Abstract:Curcumin, commonly used as a coloring agent as well as food additive, has exhibited great therapeutic efficacy for development of functional foods. In order to expand its applications in food, strategies are needed to tackle the poor water-solubility, bioavailability and bioaccessibility of curcumin. Nanoparticles made from individual biomaterials such as proteins, polysaccharides and lipids have been introduced and their properties have been comprehensively studied. Recently, there has been an increasing interest in nanocomplexes made from two or more biomaterials because of their ability to composite the advantages of various components. This review has specific focus on the nanocomplexes fabricated from proteins and polysaccharides. First, complexation methods with or without chemical covalent bond formation are summarized, while different methods with different degrees of complexation are discussed. The encapsulation strategies of curcumin, both traditional and newly invented, are also compared. Then, the structure and physicochemical properties of the resulting binary or ternary nanocomplexes are discussed in detail, as well as a comparison of the delivery efficiency. Last but not least, the current limitations and future directions in the development of curcumin-loaded biopolymer nanoparticles for the design of future food products are presented.

Abstract:ABSTR ACT Owing to the high desorption energy of colloidal particles, Pickering emulsions can effectively protect the encapsulated bioactives. In this study, zein and peach gum polysaccharide (PGP) based biopolymeric nanoparticles were fabricated to stabilize the Pickering emulsion and deliver curcumin. Effects of the PGP addition order during and after anti-solvent precipitation process (zein-PGP nanoparticles or zein/PGP nanoparticles) on the properties of nanoparticles were discussed. The zein-PGP nanoparticles were spherical with mean size of 172.8 nm, while zein/ PGP nanoparticles were rod-like with mean size of 207.5 nm. Both zein-PGP and zein/PGP nanoparticles were negatively charged with the wettability of 93.5 degrees and 90.3 degrees, respectively. Compared to the zein NPs, the zein-PGP and zein/PGP nanoparticles stabilized emulsions had smaller droplet size, better storage and environmental stabilities. The release rates of curcumin in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 23.4% and 26.7% respectively in simulated gastric fluid. The curcumin release rates in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 32.5% and 24.0% respectively in simulated intestinal fluid. These findings might promote an effective delivery system for bioactive food ingredients using zein and PGP based complex nanoparticles.
Fabrication and characterization of soy β-conglycinin-dextran-polyphenol nanocomplexes: Improvement on the antioxidant activity and sustained-release property of curcumin

Abstract:In this study, glycated soy β-conglycinin (β-CG) stabilized curcumin (Cur) composites were fabricated by a unique reversible self-assembly character of β-conglycinin-dextran conjugates (β-CG-DEX). Intrinsic fluorescence and far-UV CD spectra revealed that glycation did not affect the self-assembly property of β-CG in the pH-shifting treatment. The structure of β-CG-DEX could be unfolded at pH 12.0 and reassembled during acidification (from pH 12.0 to 7.0). Meanwhile, β-CG-DEX-3d, which was incubated at 60 °C for 3 days, exhibited a high loading capacity (123.4 mg/g) for curcumin, which far exceeds that (74.90 mg/g) of β-CG-Cur. Moreover, the reassembled β-CG-DEX-3d-Cur showed eminent antioxidant activity of approximately 1.5 times higher than that of free curcumin. During the simulated gastrointestinal condition, compared with β-CG-Cur, β-CG-DEX-3d-Cur nanoparticles showed a more stable and sustained release of curcumin. Thus, β-CG-DEX has immense potential to become a new delivery carrier for hydrophobic food components by means of a self-assembly strategy.
Fabrication and characterization of novel TGase-mediated glycosylated whey protein isolate nanoparticles for curcumin delivery

Abstract:The aim of this study was to fabricate novel transglutaminase (TGase)-mediated glycosylated whey protein isolate (WPI) nanoparticles for the encapsulation and delivery of curcumin. The influences of glycosylation on the physiochemical properties, stability, bioavailability, and antioxidant properties of WPI nanoparticles loaded with curcumin were investigated. Composite nanoparticles exhibited uniform distribution and small particle sizes. The main driving forces for the formation of curcumin nanoparticles were electrostatic interactions, hydrogen bonding, and hydrophobic interactions. The encapsulation and loading efficiency of curcumin after TGase-type glycosylation were significantly increased in comparison to WPI-curcumin nanoparticles. Glycosylated WPI-curcumin nanoparticles had stronger antioxidant properties and stability to resist external environmental changes than WPI-curcumin nanoparticles. In addition, glycosylated WPI-curcumin nanoparticles showed a controlled release and enhanced curcumin bioavailability in vitro gastrointestinal digestion. This study provides novel insights for self-assembled glycosylated protein nanoparticles as delivery systems for protecting hydrophobic nutrients.
Maintained particulate integrity of soy protein nanoparticles during gastrointestinal digestion via genipin crosslinking enhancing stability and bioavailability of curcumin

