Abstract:The alternating aerobic/anaerobic biofilm system had been applied for phosphorus (P) enrichment and recovery because of the advantage of low energy consumption and high efficiency. The metal ions and N-acyl-L-homoserine lactones (AHLs) in system were studied to better clarify the mechanism of P uptake/release under metal ion stress. The results indicated that the increase of metal ions stimulated the release of AHLs, and AHLs-guided quorum sensing (QS) enhanced P uptake. Moreover, biomineralization could stimulate the increase of P content in biofilm (P biofilm ). Meanwhile, some ortho-p was converted to short-chain poly-p in extracellular polymer substance (EPS), and others were transferred into cell through EPS to synthesize poly-p. With the P biofilm increased, more P could be absorbed/released due to the shift in the metabolic model of polyphosphate accumulating organisms (PAOs). The release of AHLs between microorganisms was also inhibited when PAOs reached the state of P saturation (75.6 ± 2.5 mg/g SS), which meant that the effect of signaling function would tend to stabilize, and the 169.2 ± 2.6 mg/L P concentration in the enriched solution was obtained due to the P release was inhibited. Moreover, P was rapidly transferred to the new enriched solution after the P was recovered, and PAOs restored its capability of P uptake/release. In addition, 31 P-NMR analysis demonstrated that EPS played a major role in PAOs compared to cell, and inorganic phosphorus (IP) played an essential role in the uptake/release of P compared to organic phosphorus (OP). Furthermore, the microbiological analysis showed that Candidatus Accumulibacter was positively correlated with AHLs (P < 0.05). This study provided essential support for clarifying the P metabolism mechanism of PAOs.

Abstract:The formation of dominant phosphate-accumulating organisms (PAOs) is essential for the high enrichment of phosphate in biofilm sequencing batch reactors (BSBR) for phosphorus recovery. The dominant PAOs in the biofilm process have not been isolated and purified, and the key metabolic pathways that promote the formation of dominant PAOs are still unclear. In this study, four strains of highly-efficient PAOs were obtained by an innovative isolation procedure. The relationship between the abundance of highly-efficient and dominant PAOs and the phosphate removal ability was compared. We found that the abundance of PAOs was positively correlated with the phosphate removal efficiency in vitro pure culture and complex biofilm process. Metagenomics analysis revealed that compared with highly-efficient PAOs cultured in vitro , dominant PAOs in biofilms had unique key metabolic pathways, F-ATPases and N-Acyl homoserine lactones (AHLs). F-ATPases are important for maintaining the proton motive force (PMF) required for the uptake of carbon sources by PAOs, and AHLs are participating in phosphate metabolism through quorum sensing (QS) mediated secretion of extracellular polymeric substance (EPS). The formation of dominant PAOs was promoted by optimizing carbon source uptake and phosphate metabolism. This study revealed that the difficult isolation of dominant PAOs was due to the AHLs-mediated QS, and we identified the key pathways regulating the formation of dominant PAOs in biofilms through genomics analysis. Our findings provide insights in enhancing phosphate enrichment in BSBR by modulating the components of microbial community under the low concentration of carbon source consumption.

Synergistic Effect of Chitosan-Based Sludge Aggregates CS@NGS Inoculum Accelerated the Start-Up of Biofilm Reactor Treating Aquaculture Effluent: Insights into Performance, Microbial Characteristics, and Functional Genes.

Abstract:The moving bed bioreactor (MBBR) process has drawn more attention as a promising biological wastewater treatment process. Nevertheless, achieving quick start-up and microbial biofilm formation remains a significant challenge. Consequently, the present study investigated a novel chitosan-based natural sludge (CS@NGS) seeding strategy for the accelerated start-up of MBBR. Three identical bioreactors were employed; the first bioreactor was without sludge seed as the control (BR1), the second was inoculated only with sludge (BR2), and the third was inoculated with CS@NGS according to the proposed seeding method (BR3). All bioreactors were utilised to treat simulated recirculating aquaculture systems (RAS) effluent. Resultantly, the CS@NGS shortened the start-up period from over twenty to seven days due to the enhanced initial microbial adhesion and biofilm formation. Under optimal conditions, the ammonium removal in BR3 approached 100%, which was relatively higher than BR2 (96.35 +/- 1.12%) and BR1 (92.56 +/- 2.17%). Moreover, a low nitrite accumulation was exhibited in the effluents, approximately <= 0.03 mg L-1. The process performance correlated positively with core bacteria from the genera Nakamurella , Hyphomicrobium , Nitrospira , Paenarthrobacter , Rhodococcus , and Stenotrophobacter. The quantitative polymerase chain reaction (qPCR) results demonstrated that the CS@NGS enhanced the expressions of amoA, nxrB, nirK, nirS, narG, and napA nitrogen metabolism-related functional genes to varying degrees. The present study findings can assist the rapid start-up of aquaculture biofilters utilised to solve high nitrite and ammonia accumulation in recirculated water from industrial RAS.

