Abstract:Introduction: This study assesses the environmental impacts of mannosylerythritol lipids (MELs) production for process optimization using life cycle assessment (LCA). MELs are glycolipid-type microbial biosurfactants with many possible applications based on their surface-active properties. They are generally produced by fungi from the family of Ustilaginaceae via fermentation in aerated bioreactors. The aim of our work is to accompany the development of biotechnological products at an early stage to enable environmentally sustainable process optimization. Methods: This is done by identifying hotspots and potentials for improvement based on a reliable quantification of the environmental impacts. The production processes of MELs are evaluated in a cradle-to-gate approach using the Environmental Footprint (EF) 3.1 impact assessment method. The LCA model is based on upscaled experimental data for the fermentation and purification, assuming the production at a 10 m3 scale. In the case analyzed, MELs are produced from rapeseed oil and glucose, and purified by separation, solvent extraction, and chromatography. Results: The results of the LCA show that the provision of substrates is a major source of environmental impacts and accounts for 20% of the impacts on Climate Change and more than 70% in the categories Acidification and Eutrophication. Moreover, 33% of the impacts on Climate Change is caused by the energy requirements for aeration of the bioreactor, while purification accounts for 42% of the impacts respectively. For the purification, solvents are identified as the main contributors in most impact categories. Discussion: The results illustrate the potentials for process optimization to reduce the environmental impacts of substrate requirements, enhanced bioreactor aeration, and efficient solvent use in downstream processing. By a scenario analysis, considering both experimental adaptations and prospective variations of the process, the laboratory development can be supported with further findings and hence efficiently optimized towards environmental sustainability. Moreover, the presentation of kinetic LCA results over the fermentation duration shows a novel way of calculating and visualizing results that corresponds to the way of thinking of process engineers using established environmental indicators and a detailed system analysis. Altogether, this LCA study supports and demonstrates the potential for further improvements towards more environmentally friendly produced surfactants.

Abstract:Enzyme biocatalysis is well known for its biodegradability and environmentally friendly conditions. Its novel properties such as high reaction selectivity and mild reaction conditions prove that it is a promising alternative that can overcome the global resource scarcity of the conventional chemical catalysts that are of a nonrenewable source. In this study, we documented the advancements and breakthroughs in emerging biocatalysis reactor technologies spanning over a decade. We aim to encapsulate the remarkable progress and innovations witnessed within this dynamic field. Thus, we reviewed 48 articles related to enzyme biocatalysis in the production of glycerides using reactors. The reaction parameters using a biocatalysis route were reviewed systematically and discussed in our study. In the biocatalysis transformation, the processes mainly focus on the conversion of raw material into product. In the optimization of biocatalysis, temperature and residence time parameters have been thoroughly researched in order to maximize the productivity of the enzyme activity. In the selection of reactors, the type of reactor and its agitator play a vital role in enzyme activity. We anticipate that the greenness of biocatalysis transformation could be improved if the enzymes in waste streams could be converted from another reaction to functional products. This work may serve as a stepping stone for the biocatalysis industry, acting as a precursor for industries or individuals interested in exploring this evergreen field.

Abstract:Using selected bio‐based feedstocks as alternative to fossil resources for producing biochemicals and derived materials is increasingly considered an important goal of a viable bioeconomy worldwide. However, to ensure that using selected bio‐based feedstocks is aligned with the global sustainability agenda, impacts along the entire life cycle of biochemical production systems need to be evaluated. This will help to identify those processes and technologies, which should be targeted for optimizing overall environmental sustainability performance. To address this need, we quantify environmental impacts of biochemical production using distinct bio‐based feedstocks, and discuss the potential for reducing impact hotspots via process optimization. Lactic acid was used as example biochemical derived from corn, corn stover and Laminaria sp. as feedstocks of different technological maturity. We used environmental life cycle assessment, a standardized methodology, considering the full life cycle of the analyzed biochemical production systems and a broad range of environmental impact indicators. Across production systems, feedstock production and biorefinery processes dominate life cycle impact profiles, with choice in energy mix and biomass processing as main aspects influencing impact results. Results show that uncertainty increases with decreasing technological maturity. When using Laminaria sp. (least mature among selected feedstocks), impacts are mainly driven by energy utilities (up to 86%) due to biomass drying. This suggests to focus on optimizing or avoiding this process for significantly increasing environmental sustainability of Laminaria sp.‐based lactic acid production. Our results demonstrate that applying life cycle assessment is useful for identifying environmental impact hotspots at an earlier stage of technological development across biochemical production systems without the aim of identifying the most environmentally friendly bio‐feedstock. With that, our approach contributes to improving the environmental sustainability of future biochemicals production as part of moving toward a viable bioeconomy worldwide.