Abstract:Poor stability during gastrointestinal digestion is a major challenge for the applications of protein-based nanoparticles as oral delivery systems. In this work, genipin was used to crosslink the partially enzymatic hydrolyzed soy protein nanoparticles, aiming to improve their performance in gastrointestinal tract as delivery carrier. Results showed that the obtained genipin-crosslinked soy protein nanoparticles (GSPNPs) were still spherically monodisperse with a diameter around 60 nm. Encapsulation with GSPNPs significantly improved the solubility of curcumin (Cur) and its stability against UV light as well as long-term storage. Compared to those un-crosslinked nanoparticles, particles crosslinked by genipin had a more compact structure less sensitive to ionic effect and digestive enzymes, showing enhanced digestion stability. The well-maintained nanoparticulate structure of GSPNPs further contributed to the enhanced bioaccessibility and facilitated absorption by epithelial cells. Furthermore, in vivo experiment on rats showed that Cur encapsulated in GSPNPs exhibited a slowed down and sustained absorption manner with an 8.11-fold improvement in its bioavailability. These suggested that GSPNPs could be a promising nanocarrier to enhance the bioavailability of functional factors.

Abstract:In this study, a novel and simple strategy is proposed based on 3D network formed by easily blending polysaccharide carrageenan (Car) and fucoidan (Fuc) without a crosslinker. The Fuc/Car dual coating effectively assists the self-assembly of soy protein-isolated (SPI)/curcumin (Cur, C) composite microcapsules (SPI/C) and achieves an excellent curcumin encapsulation efficiency (EE) up to 95.28% with a 4.16% loading capacity (LC) under optimal conditions. The resulting nanocomposites achieved a satisfying redispersibility in aqueous solution and enhanced the water solubility with a lower size dispersity index (PDI) of 0.12 and a larger zeta potential of −29.67 mV. The Fuc/Car double-layer network not only dramatically improved its thermal stability and photostability, but also provided controlled release and enhanced antioxidant activity in in vitro conditions. The underlying mechanism of the self-assembly of the curcumin-loaded nanoparticles was also addressed. The results proved the feasibility of the encapsulation of unstable hydrophobic bioactive substances (curcumin) with the dual anionic polysaccharide Fuc/Car co-stabilized SPI nanoparticles. This study paves the way for an alternative way of developing novel curcumin delivery systems and will have broad prospects in the pharmaceutical industries.

Abstract:In this study, the curcumin was firstly encapsulated in gelatin (GLT) and/or cellulose nanocrystals (CNC) stabilized emulsions, then further mixed with sodium alginate (SA) to form emulsion-filled hydrogel beads loaded with curcumin (Cur). The Cur-loaded emulsions showed a droplet size of 20.3-24.4 μm with a uniform distribution. Introducing CNC and/or SA increased the viscosity of emulsions accompanied by viscoelastic transition, while the modulus was reduced due to destruction of GLT gel. Cur was doubly immobilized in the hydrogel beads with >90 % of encapsulation efficiency. The results of simulated gastrointestinal tract experiments revealed that the beads possessed a good pH sensitivity and controlled release behavior to prolong the retention of Cur in the gastrointestinal tract. After 6 h of UV irradiation, the Cur-loaded emulsion-filled hydrogel beads showed a higher antioxidant activity than that of pure Cur, effectively delaying the photodegradation of Cur. In addition, the beads had better stability in aqueous and acidic environments, which was favorable for prolonging the release of Cur. These results suggest that the emulsion-filled hydrogel beads have great potential for the delivery of lipophilic bioactive molecules.

Abstract:Natural biopolymers are biocompatible and biodegradable, and have been a focus on the research of biologically active carrier materials. In this manuscript, the nanoparticles of ovalbumin/kappa-carrageenan (OVA-CRG) complex were fabricated to encapsulate and protect curcumin under moderate conditions, forming the ovalbuminbccarrageenan/curcumin (OVA-CRG-CUR) complex nanoparticles. The OVA-CRG complex nanoparticles show good curcumin entrapping performance, and the addition of curcumin does not affect the emulsifying stability (ES) and emulsifying activity (EA) of OVA-CRG complex nanoparticles. The maximum ES of OVA-CRG-CUR is reached at 36 h with the value of 84.6 min, and the maximum EA is reached at 48 h with the value of 74.2 m(2)/g. The encapsulated curcumin demonstrates high antioxidation activity, while its thermal and light stabilities of curcumin are both increased. The release behavior of curcumin is further studied in simulated gastric and intestinal fluids. The lower release rate of curcumin in simulated gastric fluid with the final value of 12.9% after 3 h indicates that the OVA-CRG complex nanoparticles are good carriers for improving the stability of curcumin in acidic environment, and are able to enhance the bioavailability of curcumin by a gradual release in intestine. Present researches elucidate that proteins-polysaccharides complex nanoparticles are feasible to solubilize and protect sensitive amphiphilic bioactive compounds and have extensive potential in food and medicinal applications.

