Abstract:Microplastics (MPs) and the heavy metal cadmium (Cd) have attracted global attention for their toxicological interactions in aquatic organisms. The purpose of this investigation was evaluating the effect of MPs (1 mg L<sup>-1</sup>) and Cd (5 mg L<sup>-1</sup>) on the liver function, immune response of crucian carp (Carassius carassius) after 96 h exposure, and intestinal microbiota after 21 days, respectively. Co-exposure to MPs and Cd significantly enhanced MP accumulation in the liver of the crucian carp compared to the accumulation with exposure to MPs alone. Co-exposure to MPs and Cd triggered notable histopathological alterations accompanied by increased hepatic cell necrosis and inflammation, and was associated with higher aspartate aminotransferase and alanine aminotransferase levels, lower superoxide dismutase and catalase activity levels, but higher malondialdehyde content and total antioxidant capacity in the liver. Moreover, the combined treatment of MPs and Cd led to the up-regulated transcription of genes related to immune response, such as interleukin 8 (il-8), il-10, il-1β, tumor necrosis factor-α, and heat shock protein 70, both in the liver and spleen. Co-exposure to MPs and Cd reduced the variety and abundance of the intestinal microbiota in the crucian carp. Our research indicates that the combined exposure to MPs and Cd may exert synergistic toxic effects on crucian carp, which could impede the sustainable growth of the aquaculture industry and pose potential risks to food safety.

Abstract:As the accumulation of microplastics (MPs) in the environment continues to rise, more concerns focus on the health risk of combined exposure to MPs and other contaminants. The aim of this study is to investigate the influences of MPs on the tissue-accumulation of cadmium (Cd) in zebrafish and explore the related chronic toxic effects induced by combined exposure of Cd and MPs. After co-exposure to MPs and Cd for 3 weeks, 20 and 200 μg/L MPs increased the accumulation of Cd in zebrafish livers (46% and 184%), guts (10% and 25%) and gills (9% and 46%). The Cd accumulation was gill > gut > liver. Comprehensive analyzes of biochemical biomarkers, histopathological observation and functional gene expression firstly demonstrated that the presence of MPs enhanced the toxicity of Cd on zebrafish and the combined exposure caused oxidative damage and inflammation in zebrafish tissues. Collectively, our results highlight the chronic effects of combined exposure to MPs and heavy metals.

Abstract:Microplastics (MPs) and methylmercury (MeHg) have attracted increasing attention due to ubiquitous occurrence and toxicity. This study aimed to investigate whether MPs could absorb MeHg and thus modify its bio-concentration and neurotoxicity in the zebrafish larvae (Danio rerio). The pseudo-second-order model (R-2 = 0.989) was found to be suitable for describing the adsorption kinetics of MeHg onto MPs. Compared with Freundlich and Temkin models, the Langmuir isotherm model provided a better fit with the experimental data exhibiting a maximum monolayer adsorption capacity of 54.945 mg/g. These results suggested that adsorption occurs mainly by a chemical process dominated by monolayer adsorption. MPs adsorbed MeHg to form MPs/ MeHg complex, which was ingested by zebrafish larvae, and promoted accumulation of MeHg. Thus, the presence of MPs aggravated the reduction of locomotor activity induced by MeHg, and downregulation of neuro-transmitters related genes, such as ache, gfap and scl1A3b. Metabolome analysis also revealed disrupted glutathione (GSH) metabolism upon exposure of MeHg alone and in combination with MPs, as reflected by the increased in the ratio of GSH and oxidized glutathione. These effects were also confirmed by upregulation of oxidative stress-related genes, such as sod, sod mt and gpx4a. Collectively, these results indicated that MPs could act as a carrier of MeHg and enhance its accumulation in zebrafish, thereby disrupting locomotor activity by excessive oxidative stress. This study provides a scientific basis for improving health risk assessment of environmental pollutants, particularly those potentially able to adsorb to MPs by virtue of their chemical nature.

