Abstract:Probiotic supplementation in fish feed is becoming popular as a viable alternative to curb the indiscriminate use of antibiotics in aquaculture. The effect of inactivated Lactobacillus plantarum on growth performance and health related indices of black sea bream (Acanthopagrus schlegelii) fingerlings was studied, using different concentrations of a commercial inactivated L. plantarum product (Immuno-LP20): D1 (control, 0 mg/kg), D2 (100 mg/kg), D3 (200 mg/kg), D4 (400 mg/kg), D5 (800 mg/kg), and D6 (1600 mg/kg). After an eight-week feeding trial during which the diets were fed to triplicate groups of fish (initial weight: 1.58 ± 0.05 g), D2, D3, and D4 produced fish with significantly higher growth parameters than the control fish (P < 0.05), whilst all fish fed the diets with L. plantarum had significantly improved feed utilization parameters than the control group. Serum total protein content, and lysozyme activity significantly increased in fish fed D4 than the control fish, whilst complement 4 content was higher in fish fed almost all the inactivated probiotic diets (except D5) than the control fish. Serum superoxide dismutase activity was significantly higher in fish fed all the Immuno-LP20 diets except D2, and liver glutathione peroxidase activity in D2, D3, and D4 fish were significantly higher than in the control fish. Intestinal structure was also significantly improved in fish fed the Immuno-LP20 diets. According to these results, using inactivated L. plantarum as dietary supplement could be beneficial in the production of black sea bream at an optimum inclusion rate similar to 100 mg/kg ≤ 400 mg/kg Immuno-LP20.
Sustainable plant-based diets promote rainbow trout gut microbiota richness and do not alter resistance to bacterial infection

Abstract:Abstract Background Farmed fish food with reduced fish-derived products are gaining growing interest due to the ecological impact of fish-derived protein utilization and the necessity to increase aquaculture sustainability. Although different terrestrial plant proteins could replace fishmeal proteins, their use is associated with adverse effects. Here, we investigated how diets composed of terrestrial vegetal sources supplemented with proteins originating from insect, yeast or terrestrial animal by-products affect rainbow trout ( Onchorynchus mykiss ) gut microbiota composition, growth performance and resistance to bacterial infection by the fish pathogen Flavobacterium psychrophilum responsible for frequent outbreaks in aquaculture settings. Results We showed that the tested regimes significantly increased gut bacterial richness compared to full vegetal or commercial-like diets, and that vegetal diet supplemented with insect and yeast proteins improves growth performance compared to full vegetal diet without altering rainbow trout susceptibility to F. psychrophilum infection. Conclusion Our results demonstrate that the use of insect and yeast protein complements to vegetal fish feeds maintain microbiota functions, growth performance and fish health, therefore identifying promising alternative diets to improve aquaculture’s sustainability.
Effects of dietary co-exposure to fungal and herbal functional feed additives on immune parameters and microbial intestinal diversity in rainbow trout (Oncorhynchus mykiss)

Abstract:Misuse and overuse of antibiotics in aquaculture has proven to be an unsustainable practice leading to increased bacterial resistance. An alternative strategy involves the inclusion of immunostimulants in fish diets, especially fungal and herbal compounds already authorized for human consumption, hence without environmental or public health concerns. In this study, we used a holistic and cross-disciplinary pipeline to assess the immunostimulatory properties of two fungi: Trametes versicolor and Ganoderma lucidum; one herbal supplement, capsaicin in the form of Espelette pepper (Capsicum annuum), and a combination of these fungal and herbal additives on rainbow trout (Oncorhynchus mykiss). We investigated the impact of diet supplementation for 7 weeks on survival, growth performance, cellular, humoral, and molecular immune parameters, as well as the intestinal microbial composition of the fish. Uptake of herbal and fungal compounds influenced the expression of immune related genes, without generating an inflammatory response. Significant differences were detected in the spleen-tlr2 gene expression. Supplementation with herbal additives correlated with structural changes in the fish intestinal microbiota and enhanced overall intestinal microbial diversity. Results demonstrated that the different treatments had no adverse effect on growth performance and survival, suggesting the safety of the different feed additives at the tested concentrations. While the mechanisms and multifactorial interactions remain unclear, this study provides insights not only in regard to nutrition and safety of these compounds, but also how a combined immune and gut microbiota approach can shed light on efficacy of immunostimulant compounds for potential commercial inclusion as feed supplements.

