of human exposures. Here, we take advantage of serum samples collected as part of a clinical trial testing the efficacy of a dietary fiber intervention to reduce serum cholesterol to investigate exposure to legacy and replacement PFAS chemicals in Canadian participants. Serum samples were collected from 72 participants in 2019-2020 at baseline and after 4 weeks of the intervention and were analyzed for 17 PFAS species. The highest geometric mean concentrations of PFAS measured at baseline corresponded to PFOSA (7.1 ng/ml), PFOS (4.2, ng/ml), PFOA (1.8 ng/ml) and PFHxS (1.3 ng/ml). Short chain PFAS including PFBuA, PFHxS and/or PFHpA were detected in 100% of participants and GenX was detected in 70% of participants. Analyses of associations between PFAS serum concentrations and biomarkers of adverse health outcomes showed the PFBuA, PFHxA, PFDA and PFOSA were associated with higher serum gamma-glutamyl transferase concentrations but not with measures of serum total or low-density lipoprotein cholesterol. Comparison of PFAS concentrations at baseline and after a 4-week follow-up showed that total PFAS concentrations decreased in both the control and cholesterol intervention groups. However, the suite of PFAS of concern identified by the United States National Academies of Sciences, Engineering, and Medicine, significantly decreased only in the cholesterol intervention group. This observation suggests that a dietary fiber intervention may reduce PFAS body burden, but future intervention studies need to control for PFAS exposure sources and extend beyond 4 weeks. Overall, the results show that exposures to short-chain and alternative PFAS are common in this Canadian population.

Abstract:Abstract Background There is a concern that continued emissions of man-made per- and polyfluoroalkyl substances (PFASs) may cause environmental and human health effects. Now widespread in human populations and in the environment, several PFASs are also present in remote regions of the world, but the environmental transport and fate of PFASs are not well understood. Phasing out the manufacture of some types of PFASs started in 2000 and further regulatory and voluntary actions have followed. The objective of this review is to understand the effects of these actions on global scale PFAS concentrations. Methods Searches for primary research studies reporting on temporal variations of PFAS concentrations were performed in bibliographic databases, on the internet, through stakeholder contacts and in review bibliographies. No time, document type, language or geographical constraints were applied in the searches. Relevant subjects included human and environmental samples. Two authors screened all retrieved articles. Dual screening of 10% of the articles was performed at title/abstract and full-text levels by all authors. Kappa tests were used to test consistency. Relevant articles were critically appraised by four reviewers, with double checking of 20% of the articles by a second reviewer. Meta-analysis of included temporal trends was considered but judged to not be appropriate. The trends were therefore discussed in a narrative synthesis. Results Available evidence suggests that human concentrations of perfluorooctane sulfonate (PFOS), perfluorodecane sulfonate (PFDS), and perfluorooctanoic acid (PFOA) generally are declining, while previously increasing concentrations of perfluorohexane sulfonate (PFHxS) have begun to level off. Rapid declines for PFOS-precursors (e.g. perfluorooctane sulfonamide, FOSA) have also been consistently observed in human studies. In contrast, limited data indicate that human concentrations of PFOS and PFOA are increasing in China where the production of these substances has increased. Human concentrations of longer-chained perfluoroalkyl carboxylic acids (PFCAs) with 9–14 carbon atoms are generally increasing or show insignificant trends with too low power to detect a trend. For abiotic and biological environmental samples there are no clear patterns of declining trends. Most substances show mixed results, and a majority of the trends are insignificant with low power to detect a trend. Conclusions For electrochemically derived PFASs, including PFOS and PFOA, most human studies in North America and Europe show consistent statistically significant declines. This contrasts with findings in wildlife and in abiotic environmental samples, suggesting that declining PFOS, PFOS-precursor and PFOA concentrations in humans likely resulted from removal of certain PFASs from commercial products including paper and board used in food packaging. Increasing concentrations of long-chain PFCAs in most matrices, and in most regions, is likely due to increased use of alternative PFASs. Continued temporal trend monitoring in the environment with well-designed studies with high statistical power are necessary to evaluate the effectiveness of past and continuing regulatory mitigation measures. For humans, more temporal trend studies are needed in regions where manufacturing is most intense, as the one human study available in China is much different than in North America or Europe.