Abstract:BackgroundCellulase enzymes have been reported to contribute with a significant share of the total costs and greenhouse gas emissions of lignocellulosic ethanol production today. A potential future alternative to purchasing enzymes from an off-site manufacturer is to integrate enzyme and ethanol production, using microorganisms and part of the lignocellulosic material as feedstock for enzymes. This study modelled two such integrated process designs for ethanol from logging residues from spruce production, and compared it to an off-site case based on existing data regarding purchased enzymes. Greenhouse gas emissions and primary energy balances were studied in a life-cycle assessment, and cost performance in a techno-economic analysis.ResultsThe base case scenario suggests that greenhouse gas emissions per MJ of ethanol could be significantly lower in the integrated cases than in the off-site case. However, the difference between the integrated and off-site cases is reduced with alternative assumptions regarding enzyme dosage and the environmental impact of the purchased enzymes. The comparison of primary energy balances did not show any significant difference between the cases. The minimum ethanol selling price, to reach break-even costs, was from 0.568 to 0.622 EUR L−1 for the integrated cases, as compared to 0.581 EUR L−1 for the off-site case.ConclusionsAn integrated process design could reduce greenhouse gas emissions from lignocellulose-based ethanol production, and the cost of an integrated process could be comparable to purchasing enzymes produced off-site. This study focused on the environmental and economic assessment of an integrated process, and in order to strengthen the comparison to the off-site case, more detailed and updated data regarding industrial off-site enzyme production are especially important.

Early-stage economic and environmental impact assessment for optimized bioprocess development: Monoterpenoid indole alkaloids

Abstract:Microbial refactoring offers sustainable production of plant-sourced pharmaceuticals associated with high production costs, ecological harms, and supply chain dependencies. Here, microbial tabersonine production in Saccharomyces cerevisiae is modeled during early-stage development (TRL: 3-5), guiding decisions for process-scale economic and environmental optimization. The base-case 0.7 mg/L titer indicated a minimum selling price (MSP) of $3,910,000/kg and global warming potential (GWP) of 2,540 kgCO2eq/g. The industrial process at 1 g/L resulted in an MSP of 4,262 $/kg and a GWP of 6.36 kgCO2eq/g. Location analysis indicated a sustainability trade-off between France, USA, Poland, and China, with the written order of declining MSP and increasing GWP. Continuous processing promised reducing the MSP by 18-27 %, and the GWP by 17-31 %. In-situ product extraction during fermentation was estimated to lower the MSP by 41-61 %, and the GWP by 30-75 %. In addition to showcasing a combined TEA-LCA on biopharmaceuticals, the early-stage assessment approach guides bioprocess optimization.
Environmental Impact Assessment of a Plant Cell-Based Bio-Manufacturing Process for Producing Plant Natural Product Ingredients

Abstract:Purpose: This study employed a Life Cycle Assessment (LCA) methodology to evaluate the environmental impacts of a novel plant cell-based biomanufacturing process for producing plant natural product ingredients. The primary purpose was to assess the relative sustainability of the process and to provide insights into potential areas of improvement in the biomanufacturing process. Method: The LCA method used an MS Excel (Ver. 2407) -based approach with a cradle-to-gate system boundary covering raw material sourcing (A1), raw material transportation (A2), and product extract manufacturing (A3) stages. Energy use and material inventory data are presented for different unit operations, and environmental impact factors were obtained from the Ecoinvent database. The study included a Material Circularity Index (MCI) calculation to assess the circularity of the biomanufacturing process for the production of saponin emulsifiers that are normally extracted from the woody tissue of the Chilean soapbark tree (Quillaja saponaria). Comparative analyses were performed against a wild-harvest approach for plant tannin extraction from spruce (Picea abies) tree bark. Key Results: The environmental impact assessment focused on determining relative Global Warming Potential (GWP), Acidification Potential (AP), Freshwater Eutrophication (FE), Particulate Matter Formation (PMF), and Ozone Depletion Potential (ODP). Results indicated that the extract manufacturing stage (A3) contributed significantly to adverse environmental impacts, with varying levels of effects based on the energy source used. Comparative analysis with the wild harvest approach highlights the lower environmental impact of the alternative biomanufacturing process. The biomanufacturing process showed a 23% reduction in GWP, AP, and FE and a 25% reduction in PMF and ODP relative to the wild harvest approach. However, the MCI for the biomanufacturing process was estimated to be 0.186, indicating a low material circularity. Conclusions: The results revealed that the extract manufacturing stage, particularly energy consumption, significantly influences the relative environmental impacts of the alternative production processes. Different energy sources exhibit varying effects, with renewable energy sources showing lower environmental impacts. The Material Circularity Index indicated a low circularity for the biomanufacturing process, suggesting opportunities for improvement, such as incorporating recycled or reused materials. Compared with the tannin extraction process, the plant cell-based biomanufacturing process demonstrated lower environmental impacts, emphasising the importance of sustainable practices and the use of renewable energy sources in future plant natural product sourcing. Recommendations include implementing more sustainable practices, optimising raw material choices, and extending product life spans to enhance circularity and overall environmental benefits.