Abstract:The effects of replacing fishmeal and fish oil with a plant-based diet were studied in juvenile (10g) and ongrowing (250-350g) rainbow trout from first-feeding. Feed-related differences in the intestinal and hepatic transcriptome were examined in juveniles after 7 months of feeding at 7°C. Based on microarray results obtained for juveniles, the expression of selected genes related to lipid, cholesterol and energy metabolisms, was assessed by RT-qPCR in ongrowing trout after 6 additional months of feeding at 17°C. Plasma glucose and cholesterol, lipid content and fatty acid profile of whole body were analyzed at both stages. After 7 months at 7°C, all juveniles reached the same body weight (10g), while at 13 months ongrowing fish fed the totally plant-based diet exhibited lower body weight (234 vs 330-337g). Body lipid content was higher in juveniles fed the totally plant-based diet (13.2 vs 9.4-9.9%), and plasma cholesterol was about 2-times lower in trout fed the plant-based diets at both stages. Fatty acid profile mirrored that of the respective diet, with low proportions of long-chain n-3 polyunsaturated fatty acids in fish fed plant-based diets. Genes involved in protein catabolism, carbohydrate metabolism and trafficking were down-regulated in the intestines of juveniles fed the plant-based diets. This was not true for ongrowing fish. Genes involved in lipid and cholesterol metabolisms were up-regulated in the livers of fish fed plant-based diets for both stages. In this study, feeding trout a totally plant-based diet from first-feeding affect a relatively low proportion of metabolism-related genes. In the longer term, when fish were reared at a higher temperature, only some of these changes were maintained (i.e. up-regulation of lipid/cholesterol metabolism). Although the plant-based diets tested in this study had no major deficiencies, small adjustments in the feed-formula are needed to further optimize growth performance while sparing marine resources.

Effects of increasing dietary level of organic acids and nature-identical compounds on growth, intestinal cytokine gene expression and gut microbiota of rainbow trout (Oncorhynchus mykiss) reared at normal and high temperature

Abstract:Organic acids (OA) and nature-identical compounds (NIC) such as monoterpenes and aldehydes are well-known growth and health promoters in terrestrial livestock while their application for fish production is recent and their mechanisms of action require further study. Hence, this study tested the increasing dietary level (D0, D250, D500, D1000; 0, 250, 500 and 1000 mg kg feed-1 respectively) of a microencapsulated blend containing citric and sorbic acid, thymol and vanillin over 82 days on rainbow trout to assess the effects on growth, feed utilization, intestine cytokine gene expression and gut microbiota (GM). Furthermore, the effects on intestinal cytokine gene expression and GM were also explored after one week at high water temperature (23 °C). OA and NIC improved specific growth rate (SGR) and feed conversion rate (FCR) during the second half (day 40-82) of the feeding trial, while at the end of the trial protein (PER) and lipid efficiency (LER) increased with increasing dietary level. GM diversity and composition and cytokine gene expression analysis showed no significant differences in fish fed with increasing doses of OA and NIC (82 days) demonstrating the absence of inflammatory activity in the intestinal mucosa. Although there were no statistical differences, GM structure showed a tendency in clustering D0 group separately from the other dietary groups and a trend towards reduction of Streptococcus spp. was observed in the D250 and D1000 groups. After exposure to high water temperature, lower GM diversity and increased gene expression of inflammatory intestinal cytokines were observed for both inclusions (D0 vs. D1000) compared to groups in standard condition. However, the gene up-regulation involved a limited number of cytokines showing the absence of a substantial inflammation process able to compromise the functional activity of the intestine. Despite further study should be conducted to fully clarify this mechanism, cytokines up-regulation seems to be concomitant to the reduction of the GM diversity and, particularly, to the reduction of specific lactic acid bacteria such as Leuconostoc. The application of the microencapsulate blend tested can be a useful strategy to improve growth and feed utilization in rainbow trout under normal temperature conditions. According to the results organic acids and nature-identical compounds did not revert the effects triggered by the increased temperature of water.
Supplementation of a Multienzyme Complex, an Organic Acid-Essential Oil Complex, and Prebiotic Alone or in Combination Affects Growth, Nutrient Utilization, and Immune Function of Rainbow Trout (Oncorhynchus mykiss)

