Abstract:As a measure towards achieving carbon neutrality and sustainability in the wastewater treatment industry, an autotrophic PN/HAP-A (airlift reactor/EGSB reactor) system is proposed in this study. The undiluted effluent from an AnMBR digesting municipal organic waste was applied for the practical validation of the system. With continuous feeding mode, the high efficiency, stability and operational simplicity were confirmed. In both reactors, the sludge manifested macro-aggregates and HAP-anammox granules with a satisfactory settling velocity and well-functional microorganisms. The alkalinity/IC consumptions and the buffer capacity were given individually and systematically. The elemental flow and potentials of precipitations indicated the high possibility and viability of phosphorus recovery by the formation of HAP, and in turn, the accumulation of precipitates in the biomass enhanced the settling velocity of the sludge. It is confirmed that the PN/HAP-A system is a promising approach to achieving nitrogen removal and phosphorus recovery from high-strength wastewater.
Nutrients removal from low C/N actual municipal wastewater by partial nitritation/anammox (PN/A) coupling with a step-feed anaerobic-anoxic-oxic (A/A/O) system

Abstract:In this study, a novel combined strategy was successfully established by partial nitritation/anammox (PN/A) within a step-feed A/A/O process integrated with fixed-biocarriers to treat municipal sewage for 200 days. The excellent nutrients removal performance of this system compared with national level of discharging standard were achieved: low total inorganic nitrogen (7.1 mg/L) and phosphorus (0.3 mg/L) in the effluent with the influent (51.1 and 4.2 mg/L) at C/N ratios of 3.4 ± 0.5, mainly attributed to the stable PN (oxic zone) and subsequently anammox effect (anoxic zone). Nitrogen mass balance indicated that anammox contribution in anoxic zones to nitrogen loss could be up to 42% at stable phase. Therefore, aeration and carbon cost could be greatly reduced under low DO, low C/N and aerobic hydraulic retention time (HRT) of 7.4 h condition. The low DO and anammox bacteria retention in anoxic chambers promoted the washout of NOB and combination of anammox and partial nitritation process. During long-term operation, the activity of AOB effectively maintained while that of NOB drastically reduced to 0.1 mg N / g MLSS / h resulting in high and stable nitrite accumulation ratios (about 90%). The achievement of partial nitritation was mainly due to low DO (0.4–0.5 mg/L) and effective retention of anammox bacteria even with a low temperature (14.5 °C). Notedly, anammox activity gradually increased both on the biocarriers and in the flocs while a higher anammox abundance was observed on the biocarriers (2.48%) than that in suspend flocs (0.03%). As above, this study indicated that the novel combined strategies could be applicable to mainstream anammox, and a pilot-scale reactor will be established to verify and promote the industrial application of mainstream anammox.

Abstract:A novel process that combines partial nitrification, fermentation and Anammox-partial denitrification (NFAD) was proposed to co-treat ammonia rich sludge supernatant (NH4+-N=1194.1 mg/L), external WAS (MLSS=22092.6 mg/L) and WWTP secondary effluent (NO3--N=58.6 mg/L). Three separated reactors were used for partial nitrification (PN-SBR), integrated fermentation and denitrification (IFD-SBR) and combined Anammox-partial denitrification (AD-UASB), respectively. The process resulted in excellent nitrogen removal efficiency (NRE) of 98.7%, external sludge reduction efficiency (SRE) of 44.6% and external sludge reduction rate of 4.1 kg/m3 after 200 days of continuous operation. IFD-SBR and AD-UASB contributed towards 89.4% and 9.2% nitrogen removal, respectively. In AD-UASB, cooperation between Anammox bacteria (4.1% Candidatus Brocadia) and partial denitrifying bacteria (3.2% Thauera) resulted in significant stability of Anammox pathway, which contributed up to 84.1% nitrogen removal in the combined Anammox-partial denitrification process. NFAD saved up to 100% organic resource demand and 25% of aeration consumption compared with the traditional nitrification-denitrification process.

