Abstract:Introduction & A 5-year old, otherwise healthy boy presented with an indolent, pustular plaque at his head. A preceding injury at this site, close contact to animals or preexisting skin disorders either in the patient or his family were denied. Initially, a scaly mass was found at the occipital capillitium. Besides cervical lymph node swelling further clinical examination was without any pathological fi ndings. Additional investigation using the wood lamp (UVA-light 365 nm) showed no fl uorescence. After scaling off the crusty matted plaque, an erythematous and oedematous, alopecic area interspered with follicular pustules and erosions became obvious ( ● ▶ Fig. 1 ). On manual pressure, pus was discharged, and the surrounding hair could be easily epilated. Bacteriological smear of the pustules revealed plentiful Staphylococcus aureus ( ● ▶ Fig. 2 ), sensitive to all tested antibiotics. After application of 70 % ethanol, hair and scales were asserved for microscopic analysis and culturing. Microscopy was performed according to standard procedures, i.e. after specimen incubation in 10 % potash solution for 30 min and with lactolphenol blue tease mounts. Microscopic analysis was negative for fungi. Consequently, we suspected impetigo follicularis and therefore refrained from further diagnostic steps including biopsy taking. Flucloxacillin sirup (staphylex ® , Glaxo Smith Kline) was given (50 mg / kg 3 times daily) over 10 days combined with locally applied fusidic acid cream (fucidine ® , Leo Pharma). Under this regimen, the purulent character improved and the erythema and oedema were regressive. However, clinical resolution was not achieved and hair loss progressed. To note, 14 days later the mycological cultures both of hair and scales, which were grown on agar plates with admixture of antibiotics and cycloheximide for preventing growth of bacteria and mould fungi, appeared positive. Agar plates showed powdery colonies ( ● ▶ Fig. 3 ) with smooth yellow colour from the reverse. On microscopy, septate hyphae, abundant macroconidia and fewer microconidia were detected ( ● ▶ Fig. 3 inlet ). Finally, taking together the clinical presentation and microbiological fi ndings, tinea capitis due to Microsporum gypseum was diagnosed – with Staphylococcus aureus superinfection. Therefore, systemic antimycotic therapy with terbinafi ne tablets (lamisil ® , Novartis) (62,5 mg daily) was initiated after excluding any contraindications and after detailed discussion and informed consent of the parents about the off -label use of terbinafi ne in childhood tinea. Systemic therapy was subsidized by hygienic advice (desinfection of the patient ́ s combs, use of separate towels and clothing as well as hot washing of the patient ́ s lining) and topical antimycotic application of ciclopiroxolamine cream (batrafen ® , Aventis). Familiy members were free of symptoms. Under this therapy, the infl ammation now completely resolved and fortunately fi rst hair regrowth could be documented after 4 weeks. No adverse events, especially no laboratory or gastrointestinal side effects, were noted. Local and systemic antimycotic therapy was continued over a total duration of 8 weeks. Mycological cultures were taken at week 4, showing no pathogenic fungi after 4 weeks culturing; hence, therapy was discontinued. At the fi nal follow-up controls 10 and 14 weeks after therapy start, mycological cultures remained negative, signifi cant hair growth was documented fortunately without scarring ( ● ▶ Fig. 4 ), and the patient presented in an unchanged, excellent general condition.

Abstract:Seaweed draws a lot of attention for its vital role in aquaculture as it contains beneficial biological compounds that undoubtedly might help in the development of this field. The current study sheds light on the potential efficiency of dietary supplements of Grateloupia acuminata and G. doryphore (Halymeniaceae) nanoparticles (GNS) at different levels with bionanocomposite cellulose acetate membranes (CA/bio-AgNps) on improved growth performance, digestive enzyme activity, immunity, antioxidative, resistance against infectious pathogens, and characterization of water quality treated with CA/bio-AgNps that is used in rearing Nile tilapia ( Oreochromis niloticus ). Four concentrations (0.1, 0.25, 0.5, and 1.0 ml/L) of GNS extract were tested as potential anti-bacterial and for the efficacy of being parasitic. Fish with an average weight (24.46 ± 0. 50 g) were apportioned into six experimental groups (T0, T1, T2, T3, T4, and T5) represented as 0.0%, 0.0%, 0.1%, 0.25%, 0.5%, and 1.0% GNS in diets with CA/bio-AgNps, respectively. Injection of fish with Aeromonas hydrophila was performed at the end of the trial. Chemical and bacteriological water indices significantly showed improvement after being treated with CA/bio-AgNps than the control group. Growth, carcass composition, digestive enzyme, and hematological and biochemical indices were significantly noticed positive ( p< 0.05), especially T4 and T5, than the control group. In parallel, a significant improvement was noticed in serum lysozyme, total immunoglobulin, complement C3, antioxidative enzyme, and the relative expression of hepatic and inflammatory genes with an increased level of GNS ( p< 0.05) are upregulated than the control group. Remarkably, GNS-supplemented diets and extracts provided positive efficacy against A. hydrophila with a decreased percentage of fish mortality, besides efficacy on antibacterial strains and Cichlidogyrus tilapiae , respectively. To sum up, the seaweed extract with CA/bio-AgNps resulted in better growth performance of fish, antipathogenic effect, and health status. Furthermore, CA/bio-AgNps were vital in improving water characteristics. They should be studied and applied more in the future.

