Abstract:Objective: Andaliman (Zanthoxylum acanthopodium) is a potent medicinal plant in Asia. This present study aimed to reveal the effectivity of Andaliman fruit extract in alleviating hyperglycemia, sensory and motoric balance disorders, histopathology of the cerebellum, and tissue oxidative stress in diabetic mice induced by alloxan. Materials and Methods: Diabetes induction was performed by intraperitoneally injecting alloxan monohydrate [200 mg/kg body weight (BW)]. Subsequently, the mice were treated daily with an ethanolic extract of Andaliman fruit (0, 150, 300, 450 mg/kg BW per oral) for 28 days, followed by measurements of blood glucose, paw sensitivity, motoric balance, histopathology of the cerebellum, and malondialdehyde (MDA) levels. Moreover, the phytochemical constituents of the extract were elucidated by liquid chromatography. Results: Higher doses of Andaliman fruit extract could significantly attenuate the elevation of random and fasting blood glucose (p < 0.05) and improve paw sensitivity responses (p < 0.05) and motoric balances (p < 0.05) in diabetic mice. Moreover, Andaliman fruit extract could significantly attenuate the degeneration of cerebellar Purkinje cells (p < 0.05) and suppress MDA levels in the blood (p < 0.05) while blunting the MDA in the brain tissue (p < 0.05). Phytochemical screening revealed 39 compounds in the Andaliman extract belonging to the groups of alkaloids (26 compounds), flavonoids (12 compounds), and terpenoids (1 compound). Conclusion: The ethanolic extract of Andaliman fruit is capable of ameliorating diabetic neuropathy, motor balance disorders, and Purkinje cell degeneration while also reducing oxidative stress in the peripheral system. Hence, Andaliman extract is a promising candidate for formulation as an herbal remedy against the detrimental outcomes of diabetes mellitus.

Abstract:Following the observation of increased trough whole blood cyclosporin A (CyA) concentrations and reduced renal function in a patient with recalcitrant generalized pustular psoriasis treated with a combination of CyA and etretinate. the effect of vitamin A analogues on human microsomal cytochrome P450‐dependent CyA metabolism was investigated in vitro. In addition, the effect of terbinafine, a new allylamine antifungal agent, was also tested. Etretinate, its major metabolite acitretin. and isotretinoin, each at a single concentration of 100 μ, inhibited total hepatic microsomal CyA metabolism to a similar extent (33–45%, compared with control values). The generation of total primary and total secondary CyA metabolites was also inhibited to a similar extent by each of the retinoids. Conversely, terbinafine was without significant effect on CyA metabolism in vitro. The results, which suggest that inhibition of hepatic CyA metabolism by retinoids may contribute to increased circulating CyA concentrations, are discussed in relation to other potential drug interactions, and to the use of etretinate in reducing the CyA administered dose.

Abstract:Objective: Dementia is a common aging-related neurodegenerative disease in the elderly worldwide. Alterations in neurogenesis and angiogenesis factors have been linked to cognitive impairment in neurological disorders. However, synthetic drugs to improve memory disorders have uncomfortable side effects. The purpose of this study is to explore the neuroprotective potential of the fruit ethanol extract of andaliman (Zanthoxylum acanthopodium DC) [Andaliman fruit ethanol extract (AEE)] on brain-derived neurotrophic factor (BDNF), vascular endothelial growth factor (VEGF), and spatial memory in rat models of aging. Materials and methods: This study had an experimental design with AEE. The 4 groups were treated as follows: N (normal), M (served as positive control), P1 (AEE 150 mg/kg bw), and P2 (AEE 300 mg/kg BW) for 8 weeks. Aged model rats (M, P1, and P2) were obtained by inducing D-galactose (150 mg/kg bw). BDNF and VEGF expression were determined by RT-PCR, and spatial memory was assessed using the test of the Moris Water Maze (MWM). The Kruskal-Wallis and Mann-Whitney tests were used to assess the statistical analysis. Results: AEE had a tendency to increase BDNF in P2 compared to the normal group (1.98 versus 1). VEGF expression increased in P1 and P2 compared to the normal group (1.14 and 1.29 versus 1). AEE at a dose of 300 mg/kg bw significantly improved spatial memory (p = 0.026). Conclusion: For eight weeks, AEE at a dose of 300 mg/kg bw considerably increased the potential to enhance VEGF and BDNF expression as well as spatial memory.

