Abstract:A novel electrochemical microbial biosensor for the rapid monitoring of p-nitrophenyl-substituted organophosphates (OPs) compounds based on glass carbon electrode (GCE) modified with both ordered mesopore carbons (OMCs) and cell surface-expressed organophosphorus hydrolase (OPH) (OPH-bacteria/OMCs/GCE) was described in this paper. The genetically engineered Escherichia coli strain surface displayed mutant OPH (S5) with improved enzyme activity and favorable stability was constructed using a newly identified N-terminal of ice nucleation protein as an anchoring motif, which can be used directly without further time-consuming enzyme-extraction and purification, thereafter greatly improved the stability of the enzyme. Compared to OPH-bacteria modified GCE (OPH-bacteria/GCE), the OPH-bacteria/OMCs/GCE not only significantly enhanced the current response but also reduced the oxidation overpotential towards oxidizable p-nitrophenol (p-NP), which was the hydrolysate of p-nitrophenyl-substituted OPs. Under the optimized experimental conditions, at +0.84V (vs. SCE), the current–time curve was performed with varying OPs concentration. The current response was linear with paraoxon concentration within 0.05–25μM. Similarly, linear range of 0.05–25μM was found for parathion, and 0.08–30μM for methyl parathion. The low limits of detection were evaluated to be 9.0nM for paraoxon, 10nM for parathion and 15nM for methyl parathion (S/N=3). Thus, a highly specific, sensitive and rapid microbial biosensor was established, which holds great promise for on-site detection of trace p-nitrophenyl-substituted OPs.

Abstract:In modern agricultural practices, organophosphorus pesticides or insecticides (OPs) are regularly used to restrain pests. Their limits are closely monitored since their residual hinders the capability of acetylcholinesterase (AChE) and brings out a threatening accumulation of the neurotransmitter acetylcholine (ACh), which affects human well-being. Therefore, spotting OPs in food and the environment is compulsory to prevent human health. Several techniques are available to identify OPs but encounter shortcomings like time-consuming, operating costs, and slow results achievement, which calls for further solutions. Herein, we present a rapid colorimetric sensor for quantifying OPs in foods using TMB as a substrate, a multi-enzyme cascade system, and the synergistic property of core-shell Palladinum@Platinum (Pd@Pt) nanoparticles. The multi-enzyme cascade response framework is a straightforward and effective strategy for OPs recognition and can resolve the previously mentioned concerns. Numerous OPs, including Carbofuran, Malathion, Parathion, Phoxim, Rojor, and Phosmet, were successfully quantified at different concentrations. The cascade method established using Pd@Pt had a simple and easy operation, a lower detection limit range of (1–2.5 ng/mL), and a short detection time of about 50 min. With an R2 value of over 0.93, OPs showed a linear range of 10–200 ng/mL, portraying its achievement in quantifying pesticide residue. Lastly, the approach was utilized in food samples and recovered more than 80% of the residual OPs.

Abstract:Organophosphorus pesticides (OPs) are widely used in pest control. The residual OPs can cause pollution to the environment and harm to the human body. Although OPs can inhibit the activity of acetylcholinesterase (AChE) and lead to neurological diseases, it also provides enlightenment for the development of a simple and sensitive method for the detection of OPs. Herein, we report a colorimetric sensor array using nanozymes (Fe-N-C nanozymes and Cu-N-C nanozymes) combined with natural enzymes to discriminate eight OPs (Iprobenfos (Ipr), Diazinon (Dia), Crotoxyphos (Cro), Bromophos ethyl (Bro), Dipterex (Dip), Chlorpyrifos methyl (Chl), Ethion (Eth), and Omethoate (Ome)) through a cascade reaction system. Surprisingly, eight OPs were successfully identified in tap water at 1 ng mL−1.