Abstract:Two biofilm sequencing batch reactors (BSBRs) were conducted under alternating aerobic/anaerobic (Ae/An) sequence and alternating anaerobic/aerobic (An/Ae) sequence to investigate the effects of operation sequences on phosphorus removal and recovery performance. The Ae/An BSBR had advantages in phosphate enrichment, phosphorus concentration of enrichment solution can reach 99.08 mg/L. While An/Ae BSBR exhibited a higher kinetic rate for phosphate uptake and release up to 0.48 mmol/gVSS. In An/Ae BSBR, the remarkable increase of phosphate concentration in aerobic stage put a great strain on the phosphate uptake of biofilms that led to an increase of phosphorus accumulation in the biofilm (P biofilm ). However, the poly-P proportion in TP biofilm decreased which triggered the metabolisim of polyphosphate accumulating organisms (PAOs) shifting from polyphosphate accumulating metabolism (PAM) to glycogen accumulating metabolism (GAM) and suppressed the phosphorus enrichment performance of the An/Ae BSBR. The phosphate uptake rate (PUR) of biofilms was in alignment with the bulk phosphate concentration. PAOs were dominated by Rhodoplanes and Saprospiraceae in the Ae/An BSBR, whereas Thiothrix was predominant in the An/Ae BSBR. Notably, the Candidatus Competibacter, considered as a denitrifying glycogen accumulating organisms (DGAOs), showed a dominant proportion in the microbial community of both BSBRs. The higher expression of actP in the An/Ae system indicated a greater capacity for the uptake of acetate, while the Ae/An system was is more adept for degrading acetate represented by the higher relative abundance of ackA , pta and acsA genes The high phosphorus loading in the An/Ae system suppressed the ppk expression but enhanced the ppx expression.

Abstract:Biofilm sequencing batch reactor (BSBR) can achieve efficient phosphate (P) removal and enrichment, but its process performance and metabolic mechanisms for P removal and enrichment of municipal wastewater remain largely unclear. In the present study, we assessed the P removal and enrichment of municipal wastewater at influent P concentrations of 2.5 mg/L and 10 mg/L. The efficiency of P removal and enzyme activity in polyphosphate-accumulating organisms (PAOs) and glycogen-accumulating organisms (GAOs) were compared, and the growth and metabolic characteristics of dominant PAOs and GAOs at different influent P concentrations were studied with the macro-sequencing technology. The results showed that the P recovery efficiencies were 70.03% and 76.19% when the influent P concentration was 2.5 mg/L and 10 mg/L in BSBR, respectively, and the maximum P concentration of recovery liquid was 81.29 mg/L and 173.12 mg/L, respectively. There were no phosphate kinase (PPK) and phosphate hydrolase (PPX) in extracellular polymeric substances (EPS). The dominant PAOs were Candidatus_Contendobacter, Dechloromonas, and Flavobacterium, and the dominant GAO was Candidatus_Competibacter. The abundance of Candidatus_Contendobacter was the highest with the most potential contribution to P removal. PAOs had competitive advantages in carbon (C) source uptake, glycogen metabolism, P metabolism, and adenosine triphosphate (ATP) metabolism. HMP was unique to PAOs, EMP had the highest abundance in glycogen metabolism, and ED was contained in PAOs of BSBR. These results indicated that BSBR provided sufficient reducing power and ATP for PAOs through different glycogen decomposition pathways to promote P uptake and obtained competitive advantages in P metabolism, C source uptake, and ATP utilization to achieve efficient P removal and enrichment. Collectively, our current findings provided valuable insights into the P removal and enrichment mechanism of BSBR in municipal sewage.

Abstract:This study assessed the impact of the operating conditions of the biofilm sequencing batch reactor (BSBR) on the community structure and the growth/metabolic pathways of its polyphosphate-accumulating organisms (PAOs). There are significant difference with reference to the enhanced biological phosphorus removal (EBPR) process. The leading PAOs in BSBR generally are capable of high affinity acetate metabolism, gluconeogenesis, and low affinity phosphate transport, and have various carbon source supplementation pathways to ensure the efficient circulation of energy and reducing power. A new model of the metabolic mechanism of PAOs in the BSBR was formulated, which features low glycogen metabolism with simultaneous gluconeogenesis and glycogenolysis and differs significantly from the classic mechanism based on Candidatus_Accumulibacter and Tetrasphaera . The findings will assist the efficient recovery of low concentration phosphate in municipal wastewater.

