Abstract:Differences in the rhamnolipid structures must result in its different activities, thus affecting its application effect. The rhlC gene in Pseudomonas aeruginosa SG was knocked out to construct strain P. aeruginosa SGΔrhlC. Rhamnolipid production was enhanced by 23.3% through knocking out rhlC gene. P. aeruginosa SGΔrhlC produced 14.22 g/L of rhamnolipid using glycerol and nitrate. Five kinds of mono-rhamnolipid but no di-rhamnolipid were produced by strain SGΔrhlC. The main rhamnolipid homologues were Rha-C10-C10, Rha-C10-C12:1 and Rha-C10-C12. Mono-rhamnolipid exhibited better antimicrobial activity to Escherichia coli, Staphylococcus aureus, Aspergillus niger and Penicillium chrysogenum. Rhamnolipid produced from strain SGΔrhlC showed greater emulsifying activity to crude oil with EI24 of 84.73%. Rhamnolipid produced from strain SGΔrhlC efficiently washed oily sludge at 35 °C. High-producing strain P. aeruginosa SGΔrhlC and its produced mono-rhamnolipid are more promising in agriculture and petroleum industry. This study is a step forward to the tailor-made biosynthesis and application of rhamnolipid.

Abstract:OBJECTIVE:We amplified and overexpressed the FHL activator (fh1A) in E. aerogenes ATCC13408 to enhance hydrogen production.METHODS:By using universal primers and genome walking, we cloned the full open reading frame (ORF) of fh1A gene. We inserted it into the glutathion S-transferase (GST) fusion expression vector pGEX4T-2-Cat, and transformed the recombinant plasmid into E. aerogenes ATCC13408 via electroporation for expression. Then we measured the hydrogen production of the recombinant strain in a batch culture.RESULTS:We found that the ORF of fh1A was 2073 base pair in length, potential to encode a 690 amino acid peptide (GenBank accession GU188474). The Fh1A protein from E. aerogenes ATCC13408 shared high amino acid identities with those from other bacterial species. By using SDS-PAGE and Western blot analysis, we confirmed that the fh1A gene had successfully expressed in the strain. The hydrogen yield of the recombinant strain was increased from 1.23 to 1.48 mol H2/mol glucose. [ Conclusion ] Enhancement of hydrogen productivity was attained under anaerobic conditions with the recombinant strain.
A Rare Mono-Rhamnolipid Congener Efficiently Produced by Recombinant Pseudomonas aeruginosa YM4 via the Expression of Global Transcriptional Regulator irrE

Abstract:Rhamnolipids (RLs) are widely used biosurfactants produced mainly by Pseudomonas aeruginosa and Burkholderia spp. in the form of mixtures of diverse congeners. The global transcriptional regulator gene irrE from radiation-tolerant extremophiles has been widely used as a stress-resistant element to construct robust producer strains and improve their production performance. A PrhlA-irrE cassette was constructed to express irrE genes in the Pseudomonas aeruginosa YM4 of the rhamnolipids producer strain. We found that the expression of irrE of Deinococcus radiodurans in the YM4 strain not only enhanced rhamnolipid production and the strain's tolerance to environmental stresses, but also changed the composition of the rhamnolipid products. The synthesized rhamnolipids reached a maximum titer of 26 g/L, about 17.9% higher than the original, at 48 h. The rhamnolipid production of the recombinant strain was determined to be mono-rhamnolipids congener Rha–C10–C12, accounting for 94.1% of total products. The critical micelle concentration (CMC) value of the Rha–C10–C12 products was 62.5 mg/L and the air-water surface tension decreased to 25.5 mN/m. The Rha–C10–C12 products showed better emulsifying activity on diesel oil than the original products. This is the first report on the efficient production of the rare mono-rhamnolipids congener Rha–C10–C12 and the first report that the global regulator irrE can change the components of rhamnolipid products in Pseudomonas aeruginosa.