Abstract:The relation of the physiological releases of growth hormone-releasing hormone (GHRH) and growth hormone (GH) into the circulation in various conditions was investigated using a sensitive and specific radioimmunoassay for plasma GHRH. The mean fasting plasma level of immunoreactive (IR)-GHRH in 72 normal adults was 10.3 +/- 0.5 (mean +/- SEM) pg/ml and there was no significant sex difference in the level. The concentrations of IR-GHRH in plasma from the umbilical artery and umbilical vein were 107.3 +/- 20.5 pg/ml and 33.6 +/- 3.8 pg/ml, respectively, and a marked arterio-venous gradient was observed in all 12 individuals examined. The plasma level of IR-GHRH in the maternal vein was significantly lower than that in the cord blood, but was similar to that in non-pregnant women. In normal adults, although there was no apparent fluctuation in the level of plasma IR-GHRH or of plasma GH during bed rest, a significant increase of plasma IR-GHRH was detected followed by, or synchronized with the surge of plasma GH after oral administration of L-dopa. In contrast, on L-arginine infusion, no proportional elevation of plasma IR-GHRH with increase in plasma GH was observed. During and after intravenous infusion of somatostatin, the circulating IR-GHRH level did not increase, but on stopping the infusion there was an immediate and marked rebound surge of GH. We conclude that 1) the elevated IR-GHRH in the cord blood plasma originates from the fetus and may have a primary role in enhancing secretion of GH which promotes growth in early human life, and 2) the participations of GHRH in the mechanisms of GH secretion seen after administrations of L-dopa, L-arginine and somatostatin are different.

Abstract:On-site quantitative detection of organophosphorus pesticides (OPs) is crucial for safeguarding food and public safety. This study presents a novel acetylcholinesterase (AChE)-mediated paper-based Au3+-etching of gold nanobipyramids (AuNBPs) system. The system employs a long-term storable AuNBPs-deposited nylon membrane embedded within a portable and temperature-controlled paper-based analytical device. This system, coupled with a colorimeter-based quantitative method, enables the development of a practical paper-based multicolor sensor (PMS) for on-site quantitative detection of three common OPs (paraoxon, dichlorvos, and trichlorfon). In the absence of OPs, AChE hydrolyzes acetylthiocholine to thiocholine, which reduces Au3+ to Au+. The presence of OPs inhibits AChE activity, thereby preserving Au3+ to etch AuNBPs on nylon membranes, accompanied by multicolor changes. These color changes can be simply quantified by measuring the a∗ parameter of the CIELab color space using a portable colorimeter. Under optimal conditions, the PMS displayed eight OPs-corresponding color changes with a minimum detectable concentration of 1.0-10 μg/L (visual observation) and limits of detection of 0.8-7.2 μg/L (colorimeter) and 0.2-3.4 μg/L (UV-vis spectrometry). The PMS successfully determined the OPs in vegetable and rice samples with recoveries of 89.0-109 % and RSDs (n = 5) of <6 %. These results were consistent with those obtained using the HPLC-MS method. The PMS demonstrates excellent portability, AuNBPs stability, detection sensitivity, and reproducibility, making it a promising tool for the on-site quantitative detection of OPs residues in food. Furthermore, the paper-based etching system coupled with the colorimeter-based quantitative method provides a valuable reference to develop practical PMSs for various targets in diverse fields.

Abstract:A multicolor enzyme-linked immunosorbent sensor was constructed to detect organophosphates pesticides (OPs) on base of TMB2+-mediated AuNRs etching. As the etching progresses, a rainbow-like color responses was generated. The naked eye limit of detection (LOD) of the sensing system is 0.25 ng mL-1. In particular, the longitudinal localized surface plasmon resonances (LSPR) peak shift of AuNRs is linearly correlated with the logarithm of OPs concentration in range of 104 to 5×106 pg mL-1 for parathion-methyl and fenitrothion, 5×104 to 107 pg mL-1 for fenthion, respectively. Compared with reported methods, this sensor displayed satisfactory linear ranges and detection limits. Average recoveries of this method ranged from 73.28% to 98.08% and relative standard deviations (RSDs) were below 8.0% as for food samples analysis without complicated pretreatments. Remarkably, the developed multicolor enzyme-linked immunosorbent sensor is capable of providing accurate and reliable assay for OPs in complex food samples.

