Sw. is a perennial herbaceous plant with high gardening and herbal value. In August 2014, a severe anthracnose disease was observed on C. ensifolium leaves in Changchun (125.24°E, 43.48°N), Jilin Province, China. More than 80% of the plants were symptomatic. In the early stage of disease development, small brown lesions (1 mm in diameter) were visible on infected leaves, which subsequently developed into oval spots and turned light brown in the center. Lesions ranged from 5 to 15 mm in diameter. As lesions enlarged and coalesced, some leaves dried up. When the humidity was high, the surfaces of the lesions were covered with dark brown acervuli. Small pieces of infected leaves were surface-sterilized in 75% ethanol for 60 s, sterilized for 1 min in 1.5% NaClO, rinsed twice with sterilized distilled water, and plated on potato dextrose agar (PDA) in Petri dishes at 25 ± 1°C for 7 days. Ten fungal isolates were transferred from PDA to synthetic nutrient-poor agar (SNA). White colonies were observed in 2 days with smooth margins, and they covered the Petri dishes after 7 days. Perithecia (n = 100) were brown to dark brown, globose to subglobose, and measured 136.24 to 154.5 × 145.42 to 236.68 µm. Asci and ascospores were absent. Conidiophores and setae were formed directly on the media. Conidiophores (n = 100) were cylindrical, septate, branched, and measured 5.35 to 18.28 × 2.43 to 6.56 µm. Conidia (n = 100) were hyaline, aseptate, smooth, cylindrical, contents granular, and were 12.21 to 18.42 µm in length, and 4.40 to 7.90 µm in width. Setae (n = 100) were brown, cylindrical to conical, measuring 40.62 to 160.58 µm in length, and were 1 to 4 septate. Based upon these morphological and cultural characteristics, the causal agent was identified as Colletotrichum cymbidiicola. Five sequences of the internal transcribed spacer (ITS, KX058529), glyceraldehyde-3-phosphate dehydrogenase (GAPDH, KX068532), actin (ACT, KX058531), and beta-tubulin (TUB, KX058530) regions were obtained and deposited in GenBank. BLAST search results revealed that all sequences showed 99% identity with C. cymbidiicola sequences. Pathogenicity of 10 isolates of C. cymbidiicola from C. ensifolium were confirmed by inoculating healthy leaves of C. ensifolium plants with a spore suspension (1 × 106 spores/ml) produced on PDA. Each isolate was inoculated onto five plants and another five healthy plants were sprayed with sterilized water as controls. All plants were enclosed in plastic bags for 48 h in a greenhouse at 28°C and 12 h/day light cycle. After 7 days, inoculated plants showed similar symptoms to those on the original diseased plants, while control plants remained symptomless. C. cymbidiicola was successfully reisolated from symptomatic plants to fulfill Koch’s postulates. C. cymbidiicola causing disease was reported on cymbidium in Australia, New Zealand, Japan (Damm et al. 2012; Hou et al. 2016), and India (Cannon et al. 2012). Robustness in Speech, Speaker, and Language Recognition: "You've Got to Know Your Limitations"

Abstract:In the field of speech, speaker and language recognition, significant gains have and are being made with new machine learning strategies along with the availability of new and emerging speech corpora. However, many of the core scientific principles required for effective speech processing research appear to be drifting to the sidelines with the assumptions that access to larger amounts of data can address a growing range of issues relating to new speech/speaker/language recognition scenarios. This study focuses on exploring several challenging domains in formulating effective solutions in realistic speech data, and in particular the notion of using naturalistic data to better reflect the potential effectiveness of new algorithms. Our main focus is on mismatch/speech variability issues due to (i) differences in noisy speech with and without Lombard effect and a communication factor, (ii) realistic field data in noisy/increased cognitive load conditions, and (iii) dialect identification using found data. Finally, we study speaker–noise and speaker–speaker interactions in a newly established, fully naturalistic Prof-Life-Log corpus. The specific outcomes from this study include an analysis of the strengths and weaknesses of simulated vs. actual speech data collection for research.

First Report of Powdery Mildew Caused by Podosphaera fusca on Helianthus tuberosus in China

Abstract:Helianthus tuberosus L. (Jerusalem artichoke) is an herbaceous perennial plant in the Asteraceae that is native to North America but introduced to China in the 17th century. The tubers of H. tuberosus are used as a vegetable, for the pharmaceutical production of inulin and as a source of ethanol biofuel, several B vitamins and minerals。From June to September 2009, severe powdery mildew infection was observed on H. tuberosus in a vegetable garden at Shangqiu Normal University, Shangqiu, China. Approximately 60% of leaves on individual plants were symptomatic, and almost 70% of the plants were infected. Initially, discrete powdery mildew colonies appeared on the upper surface of the leaves and on stems of the plants. Later, mycelial growth was amphigenous, thick, forming irregular white patches, covering the whole leaf surface. Eventually, leaves turned yellow, withered, and abscissed. Microscopic observations showed that conidia on infected leaves were ellipsoid-ovoid to barrel-shaped, with distinct fibrosin bodies visible in their cytoplasm, measuring 28 to 38 × 15 to 22 μm (n = 40). Conidiophores were unbranched, straight, 80 to 210 × 8 to 14 µm (n = 40) in size, and produced two to six immature conidia in chains with intercellular diaphragms. Foot cells of conidiophores were cylindrical and 45 to 65 μm × 8 to 14 µm (n = 40), with slight constrictions at basal septa, and followed by one to three short cells. Fungal hyphae were septate, branched, and flexuous to straight and 5 to 8 µm wide with indistinct to slightly nipple-shaped appressoria. These structures are typical of the genus Podosphaera, although chasmothecia were not observed. The sequence of ITS1-5.8s-ITS2 region of rDNA were amplified from conidia collected from infected leaves with universal primers ITS1F and ITS4 (White et al. 1990), sequenced and analyzed using the BLASTn search of GenBank. Amplicons were 603 bp (GQ927254) and exhibited 99.83% sequence identity with sequence of P. fusca from Cucurbita pepo (KJ698669) in Italy (Pirondi et al. 2015). The fungal species was identified as P. fusca (synonym P. xanthii ) by morphological characteristics and molecular analysis (Braun and Takamatsu 2000; Braun and Cook 2012). Pathogenicity tests were conducted by gently pressing the infected leaves onto leaves of six healthy H. tuberosus plants while six noninoculated plants served as controls. Plants were maintained in a greenhouse at 25 ± 2°C. Eight days after inoculation, symptoms similar to those observed under natural conditions developed on the inoculated leaves of H. tuberosus plants, whereas the control plants remained symptomless. The fungus on inoculated leaves was morphologically identical to that first observed in the field. P. fusca parasitizes a large number of asteraceous species including Euryops pectinatus (Saenz et al. 2000), Coreopsis lanceolata (Garibaldi et al. 2007), Cosmos caudatus (Siddiqui et al. 2011), Herba eupatorii (Ding et al. 2013) and etc. Powdery mildew caused by P. fusca has been reported on H. tuberosus in Russia (Farr and Rossman 2019). Golovinomyces ambrosiae was previously recorded on H. tuberosus in China (Huang et al. 2017; Radisek et al. 2018). This is the first report to our knowledge of powdery mildew P. fusca on H. tuberosus in China. It could cause significant yield losses and become a threat to production of H. tuberosus .
First Report of Fusarium Annulatum Causing Blight on Bletilla Striata (baiji) in China.