Environmental performance of new processes for the production of fructo-and galacto-oligosaccharides (FOS and GOS)

Abstract:The prebiotics like FOS and GOS are receiving special attention in the food industry due to their health benefits. They can be produced by enzymatic synthesis by using disaccharides or other substrates as raw materials or by extraction and hydrolysis from different natural sources (roots, legumes). The environmental footprints of these different production schemes are lacking to provide guidance for the ecodesign of such new production processes. In this work, Life Cycle Assessment (LCA) was undertaken to analyze and compare the production of FOS and GOS by enzymatic synthesis from glucose (to get FOS) or lactose (to get GOS) and hydrolytic production from extraction of yacon potato (to get FOS) or chickpea (to get GOS). A cradle-to-gate approach was considered in the two scenarios under assessment (the phases of use and/or final disposal of FOS/GOS were not considered). The functional unit was defined as 100 g of FOS/GOS produced. LCAs were performed using data collected at the laboratory scale, supplemented with data from Ecoinvent database. SimaPro was used for the LCA modeling with the midpoint impact EF2.0 characterization method. Results showed that the main environmental hotspot was the production of yacon potato or chickpea used in the hydrolysis process. For this reason, the hydrolytic process caused higher environmental burdens than the enzymatic synthesis process. Chickpea production causing more impacts than yacon potato production, GOS production generated more environmental impacts than FOS production. When produced by enzymatic synthesis, FOS and GOS were the sources of similar environmental impacts. From a process point of view, special attention must be paid on three specific stages of production: time of synthesis, freeze-drying and purification of the final product. The environmental load of these stages was associated to high energy consumption and huge amount of ethanol requirement. The results from this study helped to identify the stages requiring special efforts to ecodesign the production of FOS and GOS at pilot scale in the future. Further research should primarily be focused in the reduction of the biomass used and corresponding solid waste generated during the hydrolytic production. Furthermore, environmental assessment should be included at each development of the process to ensure its efficient eco-design.

Assessing the environmental sustainability of glucose from wheat as a fermentation feedstock

Abstract:In recent years, the need to substitute fossil fuels with renewable biomass has been a key driver in the development of the biorefinery concept. One of the possible routes towards the development of bioproducts under this scheme is through a sugar intermediate. Sugars, such as glucose, can be produced through starch crops, for instance wheat. While there are many environmental assessment studies that consider sugar as a platform for biofuel production, the main focus is on the end product of the value chain (typically bioethanol), but not on sugars as the basic feedstock. Taking the bottom-up perspective as a roadmap, the assessment of technological, economic and environmental barriers in the biorefinery scheme must take into account the sustainability of sugar production with the aim of improving its current framework or finding novel technologies. This study investigates the environmental sustainability of wheat cultivation and grain processing in different European countries by applying the life cycle assessment (LCA) methodology with a cradle to gate approach. Moreover, 1 kg of glucose at the factory gate has been considered as a functional unit for reporting the environmental results. The chosen impact categories are climate change (CC), particular matter (PM), human toxicity (HT), freshwater eutrophication (FE), terrestrial eutrophication (TE), acidification (AC) and abiotic depletion (AD). Mass and economic allocations are evaluated as the processing of grain wheat generates different valuable by-products, namely wheat bran, gluten meal and gluten feed. The results show that agricultural activities play an important role in the environmental impacts, predominantly due to the production of agrochemicals and field emissions derived from fertilisation. In addition, the choice of allocation has a greater environmental influence (both positive or negative) on the by-products than the main product under investigation.