Phosphate recovery from urban sewage by the biofilm sequencing batch reactor process: Key factors in biofilm formation and related mechanisms

Abstract:The biofilm sequencing batch reactor (BSBR) technique has been deployed in the laboratory to enrich phosphorus from simulated wastewater, but it is still not clear what its performance will be when real world sewage is used. In this work, the effluent from the multi-stage anoxic-oxic (AO) activated sludge process at a sewage plant was used as the feed water for a BSBR pilot system, which had three reactors operating at different levels of dissolved oxygen (DO). The phosphorus adsorption and release, the biofilm growth, and the extracellular polymeric substances (EPS) components and contents were examined. The microbial communities and the signaling molecules N-acyl-l-homoserine lactones (AHLs) were also analyzed. Gratifyingly, the BSBR process successfully processed the treated sewage, and the biofilm developed phosphorus accumulation capability within 40 days. After entering stable operation, the system concentrated phosphate from 2.59 ± 0.77 mg/L in the influent to as much as 81.64 mg/L in the recovery liquid. Sludge discharge had profound impacts on all aspects of BSBR, and it was carried out successfully when the phosphorus absorption capacity of the biofilm alone was comparable to that of the reactor containing the activated sludge. Shortly after the sludge discharge, the phosphate concentration of the recovery liquid surged from 50 to 140 mg/L, the biofilm thickness grew from 20.56 to 67.32 μm, and the diversity of the microbial population plunged. Sludge discharge stimulated Candidatus competibacter to produce a large amount of AHLs, which was key in culturing the biofilm. Among the AHLs, both C10-HSL and 3OC12-HSL were significantly positively correlated with EPS and the abundance of Candidatus competibacter. The current results demonstrated BSBR as a viable option to enrich phosphorus from real world sewage with low phosphorus content and fluctuating chemistry. The mechanistic explorations also provided theoretical guidance for cultivating phosphorus-accumulating biofilms.

Abstract:A high phosphate (P) recovery concentration was achieved in pilot-scale biofilm sequencing batch reactor (BSBR) with a low carbon source (C) cost. Especially, a high-abundance glycogen-accumulating organisms (GAOs) (13.93–31.72%) was detected that was accompanied by a high P recovery concentration of BSBR. High-abundance GAOs obtain additional C through various C compensation pathways (split tricarboxylic acid cycle (TCA cycle), glyoxylate shunt and gluconeogenesis), thus reducing the need to compete with polyphosphate-accumulating organisms (PAOs) for C and weakening the adverse effects on P recovery by PAO cells. Under the action of N -acyl homoserine lactones (AHLs)-mediated quorum sensing (QS), GAOs promoted the secretion of a large amount of extracellular polymeric substances (EPS), which helped to realize the P recovery of EPS-dominated biofilms (68.02%–96.89%). This study provides a low-carbon technology for the recovery of high concentration P from municipal wastewater, and improves the ecological theory of P recovery in collaboration with GAOs and PAOs.

Abstract:The biofilm sequencing batch reactor (BSBR) can simultaneously remove and enrich phosphorus from wastewater highly efficiently. However, the mechanism of phosphorus transformations in the biofilm remains to be clarified. This present work examines the biofilm in the BSBR, to reveal the changes of phosphorus in the cells and the extracellular polymeric substances (EPS) and to assess how the metabolic characteristics of intracellular polymers (polyhydroxyalkanoate, glycogen, polyphosphorus) affect the migration and transformation of phosphorus. When the BSBR reactor is under steady operation (20 °C, DO = 6 mg·L−1, the influent phosphorus concentration = 10 mg·L−1, and the aerobic phosphorus removal efficiency >95 %), the phosphorus in the biofilm is mainly distributed in the EPS, as the total phosphorus of EPS account for 83.20 %–86.97 % of the total phosphorus of the biofilm. Phosphorus speciation shows that the biofilm realizes the absorption and release of phosphorus through the combined action of EPS and cells. The cells provide the primary driving force, and the EPS play a key role. The intracellular polyphosphorus is closely related to polyhydroxyalkanoate (PHA) but not glycogen. High-throughput sequencing demonstrates that the microbial population in the biofilm is dominated by the bacteria that secrete EPS that can enhance to phosphorus removal.