Efficacy of dietary Nano-selenium on growth, immune response, antioxidant, transcriptomic profile and resistance of Nile tilapia, Oreochromis niloticus against Streptococcus iniae infection

Abstract:As recently applicable, there are few studies on the impact of using nano-selenium (nano-Se) on varied fish species. Where nothing reachable focused on its impact on tilapias so, the present analysis evaluated the efficacy of using nano-Se in tilapias on immune response, antioxidant defense compared by conventional Se form. 480 O. niloticus fingerlings were haphazardly grouped firstly into three groups with four replicates of each. The control one (CT) was fed on a basal diet. The second and third one supplemented with 0.7 mg/kg-1 Se and nano-Se respectively for ten weeks. At the start day of the ninth week, two replicates from each group were injected by Streptococcus iniae where, the remaining replicates stand without challenge. Enhancement of growth performance measurements were noted in nano-Se compared to Se or CT groups. Existed anemia in S. iniae tilapias became alleviated by using nano-Se that also, improves the alteration of leucogram induced by challenge. Elevation of aminotransferases, alkaline phosphatase, lactate dehydrogenase (ALT, AST, ALP and LDH) and creatinine in Se and CT challenged replicates that seemed nearly normal by using nano-Se. Usage of nano-Se showed more powerful antioxidant activities than Se. There were an expansion of immunoglobulin M, lysozymes, glutathione peroxidase, nitric oxide, superoxide dismutase and catalase (IgM, LYZ, GPx, NO, SOD, CAT) and their related gene expression in nano-Se with contrast in Se or CT challenged groups. Nile tilapias challenged by S. iniae disclosed substantial expansion in the percentage of mortality in CT challenged fish (93.33%), followed by the group supplemented with Se (73.33%), whereas the lowermost one at fish supplemented by nano-Se (26.66%). The mortalities have been stopped from the 5th, 12th and 14th days in, nano-Se, Se and CT respectively. It can be concluded that using of Se 0.7 mg/kg-1induce immunosuppressive, antioxidant, liver and kidneys negative impact on tilapias where the same dose from nano-Se was more potent immunomodulating and antioxidant. Also it is attend in counteracting the serious impact induced by S. iniae challenge.

Abstract:Functional trace elements and vitamins can boost immunity and anti-oxidative response in aquatic animals with effects on nutritional physiology. Nano-selenium (nano-Se) and vitamin C (VC) have been used as immunomodulators and antioxidants in fish feed. The present work was performed to determine the protective effects of diets supplemented with different combinations of nano-Se and VC on Nile tilapia (Oreochromis niloticus). Triplicate groups of 20 fish/tank (13.87 ± 0.10 g) were reared and fed with basal diet (control-T1) (without supplementation of nano-Se and VC) and three experimental diets as T2, T3, and T4 (100, 200, and 300 mg/kg VC respectively) with a pre-determined dose of nano-Se (1.0 mg/kg) for 90 days. Different immune indices, haemato-biochemical, and antioxidant activities were measured at the end of the first, second, and third months of feeding. The findings depicted that significantly (p < 0.05) higher growth was observed in T4. Red blood cells, white blood cells, and haemoglobin were found significantly (p < 0.05) higher in T4 for the third month. Serum biochemical-immunological indices (alkaline phosphatase, glucose, cholesterol, lysozyme, myeloperoxidase, total protein, albumin and globulin) followed the same trend. Furthermore, antioxidant assays such as catalase, superoxide dismutase, glutathione peroxidase, glutathione S-transferase, and malondialdehyde were significantly (p < 0.05) improved in T4 for the third month. Significantly (p < 0.05) least cumulative mortality against Aeromonas hydrophila was obtained in the fish-fed diets incorporated with nano-Se and VC. Therefore, dietary supplementation with nano-Se and VC is noteworthy for improving growth, serum biochemical status, immune response, antioxidant status, and disease resistance.

