Abstract:Understanding how aquatic species respond to extremes of DO and temperature is crucial for determining how they will be affected by climate change, which is predicted to increasingly expose them to levels beyond their optima. In this study we used novel animal-borne DO, temperature and depth sensors to determine the effect of extremes of DO and temperature on the vertical habitat use of Atlantic salmon Salmo salar in aquaculture cages. Salmon showed a preference for temperatures around 16.5 to 17.5 °C, however, selection of preferred temperatures was trumped by active avoidance of low DO (<35% saturation) at the bottom of the cage. In addition to low DO, salmon also avoided warm surface waters (>20.1 °C), which led to a considerable contraction in the available vertical habitat. Despite their avoidance behavior, fish spent a large amount of time in waters with suboptimal DO (<60% saturation). These results show that vertical habitat contraction could likely be a significant consequence of climate change if the reduction in DO outpaces the increase in hypoxia tolerance through local adaptation. They furthermore highlight that site-specific environmental conditions and stock-specific tolerance thresholds may need to be considered when determining stocking densities.

Abstract:Salmons raised in aquaculture farms around the world are increasingly subjected to sub-optimal environmental conditions, such as high water temperatures during summer seasons. Aerobic scope increases and lipid metabolism changes are known plasticity responses of fish for a better acclimation to high water temperature. The present study aimed at investigating the effect of high water temperature on the regulation of fatty acid metabolism in juvenile Atlantic salmon fed different dietary ARA/EPA ratios (arachidonic acid, 20:4n-6/ eicosapentaenoic acid, 20:5n-3), with particular focus on apparent in vivo enzyme activities and gene expression of lipid metabolism pathways. Three experimental diets were formulated to be identical, except for the ratio EPA/ARA, and fed to triplicate groups of Atlantic salmon (Salmo salar) kept either at 10°C or 20°C. Results showed that fatty acid metabolic utilisation, and likely also their dietary requirements for optimal performance, can be affected by changes in their relative levels and by environmental temperature in Atlantic salmon. Thus, the increase in temperature, independently from dietary treatment, had a significant effect on the β-oxidation of a fatty acid including EPA, as observed by the apparent in vivo enzyme activity and mRNA expression of pparα -transcription factor in lipid metabolism, including β-oxidation genes- and cpt1 -key enzyme responsible for the movement of LC-PUFA from the cytosol into the mitochondria for β-oxidation-, were both increased at the higher water temperature. An interesting interaction was observed in the transcription and in vivo enzyme activity of Δ5fad-time-limiting enzyme in the biosynthesis pathway of EPA and ARA. Such, at lower temperature, the highest mRNA expression and enzyme activity was recorded in fish with limited supply of dietary EPA, whereas at higher temperature these were recorded in fish with limited ARA supply. In consideration that fish at higher water temperature recorded a significantly increased feed intake, these results clearly suggested that at high, sub-optimal water temperature, fish metabolism attempted to increment its overall ARA status -the most bioactive LC-PUFA participating in the inflammatory response- by modulating the metabolic fate of dietary ARA (expressed as % of net intake), reducing its β-oxidation and favouring synthesis and deposition. This correlates also with results from other recent studies showing that both immune- and stress- responses in fish are up regulated in fish held at high temperatures. This is a novel and fundamental information that warrants industry and scientific attention, in consideration of the imminent increase in water temperatures, continuous expansion of aquaculture operations, resources utilisation in aquafeed and much needed seasonal/adaptive nutritional strategies.