Abstract:Traditional methods for detecting organophosphorus pesticides (OPs) are costly and complex, and also require expensive instruments. Although many colorimetric methods have been established to detect OPs, unfortunately, these methods are based on the inhibition of the activity of natural enzymes by pesticides, which is prone to false positive signals. In addition, a variety of OPs often co-exist in the actual detection, which will interfere with each other, resulting in unsatisfactory detection results. To avoid this occasion, we developed a nanozyme sensor array based on Ag 2 O nanospheres to achieve simultaneous discrimination and efficient detection of a variety of OPs without the participation of natural enzymes. The nanozyme sensor array was constructed for detecting six OPs simultaneously, which took advantage of Ag 2 O with oxidase-like activity to catalyze the TMB, OPD and ABTS reaction system. Additionally, the colorimetric results of the sensor array for OPs detection were recorded and analyzed using a smartphone with a camera. This work not only provides a convenient and high-efficiency method to identify OPs, but it also advances food safety and environmental monitoring.

Abstract:Herein, we develop a dual-mode biosensor for photoelectrochemical and colorimetric detection of organophosphate pesticides (OPPs) based on ultrathin-FeOOH-coated MnO2 (MO@FHO) nanozyme. In this biosensor, OPPs can inhibit the alkaline phosphatase (ALP) activity and hinder the dephosphorylation of l-ascorbic acid-2-phosphate, preventing the decomposition of MO@FHO nanozyme and inducing both a photoelectrochemical (PEC) signal and the colorimetric change. The MO@FHO nanozyme not only possesses an enhanced catalase-like activity to degrade H2O2 for the generation of an improved cathodic photocurrent, but also exhibits an excellent oxidase-like activity to oxidize 3,3,5,5-tetramethylbenzidine with high catalytic efficiency. This biosensor displays a detection limit of 50 pmol/L for the PEC mode and a detection limit of 0.8 nmol/L for the colorimetric mode. Moreover, this biosensor exhibits excellent performance in complex biological matrices, and the smartphone-based visual sensing platform facilitates rapid and sensitive detection of OPPs, holding promising applications in food safety monitoring, and on-site detection.

Abstract:Given the importance of food safety, it is highly desirable to develop a convenient, low-cost, and practical sensor for organophosphorus pesticides (OPs) detection. Here, a fluorescent paper analytical device (FPAD) based on aggregation-induced emission (AIE) nanoparticles (PTDNPs-0.10) and two-dimension MnO2 nanoflakes (2D-MnNFs) was developed for instrument-free and naked-eye analysis of OPs. PTDNP-MnNFs composites were obtained through 2D-MnNFs and PTDNPs-0.10 by electrostatic interaction. and the fluorescence emission of PTDNPs-0.10 was quenched through fluorescence resonance energy transfer (FRET). When acetylcholinesterase (AChE) was present, acetylthiocholine (ATCh) was catalytically hydrolyzed into thiocholine, which reduced MnO2 of PTDNP-MnNFs into Mn2+, subsequently blocking the FRET and enhancing the fluorescence. Upon the addition of OP, AChE activity was depressed and thus the FRET between 2D-MnNFs and PTDNPs-0.10 was not affected, resulting in a slight change in fluorescence. On the basis of the variation in fluorescence intensity, highly sensitive detection of OP was readily achieved with a detection limit of 0.027 ng/mL; on the basis of the variation in brightness of FPAD, instrument-free and visual detection of OP was realized using a smartphone with a detection limit of 0.73 ng/mL. The application of FPAD has significantly simplified the detection procedure and decreased the test cost, supplying a new approach for on-site detection of OPs.