Abstract:Per- and polyfluoroalkyl substances (PFAS) represent a class of persistent synthetic chemicals extensively utilized across industrial and consumer sectors, raising substantial environmental and human health concerns. Epidemiological investigations have robustly linked PFAS exposure to a spectrum of adverse health outcomes. Altered metabolites stand as promising biomarkers, offering insights into the identification of specific environmental pollutants and their deleterious impacts on human health. However, elucidating metabolic alterations attributable to PFAS exposure and their ensuing health effects has remained challenging. In light of this, this review aims to elucidate potential biomarkers of PFAS exposure by presenting a comprehensive overview of recent metabolomics-based studies exploring PFAS toxicity. Details of PFAS types, sources, and human exposure patterns are provided. Furthermore, insights into PFAS-induced liver toxicity, reproductive and developmental toxicity, cardiovascular toxicity, glucose homeostasis disruption, kidney toxicity, and carcinogenesis are synthesized. Additionally, a thorough examination of studies utilizing metabolomics to delineate PFAS exposure and toxicity biomarkers across blood, liver, and urine specimens is presented. This review endeavors to advance our understanding of PFAS biomarkers regarding exposure and associated toxicological effects.

Abstract:In this work, we demonstrate that a nonlethal dose of arsenite administered to quiescent C3H10T1/2 fibroblasts can enhance the mitogenic effect of suboptimal concentrations of serum. The mitogenic effect was dependent on the serum concentration and on the time interval between the administration of arsenite and that of serum. This suggests that mitogen sensitivity changes in time after arsenite treatment. It is shown that the concentrations of arsenite that enhance the mitogenic effect of serum also increase the mRNA levels of c‐fos, HSP68, and HSP84 and induce the specific synthesis of Heat Shock Proteins (HSPs). The physiological significance of this phenomenon is most likely to counteract the long‐term toxic effect of arsenite by early induction of compensation for cell loss. © 1993 Wiley‐Liss, Inc.

Municipal wastewater treatment plants (WWTPs) provide important conduits between waste sources and the environment. We present a meta-analysis of results reported in 44 peer-reviewed publications that include 460 influent and 528 effluent samples, collected from 21 countries, for which some or all of five perfluorinated carboxylic acids (PFCAs) and three perfluorinated sulfonic acids (PFSAs) were measured. Our meta analysis revealed global patterns and trends that, to our knowledge, have not been reported elsewhere. Regression analyses of samples collected from 2004 to 2020 quantified the temporal trends of global wastewater effluent concentrations of each of the PFAS and the corresponding mean concentration for each country. Although legacy compounds, perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS), have been reported with the highest measured concentrations, their global temporal trends are lowest of all PFAS considered. Concentrations of most PFAS analyzed in wastewater in the United States have not changed significantly with time, whereas reported PFAS concentrations in wastewater effluent from China have increased from 11% to 37% per year. In addition, our results show significant positive correlations between previous wastewater effluent concentrations of individual PFAS and the gross domestic product per capita of each country. Our analysis of this global data set also confirmed conclusions from previous studies on smaller data sets: (i) none of the PFAS studied are effectively removed by conventional treatment processes; (ii) effluents from treatment plants that include a significant industrial component to their influent tend to have higher PFAS concentrations; and (iii) the few studies that measured both aqueous concentrations and concentrations adsorbed to suspended particulate matter (SPM) indicate that PFAS adsorbed to SPM can contribute significantly to the total PFAS load.