Abstract:Bletilla striata (Thunb. ex A. Murray) Rchb (known as baiji in Chinese), a herbal plant distributed mainly in China, has become a focus of scientific attention recently due to its medicinal value (He et al. 2017). In May 2023, blight symptoms on leaves and stems were observed approximately 60% of Bletilla striata in Hangzhou, Zhejiang, China (29.80° N, 119.67° E). Brown spots initially appear on the infected leaves, which gradually decay as the spots expand. The wilting is accompanied with fading and yellowing, and eventually leading to defoliation. The infected stem initially appears brown spots, which gradually decay as the spots expand, resulting in the death of the whole plant, affecting the yield and quality of the herbs ultimately. To isolate the pathogen, small symptomatic leaves and stems (5×5 mm) were surface-disinfected with 75% ethanol for 30 s and 1% NaClO for 2 min, then rinsed in distilled water 3 times. Subsequently, the disinfected tissues were placed on PDA and incubated at 27 ℃ for 3 days. A total of 8 fungal isolates with similar morphological characteristics were obtained. The colony by single-spore purification was light purple to dark purple with abundant aerial mycelium. Macroconidia were relatively slender with a curve, mainly three to five septate and measuring 24.34 to 54.64 μm (average 40.29 μm) × 3.59 to 5.45 μm (average 4.49 μm) (n=30). Microconidia appeared obovoid to pyriform, with sizes of 5.31 to 8.43 μm (average 7.12 μm) × 2.30 to 4.29 μm (average 3.22 μm) (n=30). The morphological characteristics were consistent with Fusarium annulatum (Yilmaz et al. 2021). To further confirm the isolate's identification, the genomic DNA of isolates were extracted and identified by phylogenetic analyses of multilocus sequences of the RNA polymerase largest subunit (rpb1, primers Fa and G2R), RNA polymerase second largest subunit (rpb2, primers 7cf and 11ar) and the translation elongation factor 1-alpha (tef1, primers EF1 and EF2) (O'Donnell et al. 2022). The sequences were deposited in GenBank (rpb1: OR493933, OR493934, OR753402; rpb2: OR753398, OR753399, OR753400; tef1: OR493935, OR493936, OR753401). BLAST searches of the rpb1, rpb2, and tef1 sequences revealed 99.83% (1775/1778 nt), 99.79% (957/959 nt), and 98.98% (678/685 nt) homology with those of Fusarium annulatum CBS:258.54 from New Caledonia (rpb1: MT010944; rpb2: MT010983; tef1: MT010994). To confirm pathogenicity, one-year-old B. striata leaves and stems were disinfected with 75% ethanol, wounded with a sterile syringe on 3 healthy leaves and stems, inoculated with 5 × 5 mm mycelial discs of strain BJ-L1 and BJ-S1, respectively. And the control were treated similarly except that they were inoculated with PDA discs. The experiment was replicated 3 times. After 5 days, all inoculated leaves and stems showed similar symptoms to those initially observed on infected plants. The same pathogen was re-isolated and identified by morphological characterization and molecular analysis, confirming Koch's postulates. Thus, the pathogen causing blight of B. striata was determined to be F. annulatum. To our knowledge, this is the first report of F. annulatum causing blight on B. striata in China. F. annulatum has a wide range of hosts and has been reported to infect a wide range of crops, fruits and vegetables (Bacon et al. 1991). This study provides the basis for further research on this disease and is important for the management of this disease and the improvement of the economic benefits of B. striata.
First Report of Green Mould on Leaf of Phaseolus vulgaris Caused by Cladosporium tenuissimum in Liaoning, China

Abstract:Phaseolus vulgaris Linn. is a widely cultivated vegetable throughout the world. From spring 2019 to 2022, green mould symptoms were observed on leaves of P. vulgaris in the greenhouse in Liaoning, China, with disease incidence of 8-75% (plants) and 6-23% (leaves). Symptoms appeared as chlorotic lesions covered with dark green mould. The infections started at the apex or margin of the leaves and then spread inward with a characteristic "V" shape. Lesions exhibited curly morphology. 15 leaf samples with typical symptoms were collected from 5 different greenhouses. A total of 75 (5 replicates of each sample) leaf tissues (0.5 cm × 0.5 cm) were selected from the boundary between diseased and healthy parts. These samples were surface sterilized in 0.5% NaClO formin, rinsed 3 times in sterile distilled water and subsequently incubated at 28℃ on potato dextrose agar (PDA) supplemented with streptomycin (50 μg/ml). Numerous morphologically uniform colonies had been purified, with no other fungi observed. Afterwards, the strains were subcultured on malt extract agar (MEA). Colonies on MEA reached 70 to 80 mm diam after 14 days, smoke-grey to pale olivaceous-grey, woolly, sometimes radially wrinkled. The mycelia were pale olivaceous-grey, with hyphae measuring 1-5 μm wide (n = 20). The conidiophores were solitary or in groups of 2 to 5, and measured 50-280(-350) × 2.5-4 μm (n = 20), with 2-7 septa. The conidiogenous cells exhibited a cylindrical-oblong morphology and measured 10-44 × 5 μm (n = 20), with 0-2 septa, and the loci frequently thickened. The conidia were catenate in densely branched chains, ellipsoid to obovoid, smooth, and measured 2.5-5 × 2-3 μm (n = 50), with 0-4 septa. The morphological characteristics were similar to Cladosporium tenuissimum (Zhang 2003). The representative isolate KZ-19 was selected for molecular identification. The rDNA-ITS, translation elongation factor 1-α and actin genes were amplified, sequenced, and the resulting sequence data were submitted to GenBank (ITS: OQ931048; EF-1α: OQ954495; ACT: OQ954496). The BLAST results exhibited a 99 to 100% similarity with the sequences of C. tenuissimum type strain CBS 125995(ITS: HM148197; EF-1α: HM148442; ACT: HM148687). Furthermore, a multi-locus phylogenetic tree was constructed using the PhyloSuite (v 1. 2. 2) software, which revealed that the strains were most closely related to C. tenuissimum (Zhang et al. 2020). Based on both morphological and molecular characteristics, KZ-19 was finally identified as C. tenuissimum (Bensch 2012). Pathogenicity testing was performed on healthy 1-month-old P. vulgaris plants by inoculating the spore suspension (1×106 conidia/ml) of KZ-19 onto leaf surfaces, while control plants were simulated inoculated with sterile water, and five pots were used for each treatment. The test was performed under field conditions of 16-28°C (temperature) and 24-56% (relative humidity). Chlorotic lesions became evident within 2 days of inoculation, followed by the appearance of green mold on leaves after 7 days. No symptoms were observed in the control group. To fulfill Koch's postulates, the pathogen was re-isolated from three inoculated leaves. The morphological identification of re-isolated pathogens was similar to that of originally isolated pathogens. No infection was observed in non-inoculated control. To the best of our knowledge, this is the first report of C. tenuissimum causing green mould on P. vulgaris. As a ubiquitous saprobic hyphomycete, C. tenuissimum has been implicated in leaf mold in Punica granatum and Trifolium repens, larch bud blight, and strawberry blossom blight in previous years (He et al. 1987; Zhang et al. 2003; Zheng et al. 2010; Nam et al. 2015), presenting a potential threat to numerous crops. Therefore, an investigation of its distribution and pathogenic potential is essential in addition to the development of effective disease management strategies.
First Report of Powdery Mildew Caused by Podosphaera xanthii on Sphagneticola trilobata in Hainan Province, China