Abstract:The frequency of extreme thermal events in temperate freshwater systems is expected to increase alongside global surface temperature. The Miramichi River, located in eastern Canada, is a prominent Atlantic salmon (<i>Salmo salar</i>) river where water temperatures can exceed the proposed upper thermal limit for the species (~27°C). Current legislation closes the river to recreational angling when water temperatures exceed 20°C for two consecutive nights. We aimed to examine how natural thermal variation, representative of extreme high thermal events, affected the thermal tolerance and physiology of wild, juvenile Atlantic salmon. We acclimated fish to four thermal cycles, characteristic of real-world thermal conditions while varying daily thermal minima (16°C, 18°C, 20°C or 22°C) and diel thermal fluctuation (e.g. Δ5°C-Δ9°C). In each cycling condition, we assessed the role that thermal minima played on the acute thermal tolerance (critical thermal maximum, (CTMax)), physiological (e.g. heat shock protein 70 (HSP70), ubiquitin) and energetic (e.g. hepatic glycogen, blood glucose and lactate) status of juvenile Atlantic salmon throughout repeated thermal cycles. Exposure to 16-21°C significantly increased CTMax (+0.9°C) compared to a stable acclimation temperature (16°C), as did exposure to diel thermal fluctuations of 18-27°C, 20-27°C and 22-27°C, yet repeated exposure provided no further increases in acute thermal tolerance. In comparison to the reference condition (16-21°C), consecutive days of high temperature cycling with different thermal minima resulted in significant increases in HSP70 and ubiquitin, a significant decrease in liver glycogen, and no significant cumulative effect on either blood glucose or lactate. However, comparison between thermally taxed treatments suggested the diel thermal minima had little influence on the physiological or energetic response of juvenile salmon, despite the variable thermal cycling condition. Our results suggest that relatively cooler night temperatures in the summer months may play a limited role in mitigating physiological stress throughout warm diel cycle events.

Abstract:Ten (1–10) irritant and mild co-carcinogenic diterpene esters were isolated from the latex of Euphorbia cauducifolia L. using bioassay-guided countercurrent distribution and other chromatographic techniques. The isolated compounds were characterized on the basis of spectroscopic results and mass measurements. As an outcome, the ingenane-type esters were established with the following structures: 3-O-angeloyl-17-O-palmatoylingenol (1), 3-O-palmatoyl-5-O-angeloylingenol (2), 5-O-angeloyl-17-O-palmatoylingenol (3), 3-O-angeloyl-5-O-palmatoylingenol (4), 17-O-(2Z,4E,6Z)-2,4,6-tetradecatrienoyl-20-O-palmatoylingenol (5), 5-O-angeloyl-17-O-benzoylingenol (6), 5-O-angeloyl-17,20-diacetoxyingenol (7), 3-O-angeloyl-17-O-benzoyl-20-acetoxyingenol (8), 3-acetoxy-5-O-angeloyl-17-O-benzoylingenol (9), and 5-O-angeloyl-3,17,20-triacetoxyingenol (10). Their biological screening revealed that they are moderate irritants, and low to moderate tumor promoters compared to TPA, but hardly showed any solitary carcinogenic activity. The isolated esters represent new compounds and were not reported before from any source.

Abstract:ABSTRACT Poleward winters commonly expose animals, including fish, to frigid temperatures and low food availability. Fishes that remain active over winter must therefore balance trade-offs between conserving energy and maintaining physiological performance in the cold, yet the extent and underlying mechanisms of these trade-offs are not well understood. We investigated the metabolic plasticity of brook char (Salvelinus fontinalis), a temperate salmonid, from the biochemical to whole-animal level in response to cold and food deprivation. Acute cooling (1°C day−1) from 14°C to 2°C had no effect on food consumption but reduced activity by 77%. We then assessed metabolic performance and demand over 90 days with exposure to warm (8°C) or cold winter (2°C) temperatures while fish were fed or starved. Resting metabolic rate (RMR) decreased substantially during initial cooling from 8°C to 2°C (Q10=4.2–4.5) but brook char exhibited remarkable thermal compensation during acclimation (Q10=1.4–1.6). Conversely, RMR was substantially lower (40–48%) in starved fish, conserving energy. Thus, the absolute magnitude of thermal plasticity may be masked or modified under food restriction. This reduction in RMR was associated with atrophy and decreases in in vivo protein synthesis rates, primarily in non-essential tissues. Remarkably, food deprivation had no effect on maximum oxygen uptake rates and thus aerobic capacity, supporting the notion that metabolic capacity can be decoupled from RMR in certain contexts. Overall, our study highlights the multi-faceted energetic flexibility of Salvelinus spp. that likely contributes to their success in harsh and variable environments and may be emblematic of winter-active fishes more broadly.

