Abstract:This study aimed to improve the thermodynamic performance of nanoliposomes (NLs) using fucoidan (F) as the second-layer coating biopolymer along with chitosan (CS), to control the delivery and bioavailability of catechin (C) and juglone (J). The stabilized liposomal carrier of F/CS-conjugated JC-NL (F-CS-JC-NL) was developed with optimum concentrations of CS (0.09 wt%) and F (0.10 wt%), with the highest encapsulation efficiency of juglone (95.47%) and catechin (90.88%). Physicochemical characterization revealed that F-CS-JC-NL disclosed improved stability under different pH and ionic strengths, with the maximum juglone/catechin retention under thermal, oxidative and storage conditions. In vitro digestion revealed that NL double-coating (F-CS-JC-NL) significantly reduced compound leakage in the gastrointestinal tract, providing a controlled release and better bioavailability of juglone/catechin compared to CS-JC-NL and JC-NL. Conclusively, this study provides a novel NL-based de-livery carrier with enhanced physicochemical stability and controlled release that might have promising use in delivering functional ingredients.

Abstract:There is ongoing research on various herbal bioactives and delivery systems which indicates that both lipid nanocarriers and herbal medicines will be fine tunned and integrated for future bio-medical applications. The current study was undertaken to systematically develop NLC-DSG-yam extract for the improved efficacy of herbal Diosgenin (DSG) in the management of anti-inflammatory disorders. NLCs were characterized regarding the mean size of the particles, morphological characteristics, physical stability in time, thermal behaviour, and entrapment efficiency of the herbal bioactive. Encapsulation efficiency and in vitro antioxidant activity measured the differences between the individual and dual co-loaded-NLC, the co-loaded one assuring a prolonged controlled release of DSG and a more emphasized ability of capturing short-life reactive oxygen species (ROS). NLCs safety properties were monitored following the in vitro MTS ((3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) tetrazolium reduction assay) and RTCA (Real-Time Cell Analysis) assays. Concentrations less than 50 μg/mL showed no cytotoxic effects during in vitro cytotoxicity assays. Besides, the NLC-DSG-yam extract revealed a great anti-inflammatory effect, as the production of pro-inflammatory cytokines (TNF-alpha, IL-6) was significantly inhibited at 50 μg/mL NLC (e.g., 98.2% ± 1.07 inhibition of TNF-α, while for IL-6 the inhibition percentage was of 62% ± 1.07). Concluding, using appropriate lipid nanocarriers, the most desirable properties of herbal bioactives could be improved.

Abstract:Luteolin is one of the most common edible herbal flavonoids that has anti-inflammatory, antioxidant and anticancer effects. The aim of this study was to develop an optimized luteolin-loaded nanostructured lipid carrier (NLC) using propolis wax as the solid lipid and safflower oil as the liquid lipid via a melt emulsification-ultrasonication method. Response surface methodology was utilized to investigate the influence of total lipid concentration and luteolin content on particle size, polydispersity index (PDI), and encapsulation efficiency (EE) in order to determine the optimal NLC formulation. The optimized NLCs exhibited spherical particles with an average size of 120.904 nm, a PDI of 0.142, and an EE of 66.87%. The particles also demonstrated high physical stability over a 90-day period at 4, 25 and 37 °C. Differential scanning calorimetry, X-ray diffraction, and transform infrared spectroscopy analyses showed high compatibility and proper embedding of luteolin in NLC structure. The gastrointestinal release of luteolin from the NLC formulation followed a sustained pattern compared to free luteolin, indicating the absence of any unencapsulated form on its surface. Additionally, the antioxidant activity of luteolin-loaded NLCs (L-NLC), as measured by DPPH and Folin-Ciocalteu methods, was significantly higher than free luteolin after 30 days. The microbiological tests using the microdilution broth method showed that NLCs had more antimicrobial properties compared to free luteolin against examined foodborne pathogens. In conclusion, the developed NLCs hold great potential as a delivery system for luteolin encapsulation and can be applied in the food and drug industries.