Abstract:Abstract Background: Per- and polyfluoroalkyl substances (PFAS) are environmentally persistent synthetic chemicals detectable in the blood of most Americans. Elevated exposures to perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA), the two most prevalent PFAS, have been associated with several health outcomes including metabolic outcomes and some cancers. To evaluate possible mechanisms related to the biological effects of these chemicals, we conducted a pooled metabolome-wide association study (MWAS) of circulating PFOS and PFOA among 3,647 participants included in one of eight nested case-control serum metabolomic profiling studies conducted in the Prostate, Lung, Colorectal, and Ovarian Cancer Screening Trial (PLCO) population. Methods: All metabolomic profiling was conducted by Metabolon Inc, using liquid-phase or gas chromatography coupled with mass spectrometry. All metabolites including PFOS and PFOA were centered (mean=0, standard deviation=1) and log-transformed. We conducted multivariable linear regression analyses to estimate study-specific associations between metabolite levels and PFOS and PFOA, adjusted for age at specimen collection, race/ethnicity, body mass index, smoking, case-control status, study center, and year of specimen collection. Sex was additionally adjusted for studies including both men and women. We then conducted combined the study-specific findings through meta-analysis using random effects models. Results: The meta-analysis results of 1,040 metabolites for PFOS and 1,103 for PFOA identified 83 and 67 metabolites, respectively, associated at Bonferroni-corrected significance thresholds (P=4.81 × 10−5 for PFOS; P=4.53 × 10−5 for PFOA). For both PFOS and PFOA, the strongest positive associations were observed for the xenobiotics 3,5-dichloro-2,6-dihydroxybenzoic acid [PFOS, summary beta coefficient =0.74, 95% confidence interval=0.67,0.80; PFOA, 0.48 (0.35,0.61)] and 3-bromo-5-chloro-2,6-dihydroxybenzoic acid [0.57 (0.43,0.72) and 0.39 (0.23,0.55)]. We also observed strong positive associations with PFOS and PFOA for metabolites related to the sphingomyelin (SM) pathway, such as hydroxypalmitoyl sphingomyelin (d18:1/16:0(OH)) [PFOS, 0.44 (0.24,0.64)] and SM (d18:1/18:0) [PFOS, 0.35 (0.24,0.46); PFOA, 0.25 (0.16,0.34)], and SM (d18:2/24:2) [PFOS, 0.32 (0.17,0.47)]. The PFOS metabolite associations remained after model adjustment for PFOA; however, the PFOA associations were greatly attenuated after controlling for PFOS. Conclusions: In this MWAS of PFAS, to our knowledge the largest to date, we observed robust positive associations with serum PFOS for several xenobiotic compounds and for metabolites related to the sphingomyelin pathway. Abstract:Epidemiological evidence suggests that exposure to per- and polyfluoroalkyl substances (PFAS) is associated with lipid profile levels, but with inconsistent conclusions from different studies. The aim of this study was to conduct a meta-analysis of the relationship between PFAS exposure and lipid profile levels based on population-based epidemiological studies. Embase, PubMed, Ovid database, The Cochrane Library and Web of Science database were used to search appropriate studies (before September 6, 2022) on the correlation between PFAS exposure and lipid profile levels. β value, odd ratio (OR) and 95 % confidence intervals (CIs) were extracted from studies. In this study, we found that higher low-density lipoprotein (LDL) levels were associated with exposure to perfluoroundecanoic acid (PFUnDA) ( β value=0.13, 95 % CIs: 0.02, 0.24) and perfluorooctane sulfonic acid (PFOS) ( β value=0.13, 95 % CIs: 0.04, 0.21). PFOA, PFOS and PFNA exposure were significantly related to the higher levels of total cholesterol (TC) with the pooled effect estimates of 0.08 (95 % CI: 0.02, 0.14), 0.13 (95 % CI: 0.05, 0.21) and 0.14 (95 % CI: 0.08, 0.20) respectively. In sum, our results identified that PFOA, PFOS, PFNA and PFUnDA were the most important risk factors for abnormal levels of lipid profile, indicating that we should prevent cerebrovascular disease by reducing and controlling PFAS exposure.

