Abstract:Single-atom nanozymes (SAzymes) have been wildly used to design high-performance biosensors for the quantification of organophosphorus pesticides (OPs) recently. However, the low yield and low metal loading of SAzymes often limit their practical use in biosensing field. Herein, a large-scale synthesis (6.60 g) of Fe-N /C SAzymes (Fe loading of 3.9 wt.%) was achieved by using carbon black as a template. This prepared Fe-N/C SAzymes exhibit efficient oxidase-like activity that could catalyze the oxidation reaction of 3,3',5,5'-tetramethylbenzidine (TMB) along with color variation. On this basis of Fe-N/C SAzymes, a colorimetric biosensor was designed for OPs detection. Then a smartphone combined with RGB analysis was used to digitize the color information. The B/ (R + G) value shows good linear detection range (1-100 nM) and low detection limit (0.4177 nM). More importantly, the large-scale synthetic approach and excellent catalytic performance of Fe-N/C SAzymes are significant for promoting the marketability of biosensors.

Ultrathin-FeOOH-coated MnO2 nanozyme with enhanced catalase-like and oxidase-like activities for photoelectrochemical and colorimetric detection of organophosphorus pesticides

Abstract:Herein, we develop a dual-mode biosensor for photoelectrochemical and colorimetric detection of organophosphate pesticides (OPPs) based on ultrathin-FeOOH-coated MnO2 (MO@FHO) nanozyme. In this biosensor, OPPs can inhibit the alkaline phosphatase (ALP) activity and hinder the dephosphorylation of l-ascorbic acid-2-phosphate, preventing the decomposition of MO@FHO nanozyme and inducing both a photoelectrochemical (PEC) signal and the colorimetric change. The MO@FHO nanozyme not only possesses an enhanced catalase-like activity to degrade H2O2 for the generation of an improved cathodic photocurrent, but also exhibits an excellent oxidase-like activity to oxidize 3,3,5,5-tetramethylbenzidine with high catalytic efficiency. This biosensor displays a detection limit of 50 pmol/L for the PEC mode and a detection limit of 0.8 nmol/L for the colorimetric mode. Moreover, this biosensor exhibits excellent performance in complex biological matrices, and the smartphone-based visual sensing platform facilitates rapid and sensitive detection of OPPs, holding promising applications in food safety monitoring, and on-site detection.

Abstract:A novel electrochemical microbial biosensor for the rapid monitoring of p-nitrophenyl-substituted organophosphates (OPs) compounds based on glass carbon electrode (GCE) modified with both ordered mesopore carbons (OMCs) and cell surface-expressed organophosphorus hydrolase (OPH) (OPH-bacteria/OMCs/GCE) was described in this paper. The genetically engineered Escherichia coli strain surface displayed mutant OPH (S5) with improved enzyme activity and favorable stability was constructed using a newly identified N-terminal of ice nucleation protein as an anchoring motif, which can be used directly without further time-consuming enzyme-extraction and purification, thereafter greatly improved the stability of the enzyme. Compared to OPH-bacteria modified GCE (OPH-bacteria/GCE), the OPH-bacteria/OMCs/GCE not only significantly enhanced the current response but also reduced the oxidation overpotential towards oxidizable p-nitrophenol (p-NP), which was the hydrolysate of p-nitrophenyl-substituted OPs. Under the optimized experimental conditions, at +0.84V (vs. SCE), the current–time curve was performed with varying OPs concentration. The current response was linear with paraoxon concentration within 0.05–25μM. Similarly, linear range of 0.05–25μM was found for parathion, and 0.08–30μM for methyl parathion. The low limits of detection were evaluated to be 9.0nM for paraoxon, 10nM for parathion and 15nM for methyl parathion (S/N=3). Thus, a highly specific, sensitive and rapid microbial biosensor was established, which holds great promise for on-site detection of trace p-nitrophenyl-substituted OPs.
A colorimetric smartphone-based platform for pesticides detection using Fe-N/C single-atom nanozyme as oxidase mimetics

