Abstract:The use of efficient green cleaning agents, such as biosurfactants, is important in oil sludge treatment. Enhanced oil recovery from oily sludge by different rhamnolipids was comparatively evaluated. Using crude glycerol, the wild-type strain Pseudomonas aeruginosa SG and the recombinant strains P. aeruginosa PrhlAB and P. stutzeri Rhl produced 1.98 g L-1, 2.87 g L-1 and 0.87 g L-1 of rhamnolipids, respectively. The three bacterial strains produced different rhamnolipid mixtures under the same conditions. The proportions of mono-rhamnolipids in the three rhamnolipid products were 55.92%, 94.92% and 100%, respectively. These rhamnolipid products also possessed different bioactivities. Emulsifying activity became higher as the proportion of mono-rhamnolipids increased. The three rhamnolipid products were stable at temperatures lower than 121 °C, pH values from 5-11 and NaCl concentrations from 0-15%. All three rhamnolipid products could recover oil from oily sludge, but oil recovery efficiency was positively related to the proportion of mono-rhamnolipids. Mono-rhamnolipids produced by the recombinant strain Rhl exhibited the best oil recovery efficiency (53.81%). The results reveal that mono-rhamnolipids are the most promising for oil recovery from oily sludge.
High Di-rhamnolipid Production Using Pseudomonas Aeruginosa KT1115, Separation of Mono/Di-rhamnolipids, and Evaluation of Their Properties

Abstract:Rhamnolipids (RLs) are important bioproducts that are regarded as promising biosurfactant for applications in oil exploitation, cosmetics, and food industry. In this study, the newly isolated Pseudomonas aeruginosa KT1115 showed high production of di-RLs. The highest yield of RLs by P. aeruginosa KT1115, reaching 44.39 g/L after 8 days of fermentation in a 5 L bioreactor, was obtained from rapeseed oil-nitrate medium after process optimization. Furthermore, we established a new separation process that achieved up to 91.82% RLs recovery with a purity of 89% and further obtained mono/di-rhamnolipids. Finally, ESI-MS analysis showed that the RLs produced by strain KT1115 have a high proportion of di-RLs (mono-RLs: di-RLs = 11.47: 88.53), which have a lower critical micelle-forming concentration (8 mN/m) and better emulsification ability with kerosene (52.1% EI24) than mono-RLs (167 mN/m and 41.4% EI24, respectively). These results demonstrated that P. aeruginosa KT1115 is a potential industrial producer of di-RLs, which have improved applicability and offer significant commercial benefits.

Abstract:Differences in the rhamnolipid structures must result in its different activities, thus affecting its application effect. The rhlC gene in Pseudomonas aeruginosa SG was knocked out to construct strain P. aeruginosa SGΔrhlC. Rhamnolipid production was enhanced by 23.3% through knocking out rhlC gene. P. aeruginosa SGΔrhlC produced 14.22 g/L of rhamnolipid using glycerol and nitrate. Five kinds of mono-rhamnolipid but no di-rhamnolipid were produced by strain SGΔrhlC. The main rhamnolipid homologues were Rha-C10-C10, Rha-C10-C12:1 and Rha-C10-C12. Mono-rhamnolipid exhibited better antimicrobial activity to Escherichia coli, Staphylococcus aureus, Aspergillus niger and Penicillium chrysogenum. Rhamnolipid produced from strain SGΔrhlC showed greater emulsifying activity to crude oil with EI24 of 84.73%. Rhamnolipid produced from strain SGΔrhlC efficiently washed oily sludge at 35 °C. High-producing strain P. aeruginosa SGΔrhlC and its produced mono-rhamnolipid are more promising in agriculture and petroleum industry. This study is a step forward to the tailor-made biosynthesis and application of rhamnolipid.

