Abstract:A rapid and novel two-step dispersive liquid-liquid microextraction and dispersive micro-solid phase extraction method was established for the separation and enrichment of trace cadmium, nickel, and copper in food and environmental water prior to determination by inductively coupled plasma-mass spectrometry (ICP-MS). In the first microextraction step, carbon tetrachloride was employed to extract metal-diethyldithiocarbamate chelates from aqueous solution with ultrasound. In the following step, Fe3O4 magnetic nanoparticles were added and used to collect the analytes in the organic solvent. The sample pH, type and volume of extraction solvent, mass of magnetic nanoparticles, concentration of the chelating agent, concentration of sodium chloride, and sonication time were optimized. The linear dynamic range was from 0.01 to 20 micrograms per liter with correlation coefficients between 0.9990 and 0.9992. Enrichment factors were 78, 79, and 81 for cadmium, nickel, and copper, respectively. The limits of detection for cadmium, nickel, and copper were 0.007, 0.009, and 0.017 micrograms per liter, with relative standard deviations from 1.1 to 2.6 percent. The developed method was validated by the determination of cadmium, nickel, and copper in rice and mussel certified reference materials, food, and environmental water with satisfactory results.

Abstract:Plants can absorb and accumulate engineered nanomaterials (ENMs) through water and soil, providing a potential way for nanoparticles to be enriched in humans through the food chain. In this paper, a combination of enzymatic digestion method and SP-ICP-MS analysis was used to quantitatively characterize the enriched AuNPs in mustard and lettuce plants. The results showed that Macerozyme R-10 enzyme can extract AuNPs from plants without obvious aggregation/dissolution. Both mustard and lettuce plants can absorb and enrich the complete AuNPs to the above-ground organs, and the particle number concentrations detected are 1.24 × 107 particles L−1 and 4.39 × 107 particles L−1, respectively. With different exposure level of AuNPs(0.5 mg L−1,), a particle number concentration of 2.32 × 107 particles L−1 was detected in the stems of lettuce plants, while the mustard failed to transport AuNPs to the above-ground organs. The transport efficiency of Au ions by plants is higher than that of AuNPs, and the plants have stronger bioavailability for ions.

Abstract:The increasing use of nanopesticides has raised concerns about their effects on crop plants and the impact of human health as well as ecological effects. While increased uptake of metal ions has been observed before, to date, very few studies have demonstrated the presence of nanoparticles in edible tissues. Single-particle inductively coupled plasma–mass spectrometry (sp-ICP-MS) has been suggested as a powerful tool to detect inorganic nanoparticles (NPs) in environmental samples. Here, we exposed edible plant tissues from lettuce, kale, and collard green to nano-CuO, simulating its use as a nanopesticide. We applied sp-ICP-MS to demonstrate the presence of nanoparticles, both in the water used to rinse crop leaf surfaces exposed to nano-CuO and within the leaf tissues. Lettuces retained the highest amounts of nCuO NPs on the leaf surface, followed by collard green and then kale. Surface hydrophilicity and roughness of the leaf surfaces played an important role in retaining nano-CuO. The results indicate that most of the nanoparticles are removed via washing, but that a certain fraction is taken up by the leaves and can result in human exposure, albeit at low levels.

Abstract:Due to the enormous interest in plants related to bioscience, environmental and toxicological research, analytical methods are expected with the ability of getting information on elemental transfer, distribution and contents in plants. In this work, a mixture of gelatin (GA) and hydroxypropyl methyl cellulose (HPMC) was prepared to simulate plant matrix, a method based on laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) with matrix-matching external calibration was proposed for direct quantification of multiple elements in plants. The pH and composition of GA&HPMC substrate was optimized, such as the concentration of spiked nitric acid, the mass fraction of both GA and HPMC in the substrate and the mass ratio of GA: HPMC. After spiking elemental solution, coating the mixture onto a glass slide and drying overnight at room temperature, GA&HPMC substrate was obtained. The substrate obtained with GA: HPMC of 8: 2 was used to fabricate the standard series, which exhibited good elemental homogeneity and similar elemental signal intensities in LA-ICP-MS detection to that obtained for plant Certified Reference Material (CRM). CRMs of different plants including Citrus leaf (GBW10019), Tea (GBW07605), Beans (GBW10021) and Scallions (GBW10049) were further pressed into pellets and subjected to the proposed method, and the quantification accuracy was demonstrated. The limits of detections of this method were found to be 0.003 (Ce)-104 (Ca) μg g −1 , with a wide linear range (0.01–10000 μg g −1 ) for 17 target elements. The application potential of the method was further demonstrated by performing elemental imaging in Trigonotis peduncularis leaves. Rapid in-situ quantitative imaging of Zn, Cu, Sr and Mn was achieved, and the elemental quantitative distributions were discussed. The constructed substrate helps direct elemental quantification in plants. It provided a powerful and efficient tool for the investigation of the distribution and transfer of elements in plants, favoring further exploration of elemental bioavailability, transport and toxicity mechanisms.