Abstract:Whether the residual aerobic scope decreases while digesting and is related to bodily functions are context‐dependent and species‐specific. There was no common pattern in postprandial changes in the different physiological functions, but digestion generally showed a stronger effect on bodily function in fish species from slow‐flow habitats. A precise prediction of how the freshwater fish population responds to global climate change needs to incorporate the data of postprandial fishes. Limited aerobic scope (AS) during digestion might be the main constraint on the performance of bodily functions in water‐breathing animals. Thus, investigating the postprandial changes in various physiological functions and determining the existence of a shared common pattern because of possible dependence on residual AS during digestion in freshwater fish species are very important in conservation physiology. All species from slow‐flow habitats showed impaired swimming speed while digesting, whereas all species from fast‐flow habitats showed strong swimming performance, which was unchanged while digesting. Only two species from slow‐flow habitats showed impaired heat tolerance during digestion, suggesting that whether oxygen limitation is involved in the heat tolerance process is species‐specific. Three species from slow‐ or intermediate‐flow habitats showed impaired hypoxia tolerance during digestion because feeding metabolism cannot cease completely under hypoxia. Overall, there was no common pattern in postprandial changes in different physiological functions because: (1) the digestion process was suppressed under oxygen‐limiting conditions, (2) the residual AS decreased during digestion, and (3) performance was related to residual AS, while digestion was context‐dependent and species‐specific. However, digestion generally showed a stronger effect on bodily functions in species from slow‐flow habitats, whereas it showed no impairment in fishes from fast‐flow habitats. Nevertheless, the postprandial change in physiological functions varies with habitat, possibly due to divergent selective pressure on such functions. More importantly, the present study suggests that a precise prediction of how freshwater fish populations will respond to global climate change needs to incorporate data from postprandial fishes.

Summer Is Coming! Tackling Ocean Warming in Atlantic Salmon Cage Farming

Abstract:Atlantic salmon (Salmo salar) cage farming has traditionally been located at higher latitudes where cold seawater temperatures favor this practice. However, these regions can be impacted by ocean warming and heat waves that push seawater temperature beyond the thermo-tolerance limits of this species. As more mass mortality events are reported every year due to abnormal sea temperatures, the Atlantic salmon cage aquaculture industry acknowledges the need to adapt to a changing ocean. This paper reviews adult Atlantic salmon thermal tolerance limits, as well as the deleterious eco-physiological consequences of heat stress, with emphasis on how it negatively affects sea cage aquaculture production cycles. Biotechnological solutions targeting the phenotypic plasticity of Atlantic salmon and its genetic diversity, particularly that of its southernmost populations at the limit of its natural zoogeographic distribution, are discussed. Some of these solutions include selective breeding programs, which may play a key role in this quest for a more thermo-tolerant strain of Atlantic salmon that may help the cage aquaculture industry to adapt to climate uncertainties more rapidly, without compromising profitability. Omics technologies and precision breeding, along with cryopreservation breakthroughs, are also part of the available toolbox that includes other solutions that can allow cage farmers to continue to produce Atlantic salmon in the warmer waters of the oceans of tomorrow.