Abstract:Sphagneticola trilobata (L.) Pruski is a perennial creeping herb of the Asteraceae family, which is native to South America. It was introduced into Southern China as a groundcover in the 1970s (Zhang et al. 2023). Now it is mainly used for folk medicine to treat various kinds of inflammatory, incuding joint pain, rheumatic diseases, arthritis, in addition to treating persistent wounds, ulcers, and edemas (Gonçalves et al. 2022). In February and November 2023, powdery mildew symptoms were observed on 60% of S. trilobata plants on the Hainan Medical University campus (19° 58' 53″ N; 110° 19' 47″ E) in Haikou, Hainan Province, China. Powdery mildew colonies covered the leaf surfaces and stems of affected plants, causing discoloration and defoliation. Mycelia were superficial and hyphal appressoria were nipple-shaped. Conidiophores (n =30) were unbranched, cylindrical, 74 to 161 × 10 to 14 µm, and produced three to five immature conidia in chains with a crenate outline. Foot cells (n =30) were cylindrical, straight or sometimes curved at the base, and 27 to 56 µm long. Conidia (n =100) were ellipsoid-ovoid to doliiform, 17 to 30 ×14 to 28 µm (length/width ratio = 1.1 to 1.9), with well-developed fibrosin bodies, and produced germ tubes from the lateral position. Based on these morphological characteristics, the pathogen was provisionally identified as Podosphaera xanthii (Braun and Cook 2012). The teleomorph was not observed. A specimen was deposited in the Hainan Medical University Plant Pathology Herbarium as HMST-23. To confirm the genus identification and ascertain a putative species, genomic DNA was extracted from mycelium, conidiophores, and conidia using a fungal DNA kit (Omega Bio-Tek, USA). The rDNA internal transcribed spacer (ITS) region was amplified with primers ITS1/ITS4 (White et al. 1990) and sequenced directly. The resulting 577-bp sequence was deposited in GenBank (accession no. OR784549). A BLASTn search in GenBank of this sequence showed 100% similarity with the ITS sequences of P. xanthii isolates from China (MT260063, MN203658, OP765400, and MT739423), Thailand (LC270780), and Vietnam (KM260731, KM260730, and KR779870). Additionally, the 28S rDNA region was amplified using the primer pairs NL1 and NL4 (O´Donnell 1993; accession no. OR784550). This region shared 100% similarity with P. xanthii isolates (LC371334, LC270782, AB936277, and OP765401) as well. Powdery mildew from Hainan sample belonged to the P. xanthii group with strong bootstrap values support 99% in maximum likelihood phylogenetic tree based on ITS and 28S gene sequences. To confirm pathogenicity, five healthy potted plants of S. trilobata were inoculated by gently pressing a powdery mildew-infected leaf onto 15 young leaves. Five non-inoculated plants served as controls. All plants were maintained in a greenhouse at 24 to 30°C, 70% relative humidity, with a 16-h photoperiod. After 7 days, inoculated leaves showed powdery mildew symptoms whereas no symptoms were observed on control plants. The fungal colonies observed on inoculated plants were morphologically identical to those found on the originally infected leaves collected from Hainan Province. Based on the morphological characteristics and molecular identification, the fungus was identified as P. xanthii. In different countries and regions, P. xanthii has been previously reported on S. trilobata in Taiwan (Yeh et al. 2021). To our knowledge, this is the first record of P. xanthii infecting S. trilobata in Hainan Province, China. S. trilobata is often planted as an ornamental plant on both sides of the road, and we are concerned that it may serve as a new host, spreading this pathogen to other economic crops.
Powdery Mildew of Xanthium strumarium Caused by Podosphaera xanthii in central China

Abstract:Xanthium strumarium, known as cocklebur, is an annual herb and has been used in traditional Chinese medicine. In October 2020, powdery mildew-like disease signs and symptoms were observed on X. strumarium grown in a crop field, Xinxiang city, Henan Province, China (35.36076° N, 113.93467° E). The specimen (PX-XS2023) was stored in Xinxiang Key Laboratory of Plant Stress Biology. White colonies in irregular or coalesced circular shaped-lesions were abundant on both ad- and abaxial surfaces of leaves and covered up to 99 % of the leaf area. Some of the infected leaves were senesced. More than 70 % of plants (n = 130) exhibited these signs and symptoms. Conidiophores were straight or slightly curved, 55 to 160 × 11 to 13 μm composed of foot-cells, shorter cells and conidia. Conidia were ellipsoid to oval, 29 to 40 × 14 to 20 μm (n = 50), with a length/width ration of 2.0 to 2.5, containing fibrosin bodies. Dark brown to black chasmothecia were found on infected leaves. The appendages were mycelium-shaped and at the base of scattered or gregarious chasmothecia (n = 50, 70 to 120 μm in diameter). Asci were 55 to 80 × 50 to 65 μm (n=30). These morphological characteristics were consistent with those of Podosphaera xanthii (Braun and Cook 2012). The internal transcribed spacer (ITS) region and Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) region of the fungus (PX-XS2023) were amplified and sequenced with primers ITS1/ITS4 (White et al. 1990) and GAPDH1/GAPDH3R (Bradshaw et al. 2022) according to a previously reported method (Zhu et al. 2022). The resulting sequences were respectively deposited into GenBank (Accession No. MW300956 and PP236083). BLASTn analysis indicated that the sequences were respectively 99.82 % (564/565) and 100% (272/272) identical to P. xanthii (MT260063 and ON075658). The phylogenetic analysis indicated that the strain PX-XS2023 and P. xanthii were clustered into a same branch. Therefore, the causal agent of powdery mildew on X. strumarium was P. xanthii. To conduct pathogenicity assays, mature leaves of five healthy X. strumarium (height in 50 centimeters) were inoculated with fungal conidia by gently pressing surfaces of infested leaves onto leaves of healthy plants (Zhu et al. 2020). Five untreated plants served as controls. The controls and inoculated plants were separately maintained in greenhouses (humidity, 60%; light/dark, 16 h/8 h; temperature, 18°C). Eight days post-inoculation, signs of powdery mildew were detectable on inoculated plants, however, the controls were asymptomatic. Thus, the fungal pathogen was morphologically and molecularly identified and confirmed as P. xanthii. This powdery mildew caused by P. xanthii was previously reported on X. strumarium in Korea, Russia and India (Farr and Rossman, 2021). In addition, P. xanthii was recorded on X. strumarium in Xinjiang Province, China (Tai 1979). However, this is the first report of P. xanthii on X. strumarium in central China, where is around 3000 km away from Xinjiang Province with geographically differences. The sudden presence of powdery mildew caused by P. xanthii may adversely affect plant health and thus reduce medical value of X. strumarium. Therefore, the identification and confirmation of P. xanthii infecting X. strumarium enhance the knowledge on the hosts of this pathogen in China and will provide fundamental information for disease control in the future.