Abstract:In this study, a novel highly sensitive colorimetric platform has been designed for malathion assay based on Fe-N/C SAzyme. The as-synthesized SAzyme can directly oxidize 3,3 ́,5,5 ́-tetramethylbenzidine (TMB) to generate blue colored oxidized TMB. L-ascorbic acid-2-phosphate (AA2P), a substrate of acid phosphatase (ACP), could be hydrolyzed to AA, thereafter inhibit the oxidization reaction of TMB, leading to a conspicuous blue color fading. With the addition of malathion hindered the ACP activity and limited the AA production, resulting in the recovery of the catalytic activity of single-atom nanozyme. Under optimized operational conditions, a novel colorimetric assay has been designed for malathion detection with LOD of 0.42 nM. Besides, quantification of malathion in environmental and food samples was achieved based on the proposed strategy. In addition, the successfully integrated paper/smartphone sensor provided sensitive, and rapid, reliable detection of malathion with a LOD of 1 nM.

Abstract:The relation of the physiological releases of growth hormone-releasing hormone (GHRH) and growth hormone (GH) into the circulation in various conditions was investigated using a sensitive and specific radioimmunoassay for plasma GHRH. The mean fasting plasma level of immunoreactive (IR)-GHRH in 72 normal adults was 10.3 +/- 0.5 (mean +/- SEM) pg/ml and there was no significant sex difference in the level. The concentrations of IR-GHRH in plasma from the umbilical artery and umbilical vein were 107.3 +/- 20.5 pg/ml and 33.6 +/- 3.8 pg/ml, respectively, and a marked arterio-venous gradient was observed in all 12 individuals examined. The plasma level of IR-GHRH in the maternal vein was significantly lower than that in the cord blood, but was similar to that in non-pregnant women. In normal adults, although there was no apparent fluctuation in the level of plasma IR-GHRH or of plasma GH during bed rest, a significant increase of plasma IR-GHRH was detected followed by, or synchronized with the surge of plasma GH after oral administration of L-dopa. In contrast, on L-arginine infusion, no proportional elevation of plasma IR-GHRH with increase in plasma GH was observed. During and after intravenous infusion of somatostatin, the circulating IR-GHRH level did not increase, but on stopping the infusion there was an immediate and marked rebound surge of GH. We conclude that 1) the elevated IR-GHRH in the cord blood plasma originates from the fetus and may have a primary role in enhancing secretion of GH which promotes growth in early human life, and 2) the participations of GHRH in the mechanisms of GH secretion seen after administrations of L-dopa, L-arginine and somatostatin are different.
Ultra-small CuOx/GDYO nanozyme with boosting peroxidase-like activity via electrochemical strategy: Toward applicable colorimetric detection of organophosphate pesticides

Abstract:In this paper, an ultra-small-sized CuOx/GDYO nanozyme in situ grown on ITO glass was rationally synthesized from mixed precursors of graphdiyne oxide (GDYO) and copper based infinite coordination polymer (Cu-ICP, consisting of Cu ions and two organic ligands 3,5-di-tert-butylcatechol and 1,4-bis(imidazole-1-ylmethyl)benzene) via mild and simple electrochemical strategy. On one hand, the preferential electro-reduction of Cu-ICP enabled the formation of ultra-small CuOx with Cu(I) as the main component and avoided the loss of oxygen-containing functional groups and defects on the surface of GDYO; on the other hand, GDYO can also serve as electroless reductive species to facilitate the electrochemical deposition of CuOx and turn itself to a higher oxidation state with more exposed functional groups and defects. This one-stone-two-birds electrochemical strategy empowered CuOx/GDYO nanozyme with superior peroxidase-mimicking activity and robust anchoring stability on ITO glass, thus enabled further exploration of the portable device with availability for point-of-use applications. Based on the organophosphorus pesticides (OPs) blocked acetylcholinesterase (AChE) activity, the competitive redox reaction was regulated to initiate the chromogenic reaction of 3,3',5,5'-tetramethylbenzidine (TMB) catalyzed by CuOx/GDYO peroxidase-like nanozyme, which laid out a foundation for the detection of OPs (with chlorpyrifos as an example). With a detection of limit low to 0.57 nM, the OPs residues during agricultural production can be directly monitored by the portable device we developed.