Impacts of dietary zinc oxide nanoparticles on the growth and immunity of Nile tilapia could be ameliorated using Nigella sativa oil

Abstract:Background: Zinc nanoparticles are documented to be harmful to fish because their accumulation in fish bring about many irreversible changes in their health. Nigella sativa and its oil have been endorsed in aquaculture to improve fish health. Methods: Two hundred seventy experimental fish (113 ± 5 g body weight) were divided into 6 groups G1-6; control fish fed a diet without any treatment (G1), 0.3% of NSO (G2), 0.5% of NSO (G3), ZnO NPs (40 mg/kg diet) (G4), 0.3% of NSO and ZnO NPs (40 mg/kg diet) (G5), 0.5% of NSO and ZnO NPs (40 mg/kg diet) (G6), the trial lasted for six weeks. Results: Growth performance was enhanced in fish received diets containing NSO, final weight (FW), weight gain (WG), daily weight gain (DWG), and relative growth rate (RGR) were significantly increased with lower food conversion ratios (FCR) compared to the control. The hepatic glutathione peroxidase (GPx), catalase (CAT), and metallothionein (MT) were increased in response to ZnO NPs stress and only 0.5% NSO supplementation could ameliorate such increment. The immune-related genes [interleukin1-beta (IL-1β), tumor necrosis factor-beta (TNF-β), transforming growth factor-beta 2 (TGF-β2) and C-type lysozyme] as well as growth-related gene [insulin-like growth factor 1 (IGF1)] in liver showed an upregulation in fish fed with NSO diets. Administration of ZnO NPs lowered the resistance of Oreochromis niloticus against bacterial infection with Aeromonas hydrophila and NSO could enhance the immunity in the highest tested concentration (0.5%) (G6). Conclusions: The obtained results implied that NSO could enhance the oxidative and immune status of O. niloticus which could compensate ZnO NPs stress as well as experimental infection of a virulent strain of A. hydrophila. Our results revealed that NSO might increase fish growth and immunity only at a high dose (0.5%).

Abstract:Recently, nanotechnology has become an important research field involved in the improvement of animals' productivity, including aquaculture. In this field, silver nanoparticles (AgNPs) have gained interest as antibacterial, antiviral, and antifungal agents. On the other hand, their extensive use in other fields increased natural water pollution causing hazardous effects on aquatic organisms. Quercetin is a natural polyphenolic compound of many plants and vegetables, and it acts as a potent antioxidant and therapeutic agent in biological systems. The current study investigated the potential mitigative effect of quercetin nanoparticles (QNPs) against AgNPs-induced toxicity in Nile tilapia via investigating liver function markers, hepatic antioxidant status, apoptosis, and bioaccumulation of silver residues in hepatic tissue in addition to the whole-body chemical composition, hormonal assay, intestinal enzymes activity, and gut microbiota. Fish were grouped into: control fish, fish exposed to 1.98 mg L−1 AgNPs, fish that received 400 mg L−1 QNPs, and fish that received QNPs and AgNPs at the same concentrations. All groups were exposed for 60 days. The moisture and ash contents of the AgNP group were significantly higher than those of the other groups. In contrast, the crude lipid and protein decreased in the whole body. AgNPs significantly increased serum levels of ALT, AST, total cholesterol, and triglycerides and decreased glycogen and growth hormone (*** p < 0.001). The liver and intestinal enzymes' activities were significantly inhibited (*** p < 0.001), while the oxidative damage liver enzymes, intestinal bacterial and Aeromonas counts, and Ag residues in the liver were significantly increased (*** p < 0.001, and * p < 0.05). AgNPs also significantly upregulated the expression of hepatic Hsp70, caspase3, and p53 genes (* p < 0.05). These findings indicate the oxidative and hepatotoxic effects of AgNPs. QNPs enhanced and restored physiological parameters and health status under normal conditions and after exposure to AgNPs.

