Abstract:The effects of feed ingredient particle size on apparent digestibility. growth and excretion responses of rainbow trout were investigated in pilot production systems. Particle size of the feed ingredients (ground using a hammer mill with 0.6 and 3.0 mm screen sizes) had no significant effect (P > 0.05, d.f. = 1/6) on the apparent digestibility of dry matter, phosphorus and crude protein in cold-extruded diets. Feed ingredient particle size had an impact on the feed conversion ratio over an 85-day feeding trial when key ingredients (78.65% of diet) were ground using a pulverizer and a hammer mill with 3.0 ram screen size and heat extruded. No significant differences (P > 0.05, d.f. = 1/12) were observed in the excretion ratios of TAN (total ammonia nitrogen), TKN (total Kjeldahl nitrogen), TP (,total phosphorus), BOD5 (5-day biochemical oxygen demand) and TSS (total suspended solids) among three feed types, which included a wide ingredient particle size distribution.
Long-term dietary replacement of fishmeal and fish oil in diets for rainbow trout (Oncorhynchus mykiss): Effects on growth, whole body fatty acids and intestinal and hepatic gene expression

Abstract:The effects of replacing fishmeal and fish oil with a plant-based diet were studied in juvenile (10g) and ongrowing (250-350g) rainbow trout from first-feeding. Feed-related differences in the intestinal and hepatic transcriptome were examined in juveniles after 7 months of feeding at 7°C. Based on microarray results obtained for juveniles, the expression of selected genes related to lipid, cholesterol and energy metabolisms, was assessed by RT-qPCR in ongrowing trout after 6 additional months of feeding at 17°C. Plasma glucose and cholesterol, lipid content and fatty acid profile of whole body were analyzed at both stages. After 7 months at 7°C, all juveniles reached the same body weight (10g), while at 13 months ongrowing fish fed the totally plant-based diet exhibited lower body weight (234 vs 330-337g). Body lipid content was higher in juveniles fed the totally plant-based diet (13.2 vs 9.4-9.9%), and plasma cholesterol was about 2-times lower in trout fed the plant-based diets at both stages. Fatty acid profile mirrored that of the respective diet, with low proportions of long-chain n-3 polyunsaturated fatty acids in fish fed plant-based diets. Genes involved in protein catabolism, carbohydrate metabolism and trafficking were down-regulated in the intestines of juveniles fed the plant-based diets. This was not true for ongrowing fish. Genes involved in lipid and cholesterol metabolisms were up-regulated in the livers of fish fed plant-based diets for both stages. In this study, feeding trout a totally plant-based diet from first-feeding affect a relatively low proportion of metabolism-related genes. In the longer term, when fish were reared at a higher temperature, only some of these changes were maintained (i.e. up-regulation of lipid/cholesterol metabolism). Although the plant-based diets tested in this study had no major deficiencies, small adjustments in the feed-formula are needed to further optimize growth performance while sparing marine resources.

Abstract:This study aimed to investigate the effects of phenylalanine on the growth, digestive and absorptive capacity, antioxidant capacity, and intestinal health of triploid rainbow trout (Oncorhynchus mykiss) fed a low fish meal diet (15%). Five isonitrogenous and isoenergetic diets with different dietary phenylalanine levels (1.82%, 2.03%, 2.29%, 2.64%, 3.01%) were fed to triplicate groups of 20 fish (initial mean body weight of 38.65 ± 0.62 g). Weight gain rate (WGR) and specific growth rate (SGR) were significantly lower (P<0.05) in the 3.01% group. The trypsin activity in the 2.03% group was significantly higher than that in the control group (P<0.05). Serum superoxide dismutase (SOD) activity and liver peroxidase (CAT) activity both reached their maximum in the 2.03% group. Interleukin factors IL-1β and IL-6 both reached a minimum value in the 2.03% group and were significantly lower than the control group, while IL-10 reached a maximum value in the 2.03% group (P<0.05). The tight junction protein-related genes occludin, claudin-1, and ZO-1 all reached a maximum in the 2.03% treatment group and were significantly higher than the control group (P<0.05). There were also significant differences in intestinal villi length and muscle layer thickness among the groups (P<0.05). In conclusion, dietary phenylalanine effectively improved the growth, digestion and absorption capacity, antioxidant capacity, and intestinal health of O. mykiss. Using a quadratic curve model analysis based on WGR, dietary phenylalanine requirement of triploid O. mykiss fed a low fish meal diet (15%) was 2.13%.

