Abstract:In the circular economy, reusing agricultural residues, treated biowaste, and sewage sludges—commonly referred to as soil conditioners—in agriculture is essential for converting waste into valuable resources. However, these materials can also contribute to the spread of antimicrobial-resistant pathogens in treated soils. In this study, we analyzed different soil conditioners categorized into five groups: compost from source-separated biowaste and green waste, agro-industrial digestate, digestate from anaerobic digestion of source-separated biowaste, compost from biowaste digestate, and sludges from wastewater treatment plants. Under Italian law, only the first two categories are approved for agricultural use, despite Regulation 1009/2019/EU allowing the use of digestate from anaerobic digestion of source-separated biowaste in CE-marked fertilizers. We examined the bacterial community and associated resistome of each sample using metagenomic approaches. Additionally, we detected and isolated various pathogens to provide a comprehensive understanding of the potential risks associated with sludge application in agriculture. The compost samples exhibited higher bacterial diversity and a greater abundance of potentially pathogenic bacteria compared to other samples, except for wastewater treatment plant sludges, which had the highest frequency of Salmonella isolation and resistome diversity. Our findings suggest integrating omics and cultivation-dependent methods to accurately assess the biological risks of using sludge in agriculture.

Abstract:Topicality. Spreading the philosophy of sustainable development, where activities preserve natural and social resources for use by future generations without harming the business economy, as well as significant increases in the price of chemical fertilizers due to rising gas prices require ways to reduce the chemical burden on agriculture. The trend of abandoning intensive technologies will be exacerbated by economic leverage - European Union regulations are increasingly banning agrochemicals and introducing a "carbon tax". Therefore, the use of sewage sludge as fertilizer for agricultural production is becoming an urgent issue. Therefore, the use of sewage sludge as fertilizer for agricultural production on the basis of economic efficiency is becoming an urgent issue.Aim and tasks. The aim of the article is to consider the economic and technological principles aimed at improving the bioavailability of nutrients and reducing the risk of using biowaste before entering the soil, in the context of the idea of sustainable development.Research results. Sewage sludge (SS) application in agriculture has a beneficial effect on soil organic matter content, sorption capacity and an overall improvement in physical properties. Conversion of SS to a soil amendment can be performed by a broad spectrum of methods, which greatly differ by substrate/amendment composition, treatment time, and physicochemical conditions. Sanitary and epidemiological safety is essential, which is why bio-wastes require processing according to selected technologies that aim to improve the bioavailability of nutrients and reduce hazards before entering the soil. This review provides a more complete overview of the present status of the methods for SS disinfection. The review is focused on i) environmental and legislative aspects of SS application in agriculture; ii) risk factors related to the abundance of bacterial, viral, protozoan and other pathogens in SS and methods of SS hygienization by various physical and chemical treatments; iii) risks of soil pollution with biologically active compounds (e.g., antibiotic resistance genes and other emerging contaminants). For the life cycle assessment, an environmental performance and pathogen risk was considered. The results of such consideration have direct impact on the nature of the applied economic and technological measures. Conclusions and perspectives in this field were formulated, using 102 references, including 49 citations dated by the last five years.Conclusion. Numerous technological approaches on SS treatment have their particular advantages, although disinfection efficiency remains unsatisfactory. Legislative requirements are still based on less resistant indicator organisms. Further comprehensive research on SS treatment should be focused on combination of different physical (especially, thermal) and chemical processes, which would convert SS into a qualitative fertilizer with safe microbiological characteristics. The decrease in energy consumption during drying and the reduction of the management costs of these residues can be relevant economic gains. In general, the application of drying to remove water from sewage sludge should be a balance between energy costs in the process and the management costs without drying.

Abstract:Disposal of organic sludge and liquid agricultural wastes is a universal problem. Their production cannot be halted and as steps are taken to maintain or improve the quality of rivers and lakes it grows in quanti ty. The Commission's early awareness of the need for action to prepare for substantial growth in the Community's' sludge disposal problem led to the setting up of the COST 68 project to coordinate and guide European research and development work with particular emphasis on recycling sludge to agricultural land. Two years ago the field of research activi ties was extended to liquid agricultural wastes. This Symposium is the latest opportunity to provide a comprehensive review of the results of the project, to define current trends in practice and to establish by discussion the priorities for research over the next few years. The development of instrumentation and of analytical techniques during the period has extended our knowledge of the organic and inorganic constituents of sewage sludge and agricultural wastes and enabled us more readily to identify and measure the risks to which our general environment may be exposed when disposing of it. This evolution of understanding is a continuing process and an essential guide to the modification of disposal practices to achieve safer and more efficient operations. However, it is important to take a broad view of the application of research findings in the light of the considerable contrast in conditions in different parts of the world.