Anaerobic biosynthesis of rhamnolipids by Pseudomonas aeruginosa: performance, mechanism and its application potential for enhanced oil recovery

Abstract:Background: Pseudomonas aeruginosa, the rhamnolipids-producer, is one of dominant bacteria in oil reservoirs. Although P. aeruginosa strains are facultative bacteria, the anaerobic biosynthesis mechanism of rhamnolipids is unclear. Considering the oxygen scarcity within oil reservoirs, revealing the anaerobic biosynthesis mechanism of rhamnolipids are significant for improving the in-situ production of rhamnolipids in oil reservoirs to enhance oil recovery. Results: Pseudomonas aeruginosa SG anaerobically produced rhamnolipids using glycerol rather than glucose as carbon sources. Two possible hypotheses on anaerobic biosynthesis of rhamnolipids were proposed, the new anaerobic biosynthetic pathway (hypothesis 1) and the highly anaerobic expression of key genes (hypothesis 2). Knockout strain SGΔrmlB failed to anaerobically produce rhamnolipids using glycerol. Comparative transcriptomics analysis results revealed that glucose inhibited the anaerobic expression of genes rmlBDAC, fabABG, rhlABRI, rhlC and lasI. Using glycerol as carbon source, the anaerobic expression of key genes in P. aeruginosa SG was significantly up-regulated. The anaerobic biosynthetic pathway of rhamnolipids in P. aeruginosa SG were confirmed, involving the gluconeogenesis from glycerol, the biosynthesis of dTDP-L-rhamnose and β-hydroxy fatty acids, and the rhamnosyl transfer process. The engineered strain P. aeruginosa PrhlAB constructed in previous work enhanced 9.67% of oil recovery higher than the wild-type strain P. aeruginosa SG enhancing 8.33% of oil recovery. Conclusion: The highly anaerobic expression of key genes enables P. aeruginosa SG to anaerobically biosynthesize rhamnolipids. The genes, rmlBDAC, fabABG, rhlABRI, rhlC and lasI, are key genes for anaerobic biosynthesis of rhamnolipid by P. aeruginosa. Improving the anaerobic production of rhamnolipids better enhanced oil recovery in core flooding test. This study fills the gaps in the anaerobic biosynthesis mechanism of rhamnolipids. Results are significant for the metabolic engineering of P. aeruginosa to enhance anaerobic production of rhamnolipids.

Abstract:Pseudomonas putida KT2440, a GRAS strain, has been used for synthesizing bulk and fine chemicals. However, the gene editing tool to metabolically engineer KT2440 showed low efficiency. In this study, a novel sacB -based system pK51mobsacB was established to improve the efficiency for marker-free gene disruption. Then the rhamnolipid synthetic pathway was introduced in KT2440 and genes of the competitive pathways were deleted to lower the metabolic burden based on pK51mobsacB. A series of endogenous and synthetic promoters were used for fine tuning rhlAB expression. The limited supply of dTDP-L-rhamnose was enhanced by heterologous rmlBDAC expression. Cell growth and rhamnolipid production were well balanced by using glucose/glycerol as mixed carbon sources. The final strain produced 3.64 g/L at shake-flask and 19.77 g/L rhamnolipid in a 5 L fermenter, the highest obtained among metabolically engineered KT2440, which implied the potential of KT2440 as a promising microbial cell factory for industrial rhamnolipid production.

Heterologous production of rhamnolipids in Pseudomonas chlororaphis subsp chlororaphis ATCC 9446 based on the endogenous production of N-acyl-homoserine lactones

Abstract:Rhamnolipids (RL) are biosurfactants naturally produced by the opportunistic pathogen Pseudomonas aeruginosa. Currently, RL are commercialized for various applications and produced by Pseudomonas putida due to the health risks associated with their large-scale production by P. aeruginosa. In this work, we show that RL containing one or two rhamnose moieties (mono-RL or di-RL, respectively) can be produced by the innocuous soil-bacterium Pseudomonas chlororaphis subsp chlororaphis ATCC 9446 at titres up to 66 mg/L (about 86% of the production of P. aeruginosa PAO1 in the same culture conditions). The production of RL depends on the expression of P. aeruginosa PAO1 genes encoding the enzymes RhlA, RhlB and RhlC. These genes were introduced in a plasmid, together with a transcriptional regulator (rhlR) forming part of the same operon, with and without RhlC. We show that the activation of rhlAB by RhlR depends on its interaction with P. chlororaphis endogenous acyl-homoserine lactones, which are synthetized by either PhzI or CsaI autoinducer synthases (producing 3-hydroxy-hexanoyl homoserine lactone, 3OH-C6-HSL, or 3-oxo-hexanoyl homoserine lactone, 3O-C6-HSL, respectively). P. chlororaphis transcriptional regulator couple with 3OH-C6-HSL is the primary activator of gene expression for phenazine-1-carboxylic acid (PCA) and phenazine-1-carboxamide (PCN) production in this soil bacterium. We show that RhlR coupled with 3OH-C6-HSL or 3O-C6-HSL promotes RL production and increases the production of PCA in P. chlororaphis. However, PhzR/3OH-C6-HSL or CsaR/3O-C6-HSL cannot activate the expression of the rhlAB operon to produce mono-RL. These results reveal a complex regulatory interaction between RhlR and P. chlororaphis quorum-sensing signals and highlight the biotechnology potential of P. chlororaphis ATCC 9446 expressing P. aeruginosa rhlAB-R or rhlAB-R-C for the industrial production of RL.

