Abstract:The antidiabetic activity of the total lignan from the plant Fructus Arctii, used in China for the control of diabetes, was investigated in models of alloxan-induced diabetic mice and hyperglycemic-hyperlipidemic diabetic rats. The biochemical parameters studied were: blood glucose, glucose tolerance, serum insulin, serum triglycerides, total cholesterol and high density lipoprotein (HDL). Total lignan was given to mice and rats daily for 10 days at doses of 2.0, 1.0, 0.5 g/kg and 1.38, 0.69, 0.35 g/kg respectively. The alloxan-diabetic animals showed significant reductions in plasma glucose, triglycerides and total cholesterol after treatment with the total lignan from the plant Fructus Arctii and glibenclamide (used as standard) compared with the diabetic controls, while the glucose tolerance, serum insulin level and HDL-cholesterol were elevated without the risk of hypoglycemia. In conclusion, the total lignan from the plant Fructus Arctii has been proven to be a safer antidiabetic agent and might help to prevent diabetic complications. It can serve as a good adjuvant in the present armamentarium of antidiabetic drugs. Copyright (c) 2007 John Wiley & Sons, Ltd.

Abstract:Abstract Background In diabetic animals, there is a significant increase in plasma glucose, serum total cholesterol, triglyceride, and low-density lipoprotein levels, and decreased body weight, liver and muscle glycogen, and high-density lipoprotein. Effective treatment of diabetes mellitus is not yet known, even though the management of diabetes mellitus is considered a global concern. Plants and herbs have played an important role in the healthcare of many societies throughout history. Today’s researchers are investigating the potential for using these nonpharmaceutical approaches to treat and control diabetes, either in conjunction with standard treatments or as an alternative to them. Herbal formulations are favored because to lower cost and fewer side effects compared to other methods for alleviating diabetes and its consequences. In ethnomedicinal practices, different parts of Mangifera indica are used to treatment of diabetes. The present investigation was undertaken to evaluate the antidiabetic activity of an ethanolic extract of Mangifera indica and mangiferin in alloxan-induced diabetic rats. This experiment was conducted in a set of two with four groups of animals namely control (Tc), treatment alloxan (Ta), treatment extract (Tae), and treatment mangiferin (Tam). To develop diabetes, Wistar rats treated with 150 mg/kg b.w. of alloxan monohydrate were injected intraperitoneally. Tae and Tam’s groups received a freshly prepared single dose of extract and mangiferin in distilled water via the oral route. All experimental groups received laboratory pallet feed diet and drinking water ad libitum. Diabetic rats were treated for 21 days with an ethanolic extract of mango peel and pure mangiferin orally daily at rates of 200 mg/kg b.w. and 20 mg/kg b.w. Results An alloxan-induced diabetic rat treated with mango peel extract and mangiferin significantly improved the overhead impact due to diabetes. There was a significant ( p < 0.05) body weight loss in the alloxan-induced diabetic rats (Ta), whereas animals given mango peel extract and mangiferin showed a significant increase in body weight from 2 weeks onwards in comparison with control. Alloxan-induced rats (Ta) group have higher blood glucose levels and are significantly different ( p < 0.01) from the control group. Mango peel extract and mangiferin significantly reduced the levels of fasting glucose after 21 days of treatment in comparison with diabetic animals. Mango peel extract and mangiferin influence the glycogen synthesis pathway in diabetes groups by increasing glycogen levels in muscle and liver. mango peel extract and mangiferin were found to have a nonsignificant impact on plasma cholesterol and HDL levels compared with the control group. Mango peel extract was found to have a significant difference ( p < 0.05) in LDL levels compared with the control group. Mangiferin was found to have a significant difference ( p < 0.05) in triglyceride and VLDL levels when compared with the control group. Histopathological examination of the pancreas in rats with type I diabetes caused by alloxan found that therapy with an ethanolic extract of mango peel and mangiferin restored beta cell function as well as rejuvenation of Islets of Langerhans. Conclusions Mango peel extract and mangiferin have antidiabetic, glycogenesis, and hypolipidemic properties when administered to alloxan-induced diabetic rats. Graphical abstract

