Abstract:The aim of this study is to fabricate novel chitosan-based film loaded with curcumin loaded metal-phenolic nanoparticles (MPN@Cur NPs) as an active agent for food preservation. The average particle size and zeta-potential of the MPN@Cur NPs were determined to be 205.9 nm and -26.8 mV, respectively. The impact of incorporating MPN@Cur NPs upon the morphology, microstructure, physicochemical characteristics, and functional activities of CS-based films were investigated through scanning electron microscope, infrared spectrometer, X-ray diffractometer, ultraviolet spectrometer, antioxidant activity and antibacterial activity analysis. The outcomes indicated that MPN@Cur NPs exhibited good mechanical compatibility with chitosan, effectively filling voids within the film matrix and significantly enhancing its mechanical characteristics (e.g., elongation at break improved from 22.6% to 37.3%; tensile strength improved from 31.1 MPa to 43.6 MPa). Additionally, films incorporating MPN@Cur NPs displayed superior barrier properties (e.g., water vapor and oxygen transmission), enhanced thermal stability, and improved UV-blocking capabilities compared to pure CS films (transmittance for UV decreased from 93.6% to 35.3%). Moreover, these films exhibited a marked increase in antibacterial efficacy against the foodborne pathogens Staphylococcus aureus and Escherichia coli, as well as elevated antioxidant activity (e.g., scavenging capability for DPPH increased from 9.22% to 85.19%, ABTS increased from 8.62% to 80.02%). Ultimately, the CS-based film significantly prolonged the shelf life of milk stored at 25 degrees C. Therefore, nanocomposite films provide an attractive and promising pathway for the high value-added commercial use of chitosan in food conservation.
Protein-polysaccharide nanocomplexes as nanocarriers for delivery of curcumin: a comprehensive review on preparation methods and encapsulation mechanisms

Abstract:Curcumin, commonly used as a coloring agent as well as food additive, has exhibited great therapeutic efficacy for development of functional foods. In order to expand its applications in food, strategies are needed to tackle the poor water-solubility, bioavailability and bioaccessibility of curcumin. Nanoparticles made from individual biomaterials such as proteins, polysaccharides and lipids have been introduced and their properties have been comprehensively studied. Recently, there has been an increasing interest in nanocomplexes made from two or more biomaterials because of their ability to composite the advantages of various components. This review has specific focus on the nanocomplexes fabricated from proteins and polysaccharides. First, complexation methods with or without chemical covalent bond formation are summarized, while different methods with different degrees of complexation are discussed. The encapsulation strategies of curcumin, both traditional and newly invented, are also compared. Then, the structure and physicochemical properties of the resulting binary or ternary nanocomplexes are discussed in detail, as well as a comparison of the delivery efficiency. Last but not least, the current limitations and future directions in the development of curcumin-loaded biopolymer nanoparticles for the design of future food products are presented.
New Insights into Ultrasound-Assisted Noncovalent Nanocomplexes of Β-Lactoglobulin and Neochlorogenic Acid/cryptochlorogenic Acid and Its Potential Application for Curcumin Loading

Abstract:The cross-linking sites and structure of protein–polyphenol complexes are susceptible to the type, structure, weight of polyphenols under nonthermal process. The low bioavailability and poor gastrointestinal instability of curcumin (CUR) hampers its application. Hence, changes in binding mechanism, structural and functional properties between β-lactoglobulin (β-LG) with two different configurations of chlorogenic acids (neochlorogenic acids (3-CQA) and cryptochlorogenic acids (4-CQA) by non-covalent binding under ultrasonic treatment, and the potential capacity for loading CUR were researched. The binding affinity scores of β-LG-4CQA was −7.1 kcal/mol. It is higher than β-LG-3CQA (−6.8 kcal/mol), which implied that the interaction between β-LG and 4-CQA was stronger. Circular dichroism calculations showed that the sonicated complex of the β-LG and 4-CQA with a decreased content of α-helices by 5.4 %, β-sheets by 4.6 %, and an increased content of irregular curls by 8.4 % (p < 0.05). The result demonstrated ultrasound and the binding of β-LG to 3/4-CQA improved the hydrophilicity, thermal stability, and antioxidant property of β-LG. Furthermore, the embedding rate of CUR in the ultrasound-assisted β-LG-4-CQA complex could reach 71.56 %. Consistent with the structural characterization results, the CUR release rate of ULG-4-CQA + CUR complex reached 17.36 % in simulated intestinal digestion, which was 8.09 % higher than free CUR. Indicating that after embedding with protein–polyphenol complexes, the stability and bioaccessibility of CUR was improved. This study reveals the potential application of ultrasound-assisted protein–polyphenol complexes for loading CUR.