Abstract:Biosurfactants have significant applications in various industries, including microbial-enhanced oil recovery (MEOR). While the state-of-the-art genetic approaches can generate high-yield strains for biosurfactant production in fermenters, there remains a critical challenge in enhancing biosurfactant-producing strains for use in natural environments with minimal ecological risks. The objectives of this work are enhancing the strain's capacity for rhamnolipids production and exploring the genetic mechanisms for its improvement. In this study, we employed atmospheric and room-temperature plasma (ARTP) mutagenesis to enhance the biosynthesis of rhamnolipids in Pseudomonas sp. L01, a biosurfactant-producing strain isolated from petroleum-contaminated soil. Following ARTP treatment, we identified 13 high-yield mutants, with the highest yield of 3.45 ± 0.09 g/L, representing a 2.7-fold increase compared to the parent strain. To determine the genetic mechanisms behind the enhanced rhamnolipids biosynthesis, we sequenced the genomes of the strain L01 and five high-yield mutants. A comparative genomic analysis suggested that mutations in genes related to the synthesis of lipopolysaccharides (LPS) and the transport of rhamnolipids may contribute to the improved biosynthesis. To the best of our knowledge, this is the first instance of utilizing the ARTP approach to improve rhamnolipid production in Pseudomonas strains. Our study provides valuable insights into the enhancement of biosurfactant-producing strains and the regulatory mechanisms of rhamnolipids biosynthesis.

Abstract:An indigenous strain Pseudomonas aeruginosa S2 (P. aeruginosa S2), isolated from diesel-contaminated soil, produced extracellular surface-active material identified as rhamnolipid. Due to its excellent surface activity, rhamnolipid is known to be well-suited for stimulating the bioremediation efficiency of oil contaminated sites. To improve production yield of rhamnolipid with P. aeruginosa S2, various carbon and nitrogen sources were screened to select favorable ones leading to better biosurfactant production yield. It was found that using 4% glucose could attain better rhamnolipid yield, while 50 mM NH4NO3 appeared to be the most preferable nitrogen source. Meanwhile, the effect of carbon to nitrogen ratio (C/N ratio) on rhamnolipid yield was also investigated, and the optimal C/N ratio was identified as approximately 11.4. Moreover, response surface methodology (RSM) was applied to optimize the trace element concentration for rhamnolipid production. Results from two-level design indicate that concentrations of MgSO4 and FeSO4 were the most significant factors affecting rhamnolipid production. Using steepest ascent method and RSM analysis, an optimal medium composition was determined, giving a rhamnolipid production yield of 2.37 g/L in 100 h at 37 degrees C and 200 rpm agitation. Scale-up production of rhamnolipid in a well-controlled 5 L jar fermentor using the optimal medium and operating condition (at 37 degrees C and pH 6.8) further elevated the biosurfactant production yield to 5.31 g/L (in 97 h), which is over 2-fold higher than the best results obtained from shake-flask tests.

Abstract:Surfactants are amphiphilic molecules that are capable of mixing water and oil. Biosurfactants are eco-friendly, low-toxicity, and stable to a variety of environmental factors. Optimizing conditions for microorganisms to produce biosurfactants can lead to improved production suitable for scaling up. In this study, we compared heterologous expression levels of the luminescence system luxCDABE operon controlled by regulatable promoters araC-PBAD and its strong version araC-PBAD-SD in Escherichia coli K12, Pseudomonas aeruginosa PAO1, and P. putida KT2440. Real-time monitoring of luminescence levels in the three strains indicated that luxCDABE controlled by araC-PBAD-SD promoter with 0.2% arabinose supplementation in P. putida produced the highest level of luminescence. By using the araC-PBAD-SD promoter-controlled rhlAB expression in P. putida, we were able to produce mono-rhamnolipid at a level of 1.5 g L-1 when 0.02% arabinose was supplemented. With the same system to express olsB, lyso-ornithine lipid was produced at a level of 10 mg L-1 when 0.2% arabinose was supplemented. To our knowledge, this is the first report about optimizing conditions for lyso-ornithine lipid production at a level up to 10 mg L-1. Taken together, our results demonstrate that regulatable araC-PBAD-SD promoter in P. putida KT2440 is a useful system for heterologous production of biosurfactants.
Monorhamnolipids and 3-(3-hydroxyalkanoyloxy)alkanoic acids (HAAs) production using Escherichia coli as a heterologous host