Abstract:Introduction Acacia leucophloea (AL) has been used traditionally to treat diabetes, cancer, inflammation, ophthalmia, haemorrhoids and leukoderma, however the scientific evidence validating its antidiabetic activity is limited. Employing a multi-targeted therapeutic strategy could be advantageous for the management of type 2 diabetes mellitus which is strongly correlated with oxidative stress-induced pathways. The present study was intended to evaluate the antidiabetic and antioxidant potential of AL via in vitro assays, validating its traditional medicinal significance. Methods Anti-diabetic potential of the AL extract, its fractions and isolated compounds was assessed through the determination of α-glucosidase, DPP-IV inhibition, anti-glycation activity, and glucose uptake using 2-NBDG in differentiated L6 myoblast, along with cytotoxicity studies. Total phenolic and flavonoid content was measured and the antioxidant potential was evaluated using in vitro assays i.e., DPPH, ABTS and FRAP. In addition, quantitative determination of isolated compounds and their purity was analysed using HPLC-DAD. Results Fractions with the highest phenolic/flavonoid content (VI to XIII of AL) demonstrated substantial antioxidant and antidiabetic potential across selected therapeutic targets and these active fractions yielded six compounds upon fractionation using column chromatographic techniques. Structures of these compounds were examined using spectroscopic techniques and were identified as isoquercitrin, quercitrin, luteolin, epigallocatechin-3-O-(3″-O-methyl)-gallate, apigenin-7-O-glucoside and ethyl gallate. Isolated compounds showed significant DPP-IV inhibitory activity in a concentration-dependent manner with the highest activity with minimum IC50 being possessed by isoquercitrin (IC50 4.28 ± 0.32 µM). At a concentration of 200 µM, apigenin-7-O-glucoside displayed a considerable glycation inhibitory effect (81.57 ± 2.66 %), surpassing the effect of other compounds. Moreover, at a concentration of 50 μM, the compounds apigenin-7-O-glucoside and isoquercitrin have increased the glucose uptake in L6 myotubes by 11.5 % and 11 % respectively, compared to a 6.1 % increase by the standard drug metformin. Notably, this study reveals for the first time the potential of both apigenin-7-O-glucoside and isoquercitrin to enhance glucose uptake in L6 myotubes. Conclusion AL and its isolated compounds exhibit promising effects in ameliorating post-prandial hyperglycemia. Their multifaceted therapeutic potential prompts further investigation to elucidate the underlying molecular mechanisms.

Abstract:This work aimed to determine the phytochemical composition of the aqueous extract of leaves of Ficus vallis-choudae (AEFV) and to evaluate its antidiabetic properties on a model of type 2 diabetes induced by a high-fat diet (HFD) and a low dose of streptozotocin (STZ). The phytochemical analysis was carried out according to several methods using the standard of each bioactive compound. Type 2 diabetes was induced by feeding rats for 4 weeks with HFD lard followed by injection of a low dose of STZ (35 mg/kg). After induction, the rats were divided into groups and treated for 28 days with metformin (40 mg/kg) and the AEFV at doses of 110, 220, and 440 mg/kg. The results showed that the AEFV contains saponins, flavonoids, tannins, and total polyphenols. In addition, it dramatically reduced body mass, body mass index (BMI), atherogenic index (AI), coronary heart risk index (CRI), and abdominal fat and increased homeostatic model assessment of β-cell function (HOMA-β), high-density lipoprotein cholesterol (HDL-c) levels, and cardioprotective index (CI). The AEFV also lowered blood glucose levels, insulinemia, homeostatic model assessment of insulin resistance (HOMA-IR) index, and total cholesterol (TC), triglycerides (TG), low-density lipoproteins cholesterol (LDL-c), and very-low-density lipoproteins cholesterol (VLDL-c) levels. There was a decrease in alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity and in urea and serum creatinine levels following the administration of AEFV. The AEFV caused increased superoxide dismutase (SOD) and catalase (CAT) activities, reduced glutathione (GSH) levels, and decreased malondialdehyde (MDA) levels in the liver, kidneys, and heart of rats. The AEFV has hypoglycemic, antioxidant, and cardioprotective properties, thus validating its use in traditional medicine for the treatment of type 2 diabetes and its complications.