Abstract:The present study was conducted to investigate the effects of dietary inclusion of protein hydrolysates on growth performance, digestive enzyme activities, protein metabolism, and intestinal health in larval largemouth bass (Micropterus salmoides). The experimental feeding trial presented in this study was based on five isonitrogenous and isolipidic diets formulated with graded inclusion levels of protein hydrolysates, and it showed that protein hydrolysates improved growth performance, reduced larval deformity rate, and increased the activity of digestive enzymes, including pepsin and trypsin. Gene expression results revealed that the supplementation of protein hydrolysates upregulated the expression of intestinal amino acid transporters LAT2 and peptide transporter 2 (PepT2), as well as the amino acid transporters LAT1 in muscle. Dietary provision of protein hydrolysates activated the target of rapamycin (TOR) pathway including the up-regulation of TOR and AKT1, and down-regulation of 4EBP1. Additionally, the expression of genes involved in the amino acids response (AAR) pathway, ATF4 and REDD1, were inhibited. Protein hydrolysates inhibited the transcription of some pro-inflammatory cytokines, including IL-8 and 5-LOX, but promoted the expression of anti-inflammatory cytokines TGF-beta and IL-10. The 16S rRNA analysis, using V3-V4 region, indicated that dietary protein hydrolysates supplementation reduced the diversity of the intestine microbial community, increased the enrichment of Plesiomonas and reduced the enrichment of Staphylococcus at the genus level. In summary, protein hydrolysates have been shown to be an active and useful supplement to positively complement other protein sources in the diets for largemouth bass larvae, and this study provided novel insights on the beneficial roles and possible mechanisms of action of dietary protein hydrolysates in improving the overall performance of fish larvae.

Abstract:Abstract Replacement of fishmeal in feeds is critical for sustainable aquaculture growth. However, replacement with plant protein concentrates reduces fish performance. A rainbow trout strain selected for high performance on a plant protein diet was compared to a non-selected strain to identify physiological mechanisms associated with improved performance. Nutrient digestibility in fishmeal and plant protein diets was assessed and no strain differences were found. Levels of amino acids in the hepatic portal vein and caudal vein were measured at intervals after a single force-feeding of fishmeal, four plant protein concentrates, and a mixture of the concentrates with or without supplementation of three limiting amino acids. Each ingredient affected plasma amino acid levels in a singular manner when fed individually but without predictable additive effects when fed as a mixture. Amino acid supplementation altered uptake and plasma concentrations of all the essential amino acids. The selected trout strain fed the plant protein mixture with amino acids showed a synchronous and homogenous pattern for essential amino acids over time in the hepatic portal vein in contrast to that of the non-selected strain. The results demonstrate that selection favorably altered temporal dynamics of plant protein digestion.

Effects of increasing dietary level of organic acids and nature-identical compounds on growth, intestinal cytokine gene expression and gut microbiota of rainbow trout (Oncorhynchus mykiss) reared at normal and high temperature

Abstract:Organic acids (OA) and nature-identical compounds (NIC) such as monoterpenes and aldehydes are well-known growth and health promoters in terrestrial livestock while their application for fish production is recent and their mechanisms of action require further study. Hence, this study tested the increasing dietary level (D0, D250, D500, D1000; 0, 250, 500 and 1000 mg kg feed-1 respectively) of a microencapsulated blend containing citric and sorbic acid, thymol and vanillin over 82 days on rainbow trout to assess the effects on growth, feed utilization, intestine cytokine gene expression and gut microbiota (GM). Furthermore, the effects on intestinal cytokine gene expression and GM were also explored after one week at high water temperature (23 °C). OA and NIC improved specific growth rate (SGR) and feed conversion rate (FCR) during the second half (day 40-82) of the feeding trial, while at the end of the trial protein (PER) and lipid efficiency (LER) increased with increasing dietary level. GM diversity and composition and cytokine gene expression analysis showed no significant differences in fish fed with increasing doses of OA and NIC (82 days) demonstrating the absence of inflammatory activity in the intestinal mucosa. Although there were no statistical differences, GM structure showed a tendency in clustering D0 group separately from the other dietary groups and a trend towards reduction of Streptococcus spp. was observed in the D250 and D1000 groups. After exposure to high water temperature, lower GM diversity and increased gene expression of inflammatory intestinal cytokines were observed for both inclusions (D0 vs. D1000) compared to groups in standard condition. However, the gene up-regulation involved a limited number of cytokines showing the absence of a substantial inflammation process able to compromise the functional activity of the intestine. Despite further study should be conducted to fully clarify this mechanism, cytokines up-regulation seems to be concomitant to the reduction of the GM diversity and, particularly, to the reduction of specific lactic acid bacteria such as Leuconostoc. The application of the microencapsulate blend tested can be a useful strategy to improve growth and feed utilization in rainbow trout under normal temperature conditions. According to the results organic acids and nature-identical compounds did not revert the effects triggered by the increased temperature of water.
Supplementation of a Multienzyme Complex, an Organic Acid-Essential Oil Complex, and Prebiotic Alone or in Combination Affects Growth, Nutrient Utilization, and Immune Function of Rainbow Trout (Oncorhynchus mykiss)

