Abstract:Yanhusuo (Corydalis yanhusuo (Y. H. Chou & Chun C. Hsu) W. T. Wang ex Z.Y. Su & C.Y. Wu), a perennial herbaceous plant of the Papaveraceae family and genus Corydalis, is also known as Yuanhu and used as medicine by its tuberous roots. It is mainly planted in Zhejiang, Jiangsu and Anhui provinces of China, with the best quality produced in Panan County of Zhejiang province. Yanhusuo has the effects of promoting blood circulation, invigorating the flow of qi and relieving pain, and is widely used in Chinese traditional medicines. In surveys carried out in summer of 2020-2023, grey mold disease was found occurred on C. yanhusuo in Panan County. This disease begins at April, and lasts to July, with incidence of 20% to 70%. The diseased plants showed a large number of gray mold layers adhere to the leaves. When the disease infects from the leaf tips, it form V-shaped lesions; when the leaves are severely infected, the entire leaves die, shrink, curl, and have a large number of gray mold layers on the surface. To identify the causal agent of this leaf disease, diseased leaves were collected from Yanhusuo field at Panan County of Zhejiang province in China since 2020, and tissues at the junction of the healthy and diseased areas were cut off, disinfected with 75% ethanol for 30 seconds, rinsed with sterile water for 3 times for 1 minute each time, air-dried under sterile conditions, and then were inoculated in PDA medium and cultured in a 25℃ incubator. After 2-3 days, picked the edge hyphae of the fungi that grew on the PDA plate and cultured them on a new PDA plate. After 5 days, picked the single spore and inoculated on a new PDA plate for continuous cultivation until pure culture strains were obtained. Thirty strains were isolated from 30 samples that collected from 3 Yanhusuo fields in Panan County. One of the thirty purified strains was named "YH8" for further identification. When cultured on PDA medium, mycelia were initially whitish and turned gray with age. The hyphae accumulate into clusters, and no sclerotia are produced during the cultivation. The conidiophores are slender, septate. The base of the conidiophore is enlarged or slightly enlarged. The conidiophore often has branches and produces a large number of conidia, which are similar to grape clusters. The conidia are monosporous, ovoid, and colorless, 6.08 μm-12.76 μm×8.42 μm-19.34 μm, with an average size of 9.55 μm×14.50 μm. To further identify the species, YH8 genomic DNA was extracted, and the internal transcribed spacer (ITS), heat shock protein (HSP60), and glyceraldehyde-3-phosphate dehydrogenase (G3PDH) genes were amplified with the primers ITS1/4 (White et al. 1990), HSP60-F/HSP60-R, and G3PDH-F/G3PDH-R (Staats et al. 2005), respectively. A multilocus phylogenetic tree was constructed with the ITS, HSP60, and G3PDH reference sequences, and the sequences of PCR amplicons (Genbank number: PP388281, PP376066 and PP376067) were 100% (518 bp out of 518 bp), 99% (994 bp out of 995 bp) and 100% (880 bp out of 880 bp) identical to the Botrytis cinerea strain 5-3, respectively, and the grouping of strain YH8 was supported by 99% bootstrap value. To fulfill the Koch's postulates, spore suspension (approximately 103 CFU/mL) of YH8 was sprayed onto leaves of 3-week Yanhusuo seedlings, and sterile water was sprayed as negative control, 15 seedlings for each treatment, and the experiments was repeated for times. The seedlings were incubated in a growth chamber under 28℃ and 80% humidity. Seven days after inoculation, leaves of noninoculated controls were green and healthy, while the seedlings inoculated with spore suspension of YH8 showed lesions and molds, which were same with field symptoms. The causal pathogen was then reisolated from the lesions, and the gained pathogen showed same colony and spore morphology with YH8, which suggested the confirmation of Koch's postulates. Based on the morphological characteristics and molecular identification, the strain YH8 was identified as B. cinerea. To our knowledge, this is the first report of B. cinerea causing gray mold on the Corydalis yanhusuo in China. This report will provide guide to growers and local technicians for diagnostic and controlling grey mold disease of Yanhusuo.

Sw. is a perennial herbaceous plant with high gardening and herbal value. In August 2014, a severe anthracnose disease was observed on C. ensifolium leaves in Changchun (125.24°E, 43.48°N), Jilin Province, China. More than 80% of the plants were symptomatic. In the early stage of disease development, small brown lesions (1 mm in diameter) were visible on infected leaves, which subsequently developed into oval spots and turned light brown in the center. Lesions ranged from 5 to 15 mm in diameter. As lesions enlarged and coalesced, some leaves dried up. When the humidity was high, the surfaces of the lesions were covered with dark brown acervuli. Small pieces of infected leaves were surface-sterilized in 75% ethanol for 60 s, sterilized for 1 min in 1.5% NaClO, rinsed twice with sterilized distilled water, and plated on potato dextrose agar (PDA) in Petri dishes at 25 ± 1°C for 7 days. Ten fungal isolates were transferred from PDA to synthetic nutrient-poor agar (SNA). White colonies were observed in 2 days with smooth margins, and they covered the Petri dishes after 7 days. Perithecia (n = 100) were brown to dark brown, globose to subglobose, and measured 136.24 to 154.5 × 145.42 to 236.68 µm. Asci and ascospores were absent. Conidiophores and setae were formed directly on the media. Conidiophores (n = 100) were cylindrical, septate, branched, and measured 5.35 to 18.28 × 2.43 to 6.56 µm. Conidia (n = 100) were hyaline, aseptate, smooth, cylindrical, contents granular, and were 12.21 to 18.42 µm in length, and 4.40 to 7.90 µm in width. Setae (n = 100) were brown, cylindrical to conical, measuring 40.62 to 160.58 µm in length, and were 1 to 4 septate. Based upon these morphological and cultural characteristics, the causal agent was identified as Colletotrichum cymbidiicola. Five sequences of the internal transcribed spacer (ITS, KX058529), glyceraldehyde-3-phosphate dehydrogenase (GAPDH, KX068532), actin (ACT, KX058531), and beta-tubulin (TUB, KX058530) regions were obtained and deposited in GenBank. BLAST search results revealed that all sequences showed 99% identity with C. cymbidiicola sequences. Pathogenicity of 10 isolates of C. cymbidiicola from C. ensifolium were confirmed by inoculating healthy leaves of C. ensifolium plants with a spore suspension (1 × 106 spores/ml) produced on PDA. Each isolate was inoculated onto five plants and another five healthy plants were sprayed with sterilized water as controls. All plants were enclosed in plastic bags for 48 h in a greenhouse at 28°C and 12 h/day light cycle. After 7 days, inoculated plants showed similar symptoms to those on the original diseased plants, while control plants remained symptomless. C. cymbidiicola was successfully reisolated from symptomatic plants to fulfill Koch’s postulates. C. cymbidiicola causing disease was reported on cymbidium in Australia, New Zealand, Japan (Damm et al. 2012; Hou et al. 2016), and India (Cannon et al. 2012). First Report of Phytophthora Nicotianae Causing Blight of Dendrobium Candidum in Zhejiang Province, China