It is planted in about 10,000 ha in Zhejiang, Jiangsu, and Hubei provinces. In the past 3 years, a serious disease like southern blight occurred in hangju fields in Macheng, Hubei (31°17′14.45″ N, 115°03′13.167″ E). The disease mainly occurred in July after the rainy season. Disease incidence ranged from 30 to 50%, causing huge economic losses to growers. Disease symptoms appeared on the stem bases, followed quickly by wilting of the whole plants. White mycelia covered the stem, and numerous whitish to tan sclerotia developed on the colonized tissues. To determine the cause, plants with typical symptoms were collected from fields of Macheng and Wuhan, and isolates were obtained from the samples by culturing on potato dextrose agar (PDA) at 25°C. Mycelia grew radially over the whole plate after 3 days, and sclerotia formed after 5 days. Sclerotia were initially white and gradually turned brown as they matured. Mature sclerotia were globoid, ellipsoidal, or irregular in shape and measured 1.4 to 4.2 mm (2.78 ± 0.56 mm; n = 50) in diameter. The number of sclerotia per plate ranged from 61 to 133 (mean 87; n = 10). The hyphae formed septa, but no spores were observed. Genomic DNA was extracted from isolate J-12 using the Ezup Column Fungi Genomic DNA Purification Kit (Sangon Biotech, Shanghai, China). The 18S rDNA fragment was amplified using primers NS1 (5′-GTAGTCATATGCTTGTCTC-3′) and NS6 (5′-GCATCACAGACCTGTTATTGCCTC-3′) (Li et al. 2014) and sequenced by Sangon Biotech. The sequence (GenBank MN104721) was 99.85 and 99.98% similar to isolates of Athelia rolfsii (Curzi) C. C. Tu & Kimbr (JF819726.1 and AY665774.1, respectively). The isolate was also amplified with primers EF595F (5′-CGTGACTTCATCAAGAACATG) and EF1160R (5′-CCGATCTTGTAGACGTCCTG) for the translation elongation factor (TEF-1α) (Wendland and Kothe 1997) and sequenced. The TEF-1α sequence (MN106270) was 99.02% similar to A. rolfsii strain Sr_12 (JF267794.1). A phylogenetic tree constructed based on the TEF-1α sequence using the neighbor-joining method in MEGA6 (Tamura et al. 2013) showed that J-12 was very close to 15 GenBank accessions, all from A. rolfsii. Thus, J-12 was identified as A. rolfsii, corroborating the identification of Sclerotium rolfsii Sacc., the anamorph, based on cultural and morphological features. To demonstrate the pathogenicity, mature sclerotia were placed on the upper, middle, and lower parts of leaves (n = 6), stems (n = 6), and roots (n = 6) of heathy hangju seedlings rinsed with sterile water, and the inoculated stems, leaves, and roots were incubated in a moist chamber for 9 days at 25 ± 2°C (Mahadevakumar et al. 2018). Samples inoculated with sterile water were used as a control. Two days after inoculation, sclerotia began to germinate and infect plant tissues. Five days after inoculation, the detached stems were completely covered by mycelia, and leaves and roots were rotten. Sclerotia formed 6 days and matured 9 days after inoculation. In another experiment, 20-day-old healthy seedlings with nine true leaves were transplanted and grown in a growth chamber at 28 ± 2°C (n = 6). Stems were pierced with a sterile needle, and three of them were inoculated with 5 ml of J-12 mycelium suspension, and the other three seedlings were treated with 5 ml of sterile water as a control. After 3 days, mycelia began to grow at the base of inoculated stems. The surface of the stem base became black and withered, and the lower leaves of inoculated plants also became necrotic and wilted. The inoculated plants were completely wilted 10 days after inoculation. Fungal cultures were isolated from the inoculated sites and confirmed as S. rolfsii based on cultural and morphological features. First Report of Powdery mildew caused by Golovinomyces ambrosiae on Bidens pilosa in Guangdong Province, China

Abstract:Bidens pilosa L., an annual herbaceous plant with a wide distribution, possesses novel medicinal properties. In January 2021, a powdery mildew disease outbreak was documented on B. pilosa plants located in the roadside areas in Shenzhen, Guangdong Province, China, with 60 to 80% disease incidence. Initial symptoms manifested as small, irregular white powdery patches, primarily on the adaxial surfaces of leaves. Subsequently, the colonies expanded, forming coalescent colonies that spread across the leaves, petioles, and stems, eventually leading to the distortion and senescence of leaves. Hyphae are hyaline, flexuous to straight, septate, with thin walls and a width ranging from 2 to 8 μm. Hyphal appressoria are nipple-shaped. Conidophores are erect or slightly flexuous, ranging from 80 to 150 µm in length and 12 to 18 µm in width (n = 30). Typically, these conidophores bear chains of 2 to 5 immature conidia, displaying a sinuate outline. Foot-cells, located at the base of conidophores, are cylindrical and erect, approximately 33 to 100 µm in length and 6 to 10 µm in width (n = 30). Conidia are hyaline, ellipsoid-ovoid to barrel-shaped, and lack fibrosin bodies. Primary conidia are ellipsoid-ovoid in shape, characterized by a rounded apex and a subtruncate base, 25 to 40 µm × 15 to 22 µm in width. Secondary conidia are barrel-shaped with truncate or subtruncate ends, 27 to 35 µm × 15 to 20 µm in width. Germ tubes exhibit a longitubus pattern and are prominently produced at the perihilar or apical region of the conidia. No chasmothecia were observed in the collected samples. In order to conduct a molecular-level identification, mycelium and conidia were collected from B. pilosa leaves. Genomic DNA was subsequently extracted from these samples. The internal transcribed spacer (ITS), intergenic spacer (IGS) and beta-tubulin (tub2) sequences were performed using primer pairs ITS1/ITS4, IGS-12a/NS1R, and tub2, respectively (Carbone and Kohn 1999; Scholin et al. 1994; White et al.,1990). A 568-bp ITS, a 366-bp IGS, and a 354-bp tub2 sequences (GenBank accession nos. OR647592, OR978282 and OR978283) were obtained. The ITS sequence exhibited over 99.6% similarity to Golovinomyces ambrosiae (MT929773) and G. cichoracearum (MH590731). The IGS sequence displayed 100% similarity to G. ambrosiae (MK383490) and G. ambrosiae (OK349420). The tub2 sequence displayed 100% similarity to G. ambrosiae (MW981257) and G. ambrosiae (MW981256). Phylogenetic analysis of IGS, ITS and tub2 also grouped obtained sequences within the G. ambrosiae complex. Based on the analysis of morphological characteristics and sequence identity, the pathogen was identified as G. ambrosiae. In order to satisfy Koch's postulates, an infected leaf was carefully pressed onto leaves of six healthy young B. pilosa plants, each grown in a separate pot. Additionally, a control group consisted of six non-inoculated plants. All plants were placed in a greenhouse: 25°C, 14/10-h light/dark photoperiod, and relative humidity 50%. After 10 days, the inoculated leaves exhibited powdery mildew colonies similar to those observed in the original infected plants. At 16 days, the inoculated leaves exhibited discoloration and premature leaf drop. The pathogenicity test was conducted twice. Microscopic observation and sequencing confirmed that isolated fungus was identical to the original pathogen. G. ambrosiae has previously been documented on B. pilosa in Fuzhou, Fujian Province, China (Mukhtar et al., 2022). However, to the best of our knowledge, this study represents the first report of powdery mildew caused by G. ambrosiae on B. pilosa in Shenzhen, Guangdong Province, China.

