Abstract:This study investigated the aroma-active compounds and compared the differences of three different grades of sesame-flavor Baijiu by headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-olfactometry-mass spectrometry (GC-O-MS). A total of 54 aroma-active compounds were detected. Principal component analysis showed that JZ1, JZ2, and JZ3 were well separated from each other. JZ1 as the premium-grade Baijiu had the highest aroma intensities, concentrations. According to aroma intensities and concentrations, dimethyl trisulfide, butanoic acid, phenylacetaldehyde, 2-furylmethanethiol, ethyl hexanoate, 2,6-dimethylpyrazine, etc. could be potentially applied as volatile makers to distinguish the three different grades of sesame-flavor Baijiu as their significant difference (P < 0.05) in three Baijiu samples. Roasted aroma had the significant difference (P < 0.05) in three sample base on aroma profiles. Meanwhile, 2-furylmethanethiol, 2,6-dimethylpyrazine were related to the roasted aroma, they may be had a significant contribution to the differences of three different grades of sesame-flavor Baijiu. Extraction of Fish Sauce Flavor Compounds and Optimization of Gas Chromatographic Separation Conditions

Abstract:The extraction solvent and analysis conditions of gas chromatography (GC) for fish sauce flavor compounds were studied. The data showed that ethyl acetate is the most suitable solvent for the fish sauce flavor extraction. A capillary column CP-Sil -8cb-30 m-0.32 mm was selected to quantitatively detect the fish sauce flavor compounds. The optimal temperatures of sample receiver and FID detector are 200 ℃ and 250 ℃, respectively, with nitrogen (purity >99.999%) as the carrier at the speed of 3 ml/min, at the shunting ratio100:1. The starting temperature is 30 ℃ for 2 min and programmed to rise from 30 ℃ to 120 ℃ by 5 ℃/min, continually to 180℃ by 12 ℃/min, then hold for 4 min. Under these optimized conditions, total 44 compounds are separated, and 24 of them can be separated from the baseline.

Abstract:The complexity of flavor compound interactions significantly influences the overall flavor characteristics of alcoholic beverages. The vast diversity of these compounds, however, makes the screening for interactions both labor-intensive and time-consuming. To address this challenge, our study aimed to develop a rapid and visual approach using Headspace Gas Chromatography-Ion Mobility Spectrometry (GC-IMS), optimized via a Box-Behnken design and Response Surface Methodology (RSM). This method was validated through the investigation of interactions between lactic acid and 13 key flavor compounds in Baijiu. Furthermore, Gas Chromatography-Mass Spectrometry was utilized to quantify the impact of lactic acid on the volatility of these compounds, corroborating the findings from GC-IMS. Additionally, molecular docking techniques were employed to explore the interaction mechanisms between lactic acid and ethyl lactate, 3-hydroxy-2-butanone, and 2-methoxy-4-methylphenol, at the molecular level, offering insights into their potential binding configurations. Our findings offer a streamlined approach for screening flavor compound interactions, enhancing the understanding of flavor formation and facilitating future research and development in complex food matrices.

Abstract:A method for detection of flavor compounds in soy sauce by SPME-GC was developed.The analysis parameters of extraction time,sample temperature,content of sodium chloride,desorption time were optimized.The optimized method was as follows:10mL soy sauce was placed into 20mL vial and saturated with 1g sodium chloride.The sample was extracted using 85μm PA fiber for 30min at 40℃,then the volatiles were desorbed from the fiber at temperature of 230℃ for 3min.The method is simple,quick,reproducible and sensitive.

Abstract:Doubanjiang, a Chinese traditional fermented red pepper paste, is eaten worldwide for its unique flavor. The objective of this study was to evaluate the aroma quality of doubanjiang using solvent-assisted flavor evaporation (SAFE) and headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-olfactometry (GC-O) and aroma extract dilution analysis (AEDA). A total of 165 volatile compounds, belonging to 13 chemical classes, were identified. Esters and hydrocarbons were the predominant groups. Thirteen aroma-active compounds were detected by AEDA of SAFE and HS-SPME, and their odor activity values (OAVs) were calculated by dividing their concentration by their odor threshold in water. Among them, ethyl isovalerate, β-damascenone, 3-isobutyl-2-methoxypyrazine (IBMP), and sotolone had the highest OAVs (>1000). In addition, sotolone, methional, β-damascenone, 3-isobutyl-2-methoxypyrazine, ethyl isovalerate, phenylethyl alcohol and linalool had high flavor dilution (FD) factors. Sotolone, β-damascenone and 3-isobutyl-2-methoxypyrazine were identified for the first time in doubanjiang and played significant roles in its aroma quality.