Abstract:Dietary supplementation of functional additives is an effective strategy to improve the nutritive value of low fish meal diets for fishes. The present study is aimed at investigating the dietary effects of a multienzyme complex, an organic acid-essential oil complex and prebiotic on growth, immune function, and intestinal health of rainbow trout (Oncorhynchus mykiss). A positive control diet (PC) and a negative control diet (NC) were formulated to contain 200 g/kg and 100 g/kg of fishmeal, respectively. Seven other diets were formulated by supplementing a multienzyme complex (MC, 0.125 g/kg), organic acid-essential oil complex (OEC, 0.5 g/kg), and prebiotic (P, 2 g/kg) alone or in combination to the NC diet (MC, OEC, P, MC+OEC, MC+P, OEC+P, MC+OEC+P). All diets were formulated to be isoproteic (434.3 g/kg-439.1 g/kg) and isolipidic (119.2 g/kg-125.9 g/kg) and fed to rainbow trout of 7.66 ± 0.05 g for 56 days. The weight gain, apparent digestibility coefficient of dry matter, protein efficiency ratio, and protein retention efficiency were significantly increased, and feed conversion ratio was significantly decreased by the three additives alone or in combination ( P < 0.05 ) compared to the NC. No significant differences in growth performance and feed utilization were observed among the treatment groups. Compared to the NC group, serum alanine transaminase, aspartate transferase activities, and malondialdehyde content reduced significantly, while serum superoxide dismutase increased in all except for the MC group, and alkaline phosphatase and lysozyme activity increased in all except for the MC and MC+P groups ( P < 0.05 ). The foregut protease activity of the MC+P and OEC+P groups and the amylase activity of the OEC and MC+OEC groups were significantly higher than those of the NC group ( P < 0.05 ). The villus height of P, MC+OEC, and OEC+P groups, the villus width of MC group, and the muscle thickness of MC, MC+OEC, and MC+OEC+P groups were also significantly higher than those of the NC group ( P < 0.05 ). Compared to the PC group, the richness and diversity of intestinal microorganisms in the NC group and all the supplemented groups were significantly reduced ( P < 0.05 ), but no differences among them ( P > 0.05 ). In conclusion, the individual or combined supplementation of multienzyme complex, organic acid-essential oil complex, and prebiotic in a low fish meal diet (100 g/kg) improved the growth performance, nutrient utilization, and immune function of rainbow trout, but no synergistic effects were observed in the combination of the three supplements.

However, the limited resources and expense of fish meal and oil have become restrictive factors for the development of the rainbow trout related industry. To solve this problem, plant-derived proteins and vegetable oils have been developed as alternative resources. The present study focuses on evaluating the effects of two experimental diets, FMR (fish meal replaced with plant-derived protein) and FOR (fish oil replaced with rapeseed oil), through the alteration of the gut microbiota in triploid rainbow trout. Abstract:A 70-day feeding trial investigated the effects of dietary inclusion of different types of non-starch polysaccharides (NSPs) on gut microbiota and serum metabolome of rainbow trout. Four practical feeds (42% crude protein, 17% crude lipid) were prepared with 8% insoluble NSP (INSP, cellulose), 16.8% soluble NSP (SNSP, composed of 1.12% β-glucan, 1.28% mannan, 4.8% arabinoxylan, and 9.6% pectin), 24.8% NSPs (8% INSP + 16.8% SNSP), or no NSPs inclusion, respectively. Dietary NSPs inclusion had no significant influence on the Shannon, Simpson, ACE, and Chao1 indices of gut microbiota but induced a significant increase in the abundance of Pseudomonas aeruginosa and Photobacterium kishitanii, and a decrease in Firmicutes and Alistipes finegoldii. Besides, dietary SNSP upregulated the carnitine synthesis metabolic pathway. Our data suggest that dietary NSPs are detrimental to gut microbiota homeostasis and the health of rainbow trout, and dietary SNSP exhibit a stronger ability to interfere with physiological metabolism of rainbow trout than INSP. Therefore, the physiological effects of dietary NSPs, especially SNSP, should be carefully considered when designing the commercial feed formulations of rainbow trout.