Abstract:BACKGROUND The transformation of nanoparticles (NPs) internalized in plant tissues is the human digestive system that can provide a better understanding of the impact of NPs on the human system. The presented methodology was developed to study the bioaccessibility of cerium oxide (CeO2) and copper oxide (CuO) NPs from radish after thein vitrosimulation of gastrointestinal digestion using single-particle inductively coupled plasma mass spectrometry (SP-ICP-MS). RESULTS Radish plants were cultivated hydroponically in a growth medium containing: (i) CeO(2)NPs and (ii) CuO NPs. Both cerium (Ce) and copper (Cu) were found in all organs of the radish plants after analysis by standalone ICP-MS. This confirms the bioaccumulation of CeO(2)and CuO NPs and the translocation of their Ce and Cu to the aerial parts of the plant. Less Ce (4.095 mu g g(-1)) has been detected in leaves than in roots (1.156 mg g(-1)) while Cu content in leaves was 5.245 mu g g(-1)and in roots was 10.41 mu g g(-1). Analysis of the digestive extracts obtained after thein vitrosimulation of gastro (pepsin) and gastrointestinal (pancreatin) digestion showed that Ce has easy access to human system at least by 73%. CONCLUSION The size of CeO(2)NPs in digestive extracts showed no significant changes. However, the results obtained for CuO NPs digestion were variable and suggested that CuO NPs dissolved during the digestion process. Abstract:A one-step ultrasound/microwave-assisted solid-liquid-solid dispersive extraction procedure was used for the simultaneous determination of eight neonicotinoids (dinotefuran, nitenpyram, thiamethoxam, clothianidin, imidacloprid, acetamiprid, thiacloprid, imidaclothiz) in dried Dendrobium officinale by liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry in multiple reaction monitoring mode. The samples were quickly extracted by acetonitrile and cleaned up by the mixed dispersing sorbents including primary secondary amine, C-18, and carbon-GCB. Parameters that could influence the ultrasound/microwave-assisted extraction efficiency such as microwave irradiation power, ultrasound irradiation power, temperature, and solvent were investigated. Recovery studies were performing well (70.4-113.7%) at three examined spiking levels (10, 50, and 100 mu g/kg). Meanwhile, the limits of quantification for the neonicotinoids ranged from 0.87 to 1.92 mu g/kg. The method showed good linearity in the concentration range of 1-100 mu g/L with correlation coefficients >0.99. This quick and useful analytical method could provide a basis for monitoring neonicotinoid insecticide residues in herbs.
Evaluation of extraction and storage conditions for quantification and characterization of silver nanoparticles in complex samples by single particle-ICP-MS