Abstract:Sake is a Japanese traditional alcoholic beverage, which is produced by simultaneous saccharification and alcohol fermentation of polished and steamed rice by Aspergillus oryzae and Saccharomyces cerevisiae. About 300 compounds have been identified in sake, and the contribution of individual components to the sake flavor has been examined at the same time. However, only a few compounds could explain the characteristics alone and most of the attributes still remain unclear. The purpose of this study was to examine the relationship between the component profile and the attributes of sake. Gas chromatography coupled with mass spectrometry (GC/MS)-based non-targeted analysis was employed to obtain the low molecular weight component profile of Japanese sake including both nonvolatile and volatile compounds. Sake attributes and overall quality were assessed by analytical descriptive sensory test and the prediction model of the sensory score from the component profile was constructed by means of orthogonal projections to latent structures (OPLS) regression analysis. Our results showed that 12 sake attributes [ginjo-ka (aroma of premium ginjo sake), grassy/aldehydic odor, sweet aroma/caramel/burnt odor, sulfury odor, sour taste, umami, bitter taste, body, amakara (dryness), aftertaste, pungent/smoothness and appearance] and overall quality were accurately explained by component profiles. In addition, we were able to select statistically significant components according to variable importance on projection (VIP). Our methodology clarified the correlation between sake attribute and 200 low molecular components and presented the importance of each component thus, providing new insights to the flavor study of sake.

Abstract:Background: Soybeans and defatted soybeans, commonly used as protein ingredients, have different flavors of their fermented soy sauce. Clarifying the differences between the two soy sauces, as well as the formation mechanism, is an important prerequisite for improving the flavor of defatted soybean soy sauce. To this goal, the aroma characteristics of two soy sauces and their volatile profiles were compared by sensory evaluation and gas chromatography-mass spectrometry, and eight enzyme activities and volatile profiles of matured koji were determined. Results: Sensory results showed that the acids, fruity and cooked potato-like attributes were higher in whole soybean fermented soy sauce, whereas defatted soybean soy sauce exhibited higher smoky and malty attributes, closely related to the contents of aroma-active compounds in soy sauce, such as isobutyl acetate, 2/3-methylbutanal, acetic acid and 2/3-methylbutanoic acid. The content of most volatiles in the matured kojis showed a consistent trend with that of soy sauce: alcohols, acids, furan(one)s and ketones. Interestingly, acid protease and cellulase activities were 3.3 and 1.6 times higher in the whole soybean koji than in defatted soybean koji, respectively, whereas neutral protease, aminopeptidase, glucoamylase and β-glucosidase were approximately 2.0 times higher in defatted soybean koji. Conclusion: In summary, the flavor differences between soybean and defatted soybean fermented soy sauce were not only caused by the differences in the content of flavor precursors in the materials, but also closely related to the differences in the enzymatic profiles accumulated during the koji-making process. © 2022 Society of Chemical Industry.

Abstract:In the production of Moutai-flavor liquor (MFL), the quality of base liquor significantly impacts the final product's quality. And the low-quality liquor may cause health problems. The different quality grades base liquors were analyzed to investigate the relationship between quality and key compounds in MFL. First, by sensory evaluation, the sauce, floral and fruity, fermented aromas and taste indicators (softness, fullness, harmony, purity and persistence) were positively correlated with the quality grade of base liquors. The electronic nose (E-nose) could distinguish the different quality grade base liquors well. It was found that the higher quality grade of the base liquors, the richer the content of aromatics, alcohols, aldehydes and ketones. Second, differential compounds were identified via untargeted metabolome based on the headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). 16 common differential compounds were shared in the base liquor at different fermentation rounds, including 11 esters, 1 alcohol, 2 aldehydes and 2 ketones. At last, by correlation analysis of differential compounds and sensors responses of E-nose, furfuryl ethyl ether, butanoic acid ethyl ester, isopentyl hexanoate, nonanoic acid ethyl ester and 3-methyl-1-butanol had a significant effect on the response intensity of E-nose sensors. Thus, quantitative data on sensory differences of base liquor can be realized.