Abstract:Dietary supplementation of functional additives is an effective strategy to improve the nutritive value of low fish meal diets for fishes. The present study is aimed at investigating the dietary effects of a multienzyme complex, an organic acid-essential oil complex and prebiotic on growth, immune function, and intestinal health of rainbow trout (Oncorhynchus mykiss). A positive control diet (PC) and a negative control diet (NC) were formulated to contain 200 g/kg and 100 g/kg of fishmeal, respectively. Seven other diets were formulated by supplementing a multienzyme complex (MC, 0.125 g/kg), organic acid-essential oil complex (OEC, 0.5 g/kg), and prebiotic (P, 2 g/kg) alone or in combination to the NC diet (MC, OEC, P, MC+OEC, MC+P, OEC+P, MC+OEC+P). All diets were formulated to be isoproteic (434.3 g/kg-439.1 g/kg) and isolipidic (119.2 g/kg-125.9 g/kg) and fed to rainbow trout of 7.66 ± 0.05 g for 56 days. The weight gain, apparent digestibility coefficient of dry matter, protein efficiency ratio, and protein retention efficiency were significantly increased, and feed conversion ratio was significantly decreased by the three additives alone or in combination ( P < 0.05 ) compared to the NC. No significant differences in growth performance and feed utilization were observed among the treatment groups. Compared to the NC group, serum alanine transaminase, aspartate transferase activities, and malondialdehyde content reduced significantly, while serum superoxide dismutase increased in all except for the MC group, and alkaline phosphatase and lysozyme activity increased in all except for the MC and MC+P groups ( P < 0.05 ). The foregut protease activity of the MC+P and OEC+P groups and the amylase activity of the OEC and MC+OEC groups were significantly higher than those of the NC group ( P < 0.05 ). The villus height of P, MC+OEC, and OEC+P groups, the villus width of MC group, and the muscle thickness of MC, MC+OEC, and MC+OEC+P groups were also significantly higher than those of the NC group ( P < 0.05 ). Compared to the PC group, the richness and diversity of intestinal microorganisms in the NC group and all the supplemented groups were significantly reduced ( P < 0.05 ), but no differences among them ( P > 0.05 ). In conclusion, the individual or combined supplementation of multienzyme complex, organic acid-essential oil complex, and prebiotic in a low fish meal diet (100 g/kg) improved the growth performance, nutrient utilization, and immune function of rainbow trout, but no synergistic effects were observed in the combination of the three supplements.

Abstract:This study compared the nutrient-energy retention, digestive function, growth performance, and welfare of rainbow trout (ibw 54 g) fed isoproteic (42%), isolipidic (24%), fishmeal-free diets (CV) over 13 weeks. The diets consisted of plant-protein replacement with graded levels (10, 30, 60%) of protein from poultry by-product (PBM) and black soldier fly H. illucens pupae (BSFM) meals, either singly or in combination. A fishmeal-based diet was also tested (CF). Nitrogen retention improved with moderate or high levels of dietary PBM and BSFM relative to CV (p < 0.05). Gut brush border enzyme activity was poorly affected by the diets. Gastric chitinase was up-regulated after high BSFM feeding (p < 0.05). The gut peptide and amino acid transport genes were differently regulated by protein source and level. Serum cortisol was unaffected, and the changes in metabolites stayed within the physiological range. High PBM and high BSFM lowered the leukocyte respiratory burst activity and increased the lysozyme activity compared to CV (p < 0.05). The BSFM and PBM both significantly changed the relative percentage of lymphocytes and monocytes (p < 0.05). In conclusion, moderate to high PBM and BSFM inclusions in fishmeal-free diets, either singly or in combination, improved gut function and nutrient retention, resulting in better growth performance and the good welfare of the rainbow trout.