Abstract:Pseudomonas aeruginosa produces the biosurfactants rhamnolipids and 3-(3-hydroxyalkanoyloxy)alkanoic acids (HAAs). In this study, we report the production of one family of rhamnolipids, specifically the monorhamnolipids, and of HAAs in a recombinant Escherichia coli strain expressing P. aeruginosa rhlAB operon. We found that the availability in E. coli of dTDP-l-rhamnose, a substrate of RhlB, restricts the production of monorhamnolipids in E. coli. We present evidence showing that HAAs and the fatty acid dimer moiety of rhamnolipids are the product of RhlA enzymatic activity. Furthermore, we found that in the recombinant E. coli, these compounds have the same chain length of the fatty acid dimer moiety as those produced by P. aeruginosa. These data suggest that it is RhlAB specificity, and not the hydroxyfatty acid relative abundance in the bacterium, that determines the profile of the fatty acid moiety of rhamnolipids and HAAs. The rhamnolipids level produced in recombinant E. coli expressing rhlAB is lower than the P. aeruginosa level and much higher than those reported by others in E. coli, showing that this metabolic engineering strategy lead to an increased rhamnolipids production in this heterologous host.

Optimization of mineral nutrients to improve rhamnolipid production by Pseudomonas aeruginosa 6 K‐11

Abstract:BACKGROUND Rhamnolipids are biosurfactants that have their main application in bioremediation processes. They are able to increase the biodegradation and detoxification rates from industrial effluents. The north of Peru and Iquitos have been not only major oil producers but also have been responsible for oil spills. This study therefore focused on the optimization of mineral nutrients such as nitrogen, phosphorous, calcium and iron to improve rhamnolipids production by Pseudomonas aeruginosa 6 K‐11, which was isolated from oil soil in Peru. RESULTS Pseudomonas aeruginosa 6 K‐11 was reactivated in Trypticase Soy Broth by scaling from 5 to 200 mL of culture medium for 8 h at each stage. The culture was centrifuged and placed in 100 mL Siegmund–Wagner medium for 36 h in a 500 mL Erlenmeyer flask. First, it was determined that sodium nitrate, monopotassium phosphate, calcium chloride and iron sulfate were the best sources of mineral nutrients. Then, by using corn oil as carbon source, the ratios obtained for carbon‐to‐nitrogen and carbon‐to‐phosphorous were 21.172 and 16.279, respectively. In addition, it was found that the optimal concentration for calcium was 0.046 g L−1 and for iron was 0.003 g L−1. The most influential variables were carbon‐to‐nitrogen and carbon‐to‐phosphorus ratio. Finally, 35.124 g L−1 rhamnolipids was produced after 168 h of fermentation with the optimal parameters. CONCLUSION The optimization of mineral nutrients was an important achieved goal for rhamnolipid production. In addition, this optimization can be used for modifying the culture medium to grow the biosurfactant producer strain. © 2024 Society of Chemical Industry (SCI).

Enhancing rhamnolipid production through a two-stage fermentation control strategy based on metabolic engineering and nitrate feeding

Abstract:Nitrate plays a crucial role in the high-efficient fermentation production of rhamnolipids (RLs). However, the underlying mechanism remains unclear. Firstly, by knocking out the restriction endonuclease PaeKI and utilizatiing the endogenous CRISPR-Cas-mediated single-plasmid recombineering system, a genome editing system for P. aeruginosa KT1115 has been established. Secondly, an engineered strain KT1115ΔpaeKIΔnirS was obtained with a 87% of reduction in nitric oxide (NO) accumulation and a 93% of reduction in RLs production, revealing the crucial role of NO signaling molecule produced from nitrate metabolism in RLs production. Finally, by combining metabolic engineering of the nitrate metabolism pathway with nitrogen feeding, a new two-stage fermentation process was developed. The fermentation production period was reduced from 168 h to 120 h while achieving a high yield of 0.8 g/g, and the average productivity increased by 55%. In all, this study provides a novel insights in the RLs biosynthesis and fermentation control strategy.