Abstract:This study compared the nutrient-energy retention, digestive function, growth performance, and welfare of rainbow trout (ibw 54 g) fed isoproteic (42%), isolipidic (24%), fishmeal-free diets (CV) over 13 weeks. The diets consisted of plant-protein replacement with graded levels (10, 30, 60%) of protein from poultry by-product (PBM) and black soldier fly H. illucens pupae (BSFM) meals, either singly or in combination. A fishmeal-based diet was also tested (CF). Nitrogen retention improved with moderate or high levels of dietary PBM and BSFM relative to CV (p < 0.05). Gut brush border enzyme activity was poorly affected by the diets. Gastric chitinase was up-regulated after high BSFM feeding (p < 0.05). The gut peptide and amino acid transport genes were differently regulated by protein source and level. Serum cortisol was unaffected, and the changes in metabolites stayed within the physiological range. High PBM and high BSFM lowered the leukocyte respiratory burst activity and increased the lysozyme activity compared to CV (p < 0.05). The BSFM and PBM both significantly changed the relative percentage of lymphocytes and monocytes (p < 0.05). In conclusion, moderate to high PBM and BSFM inclusions in fishmeal-free diets, either singly or in combination, improved gut function and nutrient retention, resulting in better growth performance and the good welfare of the rainbow trout.

Abstract:A high-carbohydrate diet could achieve a protein-sparing effect, but it may cause negative impacts on the growth condition of fish due to their poor utilisation ability of carbohydrate. How to reduce the adverse effects caused by a high-carbohydrate diet is important for the development of aquaculture. In the present study, we aimed to identify whether inulin could attenuate the metabolic syndrome caused by a high-carbohydrate diet in fish. Nile tilapia (Oreochromis niloticus) (1·19 (sd 0·01) g) were supplied with 35 % carbohydrate (CON), 45 % carbohydrate (HC) and 45 % carbohydrate + 5 g/kg inulin (HCI) diets for 10 weeks. The results showed that addition of inulin improved the survival rate when fish were challenged with Aeromonas hydrophila, indicating that inulin had an immunostimulatory effect. Compared with the HC group, the HCI group had lower lipid accumulation in liver and the gene expression analyses indicated that addition of inulin down-regulated genes related to lipogenesis and up-regulated genes relevant to β-oxidation significantly (P < 0·05). Higher liver glycogen and glucose tolerance were found in the HCI group compared with the HC group (P < 0·05). These results indicated that inulin could alleviate the metabolic syndrome induced by a high-carbohydrate diet. Furthermore, addition of inulin to a high-carbohydrate diet changed the intestinal bacterial composition and significantly increased the concentration of acetic acid and propionic acid in fish gut which have the potential to increase pathogen resistance and regulate metabolic characteristics in fish. Collectively, our results demonstrated a possible causal role for the gut microbiome in metabolic improvements induced by inulin in fish.

Abstract:A feeding trial was conducted to study the effect of inulin on growth, body composition and gut microbiota of Lates calcarifer fingerlings (average weight: 7.14±0.05g). Inulin was supplemented at five different concentrations 0, 5, 10, 15 and 20gkg−1 in the diet (400gkg1 protein and 90gkg−1 lipid) of L. calcarifer. The results of the 60 days feeding trial revealed that dietary inulin supplementation had a significant effect on specific growth rate (SGR) (linear, P=0.029; quadratic, P=0.022) and feed conversion ratio (FCR) (linear, P=0.033; quadratic, P=0.003) between control and treatments. However, the final body weight (FBW) (quadratic, P=0.0138) and weight gain (WG) (quadratic, P=0.0150) followed a quadratic pattern. Supplementation of inulin did not affect whole body moisture and lipid. However, crude protein increased both linearly (linear, P=0.001) and quadratically (quadratic, P=0.001) in fish fed with 20gkg−1 inulin supplemented diets and a quadratic pattern in ash (P<0.05) was recorded. Similarly hepatosomatic index (HSI) significantly increased (linear, P=0.008; quadratic, P=0.048) in fish fed 15gkg−1 inulin. Polymerase chain reaction and denaturing gradient gel electrophoresis (PCR-DGGE) analysis of gut samples from inulin supplemented diet revealed the change in the gut microbial community of Asian seabass. It could therefore be inferred that inulin supplementation is beneficial in the diet of Asian seabass and supplementation at 15gkg −1 level is optimal for enhancing growth.