Abstract:This paper presents the current sewage sludge legislation in Europe and expected developments regarding the coming directives on the application of the “End-of-waste” criteria and on fertilizers. Discussion on sludge production and processing is also included. The Directive 86/278 has regulated the use in agriculture of residual sludge from domestic and urban wastewater. After 1986, this directive was transposed in the different member state legislation and currently the national limit values on heavy metals, some organic micropollutants and pathogens are placed in a rather wide range. This seems the inevitable consequence of different attitudes towards sludge management practices in the member states. The discussion by the European Joint Research Center (JRC) in Seville regarding application of end-of-waste criteria for compost and digestate has produced a final document (IPTS 2014) where sludge was excluded from the organic wastes admitted for producing an end-of-waste compost. Sludge processing in Europe seems addressed to different goals: sludge minimization, full stabilization and hygienization by thermal hydrolysis processes before anaerobic digestion, and on-site incineration by fluidized bed furnace. Thermophilic anaerobic digestion was applied with success on the Prague WWTP with a preliminary lysimeter centrifugation. Coming techniques, like wet oxidation and pyrolysis, are applied only on very few plants.

Abstract:Wastewater treatment plants (WWTP) are facilities where municipal wastewater undergoes treatment so that its organic load and its pathogenic potential are minimized. Sewage sludge is a by-product of this process and when properly treated is preferentially called "biosolids". These treatments may include some or most of the following: thickening, dewatering, drying, digestion, composting, liming. Nowadays it is almost impossible to landfill biosolids, which however can well be used as crop fertilizers. Continuous or superfluous biosolids fertilization may negatively affect non-target organisms such as soil macro-organisms or even plants. These effects can be depicted through bioassays on terrestrial animals and plants. It has been shown that earthworms have been affected to various degrees on the following endpoints: pollutants' bioaccumulation, viability, reproduction, avoidance behavior, burrowing behavior. Collembola have been affected on viability, reproduction, avoidance behavior. Other terrestrial organisms such as nematodes and diplopods have also shown adverse health effects. Phytotoxicity have been caused by some biosolids regimes as measured through the following endpoints: seed germination, root length, shoot length, shoot biomass, root biomass, chlorophyll content, antioxidant enzyme activity. Very limited statistical correlations between pollutant concentrations and toxicity endpoints have been established such as between juvenile mortality (earthworms) and As or Ba concentration in the biosolids, between juvenile mortality (collembola) and Cd or S concentration in the biosolids, or between phytotoxicity and some extractable metals in leachates or aquatic extracts from the biosolids; more correlations between physicochemical characteristics and toxicity endpoints have been found such as between phytotoxicity and ammonium N in biosolids or their liquid extracts, or between phytotoxicity and salinity. An inverse correlation between earthworm/collembola mortality and stable organic matter has also been found. Basing the appropriateness of biosolids only on chemical analyses for pollutants is not cost-effective. To enable risk characterization and subsequent risk mitigation it is important to apply a battery of bioassays on soil macro-organisms and on plants, utilizing a combination of endpoints and established protocols. Through combined analytical quantification and toxicity testing, safe use of biosolids in agriculture can be achieved.

Abstract:The increasing scarcity of natural resources makes the linear production model unsustainable, highlighting the need for more sustainable practices under the umbrella of circular economic principles. Sewage sludge emerges as a promising solution to provide soil organic matter (SOM) and nutrients. This meta-analysis evaluated the impacts of three levels of sludge application (low, medium, and high) on organic matter (OM), nitrogen (N), phosphorus (P), copper (Cu), and zinc (Zn) contents, considering different areas and experimental conditions worldwide. The analysis included 37 studies and 355 comparisons, after screening 7625 records, following the 2020 PRISMA protocol. The effects of sludge concentrations, continents, and types of experiment (field or greenhouse) were evaluated. Sewage sludge application significantly increased OM, N, Cu, and Zn levels, mainly at medium and high rates. The largest effects were observed in greenhouses, suggesting variation by location and environment. Moderators explained a part of the variation in the results, but the residual heterogeneity test revealed that there is still unexplained variability.

Identification of Compounds that Selectively Stabilize Specific G-Quadruplex Structures by Using a Thioflavin T-Displacement Assay as a Tool.