Abstract:Declining body size in fishes and other aquatic ectotherms associated with anthropogenic climate warming has significant implications for future fisheries yields, stock assessments and aquatic ecosystem stability. One proposed mechanism seeking to explain such body-size reductions, known as the gill oxygen limitation (GOL) hypothesis, has recently been used to model future impacts of climate warming on fisheries but has not been robustly empirically tested. We used brook trout ( Salvelinus fontinalis ), a fast-growing, cold-water salmonid species of broad economic, conservation and ecological value, to examine the GOL hypothesis in a long-term experiment quantifying effects of temperature on growth, resting metabolic rate (RMR), maximum metabolic rate (MMR) and gill surface area (GSA). Despite significantly reduced growth and body size at an elevated temperature, allometric slopes of GSA were not significantly different than 1.0 and were above those for RMR and MMR at both temperature treatments (15°C and 20°C), contrary to GOL expectations. We also found that the effect of temperature on RMR was time-dependent, contradicting the prediction that heightened temperatures increase metabolic rates and reinforcing the importance of longer-term exposures (e.g. >6 months) to fully understand the influence of acclimation on temperature–metabolic rate relationships. Our results indicate that although oxygen limitation may be important in some aspects of temperature–body size relationships and constraints on metabolic supply may contribute to reduced growth in some cases, it is unlikely that GOL is a universal mechanism explaining temperature–body size relationships in aquatic ectotherms. We suggest future research focus on alternative mechanisms underlying temperature–body size relationships, and that projections of climate change impacts on fisheries yields using models based on GOL assumptions be interpreted with caution.

Abstract:Abstract Temperature in many natural aquatic environments follows a diel cycle, but to date, we know little on how diel thermal cycles affect fish biology. The current study investigates the growth, development and physiological performance of wild Atlantic salmon collected from the Miramichi and Restigouche rivers (NB, Canada). Fish were collected as parr and acclimated to either 16–21 or 19–24°C diel thermal cycles throughout the parr and smolt life stages. Both Miramichi and Restigouche Atlantic salmon parr grew at similar rates during 16–21 or 19–24°C acclimations. However, as smolts, the growth rates of the Miramichi (−8% body mass day−1) and Restigouche (−38% body mass day−1) fish were significantly slower at 19–24°C, and were in fact negative, indicating loss of mass in this group. Acclimation to 19–24°C also increased Atlantic salmon CTmax. Our findings suggest that both life stage and river origin impact Atlantic salmon growth and performance in the thermal range used herein. These findings provide evidence for local adaptation of Atlantic salmon, increased vulnerability to warming temperatures, and highlight the differential impacts of these ecologically relevant diel thermal cycles on the juvenile life stages in this species.

Temperature-dependent plasticity mediates heart morphology and thermal performance of cardiac function in juvenile Atlantic salmon (Salmo salar)

Abstract:In many fishes, upper thermal tolerance is thought to be limited in part by the heart's ability to meet increased oxygen demands during periods of high temperature. Temperature-dependent plasticity within the cardiovascular system may help fish cope with the thermal stress imposed by increasing water temperatures. In this study, we examined plasticity in heart morphology and function in juvenile Atlantic salmon (Salmo salar) reared under control (+0°C) or elevated (+4°C) temperatures. Using non-invasive Doppler echocardiography, we measured the effect of acute warming on maximum heart rate, stroke distance and derived cardiac output. A 4°C increase in average developmental temperature resulted in a >5°C increase in the Arrhenius breakpoint temperature for maximum heart rate and enabled the hearts of these fish to continue beating rhythmically to temperatures approximately 2°C higher than for control fish. However, these differences in thermal performance were not associated with plasticity in maximum cardiovascular capacity, as peak measures of heart rate, stroke distance and derived cardiac output did not differ between temperature treatments. Histological analysis of the heart revealed that while ventricular roundness and relative ventricle size did not differ between treatments, the proportion of compact myocardium in the ventricular wall was significantly greater in fish raised at elevated temperatures. Our findings contribute to the growing understanding of how the thermal environment can affect phenotypes later in life and identify a morphological strategy that may help fishes cope with acute thermal stress.
Limited Capacity for Faster Digestion in Larval Coral Reef Fish at an Elevated Temperature