Abstract:ABSTR ACT Owing to the high desorption energy of colloidal particles, Pickering emulsions can effectively protect the encapsulated bioactives. In this study, zein and peach gum polysaccharide (PGP) based biopolymeric nanoparticles were fabricated to stabilize the Pickering emulsion and deliver curcumin. Effects of the PGP addition order during and after anti-solvent precipitation process (zein-PGP nanoparticles or zein/PGP nanoparticles) on the properties of nanoparticles were discussed. The zein-PGP nanoparticles were spherical with mean size of 172.8 nm, while zein/ PGP nanoparticles were rod-like with mean size of 207.5 nm. Both zein-PGP and zein/PGP nanoparticles were negatively charged with the wettability of 93.5 degrees and 90.3 degrees, respectively. Compared to the zein NPs, the zein-PGP and zein/PGP nanoparticles stabilized emulsions had smaller droplet size, better storage and environmental stabilities. The release rates of curcumin in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 23.4% and 26.7% respectively in simulated gastric fluid. The curcumin release rates in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 32.5% and 24.0% respectively in simulated intestinal fluid. These findings might promote an effective delivery system for bioactive food ingredients using zein and PGP based complex nanoparticles.

Abstract:The practical application of curcumin (CUR) is greatly limited due to its instability, high hydrophobicity, low bioavailability, and inability to cross the mucosal barrier of gastrointestinal tract. To overcome these disadvantages, several delivery systems have been explored to formulate CUR for oral administration. Nanoparticles (NPs) can significantly enhance oral absorption, bioavailability and therapeutic efficacy of drug, however, NPs are limited by the gastrointestinal degradation, mucosal and epithelial barriers. A novel amphiphilic quaternary ammonium chitosan (N-2-HACC) based NP delivery carrier was prepared using palmitic acid (PA) to encapsulate CUR. Palmitoyl chitosan (PA-N-2-HACC) was characterized including FT-IR, 1H NMR, TGA, and CAC. The particle size of PA-N-2-HACC NPs and PA-N-2-HACC NPs loaded with CUR (CUR@PA-N-2-HACC NPs) was 231.6 ± 9.24 nm and 264.5 ± 4.31 nm. The encapsulation efficiency and loading capacity of CUR@PA-N-2-HACC NPs was 75.43 ± 1.25 % and 6.81 ± 0.16 %. CUR@PA-N-2-HACC NPs exhibited sustained and controlled release. Compared with the CUR, minimum inhibitory concentration of the CUR@PA-N-2-HACC NPs against Escherichia coli, Staphylococcus aureus and Candida albicans was reduced by 4.2 times, 1.6 times and 4.6 times, respectively. Moreover, the antioxidant activity of CUR@PA-N-2-HACC NPs was dose-dependent and higher than that of free CUR. The PA-N-2-HACC NPs show little toxicity and are a promising delivery system for encapsulating hydrophobic drugs.