Abstract:The present study examined the impacts of varying amounts of selenium nanoparticles (Se-NPs) as a natural antioxidant and metabolic regulator on growth performance, antioxidant capacity, digestive enzymes and immune resistance of Oreochromis niloticus challenged to Aspergillus flavus infection. For 60 days, fish were fed the experimental diet of 30% crude protein up to apparent satiation three times a day. Se-NPs were added to the control diet at different levels of 0.0 (control), 0.5 (T1), 1.0 (T2) and 1.5 (T3) mg/kg diet to fed monosex Nile tilapia fingerlings with average initial weight (4.5 ± 0.5 g). Fish were randomly distributed in 12 tanks 100 L at a density of 20 fish per tank after 2-week acclimation to represent four treatments in triplicates. The results indicated that the growth indices, feed efficiency and survival rate were significantly enhanced ( P < 0.05) by incorporating Se-NPs up to 1 mg/kg diet. Furthermore, the haemato-biochemical parameters, digestive enzymes activity and antioxidant capacity of the fish were significantly improved ( P < 0.05) at T2. When the fish were challenged with Aspergillus flavus , a decreasing mortality rate was observed, which clearly shows that selenium nanoparticles boosted the fish's immune response at T2. The intestinal morphology as villus length, villus width, muscular fibres layer thickness and absorption intestinal zone after challenged with A. flavus infection were considerably enhanced ( P < 0.05) by incorporating all Se-NP levels. Histopathological score significantly improved ( P < 0.05) for the hepatopancreatic, intestinal, gills and muscle tissues at T2. The present study concludes that selenium nanoparticles up to 1.0 mg/kg diet can be efficiently used in tilapia feed to help boost fish production, immune system response and histopathological parameters.

Abstract:This account bf our examination of different molecular forms of lactic dehydrogenase, which we present as a preliminary report, does not serve the purpose of finding new diagnostic possibilities in dermatoses, but it has the aim of answering open and frequently problematic questions on the enzyme metabolism of the human skin. Substantially four problems are concerned and, because of the different character of each, they can be treated only independently. (1) Are the enzymes and enzyme systems found in the human epidermis produced by the cells of the epidermis themselves or do they diffuse from the blood vessels of the dermis into these cells? Weber and his co-workers could demonstrate'-8 by extensive bioenzymatical investigation that different metabolites and enzymes of the glycolysis-among them also the lactic dehydrogenase-are found in high concentration in the cells of the epidermis, and that these diminish continuously in the direction of the peripheral bloodstream (FIGURE 1). From this decrease of concentration Weber and his associates inferred that the enrichment of enzymes in the epidermis is caused by the production of enzymes in the cells of the epidermis, and not by diffusion from the bloodstream, which would tend to accentuate the concentration decrease. Although these results represent convincing arguments, definite proof for the correctness of this thesis was still lacking. This proof could be obtained by an electrophoretic determination of lactic dehydrogenase in a homogenate of the epidermis, of the dermis, and of erythrocytes which, according to Hess,14 contain the main portion of serum lactic dehydrogenase. As to method, we have followed Wieland et a1.12 As shown in FIGURE 2, there are such significant differences between the distribution patterns of lactic dehydrogenase in the epidermis and in the erythrocytes that from this fact alone the autochthonous production of lactic dehydrogenase in the cells of epidermis can be assumed with certainty. (2) Is there any difference between the molecular forms of lactic dehydrogenase in the epidermis and in the dermis? I t would be necessary first to find if the concentration of lactic dehydrogenase in the epidermis is much higher than in the dermis. Comparing the distribution patterns of this enzyme in both tissues (FIGURE 2), a distinct shift to the right results for the epidermis, that is, column S is accentuated, whereas the distribution pattern of enzyme in the dermis shows a shift to the left, that is, a more distinct accentuation to column 1. According to Pfleiderer and WachsmuthI3 the strictly anaerobic metabolizing tissues show a shift to the right, that is, the maxima are directed to column 5 , whereas the tissues that have a high oxygen consumption are characterized by a shift to the left: that is, by maxima directed to column 1. The conclusion i s that, in the case of cells of the epidermis, chiefly anaerobic metabolizing tissue is to be assumed: in the case of the cells of the dermis, a more aerobic metabolizing tissue is indicated.
Physiological and Neurobehavioral Disturbances Induced by Al2O3 Nanoparticle Intoxication in Nile Tilapia Fish: Benefits of Dietary Chamomile Essential Oil

Abstract:Despite the usage of nanoparticles (NPs) is rapidly increasing, several experts have noted the risk of their release into ecosystems and their potential negative impacts on biological systems. However, the available studies on the neurobehavioral impacts of aluminum oxide nanoparticles (Al2O3NPs) on aquatic organisms are little. Hence, this study targeted to ascertain the harmful effects of Al2O3NPs on behavioral characteristics and genotoxic and oxidative damages in Nile tilapia fish. In addition, the beneficial role of chamomile essential oil (CEO) supplementation in reducing these effects was also investigated. In the current study, fish were distributed into 4 equal groups ( n = 60 fish per group). The control group was fed a plain diet only, the CEO group received a basic diet complemented with CEO at a level of 2 mg/kg diet, the ALNP group received a basic diet and was exposed to an approximate concentration of 1/10th LC50 of ALNPs nearly 5.08 mg/L, and the combination group (ALNPs/CEO group) received a basal diet coadministered with ALNPs and CEO at the aforementioned percentages. The findings revealed that O. niloticus exhibit neurobehavioral changes along with changes in the level of GABA, monoamines in the brain tissue, and serum amino acid neurotransmitters, besides a reduction of AChE and Na+/K+-ATPase activities. In addition to brain tissue oxidative damage with upregulation of proinflammatory and stress genes, such as HSP70 and caspase-3, supplementation of CEO significantly reduced the negative impacts of ALNPs. These results showed that CEO has neuroprotective, antioxidant, genoprotective, anti-inflammatory, and antiapoptotic properties in fish that have been exposed to ALNPs. Therefore, we advise its usage as a valuable addition to fish diet.