Abstract:The need for fish meal constrains fish farming and significantly impacts sustainability of the aquaculture industry. Hence, it is important to investigate the use of plant-based protein sources in fish diets. The present study was conducted to determine the effects of different levels of fish meal (FM) replacement by pea protein (PP) in a 60-day feeding experiment in rainbow trout, Oncorhynchus mykiss. Effects on growth performance, body composition, hematology, serum biochemistry and immunology, and immune (TNF-α, IL1-ß and Il-8) and growth-related (GH and IGFI) gene expression were investigated. Five experimental diets (45% protein and 20% lipid) differed in replacement level of FM by PP at rates of 0% (control (PP0)), 25% (PP25), 50%(PP50), 75%(PP75) and 100%(PP100). Fish were fed with experimental diets in triplicate twice daily. The best growth performance was obtained in PP0 and PP25 groups. While fat ratios of fish fillets significantly differed (p < 0.05), there was no significant effects on protein ratios (p < 0.05). There was no significant change in the hematological values of fish, except those fed the PP100 diets, which displayed a reduction in eyrthocyte counts, hemoglobin content and hematocrit. As PP supplementation increased fish showed elevated serum glucose, total protein, cholesterol and myeloperoxidase activity and decreased glutamic pyruvic transaminase and alkaline phosphatase activity. Fish fed diets with between 25 and 75% replacement showed a decline in lactic acid bacteria in the gut. Significant increases in expression were observed in the liver of the PP25 fish relative to the 0% control for all immune and growth-related genes except for IL1-ß. These data suggest that up to 25% of FM can be replaced by PP without any adverse effects on rainbow trout.

Abstract:This study investigated the dietary effects of lipid and protein levels on growth performance, feed utilization, body composition, lipid metabolism, and antioxidant capacity of triploid rainbow trout, Oncorhynchus mykiss. A 3 × 2 two-factor design was conducted with three crude lipid levels of 4%, 9%, and 14% (L4, L9, and L14) and two crude protein levels of 44%, 49% (P44, P49). Therefore, a total of six diets were prepared as P44/L4, P44/L9, P44/L14, P49/L4, P49/L9, and P49/L14. Triploid rainbow trout (initial body weight 65.0 ± 0.1 g) were fed one of the six diets for 80 days. The results showed that weight gain (WG), protein retention (PR), and protein efficiency rate (PER) significantly increased with increasing the dietary lipid level at the same crude protein level, while feed conversion ratio (FCR) and hepatosomatic index significantly decreased ( P < 0.05 ). At the same lipid level, there was no difference in WG, FCR, PR, PER between 44% and 49% crude protein group ( P > 0.05 ). The P49/L14 group had the highest WG (374.6%) and lowest FCR (1.25), while P44/L14 group had the highest PER (1.80) and PR (25.06%) with similar WG and FCR to P49/L14 group. The crude lipid contents in whole fish were significantly higher in the L14 group than those in the L4 and L9 groups ( P < 0.05 ). Muscle n-3 PUFAs, n-6 PUFAs, and PUFAs levels were positively correlated with dietary lipid level, while n-6 PUFAs was negatively correlated with dietary protein level. Dietary protein, dietary lipid, and their interaction significantly affected hepatic malondialdehyde (MDA) content, aspartate aminotransferase, lipase (LPS), and fatty acid synthase (FAS) activities ( P < 0.05 ). In both P44 and P49 groups, LPS and FAS activities increased with increasing the dietary lipid level. MDA content significantly decreased in the P44 group and increased in the P49 group with increasing the dietary lipid level ( P < 0.05 ). As dietary protein level increased, serum total cholesterol level increased, while hepatic phosphoenolpyruvate carboxykinase activity decreased. With increasing the dietary lipid level, total superoxide dismutase, catalase, total nitric oxide synthase, and fructose-1,6-bisphosphatase activities showed an increasing trend, while the opposite was true for alanine aminotransferase activity. In conclusion, based on growth performance and feed utilization, dietary protein level of 44% and dietary lipid level of 14% (measured value, 43.71% and 13.62%) were suggested for young triploid rainbow trout.