Abstract:Volatile flavor compounds created in the mixture of enzymatic hydrolysates of soy sauce residue and defatted soybean by reaction flavor technology (RFT) were analyzed and compared using solid phase micro-extraction/gas chromatography/mass selective detector to develop a seasoning sauce. Using response surface methodology analysis, RFT was performed by adding 0.50% fructose, 0.33% glutamic acid, 0.68% arginine, 0.37% methionine, and 0.86% glycine in the base and reaction conditions at 93°C for 120 min. A total of 57 compounds, 43 in RFT and 45 in control, were detected, including 8 aldehydes and ketones, 6 aromatic hydrocarbons, 3 acids, 12 alcohols, 6 esters, 4 furans, 9 nitrogen-containing compounds, 4 sulfur-containing compounds, and 5 miscellaneous. In RFT samples, aldehydes and ketones, aromatic hydrocarbons, alcohols, esters, and S-containing compounds were significantly increased. Sulfur-containing compounds were increased by 687 fold compared to control samples (<i>P</i><0.05). Among these, the main contributors to the aroma activity of RFT samples were considered to be, with a very low threshold, the newly generated compounds, dimethyl disulfide (roasted onion/garlic-like/meaty odor), dimethyl trisulfide (roasted garlic-like/meaty odor), and methional (roasted potato/potato chip-like odor).
GC-MS Characterization of Volatile Flavor Compounds in Stinky Tofu Brine by Optimization of Headspace Solid-Phase Microextraction Conditions

Abstract:This study optimized the headspace solid phase microextraction (HS-SPME) conditions for the analysis of the volatile flavor compounds of Chinese south stinky tofu brine by gas chromatography-mass spectrometry (GC-MS). The optimum HS-SPME conditions established were as follows: polar column CD-WAX, white 85 μm polyella extractor, extraction temperature 60 °C, equilibrium time 20 min, extraction time 40 min. Under these conditions, a total of 63 volatile flavor compounds in five stinky tofu brines were identified. The offensive odor of the stinky tofu may be derived from some of the volatile flavor compounds such as phenol, p-cresol, 3-methylindole, indole, acetic acid, propionic acid, isobutyric acid, n-butyric acid and 3-methylbutanoic acid. The volatile flavor substances data was examined by principal component analysis (PCA) to visualize the response patterns in the feature space of principal components (PC). PCA analysis results revealed that the Chengshifu brine (STB1) and Baise jingdian brine (STB4) are similar in PC 1, 2, and 3, and the two brines have a similar flavor. Results also indicate that the Huogongdian brine (STB2) and Wangcheng brine (STB3) can be grouped in the same class as they are similar in PC 3. However, PC 1, 2, and 3 of the Luojia brine (STB5) and other brands of brine are different as is the flavor.

Abstract:The volatile compounds in foxtail millet sake were extracted by headspace solid-phase microextraction (HS-SPME) and analyzed using gas chromatography-mass spectroscopy (GC-MS). Different methods of sample preparation were used to optimize this method (SPME fiber types, sample amount, extraction time, extraction temperature, content of NaCl, and rotor speed). For final method of sample preparation, 8 mL of sake was placed in a 15 mL headspace vial with addition of 1.5 g of NaCl; a 50/30 μ m DVB/CAR/PDMS SPME fiber was used for extraction at 50°C for 30 min with 10 rpm continuous stirring. A total of 41 volatile compounds were identified from the sake sample, including 9 esters, 6 alcohols, 4 acids, 4 aldehydes, 9 hydrocarbons, 7 benzene derivatives, and 2 others. The main volatile compounds were ethyl acetate, phenylethyl alcohol, butanedioic acid diethyl ester, and hexadecane. According to their odors active values (OAVs), 10 volatile compounds were established to be odor active compounds and to contribute to the typical foxtail millet sake aroma. Hexanoic acid ethyl ester was the most prominent odor active compound.

