Approximately 740 million symptomatic patients are affected by otitis media every year. Being an inflammatory disease affecting the middle ear, it is one of the primary causes of tympanic membrane (TM) perforations, often resulting in impaired hearing abilities. Antibiotic therapy using broad-spectrum fluoroquinolones, such as ciprofloxacin (CIP), is frequently employed and considered the optimal route to treat otitis media. However, patients often get exposed to high dosages to compensate for the low drug concentration reaching the affected site. Therefore, this study aims to integrate tissue engineering with drug delivery strategies to create biomimetic scaffolds promoting TM regeneration while facilitating a localized release of CIP. Distinct electrospinning (ES) modalities were designed in this regard either by blending CIP into the polymer ES solution or by incorporating nanoparticles-based co-ES/electrospraying. The combination of these modalities was investigated as well. A broad range of release kinetic profiles was achieved from the fabricated scaffolds, thereby offering a wide spectrum of antibiotic concentrations that could serve patients with diverse therapeutic needs. Furthermore, the incorporation of CIP into the TM patches demonstrated a favorable influence on their resultant mechanical properties. Biological studies performed with human mesenchymal stromal cells confirmed the absence of any cytotoxic or anti-proliferative effects from the released antibiotic. Finally, antibacterial assays validated the efficacy of CIP-loaded scaffolds in suppressing bacterial infections, highlighting their promising relevance for TM applications. Abstract:Halloysite nanotube (HNT)-reinforced alginate-based nanofibrous scaffolds were successfully fabricated by electrospinning to mimic the natural extracellular matrix (ECM) structure which is beneficial for tissue regeneration. An antiseptic drug, cephalexin (CEF)-loaded HNT, was incorporated into the alginate-based matrix to obtain sustained antimicrobial protection and robust mechanical properties, the key criteria for tissue engineering applications. Electron microscopic imaging and drug release studies revealed that CEF had penetrated into the lumen space of the HNT and also deposited on the outer walls, with a total loading capacity of 30 wt %. Moreover, the diameter of alginate-based nanofibers of the scaffolds ranged from 40 to 522 nm with well-aligned HNTs, resulting in superior mechanical properties. For instance, the addition of 5% (w/w) HNT improved the tensile strength (σ) and elastic modulus by 3-fold and 2-fold, respectively, compared to those of the alginate-based scaffolds without HNT. The fabricated scaffolds exhibited remarkable antimicrobial properties against both Gram-negative and Gram-positive bacteria, and the cytotoxicity studies confirmed the nontoxicity of the fabricated scaffolds. Drug release kinetics showed that CEF inside HNTs diffuses within 24 h and that the diffusion of the drug is delayed by 7 days once the CEF-loaded HNTs are incorporated into the alginate-based nanofibers. These fabricated alginate-based electrospun scaffolds with enhanced mechanical properties and sustained antimicrobial protection hold great potential to be used as artificial ECM scaffolds for tissue engineering applications.
Identification of Sites in Domain I of Perlecan That Regulate Heparan Sulfate Synthesis*

Abstract:Perlecan is primarily a heparan sulfate containing proteoglycan found in all basement membranes. Rotary shadowed images of perlecan show it to contain three glycosaminoglycan (GAG) side chains extending from one end of its core protein. Domain I is at the N terminus of perlecan and contains three closely spaced Ser-Gly-Asp sequences that may serve in GAG attachment. We evaluated the serines in these three sequences for GAG attachment by preparing a cDNA construct encoding for the N-terminal half (domains I, II, and III) of perlecan and then a series of constructs containing deletions and mutations within domain I of the domain I/II/III construct, expressing these constructs in COS-7 cells, and then analyzing the recombinant product for GAG side chains and GAG type. The results showed that all three serine residues in the Ser-Gly-Asp sequences in domain I can accept both chondroitin and heparan sulfate side chains but that a cluster of acidic residues N-terminal to these sequences is the primary determinant responsible for targeting these sites for heparan sulfate. Furthermore, there are two elements that can enhance heparan sulfate synthesis at a targeted site: 1) the presence of a the SEA module in the C-terminal region of domain I and 2) the presence of multiple acceptors in close proximity. These results indicate that the proportion of heparan and chondroitin sulfate at any one site in domain I of perlecan is regulated by multiple factors.
Fabrication of Ciprofloxacin-Loaded Sodium Alginate Nanobeads Coated with Thiol-Anchored Chitosan Using B-390 Encapsulator Following Optimization by DoE