Abstract:By adjusting the coordination environment of single-atom catalysts, the enzyme-like activity can be finely tuned for highly sensitive biosensing. Herein, we demonstrated that coordinatively unsaturated cobalt-nitrogen sites doped within porous carbon (SA-CoN 3 ) could serve as highly efficient oxidase mimic. Compared with the typical planar four-coordination structure (SA-CoN 4 ), the as-obtained single-atom Co nanozymes anchored by three nitrogen atoms are found to display much higher oxidase-like catalytic efficiency. Combined theoretical and experimental analysis revealed that the coordinatively unsaturated Co sites could facilitate adsorption and activation of O 2 molecule and thus improve their oxidase-like activity. Based on the enhanced oxidase-like activity of SA-CoN 3 , a paper/smartphone sensor for organophosphorus pesticides (OPs) was successfully constructed and used to quantify glyphosate in environmental and food samples with a low detection limit of 0.66 μM. This work not only highlights the important role of coordination unsaturation of SA nanozymes for promoting oxidase-like activity, but also provides an easy and cost-effective way to conduct effective quantification of OPs in the field.

Redox interference-free bimodal paraoxon sensing enabled by an aggregation-induced emission nanozyme catalytically hydrolyzing phosphoesters specifically

Abstract:In view of the current serious situation of organophosphorus pesticides (OPs) residue contamination, developing rapid and accurate OPs sensors is a matter of urgency. Redox-nanozyme based colorimetric sensors have been widely researched and utilized in OPs residue determination, but overcoming the interference of external redox substances and the effect of single-signal modes on detection performance is still a challenge. Here we fabricated a Zr-based metal-organic framework (MOF) featuring specific phosphatase-like activity and strong aggregation-induced emission (AIE) fluorescence for redox interference-free bimodal pesticide sensing. In the MOF, the activity-tunable Zr4+ node offered high hydrolytic activity and affinity toward P-O containing substrates, and the rigid framework structure effectively enhanced the fluorescence emission of the ligand 1,1,2,2-tetra(4-carboxylphenyl)ethylene. The developed AIEzyme could efficiently catalyze the hydrolysis of paraoxon to yellow p-nitrophenol, which further reduced the intrinsic AIE fluorescence of AIEzyme through internal filtration effect. Thereby, a natural enzyme-free dual-mode colorimetric/fluorescence approach was established for paraoxon detection with no interference from redox substances, and a smartphone-assisted portable platform was further developed to enable the facile, rapid, and high-performance sensing of the pesticide in complex practical matrices.

Abstract:Given the importance of food safety, it is highly desirable to develop a convenient, low-cost, and practical sensor for organophosphorus pesticides (OPs) detection. Here, a fluorescent paper analytical device (FPAD) based on aggregation-induced emission (AIE) nanoparticles (PTDNPs-0.10) and two-dimension MnO2 nanoflakes (2D-MnNFs) was developed for instrument-free and naked-eye analysis of OPs. PTDNP-MnNFs composites were obtained through 2D-MnNFs and PTDNPs-0.10 by electrostatic interaction. and the fluorescence emission of PTDNPs-0.10 was quenched through fluorescence resonance energy transfer (FRET). When acetylcholinesterase (AChE) was present, acetylthiocholine (ATCh) was catalytically hydrolyzed into thiocholine, which reduced MnO2 of PTDNP-MnNFs into Mn2+, subsequently blocking the FRET and enhancing the fluorescence. Upon the addition of OP, AChE activity was depressed and thus the FRET between 2D-MnNFs and PTDNPs-0.10 was not affected, resulting in a slight change in fluorescence. On the basis of the variation in fluorescence intensity, highly sensitive detection of OP was readily achieved with a detection limit of 0.027 ng/mL; on the basis of the variation in brightness of FPAD, instrument-free and visual detection of OP was realized using a smartphone with a detection limit of 0.73 ng/mL. The application of FPAD has significantly simplified the detection procedure and decreased the test cost, supplying a new approach for on-site detection of OPs.

