Abstract:The empty cup aroma in soy sauce aroma type baijiu (SSB) is distinct, but the specific compounds responsible for its unique aroma remain unknown. The aroma characteristics of SSB and the empty cup were investigated using molecular sensory science. Fifty-three and 27 aroma active compounds were identified in SSB and empty cup aroma, respectively. AEDA of the empty cup showed ethyl 3-phenylpropanoate, phenylethyl alcohol, sotolon, p-cresol, and 2,3-dimethyl-5-ethyl pyrazine could be the most important aroma contributors to the empty cup aroma due to their high FD values. Sotolon, characterized by its seasoning-like and herbal aroma, was identified as a crucial aroma compound for the empty cup aroma for the first time. Lactic acid was found to decrease the olfactory threshold of sotolon markedly in both 53% ethanol water solution and empty cup, promoting the contribution of sotolon to the empty cup aroma.

Abstract:This study investigated the aroma-active compounds and compared the differences of three different grades of sesame-flavor Baijiu by headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-olfactometry-mass spectrometry (GC-O-MS). A total of 54 aroma-active compounds were detected. Principal component analysis showed that JZ1, JZ2, and JZ3 were well separated from each other. JZ1 as the premium-grade Baijiu had the highest aroma intensities, concentrations. According to aroma intensities and concentrations, dimethyl trisulfide, butanoic acid, phenylacetaldehyde, 2-furylmethanethiol, ethyl hexanoate, 2,6-dimethylpyrazine, etc. could be potentially applied as volatile makers to distinguish the three different grades of sesame-flavor Baijiu as their significant difference (P < 0.05) in three Baijiu samples. Roasted aroma had the significant difference (P < 0.05) in three sample base on aroma profiles. Meanwhile, 2-furylmethanethiol, 2,6-dimethylpyrazine were related to the roasted aroma, they may be had a significant contribution to the differences of three different grades of sesame-flavor Baijiu. Abstract:Soy sauce aroma type baijiu (SSAB) is one of the four basic baijiu flavor types. It has an elegant sauce flavor, empty cup aroma, lasting, mellow and sweet entrance, soft with obvious acidity, delicate taste and long aftertaste. However, to date, no study has identified the key aroma compounds in SSAB. This study introduces SSAB and its production process, and summarizes the research status on flavor compounds. Through analysis and review, some assumptions are made to explain why the key aroma compound of SSAB remains unidentified. We provide insights for the analysis of key aroma compounds in SSAB and lay a theoretical foundation for the improvement and rapid development of SSAB quality.

Abstract:Sauce-aroma Baijiu (SAB) is one of the most famous Baijius in China; SAB has more than 500 aroma compounds in it. However, the key aroma compound in SAB flavor remains unclear. Volatiles play an important role in SAB aroma and are highly correlated to SAB quality. In the present study, 63 volatile compounds were quantified among 66 SAB samples using gas chromatography with flame ionization detector (GC-FID). The authors analyzed odor contributions and volatile compound correlations in two quality groups of SAB samples. Moreover, an odor activity value (OAV) ratio-based random forest classifier was used to explain the volatile compound relationship differentiations between the two quality groups. Our results proved higher quality SABs had richer aromas and indicated a set of fruity-like ethyl valerate, green- and malt-like isobutyraldehyde and malt-like 3-methylbutyraldehyde and sweet-like furfural, had closer co-abundance correlations in higher quality SABs. These results indicated that the aroma and contributions of volatile compounds in SABs should be analyzed not only with compound odor activity values, but also the correlations between different aroma compounds.