Abstract:The prevalence of extreme, short-term temperature spikes in coastal regions during summer months is predicted to increase with ongoing climate change. In tropical systems, these changes are predicted to increase the metabolic demand of coral reef fish larvae while also altering the plankton communities upon which the larvae feed during their pelagic phase. The consequences of these predictions remain speculative in the absence of empirical data on the interactive effects of warm temperatures on the metabolism, postprandial processes and growth responses of coral reef fish larvae. Here, we tested the effect of increased temperature on the metabolism, postprandial performance and fine-scale growth patterns of a coral reef fish (Amphiprion percula) in the latter half of its ~11-d larval phase. First, we measured the length and weight of fed versus fasted larvae (N = 340; mean body mass 4.1±0.05 mg) across fine temporal scales at a typical current summer temperature (28.5°C) and a temperature that is likely be encountered during warm summer periods later this century (31.5°C). Second, we measured routine metabolic rate (Mo2 routine) and the energetics of the postprandial processes (i.e., digestion, absorption and assimilation of a meal; termed specific dynamic action (SDA)) at both temperatures. Larvae fed voraciously when provided with food for a 12-hour period and displayed a temperature-independent increase in mass of 40.1% (28.5°C) and 42.6% (31.5°C), which was largely associated with the mass of prey in the gut. A subsequent 12-h fasting period revealed that the larvae had grown 21.2±4.8% (28.5°C) and 22.8±8.8% (31.5°C) in mass and 10.3±2.0% (28.5°C) and 7.8±2.6% (31.5°C) in length compared with pre-feeding values (no significant temperature effect). Mo2 routine was 55±16% higher at 31.5°C and peak Mo2 during the postprandial period was 28±11% higher at 31.5°C, yet elevated temperature had no significant effect on SDA (0.51±0.06 J at 28.5°C vs. 0.53±0.07 J at 31.5°C), SDA duration (6.0±0.6 h vs. 6.5±0.5 h), or the percent of total meal energy used for SDA (SDA coefficient: 10.1±1.3% vs. 13.0±1.7%). Our findings of higher Mo2 routine but similar SDA coefficient at high temperature provide the first empirical evidence that coral reef fish larvae may have to secure more food to attain similar growth rates during warm summer periods, and perhaps with chronically warmer conditions associated with climate change.

Interactive Effect of Thermal and Hypoxia on Largemouth Bass (micropterus Salmoides) Gill and Liver: Aggravation of Oxidative Stress, Inhibition of Immunity and Promotion of Cell Apoptosis

Abstract:High temperatures and low oxygen in aquatic environments, such as intensive aquaculture or in natural watersheds, inevitably cause stress in fish. Fish are exposed to high temperatures during the summer, which exacerbates hypoxia. Hypoxia (1.2 +/- 0.2 mg/L) under 20 degrees C (20 HG) and 26 degrees C (26 HG) was simulated to induce stress in largemouth bass (Micropterus salmoides). Related enzymes and genes involved in antioxidant, immune, and apoptotic responses were selected to explore the interactive effects of temperature and hypoxia on largemouth bass. The results showed that malondialdehyde (MDA) levels in plasma, gill, and liver increased in the 26 HG (p < 0.05). Liver superoxide dismutase (SOD) activity increased in the 26 HG. Peak SOD (SOD1, SOD2, SOD3a, and SOD3b), CAT, and GSH-Px mRNA levels in the gill and liver were observed at 12-24 h of stress. The levels of gill and liver total antioxidant capacity, catalase (CAT), glutathione peroxidase (GSH-Px) activities and other enzyme activities and genes in the 26 HG were higher than those in the 20 HG (p < 0.05). The gill and liver add phosphatase and alkaline phosphatase activities increased with time in the 26 HG (p < 0.05), while gill and liver lysozyme activities in the 26 HG were lower than those in the 20 HG (p < 0.05). Tumor necrosis factor-alpha mRNA level was upregulated in the gill and downregulated in the liver at 24 h in the 26 HG. Interleukin (IL)-1 beta and IL-8 mRNA levels were upregulated in the gill and liver in the 26 HG at 24 h, whereas IL-15 mRNA level was downregulated in the 26 HG at 12 h. Transforming growth factor-beta 1 mRNA level was upregulated in the gill in the 20 HG at 24 h, but downregulated in gill and liver in the 26 HG at 24 h. Similarly, IL-10, Hepcidin-1, and Hepcidin-2 showed lower expression levels in the 26 HG. Gill and liver caspase-3 activities were higher in the 26 HG (p < 0.05), and gill caspase-3 activity was higher than that in the liver. The mRNA levels of proapoptotic genes (caspase-3, caspase-8, and caspase-9) were higher in the 26 HG. The present study demonstrates the interactive effects of temperature and hypoxia on stress in largemouth bass gill and liver. These results will be helpful to understand the mechanisms of stress induced by temperature and hypoxia in fish and provide a theoretical basis for aquaculture management.