Abstract:The present study focused on investigating the effect of dissolved oxygen (DO) concentration on phosphorus (P) transformation and metabolism in the polyphosphate-accumulating organisms (PAOs)-biofilm system. Two biofilm sequencing batch reactors (BSBRs) with high-DO (6 mg/L, BSBR-H) versus low-DO (2 mg/L, BSBR-L) concentrations were typically operated for 90 days. Collectively, the high DO concentration in aerobic phase would force PAOs to preferentially and rapidly perform phosphate metabolism and be beneficial for them to establish the dominant ecological niche. Clear differences in stoichiometry and kinetics indicated a glycolytic pathway in BSBR-L, while it shifted to polyphosphate hydrolysis in BSBR-H. The expression of functional genes suggested that the high-affinity P transport system was mainly responsible for P transformation in BSBR-H while the low-affinity P transport system was accountable for BSBR-L. Moreover, the BSBR-H showed relatively higher abundances of Pseudomonas while Tetrasphaera were the most dominant PAOs in BSBR-L. Dechloromonas contributed substantially to poly-P metabolism by occupying the largest proportion of ppk1/2 and ppx genes in both BSBRs.

Abstract:We report on magneto-oscillations in differential resistivity of a two-dimensional electron system subject to intense microwave radiation. The period of these oscillations is determined not only by microwave frequency but also by its intensity. A theoretical model based on quantum kinetics at high microwave power captures all important characteristics of this phenomenon which is strongly nonlinear in microwave intensity. Our results demonstrate a crucial role of the multiphoton processes near the cyclotron resonance and its harmonics in the presence of strong dc electric field and offer a unique way to reliably determine the intensity of microwaves acting on electrons.
Bacterial Enrichment in Highly-Selective Acetate-Fed Bioreactors and Its Application in Rapid Biofilm Formation.

Abstract:In this study, we systematically investigated the bacterial community dynamics in highly-selective (strong hydraulic selection pressure and high organic loading rate) bioreactors with acetate as the sole carbon source. 16S rRNA gene high-throughput sequencing and metagenomic sequencing results showed that phenolics-degrading bacteria (PDB), which were mainly Acinetobacter species, in the newly-formed aerobic granules could account for >70% of the total bacteria. Near full-length 16S rRNA gene sequences obtained by cloning suggest that the PDB are potentially novel species because they are distantly related to known Acinetobacter species. However, these PDB only temporarily appeared in the early stage of the granule formation and their abundance quickly decreased along the reactor operation. To retain these PDB, we demonstrated that the newly-formed aerobic granules could accelerate biofilm formation in moving bed biofilm reactors (MBBRs), and the biofilm carriers showed gradually-increased phenol degradation performance in the MBBRs. While, the bacterial community in biofilm significantly changed during the operation process of the MBBRs and the community structure became more complicated than that in the aerobic granules. Collectively, this study provides new insights into the microbial ecology of sludge granulation and biofilm formation process in the wastewater treatment systems for remediating phenolic matters.
Clarification of the phosphorus release mechanism for recovering phosphorus from biofilm sludge in alternating aerobic/anaerobic biofilm system

Abstract:A novel wastewater treatment plant process was constructed to overcome the challenge of simultaneous nitrate removal and phosphorus (P) recovery. The results revealed that the P and nitrate removal efficiency rose from 39.0 % and 48.4 % to 92.8 % and 93.6 % after 136 days of operation, and the total P content in the biofilm (TP biofilm ) rose from 15.8 mg/g SS to 57.8 mg/g SS. Moreover, the increase of TP biofilm changed the metabolic mode of denitrifying polyphosphate accumulating organisms (DPAOs), increasing the P concentration of the enriched stream to 172.5 mg/L. Furthermore, the acid/alkaline fermentation led to the rupture of the cell membrane, which released poly-phosphate and ortho-phosphate of cell/EPS in DPAOs and released metal‐phosphorus (CaP and MgP). In addition, high-throughput sequencing analysis demonstrated that the relative abundance of DPAOs involved in P storage increased, wherein the abundance of Acinetobacter and Saprospiraceae rose from 8.0 % and 4.1 % to 16.1 % and 14.0 %. What's more, the highest P recovery efficiency (98.3 ± 1.1 %) could be obtained at optimal conditions for struvite precipitation (pH = 7.56 and P: N: Mg = 1.87:3.66:1) through the response surface method (RSM) simulation, and the precipitates test analysis indicated that P recovery from biofilm sludge was potentially operable. This research was of great essentiality for exploring the recovery of P from biofilm sludge.

