Abstract:In this letter, we demonstrate a spherical nanocavities fabrication using an "air-molding" method, which is implemented by modulating the pressure difference across air-liquid interfaces in nanoholes on the mold. The cavities formation is theoretically considered and experimentally verified at macroscale first, and then a series of experiments are performed over a patterned surface with sub-300 nm holes by varying the pressure difference by sending a PDMS prepolymer coated mold into a vacuum chamber with changeable pressure. Results show that the air-molding method for spherical cavities fabrication is feasible not only at macroscale, but also at the nanoscale when introducing a pressure difference across the air-liquid interface. And the cavities shape is easily controlled by modulating the pressure in the vacuum chamber. The spherical cavities in this paper have application potential in the optical field and in micro- and nanofluidics.

Abstract:Here, we report a novel method for the fabrication of polydimethylsiloxane microdevices with complicated 3-D structures, such as concave and crater shapes, using an easily machined polymethyl methacrylate mold combined with a one-step molding process. The procedure presented here enables rapid preparation of complex 3-D microstructures varying in shape and dimensions. To regulate embryoid body (EB) formation, we fabricated a microfluidic device with an array of concave microwells and found that EBs growing in microwells maintained their shape, viability, and a high degree of homogeneity. We believe that this novel method provides an alternative for rapid prototyping, especially in fabricating devices with curved 3-D microstructures.
Standing Surface Acoustic Wave-Assisted Fabrication of Region-Selective Microstructures Via User-Defined Waveguides.

Abstract:Polymer-based substrate with region-selective microstructures are crucial for many biomedical applications. Here, we explored a novel method based on standing surface acoustic waves (SSAWs) for the fabrication of localized polymer-based microstructures via a predefined waveguide. When the SSAWs are excited, the generated acoustic pressure field can be controlled in a predetermined region of the fluid surface through controlling the size and shape of the waveguide geometry. On the basis of the capillary wave motion, the generated acoustic pressure field can excite microwavy patterned structures on the surface. Then with use of ultraviolet (UV) solidification, the polymer-based substrates with region-selective patterned microstructures can be successfully fabricated. Both finite element modeling and experimental studies demonstrated that the polymer substrate with different region-selective microstructures can be achieved by selecting the pairs of interdigital transducers (IDTs) and shapes of the predefined waveguides. The results showed that the proposed method is effective for fabricating polymer-based substrate with region-selective microstructures and may have potential in cell-laden chips for tissue engineering, cell-cell interactions, and other biomedical applications.
Heterogeneous spheroids with tunable interior morphologies by droplet-based microfluidics

Abstract:Abstract Heterogeneous spheroids that mimic the complex three-dimensional environment of natural tissues are needed in various biomedical applications. Geometric cues from cellular matrix play invaluable roles in governing cell behavior and phenotype. However, the structural complexity of interior morphologies of spheroids is currently limited due to poor spatial resolution of positioning/orientation of cellular constructs. Here, a coaxial capillary microfluidic device is developed to generate gelatin methacrylate (GelMA) microspheres with tunable dimensions and interior morphologies, such as core-shell, or microspheres with interior undulated wavy, or spiral canals, by manipulating the two-phase flow of hydrogel precursor solution and methylcellulose solution. The formation of diverse and exquisite interior morphologies is caused by the interacting viscous instabilities of the two-phase flow in the microfluidic system, followed by water-in-oil emulsion and photo-initiated polymerization. Polyethylene glycol diacrylate (PEGDA) is incorporated into the GelMA solution to tune the mechanical properties of the fabricated microspheres, and an optimized concentration of PEGDA is confirmed by evaluating the in vitro proliferation and vascularization of human umbilical vein endothelial cells. Further, a heterogeneous spheroid with spiral blood vessel lumen is constructed to demonstrate the versatility and potential of the proposed droplet-based microfluidic approach for building functional tissue constructs.

Abstract:Microarrays of spherical vessel-like colloids such as liposomes, polymerized vesicles and polyelectrolyte capsules may find diverse applications in bioanalysis, biosensing, and combinatorial chemistry, for their capabilities in encapsulating chemical species such as drugs, biomolecules, probes, polymers and nanoparticles. This review reports the advances on methods for fabricating microarrays of the various hollow colloids. Related strategies are described in detail, including patterning techniques, surface modification methods, and tethering approaches such as oligonucleotide hybridization, receptor-ligand binding, covalent coupling and electrostatic interaction. The preliminary developments of functionalities of these arrays serving as sensor chips, microcarriers and microreactors are summarized as well.
Fabricating SU-8 Photoresist Microstructures with Controlled Convexity–Concavity and Curvature through Thermally Manipulating Capillary Action in Poly(dimethylsiloxane) Microholes