Abstract:A metal-organic-framework (MOF) fluorescent sensor is reported based on NH2-MIL-101(Fe) propelled pesticide and alkaline phosphatase (ALP) catalytic reaction. Different from previous reports, a cascade reaction system combined with MOF structural changes to generate fluorescence was employed. The rationale is that ALP can hydrolyze L-ascorbic acid 2-phosphate (AAP) into L-ascorbic acid (AA), which can reduce Fe3+ to decompose structurally NH2-MIL-101(Fe), resulting in 2-aminoterephthalic acid (NH2-BDC) with intense fluorescence. The fluorescence can be decreased to different degrees due to inhibition of organophosphate pesticides (OPPs) on the activity of ALP. By taking chlorpyrifos (CPY) as the model compound of an OPP pesticide and adding ALP and CPY into the NH2-MIL-101(Fe) framework, the resulting cascade reaction fluorescence sensors exhibit a good sensitivity for CPY and ALP sensing. The working ranges are 0.02-2 μg/L and 0.2-20 mU/mL with detection limits (LOD) of 5.31 ng/L and 0.05 mU/mL, respectively. The proposed sensor has been actually applied to satisfactory detection of CPY and ALP in food and serum samples. This fluorescence-based assay may extend the application of MOF-based biosensors.

Abstract:In this study, a nanozyme (ZIF-Co-Cys) with high oxidase-like catalytic activity was prepared, and a ratiometric fluorescent/photothermal dual-mode probe was constructed for organophosphorus pesticides (OPs) detection based on the competitive effect of ZIF-Co-Cys and the enzymatic reaction product of acid phosphatase (ACP) on o-phenylenediamine and the inhibition effect of OPs on ACP activity. Using dimethyl dichloroviny phosphate (DDVP) as the model, both the fluorescence intensity ratio and the temperature change of the probe solution exhibited an excellent correlation with OPs concentration. The detection limits were 1.64 ng/mL and 0.084 ng/mL, respectively. Additionally, the detection of DDVP residues in real samples verified the outstanding anti-interference and accuracy of the probe. This work not only provided a complementary dual-mode method for the accurate and rapid detection of OPs residues in complex samples, but also supplied a new insight into the design of a multi-mode sensing platform based on the cascade reaction of nanozyme.

Abstract:Organophosphorus pesticides were easily remained in fruits and vegetables which would be harm to the environmental safety and human health. In recent years, due to the simple preparation process, fast response and high sensitivity, the electrochemical (bio)sensors have received increasing attention, which were extensively used as the sensing platform for the detection of OPPs. The mechanisms for the determination of OPPs mainly included redox of nitrophenyl OPPs, enzyme hydrolysis and inhibition, immunosensor, aptasensor. Nowadays, the mainly explored electrode material has focused on metal-organic frameworks, metal and metal derivatives, carbon materials (carbon nanotube, graphene, g-C3N4), MXene, etc. These nanomaterials played important roles in the electrochemical (bio)sensors, which included: (a) as an electrocatalyst to promote the redox reaction, (b) as a carrier to load the enzyme or aptamer, (c) as a recognizer to identify the targets. The nanomaterials-based electrochemical (bio)sensor was a rapid, cost-effective methods to detect OPPs with high sensitivity. Besides, this review compared the analytical performance of different nanomaterials-based electrochemical (bio)sensors, and also identified the key challenges in the future. It would provide new ideas and insights to the further development and application of electrochemical (bio)sensors and the detection of pesticides in real samples.

Abstract:A novel dual-signal fluorescent sensor was developed for detecting organophosphorus pesticides (OPs). It relies on the catalytic activities of acetylcholinesterase (AChE) and choline oxidase (ChOx) to generate hydrogen peroxide (H 2 O 2 ) through the conversion of acetylcholine (ACh) to choline·H 2 O 2 then oxidizes ferrocene-modified tetraphenylethylene (TPE-Fc) to its oxidized state (TPE-Fc + ), resulting in enhanced cyan fluorescence due to aggregation. Simultaneously, ferrocene oxidation generates hydroxyl radicals (•OH), causing a decrease in orange fluorescence of glutathione-synthesized gold nanoclusters (GSH-AuNCs). The presence of OPs restricts AChE activity, reducing H 2 O 2 production. Increasing OPs concentration leads to decreased cyan fluorescence and increased orange fluorescence, enabling visual OPs detection. The sensor has a linear dynamic range of 10–2000 ng/mL with a detection limit of 2.05 ng/mL. Smartphone-based color identification and a WeChat mini program were utilized for rapid OPs analysis with successful outcomes.