Abstract:Traditional methods for detecting organophosphorus pesticides (OPs) are costly and complex, and also require expensive instruments. Although many colorimetric methods have been established to detect OPs, unfortunately, these methods are based on the inhibition of the activity of natural enzymes by pesticides, which is prone to false positive signals. In addition, a variety of OPs often co-exist in the actual detection, which will interfere with each other, resulting in unsatisfactory detection results. To avoid this occasion, we developed a nanozyme sensor array based on Ag 2 O nanospheres to achieve simultaneous discrimination and efficient detection of a variety of OPs without the participation of natural enzymes. The nanozyme sensor array was constructed for detecting six OPs simultaneously, which took advantage of Ag 2 O with oxidase-like activity to catalyze the TMB, OPD and ABTS reaction system. Additionally, the colorimetric results of the sensor array for OPs detection were recorded and analyzed using a smartphone with a camera. This work not only provides a convenient and high-efficiency method to identify OPs, but it also advances food safety and environmental monitoring.

Photothermal effect-enhanced peroxidase-like performance for sensitive detection of organophosphorus pesticides on a visual test strip

Abstract:Pesticides pose a serious risk to public health, even in trace amounts. The sensitive, rapid, visual, and on-site detection of pesticides in a real sample remains a challenge. Herein, we report a visual and rapid organophosphorus pesticide (OP) sensing device with excellent sensitivity, ease of application, and specificity based on a photothermal effect-based peroxidase (POD) mimetic. The ultrathin MIL-101-NH 2 (Fe) (Fe-MOF) shell-coated Au nanobipyramide (NBP) not only possesses POD-like activity, but also exhibits excellent photothermal effect under near-infrared (NIR) light. The results reveal that the excellent photo-to-thermal conversion of the Au NBPs can rapidly heat the MIL-101-NH 2 (Fe) shell, which subsequently induces a markedly enhanced POD-like activity. Using paraoxon-ethyl as model OP, the combination of the OP inhibition effect on the biocatalytic activity of acetylcholinesterase with the inhibition effect of thiocholine on the colorimetric assays of the POD-mimicking Fe-MOF leads to a significant improved selectivity for OP sensing. In particular, the NIR light irradiation increased the OP detection sensitivity by 100 times. Importantly, based on this detection strategy, a visual test strip with high sensitivity was developed and enabled real-time detection through a simple color analysis software on a smartphone. The successful detection of OP residues in fruit peel using this test strip further confirmed its application potential in real sample analysis.

Abstract:A novel dual-signal fluorescent sensor was developed for detecting organophosphorus pesticides (OPs). It relies on the catalytic activities of acetylcholinesterase (AChE) and choline oxidase (ChOx) to generate hydrogen peroxide (H 2 O 2 ) through the conversion of acetylcholine (ACh) to choline·H 2 O 2 then oxidizes ferrocene-modified tetraphenylethylene (TPE-Fc) to its oxidized state (TPE-Fc + ), resulting in enhanced cyan fluorescence due to aggregation. Simultaneously, ferrocene oxidation generates hydroxyl radicals (•OH), causing a decrease in orange fluorescence of glutathione-synthesized gold nanoclusters (GSH-AuNCs). The presence of OPs restricts AChE activity, reducing H 2 O 2 production. Increasing OPs concentration leads to decreased cyan fluorescence and increased orange fluorescence, enabling visual OPs detection. The sensor has a linear dynamic range of 10–2000 ng/mL with a detection limit of 2.05 ng/mL. Smartphone-based color identification and a WeChat mini program were utilized for rapid OPs analysis with successful outcomes.