Abstract:Background: Diabetes mellitus is a metabolic disorder associated with relative or absolute insulin deficiency or resistance, characterized by hyperglycemia. Modern prescriptions such as pioglitazone have better therapeutic potential, but its side effects and financial burden for developing countries have motivated the researchers to find alternative natural drugs to compete hyperglycemia in patients with diabetes. The present study was conducted to explore the therapeutic potential of selected medicinal plants for the treatment of diabetes as an alternative to allopathic medicines. Method: In present study, hydroalcoholic extracts of Curcuma longa, Lavandula stoechas, Aegle marmelos, and Glycyrrhiza glabra and their polyherbal preparation (PHP) as compound drug were investigated for their antihyperglycemic potential in alloxan-induced diabetic mice. The study subjects (mice) were divided into different groups as normal control, diabetic control, pioglitazone treated (standard drug), test groups (plant extract treated 50, 100, and 150 mg/kg body weight), and PHP-treated group. Blood glucose concentration of all the study animals was determined by Glucose strip test. Qualitative phytochemical analysis of all the plant extracts was also performed following standard methods. Result: It was investigated that treatment of alloxan-induced diabetic mice with hydroalcoholic extracts of studied medicinal plants showed significant (P < .05) effects on fasting blood glucose levels (from baseline to normal range) in a manner comparable to that of the reference drug, pioglitazone (1 mg/kg body weight intraperitoneal). The tested plant extracts significantly (P < .05) reduced the glucose concentration in blood of diabetes-induced mice in a dose-dependent manner. Conclusion: It could be concluded that studied medicinal plants have antihyperglycemic activity. The study findings favor the use of traditional herbal medicinal practices for the management of diabetes that might due to the presence of bioactive phytoconstituents in plants. However, larger studies are required to identify, isolate, and characterize the bioactive phytoconstituents responsible for antihyperglycemic activity of studied medicinal plants.

Abstract:Abstract Diabetes mellitus (DM), an endocrine syndrome characterized by high blood glucose levels due to abrogated insulin activity. The existing treatments for DM have side effects and varying degrees of efficacy. Therefore, it is paramount that novel approaches be developed to enhance the management of DM. Therapeutic plants have been accredited as having comparatively high efficacy with fewer adverse effects. The current study aims to elucidate the phytochemical profile, anti-hyperlipidemic, and anti-diabetic effects of methanolic extract D. salicifolia (leaves) in Alloxan-induced diabetic mice. Alloxan was injected intraperitoneally (150 mg kg-1, b.w), to induced diabetes in mice. The mice were divided into three groups (n=10). Group 1 (normal control) received normal food and purified water, Group II (diabetic control) received regular feed and clean water and group III (diabetic treated) received a methanolic extract of the plant (300 mg kg-1) for 28 days with a typical diet and clean water throughout the experiment. Blood samples were collected to checked serum glucose and concentration of LDL, TC, TG. The extract demonstrated significant antihyperglycemic activity (P<0.05), whereas improvements in mice's body weight and lipid profiles were observed after treatment with the extract. This study establishes that the extract has high efficacy with comparatively less toxicity that can be used for DM management.