Abstract:Achieving simultaneous semi-partial nitrification and deep phosphorus removal is a preferred process technology for Anammox pretreatment. In this study, semi-partial nitrification combined with in-situ phosphorus recovery (PNPR) was used to treat municipal wastewater. The SRT conflict between the nitrification and phosphorus removal was resolved by in-situ phosphorus recovery every 20 cycles of Anaerobic/Oxid, and a supernatant with more than 10 times the influent phosphorus concentration was obtained, thus achieving bio-enhanced phosphorus removal and recovery with satisfactory semi-partial-nitrification effluent. Interestingly, the results showed that phosphorus removal and recovery process could improve the activity of AOB. The PNPR system's nitrite accumulation rate (NAR) and phosphorus removal rate (PRR) were more than 90% each, whereas the relative abundance of AOB and PAOs increased from 0.04% to 0.74% and from 0.25% to 0.70%, respectively (P < 0.01). Furthermore, on average, the NO 2 – -N eff /NH 4 + -N eff value was 1.96, which laid the foundation for the subsequent anammox treatment.

Abstract:Although the partial nitrification-anammox process (PN-A) has achieved great success in nitrogen removal for the high ammonium concentration wastewater, its application is still limited in low ammonium concentration wastewater treatment due to its instability and low nitrogen removal efficiency. In this study, a sequencing batch reactor (SBR) with continuous aeration was employed to enrich ammonia oxidation bacteria (AOB) and suppress nitrite oxidation bacteria (NOB) first; then, the SBR was operated intermittently aerated SBR (IASBR), to which the anammox granular sludge (AMX) was added to achieve complete autotrophic nitrogen removal under low influent ammonium concentration of 100 mg/L. A mathematical model was used to optimize the IASBR aeration strategy to achieve sub-optimal nitrogen removal. The experimental results showed that high nitrite accumulation efficiency (above 80%) in the SBR and a fast start-up within 100 days and a stable TN (total nitrogen) removal efficiency of 70% were achieved in the IASBR. Meanwhile, the simulation results indicated that keeping aeration duration at 4 h, k L a (oxygen transfer coefficient) at 50 day −1 , or aeration duration at 2.5 h, k L a at 80 day −1 could obtain a higher total nitrogen removal efficiency (TNR) (TNR > 80%), and the TN removal could also be improved by increasing hydraulic retention time (HRT) under the optimal oxygen supply rate.

Abstract:Recently, it was reported that for synthetic low-strength wastewater, the excellent nitrogen removal rate (NRR) accompanied with phosphorus removal could be achieved through the partial nitritation/anammox (PNA)-hydroxyapatite (HAP) process. Thus, this research further investigated the performance of the pilot-scale PNA-HAP process treating the effluent of an anaerobic membrane bioreactor (AnMBR) fed with the actual municipal wastewater. The results showed that with the hydraulic retention time of 4.0 h, the influent ammonium concentration ranging from 36.0 to 41.0 mg/L, and the BOD5 ranging from 6.3 to 12.7 mg/L, the average NRR and the nitrogen removal efficiency was 0.13 kg/m3/d and 63.38%, respectively. The specific activity test of sludge confirmed that the PNA process was the main nitrogen metabolism pathway. The effluent nitrate and the BOD5 were almost zero, indicating the existence of denitrification activity in reactor. Given that the oxygenation condition, the heterotrophic organic matter oxidization activity also occurred in reactor. The sludge analysis confirmed the phosphate formation in sludge. Thus, in the reactor, four kinds of biological activities and chemical crystallization occurred harmoniously in sludge. From the mixed liquid volatile suspended solid of 2.4 g/L and the low distribution range of granule size, it was obvious that the sludge had a high dispersity. Based on the well settling ability of sludge during the operation, it was inferred that there was a close bond between biomass and HAP in sludge, which was helpful to enhance the settleability of sludge granule. Besides, the phosphorus-containing sludge was suitable as the fertilizer. In all, this study demonstrated that the PNA-HAP process is an ideal alternative treating the effluent of the AnMBR process in the municipal wastewater treatment.