Abstract:BACKGROUND: Fruits, leaves, and tuberous roots of Momordica dioica are used as a folk remedy for diabetes mellitus (DM) in India. The aqueous extract of Momordica dioica fruit possesses very good anti-diabetic activity and is having high margin of safety.OBJECTIVES: The aim of the present study was to investigate the antioxidative effect of Momordica dioica fruits in alloxan-induced diabetic Wistar rats.MATERIALS AND METHODS: Effect of aqueous extract of Momordica dioica (AEMD) on thiobarbituric acid reactive substances (TBARS), hydroperoxide (HP), non-enzymatic and enzymatic antioxidants in liver, kidney, pancreas, and serum was evaluated in diabetic rats after 21 days treatment.RESULTS: Increase in the levels of TBARS, HP and decrease in the levels of non-enzymatic antioxidants and activity of enzymatic antioxidants was observed in liver, kidney, pancreas, and serum of diabetic rats when compared with normal healthy rats. TBARS and HP levels were reduced while non-enzymatic and enzymatic antioxidant enzymes activity was increased in AEMD and glibenclamide-treated rats. Furthermore, histological examination of liver, kidney, and pancreas of diabetic rats showed degenerative changes. AEMD treatment for 21 days rejuvenated liver, kidney, and pancreas histoarchitecture.CONCLUSION: In conclusion, the present results showed the protective role of AEMD on liver, kidney, and pancreas in severe diabetic rats justifying support for its anti-diabetic use in folk medicine.
Fractionation of Xanthium strumarium L. foliage phenolics, in-vitro antioxidant activities, and in-vivo anti-diabetic potential

Abstract:Introduction: The present research aimed to fractionate Xanthium strumarium L. (XSL) foliage phenolics into a set of solvents and evaluate their antioxidant potential and in-vivo anti-diabetic activity against Alloxan monohydrate-induced diabetic mice. Methodology: For this purpose, XSL foliage was fractionated into petroleum ether, ethyl acetate, ethanol, and water via orbital type shaking and tested for the presence of phenolics, and their antioxidant and antidiabetic potential. Results and discussion: The results revealed that the ethyl acetate fraction of XSL foliage contained the highest amount of total phenolics 95.25 mg GAE/g of extract, followed by ethanol (65.14 mg GAE/g), petroleum ether (25.12 mg GAE/g), water (12.20 mg GAE/g), and XSL powder (69.13 mg GAE/g). At the end of treatment time (day 18 of oral administration of 400 mg/kg body weight of mice), the ethyl acetate fraction significantly (p ≤ 0.05) lowered blood glucose level (353 ± 10.6 to 220 ± 25.5 mg/dL) which might due to the elevated level of phenolic compounds in this fraction. Conclusion: Overall, it can be speculated that ethyl acetate and ethanol may work efficiently for the enrichment of XSL phenolic without compromising their antidiabetic potential.

Abstract:Diabetes mellitus is the most common chronic endocrine and metabolic disorder, impacts the quality of life, and has become the leading cause of death worldwide. Now a day’s, herbal drugs are in great demand for the treatment of diabetes because of their traditional acceptability, lesser side effects, and adverse effects compared with allopathic medications. Terminalia arjuna fruits were selected for the study due to their abundant availability and based on ethanomedicinal folk claims. The fruits of Terminalia arjuna have been evaluated for their antidiabetic and antihyperlipidemic activities using streptozotocin-induced diabetic Wistar albino rats. In the study, two doses of ethanolic extract of Terminalia arjuna fruits (TAF) were selected and administered to normal rats for an oral glucose tolerance test. For an antidiabetic study in streptozotocin-induced diabetic rats, the effect of the extract was observed for 28 days for blood glucose alterations. All experimental animals were observed for weight changes, due to STZ-induced diabetes, and were found to be significantly reverted to normal in all animals except the diabetic control group. After the completion of the experimental period, the collected serum samples were subjected to various biochemical parameters studied, including a lipid profile, glycosylated hemoglobin (HbA1c), urea, and creatinine level. An antioxidant study was performed using collected tissues, followed by histopathological studies of the pancreas. The research study revealed that the TAF extract has significant antidiabetic activity and the potential to revert the altered lipid profile and increase HbA1c to normal. TAF extract significantly normalized the urea, creatinine, superoxide dismutase, catalase, and glutathione peroxidase levels. Thus, the study concluded that TAF extract exhibits significant (p>0.05) antidiabetic activity in STZ-induced diabetic rats.