Abstract:Japanese soy sauce has become more acceptable by Chinese consumers due to its umami taste. However, the volatile flavor compounds and taste characters have not been fully clarified. This study aimed to explore the flavor characteristics of three kinds of Japanese soy sauce, including Koikuchi Shoyu, Usukuchi Shoyu, and Amakuchi Shoyu. The secret of volatile flavor substances was investigated by Gas Chromatography-Mass Spectrometry (GC-MS) and electronic nose, while taste compounds were investigated by silylation GC-MS and electronic tongue (E-tongue). A total of 173 volatile flavor substances and 160 taste compounds were identified. In addition, 28 aroma compounds with odor activity values (OAV) ≥ 1 were considered as the typical flavors. We found that alcohols and aldehydes were in high abundance in Japanese soy sauce, but only a small portion of pyrazines and esters were. Based on electronic nose and GC-MS analysis, Koikuchi Shoyu gives more contribution to aroma compounds, while Usukuchi Shoyu and Amakuchi Shoyu give the sourness and sweetness features based on E-tongue and silylation GC-MS analysis. In this study, 50 kinds of sugars were detected that contributed to the sweetness of soy sauce. This study will provide new insight into the flavor characteristics of Japanese soy sauce that potentially contribute to the innovation and development of soy sauce.

Abstract:Soy sauce has a rich base of non-volatile substances, but existing studies are insufficient. This study analyzed the metabolites of 19 Chinese commercial soy sauces by UPLC-Orbitrap-MS/MS and GC-Tof-MS derivatization, and detected 674 and 230 kinds of substances, respectively, that could be grouped into 12 different classes of compounds, such as peptides, amino acid derivatives, organic acids, sugars, sugar alcohols, amino acids and so on. For the first time, 215 dipeptides and 91 amino acid derivatives in soy sauce were analyzed in detail and systematically from the perspective of composition and amino acid structure. The flavor profile of soy sauce was obtained by electronic tongue analysis, and orthogonal projections to latent structures (OPLS), random forest (RF), correlation were used to screen potential compounds associated with umami. The intersection of the three methods yielded 9 substances, including 4 reported umami-taste compounds, i.e., Glu, Fru-Glu, Inosine 5'prime-monophosphate (IMP) and Arg-Ser, as well as 5 others that may potentially contribute to umami or be associated with umami-taste producing microorganisms, including His-Asn and Homoserine lactone. This study will advance the understanding of soy sauce metabolites, and provide an in-depth reference for dipeptides and amino acid derivatives in soy sauce.

Abstract:BACKGROUND Considering the important of aroma compounds on the quality of baijiu, stir bar sorptive extraction (SBSE) coupled with gas chromatography-mass spectrometry was applied to the aromas analysis of baijiu (Chinese liquor). RESULTS Based on the results of optimization, a total of 76 volatile compounds were identified from 14 Chinese liquors, including 25 esters, 10 alcohols, 9 aldehydes and ketones, 8 aromatic compounds, 5 furans, 3 nitrogen-containing compounds, 6 acids, 4 phenols, 3 terpenes, 1 sulfide-containing compound, 1 lactone, and 1 acetal. By principal component analysis based on quantification results, 14 Chinese liquor samples could be classified into three groups. CONCLUSION SBSE is a fast, simple, effective, and reliable method for extracting volatile compounds from Chinese liquor. The chemometrics approach revealed that LJ liquor with soy sauce aroma could exhibit more prominent sauce flavor through extending the time of storage, and its peculiar manufacturing practice was responsible for the soy sauce flavor.