Abstract:Enzymatic biocatalysis typically generates less waste, uses less water, and minimizes energy consumption compared to traditional chemical methods. Efficient, cell‐free biosynthesis relies on the reuse of its valuable biocatalysts. Immobilization of enzymes on solid supports, such as enzyme carrier resins (ECRs), offers a reliable and widely deployed approach to maximize enzyme turnover in cell‐free biosynthesis. We focus on two major bottlenecks associated with optimizing cell‐free biocatalysis. First, we apply our lab's 3D‐printed labware to screen ECRs in 96‐well mini‐reactors to optimize enzyme immobilization conditions. Second, we introduce inline infrared spectroscopy to monitor bioreactor output and maximize enzyme productivity. Urease provides a model system for examining immobilization conditions and continuous assessment of biocatalyst performance. As required for the high substrate concentrations to improve process efficiency and minimize waste, urease was studied in unusually high concentrations of its substrate—molar concentrations of urea. The optimized reactor processed 3.24 L of 4.00 M urea at an average volumetric productivity of 13 g·L‐1·h‐1 over 18 h and achieved an estimated productivity number of >17.4 kg urea processed per g of immobilized urease Type‐IX. This workflow can be generalized to most biocatalytic processes and could accelerate adoption of cell‐free biosynthesis for greater chemical sustainability.

Abstract:Since the industrial revolution, the rapid growth and development of global industries have depended largely upon the utilization of coal-derived chemicals, and more recently, the utilization of petroleum-based chemicals. These developments have followed a linear economy model (produce, consume, and dispose). As the world is facing a serious threat from the climate change crisis, a more sustainable solution for manufacturing, i.e., circular economy in which waste from the same or different industries can be used as feedstocks or resources for production offers an attractive industrial/business model. In nature, biological systems, i.e., microorganisms routinely use their enzymes and metabolic pathways to convert organic and inorganic wastes to synthesize biochemicals and energy required for their growth. Therefore, an understanding of how selected enzymes convert biobased feedstocks into special (bio)chemicals serves as an important basis from which to build on for applications in biocatalysis, metabolic engineering, and synthetic biology to enable biobased processes that are greener and cleaner for the environment. This review article highlights the current state of knowledge regarding the enzymatic reactions used in converting biobased wastes (lignocellulosic biomass, sugar, phenolic acid, triglyceride, fatty acid, and glycerol) and greenhouse gases (CO2 and CH4) into value-added products and discusses the current progress made in their metabolic engineering. The commercial aspects and life cycle assessment of products from enzymatic and metabolic engineering are also discussed. Continued development in the field of metabolic engineering would offer diversified solutions which are sustainable and renewable for manufacturing valuable chemicals.This article has not yet been cited by other publications.

Abstract:The signs and treatment of abnormal reactions following the use of organophosphorus systemic warble fly dressings are recorded. A review is provided of all complaints received by the manufacturers and distributors of such compounds during the dressing periods for the years 1975 to 1978. The volume of warble fly dressing sold each year was known and the number of cattle dressed was based on the calculation of the number of adults which could have been treated with the volume. Complaints were divided according to the main signs reported and the most common complaint was that of paralysis-incoordination. The incidence of death or slaughter following use of an organophosphorus compound was one in 93,000 animals treated. The majority of animals showing abnormal signs other than paralysis-incoordination were involved in incidences of skin lesions, depressed milk yield, bloat or 'other reactions'. Compared with the number of animals treated, the level of reported reaction was low (one complaint per 46,000 animals treated).

They show high potential for application as alternatives to conventional oleochemical- or petrochemical surfactants. To ensure their environmentally friendly performance, we aimed to assess CL production from a life cycle perspective at an early developmental stage to identify process steps that have the highest impact on the environment. With this information, optimisation approaches can be derived.