Abstract:Rhamnolipids (RLs) are biosurfactants with great economic significance that have been used extensively in multiple industries. Pseudomonas aeruginosa is a promising microorganism for sustainable RL production. However, current CTAB-MB based screening of RL-producing strains is time-consuming, labor-intensive, and unable to distinguish mono-and di-RL. In this study, we developed a novel transcription-associated fluorescence-activated droplet sorting (FADS) method to specifically target the di-RL hyperproducers. We first investigated critical factors associated with this method, including the specificity and sensitivity for discriminating di-RL overproducers from other communities. Validation of genotype-phenotype linkage between the GFP intensity, rhlC transcription, and di-RL production showed that rhlC transcription is closely correlated with di-RL production, and the GFP intensity is responsive to rhlC transcription, respectively. Using this platform, we screened out ten higher di-RL producing microorganisms, which produced 54-208% more di-RL than the model P. aeruginosa PAO1. In summary, the droplet-based microfluidic platform not only facilitates a more specific, reliable, and rapid screening of P. aeruginosa colonies with desired phenotypes, but also shows that intracellular transcription-associated GFP intensity can be used to measure the yield of di-RL between populations of droplets containing different environmental colonies. This method also can be integrated with transposon mutation libraries to target P. aeruginosa mutants.

Abstract:Rhamnolipids are the most promising biosurfactants, have widespread applications in many fields. However, low yield and productivity in fermentation caused a high production cost and thus prohibited the bulk applications of rhamnolipids in industry. In this study, a sequential fed-batch fermentation process with fill-and-draw operation was developed to improve rhamnolipids production. By utilizing this strategy, the total produced rhamnolipids reached over 150g/L, had a 163% and 102% increase over the traditional batch and fed-batch processes, respectively. This remarkable high production efficiency was achieved by the well-maintained high productivity of 0.4g/Lh for a period of 17 d. Astonishingly, the conversion yield was high as 84%, while this value was only 53.2% and 42.7% in the traditional batch and fed-batch process, respectively. The high-efficiency rhamnolipids production in this sequential fed-batch fermentation could be largely explained by a high presence of cell coupled with the replenishment of nutrients and dilution of toxic byproducts via fill-and-draw operation. In all, this validated fermentation strategy offers a great prospect for high-efficiency production of rhamnolipids in industry.

Abstract:To construct a derivative of the avirulent Pseudomonas aeruginosa ATCC 9027 that produces high levels of di-rhamnolipid, that has better physico-chemical characteristics for biotechnological applications than mono-rhamnolipid, which is the sole type produced by ATCC 9027. We used plasmids expressing the rhlC gene, which encodes for rhamnosyl transferase II that transforms mono- to di-rhamnolipids under different promoters and in combination with the gene coding for the RhlR quorum sensing regulator, or the mono-rhamnolipid biosynthetic rhlAB operon. The plasmids tested carrying the rhlC gene under the lac promoter were plasmid p rhlC and p rhlRC , while p rhlAB-R–C expressed this gene from the rhlA promoter, forming part of the artificially constructed rhlAB-R–C operon. We measured rhamnolipds concentrations using the orcinol method and determined the proportion of mono-rhamnolipids and di-rhamnolipids by UPLC/MS/MS. We found that the expression of rhlC in P. aeruginosa ATCC 9027 caused the production of di-rhamnolipids and that the derivative carrying plasmid p rhlAB-R–C gives the best results considering total rhamnolipids and a higher proportion of di-rhamnolipids. A P. aeruginosa ATCC 9027 derivative with increased di-rhamnolipids production was developed by expressing plasmid p rhlAB-R–C , that produces similar rhamnolipids levels as PAO1 type-strain and presented a higher proportion of di-rhamnolipids than this type-strain.

Abstract:Rhamnolipids are one of the most effective biosurfactants that are of great interest in industrial applications such as enhancing oil recovery, health care, cosmetics, pharmaceutical processes, food processing, detergents for protein folding, and bioremediation due to their unique characteristics such as low toxicity, surface active property to reduce surface/interfacial tensions, and excellent biodegradability. The genes and metabolic pathways for rhamnolipid synthesis have been well elucidated, but its cost-effective production is still challenging. Pseudomonas aeruginosa, the most powerful rhamnolipid producer, is an opportunistic pathogen, which limits its large scale production and applications. Rhamnolipid production using engineered strains other than Pseudomonas aeruginosa such as E. coli and Pseudomonas putida has received much attention. The highest yield of rhamnolipids is achieved when oil-type carbon sources are used, but using cheaper and renewable carbon sources such as lignocellulose would be an attractive strategy to reduce the production cost of rhamnolipids for various industrial applications.