Abstract:We present a simple, robust, and cheap microfabrication method, based on thermally manipulating capillary action in poly(dimethylsiloxane) (PDMS) microholes, for preparing SU-8 curved microstructures. The microstructure morphology including convexity-concavity and curvature can be controlled via tuning the formation temperature. The convex SU-8 microspherical crowns with a height of 40 μm were formed at 10 °C, whereas the concave SU-8 microspherical crowns with a height of 90 μm were formed at 100 °C. The morphology of the microstructures is dictated by the thermally controlled combination of the pressure difference across the interface, contact angle, and surface tension. The fabricated microstructures with a spherical surface can be used as a microlens array or a mold for producing a microlens array. The clear and uniform images were observed using the generated microlens arrays. The equilibrium morphology of the microstructures can be predicted by numerical simulation, which can lessen the number of experiments and thus the design cost. The proposed method has the potential to find applications in industrial fields.

Abstract:We describe new developments for controlled fabrication of monodisperse non-spherical particles using droplet microfluidics. The high degree of control afforded by microfluidic technologies enables generation of single and multiple emulsion droplets. We show that these droplets can be transformed to non-spherical particles through further simple, spontaneous processing steps, including arrested coalescence, asymmetric polymer solidification, polymerization in microfluidic flow, and evaporation-driven clustering. These versatile and scalable microfluidic approaches can be used for producing large quantities of non-spherical particles that are monodisperse in both size and shape; these have great potential for commercial applications.

Abstract:We present an innovative solvent-free micromolding technique for rapidly fabricating complex polymer microparticles with three-dimensional (3D) shapes utilizing a surface tension-induced dipping process. Our fabrication process involves loading a photocurable solution into micromolds through mold dipping. The loaded solution, induced by surface tension, undergoes spatial deformation upon mold removal caused by surface forces, ultimately acquiring an anisotropic shape before photopolymerization. Results show that the amount of photocurable solution loaded depends on the degree of capillary penetration, which can be adjusted by varying the dipping time and mold height. It enables the production of polymer particles with precisely controlled 3D shapes without diluting them with volatile organic solvents. Sequential micromolding enables the spatial stacking of the polymer domain through a bottom-up approach, facilitating the creation of complex multicompartmental microparticles with independently controlled compartments. Finally, we demonstrated the successful simultaneous conjugation of multiple model-fluorescent proteins through the biofunctionalization of microparticles, indicating functional stability and effective conjugation of hydrophilic molecules such as proteins. We also extend our capacity to create bicompartmental microparticles with distinct functionalities in each compartment, revealing spatially controlled functional structures. In summary, these findings demonstrate a straightforward, rapid, and reliable method for producing highly uniform complex particles with precise control over the 3D shape and compartmentalization, all accomplished without the use of organic solvents.

Abstract:We present adaptive and robust techniques (ART) for thermoacoustic tomography with application in breast cancer detection. TAT combines high contrast and high resolution. The current image reconstruction methods used for TAT, such as the widely used Delay-and-Sum (DAS) approach, are data-independent and suffer from low resolution, high sidelobe levels, and poor interference rejection capabilities. The data- adaptive ART have much better performance than their data- independent counterparts. By allowing certain uncertainties, ART can mitigate the amplitude and phase distortion problems encountered in TAT. The excellent performance of ART is demonstrated using both simulated and experimentally measured data.

Abstract:In this paper, we present a focused femtosecond laser Bessel beam scanning technique for the rapid fabrication of large-area 3D complex microtube arrays. The femtosecond laser beam is converted into several Bessel beams by two-dimensional phase modulation using a spatial light modulator. By scanning the focused Bessel beam along a designed route, microtubes with variable size and flexible geometry are rapidly fabricated by two-photon polymerization. The fabrication time is reduced by two orders of magnitude in comparison with conventional point-to-point scanning. Moreover, we construct an effective microoperating system for single cell manipulation using microtube arrays, and demonstrate its use in the capture, transfer, and release of embryonic fibroblast mouse cells as well as human breast cancer cells. The new fabrication strategy provides a novel method for the rapid fabrication of functional devices using a flexibly tailored laser beam.

Abstract:We report a novel and facile method for fabricating coaxial microfluidic devices processing various dimensions at low cost, in which polypropylene hollow fibers or glass capillaries are used as the tip of the dispersed phase injection tube. With this coaxial microfluidic device, monodisperse biocompatible microspheres ranging from 300 to 800 μm were obtained by collecting oil-in-water or water-in-oil emulsions and solidifying the suspended microspheres. Microsphere size could be controlled by changing the tips or tuning the concentrations of the dispersed and continuous phases. By adding functional nanoparticles into the dispersed phase, it was demonstrated that fluorescent and magnetic microspheres can be fabricated easily using these microfluidic devices.