Abstract:Per- and polyfluoroalkyl substances (PFAS) analysis has become crucial due to their presence in the environment, their persistence and potential health risks. These compounds are commonly used in food contact materials (FCM) as a coating to provide water and grease-repellent properties. One of the pathways for PFAS to enter the human body is either through direct consumption of contaminated food or indirectly through migration from FCM into food. The purpose of this study was to investigate where the initial contamination of paper FCM occurs. We analysed paper material consisting of fresh fibre and secondary materials, intended to produce food packaging for the presence of PFAS. The samples were extracted and analysed for 23 different PFAS substances using the targeted approach with LC tandem mass spectrometry (LC-MS/MS). This analytical technique detects specific, easily ionisable PFAS with high sensitivity. However, one drawback of this approach is that it allows the identification of less than 1% of the PFAS known today. For this reason, we used combustion ion chromatography (CIC) to determine the content of extractable organic fluorine compounds (EOF) and compare it to the total fluorine content. The targeted analysis using LC-MS/MS measured an average sum concentration of PFAS of 0.17 ng g −1 sample. Our research shows that the primary PFAS contamination happens during the recycling process since all of the samples in which the targeted PFAS were measured belonged to the secondary material. The most frequently detected analytes were PFOA and PFOS, detected in 90% and 62% of the samples, respectively, followed by PFBS (in 29% of the samples). CIC showed that measured PFAS via LC-MS/MS amount to an average of 2.7 × 10 −4 % of total fluorine content, whereas the EOF was under the LOD in all of the measured samples. This result highlights the complexity of the accurate determination of PFAS compounds, displaying what kind of information the chosen methods provide.

Abstract:OBJECTIVE: To summarize the application of non-targeted metabolomics in epidemiological studies that assessed metabolite and metabolic pathway alterations associated with per- and polyfluoroalkyl substances (PFAS) exposure.RECENT FINDINGS: Eleven human studies published before April 1st, 2021 were identified through database searches (PubMed, Dimensions, Web of Science Core Collection, Embase, Scopus), and citation chaining (Citationchaser). The sample sizes of these studies ranged from 40 to 965, involving children and adolescents (n = 3), non-pregnant adults (n = 5), or pregnant women (n = 3). High-resolution liquid chromatography-mass spectrometry was the primary analytical platform to measure both PFAS and metabolome. PFAS were measured in either plasma (n = 6) or serum (n = 5), while metabolomic profiles were assessed using plasma (n = 6), serum (n = 4), or urine (n = 1). Four types of PFAS (perfluorooctane sulfonate(n = 11), perfluorooctanoic acid (n = 10), perfluorohexane sulfonate (n = 9), perfluorononanoic acid (n = 5)) and PFAS mixtures (n = 7) were the most studied. We found that alterations to tryptophan metabolism and the urea cycle were most reported PFAS-associated metabolomic signatures. Numerous lipid metabolites were also suggested to be associated with PFAS exposure, especially key metabolites in glycerophospholipid metabolism which is critical for biological membrane functions, and fatty acids and carnitines which are relevant to the energy supply pathway of fatty acid oxidation. Other important metabolome changes reported included the tricarboxylic acid (TCA) cycle regarding energy generation, and purine and pyrimidine metabolism in cellular energy systems.CONCLUSIONS: There is growing interest in using non-targeted metabolomics to study the human physiological changes associated with PFAS exposure. Multiple PFAS were reported to be associated with alterations in amino acid and lipid metabolism, but these results are driven by one predominant type of pathway analysis thus require further confirmation. Standardizing research methods and reporting are recommended to facilitate result comparison. Future studies should consider potential differences in study methodology, use of prospective design, and influence from confounding bias and measurement errors.