Abstract:Abstract Background In diabetic animals, there is a significant increase in plasma glucose, serum total cholesterol, triglyceride, and low-density lipoprotein levels, and decreased body weight, liver and muscle glycogen, and high-density lipoprotein. Effective treatment of diabetes mellitus is not yet known, even though the management of diabetes mellitus is considered a global concern. Plants and herbs have played an important role in the healthcare of many societies throughout history. Today’s researchers are investigating the potential for using these nonpharmaceutical approaches to treat and control diabetes, either in conjunction with standard treatments or as an alternative to them. Herbal formulations are favored because to lower cost and fewer side effects compared to other methods for alleviating diabetes and its consequences. In ethnomedicinal practices, different parts of Mangifera indica are used to treatment of diabetes. The present investigation was undertaken to evaluate the antidiabetic activity of an ethanolic extract of Mangifera indica and mangiferin in alloxan-induced diabetic rats. This experiment was conducted in a set of two with four groups of animals namely control (Tc), treatment alloxan (Ta), treatment extract (Tae), and treatment mangiferin (Tam). To develop diabetes, Wistar rats treated with 150 mg/kg b.w. of alloxan monohydrate were injected intraperitoneally. Tae and Tam’s groups received a freshly prepared single dose of extract and mangiferin in distilled water via the oral route. All experimental groups received laboratory pallet feed diet and drinking water ad libitum. Diabetic rats were treated for 21 days with an ethanolic extract of mango peel and pure mangiferin orally daily at rates of 200 mg/kg b.w. and 20 mg/kg b.w. Results An alloxan-induced diabetic rat treated with mango peel extract and mangiferin significantly improved the overhead impact due to diabetes. There was a significant ( p < 0.05) body weight loss in the alloxan-induced diabetic rats (Ta), whereas animals given mango peel extract and mangiferin showed a significant increase in body weight from 2 weeks onwards in comparison with control. Alloxan-induced rats (Ta) group have higher blood glucose levels and are significantly different ( p < 0.01) from the control group. Mango peel extract and mangiferin significantly reduced the levels of fasting glucose after 21 days of treatment in comparison with diabetic animals. Mango peel extract and mangiferin influence the glycogen synthesis pathway in diabetes groups by increasing glycogen levels in muscle and liver. mango peel extract and mangiferin were found to have a nonsignificant impact on plasma cholesterol and HDL levels compared with the control group. Mango peel extract was found to have a significant difference ( p < 0.05) in LDL levels compared with the control group. Mangiferin was found to have a significant difference ( p < 0.05) in triglyceride and VLDL levels when compared with the control group. Histopathological examination of the pancreas in rats with type I diabetes caused by alloxan found that therapy with an ethanolic extract of mango peel and mangiferin restored beta cell function as well as rejuvenation of Islets of Langerhans. Conclusions Mango peel extract and mangiferin have antidiabetic, glycogenesis, and hypolipidemic properties when administered to alloxan-induced diabetic rats. Graphical abstract

Abstract:Andaliman (Zanthoxylum acanthopodium DC.) fruit is a spice plant widely used in North Sumatra. The chemical content in the Andaliman plant has a cardioprotective effect, with antioxidant properties that inhibit oxidative stress and free radicals. SOD (superoxide dismutase), BNP (Brain Natriuretic Peptide), and cTnT (troponin T) are measured as markers of heart damage, and histopathology is to see heart damage. Quercetin administration was used as a comparison. The hydroalcoholic extract's phytochemical content and chemical elements were analyzed using LC-HRMS and GC-MS. The findings showed that the hydroalcohol extract of Andaliman fruits affected the blood levels of SOD, BNP, and cTnT in the blood of doxorubicin-induced rats. SOD levels increased, and BNP decreased; the 300 mg/kg BW group was not significantly different from the 50 mg/kg BW quercetin group. cTnT levels also decreased; the 150 mg/kg BW and 300 mg/kg BW groups were not significantly different, and both were better than the 50 mg/kg BW quercetin group. EAF with 150 mg/kg BW and 300 mg/kg BW can also repair damage to rat heart tissue caused by doxorubicin. Andaliman fruit extract has cardioprotective effects and anti-free radical activity due to its content and potential to be developed.

Effect of the Zinc Oxide Nanoparticles and Thiamine for the Management of Diabetes in Alloxan-Induced Mice: a Stereological and Biochemical Study

Abstract:This research was carried out to evaluate the antidiabetic effects of zinc oxide nanoparticles (ZnO NPs) and thiamine following experimental diabetes. Fifty-six 6-week-old female mice were used and divided into seven groups of eight animals. Diabetes was induced in fasted mice by using intraperitoneal (IP) injection of alloxan (180 mg/kg). Groups included (I) non-diabetic control, (II) thiamine (30 mg/l, IP), (III) alloxan-induced diabetic mice, (IV) diabetes + ZnO NPs (0.1 mg/kg IP), (V) diabetes + ZnO NPs (0.5 mg/kg IP), (VI) diabetes + ZnO NPs (0.1 mg/kg IP) + thiamine (30 mg/l, IP), and (VII) diabetes + ZnO NPs (0.5 mg/kg IP) + thiamine (30 mg/l, IP). Coincident with pancreas recovery, in diabetic treated mice (groups IV to VII), the mean islet volume, islets per square micrometer, and volume density of the pancreas had increased than in alloxan-induced diabetic mice. ZnO NPs and thiamine induced a decreasing blood glucose, lower serum triglyceride (TG), LDL, and total cholesterol (TC) levels in alloxan-induced diabetic mice treated with ZnO NPs and thiamine, simultaneously increasing HDL as well. In conclusion, ZnO NPs and thiamine are potent antidiabetic factors, and that, these compound supplementation possesses hypoglycemic properties and have effect on serum lipid parameters in diabetes mice.

