Abstract:The empty cup aroma in soy sauce aroma type baijiu (SSB) is distinct, but the specific compounds responsible for its unique aroma remain unknown. The aroma characteristics of SSB and the empty cup were investigated using molecular sensory science. Fifty-three and 27 aroma active compounds were identified in SSB and empty cup aroma, respectively. AEDA of the empty cup showed ethyl 3-phenylpropanoate, phenylethyl alcohol, sotolon, p-cresol, and 2,3-dimethyl-5-ethyl pyrazine could be the most important aroma contributors to the empty cup aroma due to their high FD values. Sotolon, characterized by its seasoning-like and herbal aroma, was identified as a crucial aroma compound for the empty cup aroma for the first time. Lactic acid was found to decrease the olfactory threshold of sotolon markedly in both 53% ethanol water solution and empty cup, promoting the contribution of sotolon to the empty cup aroma.

Abstract:Soy sauce has a rich base of non-volatile substances, but existing studies are insufficient. This study analyzed the metabolites of 19 Chinese commercial soy sauces by UPLC-Orbitrap-MS/MS and GC-Tof-MS derivatization, and detected 674 and 230 kinds of substances, respectively, that could be grouped into 12 different classes of compounds, such as peptides, amino acid derivatives, organic acids, sugars, sugar alcohols, amino acids and so on. For the first time, 215 dipeptides and 91 amino acid derivatives in soy sauce were analyzed in detail and systematically from the perspective of composition and amino acid structure. The flavor profile of soy sauce was obtained by electronic tongue analysis, and orthogonal projections to latent structures (OPLS), random forest (RF), correlation were used to screen potential compounds associated with umami. The intersection of the three methods yielded 9 substances, including 4 reported umami-taste compounds, i.e., Glu, Fru-Glu, Inosine 5'prime-monophosphate (IMP) and Arg-Ser, as well as 5 others that may potentially contribute to umami or be associated with umami-taste producing microorganisms, including His-Asn and Homoserine lactone. This study will advance the understanding of soy sauce metabolites, and provide an in-depth reference for dipeptides and amino acid derivatives in soy sauce.

Abstract:A method for detection of flavor compounds in soy sauce by SPME-GC was developed.The analysis parameters of extraction time,sample temperature,content of sodium chloride,desorption time were optimized.The optimized method was as follows:10mL soy sauce was placed into 20mL vial and saturated with 1g sodium chloride.The sample was extracted using 85μm PA fiber for 30min at 40℃,then the volatiles were desorbed from the fiber at temperature of 230℃ for 3min.The method is simple,quick,reproducible and sensitive.

Abstract:Despite the fact that soy sauce has been one of the most popular seasonings in Eastern Asia, rapid and holistic methods for its authentication and discrimination are still lacking. This research is sought to develop a novel method for rapid discrimination and amino nitrogen quantitative evaluation of different qualities of soy sauces (Chinese traditional fermentation soy sauce (CTFSS), Japanese traditional fermentation soy sauce (JTFSS), high-salt liquid-state fermentation soy sauce (HLFSS), and low-salt solid-state fermentation soy sauce (LSFSS)) based on tri-step infrared spectroscopy (IR) and partial least squares (PLS). Global nutrition fingerprints (saccharides, proteins, amino acids, and other compositions) of each quality soy sauce at the three levels of enhanced spectral resolution and nutrition profile variations among different soy sauces are synthetically interpreted with fast evaluation of their relative contents based on spectral peak intensity. Complementarily, electronic sensory combined with SPME-GC-MS distinguishes four species of soy sauces effective in smell with 90 discrimination indexes. Specifically, FT-IR peak intensity at about 1409 cm−1 (C–N band) exhibits regular enhancement with the increasing concentration of amino acid nitrogen. Thus, a quantitative prediction model based on IR spectra is established by PLS (R2, 0.9956; RMSEC, 0.233). It has been demonstrated that the developed method is applicable for rapid (5 min) discrimination and quantitative analysis of amino acid nitrogen in soy sauce.