Abstract:In the present study, isolated soy protein/sodium alginate (ISP/SA) nanoparticles were fabricated for the target delivery of curcumin. Response surface method of box-Benken design was used to evaluate the effects of ISP (1.0–4.0% (w/v)), SA (0.03–0.07% (w/v)) and CaCl 2 (0.003–0.01% (w/v)) concentration on physicochemical properties of ISP/SA nanoparticles, and selection of optimum nanoparticles. The results showed that the soluble complex of ISP/SA was occurred at pH = 5.8. The optimum concentrations of ISP, SA, and CaCl 2 for the creation of ISP/SA nanoparticle were 1.6% (w/v), 0.069% (w/v), and 0.008% (w/v), respectively. The optimum nanoparticle had a stable dispersion with the zeta potential, %EE, %LC and particle size of −46.5 mV, 99.86%, 0.60%, and 120 nm, respectively. Atomic force microscope images confirmed the formation of spherical particles with a smooth surface. In simulated intestinal conditions, 25% release of curcumin from ISP/SA nanoparticles was reported. The kinetics of curcumin release was consistent with the Peppas model, based on the mechanism of erosion and diffusion. Curcumin-loaded ISP/SA nanoparticles had more thermal stability than curcumin-free (p˂0.05).

Abstract:Curcumin (Cur) is of interest due to its widely reported pharmacology, which is limited by its low water solubility and low bioavailability of clinical use. Here, we report the characterization and application of a multiple stimulus-responsive nanogel loaded with Cur. Sensitive P(AM-co-NIPAM) nanogels for pH, temperature, and reduction stimuli were prepared by reflux precipitation polymerization method using AM and NIPAM as raw materials. DLS and SEM analyses showed that the fabricated nanogels had a uniform size distribution with spherical and nucleated-shell structures. By adjusting the feeding amounts of reactive monomers, cross-linkers, and initiators, a series of monodisperse nanogels with different particle sizes could be obtained. The representative nanogels have different Cur-loaded behaviors at different temperatures, times, and initial concentrations of the drug, as well as a sensitive release to changes in pH, temperature, or reducing conditions. Cur-P(AM-co-NIPAM) nanogels showed superior antioxidant activity over the free Cur in vitro. Meanwhile, it inhibited LPS and H2O2-induced oxidative stress, which could effectively inhibit the cellular production of excessive reactive oxygen species and had a protective effect against LPS-induced cellular damage. This nanogel controlled release system significantly improved the safety limit of Cur without hemolytic behavior and had good cytocompatibility. The successful development of such Cur nanogels will lead to advanced materials that can be functionalized and optimized for the controlled delivery of small molecules or biomolecules for targeted therapeutic and biomedical applications.

Abstract:To prepare fine and stable nanocarriers for curcumin using a highly efficient and convenient method, nanoprecipitation combined with ultrasonication and a high-speed dispersion (US+HSS) method were used to prepare short amylose nanoparticles with pre-formed helical structures. Their morphology, structural characteristics, and embedding effects for curcumin were investigated. The results showed that the optimal ratio of ethanol to short amylose solution and ultrasonic time was 4:1 and 4 min, respectively. The nanoparticles showed a small size (82.43 nm), relatively high loading capacity (11.57 %), and a peak gelatinization temperature of 97.74 °C. Compared to the nanoprecipitation method, the short amylose nanoparticles prepared using the US+HSS method possessed a higher V-type crystalline structure ratio. In addition, the US+HSS method was easier to use to prepare nanoparticles with high stability against NaCl, and the stable nanoparticles showed the best in vitro sustained release effect for curcumin. The Peppas–Sahlin model was the optimal model that matched curcumin release from nanoparticles during digestion.