Abstract:The escalating impact of human activities has led to the accumulation of microplastics (MPs) and heavy metals in marine environments, posing serious threats to marine ecosystems. As essential components of oceanic ecosystems, large seaweeds such as Caulerpa lentillifera play a crucial role in maintaining ecological balance. This study investigated the effects of MPs and cadmium (Cd) on the growth, physiology, biochemistry, and Cd accumulation in C. lentillifera while elucidating the underlying molecular regulatory mechanisms. The results demonstrated that exposure to MPs alone significantly promoted the growth. In contrast, exposure to Cd either alone or in combination with MPs significantly suppressed growth by reducing stem and stolon length, bud count, weight gain, and specific growth rates. Combined exposure to MPs and Cd exhibited the most pronounced inhibitory effect on growth. MPs had negligible impact while Cd exposure either alone or combined with MPs impaired antioxidant defenses and exacerbated oxidative damage; with combined exposure being the most detrimental. Analysis of Cd content revealed that MPs significantly increased Cd accumulation in algae intensifying its toxic effects. Gene expression analysis revealed that Cd exposure down-regulated key genes involved in photosynthesis, impairing both photosynthetic efficiency and energy conversion. The combined exposure of MPs and Cd further exacerbated these effects. In contrast, MPs alone activated the ribosome pathway, supporting ribosomal stability and protein synthesis. Additionally, both Cd exposure alone or in combination with MPs significantly reduced chlorophyll B and soluble sugar content, negatively impacting photosynthesis and nutrient accumulation. In summary, low concentrations of MPs promoted C. lentillifera growth, but the presence of Cd hindered it by disrupting photosynthesis and antioxidant mechanisms. Furthermore, the coexistence of MPs intensified the toxic effects of Cd. These findings enhance our understanding of how both MPs and Cd impact large seaweed ecosystems and provide crucial insights for assessing their ecological risks.

Abstract:Microplastic particles (MPs) are widely distributed in the environment. The high surface ratio of MPs make them effective transmission mediums for many toxic pollutants. The combined toxicity of MPs and heavy metals have received increasing attention in recent years. In this study, effects of MPs (100 μg/L) on the toxicity of low (15 μg/L) and high (150 μg/L) concentrations of cadmium (Cd) to zebrafish were evaluated based on a 10-day subacute exposure. The survival rate, growth, antioxidant capacity, reactive oxygen species (ROS) accumulation, histology and Cd biological enrichment in different tissues were investigated with the objective to understand the effect and mechanism of MPs on Cd toxicity to zebrafish. The results showed that the effect of MPs on Cd toxicity mainly depended on the concentration of Cd. MPs significantly enhanced the toxicity of low concentrations of Cd (LCd), including lower antioxidant enzyme activities, higher ROS levels, more severe tissue damage, inhibited growth rate and lower survival rate. However, the effects of MPs on the toxicity of high concentrations of Cd (HCd) were exactly opposite to LCd. Cd enrichment analysis showed that MPs could significantly increase LCd accumulation in intestine, gill, skin and muscle tissues, while decrease the enrichment of HCd in liver, intestine, gill and muscle tissues. Free Cd in the exposure water was significantly decreased by MPs in the HCd and MPs combined exposure group. These results suggest that effect of MPs on Cd toxicity to zebrafish depending on Cd concentration, MPs can increase the enrichment of LCd in zebrafish and enhance its toxicity, but can decrease the enrichment of HCd in zebrafish and attenuate its toxicity. The present study will broaden our understanding of the interaction between MPs and heavy metals.

Abstract:Small heat shock proteins are a ubiquitous and diverse family of stress proteins that have in common an α-crystallin domain. Mycobacterium tuberculosis has two small heat shock proteins, Acr1 (α-crystallin-related protein 1, or Hsp16.3/16-kDa antigen) and Acr2 (HrpA), both of which are highly expressed under different stress conditions. Small heat shock proteins form large oligomeric assemblies and are commonly polydisperse. Nanoelectrospray mass spectrometry showed that Acr2 formed a range of oligomers composed of dimers and tetramers, whereas Acr1 was a dodecamer. Electron microscopy of Acr2 showed a variety of particle sizes. Using three-dimensional analysis of negative stain electron microscope images, we have shown that Acr1 forms a tetrahedral assembly with 12 polypeptide chains. The atomic structure of a related α-crystallin domain dimer was docked into the density to build a molecular structure of the dodecameric Acr1 complex. Along with the differential regulation of these two proteins, the differences in their quaternary structures demonstrated here supports their distinct functional roles.