Abstract:This work presents the fabrication of ternary nanoparticles (Z/S/C NPs) comprising zein (Z), soy protein isolate (SPI) and carboxymethylcellulose sodium (CMC-Na) through a pH-driven method. The results showed that the smallest particle size (71.41 nm) and the most stable zeta potential, measuring −49.97 mV, were achieved with the following ratio of ternary nanoparticles Z/SPI/CMC-Na (2:3:3). The surface morphology of the nanoparticles was further analyzed using transmission electron microscopy, and the synthesized nanoparticles were utilized to encapsulate curcumin (Cur), a hydrophobic, bioactive compound. The nanoparticles were characterized using a particle size analyzer, infrared spectroscopy, and X-ray diffraction (XRD) techniques. The results revealed that the formation of nanoparticles and the encapsulation of Cur were driven by electrostatic, hydrogen-bonding and hydrophobic interactions. The drug loading efficiency (EE%) of Z/S/C-cur nanoparticles reached 90.90%. The Z/S/C ternary nanoparticles demonstrated enhanced storage stability, photostability and simulated the gastrointestinal digestion of Cur. The release of Cur and variations in the particle size of nanoparticles were investigated across different stages of digestion. The biocompatibility of the Z/S/C ternary nanoparticles was assessed by conducting cell viability assays on HepG2 and L-O2 cells, which showed no signs of cytotoxicity. These results suggested that the ternary composite nanoparticles have potential in delivering nutritional foods and health-promoting bioactive substances.

Abstract:Curcumin (Cur) is a bioactive substance, but its instability limits its use in the food system. Thus, a new polysaccharide-based coating is developed by combining quaternized chitosan (QCS) with sodium alginate (SA) in this study to improve the bioaccessibility and stability of Cur. The microstructure of the emulsion was characterized by fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and low-field pulsed nuclear magnetic resonance imaging (LF-NMR). The thermal, ionic strength, and pH stability of emulsions were measured. Results indicate that QCS/SA (1:1)-Cur is more stable than other three nanoemulsion systems and the encapsulation rate of which reaches 90.62 %. It also shows higher stability in gastric fluid, excellent bioaccessibility and FFAs in intestinal fluid, as well as significant antibacterial property, antioxidant activity and water dispersibility. This study introduces a new method for obtaining stable, functional Cur-coated systems for food applications.
Encapsulation of curcumin in ZEIN-HTCC complexes: Physicochemical characterization, in vitro sustained release behavior and encapsulation mechanism

Abstract:In order to improve the water solubility and stability of curcumin (CUR) and achieve its sustained release, the ZEIN–N-(2-hydroxy) propyl-3-trimethylammonium chitosan chloride (HTCC) complexes were designed to encapsulate CUR using an antisolvent precipitation (ASP) method. The encapsulation efficiency (EE) and loading capacity (LC) of curcumin in ZEIN-HTCC complexes were 92.2% and 0.6%, respectively. After encapsulation, the resistance of curcumin to thermal, natural light and UV irradiation, as well as the antioxidative activity were improved significantly (P < 0.05). ZEIN-HTCC-CUR complexes exhibited good redispersibility and enhanced the water solubility of curcumin. In vitro simulated digestion experiments clarified that the ZEIN-HTCC complexes can control the sustained release behavior of curcumin, and the release kinetics can be fitted by the Higuchi model. Results taken from fluorescence spectra and Fourier transform infrared spectroscopy (FTIR) indicated that hydrophobic interaction, hydrogen bonding and electrostatic interactions were the main driving forces to form the ZEIN-HTCC-CUR complexes. The X-ray diffraction (XRD) results revealed that the encapsulated curcumin was in an amorphous state. The information provided by this study should be of interest to food industry to fabricate robust nanoscale delivery systems with ZEIN-HTCC complexes for CUR and other hydrophobic bioactive compounds.