Abstract:Insects have been identified as an economically sustainable high-value, and safe protein-rich alternative to fishmeal in compound feeds for farmed fish. Accordingly, the present study aimed to evaluate the effects of substitution of fishmeal with insect meal from Hermetia illucens in the diet of rainbow trout (Oncorhynchus mykiss), on fish growth performance, and gut microbiota composition. For this purpose, three diets, with increasing levels of insect prepupae meal inclusion (10%, 20% and 30%) in partial substitution of fishmeal and a control diet without insect meal were tested in a 12-weeks feeding trial. Fish growth and feed conversion ratio were evaluated. The Illumina MiSeq platform for high-throughput amplicon sequencing of 16S rRNA gene and QIIME pipeline were used to analyse and characterize the whole microbiome associated to aquafeeds, and fish gut. The number of reads taxonomically classified according to the Greengenes database was 1,140,534. We identified 450 OTUs at 97% identity in trout fecal samples; 62 OTUs constituted the core gut microbiota. Actinobacteria, Firmicutes and Proteobacteria represented the dominant phyla in both experimental groups. Among them, the abundance of Actinobacteria and Proteobacteria was significantly influenced by including insect meal in the diet. In summary, our findings clearly indicated that insect meal positively modifies fish gut microbiota, increasing its richness and diversity and in particular, increasing the amount of beneficial lactic acid-and butyrate-producing bacteria, which contribute to the global health of the host. In addition, based on our present and previous studies, we believe that the prebiotic effect of insect meal is principally due to fermentable chitin.

Abstract:In aquaculture, the period of active development of aquatic organisms is considered one of the most important for the formation of potential in the future. The issue of using natural components when growing aquatic organisms and feeding young fish is given special attention. This research investigated Spirulina (25%) + Vermiculture (50%) + Hermetia illucens (25%) as an additional source of functional active substances and protein that affect the body performance of rainbow trout in early ontogenesis. All objects were cultured in a special bioreactor, which was a component of the recirculating aquaculture model system as well as in the laboratory, where an installation was made for cultivating natural food. The results of the scientific and practical part of the work showed that after the end of the experiment, body weight was higher in the experimental fish by 7.2% than in the control group. This article presents the results of feeding rainbow trout as one of the cases of improving the morpho-functional and biochemical composition of the blood of this species: an increase in the total number of red blood cells by 11.0%, hemoglobin content by 9.7%, total protein at 22.7%, albumin by 24.2%, and creatinine by 15.5%, compared with the control group. The presented positive changes in the functional status of the rainbow trout organism were in harmony with the parameters of body weight, as well as histological indicators. Histological indicators of the liver and muscle tissue of the fish supplemented the obtained positive result of the influence of natural and active components of the feed. Feeding fish according to the scheme in the experimental group promotes the formation of layers of special adipose tissue in the endomysium. The total amount of the stromal component in the muscle tissue of rainbow trout was 25% of the mass of the entire tissue. The control group showed that the muscle fibers were polygonal in cross section, separated from each other within the bundle by delicate layers of endomysium. When using a standard feeding regimen (control group), the stromal component is a moderately expanding structure that replaces the space between the fibers. The experimental group had the most significant indicators. The intensification of individual aspects of homeostasis achieved with the help of feed components (experimental group) promotes the growth of muscle fibers in thickness due to the formation of new fibrils, but not an increase in the proportion of sarcoplasm (watery meat). As a result, neurohumoral regulation and substance synthesis processes improved in the fishes’ body, which helps to improve ontogenesis rates. The presented ingredients in the diet of rainbow trout are natural stimulants of metabolic processes and sources of nutrients. The method allows aquaculture to be defined as being as close as possible to “environmentally safe”. The next stage will be a comprehensive analysis of the qualitative and quantitative characteristics of rainbow trout meat under the influence of the feed factor.
Enzyme-treated microalgal co-product diets for rainbow trout aquaculture: Supporting fish growth, phosphorus digestibility, and reducing phosphorus waste emission