Abstract:The combination of polymorphic restriction-enzyme sites in the 3' region of the beta-globin gene cluster shows very little variation in southern-African Bantu-speaking black and Kalahari !Kung San populations. The sites of the 5' region, on the other hand, show marked variation, and two common haplotypes are present--the "Negro" type (- - - - +) and the "San" type (- + - - +)--in frequencies of .404 and .106, respectively, in the Bantu-speakers and .262 and .405, respectively, in the San. Twenty of 23 beta s-associated haplotypes in southern-African Bantu-speaking black subjects were the same as that found commonly in the Central African Republic (CAR)--i.e., the "Bantu" type--a finding providing the first convincing biological evidence for the common ancestry of geographically widely separated speakers of languages belonging to the Bantu family. The (-alpha) haplotype has a frequency of .21 in the Venda, .07 in both the Sotho-Tswana and the Nguni, and .06 among the !Kung San. These data are interpreted in the light of Plasmodium falciparum malaria selection and population movements in the African subcontinent.

Abstract:Zanthoxylum armatum DC, a medicinal plant species in India, is an exceptionally nutritious vegetable with a variety of potential uses in treating rheumatism, venomous bites, and microbial infections. In the present study, we aimed to investigated the antidiabetic and antioxidant effects of aqueous extracts of Z. armatum Stem Bark (Za-Aq) in High Fat Diet (HFD) low dose streptozotocin (STZ)-induced type 2 diabetic wistar albino rats and the objectives the were achieved through examine of blood glucose, Oral Glucose Tolerance Test (OGTT), Insulin Tolerance Test (ITT), and the levels of lipid peroxides and antioxidant enzymes, such as superoxide dismutase, glutathione peroxidase and reduced glutathione in the liver tissues of control and experimental groups. Diabetic rats were treated with 200, 400 and 800 mg/kg BW Za-Aq for 45 days and the antidiabetic effects of the extract were evaluated by measuring changes in biochemical parameters in the serum and liver tissue. Oral administration of Za-Aq to diabetic rats for 45 days significantly reduced the levels of blood glucose, OGTT, ITT and lipid peroxidation, but increased the activities of antioxidant enzymes, like superoxide dismutase, reduced glutathione and glutathione peroxidase. The progression of diabetes was significantly reduced after Za-Aq treatment. In conclusion, Za-Aq exerts protective effects against HFD-STZ-induced diabetes. The Za-Aq supplementation is useful in controlling the blood glucose level and is beneficial in preventing diabetic complications from lipid peroxidation and antioxidant systems in experimental diabetic rats; therefore, it could be useful for prevention or early treatment of diabetes mellitus active constituents may be isolated from the extract for evaluation in future clinical studies. To best of our knowledge there has been no previous comprehensive study to evaluate antidiabetic activity of stem bark from Z. armatum DC and present study adds the protective effect of Zanthoxylum armatum DC stem bark as effective antidiabetic and antioxidant agent.

Abstract:Objective: The aim of the present study is to investigate the antihyperglycemic and antioxidative properties of Dillenia indica fruits.Methods: Aqueous fruit extract and methanolic fruit extract (MFE) were prepared, and preliminary phytochemical screening was carried out. Diabetic mice were prepared with alloxan (150 mg/kg) body weight (b.w.). Antihyperglycemic study of short duration was carried out with doses (150–550) mg/kg b.w. of MFE in diabetic mice. Antioxidant enzymes (superoxide dismutase, catalase, and glutathione reductase) activity assays and histopathological analysis were done in heart tissue of mice.Results: Preliminary phytochemical screening showed that the phytoconstituents were strongly present in the MFE and therefore was considered for further studies. From the antihyperglycemic study, it was found that 350 mg/kg b.w. dose was the most effective in reduction of blood glucose level. A significant increase in the activities of the antioxidant enzymes was observed in the MFE-treated group. From the histopathological studies, it was observed that detrimental effects of oxidative stress were attenuated in the treated group.Conclusion: Concluding the studies, it could be ascertained that D. indica fruits were found to be quite effective in proving its potential against hyperglycemia and oxidative stress, and therefore, the fruits could be considered to be of therapeutic value in diabetes.