Abstract:Dendrobium candidum is a traditional Chinese medicinal herb, used raw or processed for health-care products in recent years. Due to the high economic value, there is huge demand and over-collecting of this plant, and hence the resources of wild D. candidum are endangered. In order to meet the increasing demands, tissue cultured seedlings are grown on large scale fields by local enterprises. Since 2001, a root and basal stem rot and die-back disease has been observed on D. candidum in Yiwu city, Zhejiang province, China. In the field, 1-year-old affected transplanted seedlings demonstrate typical symptoms of Phytophthora root and basal stem rot. Symptom progression is acropetal and plants die within a few days. Tap roots are completely rotted with black-brown to black sunken lesions frequently extending up the stem. In contrast, 2- to 3-year-old infected plants only have die-back symptoms with almost no root and basal stem rot. Typical black-brown lesions are observed on the tender top parts of a plant, causing top wilt and defoliation. Dendrobium candidum blight has not been observed in the wild. Five isolates of a Phytophthora species from diseased material were initially identified as Phytophthora nicotianae using morphological features (Stamps et al., 1990; Erwin & Ribeiro, 1996). No oospores were observed in the tissue. Mating types were determined by pairing known P. nicotianae tester strains. Oospores were produced following pairing tester strains and all isolates proved to be of the A2 mating type. PCR amplification of genomic DNA using the primers ITS6 and ITS4 generated sequences of approximately 890 bp. The sequences were submitted to the GenBank database (Acc. No. EF140984–88). Sequence analysis showed that the ITS sequences obtained were identical with others obtained from P. nicotianae (Estevez de Jensen et al., 2006), thus confirming the identity of the pathogen. Mycelial plugs of the five Phytophthora isolates taken from agar medium, were inoculated into the basal stem of 2-month-old transplanted seedlings grown in pasteurized potting substance (mainly chipped pine bark) and to the top parts (new shoots) of 2-year-old plants. Plants were grown in the greenhouse at 25°C, where they developed typical disease symptoms after 7 days. Non-inoculated controls did not develop disease symptoms. The pathogenic isolate was re-isolated from all diseased plants. Isolates have been deposited at the Biotechnology Institute, Zhejiang University. This is first record of phytophthora blight of D. candidum in China.

First Report of Green Mould on Leaf of Phaseolus vulgaris Caused by Cladosporium tenuissimum in Liaoning, China

Abstract:Phaseolus vulgaris Linn. is a widely cultivated vegetable throughout the world. From spring 2019 to 2022, green mould symptoms were observed on leaves of P. vulgaris in the greenhouse in Liaoning, China, with disease incidence of 8-75% (plants) and 6-23% (leaves). Symptoms appeared as chlorotic lesions covered with dark green mould. The infections started at the apex or margin of the leaves and then spread inward with a characteristic "V" shape. Lesions exhibited curly morphology. 15 leaf samples with typical symptoms were collected from 5 different greenhouses. A total of 75 (5 replicates of each sample) leaf tissues (0.5 cm × 0.5 cm) were selected from the boundary between diseased and healthy parts. These samples were surface sterilized in 0.5% NaClO formin, rinsed 3 times in sterile distilled water and subsequently incubated at 28℃ on potato dextrose agar (PDA) supplemented with streptomycin (50 μg/ml). Numerous morphologically uniform colonies had been purified, with no other fungi observed. Afterwards, the strains were subcultured on malt extract agar (MEA). Colonies on MEA reached 70 to 80 mm diam after 14 days, smoke-grey to pale olivaceous-grey, woolly, sometimes radially wrinkled. The mycelia were pale olivaceous-grey, with hyphae measuring 1-5 μm wide (n = 20). The conidiophores were solitary or in groups of 2 to 5, and measured 50-280(-350) × 2.5-4 μm (n = 20), with 2-7 septa. The conidiogenous cells exhibited a cylindrical-oblong morphology and measured 10-44 × 5 μm (n = 20), with 0-2 septa, and the loci frequently thickened. The conidia were catenate in densely branched chains, ellipsoid to obovoid, smooth, and measured 2.5-5 × 2-3 μm (n = 50), with 0-4 septa. The morphological characteristics were similar to Cladosporium tenuissimum (Zhang 2003). The representative isolate KZ-19 was selected for molecular identification. The rDNA-ITS, translation elongation factor 1-α and actin genes were amplified, sequenced, and the resulting sequence data were submitted to GenBank (ITS: OQ931048; EF-1α: OQ954495; ACT: OQ954496). The BLAST results exhibited a 99 to 100% similarity with the sequences of C. tenuissimum type strain CBS 125995(ITS: HM148197; EF-1α: HM148442; ACT: HM148687). Furthermore, a multi-locus phylogenetic tree was constructed using the PhyloSuite (v 1. 2. 2) software, which revealed that the strains were most closely related to C. tenuissimum (Zhang et al. 2020). Based on both morphological and molecular characteristics, KZ-19 was finally identified as C. tenuissimum (Bensch 2012). Pathogenicity testing was performed on healthy 1-month-old P. vulgaris plants by inoculating the spore suspension (1×106 conidia/ml) of KZ-19 onto leaf surfaces, while control plants were simulated inoculated with sterile water, and five pots were used for each treatment. The test was performed under field conditions of 16-28°C (temperature) and 24-56% (relative humidity). Chlorotic lesions became evident within 2 days of inoculation, followed by the appearance of green mold on leaves after 7 days. No symptoms were observed in the control group. To fulfill Koch's postulates, the pathogen was re-isolated from three inoculated leaves. The morphological identification of re-isolated pathogens was similar to that of originally isolated pathogens. No infection was observed in non-inoculated control. To the best of our knowledge, this is the first report of C. tenuissimum causing green mould on P. vulgaris. As a ubiquitous saprobic hyphomycete, C. tenuissimum has been implicated in leaf mold in Punica granatum and Trifolium repens, larch bud blight, and strawberry blossom blight in previous years (He et al. 1987; Zhang et al. 2003; Zheng et al. 2010; Nam et al. 2015), presenting a potential threat to numerous crops. Therefore, an investigation of its distribution and pathogenic potential is essential in addition to the development of effective disease management strategies.
First Report of Powdery Mildew Caused by Blumeria graminis f. sp. bromi on Bromus catharticus in China