Abstract:Rhamnolipids (RLs) are widely used biosurfactants produced mainly by Pseudomonas aeruginosa and Burkholderia spp. in the form of mixtures of diverse congeners. The global transcriptional regulator gene irrE from radiation-tolerant extremophiles has been widely used as a stress-resistant element to construct robust producer strains and improve their production performance. A PrhlA-irrE cassette was constructed to express irrE genes in the Pseudomonas aeruginosa YM4 of the rhamnolipids producer strain. We found that the expression of irrE of Deinococcus radiodurans in the YM4 strain not only enhanced rhamnolipid production and the strain's tolerance to environmental stresses, but also changed the composition of the rhamnolipid products. The synthesized rhamnolipids reached a maximum titer of 26 g/L, about 17.9% higher than the original, at 48 h. The rhamnolipid production of the recombinant strain was determined to be mono-rhamnolipids congener Rha–C10–C12, accounting for 94.1% of total products. The critical micelle concentration (CMC) value of the Rha–C10–C12 products was 62.5 mg/L and the air-water surface tension decreased to 25.5 mN/m. The Rha–C10–C12 products showed better emulsifying activity on diesel oil than the original products. This is the first report on the efficient production of the rare mono-rhamnolipids congener Rha–C10–C12 and the first report that the global regulator irrE can change the components of rhamnolipid products in Pseudomonas aeruginosa.

Abstract:Biosurfactant rhamnolipids (RLs) have gained global interests owing to their fully green properties, potentially wide applications in diverse fields, as well as high stabilities under various harsh conditions. Nevertheless, we doubted the reputed stability of RLs in considering their natural structure of carbohydrate heads and lipid tails. This study, for the first time, systematically investigated the stability of RLs at varying temperatures and pH. As found, the concentration of RLs in an aqueous solution was significantly reduced when the pH was over 11 at room temperature, and this was much more severe with the increase in temperature and preservation time. According to the high-performance liquid chromatography-mass spectrometry (HPLC-MS) analysis, degradation yielded other RL congeners, 3-hydroxy fatty acids, rhamnose, methyl furfural, and organic acids. The newly generated RL congeners and fatty acids still possessed equivalent surface activities in reducing the surface tension of the aqueous solution, well explaining the previously claimed high stability of RLs. The finding will be greatly valued for commercially developing the industrial applications of RLs and other biosurfactants.
Mechanism Study on the Severe Foaming of Rhamnolipid in Fermentation

Abstract:Although the biosurfactant rhamnolipid has been previously characterized as having low foam ability, its fermentation is largely impeded by severe foaming. Hence, the investigation of this paradox is critically important for improving the mass production of rhamnolipid. Unexpectedly, the hydrophobic cell, instead of rhamnolipid, has been claimed to explain such severe foaming in rhamnolipid fermentation. This study tried to systematically investigate the severe foaming in fermentation, aiming to propose an effective strategy for foam control. The overflowing foam sustained a super high stability in terms of half-time for over 30 min. The major product of rhamnolipid largely contributed to the severe foaming in the fermentation process whereas other products like cells elicited much more limited effects. Furthermore, the foam stability of the fermentation broth increased with rhamnolipid concentration and noticeably increased with agitation speed. In the classic Bikerman foam test system without stirring, rhamnolipid showed foam stability as low as Tween 20 which is well known for its poor foam stability. However, in a stirring Bikerman system, rhamnolipid exhibited a foam stability almost as high as sodium dodecyl sulfate (SDS) at 10 g/L and even surpassed SDS at a higher concentration of 20 g/L. Hence, the extraordinarily increased foam stability of rhamnolipid with both agitation and concentration could explain the severe foaming at its late-stage fermentation when rhamnolipid-rich solution is mechanically agitated.

Abstract:To construct a derivative of the avirulent Pseudomonas aeruginosa ATCC 9027 that produces high levels of di-rhamnolipid, that has better physico-chemical characteristics for biotechnological applications than mono-rhamnolipid, which is the sole type produced by ATCC 9027. We used plasmids expressing the rhlC gene, which encodes for rhamnosyl transferase II that transforms mono- to di-rhamnolipids under different promoters and in combination with the gene coding for the RhlR quorum sensing regulator, or the mono-rhamnolipid biosynthetic rhlAB operon. The plasmids tested carrying the rhlC gene under the lac promoter were plasmid p rhlC and p rhlRC , while p rhlAB-R–C expressed this gene from the rhlA promoter, forming part of the artificially constructed rhlAB-R–C operon. We measured rhamnolipds concentrations using the orcinol method and determined the proportion of mono-rhamnolipids and di-rhamnolipids by UPLC/MS/MS. We found that the expression of rhlC in P. aeruginosa ATCC 9027 caused the production of di-rhamnolipids and that the derivative carrying plasmid p rhlAB-R–C gives the best results considering total rhamnolipids and a higher proportion of di-rhamnolipids. A P. aeruginosa ATCC 9027 derivative with increased di-rhamnolipids production was developed by expressing plasmid p rhlAB-R–C , that produces similar rhamnolipids levels as PAO1 type-strain and presented a higher proportion of di-rhamnolipids than this type-strain.