Abstract:Japanese soy sauce has become more acceptable by Chinese consumers due to its umami taste. However, the volatile flavor compounds and taste characters have not been fully clarified. This study aimed to explore the flavor characteristics of three kinds of Japanese soy sauce, including Koikuchi Shoyu, Usukuchi Shoyu, and Amakuchi Shoyu. The secret of volatile flavor substances was investigated by Gas Chromatography-Mass Spectrometry (GC-MS) and electronic nose, while taste compounds were investigated by silylation GC-MS and electronic tongue (E-tongue). A total of 173 volatile flavor substances and 160 taste compounds were identified. In addition, 28 aroma compounds with odor activity values (OAV) ≥ 1 were considered as the typical flavors. We found that alcohols and aldehydes were in high abundance in Japanese soy sauce, but only a small portion of pyrazines and esters were. Based on electronic nose and GC-MS analysis, Koikuchi Shoyu gives more contribution to aroma compounds, while Usukuchi Shoyu and Amakuchi Shoyu give the sourness and sweetness features based on E-tongue and silylation GC-MS analysis. In this study, 50 kinds of sugars were detected that contributed to the sweetness of soy sauce. This study will provide new insight into the flavor characteristics of Japanese soy sauce that potentially contribute to the innovation and development of soy sauce.

Abstract:Soy sauce is a fermented product, and its flavor is a complex mixture of individual senses which, in combination, create a strong palatable condiment for many Eastern and Western dishes. This Review focuses on our existing knowledge of the chemical compounds present in soy sauce and their potential relevance to the flavor profile. Taste is dominated by umami and salty sensations. Free amino acids, nucleotides, and small peptides are among the most important taste-active compounds. Aroma is characterized by caramel-like, floral, smoky, malty, and cooked potato-like odors. Aroma-active volatiles are chemically diverse including acids, alcohols, aldehydes, esters, furanones, pyrazines, and S-compounds. The origin of all compounds relates to both the raw ingredients and starter cultures used as well as the parameters applied during production. We are only just starting to help develop innovative studies where we can combine different analytical platforms and chemometric analysis to link flavor attributes to chemical composition.

Abstract:An investigation by the aroma extract dilution analysis (AEDA) technique of the aroma concentrate from five different types of Japanese soy sauces, categorized according to Japan Agricultural Standards as Koikuchi Shoyu (KS), Usukuchi Shoyu (US), Tamari Shoyu (TS), Sai-Shikomi Shoyu (SSS), and Shiro Shoyu (SS), revealed 25 key aroma compounds. Among them, 3-ethyl-1,2-cyclopentanedione and 2'-aminoacetophenone were identified in the soy sauces for the first time. Whereas 3-(methylthio)propanal (methional) and 3-hydroxy-4,5-dimethyl-2(5H)-furanone (sotolon) were detected in all of the soy sauce aroma concentrates as having high flavor dilution (FD) factors, 4-ethyl-2-methoxyphenol was detected as having a high FD factor in only four of the soy sauces (KS, US, TS, and SSS). Furthermore, 5(or 2)-ethyl-4-hydroxy-2(or 5)-methyl-3(2H)-furanone (4-HEMF) and 4-hydroxy-2,5-dimethyl-3(2H)-furanone (4-HDMF), which were thought to be the key odorants in KS, were detected in KS, US, TS, and SSS, but the FD factors widely varied among them. The sensory evaluations demonstrated that the aroma descriptions of a cooked potato-like note and a caramel-like/seasoning-like note were evaluated as high scores with no significant differences among the five soy sauces. On the other hand, a burnt/spicy note was evaluated as having high scores in KS, TS, and SSS, but it was evaluated as having a low score in SS. The comparative AEDA experiments and the auxiliary sensory experiments demonstrated that the five different types of Japanese soy sauces varied in their key aroma compounds and aroma characteristics, and the key aroma compounds in KS might not always be highly contributing in the other types of Japanese soy sauces.

Abstract:Background: Soybeans and defatted soybeans, commonly used as protein ingredients, have different flavors of their fermented soy sauce. Clarifying the differences between the two soy sauces, as well as the formation mechanism, is an important prerequisite for improving the flavor of defatted soybean soy sauce. To this goal, the aroma characteristics of two soy sauces and their volatile profiles were compared by sensory evaluation and gas chromatography-mass spectrometry, and eight enzyme activities and volatile profiles of matured koji were determined. Results: Sensory results showed that the acids, fruity and cooked potato-like attributes were higher in whole soybean fermented soy sauce, whereas defatted soybean soy sauce exhibited higher smoky and malty attributes, closely related to the contents of aroma-active compounds in soy sauce, such as isobutyl acetate, 2/3-methylbutanal, acetic acid and 2/3-methylbutanoic acid. The content of most volatiles in the matured kojis showed a consistent trend with that of soy sauce: alcohols, acids, furan(one)s and ketones. Interestingly, acid protease and cellulase activities were 3.3 and 1.6 times higher in the whole soybean koji than in defatted soybean koji, respectively, whereas neutral protease, aminopeptidase, glucoamylase and β-glucosidase were approximately 2.0 times higher in defatted soybean koji. Conclusion: In summary, the flavor differences between soybean and defatted soybean fermented soy sauce were not only caused by the differences in the content of flavor precursors in the materials, but also closely related to the differences in the enzymatic profiles accumulated during the koji-making process. © 2022 Society of Chemical Industry.