Abstract:Most infectious diseases of the gastrointestinal tract can easily be treated by exploiting the already available antibiotics with the change in administration approach and delivery system. Ciprofloxacin (CIP) is used as a drug of choice for many bacterial infections; however, long-term therapy and off-site drug accumulation lead to an increased risk of tendinitis and peripheral neuropathy. To overcome this issue, nanotechnology is being exploited to encapsulate antibiotics within polymeric structures, which not only facilitates dose maintenance at the infection site but also limits off-site side effects. Here, sodium alginate (SA) and thiol-anchored chitosan (TC) were used to encapsulate CIP via a calcium chloride (CaCl2) cross-linker. For this purpose, the B-390 encapsulator was employed in the preparation of nanobeads using a simple technique. The hydrogel-like sample was then freeze-dried, using trehalose or mannitol as a lyoprotectant, to obtain a fine dry powder. Design of Experiment (DoE) was utilized to optimize the nanobead production, in which the influence of different independent variables was studied for their outcome on the polydispersity index (PDI), particle size, zeta potential, and percentage encapsulation efficiency (% EE). In vitro dissolution studies were performed in simulated saliva fluid, simulated gastric fluid, and simulated intestinal fluid. Antibacterial and anti-inflammatory studies were also performed along with cytotoxicity profiling. By and large, the study presented positive outcomes, proving the advantage of using nanotechnology in fabricating new delivery approaches using already available antibiotics.

Antibacterial Electrospun Membrane with Hierarchical Bead-on-String Structured Fibres for Wound Infections

Abstract:Chronic wounds often result in multiple infections with various kinds of bacteria and uncontrolled wound exudate, resulting in several healthcare issues. Advanced medicated nanofibres prepared by electrospinning have gained much attention for their topical application on infected chronic wounds. The objective of this work is to enhance the critical variables of ciprofloxacin-loaded polycaprolactone-silk sericin (PCL/SS-PVA-CIP) nanofibre production via the process of electrospinning. To examine the antibacterial effectiveness of PCL/SS-PVA-CIP nanocomposites, the material was tested against P. aeruginosa and S. aureus. The combination of PCL/SS-PVA-CIP exhibited potent inhibitory properties, with the most effective concentrations of ciprofloxacin (CIP) being 3 μg/g and 7.0 μg/g for each bacterium, respectively. The biocompatibility was evaluated by conducting cell reduction and proliferation studies using the human epidermal keratinocyte (HaCaT) cells and human gingival fibroblasts (HGFs) in vitro cell lines. The PCL/SS-PVA-CIP showed good cell compatibility with HaCaT and HGF cells, with effective proliferation even at antibiotic doses of up to 7.0 μg/g. The drug release effectiveness of the nanocomposites was assessed at various concentrations of CIP, resulting in a maximum cumulative release of 76.5% and 74.4% after 72 h for CIP concentrations of 3 μg/g and 7 μg/g, respectively. In summary, our study emphasizes the possibility of combining silk sericin (SS) and polycaprolactone (PCL) loading with CIP nanocomposite for wound management.
Preparation and performance characterization of electrospun tetracycline hydrochloride-loaded polyglycolic acid nanofibrous membrane