Abstract:Umbilical cord blood transplantation (UCBT) is an expanding practice for both pediatric and adult patients. Rapid availability, low risk of infectious disease transmission, lower risk of graft-versus-host disease, and lack of risk for the donor makes UCB an attractive alternative source of hematopoietic stem cells for transplantation. We review the state of the art of pediatric and adult UCBT and important aspects of UCB banking. Current strategies to improve clinical results and expand access to UCBT to a larger number of adult patients are discussed. New approaches to enhance hematopoietic recovery by the use of accessory cells or direct intra-bone marrow injection are also reviewed.
First Report of Leaf Spot Blight of Camellia oleifera Caused by Diaporthe mahothocarpus in China

Abstract:Camellia oleifera Abel. is one of the native and important natural edible oil species in China. The cultivation of C. oleifera has vigorously increased in Guizhou Province in recent years. From June to August 2022, a severe leaf spot blight was observed on C. oleifera in Longli Plantation, with an incidence of 53.5% (n=200), which caused severe defoliation, negatively affected plant growth, and led to significant economic losses. Pale yellow and sub-circular leaf spots of 2-5 mm in diameter first appeared in the margin. The center of the spots then turned grey, and the edges turned brown. The symptomatic leaves gradually developed symptoms of blight with some brown acervular conidiomata, died, and fell off, with many deep black spots on the leaves (Fig. 1A-B). The fungal isolates GZU-Y2 and GZU-Y3 were obtained from the infected leaves of five-year-old symptomatic C. oleifera trees using the tissue isolation method, and a voucher specimen was deposited in the Forest Protection Laboratory, Guizhou University. Cultures grown on potato dextrose agar medium (PDA) were incubated at 28℃, 16L/8D. A round cream-like colony was formed on PDA, with a white surface, while the back gradually turned brown (Fig. 1C-E). The aerial hypha grew vigorously with an initial milky white color before turning grayish white. At 10 days after incubation, the pycnidia were dark brown to black and spherical, with a diameter of 563.3 μm (500 to 700) (n=20). The alpha conidia were unicellular, hyaline, aseptate, oval or fusiform and measured 6.1 μm (4.1 to 8.0) × 2.6 μm (1.9 to 3.6) (n=50). However, no beta conidia were observed (Fig. 1I). For further identification, total DNA from the pure culture was extracted using a DNA extraction kit (Sangon, Shanghai, China), and the internal transcribed spacer (ITS), translation elongation factor 1-α (TEF-1α), and beta-tubulin (TUB2) were amplified by PCR using the primers ITS1/ITS4 (White et al., 1990), EF1-728F/EF1-986R (Carbone and Kohn, 1999) and Bt2a/Bt2b (Glass and Donaldson, 1995), respectively, and sequenced for a BLASTn analysis and phylogenetic tree construction. The sequences of ITS, TEF-1α and TUB2 were deposited in GenBank as accession numbers OQ168242 (99.25%), OQ689451 (99.71%) and OQ689453 (100.00%) for GZU-Y2 and OQ674554 (99.25%), OQ689452 (99.71%) and OQ689454 (100.00%) for GZU-Y3, respectively. A phylogenetic tree (Fig. 2) was constructed with the software MEGA X using the Neighbor-Joining algorithm (Felsenstein, 1985). Based on its morphological and molecular characteristics, the pathogen was identified as Diaporthe mahothocarpus, one of the synonyms of D. eres and the teleomorph of Phomopsis mahothocarpi (Gao et al., 2014 and 2015; Chaisiri et al., 2021). A pathogenicity test was conducted by spraying spore suspensions (2 × 107 spores/mL) of isolate GZU-Y2 on the leaves of 20 pots of annual C. oleifera seedlings in vivo. The same number of control seedings were sprayed with sterile water. The seedlings were placed at a constant room temperature of 28°C, with the inoculation points wrapped in Parafilm for 5 d to retain moisture. After 10 d, typical symptoms appeared on the inoculated leaves (Fig. 1F-H), and the re-isolated fungal culture was identical in morphology and ITS sequence to that originally obtained, fulfilling Koch's postulates. To our knowledge, this is the first report of D. mahothocarpus causing leaf spot blight of C. oleifera in China. In our future work, we tend to study the green prevention and control of this disease.
First Report of Corynespora cassiicola Causing Septoria Leaf Spot on Scutellaria baicalensis in China

Abstract:Scutellaria baicalensis Georgi. is a perennial herb in the Lamiaceae family, with a distribution in more than 10 provinces in China. At the current time, the cultivation area of S. baicalensis in China exceeds 58,000 hectares, with annual production approaching 28,000 tons. As a traditional Chinese herbal medicine, the root of S. baicalensis has many applications, such as anti-inflammatory, anti-neuroinflammatory and neuroprotective, anticancer, antiviral, antibacterial, and antioxidant activities, and is effective in treatment of colitis, hepatitis, pneumonia, respiratory infections, and allergic diseases. (Jang et al. 2023; Liu et al. 2023). From August to September 2022, septoria leaf spot symptoms were observed at the Institute of Medicinal Plant Development (40.04°N, 116.28°E), Beijing, China, and the incidence of this disease was up to 20% in the field through more than two weeks of continuous investigation. Initial symptoms on leaves were observed as small, dark-brown spots (0.5 to 2.0 mm), which then expanded to irregular lesions with a pale gray center surrounded by a black ring with a dark-brown edge and light brown halo (Fig. 1A1-A3). Plants were defoliated and withered in severe cases. Thirty-six symptomatic leaves of 12 diseased plants from three experimental sites were cut into 5 × 5 mm pieces, and surface sterilized with 75% ethanol for 30 s followed by 5% NaClO solution for 45 s, rinsed with sterile water three times, dried with sterile filter paper, and subsequently placed on potato dextrose agar (PDA) medium and incubated at 25°C in dark for two days. Isolates were purified by transferring hyphal tips to new PDA plates and incubated at 25°C in dark. Finally, eight isolates (A1, B3, D1, F2, E2, a4, e4 and f1) with similar colonial morphological characteristics were obtained. Colonies on PDA exhibited dense, downy, and white to grayish-green aerial mycelia and the reverse of colonies showed dark-brown in the center and grayish on the edge (Fig. 1D, E). Conidia were solitary or catenate, pale brown, obclavate to cylindrical, apex obtuse (Fig. 1B, C). The isolates were divided into two categories by examining 100 conidia (50 of each isolate), represented by isolates D1 and e4. Conidia of D1 measured 5.4 to 75.8 μm × 2.1 to 6.8 μm, mean 26.9 × 4.4 μm, had 0 to 6 pseudosepta, with 0 to 3 pseudosepta observed in 88% of conidia. Conidia of e4 measured 20.3 to 103.4 μm × 2.0 to 7.9 μm, mean 41.9 × 4.8 μm, had 0 to 6 pseudosepta, with 2 to 5 pseudosepta observed in 90% of conidia. These isolates were identified as Corynespora cassiicola based on morphology (Ellis 1971). DNA of the two isolates (D1 and e4) was extracted by the cetyltrimethylammonium bromide (CTAB) method, and internal transcribed spacer (ITS) region of rDNA, translation elongation factor 1 alpha (TEF1-α), and beta-tubulin (TUB2) gene were amplified, using the primers ITS1/ITS4 (Bandi et al. 2022), EF1-728F/EF-986R (Wang et al. 2021), and Bt2a/Bt2b (Glass and Donaldson 1995), respectively. Sequences of ITS OQ991339 (524 bp) and OR044050 (533 bp) shared 99.8% identity to C. cassiicola, with a 99% coverage to MT228951 (536 bp) and OQ991340 (546 bp) in GenBank. Sequences of TEF1-α OR047441 (304 bp) and OR047443 (306 bp) shared 99.3% identity to C. cassiicola, with a 98% and 99% coverage to ON381927 (300 bp) and ON381933 (301 bp) in GenBank, respectively. Sequences of TUB2 OR047449 (427 bp) and OR047451 (427 bp) shared 99.53% identity to C. cassiicola, with a 99% and 98% coverage to MN604075 (442 bp) in GenBank, respectively. Phylogenetic trees were computed with ITS, TEF1-α, and TUB2 sequences in MEGA 11 using the Neighbor-Joining (NJ) method (Fig. 2). The results showed that the two isolates were C. cassiicola with more than 90% bootstrap support (1000 replicates). Nine 2-year-old seedlings of S. baicalensis were used for the pathogenicity assay. Three leaves from each plant were punctured with flame-sterilized needles, and inoculated with mycelial plugs (5 mm in diameter) of D1 and e4. Plants inoculated with sterile PDA plugs were used as control. All the inoculated seedlings were incubated at 25 oC and 90% relative humidity. About 3 to 4 days after inoculation, similar symptoms to those observed in the field were present on leaves inoculated with D1 and e4, while no symptoms were observed in the uninoculated control seedlings (Supplementary Fig. 1). Isolates with vigorous, downy, and white to grayish-green aerial mycelia were reisolated from the diseased leaves inoculated with D1 and e4 and identified as C. cassiicola by DNA sequencing, fulfilling Koch's postulates. Based on morphological and multilocus phylogenetic results, these isolates were identified as C. cassiicola, a pathogen that threatens several important crops (Dixon et al. 2009; Zhang et al. 2018; Xie et al. 2021). To our knowledge, this is the first report of C. cassiicola as the causal pathogen of septoria leaf spot on S. baicalensis in China, which poses a potential threat to the production of S. baicalensis.
First Report of Fusarium acuminatum Causing Dianthus chinensis root rot and foliage blight in China