Abstract:Bromus catharticus, rescuegrass, is a brome grass that has been cultivated for herbage production, and been widely naturalized in many provinces of China, including Henan province. During April and May 2020, powdery mildew was found on leaves of Br. catharticus on the campus of Henan Normal University, Xinxiang city (35.3°N; 113.9°E), Henan Province, China. Abundant white or grayish irregular or coalesced circular powdery colonies were scattered on the adaxial surface of leaves and 70% of the leaf areas were affected. Some of the infected leaves either were chlorotic or senescent. About 60% of the observed plants showed powdery mildew symptoms. Conidiophores (n = 25) were 32 to 45 μm × 7 to 15 μm and composed of foot cells and conidia (mostly 6 conidia) in chains. Conidia (n = 50) were 25 to 35 μm × 10 to 15 μm, on average 30 × 13 μm, with a length/width ratio of 2.3. Chasmothecia were not found. Based on these morphologic characteristics, the pathogen was initially identified as Blumeria graminis f. sp. bromi (Braun and Cook 2012; Troch et al. 2014). B. graminis mycelia and conidia were collected, and total genomic DNA was extracted (Zhu et al. 2019). The rDNA internal transcribed spacer (ITS) region was amplified with primer pairs ITS1/ITS4. The amplicon was cloned and sequenced. The sequence (574 bp) was deposited into GenBank under Accession No. MT892940. BLASTn analysis revealed that MT892940 was 100% identical to B. graminis f. sp. bromi on Br. catharticus (AB000935, 550 of 550 nucleotides) (Takamatsu et al. 1998). Phylogenetic analysis of MT892940 and ITS of other B. graminis ff. spp. clearly indicated least two phylogenetically distinct clades of B. graminis f. sp. bromi and that MT892940 clustered with the Takamatsu vouchers. Leaf surfaces of five healthy plants were fixed at the base of a settling tower and then inoculated by blowing conidia from diseased leaves using pressurized air. Five non-inoculated plants served as controls. The inoculated and non-inoculated plants were maintained separately in two growth chambers (humidity, 60%; light/dark, 16 h/8 h; temperature, 18℃). Thirteen- to fifteen-days after inoculation, B. graminis signs and symptoms were visible on inoculated leaves, whereas control plants remained asymptomatic. The pathogenicity assays were repeated twice with the same results. The observed signs and symptoms were morphologically identical to those of the originally infected leaves. Accordingly, the causal organism of the powdery mildew was confirmed as B. graminis f. sp. bromi by morphological characteristics and ITS sequence data. B. graminis has been reported on Br. catharticus in the United States (Klingeman et al. 2018), Japan (Inuma et al. 2007) and Argentina (Delhey et al. 2003). To our best knowledge, this is the first report of B. graminis on Br. catharticus in China. Since hybridization of B. graminis ff. spp. is a mechanism of adaptation to new hosts, Br. catharticus may serve as a primary inoculum reservoir of B. graminis to infect other species (Menardo et al. 2016). This report provides fundamental information for the powdery mildew that can be used to develop control management of the disease in Br. catharticus herbage production.
First report of leaf spot caused by Botrytis cinerea on Bletilla striata in Anhui province, China

Abstract:Bletilla striata (Thunb.) is a perennial herb plant of the orchidaceous family and is used as an ornamental plant in Europe and the United States. Furthermore, it is important as traditional Chinese medicine (TCM) in East Asian countries, such as China, Japan, Korea, Mongolia, and Myanmar (Gou et al. 2022). In April 2023, a severe disease similar to gray mold occurred in a B. striata plantation in Anqing, Anhui province, China (N30°27'15″, E116°18'32″), causing disease on about 20% of the plants in the field. Early symptoms were characterized by brown spots or stripes on the leaves of B. striata, and as the disease progressed, large brown irregular spots appeared. Eventually disease spots coalesced, covering the entire leaf surface and causing leaf death. A gray mildew layer was observed on the senescent leaves. To investigate the causal agent, 10 plants with typical symptoms were collected from the field. Leaf pieces (5 × 5 mm) from the border of infected areas were soaked in 75% ethanol for 10 seconds, and then transferred into 0.1% mercury bichloride for three min, rinsed three times with sterile water, and transferred to PDA at 25 °C for three days. Pure cultures were obtained by single spore isolation, and the resulting colonies were morphologically similar, indicating a single pathogen, of which the representative BSFC-7 was selected for further study. BSFC-7 colonies were initially white to gray-brown, and cottony aerial hyphae grew over the entire petri dish after five days of incubation. Grayish, branched conidiophores and their terminal unicellular conidia were observed under a microscope after additional two days at 25 °C. Conidia were colorless or gray, elliptical or oval, and 7.06-12.54 × 8.33-13.55 μm (n=30). Sclerotia appeared in BSFC-7 culture up to about two weeks and were black, hard, and round or irregularly shaped (0.81-4.32 × 0.97-5.68 mm, n=20). The morphological characteristics fit the description of Botrytis cinerea (Li et al. 2016). To further identify the species, genomic DNA of BSFC-7 was extracted. PCR analysis was performed with species-specific primer pairs C729+/C729- and two nuclear genes G3PDH and RPB2 with their corresponding primer pairs G3PDH-F/G3PDH-R and RPB2-F/RPB2-R (Rigotti et al. 2002; Aktaruzzaman et al. 2018). The sequences for all three PCR products of C729, G3PDH, and RPB2 (GenBank accession nos. OR287069, OR255923, and OR255924 respectively) exhibited 99 to 100% similarity with other B. cinerea isolates. In the pathogenicity test, detached leaves of B. striata were inoculated with the BSFC-7 isolate. The leaves were soaked in sodium hypochlorite (1%) for two min, washed with sterile distilled water, and then inoculated with 10 µl of conidial suspension (106 conidia/ml). Sterile water was used as control and samples were incubated at 25 °C. After three days, all leaves inoculated with conidia showed dark brown water-soaked lesions similar to those observed in the field, while the control leaves remained healthy. The pathogen was re-isolated from the affected leaves, fulfilling Koch's postulates. B. cinerea is a common pathogen on a wide range of host plant species worldwide and has been reported to infect B. striata in Yunnan province, China (Romanazzi and Feliziani 2014; Zhang et al. 2020). To our knowledge, this is the first report of B. cinerea causing leaf spots on B. striata in Anhui province, China. This study will provide a basis for controlling the prevalence and economic losses of gray mold on B. striata.
First Report of Fusarium Annulatum Causing Blight on Bletilla Striata (baiji) in China.

Abstract:Bletilla striata (Thunb. ex A. Murray) Rchb (known as baiji in Chinese), a herbal plant distributed mainly in China, has become a focus of scientific attention recently due to its medicinal value (He et al. 2017). In May 2023, blight symptoms on leaves and stems were observed approximately 60% of Bletilla striata in Hangzhou, Zhejiang, China (29.80° N, 119.67° E). Brown spots initially appear on the infected leaves, which gradually decay as the spots expand. The wilting is accompanied with fading and yellowing, and eventually leading to defoliation. The infected stem initially appears brown spots, which gradually decay as the spots expand, resulting in the death of the whole plant, affecting the yield and quality of the herbs ultimately. To isolate the pathogen, small symptomatic leaves and stems (5×5 mm) were surface-disinfected with 75% ethanol for 30 s and 1% NaClO for 2 min, then rinsed in distilled water 3 times. Subsequently, the disinfected tissues were placed on PDA and incubated at 27 ℃ for 3 days. A total of 8 fungal isolates with similar morphological characteristics were obtained. The colony by single-spore purification was light purple to dark purple with abundant aerial mycelium. Macroconidia were relatively slender with a curve, mainly three to five septate and measuring 24.34 to 54.64 μm (average 40.29 μm) × 3.59 to 5.45 μm (average 4.49 μm) (n=30). Microconidia appeared obovoid to pyriform, with sizes of 5.31 to 8.43 μm (average 7.12 μm) × 2.30 to 4.29 μm (average 3.22 μm) (n=30). The morphological characteristics were consistent with Fusarium annulatum (Yilmaz et al. 2021). To further confirm the isolate's identification, the genomic DNA of isolates were extracted and identified by phylogenetic analyses of multilocus sequences of the RNA polymerase largest subunit (rpb1, primers Fa and G2R), RNA polymerase second largest subunit (rpb2, primers 7cf and 11ar) and the translation elongation factor 1-alpha (tef1, primers EF1 and EF2) (O'Donnell et al. 2022). The sequences were deposited in GenBank (rpb1: OR493933, OR493934, OR753402; rpb2: OR753398, OR753399, OR753400; tef1: OR493935, OR493936, OR753401). BLAST searches of the rpb1, rpb2, and tef1 sequences revealed 99.83% (1775/1778 nt), 99.79% (957/959 nt), and 98.98% (678/685 nt) homology with those of Fusarium annulatum CBS:258.54 from New Caledonia (rpb1: MT010944; rpb2: MT010983; tef1: MT010994). To confirm pathogenicity, one-year-old B. striata leaves and stems were disinfected with 75% ethanol, wounded with a sterile syringe on 3 healthy leaves and stems, inoculated with 5 × 5 mm mycelial discs of strain BJ-L1 and BJ-S1, respectively. And the control were treated similarly except that they were inoculated with PDA discs. The experiment was replicated 3 times. After 5 days, all inoculated leaves and stems showed similar symptoms to those initially observed on infected plants. The same pathogen was re-isolated and identified by morphological characterization and molecular analysis, confirming Koch's postulates. Thus, the pathogen causing blight of B. striata was determined to be F. annulatum. To our knowledge, this is the first report of F. annulatum causing blight on B. striata in China. F. annulatum has a wide range of hosts and has been reported to infect a wide range of crops, fruits and vegetables (Bacon et al. 1991). This study provides the basis for further research on this disease and is important for the management of this disease and the improvement of the economic benefits of B. striata.
First Report of Colletotrichum gloeosporioides Causing Leaf Blight on Cercis chinensis Bunge in China