Abstract:BACKGROUND: Local drug controlled-delivery systems for anti-inflammatory are gaining increasing attention for bone defect regeneration because of their extended therapeutic effects and reduced adverse effects. Electrospun polymer nanofiber is a kind of good drug delivery vehicle. OBJECTIVE: To fabricate tetracycline hydrochloride-loaded polyglycolic acid (PLGA) nanofibrous membrane with varied tetracycline hydrochloride contents using the electrospinning technique, and to study their performance in preventing bacterial infections as well as their cell compatibility. METHODS: Tetracycline hydrochloride-loaded PLGA nanofibrous membranes were synthesized at 15-20 kV under a steady flow rate of 0.3-0.5 mL/h using an electrospinning technique. Through the images of scanning electron microscopy, we obtained morphological characters of the nanofibers. Drug entrapment efficiency was determined, drug release profiles were tested and in vitro antimicrobial activities were observed from modified Kirby-Bauer test. MG-63 cells were used to test the biocompatibility of nanofibrous membrances. RESULTS AND CONCLUSION: Average diameters of PLGA nanofibers with different tetracycline hydrochloride contents (0%, 3%, 5% or 10%) were all about 360-470 nm. Tetracycline hydrochloride entrapment efficiencies were well above 80% for all the PLGA membranes. The 10% drug loaded membranes exhibited the highest initial burst release. Tetracycline hydrochloride-loaded PLGA nanofibrous membranes were effective in inhibiting growth of staphylococcus aureus but had no adverse effect on the adhesion and proliferation of MG-63 cells. Comparatively speaking, electrospun PLGA/tetracycline hydrochloride membranes with drug loading of 3% and 5% are good choices for the prevention of bacterial infections in guided tissue regeneration.
Synergistic Effect of Co-Delivering Ciprofloxacin and Tetracycline Hydrochloride for Promoted Wound Healing by Utilizing Coaxial PCL/Gelatin Nanofiber Membrane

Abstract:Combining multiple drugs or biologically active substances for wound healing could not only resist the formation of multidrug resistant pathogens, but also achieve better therapeutic effects. Herein, the hydrophobic fluoroquinolone antibiotic ciprofloxacin (CIP) and the hydrophilic broad-spectrum antibiotic tetracycline hydrochloride (TH) were introduced into the coaxial polycaprolactone/gelatin (PCL/GEL) nanofiber mat with CIP loaded into the PCL (core layer) and TH loaded into the GEL (shell layer), developing antibacterial wound dressing with the co-delivering of the two antibiotics (PCL-CIP/GEL-TH). The nanostructure, physical properties, drug release, antibacterial property, and in vitro cytotoxicity were investigated accordingly. The results revealed that the CIP shows a long-lasting release of five days, reaching the releasing rate of 80.71%, while the cumulative drug release of TH reached 83.51% with a rapid release behavior of 12 h. The in vitro antibacterial activity demonstrated that the coaxial nanofiber mesh possesses strong antibacterial activity against E. coli and S. aureus. In addition, the coaxial mats showed superior biocompatibility toward human skin fibroblast cells (hSFCs). This study indicates that the developed PCL-CIP/GEL-TH nanofiber membranes hold enormous potential as wound dressing materials.

Abstract:The design of antibacterial-releasing coatings or wrapping materials with controlled drug release capability is a promising strategy to minimise risks of infection and medical device failure in vivo. Collagen fibres have been employed as medical device building block, although they still fail to display controlled release capability, competitive wet-state mechanical properties, and retained triple helix organisation. We investigated this challenge by pursuing a multiscale design approach integrating drug encapsulation, in-situ covalent crosslinking and fibre spinning. By selecting ciprofloxacin (Cip) as a typical antibacterial drug, wet spinning was selected as a triple helix-friendly route towards Cip-encapsulated collagen fibres; whilst in situ crosslinking of fibre-forming triple helices with 1,3 phenylenediacetic acid (Ph) was hypothesised to yield Ph-Cip {\pi}-{\pi} stacking aromatic interactions and enable controlled drug release. Higher tensile modulus and strength were measured in Ph crosslinked fibres compared to state-of-the-art carbodiimide crosslinked controls. Cip-encapsulated Ph-crosslinked fibres revealed decreased elongation at break and significantly-enhanced drug retention in vitro with respect to Cip-free variants and carbodiimide-crosslinked controls, respectively. This multiscale manufacturing strategy provides new insight aiming at wet spun collagen triple helices with nanoscale-regulated tensile properties and drug release capability.
Multifunctional hydrogel based on silk fibroin/thermosensitive polymers supporting implant biomaterials in osteomyelitis