Abstract:<p>The aim of the present work was to investigate the fabrication, characterization, physical and chemical stability, and re-dispersibility of curcumin-loaded gliadin-rhamnolipid composite nanoparticles, which were prepared by pH-driven method with the novel acidifying agent (phytic acid). During the pH-driven process, the gliadin and rhamnolipid were self-assembly to form composite nanoparticles. Gliadin-rhamnolipid composite nanoparticles showed a higher encapsulation efficiency (98.70%) of curcumin than that (64.70%) of gliadin nanoparticles. Fourier transform infrared spectroscopy and potentiometric analysis revealed that hydrogen bonding, hydrophobic and electrostatic interactions were mainly involved in the formation of curcumin-loaded gliadin-rhamnolipid composite nanoparticles. Compared to the impact of the novel acidifying agent (phytic acid) and the conventional acidifying agent (hydrochloric acid) on the attribute of curcumin-loaded gliadin-rhamnolipid nanoparticles, the nanoparticles prepared with phytic acid had better thermal stability, photostability, physical stability and re-dispersibility than those prepared with hydrochloric acid. The physicochemical properties of the composite nanoparticles also depended on the mass ratio of gliadin to rhamnolipid. Furthermore, rhamnolipid exhibited an obviously positive effect on enhancing the stability of curcumin and the re-dispersibility of gliadin-based nanoparticles. Consequently, by using the pH-driven method with phytic acid, the fabricated gliadin-rhamnolipid composite nanoparticles have potential as a better nano-delivery system for curcumin in functional foods.</p>

Abstract:Curcumin is a naturally occurring hydrophobic polyphenolic compound with a rapid metabolism, poor absorption, and low stability, which severely limits its bioavailability. Here, we employed a starch–protein-based nanoparticle approach to improve the curcumin bioavailability. This study focused on synthesizing nanoparticles with a zein "core" and a carboxymethylated short-chain amylose (CSA) "shell" through anti-solvent precipitation for delivering curcumin. The zein@CSA core–shell nanoparticles were extensively characterized for physicochemical properties, structural integrity, ionic stability, in vitro digestibility, and antioxidant activity. Fourier-transform infrared (FTIR) spectroscopy indicates nanoparticle formation through hydrogen-bonding, hydrophobic, and electrostatic interactions between zein and CSA. Zein@CSA core–shell nanoparticles exhibited enhanced stability in NaCl solution. At a zein-to-CSA ratio of 1:1.25, only 15.7% curcumin was released after 90 min of gastric digestion, and 66% was released in the intestine after 240 min, demonstrating a notable sustained release effect. Furthermore, these nanoparticles increased the scavenging capacity of the 1,1-diphenyl-2-picrylhydrazyl (DPPH•) free radical compared to those composed solely of zein and were essentially nontoxic to Caco-2 cells. This research offers valuable insights into curcumin encapsulation and delivery using zein@CSA core–shell nanoparticles.

Abstract:Curcumin (CUR) has known to have poor oral bioavailability due to its poor aqueous solubility and stability in gastrointestinal fluid condition as well as its limited permeability which greatly limits its applications in food and medical fields. The present study reported the development of CUR nanocrystal suspension (CUR-NS) for enhanced bioavailability, stabilized with octenyl succinic anhydride modified starch (OSA-S), a naturally renewable and biocompatible biomolecule. This preparation was achieved via an antisolvent precipitation combined with ultrasonic dispersion method, following which the suspension was freeze-dried to form powder CUR nanocrystals (CUR-NCs). Physicochemical properties of the prepared CUR-NS and CUR-NCs powders were extensively characterized. The particle size of the nanocrystals was found to be around 322.8 ± 15.7 nm with a small polydispersity index (PDI) of about 0.230 ± 0.039. Rod like crystal shape was observed by the SEM. Results from powder X-ray diffraction (pXRD) and differential scanning calorimetry (DSC) indicated that the crystal form of CUR in CUR-NCs was a monoclinic polymorph Form 2. Stability results indicated that CUR-NCs possessed an excellent storage-, thermal-, and photo-stabilities. CUR-NCs had a significantly higher dissolution rate, a 68-fold increase in its solubility than that of raw CUR. The cytotoxicity assay demonstrated that CUR-NCs exhibited lower toxicity towards normal human kidney epithelial cells compared to raw CURs. Parallel artificial membrane permeability assay (PAMPA) characterization confirmed that the flux of CUR was significantly enhanced after forming nanocrystals with OSA-S, probably resulting from its increased solubility and dissolution rate. The study showed that OSA-S could effectively stabilize CUR-NCs, and CUR-NCs developed in the current work exhibited significant enhancement in the stability and dissolution rate leading to an improved bioavailability which might be helpful for further development of CUR based products. Abstract:Curcumin (Cur) is a phytochemical with various beneficial properties, including antioxidant, anti-inflammatory, and anticancer activities. However, its hydrophobicity, poor bioavailability, and stability limit its application in many biological approaches. In this study, a novel amphiphilic chitosan wall material was synthesized. The process was carried out via grafting chitosan with succinic anhydride (SA) as a hydrophilic group and deoxycholic acid (DA) as a hydrophobic group; 1H-NMR, FTIR, and XRD were employed to characterize the amphiphilic chitosan (CS—SA—DA). Using a low-cost, inorganic solvent-based procedure, CS—SA—DA was self-assembled to load Cur nanomicelles. This amphiphilic polymer formed self-assembled micelles with a core–shell structure and a critical micelle concentration (CMC) of 0.093 mg·mL−1. Cur-loaded nanomicelles were prepared by self-assembly and characterized by the Nano Particle Size Potential Analyzer and transmission electron microscopy (TEM). The mean particle size of the spherical Cur-loaded micelles was 770 nm. The drug entrapment efficiency and loading capacities were up to 80.80 ± 0.99% and 19.02 ± 0.46%, respectively. The in vitro release profiles of curcumin from micelles showed a constant release of the active drug molecule. Cytotoxicity studies and toxicity tests for zebrafish exhibited the comparable efficacy and safety of this delivery system. Moreover, the results showed that the entrapment of curcumin in micelles improves its stability, antioxidant, and anti-inflammatory activity.