Abstract:The extraction of nanoparticles (NPs) from complex matrices and subsequent storage can potentially alter the NPs physicochemical properties and hinder cross-study comparisons. Most NPs extraction methods are designed and tested at high NPs concentrations, although (eco)toxicological and regulatory monitoring programs require methods capable of analyzing NPs at environmentally relevant concentrations (lower ppb range). In this study, we investigated how extraction methods affect the characteristics of PVP coated and citrate-stabilized silver NPs (AgNPs) spiked into soil, sewage sludge, and biological samples at environmentally relevant concentrations using Single Particle Inductively Coupled Plasma Mass Spectrometry spICP-MS). Further we investigated the impact of storage temperature (-80 °C - 21 °C) and storage duration (1-28 days) on the particle characteristics such as particle size. We found that aqueous AgNPs samples with low ionic strength media retained their original characteristics (like particle size, particle concentration and particle-based Ag mass) when preserved at 4 °C for up to 28 days. AgNPs dispersed in high ionic strength media were however better preserved at -80 °C. Among the extraction agents, tetrasodium pyrophosphate was efficient in extracting AgNPs from soil and sewage sludge matrices, while Proteinase K was most suitable for biological samples from organisms (earthworms or fish). Although our study focused only on AgNPs, it provides crucial information to aid interlaboratory comparisons and data interpretation for (eco)toxicological studies.
High-efficiency mechanically assisted alkaline extraction of nanoparticles from biological tissues for spICP-MS analysis

Abstract:AbstractThe widespread use and increased exposure of nanoparticles call for technology to quantify their concentration and size distribution in biological matrices. As ex situ evaluation, facile extraction with high fidelity and efficiency is critical. In this work, single particle inductively coupled plasma mass spectrometry (spICP-MS) was used for nanoparticle number and distribution analysis, where a facile and highly efficient mechanically assisted alkaline digestion has been developed to extract nanoparticles at low alkali concentration. The optimization was performed using chicken tissues in vitro mixed with 30 nm gold nanoparticles, mixture of 30 nm and 60 nm gold nanoparticles, and 45 nm silver nanoparticles, respectively, which is, then, mechanically ground to form tissue homogenate and 2% TMAH is added. The nanoparticles are extracted with a recovery of more than 94% for all the spiked nanoparticle tissue samples. The extraction method has also been attempted to be applied to extract single-sized gold nanoparticles from various organs of mice mixed in vivo with the nanoparticles through intravenous injection, and led to consistent results with acid digestion. Mice injected intravenously with double-sized gold nanoparticle mixture were also studied, further showing that gold nanoparticles of 30 nm and 60 nm have no significant difference in their biodistribution in the same organ. To the best of our knowledge, this is the first attempt for multiple nanoparticles being extracted simultaneously and measured quantitatively from various organs, such as the heart, liver, spleen, lungs, and kidneys. We believe this method is beneficial to the safety assessment and toxicokinetics studies for nanoparticles in tissues.Graphical abstract

Abstract:Metal speciation analysis of microsamples in the clinical, biological and forensic fields is important for elucidating the metals' toxicity, mobility and metabolic behaviors of metals. Such analysis may be achieved by nanoliter high-performance liquid chromatography (nanoHPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). In this work, an in-column high-pressure nebulizer (ICHPN) was developed to enhance the sensitivity and the separation efficiency of the sheathless nanoHPLC-ICP-MS coupling. The ICHPN consists of two concentric fused-silica capillaries with tapered tips connected via a PEEK tee, where C-18 silica particles with a size of 5 mu m were packed in the tapered inner capillary based on the keystone effect. Combining a heated single pass spray chamber with a makeup gas, the ICHPN was capable of independently optimizing the nebulization efficiency and the transport efficiency. The ICHPN offered high sensitivities, low detection limits and good precision at nanoflow rates. Compared with commercial nebulizers, the ICHPN fabrication was simple, rapid, reproducible and inexpensive. By implementing the ICHPN, we achieved rapid separation of four mercury species, i.e., Hg2+ and methyl-, ethyl- and phenylmercuric chloride, within 8.0 min with good resolution (2.0-13.9). Detection limits of 0.044-0.13 mu g L-1 were obtained with precisions of peak heights and areas ranging from 1.5 to 3.5% for a 50 mu g L-1 standard solution. Good agreement between the determined and certified values of mer- cury species in a certified reference material of caprine blood (SRM 955c) together with good recoveries (93-101%) validated the accuracy of the method. Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.