Abstract:Cercis chinensis Bunge, commonly used as an ornamental plant, is native to southeastern China and extensively cultivated in gardens across major cities in the country. In August 2023, a new high-incidence disease was discovered at Huangshan University in Huangshan, Anhui Province, China. The symptoms initially began as small brown spots, which gradually expanded into large irregular brown spots with black-brown edges. The disease was investigated at both Jilingshan Park and Huangshan University, where C. chinensis Bunge was planted, revealing an average incidence rate of was 85 % at these sites. Seventy two leaf tissue samples (3 to 4 mm²) were collected from the margins of the lesion and subjected to surface sterilization with 75% ethanol for 30 seconds followed by 1% sodium hypochlorite for 90 seconds. Subsequently, the tissues were rinsed with sterile H2O, placed on potato dextrose agar (PDA) medium, and incubated at 25℃ for 5 days. The same fungus was isolated from 90% of the tissues, and pure cultures were obtained by monosporic isolation. Representative isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 were selected for morphological and molecular characterization. The colonies displayed a color range from white to gray, with white margins and aerial hyphae, while the reverse side of the colonies appeared gray to brown. Conidia were cylindrical, aseptate, with obtuse to slightly rounded ends, measuring 15.8±1.8×4.7±0.56 μm (n = 50). The morphological characteristics were generally consistent with those of Colletotrichum gloeosporioides species complex (Weir et al. 2012). Five conserved regions of isolates (ZJ 2-1, ZJ 2-2 and ZJ 2-3), including the internal transcribed spacer (ITS), glutamine synthase (GS), calmodulin (CAL), actin (ACT), and chitin synthase 1(CHS1) gene regions, were amplified using specific primers ITS1/ITS4 (Gardes et al. 1993), GSR1/GSF1 (Guerber et al. 2003), CL1C/CL2C (Li et al. 2018), ACT-512F/ACT-783R, and CHS-79F/CHS-345R (Zhu et al. 2019), respectively. Using the BLAST, ITS, GS, CAL, ACT and CHS1 gene sequences (GenBank accession nos. PP514751, PP448025, PP448026, PP448027 and PP448028, respectively) were 100% (594 out of 594 bp), 100% (864 out of 864 bp), 100% (299 out of 299 bp), 100% (732 out of 732 bp) and 100% (282 out of 282 bp) identical to C. gloeosporioides (GenBank accession nos. JX010152, JX010085, JX009818, JX009731 and JX009531, respectively). A Maximum Likelihood phylogenetic tree, constructed by combining all sequenced loci in MEGA7, showed that the isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 clustered within the C. gloeosporioides clade with 99% bootstrap support (Fig. S1). To fulfill Koch's postulates, five C. chinensis Bunge plants were tested for pathogenicity in the field with isolates ZJ 2-1, ZJ 2-2 and ZJ 2-3 at Huangshan University. Twelve leaves from each tree were wounded and inoculated with mycelial plugs (approximately 4 mm in diameter) and 10 μl of a spore suspension (1.0 × 106 conidia/ml) of C. gloeosporioides. Inoculation with sterile PDA plugs and pure water on leaves of each tree served as negative controls. Plastic bags were used to wrap the leaves, and sterile H2O was sprayed into the bags to maintain moisture conditions (Zhang et al.2020). The experiment was repeated two times, and within 5 days, all inoculated points displayed lesions similar to those observed in the field, whereas controls remained asymptomatic (Fig. S2). The same fungus was reisolated from these lesions with a frequency of 100%. Consequently, the pathogen responsible the disease in C. chinensis Bunge was identified as C. gloeosporioides. To the best of our knowledge, this is the first report of C. gloeosporioides causing leaf blight on C. chinensis Bunge in China. This study provides valuable insights for implementing targeted measures to control leaf blight on C. chinensis Bunge and lays a foundation for the prevention and treatment of the disease.
First Report of Botrytis cinerea Causing Stem Rot on Hydrangea macrophylla in China

Abstract:Hydrangea (Hydrangea macrophylla), commonly referred to as big leaf hydrangea, is a species within the Hydrangeaceae family notable for its ornamental value. Characterized by its vividly colored sepals and lush, striking inflorescences, this species is globally esteemed as both a potted and landscape plant. Notably, in 2022, an alarming incidence of stem rot was observed in approximately 40% of H. macrophylla plants aged between six and twelve months within 16 greenhouses situated in Nanjing City (N 31°14', E 118°22'), Jiangsu Province, China. Initial symptoms of the disease manifested as wet gray-black spots at the base of the seedlings and stems, progressing to a necrotic gray-white discoloration in the stems and accompanied by the growth of gray mold on the affected parts. This infection ultimately led to the wilting of the leaves and the death of the seedlings. For pathogen identification, stem tissues at the interface of diseased and healthy sections were excised, surface-sterilized with 75 % ethanol for 30 s, followed by a 2 - 3 min treatment with 3% sodium hypochlorite, and subsequently rinsed three times with sterile water before air drying. Sections measuring 2 - 3 mm were then cultured on potato dextrose agar (PDA) medium, supplemented with 50 mg/mL rifampicin (RFP), and incubated at 25 ℃ for 3 - 5 d (Zhou et al. 2022). Upon 2 - 3 days of incubation, notable growth of fungal colonies was observed. Mycelial clusters from the periphery of these colonies were subsequently transferred to fresh PDA plates and incubated at 25 ℃ for an additional 5 - 7 d. A particular colony, designated JSNJ2022-2 and now preserved at the Jiangsu Academy of Agricultural Sciences, was selected for detailed examination. This colony exhibited a flocculent texture, with a coloration ranging from grey-white to light brown. It was characterized by the presence of irregularly formed, hard sclerotia within the hyphae. The conidiophores were observed to be slender and erect, featuring dendritic branches at their extremities. The conidia were clustered on the conidiophore like grapes. These conidia were generally colorless or grey, oval in shape, smooth and transparent, and measured between 6.4 - 12.2 × 7.3 - 18.2 μm (n = 50). For genetic analysis, genomic DNA (gDNA) was extracted using the DNA secure Plant Kit (Tiangen Biotech, Beijing, China). Polymerase chain reaction (PCR) amplification was performed using a set of universal primers of ITS1/ITS4 (White et al. 1990), primers corresponding to the specific sequences of glyceraldehyde-3-phosphate dehydrogenase (G3PDH), heat-shock protein 60 (HSP60), and DNA-dependent RNA polymerase subunit II (RPB2) (Yang et al. 2020). The resultant PCR products were sequenced, and the resulting sequences were submitted to the GenBank database, under the accession numbers OP131597, OP142320, OP142321, and OP142322, respectively. BLAST analysis of the sequences obtained from the isolate JSNJ2022-2 revealed a high degree of genetic similarity, ranging from 99 to 100%, with known sequences of Botrytis cinerea (accessions MK051124.1, MH796662.1, MH479931.1, and KU760986.1). To elucidate the phylogenetic position of the isolate, a phylogenetic tree was constructed using the maximum likelihood method, supported by 1,000 bootstrap replications, in the Mega7 software (Kumar et al. 2016). The results of this analysis confirmed that the strains under study clustered within the same branch as B. cinerea. To establish the pathogenicity of the isolate, Koch's postulates (Falkow 1988) were employed. Healthy potted H. macrophylla seedlings, approximately three months old, were wound inoculated at the base of the seedlings with a 6 mm diameter mycelium plug of JSNJ2002-2 cultivated on PDA for 3 days, which was subsequently covered with moistened degreasing cotton. Control plants were treated with moistened degreasing cloths minus the pathogen. Post-inoculation, these plants were placed in a growth chamber maintained at 25 ℃ with a relative humidity range of 60 - 80%. After a 3-d incubation period, the inoculated plants displayed symptoms identical to those initially observed in the greenhouse. The pathogen was successfully re-isolated from these inoculated plants and was morphologically re-confirmed as B. cinerea, thus satisfying the criteria of Koch's postulates. To our knowledge, this report represents the first documented incidence of B. cinerea causing stem rot in H. macrophylla in China.