Abstract:Introduction: Diabetes is a multi-factorial metabolic disorder characterized by hyperglycemia, which is the primary factor in preventing glucose from achieving its desired levels in patients with the condition. Hyperglycemia results from a lack of insulin secretion, inadequate insulin action or both. Materials and Methods: The comparative effects of ethanolic leaf extracts of The comparative effects of ethanolic leaf extracts of Gongronema latifolium (G. latifolium) and Bryophyllum pinnatum (B. pinnatum) on some biochemical parameters in diabetic induced albino rats were investigated. Two different concentrations of ethanolic leaves extracts of both plants were used to treat alloxan-induced diabetic rats. Results: The study found that alloxan administration significantly increased blood sugar levels in diabetic rats, with the highest reduction in group 7 and the lowest in group 5. The ethanolic extracts of B. pinnatum and G. latifolium leaves showed potential hepatoprotective and renal protective activities, but mild urea level alteration was observed. The study suggests that these extracts may be beneficial for diabetic rats. Conclusion: The study reveals that alloxan monohydrate can induce diabetes in animals, and ethanolic extracts from B. pinnatum leaves and G. latifolium leaves may reduce blood sugar levels in diabetic rats. Glibenclamide showed better hypoglycaemic effects. Alloxan may cause hepatotoxicity and renal toxicity, but the extracts may also cause mild urea level alteration.
Andaliman (Zanthoxylum acanthopodium DC.) fruit ethanolic extract exerts attenuative effect on hyperglycemia, sensory and motoric function’s disorders in alloxan-induced diabetic mice

Abstract:Objective: Andaliman (Zanthoxylum acanthopodium) is a potent medicinal plant in Asia. This present study aimed to reveal the effectivity of Andaliman fruit extract in alleviating hyperglycemia, sensory and motoric balance disorders, histopathology of the cerebellum, and tissue oxidative stress in diabetic mice induced by alloxan. Materials and Methods: Diabetes induction was performed by intraperitoneally injecting alloxan monohydrate [200 mg/kg body weight (BW)]. Subsequently, the mice were treated daily with an ethanolic extract of Andaliman fruit (0, 150, 300, 450 mg/kg BW per oral) for 28 days, followed by measurements of blood glucose, paw sensitivity, motoric balance, histopathology of the cerebellum, and malondialdehyde (MDA) levels. Moreover, the phytochemical constituents of the extract were elucidated by liquid chromatography. Results: Higher doses of Andaliman fruit extract could significantly attenuate the elevation of random and fasting blood glucose (p < 0.05) and improve paw sensitivity responses (p < 0.05) and motoric balances (p < 0.05) in diabetic mice. Moreover, Andaliman fruit extract could significantly attenuate the degeneration of cerebellar Purkinje cells (p < 0.05) and suppress MDA levels in the blood (p < 0.05) while blunting the MDA in the brain tissue (p < 0.05). Phytochemical screening revealed 39 compounds in the Andaliman extract belonging to the groups of alkaloids (26 compounds), flavonoids (12 compounds), and terpenoids (1 compound). Conclusion: The ethanolic extract of Andaliman fruit is capable of ameliorating diabetic neuropathy, motor balance disorders, and Purkinje cell degeneration while also reducing oxidative stress in the peripheral system. Hence, Andaliman extract is a promising candidate for formulation as an herbal remedy against the detrimental outcomes of diabetes mellitus.