However, the limited resources and expense of fish meal and oil have become restrictive factors for the development of the rainbow trout related industry. To solve this problem, plant-derived proteins and vegetable oils have been developed as alternative resources. Abstract:Aquaculture is one of the fastest-growing food sectors, producing almost 50% of fish for human consumption worldwide. However, relying on fish meal and oil for aquaculture diets is not sustainable economically or environmentally. Aquaculture feeds also contain terrestrial plant ingredients with indigestible forms of phosphorus (P), of which 70%–80% can be released into aquatic environments. This P influx contributes to eutrophication of freshwater ecosystems that can lead to anoxic conditions. This study explores a more sustainable diet for salmonids, an important and valuable seafood. Our aim was to test ingredients with highly digestible forms of P in nutritionally balanced portions to support fish growth and reduce P loading. We determined the digestibility of three feeds containing raw, extruded, and enzymatically processed microalgal co-product of Nannochloropsis oculata compared to a conventional diet. We also quantified how much P was retained and excreted. We detected highest growth in trout fed enzymatically processed co-product feed, though it was not statistically different (p = 0.846) from growth of fish fed the reference or other co-product diets. The enzyme-treated, microalgal co-product ingredient and diet had comparable values for P digestibility and solid P excretion to the reference diet, but the lowest average solid P excretion of all test diets. Trout fed the enzyme-treated diet had the highest P retention, while the reference diet had the lowest (p = 0.0429). Trout fed the enzyme-treated diet had the lowest (p = 0.0174) and negative dissolved P excretion, while those fed the reference diet had the highest. Results showed that enzyme-treated N. oculata co-product maintains digestibility, increases P retention, and reduces dissolved P excretion compared to the reference diet in rainbow trout. These findings encourage follow-up research to design and test growth performance of diets containing enzyme-treated microalgal co-product as sustainable trout aquafeed.

Effects of yeast extract supplemented in diet on growth performance, digestibility, intestinal histology, and the antioxidant capacity of the juvenile turbot (Scophthalmus maximus)

Abstract:An 8-week feeding experiment was conducted on the juvenile turbot (Scophthalmus maximus ) to evaluate the influence of yeast extract (YE) supplementation in the diet on growth performance, feed utilization, body composition, nutrient digestibility, intestinal histology, and antioxidant capacity. Four experimental diets were formulated with graded levels of yeast extract 0 (YE0), 1% (YE1), 3% (YE3), and 5% (YE5) and fed to turbots (initial body weight: 4.2 ± 0.1 g) with three replicates per diet and 200 fish in each replicate, respectively. The results showed that turbots fed with diets YE1 and YE3 displayed a significantly higher specific growth rate and protein efficiency rate than those fed with diets YE0 and YE5, while the feed conversion ratios in YE1 and YE3 groups were lower than those in YE0 and YE5. Fish fed with diets YE3 and YE5 showed higher body crude protein contents than those in groups YE0 and YE1. The highest apparent digestibility coefficients for dry matter and crude protein, digestive enzyme activities (trypsin, lipase, and amylase), and the height of the intestinal fold were observed in the YE3 group. YE3 treatment displayed a significantly higher superoxide dismutase (SOD) activity than the YE0 group, while the malondialdehyde (MDA) content in YE1 was significantly lower than those in YE0 and YE5. No significant difference was observed in serum physiological and biochemical parameters among all treatments. Overall, appropriate dietary supplementation of the yeast extract could improve the growth performance, digestibility, and antioxidant capacity of the juvenile turbot, and the recommended yeast extract level in the feed is 2.47%.