Abstract:High solid anaerobic digestion (HSAD) achieves the benefits of high volumetric loading rates and lower reject water production, which, however, results in much more concentrated reject water with a remarkable increase in organics and nitrogen compared with that from conventional AD with low solid content. The high concentrations of ammonium (2000-3500 mg/L) and COD (3000-4000 mg/L) were reported to exert inhibition on anammox bacteria (AnAOB), posing challenges to the application of the partial nitritation/anammox (PN/A). To date, no cases of PN/A process start-up for sludge HSAD reject water were reported. This study demonstrated the start-up process of a 480 m3/d PN/A project without anammox sludge inoculation and treating HSAD reject water from a centralized dewatered sludge treatment plant. The project did not construct new infrastructures but utilized previously constructed tanks to upgrade the process from existing short-cut nitrification-denitrification to a two-stage PN/A process. Although no external anammox sludge inoculation was performed to save seeding sludge cost, the start-up was successfully achieved in about 9 months (273 days) based on a three-step method of "AnAOB enrichment - sludge acclimation - capacity doubling". During start-up, the relative abundance of AnAOB (Candidatus_Kuenenia) increased from near zero to 12.0%. After start-up, the total inorganic nitrogen (TIN) removal load reached 0.74 kgN/(m3•d), with a total nitrogen removal efficiency of over 90%. Compared to the traditional nitrification-denitrification process, the PN/A process remarkably reduces the addition of organic chemicals and aeration energy consumption, saving approximately 4.2 million yuan (RMB) in operational costs annually. In summary, this research provides a full-scale reference for the start-up of the PN/A process treating sludge HSAD reject water.
Enhancing nitrogen removal performance through intermittent aeration in continuous plug-flow anaerobic/aerobic/anoxic process treating low-strength municipal sewage

Abstract:Advanced nitrogen removal cannot be achieved through the conventional biological nitrogen removal process, which requires higher carbon sources and aeration energy. The proposal of intermittent aeration in the aerobic chambers offered an innovative approach to enhance nitrogen removal in low carbon-to-nitrogen ratio (C/N) municipal sewage, using a plug-flow reactor with anaerobic/aerobic/anoxic (AOA) process. Due to the effective utilization of internal carbon sources through the intermittent aeration, the total inorganic nitrogen removal efficiency (NRE) increased to 77.9 ± 3.2 % with the mean aerobic hydraulic retention time of only 3.2 h and a low C/N of 3.3 during the operation of 210 days. Polyhydroxyalkanoates dominated the nitrogen removal in this AOA system, accounting for 48.0 %, primarily occurring in the alternant aerobic/anoxic chambers. Moreover, the microbial community structure remained unchanged while the NRE increased to 77.9 %. This study provided an efficient and economic strategy for the continuous plug-flow AOA process.
An Innovative Process for Mature Landfill Leachate and Waste Activated Sludge Simultaneous Treatment Based on Partial Nitrification, In Situ Fermentation, and Anammox (PNFA)

Abstract:An innovative partial nitrification, in situ fermentation, and Anammox (PNFA) system was developed to achieve mature landfill leachate and waste activated sludge simultaneous treatment. Three separate sequencing batch reactors (SBRs) were used for partial nitrification (PN-SBR), integrated fermentation-denitrification (IFD-SBR), and partial nitrification-Anammox (PNA-SBR). After 200 days of continuous operation, a satisfactory nitrogen removal efficiency (NRE) of 99.2 ± 0.1% was obtained, with an effluent total nitrogen (TN) of 15.2 ± 3.2 mg/L. In IFD-SBR, the volatile fatty acids generated from fermentation drove efficient denitrification, obtaining sludge and nitrogen reduction rates of 4.2 ± 0.7 and 0.61 ± 0.04 kg/m3·day, respectively. Furthermore, unwanted fermentation metabolites (134.1 mg/L NH4+-N) were further treated by PNA-SBR using a combination of step-feed and intermittent aeration strategies. In PNA-SBR, Anammox significantly contributed to 82.1% nitrogen removal, and Anammox bacteria (Candidatus Brocadia, 2.3%) mutually benefited with partially denitrifying microorganisms (Thauera, 4.2%), with 66.3% of generated nitrate reduced to nitrite and then reutilized in situ by Anammox. Compared with the conventional nitrification-denitrification process, PNFA reduced oxygen energy consumption, external carbon source dosage, and CO2 emission by 21.3, 100, and 38.9%, respectively, and obtained 50.1% external WAS reduction efficiency.