Abstract:The complexity of flavor compound interactions significantly influences the overall flavor characteristics of alcoholic beverages. The vast diversity of these compounds, however, makes the screening for interactions both labor-intensive and time-consuming. To address this challenge, our study aimed to develop a rapid and visual approach using Headspace Gas Chromatography-Ion Mobility Spectrometry (GC-IMS), optimized via a Box-Behnken design and Response Surface Methodology (RSM). This method was validated through the investigation of interactions between lactic acid and 13 key flavor compounds in Baijiu. Furthermore, Gas Chromatography-Mass Spectrometry was utilized to quantify the impact of lactic acid on the volatility of these compounds, corroborating the findings from GC-IMS. Additionally, molecular docking techniques were employed to explore the interaction mechanisms between lactic acid and ethyl lactate, 3-hydroxy-2-butanone, and 2-methoxy-4-methylphenol, at the molecular level, offering insights into their potential binding configurations. Our findings offer a streamlined approach for screening flavor compound interactions, enhancing the understanding of flavor formation and facilitating future research and development in complex food matrices.

Abstract:The empty cup aroma is a typical feature of soy sauce aroma type baijiu (SSB). Herein, a novel method based on vacuum-assisted sorbent extraction (VASE) combined with gas chromatography–mass spectrometry (GC–MS) was established for research on volatile compounds in empty cup of SSB. A total of 81 compounds were identified in the empty cup aroma of 8 SSB samples, involving 27 common compounds, and lactic acid was found to have the highest content. High-performance liquid chromatography (HPLC) combined with a chiral column was used to accurately conduct quantitative analysis of L-lactic acid and D-lactic acid, and it was found that the contents of D-lactic acid in all of the 8 SSB empty cup were higher than L-lactic acid. A total of 8 different SSB samples could be distinguished by principal component analysis (PCA) and heatmap analysis, and 7 compounds including L-lactic acid, D-lactic acid, ethyl hexadecanoate, benzeneacetic acid, acetic acid, 1,2-propanediol, hexanoic acid were selected as potential markers of empty cup aroma due to their high variable importance in projection (VIP) values (VIP >1.0).

Abstract:Soy sauce contains a series of biogenic amines (BAs) which is a kind of bioactive organics relating to food quality and safety. High concentration of BAs may lead to remarkable physiological and toxicological influences on human bodies, including hypotension, dizziness, and headaches. Here, we systematically compared the levels of ten main BAs among 53 Chinese commercial soy sauces using an improved high-performance liquid chromatography (HPLC) method. The results showed that the brands and production regions were both important factors accounting for the BAs' content. The contents of Cad, Spm, Try, Phe, His, and Tyr in dark soy sauces were higher than those in light soy sauces. His and Phe in dark soy sauces were 3.7 and 1.84 times higher than in light samples, respectively. Besides, it was surprising that the content of BAs in soy sauces hugely varied from place to place. This work comprehensively compared the content of BAs in soy sauces, showing the relation between soy sauce processes and BAs, offering abundant information for further research on BAs control.

Abstract:The research focused on the distinctive empty cup aroma, with the aim of identifying the key aroma compounds and the formation mechanism of empty cup aroma in soy sauce aroma type baijiu (SSB). The lasting times of SSB is significantly longer than that of other types of baijiu, with an average duration of 28 days. Key compounds such as 2,3-dimethyl-5-ethylpyrazine, phenylethyl alcohol, p-cresol, sotolon, benzeneacetic acid were identified in empty cup aroma due to their highest flavor dilution factor. Molecular dynamics (MD) simulation was performed to study the mechanism of empty cup aroma on the liquid-gas interface and solid-gas interface. The results revealed the existence of hydrogen bonding and van der Waals forces between sotolon and lactic acid, a representative nonvolatile compound, which are speculated to be an important reason for the empty cup aroma.

Abstract:BACKGROUND Considering the important of aroma compounds on the quality of baijiu, stir bar sorptive extraction (SBSE) coupled with gas chromatography-mass spectrometry was applied to the aromas analysis of baijiu (Chinese liquor). RESULTS Based on the results of optimization, a total of 76 volatile compounds were identified from 14 Chinese liquors, including 25 esters, 10 alcohols, 9 aldehydes and ketones, 8 aromatic compounds, 5 furans, 3 nitrogen-containing compounds, 6 acids, 4 phenols, 3 terpenes, 1 sulfide-containing compound, 1 lactone, and 1 acetal. By principal component analysis based on quantification results, 14 Chinese liquor samples could be classified into three groups. CONCLUSION SBSE is a fast, simple, effective, and reliable method for extracting volatile compounds from Chinese liquor. The chemometrics approach revealed that LJ liquor with soy sauce aroma could exhibit more prominent sauce flavor through extending the time of storage, and its peculiar manufacturing practice was responsible for the soy sauce flavor.