Abstract:The aim of the study was to compare the extraction efficiency of headspace and solvent extraction methods on watermelon juice flavor. Solid-phase microextraction (SPME) and solvent-assisted flavor evaporation (SAFE) techniques were employed to extract the volatiles from watermelon juice. Aroma compounds were separated and analyzed through gas chromatography-olfactometry-mass spectrometry (GC-MS/O) equipped with DB-5 and DB-Wax capillary columns. Forty-seven compounds were identified by SPME (30) and SAFE (28). These compounds included 7 aldehydes, 10 olefinic aldehydes, 14 alcohols, 7 sulfur compounds, 5 ketones, and 4 miscellaneous compounds. SPME extracted the saturated aldehydes, olefinic aldehydes, alcohols, and ketones effectively, whereas SAFE exerted better efficiency to extract sulfur compounds. (Z,Z)-3,6-Nonadienal, (Z)-6-nonen-1-ol, (E)-2-hexenal, (E)-2-nonenal, hexanal, nonanal, (Z)-3-nonen-1-ol, and (E)-2-octenal were identified as the key aroma compounds through odor activity value (OAV) and aroma extract dilution analysis (AEDA). Also, the aroma compounds can be extracted comprehensively by the combination of both SPME and SAFE techniques.

Abstract:The variety of processed food products in Japan is vast compared to the rest of the world; processed soybean foods are no exception. Miso, soy sauce, soy milk, and tofu are the main processed soybean product groups. For processed food products that provide a mellow and attractive flavor, the aroma is one of the most important factors controlling the value of the products, and it is a significant challenge to achieve a high quality aroma with good palatability. Therefore, to create attractive processed soybean food products, fundamental knowledge of the key aroma constituents (potent and off-flavor odorants) in processed soybean foods and their chemical and sensory behaviors is strongly needed. In this article, I will introduce part of our research focusing on the potent odorants and their characteristics in miso as well as the topic of enhancing the effect of saltiness due to aroma.

Abstract:In this study, sensory evaluation, electronic nose, and HS-GC-IMS were used to investigate the effects of different seasonings (deionized water, onion, ginger, Sichuan pepper, and mixed seasonings) on the flavor of fish maw. The results showed that the volatile compounds of fish maw soaked in different seasonings were mainly organic sulfides and aromatic compounds. A total of 95 volatile compounds were identified, including 25 aldehydes, 23 olefins, 19 alcohols, 11 esters, 9 ketones, 3 acids, 2 sulfides, 1 furan, 1 ether and 1 ketoxime. Sichuan pepper group and mixed seasoning group had the most significant changes in volatile components, and had the most effective improvement on the flavor of fish maw compared with other groups. These findings will provide reference for producing high quality fish maw and improving its flavor quality. These findings will provide feasible theoretical support for the pretreatment and exploration of fish maw products in the future.

Comparison of Key Aroma Compounds in Five Different Types of Japanese Soy Sauces by Aroma Extract Dilution Analysis (AEDA)

Abstract:An investigation by the aroma extract dilution analysis (AEDA) technique of the aroma concentrate from five different types of Japanese soy sauces, categorized according to Japan Agricultural Standards as Koikuchi Shoyu (KS), Usukuchi Shoyu (US), Tamari Shoyu (TS), Sai-Shikomi Shoyu (SSS), and Shiro Shoyu (SS), revealed 25 key aroma compounds. Among them, 3-ethyl-1,2-cyclopentanedione and 2'-aminoacetophenone were identified in the soy sauces for the first time. Whereas 3-(methylthio)propanal (methional) and 3-hydroxy-4,5-dimethyl-2(5H)-furanone (sotolon) were detected in all of the soy sauce aroma concentrates as having high flavor dilution (FD) factors, 4-ethyl-2-methoxyphenol was detected as having a high FD factor in only four of the soy sauces (KS, US, TS, and SSS). Furthermore, 5(or 2)-ethyl-4-hydroxy-2(or 5)-methyl-3(2H)-furanone (4-HEMF) and 4-hydroxy-2,5-dimethyl-3(2H)-furanone (4-HDMF), which were thought to be the key odorants in KS, were detected in KS, US, TS, and SSS, but the FD factors widely varied among them. The sensory evaluations demonstrated that the aroma descriptions of a cooked potato-like note and a caramel-like/seasoning-like note were evaluated as high scores with no significant differences among the five soy sauces. On the other hand, a burnt/spicy note was evaluated as having high scores in KS, TS, and SSS, but it was evaluated as having a low score in SS. The comparative AEDA experiments and the auxiliary sensory experiments demonstrated that the five different types of Japanese soy sauces varied in their key aroma compounds and aroma characteristics, and the key aroma compounds in KS might not always be highly contributing in the other types of Japanese soy sauces.