Abstract:Atlantic salmon ( ) when exposed to heat stress have reduced voluntary feeding and exhibit changes in digesta consistency. This study was performed to determine what bacterial species occur and the overall bacterial abundance in the gut microbiome in heat stressed Atlantic salmon. For this Atlantic salmon in seawater tanks at 15°C were fed for 2 to 4 weeks. Tank temperatures were increased to 19°C until voluntary feeding abated at which point tank temperatures were cooled to 15°C for 4 weeks. At the end of each temperature phase the fish were stripped of feces and microbiome profiles were determined using 16S rRNA V1-V3 metabarcoding. The tank experiment was repeated three times in successive years. Vibrionaceae comprised most reads after the warm phase completed. The prominent levels of Vibrionaceae were accompanied by a large predominance of cast (sloughed intestinal mucosa) containing fecal samples. qPCR estimated Vibrionaceae cell populations increased 1.9-3.4 log units/g after the warm phase but and slightly decreased by 0.3-1.1 log units/g after the 15°C recovery phase. The results indicated heat stress induced inappetence corresponds to increased cast production accompanied by predominance of Vibrionaceae. Vibrionaceae colonizing the Atlantic salmon gut should be the focus of studies on the microbiology of thermally induced inappetence and dysbiosis in Atlantic salmon.

Abstract:A progressive inability of the cardiorespiratory system to maintain systemic oxygen supply at elevated temperatures has been suggested to reduce aerobic scope and the upper thermal limit of aquatic ectotherms. However, few studies have directly investigated the dependence of thermal limits on oxygen transport capacity. By manipulating oxygen availability (via environmental hyperoxia) and blood oxygen carrying capacity (via experimentally-induced anemia) in European perch (Perca fluviatilis, Linneaus), we investigated the effects of oxygen transport capacity on aerobic scope and the critical thermal maximum (CTmax). Hyperoxia resulted in a two-fold increase in aerobic scope at the control temperature of 23°C, but this did not translate to an elevated CTmax in comparison with control fish (34.6±0.1°C vs. 34.0±0.5°C, respectively). Anemia (∼43% reduction in haemoglobin concentration) did not cause a reduction in aerobic scope nor CTmax (33.8±0.3°C) compared with control fish. Additionally, oxygen consumption rates of anemic perch during thermal ramping increased in a similar exponential manner as in control fish, highlighting that perch have an impressive capacity to compensate for a substantial reduction in blood oxygen carrying capacity. Taken together, these results indicate that oxygen limitation is not a universal mechanism determining the CTmax of fishes.

Abstract:Ocean temperatures continue to rise annually due to the ever-growing consequences of global climate change. These temperature changes can have an impact on the immunological robustness of cultured fish, especially cold-water species such as Atlantic salmon. The salmon farming industry already loses hundreds of millions of dollars each year to infectious and non-infectious diseases. One particularly important and WOAH reportable disease is infectious salmon anemia caused by the orthomyxovirus ISAv. Considering the changing environment, it is necessary to find ways to mitigate the effect of diseases on the industry. For this study, 20 Atlantic salmon families were housed in each of 38 different tanks at the AVC, with half of the fish being kept at 10 °C and half being kept at 20 °C. Donor Atlantic salmon IP- injected with a highly virulent ISAv isolate (HPR4; TCID50 of 1 × 105/mL) were added to each tank as the source of co-habitation infection. Both temperatures were sampled at onset of mortality in co-habited fish and at resolution of mortality. Family background and temperature significantly impacted ISAv load, as assessed by qPCR, time to mortality and overall mortality. Mortality was more acute at 20 °C, but overall mortality was higher at 10 °C. Based on percent mortality calculated over the course of the study, different families demonstrated different levels of survival. The three families that demonstrated the highest percent mortality, and the three families with the lowest percent mortality were then assessed for their antiviral responses using relative gene expression. Genes significantly upregulated between the unexposed fish and ISAv exposed fish included mx1, il4/13a, il12rb2, and trim25, and these were further impacted by temperature. Understanding how ISAv resistance is impacted by temperature can help identify seasonal risks of ISAv outbreaks as well as ideal responses to be targeted through immunopotentiation.