Abstract:Aiming at improving the precision and decreasing roughness of micro-manufacturing, this work proposes a new method that combines non-uniform graphic compensation for template preparation and demolding with viscous fluid composed of mixtures of two types of polydimethylsiloxane (PDMS), Sylgard 184 and SE 1700. Notably, this method can widely apply to a bounty of micro-manufacturing methods, such as laser direct writing (LDW), 3D printing, and lithography. This paper exploits LDW to fabricate smooth convex microdome array (MA) on PDMS, and demonstrates the applications of such microstructures in electronics and optics. Specifically, the multifunctional microstructure can serve as a pressure sensor with high sensitivity and broad range, a strain senor for biaxial strain sensing and an anti-reflection film to improve photoelectric conversion efficiency of solar cells.

Abstract:The formation of microarrays on the surface of flexible materials has great potential value in fabrication of functional devices such as various kinds of flexible electronic sensors. To fabricate microarrays on the surface of flexible materials, many methods have been proposed, such as nanoimprint lithography technique, single-pulse laser interference and colloidal self-assembly. In our study, we present a simple method to fabricate silica microsphere arrays embedded on the surface of polyurethane film in a single-step. We directly solidified self assembled microspheres at the air-liquid interface, and obtained polyurethane film embedded with silica microarrays on its surface. Compared with other methods, precise operation and complicated equipment are not required. In addition, we also utilized the fabricated microarrays as a template to fabricate concave microarrays on the surface of other flexible materials.

Abstract:In this paper we demonstrate a facile and efficient way to fabricate poly(dimethylsiloxane) (PDMS) molds with hexagonal non-close-packed (ncp) arrangements of microwells by casting PDMS prepolymer onto two-dimensional (2D) ncp colloidal crystals. The templates of the 2D ncp colloidal crystals were fabricated via coupling lift-up soft lithography and solvent-swelling. We found that the depths of the microwells together with the lattice spacing can be adjusted by the sphere interstices and chemical composition of the 2D ncp colloidal crystals. The relationship of the surface character of the templates with the depths of the microwells can be explained by the wetting behavior of PDMS prepolymer on the rough surface. Contact angle measurements are consistent with the experimental results of the microwells in depth and agree well with the Cassie-Baxter theory. There are at least three advantages of the approach. First, the depth and distance of the microwells can be controlled. Second, PDMS molds can be easily peeled from the surfaces of the templates, which results in reusing the original templates to make new molds. Third, this method can be applied to other materials, such as photopolymerizable resin or thermosetting resin. The potential application of the microwells is as microlenses to make a pattern or as microvials in bioanalytical techniques.

Abstract:Colloidal particles fabricated using lithographic methods are used in micro-nanoengineering as well as biomechanical and chemical engineering. Much of the research in this field deals with close-packed colloidal particles in the form of continuous two-dimensional (2D) surface structures or membranes. The most common approach to modifying the arrangement and spacing of colloidal particles involves etching or the fabrication of micro-nanoimprinted structures at the micro- or nanoscale. In the current study, three-dimensional (3D) micro-printing was used to fabricate grid and honeycomb structures with precise control over the spatial distribution and height. We achieved a uniform distribution of polystyrene micro spheres across the surface of the structures by performing a variation of the floating assembly method referred to as drop deposition, which when implemented using methanol was shown to enhance the dispersal of microspheres in the mixture by reducing the London diverse force (LDF). The application of ultrasonic vibrations during microsphere deposition was shown to facilitate the integration of PS microspheres within the underlying lattice. We also found that methanol is highly effective in the removal of accumulated microspheres. The fabrication of grid and hexagonal structures spaced at intervals of 6, 6.5, and 7 μm followed by the deposition of PS microspheres (diameter = 6 μm) was shown to increase the water droplet contact angle from 103° (close-packed) to 110° (square arrangement) and 123° (hexagonal arrangement).

Abstract:In this paper, we proposed an efficient and high-precision process for fabricating large-area microlens arrays using thermal reflow combined with ICP etching. When the temperature rises above the glass transition temperature, the polymer cylinder will reflow into a smooth hemisphere due to the surface tension effect. The dimensional differences generated after reflow can be corrected using etching selectivity in the following ICP etching process, which transfers the microstructure on the photoresist to the substrate. The volume variation before and after reflow, as well as the effect of etching selectivity using process parameters, such as RF power and gas flow, were explored. Due to the surface tension effect and the simultaneous molding of all microlens units, machining a 3.84 × 3.84 mm2 silicon microlens array required only 3 min of reflow and 15 min of ICP etching with an extremely low average surface roughness Sa of 1.2 nm.