Abstract:Dianthus chinensis is a popular ornamental plant that is widely cultivated in China. In May 2020, a disease was found at several landscape sites in Xuanwu District, Nanjing, China, causing symptoms of foliage blight and root discoloration on approximately 52% of one-year old D. chinensis plants. To recover the causal pathogen, samples of infected roots and leaves were cut into 5×5 mm2 pieces, surface-disinfected in 75% ethanol for 30 sec, followed by 1% NaClO for 90 sec, rinsed with sterile water three times and placed on potato dextrose agar (PDA) with 0.1 mg/mL of ampicillin at 25 ⁰C. Hyphae growing on PDA were visible from both root and leaf tissues after three days. Individual hyphal tips were transferred to new PDA plates to obtain pure isolates. Three representative isolates were deposited in the China Forestry Culture Collection Center (CFCC 57545,57546, 57547). The hyphae grew radially, densely, and the aerial hyphae were velvety, white, yellow-white, or pink-white. Representative isolate Facu-DCY5 produced three types of conidia (microconidia, macroconidia, and chlamydospores). Macroconidia were sickle-shaped, measuring 25.7-55.4 µm × 3.2-4.6 µm (n=50). Microconidia were numerous, oval or kidney-shaped, measuring 6.8-11.9 µm × 3.5-4.8 µm (n=50). Conidia produced in the aerial mycelium were 16-34 × 2.2-5.3 µm (n=50). The ITS region, TEF1, calmodulin (CMDA), and RNA polymerase II second largest subunit (RPB2) were amplified with primers ITS1/ITS4, EF1/EF2, CL1/CL2A and 5F2/7CR , respectively and sequenced at Sangon Biotech (Nanjing, China). The ITS sequence of isolate Facu-DCY5 (GenBank No. ON307073.1) was identical to HQ165938.1, ON306850.1, OM964482.1. TEF1 (ON331997.1) was identical to LC546967.1, HQ165866.1, MZ158155.1. CMDA (ON331996.1) was identical to HQ412345.1, MZ921595.1 and MZ921597.1. RPB2 (ON331995.1) was identical to MZ997370.1. Maximum parsimony and maximum likelihood phylogenies of the Facu-DCY5 multilocus sequence data and those of several species within the F. tricinctum species complex identified the isolate from D. chinensis as F. acuminatum . Pathogenicity tests were performed using a conidial suspension (104 conidia/mL). Each plant (approx. 0.3 m in -height) was inoculated with 1 mL of the conidial suspension by mixing it into the potting soil (500 g). Control plants were treated with sterile distilled water. All inoculated plants (n=9) in three repeats of the assay exhibited foliage blight and root rot after 15 days, whereas all control plants (n=9) remained asymptomatic. Fusarium isolates with identical morphological features and molecular marker sequences to those of Facu-DCY5 were recovered from foliage blight and root tissues of all the inoculated plants. In China, F. acuminatum has been reported as a pathogen of Cucurbita maxima, Actinidia arguta, Polygonatum odoratumand Schisandra chinensis. This is the first report of F. acuminatum on D. chinensis in China. Considering the importance of D. chinensis to both ornamental nurseries and landscaping industries, we recommend that diseased plants be removed to prevent the spread of F. acuminatum, and that identification of the infecting isolates from D. chinensis at other sites and landscape locations be performed.
First Report of Colletotrichum gloeosporioides Causing Leaf Blight on Cercis chinensis Bunge in China

Abstract:Cercis chinensis Bunge, commonly used as an ornamental plant, is native to southeastern China and extensively cultivated in gardens across major cities in the country. In August 2023, a new high-incidence disease was discovered at Huangshan University in Huangshan, Anhui Province, China. The symptoms initially began as small brown spots, which gradually expanded into large irregular brown spots with black-brown edges. The disease was investigated at both Jilingshan Park and Huangshan University, where C. chinensis Bunge was planted, revealing an average incidence rate of was 85 % at these sites. Seventy two leaf tissue samples (3 to 4 mm²) were collected from the margins of the lesion and subjected to surface sterilization with 75% ethanol for 30 seconds followed by 1% sodium hypochlorite for 90 seconds. Subsequently, the tissues were rinsed with sterile H2O, placed on potato dextrose agar (PDA) medium, and incubated at 25℃ for 5 days. The same fungus was isolated from 90% of the tissues, and pure cultures were obtained by monosporic isolation. Representative isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 were selected for morphological and molecular characterization. The colonies displayed a color range from white to gray, with white margins and aerial hyphae, while the reverse side of the colonies appeared gray to brown. Conidia were cylindrical, aseptate, with obtuse to slightly rounded ends, measuring 15.8±1.8×4.7±0.56 μm (n = 50). The morphological characteristics were generally consistent with those of Colletotrichum gloeosporioides species complex (Weir et al. 2012). Five conserved regions of isolates (ZJ 2-1, ZJ 2-2 and ZJ 2-3), including the internal transcribed spacer (ITS), glutamine synthase (GS), calmodulin (CAL), actin (ACT), and chitin synthase 1(CHS1) gene regions, were amplified using specific primers ITS1/ITS4 (Gardes et al. 1993), GSR1/GSF1 (Guerber et al. 2003), CL1C/CL2C (Li et al. 2018), ACT-512F/ACT-783R, and CHS-79F/CHS-345R (Zhu et al. 2019), respectively. Using the BLAST, ITS, GS, CAL, ACT and CHS1 gene sequences (GenBank accession nos. PP514751, PP448025, PP448026, PP448027 and PP448028, respectively) were 100% (594 out of 594 bp), 100% (864 out of 864 bp), 100% (299 out of 299 bp), 100% (732 out of 732 bp) and 100% (282 out of 282 bp) identical to C. gloeosporioides (GenBank accession nos. JX010152, JX010085, JX009818, JX009731 and JX009531, respectively). A Maximum Likelihood phylogenetic tree, constructed by combining all sequenced loci in MEGA7, showed that the isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 clustered within the C. gloeosporioides clade with 99% bootstrap support (Fig. S1). To fulfill Koch's postulates, five C. chinensis Bunge plants were tested for pathogenicity in the field with isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 at Huangshan University. Twelve leaves from each tree were wounded and inoculated with mycelial plugs (approximately 4 mm in diameter) and 10 μl of a spore suspension (1.0 × 106 conidia/ml) of C. gloeosporioides. Inoculation with sterile PDA plugs and pure water on leaves of each tree served as negative controls. Plastic bags were used to wrap the leaves, and sterile H2O was sprayed into the bags to maintain moisture conditions (Zhang et al.2020). The experiment was repeated two times, and within 5 days, all inoculated points displayed lesions similar to those observed in the field, whereas controls remained asymptomatic (Fig. S2). The same fungus was reisolated from these lesions with a frequency of 100%. Consequently, the pathogen responsible the disease in C. chinensis Bunge was identified as C. gloeosporioides. To the best of our knowledge, this is the first report of C. gloeosporioides causing leaf blight on C. chinensis Bunge in China. This study provides valuable insights for implementing targeted measures to control leaf blight on C. chinensis Bunge and lays a foundation for the prevention and treatment of the disease.
First Report of Erysiphe astragali Causing Powdery Mildew on Astragalus mongholicus in China