Abstract:In the production of Moutai-flavor liquor (MFL), the quality of base liquor significantly impacts the final product's quality. And the low-quality liquor may cause health problems. The different quality grades base liquors were analyzed to investigate the relationship between quality and key compounds in MFL. First, by sensory evaluation, the sauce, floral and fruity, fermented aromas and taste indicators (softness, fullness, harmony, purity and persistence) were positively correlated with the quality grade of base liquors. The electronic nose (E-nose) could distinguish the different quality grade base liquors well. It was found that the higher quality grade of the base liquors, the richer the content of aromatics, alcohols, aldehydes and ketones. Second, differential compounds were identified via untargeted metabolome based on the headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). 16 common differential compounds were shared in the base liquor at different fermentation rounds, including 11 esters, 1 alcohol, 2 aldehydes and 2 ketones. At last, by correlation analysis of differential compounds and sensors responses of E-nose, furfuryl ethyl ether, butanoic acid ethyl ester, isopentyl hexanoate, nonanoic acid ethyl ester and 3-methyl-1-butanol had a significant effect on the response intensity of E-nose sensors. Thus, quantitative data on sensory differences of base liquor can be realized.

Abstract:Aroma is among the principal quality indicators for evaluating Baijiu. The aroma profiles of sauce-flavor Baijiu produced by 10 different manufacturers were determined by GC-IMS. The results showed that GC-IMS could effectively separate the volatile compounds in Baijiu, and a total of 80 consensus volatile compounds were rapidly detected from all samples, among which 29 volatile compounds were identified, including 5 alcohols, 14 esters, 2 acids, 2 ketones, 5 aldehydes, and 1 furan. According to the differences in aroma profile found by multivariate statistical analysis, these sauce-flavor Baijiu produced by 10 different manufacturers can be further divided into three types. The relative odor activity value of the identified volatile compounds indicated that seven volatile compounds contributed most to the aroma of sauce-flavor Baijiu in order of aroma contribution rate, and they were ethyl hexanoate, ethyl pentanoate, ethyl 2-methylbutanoate, ethyl octanoate (also known as octanoic acid ethyl ester), ethyl 3-methylbutanoate, ethyl butanoate, and ethyl isobutyrate. Correspondingly, the main aromas of these sauce-flavor Baijiu produced by 10 different manufacturers were sweet, fruity, alcoholic, etheral, cognac, rummy, and winey. On the one hand, this study proved that GC-IMS is well adapted to the detection of characteristic volatile aroma compounds and trace compounds in Baijiu, which is of positive significance for improving the aroma fingerprint and database of sauce-flavor Baijiu. On the other hand, it also enriched our knowledge of Baijiu and provided references for the evaluation and regulation of the flavor quality of sauce-flavor Baijiu.

Abstract:Laobaigan (LBG) Baijiu is recognized as one of the famous Chinese liquors due to its unique flavor characteristics. However, its key aroma-active volatiles are still unknown. In this study, 414 volatile chemicals in the LBG Baijiu were initially detected by comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GCxGC-TOF/MS). Then, 52 of them were determined by sample dilution analysis (SDA) for screen of aroma-active compounds. Based on their odor-activity values (OAVs) that were determined by the external standard method performed on the GCxGC-TOF/MS, 32 volatile compounds were further recognized as important odorants, which were reconstituted to simulate and validate the aroma profile of the LBG Baijiu. Moreover, omission experiments were conducted to corroborate the importance of key odorants. As a result, nine aroma compounds were finally confirmed as the key aroma-active compounds of the LBG Baijiu.