Abstract:The harm of microplastics (MPs) to aquatic ecosystems is caused by their stable and non-degradable properties. Additionally, the pollutants such as heavy metals in the water are easy to be adsorbed on their surface with their small particle size and large specific surface area, resulting in environmental pollution. Therefore, the study on the mixture toxicity of MPs and heavy metals has theoretical significance for the risk assessment of aquatic ecosystems. In the present study, 10 nm polystyrene (PS) and cadmium (Cd) were used, and their individual and mixture acute toxicities on grass carp (Ctenopharyngodon idellus) were examined. The results indicated that the mortality of the fish increased with the concentration from 10 mg L−1 to 20 mg L−1, and the existence of PS-MPs elevated the Cd concentrations in the fish and accelerated the death. Whether the Cd and/or the PS-MPs concentrations caused varying degrees of damage to the gills, kidney, liver, and muscles of the grass carp, especially under the highest concentrations (20 mg L−1 Cd + 300 μg L−1 PS-MPs). Moreover, low concentrations of PS-MPs alone (30 μg L−1 PS-MPs) significantly increased the superoxide dismutase (SOD) activity in the kidney and liver, reaching 12.43% and 14.38%, respectively (P < 0.05). The peroxidase (POD) activity was increased only in the kidney, up to 25.95% (P < 0.05). Also, significant reductions in SOD and POD activities were observed in the combination of high concentration of Cd (20 mg L−1) and 300 μg L−1 PS-MPs (P < 0.05). To the best of our knowledge, there are few studies on the impact of combined toxicity of PS-MPs and Cd on grass carp under laboratory conditions. Therefore, these findings may provide a theoretical guarantee for pollution prevention and control in the aquatic ecosystem.

Abstract:Cadmium (Cd) and microplastics (MPs) gradually increased to be prevalent contaminants in soil, it is important to understand their combined effects on different soil-plant systems. We studied how different doses of polylactic acid (PLA) and polyethylene (PE) affected Cd accumulation, pakchoi growth, soil chemical and microbial properties, and metabolomics in two soil types. We found that high-dose MPs decreased Cd accumulation in plants in red soil, while all MPs decreased Cd bioaccumulation in fluvo-aquic soil. This difference was primarily attributed to the increase in dissolved organic carbon (DOC) and pH in red soil by high-dose MPs, which inhibited Cd uptake by plant roots. In contrast, MPs reduced soil nitrate nitrogen and available phosphorus, and weakened Cd mobilization in fluvo-aquic soil. In addition, high-dose PLA proved detrimental to plant health, manifesting in shortened shoot and root lengths. Co-exposure of Cd and MPs induced the shifts in bacterial populations and metabolites, with specific taxa and metabolites closely linked to Cd accumulation. Overall, co-exposure of Cd and MPs regulated plant growth and Cd accumulation by driving changes in soil bacterial community and metabolic pathways caused by soil chemical properties. Our findings could provide insights into the Cd migration in different soil-plant systems under MPs exposure. Environmental Implication Microplastics (MPs) and cadmium (Cd) are common pollutants in farmland soil. Co-exposure of MPs and Cd can alter Cd accumulation in plants, and pose a potential threat to human health through the food chain. Here, we investigated the effects of different types and doses of MPs on Cd accumulation, plant growth, soil microorganisms, and metabolic pathways in different soil-plant systems. Our results can contribute to our understanding of the migration and transport of Cd by MPs in different soil-plant systems and provide a reference for the control of combined pollution in the future research.

Abstract:Microplastics (MPs) are pollutants that are widely distributed in the aquatic environment. Fish are directly exposed to water and are at risk of ingesting a large amount of MPs. In the present study, the grass carp were exposed to two concentrations of MPs (1000 and 100 μg/L) and fluorescence signals were detected in the liver digestion solution. Grass carp exposed to MPs for 21-days showed liver cytoplasmic vacuolation and inhibited growth. At the end of the exposure period, the fish treated with MPs exhibited inhibition of the antioxidant system and enhancement oxidative stress in comparison with the control group. The transcriptome analysis of grass carp was then performed to reveal the molecular mechanism of the response to MPs. In total, 1554 differentially expressed genes (DEGs) were identified. The results of GO and KEGG pathway analysis of the DEGs identified energy metabolism-related pathways and peroxisome proliferator-activated receptor (PPAR) signaling pathway. Taken together, the present study not only highlighted oxidative stress and metabolism disorders related to MP ingestion, but also determined the risk of MP exposure to teleost.