Abstract:Two representative rhamnolipidic fractions, RL-F1 and RL-F2, produced by the P. aeruginosa mutant strain MIG-N146, were separated and chemically characterized by TLC, HPLC-MS, and FTIR. The RL-F1 fraction is predominantly mono-rhamnolipid homologues with a high content of one or two fatty acid moieties. The RL-F2 fraction is mainly composed of di-rhamnosyl moieties with two hydrophobic tails. Micellization behavior was investigated to assess the physicochemical properties of the surfactants, RL-F1, RL-F2, and crude rhamnolipidic extracts. The variations in morphology of micelle formation and growth were examined by dynamic light scattering measurements as a function of surfactant concentration. Critical micelle concentration (CMC), average minimal surface tension (γCMC), saturated surface excess (Γm), mean surface area per molecule (S), and adsorption efficiency (pC20) were determined from the surface tension profiles and compared for the three surfactant systems. It was found that micelle growth was significantly enhanced by increasing rhamnolipid bulk concentration, which was most probably accompanied with an aggregate shape transition. Well-separated multi- or bi-modal distributions of particle size were observed in RL-F2 and the crude extracts solutions. The results of this study demonstrate that molecular architecture of different surfactant compositions profoundly influences the performance of rhamnolipidic surfactants.
Infertility and abortion among first-lactation dairy cows seropositive or seronegative for Leptospira interrogans serovar hardjo.

Abstract:OBJECTIVE To estimate the extent to which exposure to Leptospira hardjo before or at the time of first parturition was associated with infertility and abortion during the first lactation among dairy cows that had not been vaccinated for > or = 12 months. ANIMALS 207 first-lactation cows from a herd of 2,000 lactating cows. PROCEDURE Cows were tested for antibodies to L hardjo within 40 days after calving. Time from calving to first breeding, time from calving to conception, number of breedings per conception, and risk of abortion were compared between cows seropositive for L hardjo and cows that were seronegative. RESULTS For the 9 (4.3%) cows that were seropositive for L hardjo, median time from calving to conception (132.6 days) was significantly longer than time for seronegative cows (95.4 days). Cows that were seropositive were twice as likely (relative risk, 2.07) to fail to conceive as seronegative cows. Mean number of breedings required per conception for seropositive cows (3.4) was significantly higher than that for seronegative cows (2.1). The proportion of seropositive cows that aborted was not significantly different from the proportion of seronegative cows that aborted. CONCLUSIONS AND CLINICAL RELEVANCE Exposure of nonvaccinated dairy cows to L hardjo can be associated with a subsequent reduction in fertility, as indicated by a greater time from calving to conception and higher number of breedings required per conception. The efficacy of leptospiral vaccines should be assessed to determine whether vaccination will minimize herd infertility associated with L hardjo infection.
High-Yield Di-Rhamnolipid Production by Pseudomonas aeruginosa YM4 and its Potential Application in MEOR

Abstract:Rhamnolipids are a mixture of the homologs species due to variations in the rhamnose units and β-hydroxy fatty acid moieties, mainly including Rha-C10-C10, Rha-Rha-C10-C10, and Rha-C10. In this study, strain P. aeruginosa YM4 was selected for its capacity to efficiently produce di-rhamnolipid (Rha-Rha-C10-C10) as the predominant component with soybean oil and glycerol as carbon source, accounting for 64.8% and 85.7% of total products, respectively. The critical micelle concentration (CMC) of rhamnolipid products varies with the content of di-rhamnolipid, whereby lower CMC values corresponding to higher di-rhamnolipid contents. The rhamnolipids containing 85.7% di-rhamnolipid had the lowest CMC value of 50 mg/L. Accordingly the viscosity-reducing efficiency and oil-washing efficiency of rhamnolipids increased with higher di-rhamnolipid component. At a concentration of 500 mg/L, the rhamnolipids containing 85.7% di-rhamnolipid worked best and showed 82.5% oil-washing efficiency, which offered great promise for applications in enhanced oil recovery. The results showed the variation of structure and composition of rhamnolipids had a significant effect on their application.