Abstract:The search for novel antibiotic-loaded biomaterials that support the management of osteomyelitis has been extensive in current years. This study developed multifunctional hydrogel to enhance the performance of osteo-conduction and antimicrobial as supporting implant materials for osteomyelitis treatment and prevention. Ciprofloxacin-loaded thermosensitive hydrogels composed of silk fibroin and poloxamers were prepared using the cold method. F1, F2, and F3 formulations resulted in gelation time of within 3 min under 37°C. Physical evaluation showed that the formulations, especially F3, had a proper swelling ratio and 82.34% biodegradability within 14 days. Ciprofloxacin from formulations exhibited an initial burst release within 24 h and continuous delivery, with more than 84% release over 14 days. Released antibiotics showed efficacy against Staphylococcus aureus ATCC25923 and eradicating activity against biofilms. Moreover, hydrogels had no cytotoxic effect on MC3T3-E1 cells with 93–106% cell viability. This multifunctional hydrogel showed a promising approach for supporting osteomyelitis treatment and prevention.

Medicated Scaffolds Prepared with Hydroxyapatite/Streptomycin Nanoparticles Encapsulated into Polylactide Microfibers

Abstract:The preparation, characterization, and controlled release of hydroxyapatite (HAp) nanoparticles loaded with streptomycin (STR) was studied. These nanoparticles are highly appropriate for the treatment of bacterial infections and are also promising for the treatment of cancer cells. The analyses involved scanning electron microscopy, dynamic light scattering (DLS) and Z-potential measurements, as well as infrared spectroscopy and X-ray diffraction. Both amorphous (ACP) and crystalline (cHAp) hydroxyapatite nanoparticles were considered since they differ in their release behavior (faster and slower for amorphous and crystalline particles, respectively). The encapsulated nanoparticles were finally incorporated into biodegradable and biocompatible polylactide (PLA) scaffolds. The STR load was carried out following different pathways during the synthesis/precipitation of the nanoparticles (i.e., nucleation steps) and also by simple adsorption once the nanoparticles were formed. The loaded nanoparticles were biocompatible according to the study of the cytotoxicity of extracts using different cell lines. FTIR microspectroscopy was also employed to evaluate the cytotoxic effect on cancer cell lines of nanoparticles internalized by endocytosis. The results were promising when amorphous nanoparticles were employed. The nanoparticles loaded with STR increased their size and changed their superficial negative charge to positive. The nanoparticles’ crystallinity decreased, with the consequence that their crystal sizes reduced, when STR was incorporated into their structure. STR maintained its antibacterial activity, although it was reduced during the adsorption into the nanoparticles formed. The STR release was faster from the amorphous ACP nanoparticles and slower from the crystalline cHAp nanoparticles. However, in both cases, the STR release was slower when incorporated in calcium and phosphate during the synthesis. The biocompatibility of these nanoparticles was assayed by two approximations. When extracts from the nanoparticles were evaluated in cultures of cell lines, no cytotoxic damage was observed at concentrations of less than 10 mg/mL. This demonstrated their biocompatibility. Another experiment using FTIR microspectroscopy evaluated the cytotoxic effect of nanoparticles internalized by endocytosis in cancer cells. The results demonstrated slight damage to the biomacromolecules when the cells were treated with ACP nanoparticles. Both ACP and cHAp nanoparticles were efficiently encapsulated in PLA electrospun matrices, providing functionality and bioactive properties.