Abstract:Metastasis is a complex process that is regulated by multiple signaling pathways, with the focal adhesion kinase (FAK)/paxillin pathway playing a major role in the formation of focal adhesions and cell motility. N-myc downstream regulated gene-1 (NDRG1) is a potent metastasis suppressor in many solid tumor types, including prostate and colon cancer. Considering the antimetastatic effect of NDRG1 and the crucial involvement of the FAK/paxillin pathway in cellular migration and cell-matrix adhesion, we assessed the effects of NDRG1 on this important oncogenic pathway. In the present study, NDRG1 overexpression and silencing models of HT29 colon cancer and DU145 prostate cancer cells were used to examine the activation of FAK/paxillin signaling and the formation of focal adhesions. The expression of NDRG1 resulted in a marked and significant decrease in the activating phosphorylation of FAK and paxillin, whereas silencing of NDRG1 resulted in an opposite effect. The expression of NDRG1 also inhibited the formation of focal adhesions as well as cell migration and cell-collagen adhesion. Incubation of cells with novel thiosemicarbazones, namely di-2-pyridylketone 4,4-dimethyl-3-thiosemicarbazone and di-2-pyridylketone 4-cyclohexyl-4-methyl-3-thiosemicarbazone, that upregulate NDRG1 also resulted in decreased phosphorylation of FAK and paxillin. The ability of these thiosemicarbazones to inhibit cell migration and metastasis could be mediated, at least in part, through the FAK/paxillin pathway.
Evaluation of the economic and environmental benefits of partial nitritation anammox and partial denitrification anammox coupling preliminary treatment in mainstream wastewater treatment

Abstract:Anaerobic ammonium oxidation (anammox) technology is considered the best alternative for current wastewater treatment to improve resource recovery and energy efficiency. Evaluation of the economic and environmental benefits of partial denitrification anammox (PDA) and partial nitritation anammox (PNA) from a plant-wide perspective, which are the two most common mainstream anammox processes, is crucial for guiding the development direction of anammox, but research is still lacking. In this study, two plant-wide processes with chemically enhanced primary treatment-PDA (CEPT-PDA) and high rate activated sludge-PNA (HRAS-PNA) as the core units respectively, were firstly proposed and analysed, and life cycle assessment (LCA) was conducted to assess environmental impacts (EIs). The results of energy and economic analysis showed that the net energy consumption of CEPT-PDA and HRAS-PNA was 0.11 kwh/m 3 and 0.16 kwh/m 3 respectively; however, the operating cost of CEPT-PDA was 16.54% higher than that of HRAS-PNA due to the requirement for more chemicals. Compared with CEPT-PDA, the total EIs of HRAS-PNA decreased by 14.80%. The EI categories related to human toxicity and ecotoxicity contributed greatly to total EIs, accounting for 95.81% in CEPT-PDA and 96.75% in HRAS-PNA. Major environmental impacts were attributed to coagulant and energy consumption in two processes. In addition, improving the iron recycling efficiency was more beneficial for CEPT-PDA and further narrowed the gap between CEPT-PDA and HRAS-PNA on EIs. This study identifies that CEPT-PDA is competitive in the engineering application of mainstream anammox but reducing iron addition is a prerequisite.