Abstract:In the growing landscape of interest in natural surfactants, selecting the appropriate one for specific applications remains challenging. The extensive, yet often unsystematized, knowledge of microbial surfactants, predominantly represented by rhamnolipids (RLs), typically does not translate beyond the conditions presented in scientific publications. This limitation stems from the numerous variables and their interdependencies that characterize microbial surfactant production. We hypothesized that a computational recipe for biosynthesizing RLs with targeted applicational properties could be developed from existing literature and experimental data. We amassed literature data on RL biosynthesis and micellar solubilization and augmented it with our experimental results on the solubilization of triglycerides (TGs), a topic underrepresented in current literature. Utilizing this data, we constructed mathematical models that can predict RL characteristics and solubilization efficiency, represented as log P RL = f (carbon and nitrogen source, parameters of biosynthesis) and log MSR = f (solubilizate, rhamnolipid (e.g. log P RL ), parameters of solubilization), respectively. The models, characterized by robust R 2 values of respectively 0.581–0.997 and 0.804, enabled the ranking of descriptors based on their significance and impact—positive or negative—on the predicted values. These models have been translated into ready-to-use calculators, tools designed to streamline the selection process for identifying a biosurfactant optimally suited for intended applications.

High mono-rhamnolipids production by a novel isolate Pseudomonas aeruginosa LP20 from oily sludge: characterization, optimization, and potential application

Abstract:Abstract This study aims to isolate microbial strains for producing mono-rhamnolipids with high proportion. Oily sludge is rich in petroleum and contains diverse biosurfactant-producing strains. A biosurfactant-producing strain LP20 was isolated from oily sludge, identified as Pseudomonas aeruginosa based on phylogenetic analysis of 16S rRNA. High-performance liquid chromatography-mass spectrometry results indicated that biosurfactants produced from LP20 were rhamnolipids, mainly containing Rha-C8-C10, Rha-C10-C10, Rha-Rha-C8-C10, Rha-Rha-C10-C10, Rha-C10-C12:1, and Rha-C10-C12. Interestingly, more mono-rhamnolipids were produced by strain LP20 with a relative abundance of 64.5%. Pseudomonas aeruginosa LP20 optimally produced rhamnolipids at a pH of 7.0 and a salinity of 0.1% using glycerol and nitrate. The culture medium for rhamnolipids by strain LP20 was optimized by response surface methodology. LP20 produced rhamnolipids up to 6.9 g L−1, increased by 116%. Rhamnolipids produced from LP20 decreased the water surface tension to 28.1 mN m−1 with a critical micelle concentration of 60 mg L−1. The produced rhamnolipids emulsified many hydrocarbons with EI24 values higher than 56% and showed antimicrobial activity against Staphylococcus aureus and Cladosporium sp. with inhibition rates 48.5% and 17.9%, respectively. Pseudomonas aeruginosa LP20 produced more proportion of mono-rhamnolipids, and the LP20 rhamnolipids exhibited favorable activities and promising potential in microbial-enhanced oil recovery, bioremediation, and agricultural biocontrol.

Abstract:Surfactant adsorption loss seriously hinders the economy of surfactant binary flooding technology for enhancing oil recovery, especially for biosurfactants with higher manufacturing costs. Here, biosurfactant rhamnolipid (RL) is chemically modified to develop a more efficient surfactant, rhamnolipid monoethanol amide (RL-MEA), which is characterized by decreased adsorption loss and increased oil-washing efficiency for enhanced oil recovery at a laboratory scale. Synthesis and characterization of the rhamnolipid monoethanol amide are carried out using high-performance liquid chromatography (HPLC), HPLC/MS, 1H nuclear magnetic resonance (NMR), and Fourier transform infrared (FTIR) spectroscopy. The aggregation behavior is disclosed by surface tension, dynamic light scattering, and fluorescence spectra with pyrene as the probe. The applied performances of RL-MEA in the simulated enhanced oil recovery are researched, including the efficiency of oil washing, wettability to crude oil, and adsorption isotherms on silicates. Compared with the critical micelle concentration (CMC) of rhamnolipid of 14.23 × 10-5 M in pure water and 9.02 × 10-5 M in 0.2 M NaCl solution, the modified RL-MEA shows a significantly lower CMC of 7.15 × 10-5 M in pure water and 5.34 × 10-5 M in 0.2 M NaCl solution. More importantly, the modified RL-MEA reduces adsorption loss by 20% and enhanced oil-washing efficiency at higher temperatures and salt concentrations compared with the parent RLs. These findings would provide valuable information for developing efficient surfactant flooding agents for harsh reservoir geological conditions.