Abstract:The use of efficient green cleaning agents, such as biosurfactants, is important in oil sludge treatment. Enhanced oil recovery from oily sludge by different rhamnolipids was comparatively evaluated. Using crude glycerol, the wild-type strain Pseudomonas aeruginosa SG and the recombinant strains P. aeruginosa PrhlAB and P. stutzeri Rhl produced 1.98 g L-1, 2.87 g L-1 and 0.87 g L-1 of rhamnolipids, respectively. The three bacterial strains produced different rhamnolipid mixtures under the same conditions. The proportions of mono-rhamnolipids in the three rhamnolipid products were 55.92%, 94.92% and 100%, respectively. These rhamnolipid products also possessed different bioactivities. Emulsifying activity became higher as the proportion of mono-rhamnolipids increased. The three rhamnolipid products were stable at temperatures lower than 121 °C, pH values from 5-11 and NaCl concentrations from 0-15%. All three rhamnolipid products could recover oil from oily sludge, but oil recovery efficiency was positively related to the proportion of mono-rhamnolipids. Mono-rhamnolipids produced by the recombinant strain Rhl exhibited the best oil recovery efficiency (53.81%). The results reveal that mono-rhamnolipids are the most promising for oil recovery from oily sludge.

Abstract:The physiological role of Coenzyme Q10 (CoQ10) as an electron carrier suggests its association with redox potential. Overexpression of glyceraldehyde-3-phosphate dehydrogenase type I (gapA-1) in Rhodobacter sphaeroides elevated the NADH/NAD+ ratio and meanwhile enhanced the CoQ10 content by 58%, but at the sacrifice of biomass. On the other hand, Vitreoscilla hemoglobin was heterologously expressed to enhance the oxygen uptake ability of the cells, leading to 127% improvement of biomass. Subsequent coexpression of gapA-1 and vgb resulted in a CoQ10 titer of 83.24mg/L, representing 71% improvement as compared to the control strain RspMCS. When gapA-1 and vgb genes were co-expressed in a previously created strain RspMQd [1], 163.5mg/L of CoQ10 was produced. Finally, 600mg/L of CoQ10 production was achieved in fed-batch fermentation. These results demonstrated the synergic effect of redox potential regulation and oxygen uptake improvement on enhancing CoQ10 production in R. sphaeroides.

Abstract:The biodegradation of crude oil is becoming a severe problem.Biosurfacnt producer could facilitate biodegradation of crude oil via enhancing its bioavailability.This paper investigates the effect of salinity,concentration of Mg2+、Ca2+ and acclimation on the biomass and rhamnolipid productivity of Pseudomonas aeruginosa ZJU.It was found that:the acclimation of Pseudomonas aeruginosa ZJU adapted well to the high salinity condition of the S35 synthetic seawater medium which has the similar salinity as deep sea;moreover,the concentration of rhamnolipid in the S25,S35,S40 synthetic seawater medium at the seventh day reached 14.6 g/L,11.8 g/L,10.8 g/L respectively by using the vegetable oil as the carbon source.

Abstract:Significant progress has recently been made in the biosynthesis of germacrene A using microbial cell factories. Germacrene A is a crucial precursor for the synthesis of anti‐cancer active compounds. However, its hydrophobic characteristics lead to its aggregation in cell membranes and cause severe cytotoxicity. In the present study, we found that rhamnolipids (RLs), as toxicity antidotes, could promote the production of germacrene A. An optimal RLs concentration of 1.25 g L−1 resulted in an increase of over 30% in the germacrene A titer at both shake flask and bioreactor scales. Mechanistic analysis showed that the addition of RLs could dramatically reduce aqueous‐phase surface tension and cell surface hydrophobicity (CSH), and increase the cell membrane permeability. This, in turn, promoted an efficient transfer of germacrene A from cell membrane to extraction phases. The addition of RLs also increased the adenosine triphosphate (ATP) concentration and the nicotinamide adenine dinucleotide (NAD+/NADH) ratio, while reducing reactive oxygen species (ROS) levels. Correspondingly, gene transcripts for key enzymes associated with germacrene A biosynthesis, the respiratory chain, and ROS scavenging were upregulated significantly. This study provides an effective RLs‐regulated fermentation method for the biosynthesis of hydrophobic natural products.