Abstract:The intestinal digestive enzyme alpha-glucosidase plays a vital role in the carbohydrate digestion. One anti-diabetic therapeutic approach reduces the postprandial glucose level by the inhibition of the alpha-glucosidase enzyme. The present study aimed to investigate the anti-diabetic activity of extract of Ailanthus altissima bark, in vitro. Three different plant extracts were formulated by using distilled water, methanol, and a mixture of methanol and water (70:30) to compare the best effect of the extract, while distilled water and metformin were used as negative and positive controls. Four different concentrations of plant extract in all solvents were used to find percentage inhibition of alpha-glucosidase. Maximum percentage inhibition was examined against 1.5 mL of all solvent extracts but aqueous methanolic plant extract showed maximum73% inhibition that was comparable to standard drug inhibition (80%). One-way ANOVA was followed for statistical analysis to find out the P-value based on which most significant and insignificant were declared. Aqueous methanolic plant extract showed the most significant results because the value of p 0.05. The assay results suggest that the presence of bioactive compounds are responsible for the versatile medicinal properties against diabetes, the aqueous methanolic extract exhibit a dose-dependent increase in inhibitory effect on alpha-glucosidase enzyme up to 73%. The current study proves aqueous methanolic extract of A. altissima bark has anti-diabetic activity in vitro and is a strong candidate for postprandial control of blood glucose level.

Abstract:Diabetes and its complications represent a real major public health problem in the world because of its high rates of morbidity and mortality. Chronic hyperglycemia, oxidative stress, dyslipidemia and inflammation play a major role in the pathophysiology of diabetes and its vascular complications. The objective of this study was to evaluate the effect of aqueous extract of S. zenkeri on chronic hyperglycemia and its complications in a streptozotocin-induced diabetic Wistar rat model. The barks of S. zenkeri were washed, dried and crushed; the powder was dissolved in distilled water (1:10 weight/volume) then macerated and the filtrate obtained was dried in an oven. Subsequently, after quantification of the bioactive compounds (total polyphenols, flavonoids and alkaloids) present in the extract, an in vivo study was conducted in an animal model of streptozotocin-induced hyperglycemia. For this fact, the rats were divided into four groups of five rats as folow: a normoglycaemia group (NC), an untreated hyperglycaemia group (PC), two hyperglycaemia groups including a test group receiving by esophageal gavage, the aqueous extract of the bark of S zenkerii (AESZ) at a dose of 300 mg/kg body weight and a control group receiving metformin at a dose of 20 mg/kg body weight. During the treatment which lasted 21 days, the weights have been taken every two days and the blood sugar levels every week. At the end of the treatment, the rats were killed under light chloroform anesthesia; the plasma, hemolysate, serum and liver homogenate prepared were used to assay the biochemical parameters of oxidative stress (catalase, MDA), lipid profile (Triglycerides, total cholesterol and HDL-cholesterol) and immunological (CRP and NFS). It emerged that the extract limited weight loss and caused a reduction in blood sugar of -26.59% after 21 days of treatment; the extract caused an increase in the activity of erythrocyte catalase and a reduction in the concentration of hepatic MDA, as well as a very marked reduction in inflammatory cells and CRP. The extract also caused a reduction in dyslipidemiawhich was materialized by a reduction in CRR, AC, AIP and an increase in CPI. These results suggest that this extract contains bioactive compounds capable of reducing chronic hyperglycemia while preventing its complications, thus justifying its traditional use in the management of diabetes.
Hypoglycaemic effect of total alkaloids extracted from Sambucus wightiana Wall. ex Wight & Arn. in streptozotocin-nicotinamide induced diabetic rats