Impact of Streptococcus agalactiae Challenge on Immune Response, Antioxidant Status and Hepatorenal Indices of Nile Tilapia: The Palliative Role of Chitosan White Poplar Nanocapsule

Abstract:A new insight into the synthesis of the herbal plant (White poplar, Poplus alba) leave extract using chitosan nanocapsule was studied. The in vitro antibacterial activity of chitosan white poplar nanocapsule (CWPNC) against Streptococcus agalactiae (S. agalactiae) was determined. About 120 fish were categorized for 7 days into four groups. The first and second (CWPNC) groups were treated with 0 mg/L and 3 mg/L CWPNC in the water, respectively, without being challenged; the first group was a control. The third (S. agalactiae) and fourth (CWPNC + S. agalactiae) groups were treated with 0 and 3 mg/L CWPNC, respectively, and challenged with S. agalactiae (0.5 × 107 CFU/mL). The obtained results revealed that CWPNC had an in vitro antibacterial activity against S. agalactiae. Moreover, S. agalactiae infection caused a significant elevation (p < 0.05) in the lipid peroxidation (malondialdehyde) and hepatorenal biomarkers, as well as the lowest significant (p < 0.05) survival rate (33.33%). Moreover, a significant depletion (p < 0.05) in the level of antioxidants (catalase and superoxide dismutase) and the immune indicators (immunoglobulin, lysozyme activity, and complement 3) were the consequences of S. agalactiae infection. Treatment of the infected fish with 3 mg/L CWPNC alleviated these bad circumstances.

Polystyrene nanoplastic and engine oil synergistically intensify toxicity in Nile tilapia, Oreochromis niloticus

Abstract:Polystyrene nanoplastic (PS-NPs) and Engine oil (EO) pose multiple ecotoxic effects with increasing threat to fish ecosystems. The current study investigated the toxicity of 15 days exposure to PS-NPs and / or EO to explore their combined synergistic effects on Nile tilapia, Oreochromis niloticus ( O. niloticus ). Hematobiochemical parameters, proinflammatory cytokines, and oxidative stress biomarkers as well as histological alterations were evaluated. The experimental design contained 120 acclimated Nile tilapia distributed into four groups, control, PS-NPs (5 mg/L), EO (1%) and their combination (PS-NPs + EO). After 15-days of exposure, blood and tissue samples were collected from all fish experimental groups. Results indicated that Nile tilapia exposed to PS-NPs and / or EO revealed a significant decrease in almost all the measured hematological parameters in comparison to the control, whereas WBCs and lymphocyte counts were significantly increased in the combined group only. Results clarified that the combined PS-NPs + EO group showed the maximum decrease in RBCs, Hb, MCH and MCHC, and showed the maximum significant rise in interleukin-1β (IL-1β), and interleukin-6 (IL-6) in comparison to all other exposed groups. Meanwhile, total antioxidant capacity (TAC) showed a significant ( p < 0.05) decline only in the combination group, whereas reduced glutathione (GSH) showed a significant decline in all exposed groups in comparison to the control. Both malondialdehyde (MDA) and aspartate aminotransferase (AST) showed a significant elevation only in the combination group. Uric acid showed the maximum elevation in the combination group than all other groups, whereas creatinine showed significant elevation in the EO and combination group when compared to the control. Furthermore, the present experiment proved that exposure to these toxicants either individually or in combination is accompanied by pronounced histomorpholgical damage characterized by severe necrosis and hemorrhage of the vital organs of Nile tilapia, additionally extensively inflammatory conditions with leucocytes infiltration. We concluded that combination exposure to both PS-NPs and EO caused severe anemia, extreme inflammatory response, oxidative stress, and lipid peroxidation effects, thus they can synergize with each other to intensify toxicity in fish.