Abstract:Curcumin is a naturally occurring hydrophobic polyphenolic compound with a rapid metabolism, poor absorption, and low stability, which severely limits its bioavailability. Here, we employed a starch–protein-based nanoparticle approach to improve the curcumin bioavailability. This study focused on synthesizing nanoparticles with a zein "core" and a carboxymethylated short-chain amylose (CSA) "shell" through anti-solvent precipitation for delivering curcumin. The zein@CSA core–shell nanoparticles were extensively characterized for physicochemical properties, structural integrity, ionic stability, in vitro digestibility, and antioxidant activity. Fourier-transform infrared (FTIR) spectroscopy indicates nanoparticle formation through hydrogen-bonding, hydrophobic, and electrostatic interactions between zein and CSA. Zein@CSA core–shell nanoparticles exhibited enhanced stability in NaCl solution. At a zein-to-CSA ratio of 1:1.25, only 15.7% curcumin was released after 90 min of gastric digestion, and 66% was released in the intestine after 240 min, demonstrating a notable sustained release effect. Furthermore, these nanoparticles increased the scavenging capacity of the 1,1-diphenyl-2-picrylhydrazyl (DPPH•) free radical compared to those composed solely of zein and were essentially nontoxic to Caco-2 cells. This research offers valuable insights into curcumin encapsulation and delivery using zein@CSA core–shell nanoparticles.

Abstract:We successfully designed curcumin (Cur)-loaded composite nanoparticles consisting of high-hydrostatic-pressure-treated (HHP-treated) zein and pectin with a pressure of 150 MPa (zein-150 MPa-P-Cur), showing nano-spherical structure with high zeta-potential (-36.72 ± 1.14 mV) and encapsulation efficiency (95.64 ± 1.23 %). We investigated the interaction mechanism of the components in zein-150 MPa-P-Cur using fluorescence spectroscopy, molecular dynamics simulation, Fourier-transform infrared spectrometry and scanning electron microscopy techniques. Compared with zein-P-Cur, the binding sites and binding energy (-53.68 kcal/mol vs. - 44.22 kcal/mol) of HHP-treated zein and Cur were increased. Meanwhile, the interaction force among HHP-treated zein, pectin, and Cur was significantly enhanced, which formed a tighter and more stable particle structure to further improve package performance. Additionally, Cur showed the best chemical stability in zein-150 MPa-P-Cur. And the bioavailability of Cur was increased to 65.53 ± 1.70 %. Collectively, composite nanoparticles based on HHP-treated zein and pectin could be used as a promising Cur delivery system.

Abstract:Curcumin (Cur) is a phytochemical with various beneficial properties, including antioxidant, anti-inflammatory, and anticancer activities. However, its hydrophobicity, poor bioavailability, and stability limit its application in many biological approaches. In this study, a novel amphiphilic chitosan wall material was synthesized. The process was carried out via grafting chitosan with succinic anhydride (SA) as a hydrophilic group and deoxycholic acid (DA) as a hydrophobic group; 1H-NMR, FTIR, and XRD were employed to characterize the amphiphilic chitosan (CS—SA—DA). Using a low-cost, inorganic solvent-based procedure, CS—SA—DA was self-assembled to load Cur nanomicelles. This amphiphilic polymer formed self-assembled micelles with a core–shell structure and a critical micelle concentration (CMC) of 0.093 mg·mL−1. Cur-loaded nanomicelles were prepared by self-assembly and characterized by the Nano Particle Size Potential Analyzer and transmission electron microscopy (TEM). The mean particle size of the spherical Cur-loaded micelles was 770 nm. The drug entrapment efficiency and loading capacities were up to 80.80 ± 0.99% and 19.02 ± 0.46%, respectively. The in vitro release profiles of curcumin from micelles showed a constant release of the active drug molecule. Cytotoxicity studies and toxicity tests for zebrafish exhibited the comparable efficacy and safety of this delivery system. Moreover, the results showed that the entrapment of curcumin in micelles improves its stability, antioxidant, and anti-inflammatory activity.