Abstract:This work describes the evaluation of the solid-state fermentation (SSF) bioprocess utilizing brewery spent grain (BSG) and apple pomace (AP) as carbon sources and matrices for microorganism growth to produce xylanase, pectinase, and cellulase. The process was assessed at a larger scale by designing a packed column-type bioreactor equipped with sensors for monitoring critical parameters such as CO2 concentration, humidity, and temperature. Then, process simulation was used to evaluate the techno-economic feasibility of the bioprocess at an industrial scale. The analysis centered on evaluating which formulation, primarily containing xylanase (scenario 1), pectinase (scenario 2), or cellulase (scenario 3), yielded the most promising results for advancing to the commercial stage. Additionally, a sensitivity analysis was conducted to explore the influence of variations in raw material costs and enzyme prices. The obtained results at a higher scale were within the expected results obtained under optimum conditions. Scenario 1 exhibited strong economic viability with further optimization potential (base case: 5000 kg/batch with an ROI of 37.59 %, payback time of 2.66 years, IRR of 26.8%, and net present value of USD 7,325,537). The sensitivity analysis revealed that changes in enzyme prices, particularly xylanase, could significantly influence the process's profitability. This study also demonstrated the potential for cost optimization by selecting a more cost-effective inoculum media and optimizing water usage to enhance process efficiency and sustainability.

Sustainable enzymatic treatment of organic waste in a framework of circular economy

Abstract:Enzymatic treatment of food and vegetable waste (FVW) is an eco-friendly approach for producing industrially relevant value-added products. This review describes the sources, activities and potential applications of crucial enzymes in FVW valorization. The specific roles of amylase, cellulase, xylanase, ligninase, protease, pectinase, tannase, lipase and zymase enzymes were explained. The exhaustive list of value-added products that could be produced from FVW is presented. FVW valorization through enzymatic and whole-cell enzymatic valorization was compared. The note on global firms specialized in enzyme production reiterates the economic importance of enzymatic treatment. This review provides information on choosing an efficient enzymatic FVW treatment strategy, such as nanoenzyme and cross-linked based enzyme immobilization, to make the process viable, sustainable and cheaper. Finally, the importance of life cycle assessment of enzymatic valorization of FVW was impressed to prove this approach is a better option to shift from a linear to a circular economy.

Abstract:Microbial cell factories are emerging as powerful tools in addressing pressing environmental challenges and promoting sustainable biomanufacturing. This paper highlights the key role of engineered microorganisms in mitigating pollution, converting waste and pollutants into valuable products, and reducing reliance on fossil fuels. Through advancements in metabolic engineering, gene editing technologies, and synthetic biology, microbial cell factories are being optimized to enhance their efficiency in breaking down pollutants and producing renewable chemicals, such as biofuels, bioplastics, and specialty chemicals. Detailed case studies illustrate successful implementations, such as the conversion of lignocellulosic biomass, CO 2 , and industrial waste into high-value products, underscoring the practical impact of microbial cell factories in diverse sectors, including energy, materials, and chemicals.Furthermore, this research addresses the challenges faced by microbial cell factories in industrial-scale operations, such as maintaining genetic stability and optimizing growth conditions, and offers insight into emerging technological solutions to these obstacles. By providing a comprehensive overview of recent developments and identifying future research directions, this paper offers actionable recommendations for unlocking the full potential of microbial biotechnology. These efforts aim to further integrate microbial processes into industrial systems, contributing to a more sustainable and resilient global economy, with the ultimate goal of fostering a circular bioeconomy.

Abstract:A wide range of chemicals and materials can be produced from renewable feedstocks through bioconversions. An iterative, progressively detailed technology screening approach was developed to identify the most promising candidates. Initially, candidates are selected into a portfolio based on their future potential as large-volume industrial chemicals or materials. Second, the candidates are ranked with respect to a simple economic criterion based on the market value, the price of the starting material, and the product yield. Simple comparisons are then made, where possible, between producing the product via the most competitive conventional route and via the proposed bioprocessing route. Next, qualitative information is gathered from industrial experts on the advantages and disadvantages of each product with respect to energy impacts, environmental quality, and economic competitiveness. Finally, engineering and economic evaluations are performed for the most promising candidates to assess the profitability of the bioprocessing route and to identify the research and development opportunities that have the greatest impact on energy savings, environmental quality, and economics. Forty chemicals and materials that could potentially be produced from renewable feedstocks were initially selected for evaluation by this methodology. From this, succinic acid was chosen for the first more detailed evaluation based on the initial screening results. The approach described in this article could be used by potential industrial producers to complement their forecasting of the technical and economic feasibility of producing chemicals and materials from renewable resources, and by researchers to identify opportunities for focused research and development.