Abstract:The combined application of clove oil in a lipid nanocarrier opens a promising avenue for bone and joints therapy. In this study, we successfully developed a tunable controlled-release lipid platform for the efficient delivery of clove oil (CO) for the treatment of rheumatoid arthritis (RA). The ultra-small nanostructured lipid carriers co-loaded with CO (CONCs) were developed through an aqueous titration method followed by microfluidization. The CONCs appeared to be spherical (particle size of 120 nm), stable (zeta potential of −27 mV), and entrapped efficiently (84.5%). In toluene:acetone:glacial acetic acid (90:9:1 percent v/v/v) solvent systems, high-performance thin layer chromatography (HPTLC) analysis revealed the primary components in CO as eugenol (RF = 0.58). The CONCs greatly increased the therapeutic impact of CO in both in vitro and in vivo biological tests, which was further supported by excellent antiarthritic action. The CONC had an antiarthritic activity that was slightly higher than neat CO and slightly lower than standard, according to our data. The improved formulation inhibited serum lysosomal enzymes and proinflammatory cytokines while also improving hind leg function. This study provides a proof of concept to treat RA with a new strategy utilizing essential oils via nanodelivery.

Abstract:Low bioavailability and stability of 7,8-dihydroxyflavone (7,8-DHF) hinder its potential application in prevention of humans' disease. 7,8-DHF loaded nanoliposomes decorated by original, crosslinked and glycosylated lactoferrin (LF) were fabricated. The mean particle size of spherical crosslinked and glycosylated LF coating liposomes were larger than uncoated liposomes with a low polydispersity index, and the former had higher encapsulation efficiency (Above 98%). Crosslinked and glycosylated LF coating liposomes had higher antioxidant activity, storage stability, slow-release ability, bioaccessibility and transepithelial transport compared to free 7,8-DHF and 7,8-DHF loaded liposome. Effective hydrogen bonding between phospholipid and 7,8-DHF, while electrostatic interaction and hydrogen bonding between LF proteins and phospholipid existed. The encapsulated 7,8-DHF was in an amorphous state rather than a crystalline form as detected by differential scanning calorimetry. Collectively, crosslinked or glycosylated LF coating liposomes are promising delivery systems for protecting and transporting 7,8-DHF or other relative bioactive components.
Encapsulation of Oleuropein in Nanostructured Lipid Carriers: Biocompatibility and Antioxidant Efficacy in Lung Epithelial Cells

Abstract:Oxidative damage has been linked to a number of diseases. Oleuropein (OLE), a natural occurring polyphenol from olive leaves (Olea europaea L.), is known to be a potent antioxidant compound with inherent instability and compromised bioavailability. Therefore, in this work, nanostructured lipid carriers (NLCs) were proposed for OLE encapsulation to protect and improve its antioxidant efficacy. The lipid matrix, composed of olive oil and Precirol, was optimized prior to OLE encapsulation. The characterization of the optimized oleuropein-loaded NLCs (NLC-OLE) showed a mean size of 150 nm, a zeta potential of −21 mV, an encapsulation efficiency of 99.12%, sustained release profile, and improved radical scavenging activity. The cellular in vitro assays demonstrated the biocompatibility of the NLCs, which were found to improve and maintain OLE antioxidant efficacy in the A549 and CuFi-1 lung epithelial cell lines, respectively. Overall, these findings suggest a promising potential of NLC-OLE to further design a pulmonary formulation for OLE delivery in lung epithelia.

Abstract:Abstract Flavonoids are a class of polyphenolic compounds that can be extracted from citrus fruits. Because of their features, they have been considered as new bullets in cancer therapy. Flavonoids present anti-oxidant, anti-inflammatory and anti-cancer properties and some studies are pointing that their higher intake involves lower risks of cancer. The nanostructured lipid carriers (NLC) are drug delivery systems composed from a mixture of solid and liquid lipids that increase physical stability and promote the sustained release of the drug. So, the main aim of this research was to combine the potential of the flavonoid as a natural anti-oxidant compound with the nanostructured lipid carriers to form an efficient system for flavonoid delivery into the cells. These NLC were prepared through high pressure homogenization technique and were evaluated in function of several standard properties. The samples exhibited good stability through time and temperature, with sizes of around 215 nm and zeta potentials of -35 mV during a period of 105 days. The encapsulation and the loading efficiency were respectively 96% and 2.25% up to 105 days of storage. In vitro release assays showed a burst process for NLC-L pointing to their shell structure with flavonoid situated in the contour of the particles. The cell viability tests showed that flavonoid, either free or loaded into NLC, did not present in vitro toxicity to the melanoma cells tested: A375, CHL01 and SKMEL147 through 3-(4,5-dimethyiltiazol-2yl)-2,5-diphenyl tetrazolium bromide (MTT), neutral red and crystal violet assays. Hesperidin at 45 μmol L −1 was able to reduce significantly the radical forming up to 92.58 ± 0.82% in DPPH assay. On the other hand, the produced NLC were applied in a skin lotion formulation, which showed a good stability and opened up a new application for the produced nanoparticles as in anti-aging and moisturizing cosmetics. Finally, the in vitro cytotoxicity tests were carried out using several human culture cells. The flavonoid loaded and empty nanocarriers have been used to treat the cells. All data suggested that, at the level studied, non-significant toxicities on cells were observed.