Optimizing Systems for Robust Heterologous Production of Biosurfactants Rhamnolipid and Lyso-Ornithine Lipid in Pseudomonas putida KT2440

Abstract:Surfactants are amphiphilic molecules that are capable of mixing water and oil. Biosurfactants are eco-friendly, low-toxicity, and stable to a variety of environmental factors. Optimizing conditions for microorganisms to produce biosurfactants can lead to improved production suitable for scaling up. In this study, we compared heterologous expression levels of the luminescence system luxCDABE operon controlled by regulatable promoters araC-PBAD and its strong version araC-PBAD-SD in Escherichia coli K12, Pseudomonas aeruginosa PAO1, and P. putida KT2440. Real-time monitoring of luminescence levels in the three strains indicated that luxCDABE controlled by araC-PBAD-SD promoter with 0.2% arabinose supplementation in P. putida produced the highest level of luminescence. By using the araC-PBAD-SD promoter-controlled rhlAB expression in P. putida, we were able to produce mono-rhamnolipid at a level of 1.5 g L-1 when 0.02% arabinose was supplemented. With the same system to express olsB, lyso-ornithine lipid was produced at a level of 10 mg L-1 when 0.2% arabinose was supplemented. To our knowledge, this is the first report about optimizing conditions for lyso-ornithine lipid production at a level up to 10 mg L-1. Taken together, our results demonstrate that regulatable araC-PBAD-SD promoter in P. putida KT2440 is a useful system for heterologous production of biosurfactants.
Norepinephrine-induced relaxations in rat aorta mediated by endothelial beta adrenoceptors. Impairment by ageing and hypertension.

Abstract:The objective of the present work has been to analyze the influence of endothelium, ageing and hypertension in the norepinephrine (NE)-induced responses. For this purpose we used aortic rings from spontaneously hypertensive (SHR) and Wistar-Kyoto (WKY) rats of different ages. In rings with endothelium from 5-week-old WKY, cumulative addition of NE (0.01 and 0.1 microM) caused concentration-dependent contractions, whereas higher concentrations (1 and 10 microM) induced concentration-dependent relaxations. In 5-week-old SHR and 3-month-old WKY rats, these relaxant responses were observed only at 10 microM NE, and they disappeared in older animals from both strains. In endothelium denuded rings, NE induced only contractions, which were similar in WKY rats of different ages, but significantly increased in 6- and 12-month-old SHR. In both strains, the endothelium-dependent relaxant responses to NE were abolished by NG-nitro-L-arginine methyl ester and propranolol, but not modified by yohimbine or ouabain. Isoproterenol (0.01-10 microM) induced concentration-dependent vasodilation in rings from 5-week-old rats of both strains, precontracted with 1 microM NE. Isoproterenol-elicited responses were reduced by endothelium removal or by 0.1 mM NG-nitro-L-arginine methyl ester and abolished by 1 microM propranolol. These results suggest that: 1) in the aorta from young WKY and prehypertensive SHR rats, NE induces vasodilations mediated by activation of endothelial beta adrenoceptors and release of nitric oxide; 2) these responses are impaired during ageing and hypertension; and 3) there is an important negative endothelial modulation of NE-induced contraction in adult SHR rats.