Production of chemicals and utilities in-house improves the environmental sustainability of phytoplankton-based biorefinery

Abstract:Life cycle assessment was used to evaluate the environmental impacts of phytoplanktonic biofuels as possible sustainable alternatives to fossil fuels. Three scenarios were examined for converting planktonic biomass into higher-value commodities and energy streams using the alga Scenedesmus sp. and the cyanobacterium Arthrospira sp. as the species of interest. The first scenario (Sc-1) involved the production of biodiesel and glycerol from the planktonic biomass. In the second scenario (Sc-2), biodiesel and glycerol were generated from the planktonic biomass, and biogas was produced from the residual biomass. The process also involved using a catalyst derived from snail shells for biodiesel production. The third scenario (Sc-3) was similar to Sc-2 but converted CO2 from the biogas upgrading to methanol, which was then used in synthesizing biodiesel. The results indicated that Sc-2 and Sc-3 had a reduced potential (up to 60 % less) for damaging human health compared to Sc-1. Sc-2 and Sc-3 had up to 61 % less environmental impact than Sc-1. Sc-2 and Sc-3 reduced the total cumulative exergy demand by up to 44 % compared to Sc-1. In conclusion, producing chemicals and utilities within the biorefinery could significantly improve environmental sustainability, reduce waste, and diversify revenue streams.

Abstract:Enzymatic hydrolysis of organic waste is gaining relevance as a complementary technology to conventional biological treatments. Moreover, biorefineries are emerging as a sustainable scenario to integrate waste valorization and high-value bioproducts production. However, their application on municipal solid waste is still limited. This study systematically evaluates the techno-economic feasibility of the conversion of the organic fraction of municipal solid waste (OFMSW) into high-value bioproducts through enzymatic hydrolysis. Two key variables are examined: (a) the source of the enzymes: commercial or on-site produced using OFMSW, and (b) the treatment of the solid hydrolyzate fraction: solid-state fermentation (SSF) for the production of biopesticides or anaerobic digestion for the production of energy. As a result, four different biorefinery scenarios are generated and compared in terms of profitability. Results showed that the most profitable scenario was to produce enzymes on-site and valorize the solid fraction via SSF, with an internal rate of return of 13%. This scenario led to higher profit margins (74%) and a reduced payback time (6 years), in contrast with commercial enzymes that led to an unprofitable biorefinery. Also, the simultaneous production of higher-value bioproducts and energy reduced the economic dependence of OFMSW treatment on policy instruments while remaining energetically self-sufficient. The profitability of the biorefinery scenarios evaluated was heavily dependent on the enzyme price and the efficiency of the anaerobic digestion process, highlighting the importance of cost-efficient enzyme production alternatives and high-quality OFMSW. This paper contributes to understanding the potential role of enzymes in future OFMSW biorefineries and offers economical insights on different configurations.

Abstract:Background: A considerable challenge in the development of bioprocesses for producing chemicals and fuels has been the high cost of feedstocks relative to oil prices, making it difficult for these processes to compete with their conventional petrochemical counterparts. Hence, in the absence of high oil prices in the near future, there has been a shift in the industry to produce higher value compounds such as fragrances for cosmetics. Yet, there is still a need to address climate change and develop biotechnological approaches for producing large market, lower value chemicals and fuels. Results: In this work, we study ethylene glycol (EG), a novel feedstock that we believe has promise to address this challenge. We engineer Escherichia coli (E. coli) to consume EG and examine glycolate production as a case study for chemical production. Using a combination of modeling and experimental studies, we identify oxygen concentration as an important metabolic valve in the assimilation and use of EG as a substrate. Two oxygen-based strategies are thus developed and tested in fed-batch bioreactors. Ultimately, the best glycolate production strategy employed a target respiratory quotient leading to the highest observed fermentation performance. With this strategy, a glycolate titer of 10.4 g/L was reached after 112 h of production time in a fed-batch bioreactor. Correspondingly, a yield of 0.8 g/g from EG and productivity of 0.1 g/L h were measured during the production stage. Our modeling and experimental results clearly suggest that oxygen concentration is an important factor in the assimilation and use of EG as a substrate. Finally, our use of metabolic modeling also sheds light on the intracellular distribution through central metabolism, implicating flux to 2-phosphoglycerate as the primary route for EG assimilation. Conclusion: Overall, our work suggests that EG could provide a renewable starting material for commercial biosynthesis of fuels and chemicals that may achieve economic parity with petrochemical feedstocks while sequestering carbon dioxide.