Abstract:Antibiotics play a vital role in aquaculture where they are commonly used to treat bacterial diseases. However, the impact of antibiotic treatment on the gut microbiome and the development of antimicrobial resistance in Nile tilapia (Oreochromis niloticus) over time remains to be fully understood. In this study, fish were fed a single treatment of oxytetracycline (100 mg/kg/day) for eight days, followed by a 14-day withdrawal period. Changes in the distal gut microbiome were measured using 16S rRNA sequencing. In addition, the abundance of antimicrobial resistance genes was quantified using real-time qPCR methods. Overall, the gut microbiome community diversity and structure of Nile tilapia was resilient to oxytetracycline treatment. However, antibiotic treatment was associated with an enrichment in Plesiomonas, accompanied by a decline in other bacteria taxa. Oxytetracycline treatment increased the proportion of tetA in the distal gut of fish and tank biofilms of the treated group. Furthermore, the abundance of tetA along with other tetracycline resistance genes was strongly correlated with a number of microbiome members, including Plesiomonas. The findings from this study demonstrate that antibiotic treatment can exert selective pressures on the gut microbiome of fish in favour of resistant populations, which may have long-term impacts on fish health.

Abstract:This study aimed to evaluate the effect of adding liquid extract of algae (Hypnea musciformis, Grateloupia acuminata, and Sargassum muticum) (HGS) and Magnesium oxide nanoparticles (MgO NPs) using this extract to rear water of Oreochromis niloticus, on improving culture water indices, growth performance, digestive enzyme, hemato-biochemical characters, immune, antioxidative responses, and resistance after challenged by Aeromonas hydrophila with specific refer to the potential role of the mixture in vitro as resistance against three strains bacteria (Aeromonas sobria, Pseudomonas fluorescens, P. aeruginosa) and one parasite (Cichlidogyrus tilapia). The first group represented control, HGS0, whereas the other group, HGS5, HGS10, and HGS15 mL-1 of liquid extract, as well as all groups with 7.5 μg mL-1 MgO-NPs added to culture water of O. niloticus, for 60 days. Data showed that increasing levels at HGS 10 and HGS15 mL-1 in to-culture water significantly enhanced growth-stimulating digestive enzyme activity and a significantly improved survival rate of O. niloticus after being challenged with A. hydrophila than in the control group. The total viability, coliform, fecal coliform count, and heavy metal in muscle partially decreased at HGS 10 and HGS15 mL-1 than in the control group. Correspondingly, the highest positive effect on hemato-biochemical indices was noticed at levels HGS 10 and HGS15 mL-1. Fish noticed an improvement in immune and antioxidant indices compared to control groups partially at HGS 10 and HGS15 mL-1. Interestingly, fish cultured in rearing water with the mixture provided downregulated the related inflammatory genes (HSP70, TNF, IL-1β, and IL-8) partially at HGS15 mL-1. In vitro, the mixture showed positive efficiency as an antibacterial and partially antiparasitic at HGS 10 and HGS15 mL-1. This study proposes utilizing a mixture of (HGS) and (MgO-NPs) with optimum levels of 10-15 mL-1 in cultured water to improve water indices, growth, health status, and increased resistance of O. niloticus against bacterial and parasitic infection.

Abstract:In recent times, nutraceuticals have been used extensively to identify promising feed additives for the improvement of the aquaculture industry through the enhancement of growth and survival rates, potentiation of the immune responses, and fortification of the resistance against infectious bacterial diseases. In this study, Nile tilapia (Oreochromis niloticus) were fed with diets supplemented with quinoa seeds (QU) or prickly pear fruit peel (PP) at the dose levels of 10% or 20% of the diet. After 45 days of the feeding trial, the fish were exposed to Aeromonas sobria (A. sobria) challenge. The pre-challenge indices indicated that both supplements mediated a significant improvement in most of the estimated parameters, including survival rate, antioxidant status, hematological and immunological indices, and hepatoprotective potential. These effects were recorded in the groups fed with high doses of the supplements (20%). The least changes were observed in the QU10-supplemented fish. In the spleen tissue, the TGF-β gene was upregulated in the PP10-, PP20- and QU20-supplemented groups, while the expression of the IFN-γ gene remained unaffected in all the supplemented groups, except for the PP20-supplemented group, which showed an upregulation. After the challenge with A. sobria, the relative survival percentage was improved by the supplementation of PP and QU, particularly in the PP20-supplemented group, possibly via the promotion of immunological responses, hepatoprotective potency, and modulation of the studied genes. Moreover, the morphological structure of the tissues showed marked recovery. The findings suggest that Nile tilapia fed with different levels of PP peel and QU seeds, particularly at the level of 20%, enhanced the immune response in fish and improved their resistance against A. sobria infection.