Abstract:Microplastics can accumulate residual drugs and heavy metals in the environment and accumulate through the layers of the food chain, ultimately causing harm to human health. The pollution of microplastics in the freshwater environment is becoming more and more serious, which directly affects the safety of aquatic organisms. This experiment studied the effects of single and composite microplastics and Cd on the tissue damage, antioxidant, and immune response of juvenile Channa argus. Microplastics with different diameters of 80 nm, 0.5 μm (200 μg/L), and Cd (50 μg/L) were used for exposure, and four sampling points were set for 24 h, 48 h, 96 h, and clear 48 h. Under different treatments, a certain degree of gill tissue damage can be found in 96 h. Microplastics and Cd can cause oxidative stress and affect the antioxidant status, and the impact of 0.5-μm microplastics is stronger than that of 80-nm microplastics. There is an antagonistic effect between the two microplastics and Cd during compound exposure, but the activity of CAT shows a synergistic effect. Microplastics and Cd affect the expression of immune-related genes to varying degrees. When exposed together, the expression of HSP70 gene all showed mutual antagonism, while the expression of IL-1β gene was different. The expression of the MT gene can infer the ability of microplastics to accumulate Cd, and microplastics with a small diameter of 80 nm have stronger enrichment capabilities.

Abstract:Microplastic pollution of the ocean has received extensive attention as plastic pollution increases globally, but the potential ecological risks caused by microplastic interactions with trace metals still require further research. In this study, Apostichopus japonicus was used to explore the individual and combined toxicities of cadmium (Cd) and microplastics and their effects on growth, Cd tissue accumulation, digestive enzymes, and gut microbes. The body weight gain and specific growth rate of animals exposed to a combination of high concentrations of Cd and microplastics decreased. The addition of high concentrations of cadmium to the diet led to an increase in cadmium content in the respiratory tree, digestive tract and body wall. Amylase, lipase and trypsin decreased to different degrees in the group treated with high concentrations of Cd/microplastics. Firmicutes were significantly reduced across multiple treatment groups, with the order Lactobacillales being the most significantly affected. Cd is the pollutant causing the greatest negative impact, but the presence of microplastics undoubtedly increases its toxicity.

Abstract:The coexistence of microplastics (MPs) and toxic metal contaminants in soils is becoming increasingly common, thereby posing serious threat to soil–plant systems. Cadmium (Cd) is the most common metal contaminant in soil and can easily combine with MPs, thereby altering its bioavailability. However, few studies have focused on the co-pollution of MPs and Cd, particularly the complex phytotoxicity caused by their interaction and the effect of co-exposure on Cd uptake in plants. We conducted pot experiments to compare the effects of exposure to polystyrene (PS) and Cd, as well as the effects of co-exposure (PS + Cd), on the physiological characteristics of Brassica chinensis L. and explored the regulatory factors of MPs on Cd uptake in plant tissues. The results showed that plant biomass, photosynthetic parameters, and chlorophyll content significantly decreased (p < 0.05) with increasing PS doses under treatment with MPs alone. Although the negative effects of PS and Cd co-exposure on plants were higher than those of PS alone, however, the addition of MPs reduced the toxicity effects of Cd on plants and decreased the uptake and accumulation of Cd by plants compared with the Cd treatment alone. Furthermore, plants can resist the increased malondialdehyde content and oxidative stress induced by PS and Cd exposure by increasing the activities of superoxide dismutase and peroxidase. Under the PS + Cd treatment, linear models showed that soil organic carbon and sucrase activity were the key variables affecting Cd uptake by plant shoots and roots, respectively. The results of the partial least squares path modeling further showed that PS indirectly affected Cd uptake by B. chinensis by significantly affecting the physicochemical properties of soil, Cd concentration, and enzyme activity. Our results provide a new perspective and an important reference for further understanding the effects of MPs on the bioavailability and fate of heavy metals.