Abstract:Small molecules are used in the G-quadruplex (G4) research field in vivo and in vitro, and there are increasing demands for ligands that selectively stabilize different G4 structures. Thioflavin T (ThT) emits an enhanced fluorescence signal when binding to G4 structures. Herein, we show that ThT can be competitively displaced by the binding of small molecules to G4 structures and develop a ThT-displacement high-throughput screening assay to find novel and selective G4-binding compounds. We screened approximately 28 000 compounds by using three different G4 structures and identified eight novel G4 binders. Analysis of the structural conformation and stability of the G4 structures in presence of these compounds demonstrated that the four compounds enhance the thermal stabilization of the structures without affecting their structural conformation. In addition, all four compounds also increased the G4-structure block of DNA synthesis by Taq DNA polymerase. Also, two of these compounds showed selectivity between certain Schizosaccharomyces pombe G4 structures, thus suggesting that these compounds or their analogues can be used as selective tools for G4 DNA studies.

Abstract:Purpose Urbanization and industrialization in China has resulted in a dramatic increase in the volume of wastewater and sewage sludge produced from wastewater treatment plants. Problems associated with sewage sludge have attracted increasing attention from the public and urban planners. How to manage sludge in an economically and environmentally acceptable manner is one of the critical issues that modern societies are facing. Methods Sludge treatment systems consist of thickening, dewatering, and several different alternative main treatments (anaerobic digestion, aerobic digestion, drying, composting, and incineration). Agricultural application, landfill, and incineration are the principal disposal methods for sewage sludge in China. However, sewage sludge disposal in the future should focus on resource recovery, reducing environmental impacts and saving economic costs. Results The reuse of biosolids in all scenarios can be environmentally beneficial and cost-effective. Anaerobic digestion followed by land application is the preferable options due to low economic and energy costs and material reuse. Conclusion It is necessary to formulate a standard suitable for the utilization of sewage sludge in China.
Production and Destination of Sewage Sludge in the Piemonte Region (Italy): The Results of a Survey for a Future Sustainable Management

Abstract:The management of sewage sludge originated from municipal wastewater treatment plants (WWTPs) is an urgent issue. In 2019, the local authority of the Piemonte region started a survey with the aim of collecting recent data concerning wastewater and sludge management in the WWTPs located in its own territory. The survey's results revealed that 60% of the sludge (51,000 t, as dry substance, d.s.) produced by the local WWTPs was recovered or disposed of outside of the region, and a similar amount of sludge was recovered in agriculture directly or after composting. The increase in the costs to accommodate sewage sludge in recovery or disposal plants, followed to a recent Italian Sentence (27958/2017), and the more and more stringent requirements fixed by lots of European countries for the application of sludge in agriculture, are pushing the Piemonte region authority to re-organize its own network for sludge management, with solutions based onto proximity and diversification. Whether the provisions of the current German legislation are applied in the future also in Italy, approx. 90% of sewage sludge produced into the Piemonte region should be incinerated, with a subsequent step of phosphorous recovery. The new regional plan, according to the Regional Address Deed, should consider a diversification of sludge treatment and recovery practices. On this basis, a need for new plants for around 40,000 t d.s./y could be planned.

Abstract:Sewage sludge is a by-product of the sewage treatment plants. Because of its richness in nutrients and for several environmental and economic reasons, this waste by-product is widely used as a fertilizer for agricultural purposes under specific conditions. This practice might be hazardous since this waste includes many known and unknown non-biodegradable and harmful pollutants, especially emerging organic contaminants (EOCs), as well as cultivable and non-cultivable pathogens. The present work is aiming at (1) providing information on the nature of sludge in terms of persistent pollutants and cultivable and non-cultivable microbiota generated by the currently implemented treatment processes, (2) analyzing the consequences of the agricultural valorization on ecosystem biodiversity and soil fertility, and (3) addressing and discussing the economic, ethical, and environmental benefits or damage of this type of valorization. These issues need full consideration by policy makers regarding the agricultural use of this waste by-product in terms of irrigation with treated domestic wastewater or sludge land spreading as soil fertilizer and plant growth promoter.

Abstract:Sewage sludge production increases in Europe proportionally to the number of municipalities that put wastewater treatment plants into service. Recycling to soil is among the main sludge outlets, since amending with sludge meets requirements of efficient recycling of resources while providing organic matter for poor or degraded soils. As raw sludge contains substances potentially harmful for environmental and human health, it must be stabilized before application to the soil. We tested three kinds of anaerobically digested sludge, differing by the post-treatments they had undergone (dewatering, composting or thermal drying), for their effects on soil microbial activity, microarthropod density and biological soil quality quantified as QBS indicator. Our experimental treatments were set on plots located in a burnt Mediterranean forest and consisted of spreading sludge on the soil surface to achieve a dose of 6 Mg o.m.ha−1. Although amendment with any of the three kinds of sludge increased soil organic matter and nitrogen, thermally dried sludge caused a transitory increase of the soil microbial metabolic coefficient that we considered indicative of soil disturbance. One year after soil amendment, all sludge effects on the soil microbial activity had disappeared. When working with microarthropods’ density and biodiversity, no sludge improved soil biological quality, fresh digested sludge being the most detrimental, since it caused the decline of the most sensitive taxa (oribatid mites) and increased the density of soil forms resistant to disturbance and pollution (astigmatic mites and symphypleon collembolans). Sludge effects on soil microarthropods were still evident three years after soil amendment, which suggests that soil invertebrate diversity is less resilient to soil disturbance than global microbial metabolism.