Abstract:Nanovesicles produced with lipids and polymers are promising devices for drug and bioactive delivery and are of great interest in pharmaceutical applications. These nanovesicles can be engineered for improvement in bioavailability, patient compliance or to provide modified release or enhanced delivery. However, their applicability strongly depends on the safety and low immunogenicity of the components. Despite this, the use of unsaturated lipids in nanovesicles, which degrade following oxidation processes during storage and especially during the proper routes of administration in the human body, may yield toxic degradation products. In this study, we used a biopolymer (chitosan) labeled with flavonoid (catechin) as a component over a lipid bilayer for micro- and nanovesicles and characterized the structure of these vesicles in oxidation media. The purpose of this was to evaluate the in situ effect of the antioxidant in three different vesicular systems of medium, low and high membrane curvature. Liposomes and giant vesicles were produced with the phospholipids DOPC and POPC, and crystalline cubic phase with monoolein/DOPC. Concentrations of chitosan–catechin (CHCa) were included in all the vesicles and they were challenged in oxidant media. The cytotoxicity analysis using the MTT assay (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) revealed that concentrations of CHCa below 6.67 μM are non-toxic to HeLa cells. The size and zeta potential of the liposomes evidenced the degradation of their structures, which was minimized by CHCa. Similarly, the membrane of the giant vesicle, which rapidly deteriorated in oxidative solution, was protected in the presence of CHCa. The production of a lipid/CHCa composite cubic phase revealed a specific cubic topology in small-angle X-ray scattering, which was preserved in strong oxidative media. This study demonstrates the specific physicochemical characteristics introduced in the vesicular systems related to the antioxidant CHCa biopolymer, representing a platform for the improvement of composite nanovesicle applicability.

Abstract:Abstract This work aimed to compare the performance of trans -ferulic acid-encapsulated nanostructured lipid carriers (NLCs) and solid lipid nanoparticles (SLNs) for transport by Caco-2 cells. The NLC particles (diameter: 102.6 nm) composed of Compritol ® 888 ATO, ethyl oleate, Cremophor ® EL, and Transcutol ® P were larger than the SLNs (diameter: 86.0 nm) formed without liquid lipid (ethyl oleate), and the former had a higher encapsulation efficiency for trans -ferulic acid ( p < 0.05). In vitro cultured Caco-2 cell transport was used to simulate intestinal absorption, and the cellular uptake of NLCs was higher than that of SLNs ( p < 0.05). Compared to SLNs, NLCs greatly enhanced trans -ferulic acid permeation through the Millicell TM membrane ( p < 0.05). This work confirms that NLCs have better properties than SLNs in terms of increasing drug transport by Caco-2 cells. This helps to comprehend the approach by which NLC-mediated oral bioavailability of trans -ferulic acid is better than that mediated by SLNs, as shown in our previous report.

Ferulic acid-loaded nanocapsules: Evaluation of mucosal interaction, safety and antioxidant activity in human mononucleated cells

Abstract:Ferulic acid (FA) is a phenolic compound that has antioxidant, anti-inflammatory and anticarcinogenic properties besides presenting cytoprotective activity. It has limited oral bioavailability what is a challenge to its therapeutic application. In this way, this investigation aimed to develop FA-loaded nanocapsule suspensions (NC-FA) prepared with ethylcellulose and evaluate their in vitro release profile, mucoadhesion and irritation potential; scavenging capacity, cytotoxicity, cytoprotection and genoprotection against hydrogen peroxide-induced damage in hMNC (human Mononucleated Cells) culture. The nanocapsules presented physicochemical characteristics compatible with colloidal systems (NC-FA: 112 ± 3 nm; NC-B (without FA): 107 ± 3 nm; PdI < 0.2; Span<2.0 and negative zeta potential). In addition, the nanoparticulate system promoted the FA controlled release, increasing the half-life twice through the in vitro dialysis method. NC-FA and NC-B were able to interact with mucin, which is an indicative of mucoadhesive properties and the association of FA with nanocapsules showed decreased irritation by HET-CAM method. Besides, the NC-FA did not present cytotoxicity in hMNC and improved the ATBS radical scavenging capacity. Besides, it prevented, treated and reversed oxidative conditions in a H2O2-induced model in hMNC. Thus, this nanocarrier formulation is promising to perform more preclinical investigations focusing on diseases involving oxidative mechanisms.
The Potential of Caffeic Acid Lipid Nanoparticulate Systems for Skin Application: In Vitro Assays to Assess Delivery and Antioxidant Effect