Enhanced wound healing of ciprofloxacin incorporated PVA/alginate/PAA electrospun nanofibers with antibacterial effects and controlled drug release

Abstract:Novel polyvinyl alcohol (PVA), alginate (Alg), and polyacrylic acid (PAA) based nanofiber matrix incorporated with ciprofloxacin (Cip) was manufactured via electrospinning. The functional matrix could accelerate wound healing and also prevent infections by a controlled release of ciprofloxacin. Beads were observed in PVA-Cip which disappeared with the addition of Alg and PAA resulting in the thinnest diameter (141 ± 53 nm) and the most uniform surface. The polymers underwent both intermolecular hydrogen and ester bonds via abundant carboxyl and hydroxyl groups trapping ciprofloxacin inside the matrix. The tensile properties of PVA-Alg-PAA-Cip were significantly higher than the other two samples because of the complex intermolecular bonds. Although the glass transition temperatures (T g ) of the samples were similar, the melting temperature (T m ) of PVA-Alg-PAA-Cip was the highest at 339 o C due to stronger intermolecular bonding. Because of the absorbable nature of alginate, PVA-Alg-Cip swelled the most (600%) in different pH conditions. A controlled release was achieved with only 40% release of ciprofloxacin from PVA-Alg-PAA-Cip compared to 70% in the case of PVA-Cip in neutral pH after 24 h. Because of this slow release, all the samples were able to kill both S. aureus and E. coli for at least 7 days with just a single dose of the drug and PVA-Alg-PAA-Cip demonstrated higher bactericidal activity than other samples. The functional polymer matrix could induce faster healing in a murine excisional wound model, unlike the control group. Ciprofloxacin prevented bacterial infection facilitating faster re-epithelialization as observed in hematoxylin and eosin (H&E) and Masson's trichrome (MT) staining.

Photothermally Controlled Drug Release of Poly(d,l-lactide) Nanofibers Loaded with Indocyanine Green and Curcumin for Efficient Antimicrobial Photodynamic Therapy

Abstract:Chronic wound infections with antibiotic-resistant bacteria have become a significant problem for modern healthcare systems since they are often associated with high costs and require profound topical wound management. Successful wound healing is achieved by reducing the bacterial load of the wound and providing an environment that enhances cell growth. In this context, nanofibers show remarkable success because their structure offers a promising drug delivery platform that can mimic the native extracellular matrix and accelerate cell proliferation. In our study, single-needle electrospinning, a versatile and cost-efficient technique, was used to shape polymers into an applicable and homogeneous fleece capable of a photothermally triggered drug release. It was combined with antimicrobial photodynamic therapy, a promising procedure against resistant bacteria. Therefore, poly(d,l-lactide) nanofibers loaded with curcumin and indocyanine green (ICG) were produced for local antimicrobial treatment. The mesh had a homogeneous structure, and the nanofibers showed a smooth surface. Recordings with a thermal camera showed that near-infrared light irradiation of ICG increased the temperature (>44 °C) in the surrounding medium. Release studies confirmed more than 29% enhanced curcumin release triggered by elevated temperature. The antimicrobial activity was tested against the gram-positive strain Staphylococcus saprophyticus subsp. bovis and the gram-negative strain Escherichia coli DH5 alpha. The nanofibers loaded with both photosensitizers and irradiated with both wavelengths reduced the bacterial viability (~4.4 log10, 99.996%) significantly more than the nanofibers loaded with only one photosensitizer (<1.7 log10, 97.828%) or irradiated with only one wavelength ( 4.3 log10 (99.995%), which was also confirmed visually. Finally, the produced nanofibers showed good biocompatibility, proven by the cellular viability of mouse fibroblasts (L929). The data demonstrate that we have developed a new economic nanofiber formulation, which offers a triggered drug release, excellent antimicrobial properties, and good biocompatibility.

Abstract:Nanofibers prepared by biobased materials are widely used in the field of biomedicine, owing to outstanding biocompatibility, biodegradable characters, and excellent mechanical behavior. Herein, we fabricated multilayered nanofibrous scaffolds in order to improve the performance of drug delivery. The composite layer-by-layer scaffolds were incorporated by hydrophobic poly( l -lactic acid) (PLA): polycaprolactone (PCL) and hydrophilic poly(vinyl alcohol) (PVA) nanofibers via multilayer electrospinning. Morphological and structural characteristics of the developed scaffolds measured by scanning electron microscopy (SEM), and transmission electron microscopy (TEM) confirmed smooth and uniform fibers ranging in nanometer scale. The differences in contact angles and Fourier transform infrared spectrum (FTIR) between single-layered PVA nanofibers and multilayered scaffolds verified the existence of PLA: PCL surface. In vitro biodegradable and drug release analysis depicted multilayered scaffolds had good biodegradability and potential for medical application. Due to the model drug incorporation, scaffolds exhibited good antibacterial activity against Escherichia coli and Staphylococcus aureus by the zone of inhibition test. These results revealed that the multilayered scaffolds were proved to be desirable antibacterial materials for biomedical application.