Abstract:Curcumin (CUR) has known to have poor oral bioavailability due to its poor aqueous solubility and stability in gastrointestinal fluid condition as well as its limited permeability which greatly limits its applications in food and medical fields. The present study reported the development of CUR nanocrystal suspension (CUR-NS) for enhanced bioavailability, stabilized with octenyl succinic anhydride modified starch (OSA-S), a naturally renewable and biocompatible biomolecule. This preparation was achieved via an antisolvent precipitation combined with ultrasonic dispersion method, following which the suspension was freeze-dried to form powder CUR nanocrystals (CUR-NCs). Physicochemical properties of the prepared CUR-NS and CUR-NCs powders were extensively characterized. The particle size of the nanocrystals was found to be around 322.8 ± 15.7 nm with a small polydispersity index (PDI) of about 0.230 ± 0.039. Rod like crystal shape was observed by the SEM. Results from powder X-ray diffraction (pXRD) and differential scanning calorimetry (DSC) indicated that the crystal form of CUR in CUR-NCs was a monoclinic polymorph Form 2. Stability results indicated that CUR-NCs possessed an excellent storage-, thermal-, and photo-stabilities. CUR-NCs had a significantly higher dissolution rate, a 68-fold increase in its solubility than that of raw CUR. The cytotoxicity assay demonstrated that CUR-NCs exhibited lower toxicity towards normal human kidney epithelial cells compared to raw CURs. Parallel artificial membrane permeability assay (PAMPA) characterization confirmed that the flux of CUR was significantly enhanced after forming nanocrystals with OSA-S, probably resulting from its increased solubility and dissolution rate. The study showed that OSA-S could effectively stabilize CUR-NCs, and CUR-NCs developed in the current work exhibited significant enhancement in the stability and dissolution rate leading to an improved bioavailability which might be helpful for further development of CUR based products. Fabrication of Ovalbumin/κ-Carrageenan Complex Nanoparticles As a Novel Carrier for Curcumin Delivery

Abstract:Natural biopolymers are biocompatible and biodegradable, and have been a focus on the research of biologically active carrier materials. In this manuscript, the nanoparticles of ovalbumin/kappa-carrageenan (OVA-CRG) complex were fabricated to encapsulate and protect curcumin under moderate conditions, forming the ovalbuminbccarrageenan/curcumin (OVA-CRG-CUR) complex nanoparticles. The OVA-CRG complex nanoparticles show good curcumin entrapping performance, and the addition of curcumin does not affect the emulsifying stability (ES) and emulsifying activity (EA) of OVA-CRG complex nanoparticles. The maximum ES of OVA-CRG-CUR is reached at 36 h with the value of 84.6 min, and the maximum EA is reached at 48 h with the value of 74.2 m(2)/g. The encapsulated curcumin demonstrates high antioxidation activity, while its thermal and light stabilities of curcumin are both increased. The release behavior of curcumin is further studied in simulated gastric and intestinal fluids. The lower release rate of curcumin in simulated gastric fluid with the final value of 12.9% after 3 h indicates that the OVA-CRG complex nanoparticles are good carriers for improving the stability of curcumin in acidic environment, and are able to enhance the bioavailability of curcumin by a gradual release in intestine. Present researches elucidate that proteins-polysaccharides complex nanoparticles are feasible to solubilize and protect sensitive amphiphilic bioactive compounds and have extensive potential in food and medicinal applications.

A simple ionic gelation method was conducted to formulate Q-AmCs nanoparticles (NPs), using different ratios of sodium tripolyphosphate (TPP) as an ionic crosslinker. Various characterization tools were employed to investigate the structure, surface morphology, and thermal properties of the formulated nanoparticles. The formulated Q-AmCs NPs displayed a smaller particle size of 162 ± 9.10 nm, and higher surface positive charges, with a maximum potential of +48.3 mV, compared to native aminated chitosan (AmCs) NPs (231 ± 7.14 nm, +32.8 mV). The Cur-drug encapsulation efficiency was greatly improved and reached a maximum value of 94.4 ± 0.91%, compared to 75.0 ± 1.13% for AmCs NPs. Moreover, the in vitro Cur-release profile was investigated under the conditions of simulated gastric fluid [SGF; pH 1.2] and simulated colon fluid [SCF; pH 7.4]. For Q-AmCs NPs, the Cur-release rate was meaningfully decreased, and recorded a cumulative release value of 54.0% at pH 7.4, compared to 73.0% for AmCs NPs. The formulated nanoparticles exhibited acceptable biocompatibility and biodegradability. These findings emphasize that Q-AmCs NPs have an outstanding potential for the delivery and slow release of anticancer drugs.
Preparation and characterization of dextran-zein-curcumin nanoconjugate for enhancement of curcumin bioactivity