Abstract:The object of this study is a comparison between solid lipid nanoparticles and ethosomes for caffeic acid delivery through the skin. Caffeic acid is a potent antioxidant molecule whose cutaneous administration is hampered by its low solubility and scarce stability. In order to improve its therapeutic potential, caffeic acid has been encapsulated within solid lipid nanoparticles and ethosomes. The effect of lipid matrix has been evaluated on the morphology and size distribution of solid lipid nanoparticles and ethosomes loaded with caffeic acid. Particularly, morphology has been investigated by cryogenic transmission electron microscopy and small angle X-ray scattering, while mean diameters have been evaluated by photon correlation spectroscopy. The antioxidant power has been evaluated by the 2,2-diphenyl-1-picrylhydrazyl methodology. The influence of the type of nanoparticulate system on caffeic acid diffusion has been evaluated by Franz cells associated to the nylon membrane, while to evaluate caffeic acid permeation through the skin, an amperometric study has been conducted, which was based on a porcine skin-covered oxygen electrode. This apparatus allows measuring the O2 concentration changes in the membrane induced by polyphenols and H2O2 reaction in the skin. The antioxidative reactions in the skin induced by caffeic acid administered by solid lipid nanoparticles or ethosomes have been evaluated. Franz cell results indicated that caffeic acid diffusion from ethosomes was 18-fold slower with respect to solid lipid nanoparticles. The amperometric method evidenced the transdermal delivery effect of ethosome, indicating an intense antioxidant activity of caffeic acid and a very low response in the case of SLN. Finally, an irritation patch test conducted on 20 human volunteers demonstrated that both ethosomes and solid lipid nanoparticles can be safely applied on the skin.

Abstract:Niosomes are multilamellar vesicles that effectively transfer active ingredients into the skin's layers. To improve the active substance's penetration across the skin, these carriers are frequently utilized as topical drug delivery systems. Essential oils (EOs) have garnered significant interest in the field of research and development owing to their various pharmacological activities, cost-effectiveness, and simple manufacturing techniques. However, these ingredients undergo degradation and oxidation over time, leading to a loss of functionality. Niosome formulations have been developed to deal with these challenges. The main goal of this work was to create a niosomal gel of carvacrol oil (CVC) to improve its penetration into the skin for anti-inflammatory actions and stability. By changing the ratio of drug, cholesterol and surfactant, various formulations of CVC niosomes were formulated using Box Behnken Design (BBD). A thin-film hydration technique using a rotary evaporator was employed for the development of niosomes. Following optimization, the CVC-loaded niosomes had shown: 180.23 nm, 0.265, −31.70 mV, and 90.61% of vesicle size, PDI, zeta potential, and EE%. An in vitro study on drug release discovered the rates of drug release for CVC-Ns and CVC suspension, which were found to be 70.24 ± 1.21 and 32.87 ± 1.03, respectively. The release of CVC from niosomes best fit the Higuchi model, and the Korsmeyer–Peppas model suggests that the release of the drug followed the non-Fickian diffusion. In a dermatokinetic investigation, niosome gel significantly increased CVC transport in the skin layers when compared to CVC–conventional formulation gel (CVC-CFG). Confocal laser scanning microscopy (CLSM) of rat skin exposed to the rhodamine B-loaded niosome formulation showed a deeper penetration of 25.0 μm compared to the hydroalcoholic rhodamine B solution (5.0 μm). Additionally, the CVC-N gel antioxidant activity was higher than that of free CVC. The formulation coded F4 was selected as the optimized formulation and then gelled with carbopol to improve its topical application. Niosomal gel underwent tests for pH determination, spreadability, texture analysis, and CLSM. Our findings imply that the niosomal gel formulations could represent a potential strategy for the topical delivery of CVC in the treatment of inflammatory disease.