Abstract:BACKGROUND Endothelial progenitor cell (EPC) capture stent is designed to promote rapid endothelization and healing and is potentially useful in patients undergoing primary percutaneous coronary intervention (PCI) for acute myocardial infarction (AMI). We studied the intermediate-term efficacy and safety of EPC stent and compared that with sirolimus-eluting bioabsorbable polymer stent (CURA) and bare metal stent (BMS) in AMI patients. METHODOLOGY Patients presenting with AMI who underwent primary PCI with the respective stents between January 2004 and June 2006 were enrolled in the single-center clinical registry. The study end-points were major adverse cardiac events (MACE) and stent thrombosis. RESULTS A total of 366 patients (EPC = 95, CURA = 53, BMS 218) were enrolled. Baseline demographics including age, gender, diabetes, renal impairment, predischarge left ventricular ejection fraction, and creatinine kinase level were comparable among the groups. Procedural success rate was 99.5%. Post-procedural thrombolysis in myocardial infarction (TIMI) 3 flow was achieved in EPC 91.6%, CURA 96.2%, and BMS 88.5% (P = 0.209). At 2 years, the MACE rate was EPC 13.7%, CURA 15.1%, and BMS 19.7% (P = 0.383). Target vessel revascularizations (TVR) were EPC 4.2%, CURA 9.4%, and BMS 6.0% (P = 0.439). Nonfatal myocardial infarctions were EPC 1.1%, CURA 3.8%, and BMS 4.1% (P = 0.364). One patient in the EPC group had acute stent thrombosis. There was no late stent thrombosis in the EPC group. CONCLUSION EPC stent appeared to be safe and had comparable clinical efficacy with a BMS when used in the AMI setting. At 2-year follow-up, the EPC group showed favorable, single-digit TVR rate and stent thrombosis remained a low-event occurrence.

Abstract:Micellar nanocrystals (micelles with encapsulated nanocrystals) have become an emerging major class of nanobiomaterials. We describe a method of fabricating micellar nanocrystals based on combining top-down electrospray, bottom-up self-assembly, and solvent-based structure control. This method involves first using electrospray to generate uniform ultrafine liquid droplets, each of which functions as a micro-reactor in which self-assembly reaction occurs forming micellar nanocrystals, with the structures (micelle shape and nanocrystal encapsulation) controlled by the organic solvent used. This method is largely continuous and produces high quality micellar nanocrystal products with an inexpensive structure control approach. By using a water-miscible organic solvent tetrahydrofuran (THF), worm-shaped micellar nanocrystals can be produced due to solvent-induced/facilitated micelle fusion. Compared with the common spherical micellar nanocrystals, worm-shaped micellar nanocrystals can offer minimized non-specific cellular uptake, thus enhancing biological targeting. By co-encapsulating multiple nanocrystals into each micelle, multifunctional or synergistic effects can be achieved. Current limitations of this fabrication method, which will be part of the future work, primarily include imperfect encapsulation in the micellar nanocrystal product and the incompletely continuous nature of the process.

Abstract:Concave microstructures such as microwells and microgrooves are widely utilized in fields such as biochips, microfluidics, and functional devices. Previously, concave microstructure fabrication was mostly based on laser etching or lithography which is either costly or of multisteps. The inkjet etching method is a direct structuring technique, but limited by its inherent transverse ink diffusion that leads to low feature resolution. Nanoimprint lithography can reach submicro and even nano ranges, whereas an elaborate template is needed. Thus, it is still a challenge to realize controllable fabrication of concave microstructures in large areas with high efficiency and resolution. Here, a template‐free strategy to fabricate concave microstructures with high resolution by inkjet imprinting is provided. In this method, a sacrificial ink is inkjet‐printed onto a precured viscoelastic surface and imprints its shapes to construct concave microstructures. The morphology of the microstructures could be adjusted by controlling the interaction between the two immiscible phases. The microwells/microgrooves could be used to pattern single cells and functional materials such as optical, electronic, and magnetic nanoparticles. These results will open a new pathway to fabricate concave microstructures and broaden their applications in various functional devices.
Functional Shape-Morphing Microarchitectures Fabricated by Dynamic Holographically Shifted Femtosecond Multifoci

Abstract:Functional microdevices based on responsive hydrogel show great promise in targeted delivery and biomedical analysis. Among state-of-the-art techniques for manufacturing hydrogel-based microarchitectures, femtosecond laser two-photon polymerization distinguishes itself by high designability and precision, but the point-by-point writing scheme requires mechanical apparatuses to support focus scanning. In this work, by predesigning holograms combined with lens phase modulation, multiple femtosecond laser spots are holographically generated and shifted for prototyping of three-dimensional shape-morphing structures without any moving equipment in the construction process. The microcage array is rapidly fabricated for high-performance target capturing enabled by switching environmental pH. Moreover, the built scaffolds can serve as arrayed analytical platforms for observing cell behaviors in normal or changeable living spaces or revealing the anticancer effects of loaded drugs. The proposed approach opens a new path for facile and flexible manufacturing of hydrogel-based functional microstructures with great versatility in micro-object manipulation.