Abstract:Nanoplastics (NPs) are invisible to human eyes yet pose significant concerns to human health due to their wide environmental presence and high potential for biological uptake, transport, and accumulation. Conventional analytical methods suffer from low accuracy and precision in NP detection due to their limited reliability and quantitative ability. To address these challenges, we developed a dual-functional model NP that allows for in-situ imaging by surface-enhanced Raman spectroscopy (SERS) and ex-situ quantification by inductively coupled plasma-mass spectrometry (ICP-MS). In this study, the model NP has a core-shell structure with Raman reporter-functionalized gold nanoparticles as the core and a layer of plastic as the shell. The gold core can enhance the Raman reporter signals and make model PS detectable, which can be used to visualize the uptake of the model NP in plant tissue by SERS. Meanwhile, the model NP particle numbers in the collected plants can be quantified by ICP-MS based on the presence of gold in the core. The model PS demonstrated stability in structure, size, and surface charges over one year, with no indication of chemical leaching. In this study, garlic plants were used as our experimental matrix to evaluate the potential of the dual-functional model PS for application in living organisms. Our aim was to determine whether the model PS in the garlic plants could be effectively quantified and qualified by SERS and ICP-MS. Garlic plants were grown in various concentrations of model NP suspensions for a 30-day period. The results showed that the NP uptake was concentration-dependent with higher concentrations of model NP leading to higher uptake by the garlic roots. The study also investigated the changes in NP uptake over time, showing that longer NP exposure resulted in more NP uptake in garlic roots. The study also demonstrated the effective coordination between SERS and ICP-MS. In cases where SERS had limitations in detecting the presence of model PS, ICP-MS proved capable of facilitating detection in garlic tissue. This study demonstrates the potential use of this dual-functional model NP for studying NP behavior with SERS and ICP-MS in living organisms, which holds significant implications for better understanding their impact on crops for future studies.

Abstract:Current sample preparation strategies for nanomaterials (NMs) analysis in soils by means single particle inductively coupled plasma mass spectrometry have significant constrains in terms of accuracy, sample throughput and applicability (i.e., type of NMs and soils). In this work, strengths and weakness of microwave assisted extraction (MAE) for NMs characterization in soils were systematically investigated. To this end, different extractants were tested (ultrapure water; NaOH, NH 4 OH, sodium citrate and tetrasodium pyrophosphate) and MAE operating conditions were optimized by means of design of experiments. Next, the developed method was applied to different type of metallic(oid) nanoparticles (Se-, Ag-, Pt- and AuNPs) and soils (alkaline, acid, sandy, clayey, SL36, loam ERMCC141; sludge amended ERM483). Results show that Pt- and AuNPs are preserved and quantitatively extracted from soils in 6 min (12 cycles of 30 s each) inside an 800 W oven by using 20 mL of 0.1 M NaOH solution. This methodology is applicable to soils showing a wide range of physicochemical properties except for clay rich samples. If clay soil fraction is significant (>15%), NMs are efficiently retained in the soil thus giving rise to poor recoveries (<10%). The analysis of labile NMs such as Se- and AgNPs is not feasible by means this approach since extraction conditions favors dissolution. Finally, when compared to current extraction methodologies (e.g., ultrasound, cloud point extraction, etc.), MAE affords better or equivalent accuracies and precision as well as higher sample throughput due to treatment speed and the possibility to work with several samples simultaneously.

Abstract:The wide application of nano-particles (NPs) has raised a serious concern over their impact on plants. However, evaluation of the effects of NPs on plant metabolism by direct detection of chemicals inside solid tissues presents a challenge. In this study, we report on a direct ionization method in mass spectrometry, internal extractive electrospray ionization (iEESI), for the direct evaluation of phytotoxicity of three different NPs (including CdTe quantum dots (CdTe QDs), gold nano-particles (Au NPs), and silver nano-particles (Ag NPs)) both on surfaces and inside solid tissues from the mung bean seeds (Vigna radiata) that were cultured in aqueous solutions of three NPs at 50 μg mL-1. The results showed that NPs could stimulate the biological accumulation of trigonelline and the decomposition of polysaccharides/oligosaccharides to glucose and maltose within 21 h of culture. To the best of our knowledge, this is the first study to apply internal extractive electrospray ionization mass spectrometry (iEESI-MS) for the direct measurement of solid tissue samples to evaluate the phytotoxicity of NPs on mung bean sprouts. Our study lays a solid foundation for further examination of other NPs-induced damaging effects such as apoptosis/necrosis, helping us to understand the phytotoxicity of NPs on plants.