First Report of Apiospora arundinis Causing Leaf Spot on Polygonatum cyrtonema Hua in China

Abstract:Polygonatum cyrtonema Hua, which is a perennial herb of Liliaceae, can be used as clinical Chinese medicine for treating diabetes and asthma. It is widely cultivated in China, with 700 ha planted in Tonggu County, Jiangxi province (Chen et al. 2022). In June 2022, leaf spot symptoms were observed on P. cyrtonema in Tonggu county (28°71'42″N, 114°56'19″E), and the disease incidence was estimated to be above 35%. In the early stages of infection, small brown spots appear on the edge or tip of the leaves. As the lesion matures, the spots gradually expand to form wedge-shaped or elliptic to irregular lesions with brown edges and yellow halos. To identify the pathogen species, leaf pieces (5 × 5 mm) from the lesion borders were surface sterilized in 75% ethanol for 30 s, followed by 2% NaClO for 2 min, then rinsed with sterile distilled water for 3 times and dried with sterile filter paper. The tissues were placed on potato dextrose agar (PDA) and incubated at 25°C. Pure cultures were obtained by monosporic isolation, and the representative isolates, HJYB2-1, HJYB2-2 were used for morphological studies and phylogenetic analyses. Colonies on PDA of the two isolates were white with fluffy aerial mycelia. The hyphae were smooth, hyaline, and branched. The conidiophores were hyaline or pale brown and produced conidiogenous cells. The conidiogenous cells were pale brown, smooth, and ampulliform, 5.8-11.7 × 3.0-4.9 μm (n=50). The conidia were brown, smooth, and ellipsoidal to spherical, 4.7-7.3 × 2.3-4.5 μm (n=50). Morphological features were similar to Apiospora arundinis species complex (Crous et al. 2013, Pintos et al. 2021). For molecular identification, the ITS, TUB2 and TEF 1-α genes were amplified with the primer pairs ITS1/ITS4, T1/Bt-2b and EF1-728F/EF-2 (White et al. 1990), respectively. The generated sequences were deposited in GenBank with accession numbers OR229623, OR229624 (ITS), OR260104, OR260105 (TUB2), and OR290789, OR290790 (TEF1-α). Maximum likelihood and Bayesian posterior probability analyses using IQtree v. 1.6.8 and Mr. Bayes v. 3.2.6 with the concatenated sequences placed HJYB2-1 and HJYB2-2 in the clade of A. arundinis. The two isolates were identified as A. arundinis based on morphological and multilocus phylogenetic analyses. For pathogenicity testing, healthy leaves of six P. cyrtonema plants (five leaves each, n=30) in the field were pin-pricked with a sterile needle and inoculated with 100 µL spore suspension on the wound (1×106 conidia per mL). Healthy leaves of another three P. cyrtonema plants (five leaves each, n=15) in the field were pin-pricked with a sterile needle, inoculated with sterile distilled water and served as the control. All the inoculated leaves were covered with plastic bags to keep a high-humidity environment with a temperature of about 28℃. The test was repeated three times. More than 90% of inoculated leaves showed similar symptoms to those observed in the field, whereas control leaves were asymptomatic for 7 days. Apiospora arundinis was reisolated from the leaf lesions on the inoculated plants. No pathogenic fungus was isolated from the control leaves. A. arundinis has been reported causing disease on Camellia sinensis (Thangaraj et al. 2019), Prunus persica (Ji et al. 2020), Saccharum officinarum (Liao et al. 2022) but has not previously been reported causing disease on P. cyrtonema. To our knowledge, this is the first report that A. arundinis can cause leaf spot on P. cyrtonema in China. Our result should help with future monitoring and control of this disease.
Prevention of Spinal Anesthesia-Induced Hypotension in the Elderly: Comparison Between Preanesthetic Administration of Crystalloids, Colloids, and No Prehydration

Abstract:The practice of routinely prehydrating patients by infusing a crystalloid or colloid solution (up to 1.0 L/70 kg) for prevention of spinal anesthesia-induced hypotension has been challenged recently, after several reports of failure to demonstrate its efficacy in young women. We compared the incidence and frequency of hypotension and vasopressor therapy after spinal anesthesia and no prehydration with crystalloid and colloid prehydration in elderly patients. Eighty-five ASA grade I or II patients (aged 60-89 yr) for elective total hip replacement were randomized to receive 500 mL crystalloid solution (Hartmanns' [TM], n = 29), 500 mL colloid (Haemaccel [TM], n = 28), or no prehydration (n = 28) over 10 min prior to spinal anesthesia. Hypotension was defined as a 30% decrease from baseline systolic blood pressure (BP) or systolic <90 mm Hg, and was treated with ephedrine 3-mg boluses. Although absolute systolic BP readings were significantly higher in the colloid group between 6 and 30 min (P < 0.05), the incidence of hypotension was not significantly different between the groups. The incidence of ephedrine use, incidence of nausea/vomiting, and median total dose of ephedrine were similar in all groups. We conclude that, in elderly patients undergoing elective procedures, withholding prehydration is not associated with any greater degree of hypotension or need for vasopressor therapy compared with crystalloid or colloid prehydration. First report of shoot and leaf blight caused by Botryosphaeria dothidea on Taxus × media in Sichuan province, China