Abstract:Ethanolic extracts of Ananas comosus L. leaves (AC) enriched with phenols have hypoglycemic activity in diabetic rats. Here, we investigated the effect of AC on insulin sensitivity in rats and HepG2. In high-fat diet-fed and low-dose streptozotozin-treated diabetic Wistar rats subjected to challenge with exogenous human insulin, AC treatment at an oral dose of 0.40 g/kg could significantly improve sensitivity to exogenous insulin. After a sub-acute treatment, AC also could inhibit the development of insulin resistance in high-fat diet-fed and low-dose streptozotozin-treated diabetic rats following the test of loss of tolbutamide-induced blood glucose lowering action. For intravenous insulin/glucose infusion test, high-fat diet-fed and low-dose alloxan-treated Wistar rats were associated with insulin resistance, which was improved after AC or fenofibrate treatment. AC application inhibited the development of insulin resistance in HepG2 cells. The above animal models were well developed to simulate type 2 diabetes. Taken together, our results suggest that AC may improve insulin sensitivity in type 2 diabetes and could be developed into a new potential natural product for handling of insulin resistance in diabetic patients.

Abstract:Scope: Type 2 diabetes is a complex metabolic and endocrine disorder worldwide, which causes severe health and economic problems. The aim of this study is to investigate the molecular mechanisms by which arabinoxylan from Plantago asiatica L. attenuates type 2 diabetes from the perspective of urine metabolomics. Methods and Results: High-fat diet and streptozotocin-induced type 2 diabetic rats are treated with arabinoxylan, then the urine samples are collected for untargeted metabolomics analysis by UPLC-Triple-TOF/MS. Diabetes causes significant increases in the levels of acetone, glucose, 2-oxoglutarate, and leucine, and significant decreases in the concentrations of creatine, histidine, lysine, L-tryptophan, hippurate, L-cysteine, kynurenine, and arabitol as compared with normal rats (p < 0.01). And these 12 metabolites (with VIP cut-off value > 1) can be used as biomarkers in type 2 diabetes. A total of 21 urinary metabolites are significantly improved by arabinoxylan administration in diabetic rats, and these metabolites are mainly involved in TCA cycle, and metabolism of lipid and ketone body, taurine and hypotaurine, tryptophan, and branched chain amino acids. Conclusion: Arabinoxylan administration improves carbohydrate, lipid, and amino acid metabolism in type 2 diabetic rats, which provide important insights into the mechanisms underlying type 2 diabetes as well as the effects of arabinoxylan.
Aporphine alkaloids identified from Xylopia aethiopica and their potential hypoglycemic and hypolipidemic activities

Abstract:Xylopia aethiopica has been widely utilized as food condiments in West African countries, as well as traditional medicine for diabetes, but the responsible components remain unknown. In this work, the probable active components in the contributing fraction were screened using bio-affinity ultrafiltration strategy targeting α -glucosidase and pancreatic lipase. Then, 11 alkaloids were identified, 9 of which were aporphines, including a new one. Further enzyme inhibitory validation of these compounds discovered that compounds 1 , 2 , 6 and 11 showed better α-glucosidase inhibition activity than positive control, among which compounds 1 , 2 and 6 simultaneously possessed potent inhibition against pancreatic lipase. The hypoglycemic and hypolipidemic capacities of these screened compounds were verified by in vitro glucose consumption and lipid accumulation HepG2 cell models. This work provided solid evidences for the anti-diabetic benefit of X. aethiopica , and several aporphine alkaloids could be the responsible compounds for its promising hypoglycemic and hypolipidemic activities.