Abstract:In bioaugmented wastewater treatment systems, it is essential for recalcitrant pollutant-degrading bacteria to form biofilms. Inducing biofilm formation in these bacteria, however, is challenging as it involves multiple inter-related regulating pathways and environmental factors. Herein, we report the remarkable promoting effect of Ca2+ on biofilm formation of two novel pyridine-degrading bacteria with poor innate biofilm-forming capabilities, Pseudomonas sp. ZX01 and Arthrobacter sp. ZX07. The roles of Ca2+ in different biofilm development stages were investigated. Our data showed strong influences of Ca2+ on the initial attachment of the two strains onto positively charged glass surfaces by altering cell surface charge as well as the cation bridging effect. Contrary to many other biofilm promoting mechanisms, Ca2+ downregulated the extracellular polymeric substances (EPS) production per cell in both Pseudomonas sp. ZX01 and Arthrobacter sp. ZX07, while increasing biofilm biomass. This is attributed to the strong cationic bridging between Ca2+ and EPS which can elevate the efficiency of the extracellular products in binding bacterial cells. Furthermore, Ca2+ increased the protein-to-polysaccharide (PN/PS) ratio in biofilm EPS of both strains, which favored cell aggregation, and biofilm establishment by increasing the hydrophobicity of cell surfaces. More intriguingly, the intracellular c-di-GMP, which can drive the switch of bacterial lifestyle from planktonic state to biofilm state, was also elevated markedly by exogenous Ca2+. Taken together, these results would be of guidance for applying the two strains into bioaugmented biofilm reactors where Ca2+ supplement strategy can be employed to facilitate their biofilm formation on the surfaces of engineering carriers.

Abstract:To investigate the diversities of Accumulibacter phosphatis and its polyhydroxyalkanoate (PHA) synthase gene (phaC) in enhanced biological phosphorus removal (EBPR) sludge, an acetate-fed sequencing batch reactor was operated. Analysis of microbial communities using fluorescence in situ hybridization and 16S rRNA gene clone libraries showed that the population of Accumulibacter phosphatis in the EBPR sludge comprised more than 50% of total bacteria, and was clearly divided into two subgroups with about 97.5% sequence identity of the 16S rRNA genes. PAO phaC primers targeting the phaC genes of Accumulibacter phosphatis were designed and applied to retrieve fragments of putative phaC homologs of Accumulibacter phosphatis from EBPR sludge. PAO phaC primers targeting G(1PAO), G(2PAO), and G(3PAO) groups produced PCR amplicons successfully; the resulting sequences of the phaC gene homologs were diverse, and were distantly related to metagenomic phaC sequences of Accumulibacter phosphatis with 75-98% DNA sequence identities. Degenerate NPAO (non-PAO) phaC primers targeting phaC genes of non-Accumulibacter phosphatis bacteria were also designed and applied to the EBPR sludge. Twenty-four phaC homologs retrieved from NPAO phaC primers were different from the phaC gene homologs derived from Accumulibacter phosphatis, which suggests that the PAO phaC primers were specific for the amplification of phaC gene homologs of Accumulibacter phosphatis, and the putative phaC gene homologs by PAO phaC primers were derived from Accumulibacter phosphatis in the EBPR sludge. Among 24 phaC homologs, a phaC homolog (G1NPAO-2), which was dominant in the NPAO phaC clone library, showed the strongest signal in slot hybridization and shared approximately 60% nucleotide identity with the G(4PAO) group of Accumulibacter phosphatis, which suggests that G1NPAO-2 might be derived from Accumulibacter phosphatis. In conclusion, analyses of the 16S rRNA and phaC genes showed that Accumulibacter phosphatis might be phylogenetically and metabolically diverse.
Strengthening the enrichment of anaerobic ammonia oxidizing bacteria in biofilms through sludge concentration control

Abstract:Controlling sludge concentration is an effective means to achieve PN. In this article, the reactor used domestic sewage as raw water and promoted the high enrichment of anammox bacteria by controlling the MLVSS of flocs to 1000-1500 mg/L and increasing the concentration of filler sludge. The measures to reduce the concentration of flocculent sludge increased the proliferation rate of the biofilm and provided sufficient substrate for AnAOB. After 102 days of operation, the abundance of Candidatus Brocadia increased from 0.43% during inoculation to 23.56% in phase VI. The ability of the microbial community to utilize energy metabolism and produce ATP was significantly improved, and the appropriate distribution of anammox bacteria and nitrifying, denitrifying bacteria in the ecological niche led to its high enrichment. In summary, this study proposes a strategy to promote the high enrichment of anammox bacteria in mainstream domestic sewage without adding any chemicals.