Abstract:Japanese soy sauce has become more acceptable by Chinese consumers due to its umami taste. However, the volatile flavor compounds and taste characters have not been fully clarified. This study aimed to explore the flavor characteristics of three kinds of Japanese soy sauce, including Koikuchi Shoyu, Usukuchi Shoyu, and Amakuchi Shoyu. The secret of volatile flavor substances was investigated by Gas Chromatography-Mass Spectrometry (GC-MS) and electronic nose, while taste compounds were investigated by silylation GC-MS and electronic tongue (E-tongue). A total of 173 volatile flavor substances and 160 taste compounds were identified. In addition, 28 aroma compounds with odor activity values (OAV) ≥ 1 were considered as the typical flavors. We found that alcohols and aldehydes were in high abundance in Japanese soy sauce, but only a small portion of pyrazines and esters were. Based on electronic nose and GC-MS analysis, Koikuchi Shoyu gives more contribution to aroma compounds, while Usukuchi Shoyu and Amakuchi Shoyu give the sourness and sweetness features based on E-tongue and silylation GC-MS analysis. In this study, 50 kinds of sugars were detected that contributed to the sweetness of soy sauce. This study will provide new insight into the flavor characteristics of Japanese soy sauce that potentially contribute to the innovation and development of soy sauce.

Abstract:Background Laimao Baijiu is a typical soy sauce aroma type baijiu in China. Amino acids are non‐volatile compounds in baijiu and are beneficial to human health. Aroma is one of the important indicators for evaluating the quality of baijiu. The interaction between aroma‐active compounds and non‐volatile compounds can also affect the release of aroma compounds. In this study, we did a comprehensive identification of the active‐aroma compounds and amino acids in Laimao Baijiu by SBSE, GC‐O, GC‐MS, and UPLC‐MS. The interaction between amino acids and key esters was investigated by sensory analysis and partition coefficients. Results A total of 63 aroma compounds and 21 amino acids were identified, 21 esters of which were identified as the major aroma‐active ester compounds with odor activity values (OAVs) ≥ 1. Finally, the sensory analysis revealed that L‐Alanine had a significant effect on the strength of the aromas of esters, suggesting that low concentrations of amino acids were more likely to promote and high concentrations were more likely to inhibit the release of esters. The partition coefficient can be a good explanation for this phenomenon. Conclusion L‐Alanine can significantly affect the aroma intensity of key ester aroma compounds in Laimao Baijiu, and the effects of different concentrations of amino acids are different. This work shows that amino acids, as non‐volatile compounds, have a regulatory effect on the release of aroma compounds in alcoholic beverages, which may provide new technical support for the aroma modulation of alcoholic beverages. Quantitative Analysis of Pyrazines and Their Perceptual Interactions in Soy Sauce Aroma Type Baijiu

Abstract:Pyrazines are important compounds in soy sauce aroma type Baijiu (SSAB). In this work, a total of 16 pyrazines were analyzed using ultra-performance liquid chromatography coupled with tandem mass spectrometry (UPLC–MS/MS) in SSAB. The quantitative results showed that 2,3,5,6-tetramethylpyrazine, 2,6-dimethylpyrazine and 2,3,5-trimethylpyrazine were the three most concentrated pyrazines. The highest odor activity value (OAV) was determined for 2-ethyl-3,5-dimethylpyrazine. Quantitative analysis combined with descriptive sensory analysis revealed that sub-threshold pyrazines (2,3-dimethylpyrazine, 2,3-diethylpyrazine, 2,3-diethyl-5-methylpyrazine and 2-acetyl-3-methylpyrazine) are significantly correlated with the roasted aroma in SSAB. Our study focused on the impact of sub-threshold pyrazines on the perception of roasted aroma in SSAB. The effect of the sub-threshold pyrazines was detected by the addition of various pyrazines in SSAB samples, despite their sub-threshold concentrations. Furthermore, the presence of sub-threshold pyrazines in dilute alcohol solution resulted in a significant reduction in the odor thresholds of supra-threshold pyrazines. Sensory investigation indicated that pyrazines have a synergistic effect on the perception of roasted aroma. The results highlighted the contribution of some pyrazines to the roasted aroma in SSAB despite their sub-threshold concentrations.