Abstract:The advanced nitrogen removal of mature landfill leachate (MLL) with low carbon to nitrogen ratio (C/N) is a topic worthy of further research. A two-stage partial nitrification/anammox (PN/A) system for integrated MLL and kitchen waste digestion liquid (KWDL) treatment was established in this study. The nitrogen removal efficiency (NRE) of this process was 98.1 ± 1.3 %, with an effluent TIN concentration of 15.2 ± 1.8 mg/L. In the partial nitrification-sequencing batch reactor (PN-SBR), aerobic hydraulic retention time (HRT) accounted for just 36.4 % of the whole process with dissolved oxygen (DO) < 0.3 mg/L, which significantly reduced energy consumption. Furthermore, the introduction of KWDL heightened anoxic denitrification and aerobic simultaneous nitrification and denitrification (SND) processes, contributing to 74.2 ± 4.3 % and 25.8 ± 4.3 % of the total NRE, respectively. This greatly strengthened the nitrogen removal capacity of PN-SBR. Batch tests indicated that the capacity of KWDL as a carbon source to drive the process of partial denitrification coupled with anammox (PDA) was equal to that of sodium acetate. Adding KWDL in the later phase of each cycle in the anammox-sequencing batch reactor (A-SBR) facilitated achieving stable PDA and advanced nitrogen removal. The nitrate accumulation problem in the traditional two-stage PN/A process has been resolved without the addition of carbon sources. Nitrosomonas (3.7 %→7.9 %), Thauera (0.04 %→40.7 %) and a salt-tolerant AnAOB microbe, unclassified_f_Brocadiaceae (0 %→5.3 %) were dominant in the combined system. This study revealed a pioneering avenue for efficient high-strength wastewater treatment.

Abstract:In this study, an innovative partial nitrification-anammox (PN/A) and endogenous partial denitrification-anammox (EPD/A) process was developed in a single-stage integrated fixed film activated sludge sequencing batch reactor (IFAS-SBR) treating real municipal wastewater with C/N ratio below 3.2. Enhanced efficiency of total nitrogen (TN) removal reached 90.1% with low HRT of 12 h and DO of 0.4 ± 0.1 mg/L. Detailed nitrogen removal mechanism analysis of typical cycle revealed that 89.9% of TN was eliminated through anammox pathway. Anammox bacteria (Candidatus Brocadia) and endogenous denitrifying bacteria (Candidatus Competibacter) were abundant both in biofilms and suspended sludge, meanwhile ammonium-oxidizing bacteria has outcompeted nitrite-oxidizing bacteria, which all favored the synergistic effect of anammox with PN and EPD and contributed to the improvement of nitrogen removal. Overall, the above results confirmed that combined PN/A and EPD/A process is a reliable and efficient alternative for mainstream anammox process.

A sustainable integrated anoxic/aerobic bio-contactor process for simultaneously in-situ deodorization and pollutants removal from decentralized domestic sewage

Abstract:The integration of anoxic filter and aerobic rotating biological contactor shows promise in treating rural domestic sewage. It offers high efficiency, low sludge production, and strong shock resistance. However, further optimization is needed for odor control, pollutant removal, and power consumption. In this study, the investigation on a one-pump-drive lab-scale device of retention anoxic filter (RAF) integrated with hydraulic rotating bio-contactor (HRBC) and its optimal operation mode were conducted. During the 50-day operation, optimal operation parameters were investigated. These parameters included a 175 % reflux ratio (RR), 5-h hydraulic retention time in the RAF (HRTRAF), and 2.5-h hydraulic retention time in the HRBC (HRTHRBC). Those conditions characterized a micro-aerobic environment (DO: 0.6-0.8 mg/L) in RAF, inducing improved deodorization (89.3 % sulfide removal) and denitrification (85.9 % nitrate removal) simultaneously. During the operation period, 84.79 ± 3.87 % COD, 82.71± 2.06 % NH4+-N, 74.83 ± 2.06 % TN, 91.68± 2.12 % S2-, and 89.04 ± 1.68 % TON were removed in RAF-HRBC. Based on large amount of operational data, organic loading rate curves of RAF-HRBC were validated and calibrated as a crucial reference to aid in full-scale designs and applications. The richness of microbial community was improved in both RAF and HRBC. In the RAF, the autotrophic sulfide-oxidizing nitrate-reducing bacteria (a-son) and heterotrophic sulfide-oxidizing nitrate-reducing bacteria (h-son) were selectively enriched, which intensified the sulfide removal and denitrification process. In the two-stage HRBC system, the 1st stage RBC was primarily composed of organics degraders, while the 2nd stage RBC consisted mainly of ammonium oxidizers. Overall, the integrated RAF-HRBC process holds significant potential for simultaneously improving pollutant removal and in-situ odor mitigation in decentralized domestic sewage treatment. This process specifically contributes to enhancing environmental sustainability and operational efficiency.
Integrated Thp-Ad Centrate and Municipal Wastewater Treatment Via Partial Nitritation/Anammox (Pn/A) Process: A Promising Approach to Alleviate Inhibitory Effects and Enhance Nitrogen Removal