Abstract:Solid-phase microextraction and liquid–liquid extraction with gas chromatography– olfactometry–mass spectrometry (GC–O–MS) were used to analyze four award-winning high-salt liquid-state soy sauces (HLS). In this study, 110 volatile odor compounds were detected using GC–O–MS. Twenty key odor-active compounds with a flavor dilution factor ≥ 1024 were identified through aroma extract dilution analysis(AEDA), of which 11 compounds had odor activity values > 1; these were methional, 4-hydroxy-2,5-dimethyl-3(2H)-furanone (HDMF), guaiacol, 4-hydroxy-2(or 5)-methyl-5(or 2)-ethyl-3(2 H)-furanone (HEMF), 1-hydroxy-2-propanone, 3-hydroxy-4,5-dimethyl-2(5H)-furanone, 2-ethyl-6-methylpyrazine, 3-methyl-1-butanol, 2,6-dimethylpyrazine, ethyl benzoate, and 4-ethylguaiacol. Sensory evaluation showed that roasted, cooked potato-like, seasoning-like, and smoky odors were the main odor characteristics in the award-winning HLS. Electronic nose analysis indicated that the odor characteristics of the four samples were significantly different and could be distinguished from each other. This study helps to understand and explore the odor characteristics and key odor components of the award-winning HLS.

Decoding temperature-driven microbial community changes and flavor regulation mechanism during winter fermentation of soy sauce

Abstract:The flavor regulation of soy sauce fermented in winter is imminent challenge for the industry, while fermentation temperature is considered as an effective method to fortify soy sauce flavor. Thus, industrial-level fermentation systems with controlled temperature at 30°C (SSCT) and regular temperature (SSRT) in winter were designed to elucidate molecular basis and microbial regulatory mechanism of temperature-controlled flavor enhancement of soy sauce. Sensory evaluation suggested 30°C fermentation enhanced caramel-like, floral, fruity, roasted nut and smoky aroma. A total of 160 volatiles were identified, of which 39 components were evaluated for odor activity value (OAV). Eleven volatiles were determined as the odor markers distinguishing the aroma profiles of SSRT and SSCT, among which 2,5-dimethyl-4-hydroxy-3(2H)-furanone (HDMF, caramel-like), β-damascenone (floral), ethyl 2-methylpropanoate (fruity), ethyl acetate (fruity) and 2/3-methyl-1-butanol (malty, alcoholic) were largely responsible for the flavor enhancement. Moreover, high-throughput sequencing results demonstrated the temperature intervention induced more differential bacterial structure (R = 0.324, P = 0.001) than fungal structure (R = 0.069, P = 0.058). Correlation analysis revealed dominant and low-abundance genus together drove the formation and variation of volatile profile, particularly Weissella, Tetragenococcus, Starmerella and Pediococcus. Representatively, the formation pathways of key aroma substances HDMF and 5-ethyl-4-hydroxy-2-methyl-3(2H)-furanone (HEMF) were elaborated. Both temperature-mediated abiotic reactions and gene functions of microbiota were proposed to favor the yields of HDMF and C5 precursor of HEMF, whereas the small populations of Zygosaccharomyces and insufficient acetaldehyde limited the elevation of the HEMF level through the biosynthesis pathway. This study provided the practical and theoretical basis for the industrial applications of temperature control in soy sauce fermentation.