Abstract:The widespread application of micro-plastics (MP) and their release in the open environment has become a matter of worldwide concern. When interacting with contaminants such as heavy metals in the soil ecosystem, MPs can result in detrimental effects on the soil environment and plant growth and development. However, information based on the interaction between MPs and heavy metals and their effects on terrestrial plants is still limited. Keeping this in mind, the present study was conducted to explore the single and combined toxicity of polypropylene (PP) MPs (13 and 6.5 μm) and cadmium (Cd) on germination indices; root and stem growth; fresh and dry weight; and anti-oxidative enzyme activities of rice (Oryza sativa L.) seedlings. Our results indicated that a single application of PP MP and Cd on rice seedlings inhibited most of the germination indicators, while their co-occurrence (PP + Cd) showed a reduction in the overall toxicity to some extent. A single application of both the contaminants significantly inhibited root length, stem length, fresh weight and the activities of catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) enzymes in rice seedling, while no significant effect on dry weight was observed. The combined toxicity of both PP and Cd revealed that 13 μm PP + Cd had an antagonistic effect on the growth of rice seedlings, while 6.5 μm PP + Cd showed a synergistic effect. The present study revealed that smaller PP MP particles (6.5 μm) prominently affected plant growth more as compared to larger particles (13 μm). Our work reported the combined effect of PP MP and Cd on the germination and growth of rice for the first time. This study can provide the basis for future research on the combined effects of different types and sizes of MPs and heavy metals on the terrestrial ecosystem.

Abstract:As emerging contaminants, microplastics (MPs) are predicted to act as vectors for other contaminants and their combined effects are largely unknown. In this study, the combined effects of MPs and natural organic matter (NOM) on the accumulation and toxicity of copper (Cu) in zebrafish (Danio rerio) were investigated. As a result, small-size MPs could absorb more Cu than large-size MPs. The presence of NOM promoted Cu adsorption on MPs in the pH range of 6–8. Our results demonstrate that the combination of MPs and NOM increased Cu accumulation in the livers and guts in a size-depended manner. Correspondingly, the results of biochemical test showed that MPs and NOM could aggravate Cu-toxicity in the livers and guts, which is manifested in the increased levels of malonaldehyde (MDA) and metallothionein (MT) and decreased levels of superoxide dismutase (SOD). Furthermore, the results of transcriptomic analysis suggested that such aggravation of toxicity was mainly attributed to the inhibition of Cu-ion transport and the enhanced oxidative stress. Since the co-existence of MPs and NOM in the environment is inevitable, their enhancement effects on the bioaccumulation and toxicity of other pollutants such as heavy metals deserve more attention.

Abstract:Microcystin-LR (MC-LR) and microplastics (MPs) have attracted increasing attention as important new pollutants in freshwater fishery environments. However, there are few reports on the effects of long-term combined MC-LR and MPs pollution on nitrogen transformation and microbial communities in aquaculture ponds, and the resulting risks have yet to be determined. Therefore, in this study, traditional refractory MPs (polystyrene, PS), biodegradable MPs (polylactic acid, PLA) and MC-LR, which are common in freshwater fishery environments in China, were selected as pollutants to construct a microcosm that simulates freshwater aquaculture ponds. MC-LR coexposure to PS and PLA was tested to reveal the effects of these pollutants on nitrogen transformation and microbial communities in aquaculture ponds, as well as to elucidate the potential risks posed by traditional refractory MPs and biodegradable MPs to freshwater aquaculture ecosystems. The results revealed that the MPs had a relatively high adsorption rate for MC-LR and that PS presented a relatively high adsorption capacity, whereas PLA presented a relatively high desorption capacity. Single or combined MPs and MC-LR pollution disrupted the normal nitrogen cycle in the aquaculture system, causing an overall loss of nitrogen in the water, and denitrification and nitrogen fixation in the water were inhibited to a certain extent through the inhibition of nitrogen cycle-related functional genes, with the PS + MC-LR group having the greatest inhibitory effect. In addition, compared with single-pollutant exposure, combined exposure to MC-LR and MPs had a greater effect on the microbial community composition. Analysis of the integrated biomarker response (IBR) index revealed that the risk of combined exposure to MC-LR and PS was greater than that of single exposure, so this phenomenon merits further attention.

Abstract:Does exclusion failure--responding with previously presented words despite instructions to avoid doing so--demonstrate unconscious influences? This article examines exclusion-based evidence for unconscious perception. I propose an alternative signal detection theory (SDT) framework that can account for exclusion failure and ostensibly convergent qualitative differences without positing additional unconscious perceptual mechanisms. In the proposed SDT model, exclusion failure is a criterion artifact, similar to classic SDT-based critiques of subjective threshold approaches. However, it is suggested that exclusion approaches do demonstrate that response strategies are applied only to above-criterion stimuli and thereby illustrate important qualitative differences between two conscious processes: phenomenal awareness itself and higher-order (i.e., metacognitive) decision processes.