Nanozyme coating-gated multifunctional COF composite based dual-ratio enhanced dual-mode sensor for highly sensitive and reliable detection of organophosphorus pesticides in real samples

Abstract:The reliable detection of organophosphorus pesticides (OPs) in complex matrices remains an enormous challenge due to inevitable interference of sample matrices and testing factors. To address this issue, we designed a nanozyme-coated mesoporous COF with guest molecule loading, and successfully used it to construct a dual-ratio dual-mode sensor through target-regulated signal generation. The multifunctional COF-based composite (MB/COF@MnO2, MCM) featured high loading of methylene blue (MB), oxidase-like MnO2 coatings as gatekeepers, and specific recognition of thiocholine (TCh). TCh, a regulator produced from acetylcholinesterase (AChE)-catalyzed hydrolysis of acetylthiocholine, could decompose MnO2 coatings, triggering the release of abundant MB and oxidation of few o-phenylenediamine (OPD). OPs, strong inhibitors of AChE, could restrain TCh production and MnO2 decomposition, thereby controlling the release of less MB and oxidation of more OPD. This regulation boosted the dual-ratio dual-mode assay of OPs by using the released MB and oxidized OPD in the solution as testing signals, measured by both fluorescent and electrochemical methods. Experimental results demonstrated the sensitive detection of dichlorvos with LODs of 0.083 and 0.026 ng/mL via the fluorescent/electrochemical mode, respectively. This study represented a creative endeavor to develop dual-ratio dual-mode sensors for OPs detection in complex samples, offering high sensitivity, excellent selectivity, and good reliability.
A Lab-in-a-Syringe Device Integrated with a Smartphone Platform: Colorimetric and Fluorescent Dual-Mode Signals for On-Site Detection of Organophosphorus Pesticides