Abstract:Background: The inappropriate use of pesticides including fungicides creates severe biological hazards that can endanger fish health and impede sustainable aquaculture. Objective: This study investigated the negative impacts of metiram (MET), a fungicide on the health status of Nile tilapia (Oreochromis niloticus) for a 96-hour duration as an acute exposure in a static renewal system. Methods: Three hundred fish (average body weight: 37.50 ± 0.22 g) were assigned into six groups (50 fish/group) with five replicates (10 fish/replicate). Fish were exposed to various six concentrations (0, 1.5, 3, 4.5, 6, and 7.5 mg/L) of MET as a water exposure to for 96-hour without water exchange. The fish's behavior, clinical signs, and mortalities were documented every day of the exposure period. Additionally, MET's impact on blood profile, stress biomarkers, hepato-renal functions, immune-antioxidant status, and brain biomarker were closely monitored. Results: The lethal concentration (LC50) of MET estimated using Finney's probit technique was 3.77 mg/L. The fish's behavior was severely impacted by acute MET exposure, as clear by an increase in surfacing, loss of equilibrium, unusual swimming, laterality, abnormal movement, and a decline in aggressive behaviors. The survivability and hematological indices (white and red blood cell count, differential white blood cell count, hematocrit value, and hemoglobin) were significantly reduced in a concentration-dependent manner following MET exposure. Acute exposure to MET (1.5-7.5 mg/L) incrementally increased stress biomarkers (nor-epinephrine, cortisol, and glucose), lipid peroxides (malondialdehyde), and brain oxidative DNA damage biomarker (8-hydroxy-2-deoxyguanosine). A hepato-renal dysfunction by MET exposure (4.5-7.5 mg/L) was evidenced by the significant increase in the alanine and aspartate aminotransferases and creatinine values. Moreover, a substantial decline in the immune parameters (lysozyme, complement 3, serum bactericidal activity, and antiprotease activity) and antioxidant variables (total antioxidant capacity, superoxide dismutase, and glutathione peroxidase) resulted from acute MET exposure. Conclusion: According to these findings, the 96-hour LC50 of MET in Nile tilapia was 3.77 mg/L. MET exposure triggered toxicity in Nile tilapia, as seen by alterations in fish neuro-behaviors, immune-antioxidant status, hepato-renal functioning, and signifying physiological disturbances. This study emphasizes the potential ecological dangers provoked by MET as an environmental contaminant to aquatic systems. However, the long-term MET exposure is still needed to be investigated.

Abstract:Copper oxide nanoparticles (CuONPs) have attracted much interest due to their catalytic and anti-bacterial activity. The present study exhibits that Moringa oleifera synthesized CuONPs (MS-CuONPs) might provide a promising anti-bacterial substitute for conventional antibiotics to treat Staphylococcus aureus infection in African catfish. MS-CuONPs showed an in vitro anti-bacterial activity (21.33 ± 0.80 mm inhibition zone) against S. aureus . One hundred-twenty fish (average weight: 120 ± 5.36 g) were haphazardly allocated into four groups in triplicates for 10 days (control, MS-CuONPs, S. aureus , and MS-CuONPs + S. aureus ), where a concentration of 0.1 mg/L of MS-CuONPs was used. The S. aureus infection induced a marked reduction in immunological (lysozyme, complement 3, and nitric oxide), protein profile (total protein, total globulins, and albumin), and hepatic antioxidant (total antioxidant capacity, superoxide dismutase, and glutathione peroxidase) indices as well as a higher mortality rate (46.67%). Moreover, an apparent elevation in the values of tumor necrosis factor-alpha (TNF-α), hepatic malondialdehyde (MDA), stress biomarkers (cortisol and glucose), and hepato-renal indicators (aspartate aminotransferase, alkaline phosphatase, alanine aminotransferase, creatinine, and urea) were the consequences of S. aureus infection. Interestingly, treatment of S. aureus- infected fish with MS-CuONPs improved these parameters. According to these outcomes, MS-CuONPs as a water additive can protect fish against the negative impacts of S. aureus and can be used a promise anti-bacterial agent.