Abstract:Due to their poor solubility in hydrophilic matrices and chemical instability, the applications of quercetin and linseed oil in functional foods are limited. In this work we have encapsulated quercetin and linseed oil into NLC by high pressure homogenization technique. Quercetin and linseed oil co-loaded nanoparticles were less than 100 nm in diameter and spherical in shape. Nano-encapsulated quercetin was found to be presented in NLC with less ordered lipid cores as an encapsulated amorphous form. And the solubility of quercetin in hydrophilic matrices consisted of linseed oil and surfactants was improved by at least 1300 folds. The addition of linseed oil improved the in vitro antioxidant activities of quercetin loaded NLCs. Besides, the in vitro release of quercetin from NLC exhibited a sustained pattern. Lower lipid oxidation was found in quercetin and linseed oil co-loaded NLC compared with conventional linseed oil emulsion. Finally, storage study indicated that the fabricated NLCs were stable for more than 3 months at 25 °C. In conclusion, NLC could be a promising vehicle for delivery of hydrophobic bioactive compounds and ω-3 unsaturated fatty acids in food industry.

Abstract:Natural products are an invaluable source of diverse structures distinguished for their significant therapeutic potential. Specifically, essential oils (EO) are natural, complex liquids presenting various biological activities, such as antioxidant, antimicrobial and antifungal. Flavonoids are natural compounds with multiple pharmaceutical properties. The hydrophobicity and sensitivity of flavonoids and EOs, along with the high volatility of EOs impede their pharmaceutic, cosmetic and food applications, Thus, their encapsulation in a variety of carriers, in an effort to increase their bioavailability and stability is being sought out as a solution to the above problems. Solid Lipid Nanoparticles (SLNs) and Nanostructured Lipid Carriers (NLCs) are very promising drug delivery systems used for the controlled release and the effective protection of bioactive substances from external conditions, while presenting low toxicity and excellent biodegradability. This review covers the latest research on the encapsulation of essential oils and 10 different flavonoids in SLNs and NLCs, focused on the improvement of their biological responses and their potent applications.

Abstract:In the present study, nanostructured lipid carriers (NLC) composed of cetyl palmitate with various amounts of caprylic/capric triacylglycerols (as liquid lipid) were prepared and Coenzyme Q(10) (Q(10)) has been incorporated in such carriers due to its high lipophilic character. A nanoemulsion composed solely of liquid lipid was prepared for comparison studies. By photon correlation spectroscopy a mean particle size in the range of 180-240nm with a narrow polydispersity index (PI) lower than 0.2 was obtained for all developed formulations. The entrapment efficiency was 100% in all cases. The increase of oil loading did not affect the mean particle size of NLC formulations. NLC and nanoemulsion, stabilized by the same emulsifier, showed zeta potential values in the range -40/-50mV providing a good physical stability of the formulations. Scanning electron microscopy studies revealed NLC of disc-like shape. With respect to lipid polymorphism, a decrease in the ordered structure of NLC was observed with the increase of both oil and Q(10) loadings, allowing therefore high accommodation for Q(10) within the NLC. Using static Franz diffusion cells, the in vitro release studies demonstrated that Q(10)-loaded NLC possessed a biphasic release pattern, in comparison to Q(10)-loaded nanoemulsions comprising similar composition of which a nearly constant release was observed. The NLC release patterns were defined by an initial fast release in comparison to the release of NE followed by a prolonged release, which was dependent on the oil content.

Abstract:Alginate encapsulates loaded with clove essential oil (CEO) were prepared by ionic gelation, with subsequent freeze-drying. The objective of the present work was to develop a product with the ability to protect CEO against its easy volatility and oxidation. The following techniques were used to characterize the formulations: eugenol release, degree of swelling, GC/MS, TGA/DSC, and SEM. The alginate solution (1.0%) containing different concentrations of CEO (LF1: 1.0%; LF2: 0.5%; LF3: 0.1%) was dropped into a 3.0% CaCl2 solution. After lyophilization, the encapsulated samples were wrinkled and rigid, with high encapsulation power (LF3: 76.9% ± 0.5). Three chemical components were identified: eugenol (the major one), caryophyllene, and humulene. The antioxidant power (LF1: DPPH IC50 18.1 μg mL−1) was consistent with the phenol content (LF1: 172.2 mg GAE g−1). The encapsulated ones were thermally stable, as shown by analysis of FTIR peaks, eugenol molecular structure was kept unaltered. The degree of swelling was 19.2% (PBS). The release of eugenol (92.5%) in the PBS solution was faster than in the acidic medium. It was concluded that the low-cost technology used allows the maintenance of the content and characteristics of CEO in the three concentrations tested, offering a basis for further research with essential oil encapsulates.