Abstract:Two-stage Partial nitritation/Anammox (PN/A) was firstly performed for recalcitrant organics (RO)-rich thermal hydrolysis pretreated anaerobic digestion (THP-AD) centrate treatment with municipal wastewater (MW) as co-substrate. Results indicated the inhibitory effects of RO was alleviated and high nitrate issue in PN/A effluent was addressed by cotreatment strategy. Stable PN with nitrite accumulation ratio of 95% and N removal efficiency of 97.1% were well maintained at MW of 80%. Nevertheless, nitrate accumulation and anammox activity loss were observed with lowering MW proportions owing to the weakened denitrification activity and aggravated inhibitory effect. Microbial analysis revealed Nitrosomonas was the major ammonium oxidizing bacteria and the ideal PN performance was due to the effective out-selection of nitrite oxidizing bacteria. Candidatus Kuenenia was identified as the primary bacteria for nitrogen removal (82.7%), and the controlled abundance of heterotrophic denitrifiers in anammox system ensured the enhanced nitrogen removal regardless of high COD loading from THP-AD centrate.
Nitrogen degradation performance and microbial dynamic analysis for the efficient combination of partial denitrification and anammox (PD/A) process

Abstract:The coupling partial denitrification and anammox (PD/A) is a promising technology for deep-level nitrogen removal, and the quickly effective startup remains a huge challenge in real application. In this study, the stability of nitrite (NO 2 - -N) accumulation was investigated in the PD sequential batch reactor (PSBR), and the nitrogen removal performance in response to nitrogen loading rate (NLR) was compared in another independent anammox SBR (ASBR), where the combined PD/A operation was subsequently carried out based on the optimal operating conditions. In the PSBR, nitrate removal efficiency (NRE) maintained around 89–90% with a mean nitrate-to-nitrite transformation ratio (NTR) of 84.43%, and the total proportion of functional bacteria (e.g., Terrimonas , norank_f_A4b , Ellin6067 , Defluviicoccus , etc) accounted for 40.98% after long-term acclimation. In the ASBR, increased NLR (0.22 → 0.70 kgN/(m 3 ·d)) depressed the anammox activity owing to the sludge disintegration and adverse nitrogen contribution, however, the dominant Candidatus_Brocadia enriched from 0.50% to 2.05% and realized the efficient retention of functional bacteria, which also laid a good foundation for the rapid startup of PD/A process. As NLR controlled around 0.18 - 0.25 kgN/(m 3 ·d), the total nitrogen removal (Re TN ) climbed from 33.88% to 98% with anammox contribution soaring up to 65%, since the higher tight-bound extracellular polymeric substances (T-EPS) production and protein/polysaccharide (PN/PS) ratio accelerated the re-formation of anammox granular sludge. In the end, the potential significances and challenges of PD/A process were further summarized for the mainstream wastewater treatment.

Abstract:For achieving efficient and robust treatment of domestic sewage with C/N around 2.8, this study innovatively developed an integrated fermentation, partial-nitrification, partial-denitrification and anammox (IFPNDA) process based on the Anaerobic Baffled Reactor and Continuous-flow Stirred Tank Reactor (ABR-CSTR) bioreactor. Desirable N-removal efficiency of 87.5 ± 2.1% was obtained without external organics, correspondingly effluent total nitrogen (TN) concentration reached 6.1 ± 0.7 mg/L. The N-removal stability was greatly facilitated by the effective linkage between partial nitrification (PN) process and partial denitrification (PD) process in emergency. Highly enriched hydrolytic bacteria (6.9%) and acidogenic bacteria (5.7%) in A1, especially Comamonas (2.8%) and Longilinea (3.5%), induced the significant increase of volatile fatty acids (VFAs) in domestic sewage. Thauera (6.1%) in A2 and Nitrosomonas (5.4%) in A3 acted as the dominant flora of nitrite supplies for anammox in IFPNDA process. Candidatus_Brocadia (2.4%) dominated the advanced nitrogen removal. The IFPNDA process exhibited much potential for achieving energy neutrality during wastewater treatment.