Abstract:Atlantic salmon ( Salmo salar L.) undergo great alterations in physiology and gut microbiota composition throughout their life stages. This study assessed gut health and microbiota in out-of-season smolts before and after seawater transfer and modulatory effects of functional feed ingredients under commercial Arctic conditions. The fish were fed two series of diets, one without (Ref diet) and one with a mixture of functional ingredients (Test diet). Both diets varied in nutrient composition as required according to the developmental stage of the fish. For fish in freshwater, the mixture of functional ingredients contained nucleotides and immune stimulants, in seawater nucleotides, DHA, and EPA. Samples were collected four weeks before and four weeks after seawater transfer. Regardless of diet, seawater transfer (seawater compared to freshwater) significantly suppressed fish growth rate, condition factor, plasma nutrient levels, digesta bile acid concentrations, expression of genes related to gut immune functions (i.e., cytokines and T-cell markers), and increased intestinal microbial richness and diversity. Seawater transfer also reduced the symptoms of pyloric caeca lipid malabsorption with a corresponding decreasing effect on perilipin-2 ( plin2 ) expression. In the gut microbiota of fish in freshwater, the unclassified Ruminococcaceae family dominated strongly, accounting for about 85% of the total abundance, whereas in seawater the genera Lactobacillus and Photobacterium were the dominant taxa, accounting for about 90% of the total abundance. Multivariate association analysis showed that relative abundance of certain lactic acid bacteria (LAB) correlated positively with expression of important immune genes. Regarding the effects of functional feed ingredients, Test-fed fish in FW showed higher plasma cholesterol levels, and reduced symptoms of lipid malabsorption, suggesting that the mixture of nucleotides and immune stimulants enhanced digestive and absorptive capacity. However, the inclusion of nucleotides, DHA, and EPA affected the SW-fish only marginally. In conclusion, the outcomes of the present study highlight the great reductions in growth and impact on gut health biomarkers after seawater transfer, in this case observed under commercial Arctic conditions, and the promotion of metabolic capacity of Atlantic salmon fed functional ingredients in freshwater, which may shape future best-practices in salmon industry.

Abstract:Changes in growth, survival, maturation and health in Atlantic salmon post-smolts were observed under moderate and more extreme heat-wave conditions for the west coast of Scotland. A Scottish Atlantic salmon breeding programme population of 518 salmon of age 13-14 months, was observed for 4 weeks in summer, in tanks with ambient water temperature or ∼ 4 degrees and ∼ 8 degrees above ambient temperature. Data were recorded for the fish before and after the challenge. All fish were genotyped using a custom SNP (single nucleotide polymorphism) array (8,978 SNP genotypes after quality control). Temperature-dependant genotype-by-environment interactions and the potential for selective breeding to improve resilience were investigated. Statistical analyses revealed significant differences between thermal environments for body weight, average daily weight gain, and survival, while higher temperature induced earlier maturation, and an increase of gill health scores. QPCR (quantitative polymerase chain reaction) analyses revealed the presence of , a pathogen typically more prevalent in warmer climates. Using genomic relationships and animal mixed models, body weight and average daily weight gain provided moderate heritabilities, while between-tank genetic correlations were close to 1, indicating no significant re-ranking of genotypes between the different thermal environments. These findings suggest that even short-term exposure to heat stress may be sufficient to negatively impact survival and gill health, and induce earlier maturation. However, observations took place within a commercial farm where having replicates for each thermal environment was not possible, hence further experiments with larger populations, exposed to more prolonged heat stress are needed.