Abstract:Diabetes mellitus is the most prevalent metabolic disease, and almost 1200 plants are reported with hypoglycaemic potential. Some of these plants are scientifically proven for the hypoglycaemic and antidiabetic effects. This study was designed to assess the hypoglycaemic effect of Sambucus wightiana Wall. ex Wight & Arn. extract in streptozotocin-nicotinamide induced diabetic Wistar Albino rats. An acute toxicity study was performed to establish the safe dose of total alkaloids extracted from S. wightiana (TASW). TASW was administered orally (50 mg, 100 mg, and 200 mg/kg body weight per day) to diabetic rats for 21 days. Fasting blood glucose level was determined, along with other associated parameters that may fluctuate in diabetes, consisting of the lipid profile, body weight, serum liver marker enzymes and Hb level. The test groups that were administered TASW at three different doses showed a marked reduction in hyperglycaemia when compared to the control group. Moreover, body weight and food intake improved among test group animals. Administration of TASW also reduced the levels of triglycerides, cholesterol and LDL while increasing HDL and Hb levels. Serum liver enzyme activity is generally increased in diabetes, and this was reduced after dosing with TASW. The findings of this study elucidate that the total alkaloids extracted from S. wightiana are not toxic at the tested doses, and present potential hypoglycaemic activity.

Abstract:Diabetes mellitus (DM) is one of the leading worldwide public health problems. It is characterized by hyperglycemia which induces oxidative stress and inflammation, both involved in the pathogenesis of diabetes. We previously showed that Boswellia dalzielii (BD) and Hibiscus sabdariffa (HS) extracts reduced hyperglycemia and hyperlipidemia in alloxan-induced diabetic rats. In the present study, we evaluated the antioxidant and anti-inflammatory activities of both plants in alloxan-induced diabetic rats. Two sets of experiments were conducted in male Wistar rats subjected to a single intraperitoneal injection of alloxan monohydrate (150 mg/kg, b. w.). Then, diabetic rats were daily administered with either BD (1st set of experiments) or HS (2nd set of experiments) at 100, 200, and 400 mg/kg orally for 21 consecutive days. Glibenclamide (10 mg/kg) was also administered as a reference drug. At the end of the study, the animals were anesthetized, and blood samples were collected from each animal. Then, oxidative stress and inflammatory biomarkers in the serum were determined. We found that treatment with BD and HS significantly reduced malondialdehyde (MDA) and enhanced the levels of reduced glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT). These extracts also significantly decreased the inflammatory markers tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1 beta (IL-1β). From the results obtained, it can therefore be concluded that BD and HS have the potential to being developed as natural sources of antioxidant and anti-inflammatory agents that can be used for the prevention or treatment of DM.

Abstract:Background: The prevalence of diabetes mellitus (DM) is increasing overtime, potentially leading to various severe health complications and mortality. Despite therapeutic agents have currently been developed, unexpected adverse effects are inevitable. Hence, safe and effective medications such as those of plant origin are critical to prevent unexpected complication in DM sufferers. Etlingera elatior has been widely used as spice and traditional medicine to treat diabetes in Aceh Province, Indonesia. However, study regarding α-glucosidase inhibitory effect of E. elatior growing in Gayo highlands, Aceh, Indonesia, is completely lacking. The aim of this study was to evaluate in vitro α-glucosidase inhibitory effect of E. elatior ethanol extracts (EEEE) growing in Gayo highlands, Aceh Province, Indonesia. Methods: Antioxidant activity was determined using DPPH procedure, whereas α-glucosidase inhibition assay was carried out using spectrophotometric method. Data analysis was performed using One-Way Analysis of Variance (ANOVA), followed by Duncan's multiple range test at α=0.05. Results: Phytochemical analysis revealed the presence of total phenolic (TPC), total flavonoid (TFC), and total tannin (TTC) content in all E. elatior plant parts, in which the highest TPC was found in the stem (158.38 GAE/g), whereas the highest TFC and TTC was obtained in the rhizome extracts. The extract of fruit showed the strongest antioxidant activities, followed by the stem and leaf, with IC 50 of 2.381 μg/mL, 6.966 μg/mL, and 19.365 μg/mL, respectively. All E. elatior extracts revealed a significant inhibitory activity against α-glucosidase at the concentration of 500 μg/mL, in which the stem extract showed the most effective α-glucosidase inhibitory effect with IC 50 value of 5.15 μg/mL, suggesting its promising potential as antidiabetic agent. Conclusions: This study highlights E. elatior potency as a novel source of antioxidant and natural antidiabetic compounds that are useful for the prevention and treatment of diabetes.