Abstract:The genus Taxus is the natural material of the anticancer drug paclitaxel (Xiong et al. 2021). Harvesting sources of paclitaxel from the wild has greatly decreased the population of these trees. One of the taxus species, Taxus × media Rehder, a natural hybrid of taxus trees, has a higher paclitaxel content (Zhou et al. 2019). It has been introduced and cultivated in Sichuan, Chongqing, Yunnan, Zhejiang, Jiangxi, and other places in China. In 2021, approximately 20% of T. media (an average 30% of the affected area per tree) showed obvious shoot and leaf blight symptoms in a plantation of taxus trees (about 40 ha of the planting area), located in Sandaoyan county, Sichuan province, China (GPS, 103°94'60″N, 30°84'97″E). Initially, brown necrotic spots appeared on shoots. Gradually, the spots increased in number, expanded to the leaf attached to the branch, and caused wilting of the shoots and leaves. To identify the pathogen, symptomatic samples were randomly collected. Lesion margins of the diseased leaves and barks were surface sterilized for 1 min in 75% ethanol, rinsed with sterile distilled water three times, dried with sterile filter paper, placed on potato dextrose agar (PDA) amended with streptomycin sulfate (50 mg/liter), and incubated at 28°C in the dark. Six purified fungal isolates were obtained. Collected isolates with similar morphology were described as Botryosphaeria spp. (Zhang et al. 2021). The colonies were initially white, gradually became dark gray with dense erial mycelium after 5 days, and formed black pycnidia (Dimensions, 121.3 to 134.6 μm, n = 5) after 16 days. Conidia were fusiform, aseptate, transparent, and thin-walled (23.6 ± 1.2 × 7.27 ± 1.3 μm, n = 50), similar to B. dothidea (Hattori et al. 2021). For pathogenicity testing, ten 2-year-old seedlings of T. media were selected. Fungal cakes of the isolate Tmsdy-2 were applied to the punctured stems of seedlings and covered with Parafilm. Pieces of sterile medium were used as controls. All the seedlings were incubated at 25 ± 2°C, 50% relative humidity, and 16 h of light in a greenhouse. Four days later, the inoculated seedlings developed brown spots and were blighted in 14 days, with symptoms similar to the original diseased plants. The controls remained healthy. The same fungus was reisolated from the infected tissues and subsequently identified by morphological characteristics and DNA sequence analysis. The pathogenicity test was repeated three times with similar results, confirming Koch's postulates. For molecular identification, the DNA of the isolates was extracted using a Quick-DNA Extraction Kit (Tiangen Biotech, Beijing). The ITS, LSU, SSU, TUB2, and TEF 1-α genes were amplified with the primer pairs ITS1/ITS4, LR0R/LR05, NS1/NS4 (Li et al. 2018), Bt2a/Bt2b, and EF1-728F/EF1-986R (Hattori et al. 2021), respectively. The generated sequences were deposited in GenBank with accession numbers OQ179939 (ITS), OQ179940 (LSU), OQ179942 (SSU), OQ268596 (TUB2), and OQ268597 (TEF 1-α). BLAST analyses showed >99.65% identity with previously deposited sequences of B. dothidea in GenBank. Based on the maximum likelihood method, phylogenetic analysis revealed 100% bootstrap support values with B. dothidea. The fungus was identified as B. dothidea based on morphological and multilocus phylogenetic analyses. To our knowledge, this is the first report of B. dothidea causing shoot and leaf blight of T. media in China. These results will contribute to developing control strategies for this disease.
First Report of Powdery Mildew on Vitis bryoniifolia Caused by Erysiphe necator in China

Abstract:Grapes, belonging to the genus Vitis, are one of the world's most economically valuable fruit crops, which are widely used as the source of wine, raisins, and juice. The genus Vitis contains about 60 species mainly distributed in Asia, Europe, and North America to northern South America (Wan et al. 2013). In May 2022, severe powdery mildew symptoms were found on wild Vitis bryoniifolia plants at Guizhou Normal University, Guiyang, China. The incidence observed was approximately 85% among 50 V. bryoniifolia plants. Infected leaves appear white powdery patches, eventually leading to chlorosis to necrosis. Mycelia occurred on adaxial and abaxial leaf surfaces, petioles and young shoots. Upon microscopic observation hyphae were hyaline and 3.5-6 μm wide. Hyphal appressoria were solitary and lobed to multilobed. Conidiophores were erect, straight to somewhat flexuous and 80-130 µm long (n = 30). Foot cells were subcylindrical to curved-sinuous at the base, followed by 2-4 cells. Conidia formed singly (occasionally 2-6 in a chain) and were ellipsoid to ovoid in shape with dimensions of 22.5-38 × 12-19.5 µm (n = 50). No fibrosin bodies were observed on conidia. Based on these morphological characteristics, the powdery mildew fungus strongly resembled Erysiphe necator (Braun and Takamasu 2000; Zheng et al. 1987) that has been shown to cause powdery mildew on Vitis (Gadoury et al. 2012; Gaforio et al. 2011; Qiu et al. 2015). To confirm the identification, the ribosomal DNA internal transcribed spacer (ITS) and the ribosomal large subunit (LSU) region were amplified and sequenced using the ITS1/ITS4 primer pair (White et al. 1990) and the NL1/NL4 primer pair (Ziemiecki et al. 1990), respectively. The spliced 1250-bp ITS-LSU sequence (GenBank accession no. PP188565) shared 99.84-100% identity with ITS-LSU sequences of E. necator (LC028995, LC028996, ON073862, LC777882, and OM033353). Based on the phylogenetic analysis of the combined ITS-LSU dataset of Erysiphe species (Takamasu et al. 2015), PP188565 was grouped in a clade with E. necator strains MUMH530, MUMHs141, and VPRI19719. To perform pathogenicity analysis, leaves of three healthy, potted 1-year-old V. bryoniifolia plants were inoculated by gently pressing with diseased leaves. Three non-inoculated healthy plants served as controls. All plants were incubated in a greenhouse at 25 ± 2°C with 80% relative humidity. Powdery mildew symptoms, similar to field conditions were observed on inoculated plants 13 days after inoculation, whereas control plants remained symptomless. Fungus reisolated from inoculated V. bryoniifolia plants was morphologically identical to that on originally diseased plants, and the spliced ITS-LSU sequence of reisolated fungus shared 100% identity with PP188565, fulfilling Koch's postulates. Based on the morphological and molecular characterizations, the powdery mildew fungus was identified as E. necator. To our knowledge, this is the first report of powdery mildew caused by E. necator on V. bryoniifolia in China. This work further expands the host range of Erysiphe necator on Vitis species.
First Report of Anthracnose Caused by Colletotrichum spaethianum on Zephyranthes candida in China