Abstract:Yongchuan douchi is a traditional fermented soya bean product which is popular in Chinese dishes due to its unique flavor. In this study, the key aroma-active compounds of Yongchuan douchi were characterized by the combined gas chromatography–olfactometry (GC–O) and gas chromatography−mass spectrometry (GC–MS) with sensory evaluation. In total, 49 aroma compounds were sniffed and identified, and 20 of them with high flavor dilution factors (FD) and odor activity values (OAVs) greater than one were screened by applied aroma extract dilution analysis (AEDA) and quantitated analysis. Finally, aroma recombination and omission experiments were performed and 10 aroma-active compounds were thought to have contributed significantly including 2,3-butanedione (butter, cheese), dimethyl trisulfide (garlic-like), acetic acid (pungent sour), acetylpyrazine (popcorn-like), 3-methylvaleric acid (sweaty), 4-methylvaleric acid (sweaty), 2-mehoxyphenol (smoky), maltol (caramel), γ-nonanolactone (coconut-like), eugenol (woody) and phenylacetic acid (flora). In addition, sensory evaluation showed that the flavor profile of Yongchuan douchi mainly consisted of sauce-like, sour, nutty, smoky, caramel and fruity notes.

Abstract:Multispecies microbial mixed fermentation is crucial for the formation of flavors in soy sauce . The present study applied metagenomic strategies to uncover the metabolic network for flavors formation in the non-Cantonese soy sauce in China. Twenty-nine major flavor compounds were identified by GC-MS and HPLC , representing the typical flavor compounds of the non-Cantonese soy sauce, including 3 organic acids, 8 amino acids, and 18 volatile compounds. Metagenomic sequencing and KEGG enrichment analysis revealed the specific functional microbiome, which constituted seven major flavor compounds associated genera, namely Leuconostoc , Staphylococcus , Tetragenococcus , Weissella , Aspergillus , Wickerhamycesa , and Zygosaccharomyces . Aspergillus was primarily responsible for the degradation of raw materials in the early fermentation stage, including the degradation of cellulose and lecithin. It also played an irreplaceable role in the formation of 1-octene-3-ol. Weissella , Staphylococcus, and Leuconostoc played essential metabolic roles in the whole fermentation process, particularly in the metabolism of amino acids and the formation of alcohols. Staphylococcus also participated in the degradation of nitrite/nitrate. Tetragenococcus became the dominant bacteria with the highest relative abundance in the late fermentation stage, which was mainly responsible for the synthesis of acetic acid, lactic acid, and other acids. Additionally, Wickerhamycesa and Zygosaccharomyces also contributed to the formation of alcohols, acids, and aldehydes during the late fermentation stage. This study provides a deeper understanding of the diverse roles in the flavor formation of microorganisms during the traditional condiment soy sauce fermentation process.

Abstract:In the production of Moutai-flavor liquor (MFL), the quality of base liquor significantly impacts the final product's quality. And the low-quality liquor may cause health problems. The different quality grades base liquors were analyzed to investigate the relationship between quality and key compounds in MFL. First, by sensory evaluation, the sauce, floral and fruity, fermented aromas and taste indicators (softness, fullness, harmony, purity and persistence) were positively correlated with the quality grade of base liquors. The electronic nose (E-nose) could distinguish the different quality grade base liquors well. It was found that the higher quality grade of the base liquors, the richer the content of aromatics, alcohols, aldehydes and ketones. Second, differential compounds were identified via untargeted metabolome based on the headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). 16 common differential compounds were shared in the base liquor at different fermentation rounds, including 11 esters, 1 alcohol, 2 aldehydes and 2 ketones. At last, by correlation analysis of differential compounds and sensors responses of E-nose, furfuryl ethyl ether, butanoic acid ethyl ester, isopentyl hexanoate, nonanoic acid ethyl ester and 3-methyl-1-butanol had a significant effect on the response intensity of E-nose sensors. Thus, quantitative data on sensory differences of base liquor can be realized.

Abstract:The co-inoculation fermentation of different microorganisms plays a crucial role in soy sauce fermentation in terms of enriching the composition of the volatile compounds. Therefore, the present study sought to investigate the effect of co-inoculation fermentation of Aspergillus oryzae and Zygosaccharomyces rouxii on the dynamics of volatile compounds in soy sauce using the combination of gas chromatography-mass spectrometry (GC-MS) and multivariate correlation analysis. A total of 124 volatile compounds were identified by A. oryzae and Z. rouxii inoculation alone and co-inoculation, including hydrocarbons, fatty acids, phenols, fatty esters, and benzamides. The content of phenylethyl alcohol, ethyl linoleate, and ethyl palmitate was significantly increased, and the metabolic pathways of multiple volatile compounds were affected in soy sauce inoculated with Z. rouxii . The present study results potentially contribute to the optimization of soy sauce fermentation processes.
Insights into volatile flavor compound variations and characteristic fingerprints in Longpai soy sauce moromi fermentation via HS-GC-IMS and HS-SPME-GC× GC-ToF-MS