Abstract:Soy sauce has a rich base of non-volatile substances, but existing studies are insufficient. This study analyzed the metabolites of 19 Chinese commercial soy sauces by UPLC-Orbitrap-MS/MS and GC-Tof-MS derivatization, and detected 674 and 230 kinds of substances, respectively, that could be grouped into 12 different classes of compounds, such as peptides, amino acid derivatives, organic acids, sugars, sugar alcohols, amino acids and so on. For the first time, 215 dipeptides and 91 amino acid derivatives in soy sauce were analyzed in detail and systematically from the perspective of composition and amino acid structure. The flavor profile of soy sauce was obtained by electronic tongue analysis, and orthogonal projections to latent structures (OPLS), random forest (RF), correlation were used to screen potential compounds associated with umami. The intersection of the three methods yielded 9 substances, including 4 reported umami-taste compounds, i.e., Glu, Fru-Glu, Inosine 5'prime-monophosphate (IMP) and Arg-Ser, as well as 5 others that may potentially contribute to umami or be associated with umami-taste producing microorganisms, including His-Asn and Homoserine lactone. This study will advance the understanding of soy sauce metabolites, and provide an in-depth reference for dipeptides and amino acid derivatives in soy sauce.
Identification of regionalmarkers based on the flavor molecular matrix analysis of sauce-aroma style baijiu

Abstract:Background: Baijiu is a very complex system and its flavor substances are endogenous, influenced by raw materials, starter, production process, production region and other factors. The production region directly affects the composition of flavor substances and quality of baijiu. However, identification of baijiu region is challenging because the corresponding relationship between the production region and baijiu quality is not clear, and the identification of regionalmarkers is indeterminate. In this study, the differences in volatile components of sauce-aroma style baijiu from four representative regions were investigated. Results: A total of 94 volatile compounds were identified in samples tested. Additionally, it was verified that 35 potential flavor substances had important contributions to the aroma of sauce-aroma style baijiu. Meanwhile, nine potential regionalmarkers were screened through multivariate analysis. Further, based on distribution of volatile compounds and the results of sensory evaluation combined with multivariate analysis, a molecular matrix and correlation network were established according to the results of addition experiments, which showed that six substances had a significant impact on the flavor of the tested samples. Conclusion: Six key flavor substances (ethyl octanoate, ethyl 2-methylpropanoate, propyl acetate, ethyl heptanoate, 2-nonanone and butyl hexanoate) were considered as important regionalmarkers to effectively identify the production region of sauce-aroma style baijiu. © 2023 Society of Chemical Industry.
Identification of 2-Hydroxymethyl-3,6-diethyl-5-methylpyrazine as a Key Retronasal Burnt Flavor Compound in Soy Sauce Aroma Type Baijiu Using Sensory-Guided Isolation Assisted by Multivariate Data Analysis.

Abstract:The burnt flavor (overcooked rice flavor) is often retronasally perceived during the soy sauce aroma type Chinese Baijiu (Chinese distilled spirit) tasting and regarded as an important quality marker of the Baijiu. The sensory-guided isolation approach assisted by multivariate data analysis was applied to identify the compounds responsible for the retronasal burnt flavor in the Baijiu. Using two-dimensional offline high performance liquid chromatography separation and taste dilution analysis, a subfraction with the highest burnt flavor intensity was isolated from the Baijiu sample. In the subfraction, six potential flavor compounds were detected by ultra performance liquid chromatography-mass spectrometry. The most probable burnt flavor compound was screened by projection to latent structure regression analysis of the correlation between the potential compounds and their impact on burnt flavor. The target compound was identified as 2-hydroxymethyl-3,6-diethyl-5-methylpyrazine by means of high-resolution mass spectrometry and chemical synthesis. Sensory evaluation revealed a relatively low burnt flavor threshold of 2.89 μmol/L in 53% ethanol aqueous solution. On the basis of the dose-overthreshold factor and addition experiment, this compound was confirmed as the key contributor to the burnt flavor of soy sauce aroma type Baijiu.