Abstract:The great interest of modern societies in the reuse of wastes opens up new horizons in the field of wastewater, as well. In particular, the treated sludge resulting from a Wastewater Treatment Plant (WWTP) is dealed with a new perspective in the context of circular economy. The aim of this study is the characterization of its complex matrix, and the evaluation for reuse. Biosolids (BS) collected from four urban WWTPs in the Western region of Greece i.e. Agrinio (AG), Amaliada (AM), Aegio (AE) and Itea (IT). Analytical and spectroscopical methods namely TGA, ICP-OES, Fluorescence, SEM/EDS, XRD, FT-IR and NMR were the means that served this purpose. SEM along with XRD proved the amorphous nature of BS. The dominant metals detected in the samples are: Fe, Zn, Mn, with concentrations which meet the guidelines included in 86/278/EEC Directive. The inorganic load is of great importance, along with their humic acid content, adding value at the samples as fertilizers. BS appeared to be rich in organic matter with long aliphatic chains and numerous functional groups, as capturedin FT-IR spectra. The applied methods form an analytical protocol of the BS mapping, highlighting its potential as a material to be utilized in agriculture.

Abstract:The fertilizer framework in the European Union has recently been reviewed by EU Regulation 1009/2019 that excluded sewage sludge from the list of the possible constituents of organic fertilizers relying on their origin, not on their quality. This paper aimed to carry out a complete characterization of sewage sludge obtained from a pharmaceutical manufacturing process (PDSS) to demonstrate that sewage sludge obtained from a standardized and controlled manufacturing process can be safely recycled as organic fertilizer. The agronomic and environmental characteristics of the PDSS product were analyzed and compared to other organic fertilizers. Its fertilizing potential was also evaluated through plant growth trials. PDSS was characterized by a high concentration of total N (6.6% w/w), which was all present in organic form. PDSS also showed a low concentration of heavy metals, an absence of pathogens and low concentrations of organic contaminants. Plant growth trials showed that the PDSS was able to improve lettuce and carrot growth (+25 and +46% of dry weight compared to the unfertilized control), as well as their physiological status. Considering all the results, the exclusion of sewage sludge relying only on its origin and not on its quality appears to conflict with the principles of the circular bioeconomy.

Abstract:Sewage sludge handling is becoming a concern in Europe due to its increasing amount and the presence of contaminants, such as heavy metals and pharmaceutical and personal care products (PPCPs). Currently, over 70% of sludge in Europe is treated thermally by incineration or used as fertilizer in agriculture. New thermochemical methods are under development and are expected to be implemented in the near future. This paper considers the life cycle environmental impacts of the following five alternatives for sludge handling, taking into account the presence of heavy metals and PPCPs: i) agricultural application of anaerobically digested sludge; ii) agricultural application of composted sludge; iii) incineration; iv) pyrolysis; and v) wet air oxidation. The results suggest that anaerobic digestion with recovery of nutrients and electricity has the lowest environmental impacts in 11 out of 18 categories considered. For the mean to maximum resource recovery, composting is the worst alternative, followed by pyrolysis with lower recovery rates. Agricultural application of anaerobically digested sludge has the highest freshwater ecotoxicity due to heavy metals, unless their concentration is in the lowest range, as found in some European sewage sludge applied on land. Therefore, stricter control of heavy metals in the sludge is needed for this option to limit freshwater ecotoxicity to the levels comparable with the thermal processes. The results also indicate that PPCPs have a negligible contribution to freshwater ecotoxicity when compared to heavy metals in the anaerobically digested sludge. Since thermal processes are currently drawing attention due to their potential benefits, the findings of this work suggest that their adoption is environmentally beneficial only if high resource recovery rates can be achieved.