Abstract:Zephyranthes candida, an bulbous perennial plant, are planted in almost every park. In October 2023, anthracnose symptoms were observed on Z. candida leaves in Jiangxi Agricultural University (28.75° N, 115.83°E), Nanchang, Jiangxi Province, China, and the incidence of disease were up to 35% (140 of 400 plants). The lesions extended from the leaf apex to the base, appearing as a dark brown color, and later changed to yellow and became dry. To isolate the pathogen, 20 symptomatic leaves were collected and cut into small pieces (4×4 mm, one pieces per leave), surface-sterilized with 70% ethanol for 10 s and 1% NaClO for 30 s, rinsed thrice with sterile water, placed onto potato dextrose agar (PDA) plates and incubated at 25℃ for 5 days. Fifteen isolates (15 out of 20) with similar morphological characteristics were obtained. The colonies on PDA presented effuse mycelium, initially white and later pale gray. Conidia were hyaline, curved or slightly curved, aseptate, with a truncate base and acute apex, measuring 17 to 23 × 3 to 6 μm (n = 50), and were matched to Colletotrichum species (Damm et al. 2009). To further confirm species, two representative isolates (JFRL 03-2873 and JFRL 03-2874) were selected for molecular identification. The internal transcribed spacer (ITS), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), chitin synthase (CHS), histone 3 (HIS3), actin (ACT) and β-tubulin 2 (TUB2) regions were amplified and sequenced by using primers sets ITS5/ITS4, Gpd1/Gpd2, CHS-79F/CHS354R, CYLH3F/CYLH3R, ACT-512F/ACT-783R and T1/Bt2b (Tan et al. 2022), respectively. These sequences were deposited into GenBank with accession number PP425890-PP425891 (ITS), PP437551-PP437552 (GAPDH), PP437549-PP437550 (CHS), PP480643-PP480644 (HIS3), PP437547-PP437548 (ACT) and PP437553-PP437554 (TUB2). A BLASTN search revealed high similarity of 99%-100% to ITS (GU227807, 518 nt/519 nt), GAPDH (GU228199, 525 nt/526 nt), CHS (GU228297, 251 nt/251 nt), HIS3 (GU228003, 372 nt/373 nt), ACT (GU227905, 236 nt/237 nt) and TUB2 (GU228101, 490 nt/490 nt) sequences of Colletotrichum spaethianum CBS 167.49. A maximum likelihood phylogenetic tree was constructed by combining ITS, GAPDH, CHS , HIS3, ACT and TUB2 sequences in IQtree web server (Ngugen et al. 2015). The result indicated that the two representative isolates were clustered together with Colletotrichum spaethianum in a clade with 100% bootstrap support. Based on morphological observation and sequence analysis, the isolates were identified as C. spaethianum. To confirm pathogenicity, six surface-sterilized leaves of Z. candida were wounded and inoculated with 1 × 106 conidia/ml conidial suspension of JFRL 03-2873, and control leaves were inoculated with sterile water. They were incubated at 25 ℃ with 12 h photoperiod and 80% humidity, the experiment was repeated twice. After five days, all leaves inoculated with JFRL 03-2873 displayed anthracnose symptom, whereas the control leaves remained unaffected. We re-isolated C. spaethianum from the symptomatic leaves and identified it based on morphological and molecular characteristics. Previous studies reported that C. spaethianum caused anthracnose on various common herbaceous plants in China (Vieira et al. 2014, Guo et al. 2013), but to our knowledge, this is the first report of C. spaethianum causing anthracnose on Z. candida in China. Anthracnose disease caused great economic loss to the cultivation of landscape plant Z. candida. Therefore, it is necessary to pay more attention to the anthracnose disease of herbaceous plants caused by C. spaethianum and develop appropriate control strategies.
First report of Trunk Canker Caused by Botryosphaeria dothidea on Cinnamomum camphora in China

Abstract:Cinnamomum camphora, an essential woody plant in China, experienced a severe outbreak of trunk canker affecting an area of 540,000 square meters in the Xiaoshan District of Zhejiang province during the early summer of 2022. The observed symptoms included stem canker, dieback, twig blight, and extensive vascular discoloration, with an incidence rate ranging from 45% to 70%. To investigate the etiology of the disease, symptomatic plant samples were subjected to rigorous surface sterilization involving washing with running tap water, followed by surface sterilization using 75% ethanol. The samples were then rinsed twice with sterile distilled water, plated on potato dextrose agar (PDA) medium, and incubated at a temperature of 28°C. The isolated fungi exhibited characteristics consistent with the species Botryosphaeria dothidea. Fungal isolates displayed abundant white aerial mycelium, which darkened to grey after five to seven days, eventually giving rise to black pycnidia. Single hyphal tip cultures of putative two isolates were stored at the Agricultural Experiment Station of Zhejiang University. Conidia formed on pycnidia were one-celled, hyaline, aseptate, and fusiform, with dimensions of 18.34-27.12 μm x 3.77-6.84 μm (average 22.90 μm x 5.20 μm) (one hundred conidia were measured). To determine the fungal species, genomic DNA was extracted from individual isolates ZJUP0868, and subjected to DNA sequence analysis of four gene regions: the internal transcribed spacer (ITS) using the primer-pair ITS4 and ITS1 (White et al., 1990), the large subunit (LSU) gene with LR0R and LR5 (Rehner and Samuels, 1995), the small subunit (SSU) gene with SR1R and SR7 (Zoller et al., 1999), and the translation elongation factor 1-alpha (tef1) with EF1-983F and EF1-2218R (Rehner and Buckley, 2005). Furthermore, DNA sequence analysis of above four genes was performed. BLAST analysis indicated the highest nucleotide sequence identity with Botryosphaeria dothidea CBS 115476 reference sequence (ITS: 99.59%, KF766151; LSU: 99.88%, DQ377852; SSU: 100%, NG_062738; tef1: 98.93%, AY236898). Representative sequences of isolate ZJUP0868 from these regions were deposited in GenBank (ITS: Accession No. OR192838; LSU: Accession No:OR248147; SSU: Accession No:OR248174; tef1: Accession No. OR262053). A phylogenomic analysis was conducted to determine the phylogenetic position of Botryosphaeria dothidea in the family Botryosphaeriaceae. The combined morphological and molecular findings confirmed the identification of the pathogen as Botryosphaeria dothidea. Pathogenicity tests were conducted by stem inoculation of two-year-old Cinnamomum camphora baranches. Mycelial plugs (2-3 mm in diameter) from actively growing colonies of B. dothidea (PDA) were applied to bark of similar size on the middle point of the stems. Inoculated barks were wrapped with Parafilm, while control branches received sterile PDA plugs. Inoculated and control branches (7 each) were maintained in a greenhouse at 28°C. After two weeks, all inoculated plants exhibited dark vascular stem tissue, whereas the control plants remained healthy. B. dothidea was re-isolated from symptomatic tissues, thus fulfilling Koch's postulates. No symptoms were observed in the control branched, and B. dothidea was not re-isolated from their tissues. B. dothidea , the type species of Botryosphaeria (Botryosphaeriaceae, Botryosphaeriales), is commonly associated with cankers and dieback in woody plants. Previous reports have identified Botryosphaeria dothidea as a pathogen causing stem dieback and trunk canker on Glycine max (Chen et al., 2020), Camellia oleifera (Hao et al., 2022), and Gleditsia sinensis (Huang et al., 2020). Additionally, B. dothidea has been reported to cause leave wilt on various plant species in China, including Daimyo oak (Liu et al., 2023) and Cornus officinalis (Zhang et al., 2022). To the best of our knowledge, this is the first report of B. dothidea on C. camphora in China. This findings contribute to a better understanding of the pathogens affecting Cinnamomum camphora in the China.
First Report of Gibellulopsis nigrescens Causing Yellow Wilt on Chinese Cabbage in China