Abstract:Herein, a portable lab-in-a-syringe device integrated with a smartphone sensing platform was designed for rapid, visual quantitative determination of organophosphorus pesticides (OPs) via colorimetric and fluorescent signals. The device was chiefly made up of a conjugate pad labeled with cetyltrimethylammonium bromide-coated gold nanoparticles (CTAB-Au NPs) and a sensing pad modified by ratiometric probes (red-emission quantum 2 quantum dots, 2@gQDs probe), which was assembled through a disposable syringe and reusable plastic filter. In the detection system, thiocholine (Tch), the hydrolysis product of thioacetylcholine (ATch) by acetylcholinesterase (AchE), could trigger the aggregation of CTAB-Au NPs, resulting in a significant color change from red to purple. Then, CTAB-Au NPs flowed vertically upward and bound to the 2@gQDs probe on the sensing pad, reducing the fluorescence resonance energy transfer effect between CTAB-Au NPs and gQDs. Meanwhile, rQDs embedded in SiO2 NPs remained stable as internal reference fluorescence, achieving a color transition from red to green. Thus, based on the inhibition of AChE activity by OPs, a colorimetric and fluorescent dual-mode platform was constructed for on-site detection of OPs. Using glyphosate as a model, with the support of a color recognizer application (APP) on a smartphone, the ratio of red and green channel values could be utilized for accurate OP quantitative analysis ranging from 0 to 10 μM with a detection limit of 2.81 nM (recoveries, 90.8–122.4%; CV, 1.2–3.4%). Abstract:Background: Dichlorvos (DDVP), as a highly effective insecticide, is widely used in agricultural production. However, DDVP residue in foodstuffs adversely affects human health. Conventional instrumental analysis can provide highly sensitive and accurate detection of DDVP, while the need of bulky and expensive equipment limits their application in resource-poor areas and on-site detection. Therefore, the development of easily portable sensing platforms for convenient, rapid and sensitive quantification of DDVP is very essential for ensuring food safety. Result: A portable colorimetric sensing platform for rapid and sensitive quantification of DDVP is developed based on nanozyme-participated highly efficient chromogenic catalysis. The Fe-Mn bimetallic oxide (FeMnOx) nanozyme possesses excellently oxidase-like activity and can efficiently catalyze oxidation of 3, 3', 5, 5'-tetramethylbenzidine (TMB) into a blue oxide with a very low Michaelis constant (Km) of 0.0522 mM. The nanozyme-catalyzed chromogenic reaction can be mediated by DDVP via inhibiting the acetylcholinesterase (AChE) activity. Thus, trace DDVP concentration-dependent color evolution is achieved and DDVP can be sensitively detected by spectrophotometry. Furthermore, a smartphone-integrated 3D-printed miniature lightbox is fabricated as the colorimetric signal acquisition and processing device. Based on the FeMnOx nanozyme and smartphone-integrated lightbox system, the portable colorimetric sensing platform of DDVP is obtained and it has a wide linear range from 1 to 3000 ng mL-1 with a low limit of detection (LOD) of 0.267 ng mL-1 for DDVP quantification. Significance: This represents a new portable colorimetric sensing platform that can perform detection of DDVP in foodstuffs with simplicity, sensitivity, and low cost. The work not only offers an alternative to rapid and sensitive detection of DDVP, but also provides a new insight for the development of advanced sensors by the combination of nanozyme, 3D-printing and information technologies.
“Light Up” Fluorescence Visual Sensitive Detection of Organophosphorus with a Smartphone-Based Platform Utilizing a Composite Rhodamine B-Ag@Au Nanoprobe

Abstract:The present work reports a fluorescent sensor by utilizing a composite Rhodamine B-Ag@Au nanoprobe for the visual and semi-quantitative assay of organophosphorus pesticide (OP) residues via a “light up” sensing phenomenon. Core–shell structured Ag@Au NPs can directly quench the orange fluorescence of Rhodamine B (RB) at 580 nm based on Förster resonance energy transfer. Due to the stronger coordination ability of Ag@Au NPs with OPs, OP residues can displace RB from the surface of Ag@Au NP; thus, the RB fluorescence can effectively recover. Under optimum conditions, the composite fluorescent nanoprobe shows sensitive responses to OP residues in the range of 0.001–0.4 μM with a sensitive detection limit (LOD) of 7.89 nM. Moreover, combined with the smartphone color recognizer application (APP), the ratio of fluorescence intensity response is recorded on the green channel (G)/red channel (R), which has been employed for visual quantitative determination of OPs with an LOD of 16.3 nM. This proposed light up fluorescent sensor is also applied to OP residue identification in real water and agricultural samples with highly promising performance. The method reported here provides a convenient pathway for rapid and visual detection of pesticide residues utilizing organic composite nanomaterials.