Abstract:Arachidonic acid (ARA) is becoming increasingly important due to significant reductions in fishmeal (FM) and fish oil (FO), the primary sources of ARA, in aquafeeds. ARA is an essential omega-6 fatty acid in some fish species and plays a wide range of physiological roles. However, studies reported to date have demonstrated contrasting findings concerning the effects of ARA in fish growth and health, regardless of the fish's trophic level. Additionally, limited research has addressed energy allocation during reproduction and growth in sub-adult rainbow trout as they approach reproductive maturity. Therefore, a study was conducted to evaluate the effects of different levels of ARA on growth performance, the fatty acid composition of whole-body and ovary, intestinal inflammatory gene expression, gut histology, antioxidant activity, and non-specific immunity in sub-adult rainbow trout ( Oncorhynchus mykiss ). Four isonitrogenous (51% crude protein), isolipidic (15% crude lipid), and isocaloric (23 MJ/kg) diets were formulated and supplemented with ARA-enriched oil (40%), resulting in analyzed ARA levels of 0.10 (ARA 0.10 ), 3.95 (ARA 3.95 ), 7.67 ARA( 7.67 ), and 14.8% (ARA 14.8 ) of total fatty acid, respectively. Rainbow trout (307 ± 3.64 g) were fed to apparent satiation three times a day for 12 weeks to assess growth performance, feed utilization, and intestinal inflammatory response. The fish were then exposed to acute stress to evaluate circulating cortisol, prostaglandin E 2 , antioxidant activity, and non-specific immune response. The 12-week growth trial demonstrated that dietary ARA did not affect rainbow trout growth performance and feed utilization during the final grow-out. Whole-body fatty acid profiles reflected those of the diets except for eicosapentaenoic acid (EPA) content being significantly lower in the ARA 14.8 group than in the ARA 0.10 and ARA 3.95 groups. EPA and docosahexaenoic acid (DHA) contents in the ovary were less affected by dietary ARA supplementation, likely reflecting the selective deposition of those fatty acids in embryonic development. The expression of tnf-α was significantly downregulated in ARA 7.67 and ARA 14.8 groups relative to other treatment groups, and only distal intestinal villus height was significantly increased in the ARA 3.95 group, suggesting that dietary ARA supported intestinal health. Plasma superoxide dismutase, catalase, and lysozyme activity significantly increased in both pre- and post-stress groups due to incremental increases in dietary ARA. These results suggest that a moderate level of dietary ARA in low FM and FO-free enhances innate immune function and antioxidant responses while supporting intestinal health in sub-adult rainbow trout.

Abstract:The present study aimed to investigate the effects of dietary sodium propionate on growth performance, hepatic lipid deposition, and intestinal activity of digestive enzymes, inflammation, bacterial population, and antioxidant capacity in rainbow trout, Oncorhynchus mykiss . The fish were fed diets supplemented with 0 g/kg (CTL), 1 g/kg (P1), 2.5 g/kg (P2.5), or 5 g/kg (P5) sodium propionate for 70 days. The results showed significant ( P < 0.05) linear or quadratic relationships between dietary sodium propionate levels and all tested parameters. Growth performance and feed efficiency significantly increased in the P1 and P2.5 treatments, but decreased in the P5 treatment, compared to the CTL group. Hepatosomatic index significantly decreased in the P1 and P2.5 treatments, but increased in the P5 treatment, compared to the CTL group. The P1 and P2.5 treatments exhibited significantly lower viscerosomatic indices than the CTL group. The P1 and/or P2.5 treatments exhibited significantly higher intestinal amylase, lipase, trypsin, chymotrypsin, superoxide dismutase (SOD), catalase (CAT), and lower intestinal malondialdehyde (MDA), plasma glucose, and triglyceride levels than the CTL treatment. On the other hand, the P5 treatment exhibited significant decrease in intestinal lipase, SOD, CAT, and higher intestinal MDA, plasma glucose, cortisol, triglyceride, and cholesterol levels than the CTL group. The intestinal total viable bacteria significantly decreased in the propionate treatments, and the lowest level was observed in the P2.5 treatment. The P1 and P2.5 treatments had significantly higher intestinal abundance of Lactobacillus sp. and lower Aeromonas hydrophila , Yersinia ruckeri , and Streptococcus iniae than the CTL group. The pathogenic bacteria showed significant elevations in the P5 treatment, compared to the CTL group. The propionate treatment groups showed significant up-regulations in intestinal heat shock protein 70 expression than the CTL group. The intestinal expression of tumor necrosis factor-alpha and interleukin-1 beta significantly decreased in the P1 and P2.5 treatments, while increased in the P5 treatment, compared to the CTL group. The intestinal expression of defensin significantly increased in the P1 and P2.5 treatments, while decreased in the P5 treatment, compared to the CTL group. In conclusion, dietary sodium propionate can improve the growth and intestinal health of rainbow trout. Based on the results, 2.5 g/kg sodium propionate is suitable for rainbow trout, but higher levels induce growth retardation, hepatic lipid accumulation, and intestinal oxidative stress, inflammation, and dysbiosis.