Single particle inductively coupled plasma-mass spectrometry (sp-ICP-MS) is an attractive approach that can perform quantitative and qualitative analyses of ENPs. However, the application of this approach for biological samples is limited because of the lack of pretreatment methods for effectively recovering ENPs from biological tissues. In this study, we evaluated various pretreatment methods and identified the optimal pretreatment conditions for sp-ICP-MS analyses of ENPs in biological tissues using silver nanoparticles (nAg) as a model. We screened five reagents as pretreatment solvents (sodium hydroxide, tetramethylammonium hydroxide, nitric acid, hydrochloric acid, and proteinase K). Our results showed that treatment with sodium hydroxide was optimal for detecting nAg in the mouse liver. Moreover, this pretreatment method can be applied to other organs, such as the heart, lung, spleen, and kidney. Finally, we evaluated the applicability of this method by analyzing the quantity and physical properties of silver in the mouse blood and liver, after intravenous administration of nAg or silver ion. Our sp-ICP-MS method revealed that nAg administered into the blood was partially ionized and tended to be distributed in the particle form (approximately 80%) in the liver and in ionic form (approximately 95%) in the blood. In conclusion, we optimized pretreatment strategies for sp-ICP-MS evaluation of ENPs in biological tissues and demonstrated its applicability by evaluating the changes in the physical properties of nAg in the liver and blood. We also showed that partial changes from the particle form to the ionic form of nAg influences their kinetics and distribution when administered to mice.
Identification and Speciation of Nanoscale Silver in Complex Solid Matrices by Sequential Extraction Coupled with Inductively Coupled Plasma Optical Emission Spectrometry

Abstract:Nanoscale silver (n-Ag) including silver nanoparticles (Ag-NPs), silver chloride nanoparticles (AgCl-NPs), and silver sulfide nanoparticles (Ag2S-NPs) and their corresponding ionic counterpart, namely, dissolved Ag, may coexist in soils. X-ray absorption near edge spectroscopy (XANES) is used to elucidate the speciation of n-Ag in soils, whereas it possesses drawbacks like high costs, rare availability of the instrument, and providing semiquantitative data. We developed a new method for the identification and speciation of n-Ag in soils and sediments based on a sequential extraction technique coupled with inductively coupled plasma optical emission spectrometry. Extraction conditions were first evaluated, establishing the optimal extraction procedure; Ag-NPs, AgCl-NPs, and dissolved Ag in soil were simultaneously extracted by using an aqueous solution of 10 mM tetrasodium pyrophosphate, followed by selective isolation and quantification via AgCl-NPs dissolution (4.45 M aqueous ammonia), centrifugation (Ag-NPs), and detection. The Ag2S-NPs remaining in the soil were then extracted with Na2S solution at pH 7.0 through selective complexation. Optimal recoveries of Ag-NPs, AgCl-NPs, Ag2S-NPs, and dissolved Ag were 99.1 ± 2.4%, 112.0 ± 3.4%, 96.4 ± 4.0%, and 112.2 ± 4.1%, respectively. The method was validated to investigate the speciation of n-Ag in soils and sediments, exhibiting the distribution of Ag-NPs, AgCl-NPs, Ag2S-NPs, and dissolved Ag in each sample, wherein Ag2S-NPs, the major species of n-Ag, accounted for 35.42-68.87% of the total Ag. The results of n-Ag speciation in soil are comparable to those obtained through the linear combination fitting of XANES. This method thus is a powerful, yet convenient, substitute for XANES to understand the speciation of n-Ag in complex solid matrices.

Abstract:Quantitatively monitoring the presence of engineered, natural, or incidental nanoparticles (NPs) is essential to understand their potential environmental and ecotoxicological implications. In particular, a significant number of NPs may travel through wastewater treatment plants (WWTPs), as a conduit to their release to the environment, either in treated effluent or in wastewater sludge (biosolids). Here we developed a fast and simple protocol for full quantitative multielement analysis of metallic or metal-containing NPs in wastewater and sludge samples via single-particle ICP-MS. We employed centrifugation to separate NPs from wastewater sludge, with high recoveries (>84%) for Au and Ag NPs, which have rather high densities (19.3 and 10.5 g cm(-3), respectively). In wastewater samples, particle mass concentrations ranged from <1 ng/L for Cd-based NPs to almost 100 mu g/L for Mg particles. Particles from most elements detected in wastewater were <100 nm in size, although TiO2 in raw wastewater was around 250 nm in size, and Mg was >1500 nm in size, well beyond the nanoscale. The efficiency of removal of NPs throughout the WWTP was significantly dependent on the type of metal-containing particles and the influent concentration.
Extraction of Silicon-Containing Nanoparticles from an Agricultural Soil for Analysis by Single Particle Sector Field and Time-of-Flight Inductively Coupled Plasma Mass Spectrometry