Abstract:Glaucoma is the second leading cause of blindness in the world. The ultimate cause of vision loss due to glaucoma is thought to be retinal ganglion cell (RGC) apoptosis. Neuroprotection of RGC is becoming an important approach of glaucoma therapy. Several lines of evidence suggest that estrogen has neurotrophic and neuroprotective properties. In this study, we examine the role of estrogen in preventing RGC loss in DBA/2J mouse, an in vivo model of an inherited (pigmentary) glaucoma. Two‐month‐old female DBA/2J mice were anesthetized and ovariectomized with or without subcutaneous 17β‐estradiol (βE2) pellet implantation. RGC survival was evaluated from flat‐mounted whole retinas by counting retrograde‐labeled cells. The loss of nerve fibers and RGC were also evaluated in paraffin‐fixed retinal cross sections. Biochemical alterations in the retinas of DBA/2J mice in response to systemic injection of βE2 were also examined. We have made several important observations showing that: (1) βE2 treatment reduced the loss of RGC and neurofibers through inhibition of ganglion cell apoptosis, (2) βE2 activated Akt and cAMP‐responsive‐element‐binding‐protein (CREB), (3) βE2 up‐regulated thioredoxin‐1 (Trx‐1) expression, (4) βE2 reduced the increased activations of mitogen‐activated protein kinases (MAPK) and NF‐κB, (5) βE2 inhibited the increased interleukin‐18 (IL‐18) expression, and (6) treatment with tamoxifen, an estrogen receptor antagonist, blocked βE2‐mediated activation of Akt and inhibition of MAPK phosphorylation in the retinas of DBA/2J mice. These findings suggest the possible involvement of multiple biochemical events, including estrogen receptor/Akt/CREB/thioredoxin‐1, and estrogen receptor/MAPK/NF‐κB, in estrogen‐mediated retinal ganglion cell protection. © 2007 Wiley Periodicals, Inc. Abstract:Owing to their large specific surface, favorable electrical conductivity, and excellent electrocatalytic capabilities, two-dimensional transition metal dichalcogenides have received considerable attention in the field of biosensors. On the basis of these properties, we developed a portable and disposable enzyme-based biosensor for paraoxon detection using a metallic MoS2 nanosheets modified screen-printed electrode (SPE). The exfoliated ultrathin metallic MoS2 nanosheets can accelerate the electron transfer on electrode surface and contribute to the immobilization of acetylcholinesterase (AChE) via the cross-linking of glutaraldehyde. Electrodeposited gold nanoparticles (AuNPs) on SPE were used to immobilize MoS2 nanosheets through the interaction between Au atoms on AuNPs and S atoms on MoS2. Using acetylcholine as the substrate, AChE can catalyze the formation of electroactive thiocholine and further generate the redox current. In the presence of paraoxon, the activity of AChE can be inhibited, making the related electrochemical signals weaken. Under the optimized conditions, this electrochemical biosensor exhibited a favorable linear relationship with the concentration of paraoxon from 1.0 to 1000 μg L-1, with the detection limit of 0.013 μg L-1. Furthermore, this developed biosensor was successfully applied to detect paraoxon in pretreated apple and pakchoi samples, which can provide a reliable method for the rapid analysis of organophosphorus pesticides in agricultural products.

Abstract:An innovative fluorescence biosensor was successfully developed to detect organophosphorus pesticide (OPs) by utilizing smartphone technology. The assay relied on the enzymatic activity of alkaline phosphatase (ALP), which facilitated the conversion of L -ascorbic acid 2-phosphate sesquimagnesium salt hydrate (AAP) into L -ascorbic acid (AA). The AA that generated was then reacted with o -phenylenediamine (OPD) to yield a fluorescent marker identified as 3-(1,2-dihydroxyethyl)furo[3,4-b]quinoxalin-1(3H)-one (DFQ). A novel bandpass approach was specifically developed for a smartphone that was integrated with a customized portable fluorescence device to measure the fluorescence emission of DFQ. The device has a unique application that converts the fluorescence intensity into an RGB signal. In the presence of OPs, malathion was chosen as the representative of the OPs substance; the enzymatic activity of the ALP was inhibited, resulting in a decrease in fluorescence intensity, which was proportional to the concentration of malathion. Smartphones can be used to measure fluorescence emission, offering a calibration sensitivity more than 70 times higher than that of conventional spectrofluorometer. The recently developed methodology can be employed to identify malathion within the concentration range of 0.1–1 ppm, with a detection limit of 0.05 ppm. The practical applicability of the method was established using vegetable samples, and the acquired results were in good agreement with those obtained using the standard HPLC approach. This innovative method provides both portability and accuracy, while also exhibiting a notable degree of sensitivity in detecting trace amounts of OPs.