Abstract:In this work, ovalbumin-dextran nanogels were fabricated via the Maillard reaction followed by a heat-gelation process and their potential to improve curcumin bioavailability was investigated using an in vitro gastrointestinal tract. The result of electrophoresis, conjugation efficiency and yield, and far-UV circular dichroism verified the covalent attachment of dextran to ovalbumin by the Maillard reaction. The conjugates were then heated at the isoelectric point of ovalbumin to give stable nanogels. The transmission electron microscopy observation suggested that nanogels displayed a spherical structure. Curcumin was loaded into nanogels by a pH-driven method and the incorporation of curcumin affected the morphology of nanogels marginally. The transformation of curcumin in nanogels was significantly higher than that in ovalbumin nanoparticle. On the other hand, the bioaccessibility was similar for curcumin in nanogels and nanoparticle. The ovalbumin-dextran nanogels hold the potential as delivery systems for curcumin to fortify functional foods and beverages.

Abstract:Curcumin is an extract of turmeric (Curcuma longa) which possesses anti-inflammatory, anti-cancer and wound-healing effects and has been used as an active compound in biomedical research for many years. However, its poor solubility presents challenges for its use in drug delivery systems. A modified nanogel delivery system, with PNIPAM and β-cyclodextrin grafted onto hyaluronic acid (PNCDHA), was utilized to enhance the solubility. The polymer was characterized by NMR, and the inclusion complex between curcumin and β-cyclodextrin was confirmed by FTIR. The potential of this PNCDHA polymer complex as a drug delivery vehicle was supported by a curcumin encapsulation efficiency of 93.14 ± 5.6% and the release of encapsulated curcumin at 37 °C. At a concentration of 0.5% w/v in water, PNCDHA nanogels were biocompatible with fibroblast cell line (L929) up to a curcumin concentration of 50 μM. There was a direct concentration between curcumin loading and cellular internalization. A more detailed study of the cellular internalization of PNCDHA nanogel should be considered in order to clarify cellular delivery mechanisms and to assess how its viability as a carrier may be optimized.

Abstract:Curcumin has a broad-spectrum anti-tumor effect and has no toxic side effects. However, the unique diketone structure of curcumin will undergo diketo-enol tautomerism under different acid-base conditions, resulting in its instability under physiological conditions. In addition, the low biocompatibility and absorption rate of curcumin also limit the use of curcumin drugs. In this paper, curcumin was modified by substitution of acryloyl and acrylsulfonyl groups, and four kinds of nanoparticles with regular morphology were prepared using non-toxic and non-irritating acrylic resin as coating material to improve the stability and bioavailability of the compounds. Zeta potential testing shows that the composites surface carries positive charges and have good stability. In the release experiment, four complexes have the potential for slow and controlled release. Imaging of Hela cells with different channels was performed, and the imaging results showed that the complexes could enter the cells and be absorbed by them, demonstrating good imaging performance. MTT experiments have shown that the complexes have certain anti-tumor activity and low cytotoxicity. In general, the complexes synthesized in this paper have potential in the field of drug fluorescence imaging detection. At the same time, this experiment provides a new idea for the design of slow and controlled release of drugs.