Abstract:Curcumin is one of the most important polyphenolic nutrients in pharmaceutical industries. Unfortunately, its poor solubility and bioavailability have hampered its clinical application. To improve curcumin solubility and bioavailability, a natural nanocarrier made from protein-polysaccharide conjugate has been developed. Following antisolvent precipitation method, zein (Z) nanoparticles coated with dextran sulphate (DS) have been fabricated as curcumin (C) nanocarrier (DSZCNPs). The physicochemical properties of the nanoconjugate were measured using different techniques. Morphologically, DSZCNPs appeared spherical and monodispersed in scanning electron microscope (SEM) and transmission electron microscope (TEM) images. Curcumin encapsulation efficiency was ≈ 96%. DSZCNPs size was 180 nm and the polydispersity index value (PDI) 0.28. Zeta potential for DSZCNPs was −28.5 mV. DSZCNPs showed stability either for shelf storage (100 days) or at different pHs. Furthermore, DSZCNPs protected zein nanoparticles degradation in gastric environment and achieved controlled curcumin release in intestinal environment. DSZCNPs greatly enhanced the antioxidant activity of curcumin as demonstrated by DPPH assay. DSZCNPs had significant results in the reduction of colony forming unit (CFU%) against the tested microbes when compared with free curcumin. Also, the anticancer activity of DSZCNPs and free curcumin against hepatocellular carcinoma cells (HepG2) were assessed by MTT assay. IC 50 for DSZCNPs was 13 μg/ml compared to 50 μg/ml for free curcumin indicating the therapeutic impact of DSZCNPs over free curcumin. Based on the above results, the developed zein-dextran nanocomplex exhibited high stability and improved the efficacy and bioactivity of curcumin suggesting its potential utility as nanovehicle for the hydrophobic drug curcumin.

Abstract:Many liposoluble nutrients are restricted for applications in foods due to their poor water solubility, stability and bioavailability. Here, a deamidated zein peptide with a broad molecular weight distribution and high carboxyl and hydrophobic residue contents was used to solubilize curcumin. The complex nanoparticles of curcumin and the peptide were produced through self-assembly in aqueous solutions. Fluorescence and infrared spectra revealed that hydrogen bonding and hydrophobic interactions actuated the assembly. The complex nanoparticles had a curcumin loading capacity of 31.9% and completely inhibited curcumin crystallization. The peptide effectively protected curcumin from decomposition in aqueous solutions by inhibiting the reaction between dissolved oxygen and curcumin. The nanoparticles presented excellent freeze-drying/re-dispersion stability without any lyoprotectant. The curcumin bioaccessibility of the nanoparticles was 75% and the nanoparticles exerted a significant antioxidant effect after oral administration in mice. This study indicates that the nanoparticles are potentially useful as an antioxidant additive in foods.
Enhanced bioavailability of curcumin amorphous nanocomposite prepared by a green process using modified starch