Abstract:Slices of striatal tissue from newborn to eight-day-old rats were cultured for six to 47 days. Cholinergic neurons and fibres were then visualized by histochemical staining for acetylcholinesterase or immunocytochemical staining for choline acetyltransferase. GABA-containing neurons and fibres were visualized by immunocytochemical staining for glutamate decarboxylase or GABA. Corresponding to the normal postnatal development in vivo, acetylcholinesterase staining of the striatal tissue progressed from a "patchy" distribution in the six to 14 days old cultures to an almost even distribution of high acetylcholinesterase activity after 18-27 days. Extrinsic afferents were accordingly not necessary for the maintenance of a patch-matrix-like, acetylcholinesterase distribution during the first one to two weeks in culture, just as a subsequent, normal developmental change of the acetylcholinesterase staining pattern into a more homogeneous distribution also occurred without such afferents. Cholinergic, choline acetyltransferase-immunoreactive neurons were evenly distributed within the cultured striatal tissue, like in vivo, but the density of the neurons appeared to be higher in the cultures. The neurons had a morphology corresponding to the "classical", large-sized, aspiny, cholinergic interneurons in the adult rat striatum. Glutamate decarboxylase-immunoreactive and GABA-immunoreactive neurons were either lightly or darkly stained and of medium size, but some large, lightly stained glutamate decarboxylase-immunoreactive and GABA-immunoreactive neurons were also found. The difference in staining density among the medium-sized cells was observed with both antisera and hence provide evidence for the existence of two populations of medium-sized GABAergic neurons, which in vivo are intensely stained interneurons and more weakly stained, spiny projection neurons. Fibres stained better for glutamate decarboxylase than for GABA and outgrowth of glutamate decarboxylase-immunoreactive nerve fibres from the striatal slice cultures onto the coverslip was often observed. The presence at all culture periods of "protospines" on cell bodies and proximal dendrites of some glutamate decarboxylase-immunoreactive, and in particular some GABA-immunoreactive neurons, suggested that at least some developmental characteristics might be maintained for extended periods in culture. In several cultures, groups of small GABA-immunoreactive cells were observed. Similar groups were also found by staining for glutamate decarboxylase, but a smaller proportion of the cells were then positively stained. In view of their immature appearance with few or no processes, the known presence of GABA in neuroblast-like cells, and the recent demonstration of neuronal and glial progenitor cells in the adult mouse striatum, the small cells might belong to a population of undifferentiated cells surviving in the slice cultures.(ABSTRACT TRUNCATED AT 400 WORDS)

Abstract:A novel technology was developed to achieve partial nitrification at moderately low DO and short HRT, which would save the aeration cost and have the capacity to treat a wide range of low-strength real wastewater. The process enables a relatively stable whereas nitrite accumulation rate (NO2-AR) was stabilized over 94% in the last aerobic basin on average of each phase through a combination of short HRT and low DO level. Low DO did not produce sludge with poorer settleability. The morphology and internal structure of the granular sludge was observed by using a scanning electron microscope (SEM) analysis during a long-term operation. The images indicated that thick clusters of spherical cells and small rod-shaped cells (NOB and AOB are rod-shaped to spherical cells) were the dominant population structure, rather than filamentous and other bacteria under a combination of low DO and short HRT, which gives a good indication of nitrite accumulation achievement. MPN method was used to correlate AOB numbers with nutrient removal. It showed that an ammonia-oxidizing bacterium (AOB) was the dominant nitrifying bacteria, whereas high NO2-AR was achieved at AOB number of 5.33×108 cell/g MLSS. Higher pollutant removal efficiency of 86.2%, 98% and 96.1%, for TN, NH4+-N, and TP, respectively, was achieved by a novel six basin activated sludge process (SBASP) at low DO level and low C/N ratio which were approximately equal to the complete nitrification-denitrification with the addition of sodium acetate (NaAc) at normal DO level of (1.5–2.5 mg/L).