Abstract:Experimental studies of barley and maize lipid transfer proteins (LTPs) show that the two proteins bind the ligand palmitate in opposite orientations in their internal cavities. Moreover, maize LTP is reported to bind the ligand caprate in the internal cavity in a mixture of two orientations with approximately equal occupancy. Six 30 ns molecular dynamics (MD) simulations of maize and barley LTP with ligands bound in two orientations (modes M and B) have been used to understand the different ligand binding preferences. The simulations show that both maize and barley LTP could bind palmitate in the orientation observed experimentally for maize LTP (mode M), with the predominant interaction being a salt bridge between the ligand carboxylate headgroup and a conserved arginine side chain. However, the simulation of barley LTP with palmitate in the mode B orientation shows the most favorable protein-ligand interaction energy. In contrast, the simulations of maize LTP with palmitate and with caprate in the mode B orientation show no persistent ligand binding, the ligands leaving the cavity during the simulations. Sequence differences between maize and barley LTP in the AB loop region, in residues at the base of the hydrophobic cavity, and in the helix A region are identified as contributing to the different behavior. The simulations reproduce well the experimentally observed binding preferences for palmitate and suggest that the experimental data for maize LTP with caprate reflect ligand mobility in binding mode M rather than the population of binding modes M and B.
A novel dietary multi-strain yeast fraction modulates intestinal toll-like-receptor signalling and mucosal responses of rainbow trout (Oncorhynchus mykiss)

Abstract:This study was conducted to evaluate the mucosal immune responses of rainbow trout when supplementing an experimental formulated feed with multi-strain yeast fraction product ( Saccharomyces cerevisiae and Cyberlindnera jardinii ). In total, 360 fish (initial BW 23.1 ± 0.2 g) were randomly allotted into three dietary treatments in an 8-week feeding trial. The dietary treatments included basal diet (control) and control + 1.5 g/kg multi-strain yeast fraction product (MsYF) fed continuously and pulsed every two weeks between control and MsYF diet. No negative effects on growth performance of feeding the MsYF supplemented diet were observed. SGR and FCR averaged 2.30 ± 0.03%/day and 1.03 ± 0.03, respectively, across experimental groups. Muscularis thickness in the anterior intestine after 8 weeks of feeding was significantly elevated by 44.3% in fish fed the MsYF continuously, and by 14.4% in fish fed the MsYF pulsed ( P 50% increase) intestine were observed after 8 weeks of feeding the MsYF supplemented diet ( P 10% reduction). The gene expression analysis of the intestine revealed significant elevations in expression of tlr2 , il1r1 , irak4 , and tollip2 after 4 weeks of feeding the MsYF. Significant elevations in effector cytokines tnfα , il10 and tgfβ were observed after 4 weeks of feeding the MsYF regime. After 8 weeks significant elevations in the gene expression levels of il1β , ifnγ , and il12 were observed in fish fed the MsYF. Likewise, the expression of the transcription factor gata3 was significantly elevated ( P <0.01). Supplementation of the multi-strain yeast fraction product positively modulates the intestinal mucosal response of rainbow trout through interaction with toll-like receptor two signalling pathway and potential for increased capacity of delivery of antigens to the underlying mucosal associated lymphoid tissue.

Abstract:Duplicate groups of rainbow trout (mean initial weight 16.7 ± 0.1 g) were fed six isonitrogenous (43.7 % crude protein) diets for 12 weeks. A fish-meal-based diet was designated the control. In the other five diets, 75 or 100 % of the fish meal was replaced with either poultry by-product meal (PBM), hydrolyzed feather meal (FEM), and spray-dried blood meal (BM) or with defatted soybean meal and corn gluten meal. Trout fed the diets in which 75 and 100 % of the fish meal was replaced with rendered animal protein showed comparable growth performance to trout fed the control diet except in terms of the protein efficiency ratio and feed conversion ratio. The feed intake of the trout fed a combination of fish meal and rendered animal protein (either with or without plant protein) was significantly higher than the feed intake of trout on the fish-meal-based diet. Apparent crude protein digestibility coefficients were significantly higher in trout fed the diets in which 75 % of the fish meal was replaced with plant protein sources than in trout fed the diets in which 75 % of the fish meal was replaced with rendered animal protein sources (P < 0.05). These results suggest that a combination of PBM, FEM, and BM is a good substitute for most of the fish meal in practical feed for rainbow trout.