Abstract:Curcumin (Cur), a bioactive compound extracted from plants, has attracted widespread attention due to its multiple pharmacological activities. However, the low bioavailability due to the inherent limitations in water solubility, chemical stability, and permeability poses great challenges for realizing its clinical potentials. In the current study, octenyl succinic anhydride-modified starch (OSA-S), a renewable and biodegradable biopolymer, was employed to fabricate Cur amorphous composite nanoparticles (Cur/OSA-S NPs) through a solvent-free pH-driven method with the aim to enhance Cur's bioavailability by improving its solubility and stability. Cur/OSA-S NPs, with mean sizes of about 128.9 ± 8.6 nm, encapsulation efficiencies of about 90.0 %, and the drug loading capacities around 51.0 ± 0.2 %, were successfully prepared. Cur was found to be dispersed within the composite nanoparticles in amorphous state as confirmed by the XRD and DSC characterizations. In addition, Cur/OSA-S NPs offers excellent storage, thermal and light stability, excellent re-dispersibility, and approximately 92 times better solubility than the original Cur. Furthermore, studies of dissolution and the parallel artificial membrane permeability assay (PAMPA) confirmed enhanced dissolution rates and in vitro permeabilities of Cur/OSA-S NPs. Cancer cell viability and uptake experiments revealed that Cur/OSA-S NPs possessed more potent inhibitory effects on cancer cell proliferation compared to the raw Cur. The results obtained from the current study demonstrated the effectiveness of OSA-S for manufacturing Cur amorphous composite nanoparticles with enhanced solubility, stability, and permeability, which might be valuable for further development of Cur based products for treatment of various diseases.
Co-encapsulation of curcumin and quercetin with zein/HP-β-CD conjugates to enhance environmental resistance and antioxidant activity

Abstract:In this study, composite nanoparticles consisting of zein and hydroxypropyl beta-cyclodextrin were prepared using a combined antisolvent co-precipitation/electrostatic interaction method. The effects of calcium ion concentration on the stability of the composite nanoparticles containing both curcumin and quercetin were investigated. Moreover, the stability and bioactivity of the quercetin and curcumin were characterized before and after encapsulation. Fluorescence spectroscopy, Fourier Transform infrared spectroscopy, and X-ray diffraction analyses indicated that electrostatic interactions, hydrogen bonding, and hydrophobic interactions were the main driving forces for the formation of the composite nanoparticles. The addition of calcium ions promoted crosslinking of the proteins and affected the stability of the protein-cyclodextrin composite particles through electrostatic screening and binding effects. The addition of calcium ions to the composite particles improved the encapsulation efficiency, antioxidant activity, and stability of the curcumin and quercetin. However, there was an optimum calcium ion concentration (2.0 mM) that provided the best encapsulation and protective effects on the nutraceuticals. The calcium crosslinked composite particles were shown to maintain good stability under different pH and simulated gastrointestinal digestion conditions. These results suggest that zein-cyclodextrin composite nanoparticles may be useful plant-based colloidal delivery systems for hydrophobic bio-active agents.
Fabrication and characterization of soy β-conglycinin-dextran-polyphenol nanocomplexes: Improvement on the antioxidant activity and sustained-release property of curcumin

Abstract:In this study, glycated soy β-conglycinin (β-CG) stabilized curcumin (Cur) composites were fabricated by a unique reversible self-assembly character of β-conglycinin-dextran conjugates (β-CG-DEX). Intrinsic fluorescence and far-UV CD spectra revealed that glycation did not affect the self-assembly property of β-CG in the pH-shifting treatment. The structure of β-CG-DEX could be unfolded at pH 12.0 and reassembled during acidification (from pH 12.0 to 7.0). Meanwhile, β-CG-DEX-3d, which was incubated at 60 °C for 3 days, exhibited a high loading capacity (123.4 mg/g) for curcumin, which far exceeds that (74.90 mg/g) of β-CG-Cur. Moreover, the reassembled β-CG-DEX-3d-Cur showed eminent antioxidant activity of approximately 1.5 times higher than that of free curcumin. During the simulated gastrointestinal condition, compared with β-CG-Cur, β-CG-DEX-3d-Cur nanoparticles showed a more stable and sustained release of curcumin. Thus, β-CG-DEX has immense potential to become a new delivery carrier for hydrophobic food components by means of a self-assembly strategy.