Abstract:Diabetes mellitus is the most common chronic endocrine and metabolic disorder, impacts the quality of life, and has become the leading cause of death worldwide. Now a day’s, herbal drugs are in great demand for the treatment of diabetes because of their traditional acceptability, lesser side effects, and adverse effects compared with allopathic medications. Terminalia arjuna fruits were selected for the study due to their abundant availability and based on ethanomedicinal folk claims. The fruits of Terminalia arjuna have been evaluated for their antidiabetic and antihyperlipidemic activities using streptozotocin-induced diabetic Wistar albino rats. In the study, two doses of ethanolic extract of Terminalia arjuna fruits (TAF) were selected and administered to normal rats for an oral glucose tolerance test. For an antidiabetic study in streptozotocin-induced diabetic rats, the effect of the extract was observed for 28 days for blood glucose alterations. All experimental animals were observed for weight changes, due to STZ-induced diabetes, and were found to be significantly reverted to normal in all animals except the diabetic control group. After the completion of the experimental period, the collected serum samples were subjected to various biochemical parameters studied, including a lipid profile, glycosylated hemoglobin (HbA1c), urea, and creatinine level. An antioxidant study was performed using collected tissues, followed by histopathological studies of the pancreas. The research study revealed that the TAF extract has significant antidiabetic activity and the potential to revert the altered lipid profile and increase HbA1c to normal. TAF extract significantly normalized the urea, creatinine, superoxide dismutase, catalase, and glutathione peroxidase levels. Thus, the study concluded that TAF extract exhibits significant (p>0.05) antidiabetic activity in STZ-induced diabetic rats.

Abstract:Abstract Diabetes Mellitus is a metabolic disease characterized by hyperglycemia, occurring due to abnormal insulin secretion or insulin action. Alloxan induces diabetes by damaging pancreatic cells and initiating hyperglycemia. One of the plants that has the potential to decrease blood sugar is leaves of sikkam ( Bischofia javanica ), because it contains quercetin and gallic acid compounds which are classified as an antioxidant group. The objectives of this study were (1) to analyze the effect of sikkam leaf extract on the reduction in blood sugar levels of Wistar rats and improved the histology of langerhans islet and (2) determine the dose of sikkam leaf extract which has the most effective effect in reducing rat blood sugar levels. The design used is a non factorial complete random design. The samples used were 30 Wistar rats which were divided into 6 treatment groups and 5 repetitions, namely K 0 (negative control), K 1 (positive control), K 2 (dose of ethanol extract of sikkam leaf (EESL) 300 mg/Kg BW), K 3 (EESL 600 mg/kg BW), and K 4 (EESL treatment 900 mg/kg kg) and K 5 (Glibenclamid 4.5 mg). Data analysis used One-way ANOVA followed by LSD test. The results showed that there was an EESL effect on the reduction in rat blood sugar levels and improved the histology of Langerhans islet. EESL which has the best antidiabetic effect is on administering a dose of 900 mg/kg BW.

Abstract:Diabetes mellitus (DM) is a serious health issue for the general public. In this work, male diabetic rats were used to determine the impact of Citrullus colocynthis and Phyllanthus Emblica fruits ethanolic extract (EECC and EEPE). Six groups of thirty mature rats (male Sprague Dawley) were examined. In the first group, normal rats have been fed a balanced diet served as the negative control group (C-); in the second group, diabetic rats served as the control positive (C+). Diabetic rats in groups 3, 4, 5, and 6 received daily treatments of (200 and 400 mg kg-1 body weight (BW) EECC and EEPE, respectively. After four weeks of treatment, Biochemical indicators were examined in all blood samples. Streptozotocin was injected and resulted in a significant reduction in feed intake (FI), feed efficiency (FER) body weight gain (BWG), serum insulin, high-density lipoprotein (HDL-c), globulin, albumin, and total protein, as well as antioxidant enzymes, glutathione (GSH) and superoxide dismutase (SOD) in pancreatic tissue, whereas a significant increment was noted in serum glucose, triglyceride (TG), total cholesterol (TC), low & very low-density lipoprotein cholesterol (LDL-c and VLDL-c), atherogenic index (AI), Alkaline phosphatase (ALP), Aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, uric acid, creatinine, tumor necrosis factor-α, creatine phosphate kinase (CPK) and lactate dehydrogenase (LDH) and lipid peroxidation (MDA) in pancreatic tissue. The treatment of diabetic animals with different ethanolic extracts caused improvements in all previously measured parameters, with EEPE (400 mg kg-1 BW) showing the greatest improvement.