Abstract:The variations in flavor substances across the different stages of Longpai soy sauce moromi fermentation were analyzed by HS-GC-IMS combined GC × GC-ToF-MS. A total of 374 VOCs were detected from soy sauce, and the highest content of VOCs were alcohols and esters, respectively. In the early stage fermentation, alcohols and aldehydes were the main flavor compounds. The content of esters, pyrazines, furans and S -containing compounds improved with continuous fermenting process. Based on VOCs, the samples could be delineated by clustering and PCA analysis. Of these, 63 VOCs were regarded as the marker VOCs of soy sauce in different fermentation stage. Alcohol, 1-butanol and 3-methyl-2-butenal provided fruity and mellow aroma at the early stage. Caramel-like, malty and sweet aroma could be enhanced by 3-methyl-2(5H)-furanone, 3-methylbutanol, methional and acetyl valeryl in the middle stage. Furthermore, furfuryl alcohol, 2(5H)-furanone, methyl cyclopentenolone, 2-Ethyl-6-methylpyrazine could be served as marker VOCs of matured Longpai soy sauce.
Community structure of yeast in fermented soy sauce and screening of functional yeast with potential to enhance the soy sauce flavor

Abstract:Yeast plays an important role in the formation of desirable aroma during soy sauce fermentation. In this study, the structure and diversity of yeast communities in seven different soy sauce residues were investigated by ITS sequencing analysis, and then the aroma characteristics of selected yeast species were examined by a combination of gas chromatography-mass spectrometry (GC-MS), headspace solid-phase microextraction (SPME) and liquid-liquid extraction (LLE). A total of 18 yeast genera were identified in seven soy sauce residues. Among them, Candid and Zygosaccharomyces were detected in all samples, followed by Millerozyma, Wickerhamiella, Meyerozyma, Trichosporon and Wickerhamomyces, which were found in more than two-thirds of the samples. Subsequently, eight representative species, isolated from soy sauce residues, were subjected to environmental stress tolerance tests and aroma production tests. Among them, three isolated species were regarded as potential aroma-enhancing microbes in soy sauce. Wickerhamiella versatilis could increase the contents of ethyl ester compounds and alcohols, thereby improving the fruity and alcoholic aroma of soy sauce. Candida sorbosivorans enhanced sweet and caramel-like aroma of soy sauce by producing 4-hydroxy-2,5-dimethyl-3(2H)-furanone (HDMF) and 3-hydroxy-2-methyl-4h-pyran-4-one (maltol). Starmerella etchellsii could enhance the contents of 2,6-dimethylpyrazine, methyl pyrazine and benzeneacetaldehyde. This study is of great significance for the development and application of flavor functional yeasts in soy sauce fermentation.

Abstract:The changes in the concentration of glucagon and insulin in arterial plasma which occur in response to splanchnic nerve stimulation have been investigated in adrenalectomized dogs, cats and sheep. 2. In dogs, stimulation of both splanchnic nerves at a low frequency (2‐0 c/s) for 10 min produced a small but statistically significant increase in plasma glucagon concentration and appeared to inhibit the release of insulin. Stimulation at a higher frequency (10‐0 c/s) produced a much greater increase in plasma glucagon concentration, which was normally accompanied by a rise in plasma glucose concentration. 3. Qualitatively similar changes in plasma glucagon and insulin concentration were observed in both sheep and cats in response to adrenergic stimulation. 4. Intramesenteric infusions of glucagon at a dose of 5‐0 ng kg‐1 min‐1 in dogs produced a comparable rise in plasma glucagon concentration to that elicited by splanchnic nerve stimulation at high frequency (10‐0 c/s) and invariably caused a rise in plasma glucose concentration. 5. In dogs given exogenous glucose, release of glucagon in response to splanchnic nerve stimulation was unaffected by induced hyperglycaemia. Secretion of insulin was partially inhibited by stimulation at 2‐0 c/s and completely suppressed at higher frequency (10‐0c/s). 6. It is concluded that stimulation of the sympathetic innervation to the pancreatic islets, at frequencies within thephysiological range, stimulates the release of glucagon and inhibits that of insulin in each of these species.