Abstract:Fucoidans are marine algal sulfated glycans that are widely used as dietary additives in aquaculture. These glycans are recognized as beneficial supplements for their antimicrobial, anti-inflammatory, anticancer, and antiviral properties. Potassium permanganate is another commonly used chemical that is used in aquaculture to treat infections in fish. Despite their widespread use, there are few data available regarding the potential sublethal toxicity associated with fucoidan and potassium permanganate treatments of fish. In this study, we investigated the effect of each compound on the growth, intestinal health, and antioxidant status of Nile tilapia (Oreochromis niloticus). Both compounds affected the growth of experimental fish compared with untreated fish. However, while growth parameters were positively associated with the dose of fucoidan administered, growth was negatively associated with the dose of potassium permanganate in Nile tilapia. Fucoidan treatment was observed to improve the intestinal health of fish based upon increases in intestinal villous area, intestinal villous length and width, and the intraepithelial lymphocyte number and decreases in the total intestinal bacterial count compared with untreated fish. Conversely, potassium permanganate induced intestinal epithelium proliferation and villous branching, a histopathological response typically observed with chemical irritants. Both fucoidan and potassium permanganate decreased levels of oxidative and nitrosative stress markers and enhanced the antioxidant status in multiple organs. Taken together, fucoidan dietary application improved the growth, intestinal health, and antioxidant status in Nile tilapia, supporting the use of this compound as a promising feed additive for aquaculture production. Conversely, potassium permanganate baths have negative effects on fish growth at higher doses and appeared to act as a gastrointestinal irritant in tilapia. This study improves knowledge regarding the biochemical and histological responses in Nile tilapia to two widely used aquaculture-related treatments.

Abstract:Intensive aquaculture often results in immunosuppression in fish, which may cause a series of diseases. In this study, to investigate the immunosuppressive mechanisms in fish, tilapia were intrapleural injected cyclophosphamide (CTX) at the doses of 10, 25, 50, 75 and 100 mg•kg−1 to induce immunosuppression. We determined the viability of immune cells, the content of lysozyme (LZM) and immunoglobulin M (IgM), the levels of nitric oxide (NO) and antioxidant parameters. Meanwhile, the mRNA levels of complement C3 (c3), igm and the genes associated with the TLR-NF-κB signaling pathway in the head kidney (HK) and spleen were also determined. The results showed that CTX had a significant cytotoxic effect on peripheral blood leukocytes, HK macrophages and spleen cells in a dose-dependent manner. The protein and mRNA levels of C3 and IgM were down-regulated with the increase of CTX concentrations in serum, HK and/or spleen. The NO and LZM contents decreased significantly in HK and spleen after CTX treatments with 75 and 100 mg•kg−1. CTX treatments with 50, 75 and/or 100 mg•kg−1 markedly decreased the antioxidant ability and enhanced lipid peroxidation in HK and spleen. Furthermore, qPCR data showed that CTX treatments with 50-100 mg•kg−1 clearly down-regulated the mRNA levels of tlr2, myd88, irak1, traf6, nfκb1, nfκb2, il-6, il-10 and tnf-α in the HK and/or spleen. Overall results suggested that CTX treatment had a cytotoxic effect on immune cells, induced lipid peroxidation, decreased the antioxidant capacity and inhibited immune function. The immunosuppressive mechanisms of CTX may be associated with the TLR-NF-κB signaling pathway.

Abstract:This investigation attempts to evaluate the effect of diet additives via aqueous or ethanolic herbal extracts from Azadirachta indica leaves on Nile tilapia, Oreochromis niloticus. Five dietary categories were assigned to the fish: the first category (N1, with no extract) was kept under control conditions; two categories contained aqueous extract (N2 (1.0 g/kg) and N3 (2.0 g/kg); and two categories contained ethanolic extract, N4 (1.0 g/kg) and N5 (2.0 g/kg), with each group being fed for 60 days. After the feeding trial, Aeromonas hydrophila was injected intraperitoneally into fish for 14 days; fish mortality was recorded during this period. The results showed that the fish-fed dietary A. indica significantly improved growth performance and intestinal health (digestive enzymes and intestinal morphology), especially in the N4 and N5 categories. However, N4 and N5 categories demonstrated a significant decrease in AST and ALT activities and an increase in total protein, serum albumin, globulin, growth hormone (GH), leptin hormone (LEP), hemoglobin, white blood cells, and hematocrit (P < 0.05) in comparison with the control category (N1). Compared to the control category, the N4 and N5 categories have revealed a significant reduction in MDA activity and improvements in immunological activities (lysozyme, complement C3, and nitric oxide) and antioxidant enzymes (CAT, SOD, and GPX). Moreover, in tilapia-fed A. indica, the expression of IL-8, IL-1β, and Nf-κb genes was downregulated partially in the N4 and N5 categories than the control category. In contrast, the lysozyme, C3, GPX, and CAT genes were upregulated partially at N4 and N5 compared to the control category. Following the bacterial challenge, fish in the N4 and N5 categories also displayed the lowest fish mortality compared to the control category. The ethanolic extract displayed a more potent resistance against the parasite Cichlidogyrus tilapia in vitro than the aqueous and control categories, partially at 2 g/L. According to these findings, an ethanolic neem extract (2.0 g/kg feed) activates the immune system and antioxidant response in Nile tilapia fingerlings, improving growth and fish resistance to parasitic and bacterial infections.