Abstract:Objective: The nanofibre membrane with the antibacterial doxycycline (DOX) intermediate layer can direct tissue regeneration and have antibacterial properties. Through the use of electrospinning, we were able to create DOX-loaded guided tissue regeneration (GTR)/guided bone regeneration (GBR) membranes and assess their efficacy in the treatment of peri-implantitis. Methods: Electrostatic spinning was used to create DOX-containing poly(caprolactone) (PCL) nanofibres, and a scanning electron microscope (SEM) was used to examine the membranes' surface morphology. Results: The synthesised DOX-PCL nanofibres were found to have a smooth surface and a uniform diameter distribution, as revealed by scanning electron microscopy. When the percentage of DOX was raised from 15% to 25%, the fibre diameter shrank from 247.17 nm to 181.34 nm. According to differential calorimetry, the heat absorption peaks for the pre-electrospun PCL and 10% PCL electrospun membranes were most pronounced at 66 °C, while the peaks for the DOX powder showed at 171.5 °C. With the shift in drug loading, carbonization occurred at 223.5 degrees Celsius. PCL underwent a thermal reaction between 60 and 66 degrees Celsius. However, medication loading was strongly correlated with the level of suppression. Agar diffusion results showed that DOX-loaded nanofibre membranes inhibited the growth of Actinomyces (Aa) and Porphyromonas gingivalis; for both bacterial species, the diameter of the inhibition zone grew larger when more drug was added to the membranes. After 1 hour, 4 hours, 12 hours, 1 day, 7 days, and 28 days of the experiment, the cumulative drug release rates of CL+DOX were 19.14%, 36.16%, 44.37%, 59.52%, and 65.150%, respectively. After the initial steep drop during the first three days of the trial, the PCL-DOX release rate stabilised at around 1.61 percent per hour. Minimum concentration was 3.13 g/mL, while PCL-DOX release rate fell from 1.28 percent on Day 4 to 0.51 percent on Day 28. Conclusion: Combining the availability of natural polymeric materials with the reliability of manufactured polymeric materials, the three-layer nanofibre membranes use PCL-GE as a double surface layer and PCL-DOX as an intermediate layer. In addition, the physical insulating qualities of these membranes are maintained, which allows the fibre membrane to perform a sustained antibacterial action and prevents the initial, abrupt release of the medication in the tissue. Peri-implantitis may be treatable with the use of both pharmaceuticals and GTR/GBR technology.

Abstract:The development of biocompatible scaffolds is rapidly growing in several fields of human health, particularly for the treatment of bone defects. In this study, silver nanoparticle (AgNPs)-incorporated hydroxyapatite (HA), chitosan (Chi), and polyvinylpyrrolidone (PVP)-based scaffolds were fabricated using the freeze-drying method for bone tissue engineering. Physicochemical properties such as porosity, density, swelling ratio, and biodegradability, as well as blood biocompatibility, brine shrimp lethality, antibacterial activities, FTIR-ATR, tensile strength, cytotoxicity, and SEM, were conducted. A study on the in vitro release of ceftriaxone (Cef), a routinely prescribed medication for treating osteomyelitis, was done. The selected scaffolds had desirable porosity, density, swelling capability, and biodegradation rate, which would meet the basic prerequisites for osteogenesis. The scaffolds showed qualitative and quantitative antibacterial activities against Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, and Pseudomonas aeruginosa. Scaffolds were minimally cytotoxic to brine shrimp and RBC biocompatible. FTIR analysis confirmed intermolecular interaction among components of scaffolds. Scaffold had a tensile strength of 2.9 ± 0.22 N/mm2 and a non-cytotoxic effect on Vero cell lines. Surface morphology, pore shape, and sizes (98–382 μm) were observed through SEM. The percentage of drug (Cef) loading was found to be 63.16 ± 4.85%. An initial burst release of Cef was 40.21 ± 1.50% in the first 24 h. A sustained release of Cef was observed after 4 days, and about 60.36 ± 2.38% was released within 14 days. Therefore, selected AgNPs-incorporated HA-Chi-PVP scaffold could be a potential aid for bone tissue engineering.