Abstract:Rhamnolipids are one of the most effective biosurfactants that are of great interest in industrial applications such as enhancing oil recovery, health care, cosmetics, pharmaceutical processes, food processing, detergents for protein folding, and bioremediation due to their unique characteristics such as low toxicity, surface active property to reduce surface/interfacial tensions, and excellent biodegradability. The genes and metabolic pathways for rhamnolipid synthesis have been well elucidated, but its cost-effective production is still challenging. Pseudomonas aeruginosa, the most powerful rhamnolipid producer, is an opportunistic pathogen, which limits its large scale production and applications. Rhamnolipid production using engineered strains other than Pseudomonas aeruginosa such as E. coli and Pseudomonas putida has received much attention. The highest yield of rhamnolipids is achieved when oil-type carbon sources are used, but using cheaper and renewable carbon sources such as lignocellulose would be an attractive strategy to reduce the production cost of rhamnolipids for various industrial applications.

Abstract:Pseudomonas aeruginosa produces the biosurfactants rhamnolipids and 3-(3-hydroxyalkanoyloxy)alkanoic acids (HAAs). In this study, we report the production of one family of rhamnolipids, specifically the monorhamnolipids, and of HAAs in a recombinant Escherichia coli strain expressing P. aeruginosa rhlAB operon. We found that the availability in E. coli of dTDP-l-rhamnose, a substrate of RhlB, restricts the production of monorhamnolipids in E. coli. We present evidence showing that HAAs and the fatty acid dimer moiety of rhamnolipids are the product of RhlA enzymatic activity. Furthermore, we found that in the recombinant E. coli, these compounds have the same chain length of the fatty acid dimer moiety as those produced by P. aeruginosa. These data suggest that it is RhlAB specificity, and not the hydroxyfatty acid relative abundance in the bacterium, that determines the profile of the fatty acid moiety of rhamnolipids and HAAs. The rhamnolipids level produced in recombinant E. coli expressing rhlAB is lower than the P. aeruginosa level and much higher than those reported by others in E. coli, showing that this metabolic engineering strategy lead to an increased rhamnolipids production in this heterologous host.

Abstract:The effect of biosurfactant rhamnolipid (RL) on hydrolysis and acidification of waste activated sludge (WAS) was investigated. The results indicated that RL could greatly reduce the surface tension of sludge, resulting in stimulating the hydrolysis rate of WAS and enhancing the production of short-chain fatty acids (SCFAs). With the increase of RL dosage from 0.2 to 0.5 g/g DS, the maximum soluble chemical oxygen demand (SCOD), protein and carbohydrate concentration increased correspondingly. After 6 h of hydrolysis, SCOD, protein and carbohydrate concentration increased from 371.9, 93.3 and 9.0 mg/l to 3,994.5, 800.0 and 401.4 mg/l at RL 0.3 g/g DS, respectively. Furthermore, the release of NH4 +-N, PO4 3−-P and the accumulation of SCFAs also improved in the presence of RL. The maximum SCFAs was 1,829.9 mg COD/l at RL 0.3 g/g DS, while it was only 377.7 mg COD/l for the blank test. The propionic acid and acetic acid were the mainly SCFAs produced, accounting for 50–60% of total SCFAs.
Optimization of Rhamnolipid Hydrolysis Conditions Using Response Surface Methodology

Abstract:The response surface methodology (RSM) was used to optimize the hydrolysis reaction conditions of rhamnolipids for preparation of rhamnose. On the basis of single factor experiments, three-factor response surface analysis method was used to study the hydrolysis yield of rhamnolipid in the hydrolysis process using the amount of sulfuric acid, reaction temperature, and reaction time as three-levels according to the Box-Behnken central combination design principle. The regression equation between the hydrolysis yield and three factors was fitted to optimize the hydrolysis reaction conditions of rhamnolipids. The results showed that the amount of sulfuric acid and the reaction temperature had significant effect on the hydrolysis of rhamnolipid. The interaction between the amount of sulfuric acid and hydrolysis time, and the interaction between hydrolysis temperature and time both have significant effects. The quadratic effects of various factors were extremely significant. The optimal hydrolysis conditions derived from the model were:2.3％ (mass fraction) of sulfuric acid, hydrolysis temperature of 137 ℃ and the hydrolysis time of 2.8 h. Under these conditions, the actual yield of rhamnose was 81.6％, which was in agreement with the predicted value of 82.35％. Finally, the model was futher applied to 3 m3 industrial-scale hydrolysis production and the prediction error was found to be in between -8.12％ and 9.30％, indicating that the model had a good reference value for industrial production.