Abstract:Aquaculture is one of the fastest-growing food sectors, producing almost 50% of fish for human consumption worldwide. However, relying on fish meal and oil for aquaculture diets is not sustainable economically or environmentally. Aquaculture feeds also contain terrestrial plant ingredients with indigestible forms of phosphorus (P), of which 70%–80% can be released into aquatic environments. This P influx contributes to eutrophication of freshwater ecosystems that can lead to anoxic conditions. This study explores a more sustainable diet for salmonids, an important and valuable seafood. Our aim was to test ingredients with highly digestible forms of P in nutritionally balanced portions to support fish growth and reduce P loading. We determined the digestibility of three feeds containing raw, extruded, and enzymatically processed microalgal co-product of Nannochloropsis oculata compared to a conventional diet. We also quantified how much P was retained and excreted. We detected highest growth in trout fed enzymatically processed co-product feed, though it was not statistically different (p = 0.846) from growth of fish fed the reference or other co-product diets. The enzyme-treated, microalgal co-product ingredient and diet had comparable values for P digestibility and solid P excretion to the reference diet, but the lowest average solid P excretion of all test diets. Trout fed the enzyme-treated diet had the highest P retention, while the reference diet had the lowest (p = 0.0429). Trout fed the enzyme-treated diet had the lowest (p = 0.0174) and negative dissolved P excretion, while those fed the reference diet had the highest. Results showed that enzyme-treated N. oculata co-product maintains digestibility, increases P retention, and reduces dissolved P excretion compared to the reference diet in rainbow trout. These findings encourage follow-up research to design and test growth performance of diets containing enzyme-treated microalgal co-product as sustainable trout aquafeed.

Abstract:Abstract Replacement of fishmeal in feeds is critical for sustainable aquaculture growth. However, replacement with plant protein concentrates reduces fish performance. A rainbow trout strain selected for high performance on a plant protein diet was compared to a non-selected strain to identify physiological mechanisms associated with improved performance. Nutrient digestibility in fishmeal and plant protein diets was assessed and no strain differences were found. Levels of amino acids in the hepatic portal vein and caudal vein were measured at intervals after a single force-feeding of fishmeal, four plant protein concentrates, and a mixture of the concentrates with or without supplementation of three limiting amino acids. Each ingredient affected plasma amino acid levels in a singular manner when fed individually but without predictable additive effects when fed as a mixture. Amino acid supplementation altered uptake and plasma concentrations of all the essential amino acids. The selected trout strain fed the plant protein mixture with amino acids showed a synchronous and homogenous pattern for essential amino acids over time in the hepatic portal vein in contrast to that of the non-selected strain. The results demonstrate that selection favorably altered temporal dynamics of plant protein digestion.

Abstract:Abstract The rapid increase in aquaculture over the last several decades has led to concerns about the environmental impact of fish feeds relying on marine resources for fishmeal (FM). We aim to assess Nannochloropsis sp. QH25 co-product as a viable and sustainable replacement for FM in juvenile rainbow trout, Oncorhynchus mykiss , feeds. We formulated four experimental diets: a reference (FM based), 33N, 66N, and 100N diet (33%, 66%, and 100% co-product replacement). Rainbow trout were randomly assigned to one of 16 tanks and randomly assigned an experimental diet to consume throughout the experiment (64 days total), with four replicate tanks per diet. We compared the phosphorus (P) and nitrogen (N) digestibility, emissions, and growth between diets and, compared six environmental impacts (biotic resource use (BRU), global warming potential (GWP), water use, land use, marine eutrophication potential (MEP), and freshwater eutrophication potential (FEP)) of each diet. Our results indicate that replacing FM with co-product did not significantly alter growth. P digestibility of the experimental and reference diets was comparable. BRU conversion ratio was significantly lower in the experimental diets. However, there were significantly higher water and land use conversion ratios but insignificantly higher results in GWP, MEP, and FEP between the reference and 100N diet.