Abstract:The increasing amount of sewage sludge, which is mainly related to the improvement of wastewater treatment technologies and the expansion of sewage systems, has become a serious problem for sewage sludge management. Sewage sludge is a valuable source of nutrients and organic matter. The use of sewage sludge for agricultural purposes presents a sustainable method of disposal in line with circular economy strategies, which are aimed at waste prevention, the effective use of resources and energy, and a nutrient closed-loop system. However, sewage sludge can contain heavy metals and toxic organic compounds which pose threats to the environment. Therefore, sewage sludge management is still a controversial topic. Taking into account disruptions in the fertiliser market caused by the pandemic and wars, it is necessary to consider local waste with fertilising potential as an alternative raw material to reduce fertiliser import dependency and ensure food security. This review collected data confirming the agronomic value of sewage sludge, including long-term field studies and data on the presence of pollutants along with possible ways to reduce them. Particular attention is given to the technologies available for fertiliser production from sewage sludge which are operating at full scale. Ecotoxicological assessment of waste-derived organic fertilizers and long-term monitoring of fertilized soils using a multi-matrix and multi-species approach

Abstract:The present study investigates the ecotoxicity of 7 biofertilizers, including biowaste-derived organic matrices. Real-field tests were conducted to assess the impacts of soil fertilization with sewage sludge digestate from high-solid thermophilic anaerobic digestion (HSTAD) compared to those obtained on non-amended and urea-fertilized soils. The physical-chemical and ecotoxic impact of HSTAD digestate on soil was monitored for 12 months, at 5 time points and 2 soil depths, on a maize field divided in 3 portions (non-treated, fertilized with urea, amended with digestate). The chemical and physical characteristics of the soil were previously analyzed for 3 years to provide a long-term outlook of the impacts of biofertilizer application. Seven bioindicators were utilized for direct (on whole soil) and indirect (on soil elutriates) ecotoxicological tests on fertilizers and amended soils, including plant seeds (Lepidium sativum, Sorghum saccharatum, and Sinapsis alba), the aquatic organism Daphnia magna, the alga Raphidocelis subcapitata, the luminescent bacterium Aliivibrio fischeri, and the Nematode Caenorhabditis elegans. No serious negative effects on soil fertilized with HSTAD digestate were evidenced. Conversely, bioassays rather showed positive effects, encouraging the utilization of HSTAD digestate in agriculture, considering the proper concentrations of use. The obtained data were interpolated and a test battery integrated index was generated, confirming the absence of ecotoxicological risk for the soils amended with the applied fertilizers. The long-term evolution of the physical-chemical soil characteristics (including the concentrations of potential contaminants) was similar for both HSTAD digestate and urea application as well as for non-fertilized soil, indicating no negative effects due to digestate application on land. On the contrary, digestate application improved the content of stabilized organic matter and nutrients in soil. This study proposes a more correct approach to ecotoxicity assessment of fertilized soils for biofertilizer evaluation and demonstrates the long-term safe application of HSTAD digestate on agricultural soil.

Abstract:Safe disposal of municipal sewage sludge is a challenging global environmental concern. The aim of this study was to assess the response of soil microbial functional diversity to the accumulation of municipal sewage sludge during landfill storage. Soil samples of a municipal sewage sludge (SS) and from a sewage sludge landfill that was 3 m from a SS landfill (SS3) were analyzed relative to an undisturbed reference soil. Biolog EcoPlatesTM were inoculated with a soil suspension, and the Average Well Color Development (AWCD), Richness (R) and Shannon-Weaver index (H) were calculated to interpret the results. The fungi isolated from the sewage sludge were identified using comparative rDNA sequencing of the LSU D2 region. The MicroSEQ® ID software was used to assess the raw sequence files, perform sequence matching to the MicroSEQ® ID-validated reference database and create Neighbor-Joining trees. Moreover, the genera of fungi isolated from the soil were identified using microscopic methods. Municipal sewage sludge can serve as a habitat for plant pathogens and as a source of pathogen strains for biotechnological applications.

Abstract:The simultaneous requirement to manage resources and wastes in more rational way has meant that many communities worldwide have begun to search for long-term alternative solutions. Reuse and recovery of biosolids is considered to be a constant solution of circular sustainability, as waste disposal without further reuse background like fertilizer is no longer an alternative to be promoted. There have been developed many treatment methods over the years for the stabilization and sanitization of biosolids. However, the literature concludes that none of them is fully integrated by meeting all the basic criteria. Each method has its Achilles heel, and the appropriateness of the method lies in what is the goal each time. There are conventional methods with positive reciprocity in terms of sustainability, reuse indicators and technological maturity, but have high risk of microorganisms' reappearance. New advanced sustainable technologies, such as cold plasma, need to be further studied to apply on a large scale. The reuse of biosolids as construction materials is also discussed in the context of circular economy. Biosolids reuse and management legislation frame need to be revised, as a directive adopted 30 years ago does not fully meet communities' current needs.