Abstract:The increased use of silica and silicon-containing nanoparticles (Si-NP) in agricultural applications has stimulated interest in determining their potential migration in the environment and their uptake by living organisms. Understanding the fate and behavior of Si-NPs will require their accurate analysis and characterization in very complex environmental matrices. In this study, we investigated Si-NP analysis in soil using single-particle ICP-MS. A magnetic sector instrument was operated at medium resolution to overcome the impact of polyatomic interferences (e.g., 14N14N and 12C16O) on 28Si determinations. Consequently, a size detection limit of 29 ± 3 nm (diameter of spherical SiO2 NP) was achieved in Milli-Q water. Si-NP were extracted from agricultural soil using several extractants, including Ca(NO3)2, Mg(NO3)2, BaCl2, NaNO3, Na4P2O7, fulvic acid (FA) and Na2H2EDTA. The best extraction efficiency was found for Na4P2O7, for which the size distribution of Si-NP in the leachates was well preserved for at least one month. On the other hand, Ca(NO3)2, Mg(NO3)2 and BaCl2 were relatively less effective and generally led to particle agglomeration. A time-of-flight ICP-MS was also used to examine the nature of the extracted Si-NP on a single-particle basis. Aluminosilicates accounted for the greatest number of extracted NP (~46%), followed by NP where Si was the only detected metal (presumably SiO2, ~30%).

Abstract:led some experts to recommend that patients starting a weight loss program be provided a choice between empirically supported dietary approaches. For example, a Mediterranean diet tends to be one high in vegetables and fruits and low in fat diet, whereas a diet high in protein, such as the South Beach Diet, is consistent with a low-carbohydrate diet, yet both may be effective. Other types of diet have also been investigated relative to their ability to contribute to weight loss. Vegetarian diets have been found to be effective for weight management, but effects could be deleterious when energy restriction is included (Farmer, 2014). In their systematic review, Mytton, Nnoaham, Eyles, Scarborough, and Ni Mhurchu (2014) found that increased consumption of fruits and vegetables without decreasing intake of other foods did not contribute to weight gain and, in some instances, could be infl uential in weight maintenance. A randomized control trial comparing weight loss outcomes of the Mediterranean diet (Med diet) with MyPyramid in a sample of postpartum breastfeeding women found both diets resulted in weight loss during the postpartum period with weight loss change ranging from -2.31 ± 3.42 (p < 0.001) for participants in the Med diet group to -3.11 ± 3.35 (p < .001) for those in the MyPyramid group (StendellHollis, Thompson, West, Wertheim, & Thomson, 2013). Abstract:Laser ablation–inductively coupled plasma–mass spectrometry (LA–ICP–MS) is an emerging method for the analysis of metal nanoparticles (NPs) in single cells. However, two main obstacles, low analytical throughput and lack of commercial reference materials, need to be overcome. In this work, we demonstrated the principles of a new approach termed "single-cell isotope dilution analysis" (SCIDA) to remove the two obstacles. For a proof of concept, macrophage cells were chosen as a model to study the uptake of silver NPs (AgNPs) at a single-cell level. Single cells exposed to AgNPs were placed in an array by a microfluidic technique; each cell in the array was precisely dispensed with a known picoliter droplet of an enriched isotope solution with a commercial inkjet printer; accurate quantification of AgNPs in single cells was done by using isotope dilution LA–ICP–MS. The average Ag mass of 1100 single cells, 396 ± 219 fg Ag per cell, was in good accord with the average of the population of cells determined by solution ICP–MS analysis. The detection limit was 0.2 fg Ag per cell.