Abstract:Guided tissue regeneration/guided bone regeneration membranes with sustained drug delivery were developed by electrospinning drug-loaded halloysite clay nanotubes doped into poly(caprolactone)/gelatin microfibers. Use of 20 wt % nanotube content in fiber membranes allowed for 25 wt % metronidazole drug loading in the membrane. Nanotubes with a diameter of 50 nm and a length of 600 nm were aligned within the 400 nm diameter electrospun fibers, resulting in membranes with doubling of tensile strength along the collector rotating direction. The halloysite-doped membranes acted as barriers against cell ingrows and have good biocompatibility. The metronidazole-loaded halloysite nanotubes incorporated in the microfibers allowed for extended release of the drugs over 20 days, compared to 4 days when directly admixed into the microfibers. The sustained release of metronidazole from the membranes prevented the colonization of anaerobic Fusobacteria, while eukaryotic cells could still adhere to and proliferate on the drug-loaded composite membranes. This indicates the potential of halloysite clay nanotubes as drug containers that can be incorporated into electrospun membranes for clinical applications.
Polycaprolactone-gelatin nanofibers incorporated with dual antibiotic-loaded carboxyl-modified silica nanoparticles

Abstract:In this study, we used electrospun polycaprolactone (PCL) or a mixture of PCL and gelatin (Gel) in a mixed acidic solvent to develop antimicrobial electrospun nanofibers. Carboxyl-modified mesoporous silica nanoparticles (CMSNs) or CMSNs loaded with antibiotic drugs polymyxin B and vancomycin (CMSNs/ABs) were mixed with the electrospinning solution in concentrations of 1%, 2.5% and 5%. The nanofibers diameter measured between 122 and138 nm. Higher concentrations of gelatin or CMSNs increased hydrophilicity and degradability of the nanofibers. CMSNs enhanced nanofibers mechanical strength. PCL/Gel nanofibers incorporated with CMSNs/ABs (2.5% and 5%) showed high antibacterial efficiency against Pseudomonas aeruginosa and Staphylococcus aureus. Also bacterial cell adhesion decreased when 2.5% and 5% of CMSNs/ABs were incorporated in PCL/Gel mats. MTT and hemolysis assays indicated excellent biocompatibility of all types of electrospun nanofibers. This study confirms that a proper mixture of PCL, gelatin and CMSNs loaded with two antibiotics could offer antimicrobial activities with high biocompatibility and biodegradability properties.Graphic abstract

The treatment response of barrier membrane with amoxicillin-loaded nanofibers in experimental periodontitis

Abstract:Background: Infection control is a major determinant of guided tissue regeneration (GTR). This study aims to develop an antibiotic-loaded membrane to assist periodontal repair. Methods: Poly(D,L-lactic acid) (PDLLA) nanofibers encapsulating amoxicillin (PDLLA-AMX) were fabricated using the electrospinning technique, and their structures, drug encapsulation efficiency, and release characteristics were assessed. The viability and behaviors of periodontal ligament (PDL) cells on nanofibers, and antibacterial capabilities of nanofibers were evaluated in vitro. Early therapeutic efficiency of the antibiotic-loaded membranes was investigated in rats with ligature-induced experimental periodontitis, and the outcomes were evaluated by gene expression, microcomputed tomography imaging, and histology within 7 days of membrane placement. Results: AMX was successfully encapsulated in the PDLLA nanofibers and released in a sustained manner. After initial attachment was achieved, cells stretched out along with the directions of nanofibers. The viability and expression of migration-associated gene of PDL cells were significantly improved, and the growth of Streptococcus sanguinis and Porphyromonas gingivalis was significantly reduced in the PDLLA-AMX group compared with the controls. On PDLLA-AMX-treated sites, wound dehiscence and sulcular inflammation were reduced. Collagen fiber matrix deposition was accelerated with upregulated type I collagen and interleukin-1β, and downregulated matrix metalloproteinase-8, whereas periodontal bone level and the expressions of vascular endothelial growth factor and core-binding factor subunit alpha-1 were equivalent to conventional membrane treatment. Conclusions: PDLLA-AMX nanofibers inhibited bacterial growth and promoted the viability and mobility of PDL cells after initial cell attachment. Membranes with PDLLA-AMX nanofibers reduced inflammation and accelerated periodontal repair at an early stage, providing good prospects for the further development of GTR membranes.