Abstract:Astragalus mongholicus Bge. [A. membranaceus Bge. var. mongholicus (Bge.) Hsiao] is a highly valuable perennial medicinal plant mainly distributed in China, whose dry roots are known as Huangqi in traditional Chinese medicine for reinforcing vital energy, strengthening superficial resistance, and promoting tissue regeneration (Lin et al. 2000). A. mongholicus roots of high quality are produced in Northwest and North China. Since July 2021, powdery mildew outbreaks happened annually on the leaves of A. mongholicus in a plantation (123° 56' 40'' E, 47° 22' 20'' N) in Qiqihar city, Heilongjiang Province, China. Disease incidence reached 100% by October (Fig. 1A-C), causing severe impairment of growth. Powdery mildew spots of circular or irregular shapes emerged on upper surface of leaf, resulting in plentiful lesion specks. Dense white hyphae appeared chaotically intertwined. Hyphae were hyaline and highly flexuous, 5.3 - 10.7 μm in diameter (n = 20). Chasmothecia were globose or slightly ovoid-shaped and turned dark brown when matured. Chasmothecia (diameter: 135.2 - 222.9 μm, n = 20) existed abundantly on the diseased leaves in the fields. Conidiophores were 89.0 - 129.9 μm in length (n = 20) and composed of one cylindrical, straight foot cell, followed by two cells and one to three conidia. Conidia were slim ellipsoid-shaped, occasionally ovoid-shaped, measuring 14.6 - 24.7 μm by 6.4 to10.4 μm, length/width ratio was 1.8 - 3.0 (n = 30). Hyphal appressoria were nipple-shaped and appeared in singular, occasionally in pairs. Unbranched germ tube emerged reaching out of the germinating conidia while forming an acute angle with the long axis. Comprehensively, the pathogen exhibited micro-morphology of the genus Erysiphe. For molecular identification, pathogen was carefully scraped off diseased leaves for DNA extraction. We used the DNA samples of three biological replicates for the sequencing of the ITS rDNA fragment (primers by (White et al. 1990). All the samples resulted in an identical ITS sequence (deposited in GenBank as OQ390098.1). It displayed 99.83% identity with OP806835.1 of an E. astragali voucher collected in Iran (Fig. 1D-M, O). Hence, our pathogen was identified as an E. astragali stain. Additionally, we amplified the Mcm7 sequence (using primers by (Ellingham et al. 2019), deposited as OQ397582.1). We propagated 40-day-old A. mongholicus plants via germinating seeds in pot soil and performed pathogenicity tests. Firstly, we incubated detached healthy leaves of propagated plants with severely symptomatic leaves collected from the fields in petri dishes under saturated moisture content and room temperature. Powdery mildew symptoms emerged on each healthy leaf (n = 5) after two weeks. Further, we infected healthy plants (n = 5) by gently pressing and rubbing symptomatic leaves on each healthy leaf, and kept them in a greenhouse (24 ℃, 80% humidity, 16/8-hour light/dark cycle). After a month, symptoms emerged on a number of leaves of each infected plant. We performed micromorphology observation (Fig. 1N-P) and ITS sequencing to confirm that the results fulfilled Koch's postulates. Powdery mildew caused by E. astragali on A. strictus in Tibet (Wang and Jiang 2023) and on A. scaberrimus in Inner Mongolia (Sun et al. 2023) have been reported. Here we report powdery mildew caused by E. astragali on Astragalus mongholicus for the first time. These Astragalus spp. are all acknowledged to have medicinal values in China but their usages are quite different.
First report of Diaporthe tulliensis causing leaf spot on Millettia speciosa in China

Abstract:Millettia speciosa Champ, renowned for its diverse applications in traditional medicine, is extensively cultivated in the Guangxi region of China, spanning roughly 5,973 hectares. In July 2021, a plantation in Yulin, Guangxi, China (22°64'N; 110°29'E), exhibited severe leaf spot disease on M. speciosa. Notably, a 46,690 square meters area had over 40% leaf spot incidence. Initially, symptoms appeared as small, circular, pale-yellow lesions on the leaves, then turned into irregular, dark brown spots with yellow halos, leading to the wilt and defoliation of leaves. To identify the responsible pathogen, a total of five symptomatic leaves were collected and sterilized systematically. Small tissue segments (5×5 mm) from lesion peripheries were aseptically excised, then surface sterilized with 75% ethanol for 10 s, and 1% sodium hypochlorite (NaClO) for 3 min. Following this, the sterilized tissues were triple-rinsed with sterile water and cultured on potato dextrose agar (PDA) at 28 °C in the dark for 7 d. A total of seven isolates were obtained through single-spore isolation, and one representative isolate, N2-3, was selected for further analysis. After 7 d of incubation, colonies displayed flat, white, and extensively branched aerial hyphae. Over time, the reverse side of the colony changed from white to yellowish-white. The pycnidia were black with conidial droplets ranging from cream to pale yellow exuding from their ostioles. The α-conidia were one-celled, hyaline, ovoid to cylindrical, typically with one or two droplets, 2.6 to 5.9 ×1.4 to 3.9 μm (n=50). These morphological traits align with those of the genus Diaporthe, as reported by Li et al. (2022) and Crous et al. (2015). To identify the species, isolate N2-3 underwent sequencing of the internal transcribed spacer (ITS), β-tubulin (BT), and translation elongation factor 1 alpha (EF1-α) sections (Huang et al. 2021). Obtained sequences of ITS, BT and EF1-α (Genebank accessions nos. OR600532, OR662169 and OR662168) displayed a 99% similarity to Diaporthe tulliensis (Genebank accessions nos. OP219651, ON932382, OL412437, respectively). Based on the concatenated ITS, BT and EF1-α, a neighbor-joining phylogenetic analyses using MEGA7.0 clustered with D. tulliensis. Therefore, the fungus was identified as D. tulliensis (teleomorph name) based on morphological and molecular features. A pathogenicity test was conducted on 1-year-old M. speciosa seedlings by gently abrading healthy leaves with sterilized toothpicks to create superficial wounds. Wounded leaves were then inoculated with 5 mm diameter mycelial plugs, while control seedlings received PDA plugs. Three leaves per plant and five plants per treatment were selected for assessment. All seedlings were kept in a controlled greenhouse (12/12h light/dark, 25 ± 2 °C, 90% humidity). After 7 d, the inoculated leaves showed symptoms like those in the field, while control plants remained healthy. The fungus was consistently reisolated from the infected leaves, satisfying Koch's postulates. Notably, D. tulliensis has caused Boston ivy leaf spot, bodhi tree leaf spot, cacao pod rot, and jasmine stem canker (Huang et al. 2021; Li et al. 2022; Serrato-Diaz et al. 2022; Hsu et al. 2023). This discovery is significant as it marks the first report of Diaporthe tulliensis causing leaf spot on Millettia speciossa in China, which has direct implications for the development of diagnostic tools and research into potential disease management strategies.