Abstract:Based on the successful synthesis of mercaptomethamidophos as a substrate, a novel nanogold/mercaptomethamidophos multi-residue electrochemical biosensor was designed and fabricated by combining nanoscale effect, strong Au?S bonds as well as interaction between acetylcholinesterase (AChE) and mercaptomethamidophos, which can simultaneously detect 11 kinds of organophosphorus pesticides (OPPs) and total amount of OPPs using indirect competitive method. Electrochemical behavior of the modified electrode was characterized by differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). The AChE concentration and incubation time were optimized at 37.4 ?C to achieve the best detection effect. This biosensor exhibits excellent electrochemical properties with a wider linear range of 0.1 - 1500 ng?mL- 1, lower detection limit of 0.019 - 0.077 ng?mL- 1, better stability and repeatability, which realizes the rapid detection of total amount of OPPs, and can simultaneously detect a large class of OPPs rather than one kind of OPP. Two OPPs (trichlorfon, dichlorvos) were detected in actual samples of apple and cabbage and achieved satisfactory test results.

Abstract:Pesticides pose a serious risk to public health, even in trace amounts. The sensitive, rapid, visual, and on-site detection of pesticides in a real sample remains a challenge. Herein, we report a visual and rapid organophosphorus pesticide (OP) sensing device with excellent sensitivity, ease of application, and specificity based on a photothermal effect-based peroxidase (POD) mimetic. The ultrathin MIL-101-NH 2 (Fe) (Fe-MOF) shell-coated Au nanobipyramide (NBP) not only possesses POD-like activity, but also exhibits excellent photothermal effect under near-infrared (NIR) light. The results reveal that the excellent photo-to-thermal conversion of the Au NBPs can rapidly heat the MIL-101-NH 2 (Fe) shell, which subsequently induces a markedly enhanced POD-like activity. Using paraoxon-ethyl as model OP, the combination of the OP inhibition effect on the biocatalytic activity of acetylcholinesterase with the inhibition effect of thiocholine on the colorimetric assays of the POD-mimicking Fe-MOF leads to a significant improved selectivity for OP sensing. In particular, the NIR light irradiation increased the OP detection sensitivity by 100 times. Importantly, based on this detection strategy, a visual test strip with high sensitivity was developed and enabled real-time detection through a simple color analysis software on a smartphone. The successful detection of OP residues in fruit peel using this test strip further confirmed its application potential in real sample analysis.

Abstract:Nanostructured biosensors for the determination of organophosphorus pesticides (OPs) are fabricated by a layer-by-layer assembly technique. In the biosensors, bi-enzymes of acetylcholinesterase (AChE) and choline oxidase (ChOx) are used as biological receptors, while CdTe quantum dots (QDs) are explored as fluorescent probes for optical transduction of the enzymatic activity. Increasing amounts of OPs lead to a decrease of the enzymatic activity and thus a decrease in the production of hydrogen peroxide, which can quench the fluorescence of the CdTe QDs. The decrease of quenching rate is relative to the concentration of OPs. Using this biosensor, monitoring of three types of commonly used OPs (paraoxon, dichlorvos and parathion) at picomolar levels is realized. The linear range of detection covers six orders of magnitude (10(-12) to 10(-6) M). In addition, the biosensors exhibit a similar limit of detection and calibration curves for these pesticides, which allow them to be used for the accurate determination of total OPs and carbamate content (not the sum of anti-acetylcholinesterase toxicity as obtained by standard cholinesterase inhibition assay) of mixtures of OPs and carbamate pesticides. Finally, detection of OPs in fruits at very low levels has been achieved.