Abstract:Diabetes mellitus (DM) is one of the leading worldwide public health problems. It is characterized by hyperglycemia which induces oxidative stress and inflammation, both involved in the pathogenesis of diabetes. We previously showed that Boswellia dalzielii (BD) and Hibiscus sabdariffa (HS) extracts reduced hyperglycemia and hyperlipidemia in alloxan-induced diabetic rats. In the present study, we evaluated the antioxidant and anti-inflammatory activities of both plants in alloxan-induced diabetic rats. Two sets of experiments were conducted in male Wistar rats subjected to a single intraperitoneal injection of alloxan monohydrate (150 mg/kg, b. w.). Then, diabetic rats were daily administered with either BD (1st set of experiments) or HS (2nd set of experiments) at 100, 200, and 400 mg/kg orally for 21 consecutive days. Glibenclamide (10 mg/kg) was also administered as a reference drug. At the end of the study, the animals were anesthetized, and blood samples were collected from each animal. Then, oxidative stress and inflammatory biomarkers in the serum were determined. We found that treatment with BD and HS significantly reduced malondialdehyde (MDA) and enhanced the levels of reduced glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT). These extracts also significantly decreased the inflammatory markers tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1 beta (IL-1β). From the results obtained, it can therefore be concluded that BD and HS have the potential to being developed as natural sources of antioxidant and anti-inflammatory agents that can be used for the prevention or treatment of DM.

Abstract:Abstract Type 2 diabetes (T2D) is the most common metabolic disorder. The undesirable effects of synthetic drugs demand a search for safe antidiabetic agents. This study aimed to assess the antidiabetic activity of different fractions of Atriplex halimus (petroleum ether 60–80, methylene chloride, ethyl acetate, and n ‐butanol) using Drosophila melanogaster larvae. Titers of total glucose and trehalose, as well as larval weight, were measured and compared with those of control and diabetic larvae. The expression of Drosophila insulin‐like peptides (DILP2 and DILP3) and adipokinetic hormone (AKH) was evaluated. The results revealed a significant increase in total glucose, trehalose, and a decrease in body weight in the larvae fed a high‐sugar diet compared with those in the control. When larvae fed diets containing the tested fractions, the total glucose and trehalose decreased to the control level, and the body weight increased. DILP2, DILP3, and AKH exhibited significant decreases upon treatment with A. halimus ethyl acetate. Metabolomic profiling of the ethyl acetate fraction of A. halimus revealed the presence of flavonoids and flavonoid glycosides. After docking screening to predict the most powerful moiety, we discovered that flavonoid glycosides (especially eriodictyol‐7‐ O ‐neohesperidoside) have a greater affinity for the pocket than the other moieties. The results indicated the therapeutic activity of the A. halimus ethyl acetate fraction against induced T2D in Drosophila larvae. The antidiabetic activity may be attributed to flavonoids, which are the main components of the A. halimus ethyl acetate fraction.

Abstract:Zanthoxylum armatum DC, a medicinal plant species in India, is an exceptionally nutritious vegetable with a variety of potential uses in treating rheumatism, venomous bites, and microbial infections. In the present study, we aimed to investigated the antidiabetic and antioxidant effects of aqueous extracts of Z. armatum Stem Bark (Za-Aq) in High Fat Diet (HFD) low dose streptozotocin (STZ)-induced type 2 diabetic wistar albino rats and the objectives the were achieved through examine of blood glucose, Oral Glucose Tolerance Test (OGTT), Insulin Tolerance Test (ITT), and the levels of lipid peroxides and antioxidant enzymes, such as superoxide dismutase, glutathione peroxidase and reduced glutathione in the liver tissues of control and experimental groups. Diabetic rats were treated with 200, 400 and 800 mg/kg BW Za-Aq for 45 days and the antidiabetic effects of the extract were evaluated by measuring changes in biochemical parameters in the serum and liver tissue. Oral administration of Za-Aq to diabetic rats for 45 days significantly reduced the levels of blood glucose, OGTT, ITT and lipid peroxidation, but increased the activities of antioxidant enzymes, like superoxide dismutase, reduced glutathione and glutathione peroxidase. The progression of diabetes was significantly reduced after Za-Aq treatment. In conclusion, Za-Aq exerts protective effects against HFD-STZ-induced diabetes. The Za-Aq supplementation is useful in controlling the blood glucose level and is beneficial in preventing diabetic complications from lipid peroxidation and antioxidant systems in experimental diabetic rats; therefore, it could be useful for prevention or early treatment of diabetes mellitus active constituents may be isolated from the extract for evaluation in future clinical studies. To best of our knowledge there has been no previous comprehensive study to evaluate antidiabetic activity of stem bark from Z. armatum DC and present study adds the protective effect of Zanthoxylum armatum DC stem bark as effective antidiabetic and antioxidant agent.