Abstract:Biofilms are economical and efficient nitrogen removal technologies utilized in water treatment. However, their practical applications are limited by the slow colonization and growth of film-forming bacteria, as well as the long start-up process. Quorum sensing (QS) signaling molecules play a vital role in regulating various microbial behaviors. This study investigated the impact of the exogenous addition of the QS signaling molecule 3-oxo-C14-HSL on the start-up and biofilm growth of aquaculture moving bed biofilm reactors (MBBR). The results showed that introducing 400 ng/L of 3-oxo-C14-HSL significantly increased the adhesion of aquaculture effluent (AE) microorganisms to cell culture plates (72.9–74.2 %, P < 0.01). By adding 3-oxo-C14-HSL, the thickness and weight of the MBBR carrier biofilm, as well as the secretion of extracellular polymeric substances (EPS) and AHLs, were significantly increased, significantly shortening biofilm formation time. Metagenomic analysis showed that under the regulation of exogenous 3-oxo-C14-HSL, the relative abundance of major nitrogen-removing bacteria, nitrogen metabolism, and QS-related genes in the biofilm significantly increased, improving the performance of the MBBR in the treatment of AE. The total nitrogen, chemical oxygen demand, and nitrate removal rates were as high as 90.30 %, 93.47 %, and 97.35 %, respectively. This study revealed a strong correlation between QS signaling molecules based on AHLs and MRRB biofilm formation, nitrogen metabolism, and microbiota reconstruction, providing a potential biofilm enhancement strategy for MBBR treatment of AE with practical applications.

Abstract:The extremely slow growth rate of anaerobic ammonia oxidation (anammox) bacteria limits full-scale application of anammox process worldwide. In this study, extracellular polymeric substances (EPS)-coated polypropylene (PP) carriers were prepared for biofilm formation. The biomass adhesion rate of EPS-PP carrier was 12 times that of PP carrier, and EPS-PP achieved significant enrichment of E. coli BY63. The 120-day continuous flow experiment showed that the EPS-PP carrier accelerated the formation of anammox biofilm, and the nitrogen removal efficiency increased by 10.5 %. In addition, the abundance of Candidatus Kuenenia in EPS-PP biofilm was 27.1%. Simultaneously, amino acids with high synthesis cost and the metabolites of glycerophospholipids related to biofilm formation on EPS-PP biofilm were significantly up-regulated. Therefore, EPS-PP carriers facilitated the rapid formation of anammox biofilm and promoted the metabolic activity of functional bacteria, which further contributed to the environmental and economic sustainability of anammox process.

Abstract:To balance the high phosphorus concentration in recirculated solution and the stability of biofilm system, this study explored the performance and mechanism of phosphorus uptake/release for recovering phosphorus from sewage when the phosphorus content in biofilm (Pbiofilm) changed. The results showed that the maximum phosphorus concentration in the concentrated solution reached 171.2 ± 2.5 mg·L-1 in harvest 1st-5th stages. Polyphosphate accumulating organisms (PAOs) performed a metabolic shift from glycogen accumulation metabolism (GAM) to polyphosphate accumulation metabolism (PAM) when Pbiofilm increased at each phosphorus enrichment stage, and more phosphorus was absorbed/released by PAOs. Nevertheless, the release of poly-phosphate from PAOs was inhibited after phosphorus concentration stabilized, and PAOs were unable to absorb phosphorus from wastewater as it reached the phosphorus saturation stage. To maintain the stability of the system, phosphorus had to be harvested so that the saturated phosphorus in PAOs was easily released in a new recirculated solution, resulting in adequate storage space for PAOs to absorb phosphorus. Meanwhile, the 31P NMR analysis demonstrated that phosphorus was stored in EPS and cell of PAOs, whereas EPS played a significant role than cell at the anaerobic phase. Particularly, ortho-phosphate was the major component of phosphorus release by EPS and poly-phosphate was the major part of phosphorus release by cell. Furthermore, the change of Pbiofilm had no impact on biofilm characteristics and microbial communities, whereas some PAOs would be enriched, and others that were not suitable for this process would be inhibited with repeated cycles of alternating aerobic/anaerobic operation.