Abstract:Rather than speak of the precancer cell as a new problem in cancer research, perhaps we should refer to it as a “new opportunity” or new method of conducting functional research in the living subject. It is not the purpose of this paper to present the results of a concluded experiment but, rather, to present the broad panorama of research work still in progress utilizing this unique approach. ‘The introduction of exfoliative cytology into contemporary medicine by Doctor George N. Papanicolaou has been the springboard for new advances in techniques for early diagnosis. Of equal importance, though slow to be applied, is the new research approach to cancer of the cervix. It has now been widely recognized that the cervical smear or cell scraping will reveal with great accuracy the presence of carcinoma in situ in cervical tissues that may appear benign or completely normal to the naked eye. For some time, this was considered the earliest recognizable stage of cervical malignancy. The use of cervical cytology for routine cancer screening, however, especially when the surface cell biopsy technique is used, has revealed an in-between stage where the cells are neither normal nor malignant in appearance. These cells were reported by our group after the introduction of the cervical spatula ( I Y M ) , and were described as the “precancer cell complex.” This cell entity has remained highly controversial in diagnostic practice. I ts importance is greater in basic research, however, because of the opportunity afforded to explore carcinogenic influences and to examine the cell-transformation stage from benign to anaplastic. The advantage of the scraping technique in research is the consistency of the sample in serial cell studies. If one depends on physiological exfoliation, on the other hand, many variable factors influence the number of anaplastic cells per specimen; for example: the volume of cervical mucus, the point of collection of specimen, and the technique used. As a result, individual specimens will vary between positive, suspicious, doubtful, or atypical. Exploration by the research cytologist of this developing neoplasm enables precision studies of cell behavior and cell response before the potential c arcinoma reaches the dangerous stage. It is possible to trace premalignant changes back to their normal cell precursor, and true behavior patterns of regression, progression, and latency reveal the full duration and life history of epidernioid cancer. First Report of Fusarium Proliferatum Causing Root Rot Disease in Salvia Miltiorrhiza in China

Abstract:Salvia miltiorrhiza Bunge is a herb plant used as a traditional Chinese medicine to cure cardiovascular disease. In December 2018, a root rot disease was observed on S. miltiorrhiza in four surveyed counties (Song, Yuzhou, Fangcheng, and Mianchi) in Henan province in China. The disease incidence ranged from 15 to 50% in 12 surveyed fields. At the early stage, the diseased plants were wilting with purple leaves. Leaves and branches became withered and fibrous roots became brown and rotted. The main roots of severely diseased plants also became rotted. The color of the stem surface turned from red to black, and the color of the stem xylem and phloem turned from dark red to brown. Eventually, the roots of diseased plants became completely rotted and the whole plants became dead, but no stink, which is different from Fusarium solani (Mart.) Sacc. (Yuan et al. 2015). Diseased root tissues (5×5×5 mm in size) were cut from diseased plants, surface-sterilized with 1% sodium hypochlorite for 1 min followed by dipping in 75% alcohol for 30 sec, rinsed in sterile distilled water for 3 times, air-dried on a sterilized filter paper in a laminar flow hood, placed on potato dextrose agar (PDA) containing 250 mg/l of streptomycin sulfate, and incubated at 28℃. Five isolates of Fusarium were obtained and purified using the single-spore isolation method. On PDA plates, the colonies were purple in color with formation of white aerial mycelia and reached 50 to 60 mm in diameter after incubation for 5 days. The colonies produced abundant microconidia on the colonies. The microconidia were 4.3 to 12.3 (10.0) × 2.1 to 3.5 (3.1) μm in size (n = 40), hyaline, ovoid or ellipse in shape. The conidiogenous cells were polyphialides. On mung bean media, the isolates formed macroconidia with 3 to 6 septae, fusiform in shape, slightly curved, 21.8 to 32.7 (31.4) × 2.6 to 4.3 (3.4) μm in size (n = 50). The morphological features of the five isolates were consistent with the description for Fusarium proliferatum (Matsush.) Nirenberg ex Gerlach & Nirenberg (Leslie and Summerell 2006). To further define the identity of the five isolates, molecular phylogenetic analysis was performed. The genomic DNA was extracted from all five isolates using the cetyl trimethylammonium bromide (CTAB) method. Five genes [nuclear ribosomal internal transcribed spacer (ITS) region, translation elongation factor 1-α (EF1α), β-tubulin gene, partial sequence for calmodulin (PRO), and RNA-dependent DNA polymerase II subunit (RPB2)] in F. proliferatum were amplified using primers pairs ITS1/ITS4, EF1T/2T, β-tubulin 2a/b, PRO1/2, and RPB2F/R, respectively (Glass and Donaldson 1995; Liu et al. 1999; Mulè 2004; O'Donnell et al. 1998; O'Donnell et al., 2010). The sequences (GenBank accession numbers: MT371373, MT371384, MT925651, MT925652, and MT934441, respectively) showed 99.6 to 100% identities to the corresponding DNA sequences in F. proliferatum (GenBank Acc. Nos. MK243486, MN245720, KJ12896, MN245721, and MK144327, respectively). All five isolates were tested for pathogenicity to fulfill the Koch's postulates. The 45-day-old healthy plants of S. miltiorrhiza grown in sterilized soil in pots (20 cm in diameter), one plant in one pot, were inoculated with conidial suspensions (1.0 × 107 cfu/ml) by pouring 10 ml conidial suspensions around the stem base in one pot. For each isolate, four plants were inoculated. Four plants were treated with sterilized water in the same volume as a control. The tested plants were placed in a growth room at 25°C (RH > 60%) with a 12 h photoperiod of fluorescent light. The pathogenicity assay was repeated for three times. The similar wilt symptoms were observed on the roots in the inoculated plants 30 days after inoculation but were not observed in the control plants. F. proliferatum was re-isolated from the infected roots, and its identity was confirmed by PCR with the primers described above. To our knowledge, this is the first report of F. proliferatum casing root rot disease on S. miltiorrhiza in China.