Abstract:Annona reticulata L. (Bullock's heart) is a pantropic tree commonly known as custard apple, which is used therapeutically for a variety of maladies. The present research was carried out to evaluate the possible protective effects of Annona reticulata L. (A. reticulata) ethanolic seed extract on an experimentally induced type 2 diabetes rat model. Male Albino Wistar rats were randomly divided into five groups with six animals in each group viz., control rats in group I, diabetic rats in group II, diabetic rats with 50 and 100 mg/kg/bw of ethanolic seed extract of A. reticulata in groups III and IV, respectively, and diabetic rats with metformin in group V. Treatment was given for 42 consecutive days through oral route by oro-gastric gavage. Administration of A. reticulata seed extract to diabetes rats significantly restored the alterations in the levels of body weight, food and water intake, fasting blood glucose (FBG), insulin levels, insulin sensitivity, HbA1c, HOMA-IR, islet area and insulin positive cells. Furthermore, A. reticulata significantly decreased the levels of triglycerides, cholesterol, LDL, and significantly increased the HDL in diabetic rats. A. reticulata effectively ameliorated the enzymatic (ALT, AST, ALP, GGT) and modification of histopathological changes in diabetic rats. The serum levels of the BUN, creatinine levels, uric acid, urine volume, and urinary protein were significantly declined with a significant elevation in CCr in diabetic rats treated with A. reticulata. MDA and NO levels were significantly reduced with an enhancement in SOD, CAT, and GPx antioxidant enzyme activities in the kidney, liver, and pancreas of diabetic rats treated with A. reticulata. Diabetic rats treated with A. reticulata have shown up-regulation in mRNA expression levels of nuclear factor erythroid 2-related factor 2 (Nrf2), NAD(P)H:quinone oxidoreductase 1 (NQO1), Heme oxygenase-1 (HO-1) and protein expression level of Nrf2 with diminution in Keap1 mRNA expression level in pancreas, kidney, and liver. From the outcome of the current results, it can be inferred that seed extract of A. reticulata exhibits a protective effect in diabetic rats through its anti-diabetic, anti-hyperlipidemic, antioxidant and anti-inflammatory effects and could be considered as a promising treatment therapy in the treatment of diabetes mellitus.

Abstract:Type 1 diabetes mellitus is an autoimmune disorder characterized by the destruction of pancreatic beta cells, leading to impaired insulin production and regulation. Recent research has shown that medicinal plants with antioxidant and anti-inflammatory properties hold promise as potential therapeutic agents for managing complications of the disease. The aim of this study is to investigate the anti-diabetic, antioxidant, and anti-inflammatory effect of ethyl acetate extract of Annona muricata leaves in streptozotocin (STZ)-induced diabetes in Wistar rats. Thirty five (35) male Wistar rats weighing 150-180g were used for this experiment. Group 1 (Normal control), Group 2 (STZ 40 mg/kg only Diabetic control), Group 3 (STZ+ metformin 500mg/kg bw), Group 4 (STZ + ethyl acetate extract 250 mg/kg bw), Group 5 (STZ + ethyl acetate extract (500 mg/kg bw). Fasting blood samples were collected by cardiac puncture for glucose estimation and liver tissues excised for both biochemical and molecular studies. The study demonstrated the extract's ability to modulate the NF-κB pathway, leading to the suppression of inflammatory responses in diabetic rats. These findings suggest that the ethyl acetate extract of A. muricata leaves may ameliorate type 1 diabetes by promoting antioxidant defense and reducing inflammatory processes through the NRF2 and NF-κB pathways. This research sheds light on the potential of natural compounds as adjunct therapies for type 1 diabetes and provides a foundation for further investigations into the development of novel treatments targeting oxidative stress and inflammation in autoimmune diabetes.