Abstract:Chinese cabbage (Brassica rapa L. ssp. pekinensis), in the family Brassicaceae, is a widely planted crop in China valued for its nutritional benefits. In May 2023, wilt symptoms on Chinese cabbage (cv. 'Dongtian118') were observed in several commercial fields located in Sheqi County, (32.47ºN, 112.46ºE), Nanyang, Henan Province, China. A disease survey noted that disease incidence on plants was approximately 20% to 50% within observed fields. Symptoms included yellowing and wilting leaves, and vascular discoloration of the stem bases. To isolate the pathogen, ten symptomatic leaves collected from different diseased cabbage in two field were cut into small pieces (5 × 5 mm), surface disinfected with 75% ethanol for 30 s, then washed three times in sterile water. After drying, tissues were transferred onto potato dextrose agar (PDA). Plates were incubated at 28℃ for 7 days in the dark. Twelve morphologically similar fungal isolates were obtained by single-spore subculture. The mycelia on PDA were originally white, later becoming dark gray due to the formation of masses of melanized chlamydospores after 15 days of culture. Conidiophores were hyaline and most had secondary branches. In addition, verticillate branches had three to four phialides in each whorl. The conidia were hyaline, elliptical or nearly circular, measuring from 3.2 to 9.5 × 2.6 to 3.8 μm (n=40). These morphological characteristics were similar to those described for Gibellulopsis nigrescens (Zare et al. 2007). The isolates were further identified based on PCR amplification. The ITS, GAPDH, and TEF1 genes were amplified using primers ITS1/ITS4, VGPDf2/VGPDr (Inderbitzin et al. 2011) and EF-2/EF1-728F (O'Donnell et al. 1998). BLAST analysis revealed 12 isolates were highly similar to G. nigrescens, with 99.82% similarity for ITS (OR818474, KJ534578), 93.17% similarity for GAPDH (JN188192.1, JN188166.1) and 91.07% similarity for TEF1 (EF543798.1, EF543804.1). Sequences of the representative isolate BC230515 were deposited into NCBI GenBank with accession nos. OR889646 for ITS and PP135039 for GAPDH. Pathogenicity of all 12 isolates was tested on potted Chinese cabbage plants (cv. 'Dongtian118'). Twenty-four healthy Chinese cabbage plants were inoculated by applying a 10 mL conidial suspension (1×107 conidial/mL) at the artificially wounded root region of each plant. Twenty-four control plants wounded similarly were treated with sterile distilled water. All plants were kept in a growth chamber at 22~25°C (day)/18~20°C (night) , 85% relative humidity and a photoperiod of 12 h per day. After 15 days, inoculated plants exhibited wilting symptoms similar to those observed in the field, whereas control plants remained healthy. The pathogenicity test was repeated three times. The associated fungus on the artificially inoculated plants was reisolated from the symptomatic leaves, and its identity was confirmed by PCR with the primers described above. Reisolated G. nigrescens had identical morphological and molecular characteristics to the original isolates, confirming Koch's postulates. To our knowledge, this is the first report of G. nigrescens causing yellowing and wilt of Chinese cabbage in China. G. nigrescens is a destructive pathogen with multiple hosts such as beet (Zhou et al. 2017), alfalfa (Hu et al. 2011), prevention and control measures should be taken in advance.

Abstract:Egg albumin in the native, unaltered state, does not give tests characteristic of sulfhydryl groups. When, however, this protein is treated in any one of several ways, such as by heat (8, 9, 17), ultraviolet irradiation (lo), shaking (lo), or by solution in urea or other amides (7), free sulfhydryl groups make their appearance. The amount of free -SH groups appearing in egg albumin through the action of urea, guanidine hydrochloride, and their derivatives has been estimated (6). Considerable differences in the number of such groups liberated, which depend on the nature and the concentration of the reagent used, were noted. The investigation of these groups has been extended to a number of proteins whose molecular weights have been reported to be smaller in urea solutions than in water. The three seed globulins, edestin, excelsin, and amandin, have practically the same molecular weight in water (4, 5, 15, 16). Their molecular weights in 4 M urea are also almost the same (4), but are reported to be only oneeighth as great as in water. Burk and Greenberg (5) showed that the molecular weight of horse hemoglobin in urea solution was 34,000, a value just half the molecular weight of this protein in water. The results of these investigators were confirmed by Wu and Yang (18) and by Steinhardt (14), the latter showing further that the molecular weight in acetamide solutions was comparable to that in urea. It is planted in about 10,000 ha in Zhejiang, Jiangsu, and Hubei provinces. In the past 3 years, a serious disease like southern blight occurred in hangju fields in Macheng, Hubei (31°17′14.45″ N, 115°03′13.167″ E). The disease mainly occurred in July after the rainy season. Disease incidence ranged from 30 to 50%, causing huge economic losses to growers. Disease symptoms appeared on the stem bases, followed quickly by wilting of the whole plants. White mycelia covered the stem, and numerous whitish to tan sclerotia developed on the colonized tissues. To determine the cause, plants with typical symptoms were collected from fields of Macheng and Wuhan, and isolates were obtained from the samples by culturing on potato dextrose agar (PDA) at 25°C. Mycelia grew radially over the whole plate after 3 days, and sclerotia formed after 5 days. Sclerotia were initially white and gradually turned brown as they matured. Mature sclerotia were globoid, ellipsoidal, or irregular in shape and measured 1.4 to 4.2 mm (2.78 ± 0.56 mm; n = 50) in diameter. The number of sclerotia per plate ranged from 61 to 133 (mean 87; n = 10). The hyphae formed septa, but no spores were observed. Genomic DNA was extracted from isolate J-12 using the Ezup Column Fungi Genomic DNA Purification Kit (Sangon Biotech, Shanghai, China). The 18S rDNA fragment was amplified using primers NS1 (5′-GTAGTCATATGCTTGTCTC-3′) and NS6 (5′-GCATCACAGACCTGTTATTGCCTC-3′) (Li et al. 2014) and sequenced by Sangon Biotech. The sequence (GenBank MN104721) was 99.85 and 99.98% similar to isolates of Athelia rolfsii (Curzi) C. C. Tu & Kimbr (JF819726.1 and AY665774.1, respectively). The isolate was also amplified with primers EF595F (5′-CGTGACTTCATCAAGAACATG) and EF1160R (5′-CCGATCTTGTAGACGTCCTG) for the translation elongation factor (TEF-1α) (Wendland and Kothe 1997) and sequenced. The TEF-1α sequence (MN106270) was 99.02% similar to A. rolfsii strain Sr_12 (JF267794.1). A phylogenetic tree constructed based on the TEF-1α sequence using the neighbor-joining method in MEGA6 (Tamura et al. 2013) showed that J-12 was very close to 15 GenBank accessions, all from A. rolfsii. Thus, J-12 was identified as A. rolfsii, corroborating the identification of Sclerotium rolfsii Sacc., the anamorph, based on cultural and morphological features. To demonstrate the pathogenicity, mature sclerotia were placed on the upper, middle, and lower parts of leaves (n = 6), stems (n = 6), and roots (n = 6) of heathy hangju seedlings rinsed with sterile water, and the inoculated stems, leaves, and roots were incubated in a moist chamber for 9 days at 25 ± 2°C (Mahadevakumar et al. 2018). Samples inoculated with sterile water were used as a control. Two days after inoculation, sclerotia began to germinate and infect plant tissues. Five days after inoculation, the detached stems were completely covered by mycelia, and leaves and roots were rotten. Sclerotia formed 6 days and matured 9 days after inoculation. In another experiment, 20-day-old healthy seedlings with nine true leaves were transplanted and grown in a growth chamber at 28 ± 2°C (n = 6). Stems were pierced with a sterile needle, and three of them were inoculated with 5 ml of J-12 mycelium suspension, and the other three seedlings were treated with 5 ml of sterile water as a control. After 3 days, mycelia began to grow at the base of inoculated stems. The surface of the stem base became black and withered, and the lower leaves of inoculated plants also became necrotic and wilted. The inoculated plants were completely wilted 10 days after inoculation. Fungal cultures were isolated from the inoculated sites and confirmed as S. rolfsii based on cultural and morphological features.