Abstract:Antibiotic medications have been found to hinder the success of regenerative endodontic treatment due to the rapid degradation of the drug, and the acidic nature of ciprofloxacin (CIP) can be harmful to stem cells of the apical papilla (SCAPs), the cells responsible for regeneration. In this study, a nanocarrier system was used for controlled drug release for longer drug activity and less cytotoxicity to the cells. CIP was loaded in poly (ethylene glycol) methyl ether- block -poly (lactide- co -glycolide) (PEG–PLGA) nanoparticles (NPs) with an ion-pairing agent. The NPs demonstrated a monodispersed spherical morphology with a mean diameter of 120.7 ± 0.43 nm. The encapsulation efficiency of the CIP-loaded PEG–PLGA NPs was 63.26 ± 9.24%, and the loading content was 7.75 ± 1.13%. Sustained CIP release was achieved over 168 h and confirmed with theoretical kinetic models. Enhanced NP bactericidal activity was observed against Enterococcus faecalis . Additionally, CIP-loaded PEG–PLGA NPs had a low cytotoxic effect on SCAPs. These results suggest the use of a nanocarrier system to prolong the antibiotic activity, provide a sterile environment, and prevent reinfection by the bacteria remaining in the root canal during regenerative endodontic treatment.

Abstract:The development of wound-dressing materials has attracted significant research interests in recent years. With the advancement of nanofabrication, the application of nanoparticles (NPs) in drug delivery systems has become feasible. However, most existing work focuses on incorporation of metal, metal/semi-metal oxide, or organic particles into nanofiber scaffolds. There has been a lack of work on the incorporation of drug-encapsulated polymeric particles into nanofiber scaffolds. In this study, gentamicin-encapsulated poly(lactic-co-glycolic acid) (PLGA) NPs were synthesized via a double emulsion solvent evaporation method. Electrospinning was used to incorporate gentamicin-encapsulated PLGA NPs into nanofiber scaffolds. Atomic force microscopy (AFM), dynamic light scattering, scanning electron microscopy (SEM), ultraviolet–visible spectroscopy (UV–vis), and an agar diffusion method were utilized to characterize the morphologies, release profiles, and antibacterial activities of various gentamicin-loaded PLGA NP-incorporated nanofiber scaffolds. The results indicated that the PLGA NPs had a spherical morphology with an average diameter of 130 nm. Purification of PLGA NPs was essential to eliminate the residual poly(vinyl alcohol) (PVA) and to prevent particle agglomeration. The purified PLGA NPs were uniformly and individually incorporated into the polyurethane (PU)/ poly(ethenyl oxide) (PEO) or PEO-only nanofiber scaffolds but nearly none into the PU-only fiber scaffolds. PEO served as a continuous phase in the PU/PEO mixture, which significantly improved the compatibility of PLGA NPs and PU, resulting in a well-dispersed distribution of PLGA NPs in the monolithic nanofiber scaffolds. Excellent antibacterial properties against Escherichia coli were found in both PU/PEO and PEO nanofiber scaffolds. This study of incorporating gentamicin-encapsulated PLGA NPs into fiber scaffolds provides insights for achieving successful incorporation of drug-encapsulated polymeric NPs into fiber scaffolds. This offers a promising microfabrication technology for delivery of therapeutic molecules with controlled release for biomedical applications.
