Abstract:Abstract Diet has an important role in shaping the structure of rumen microbial communities. However, little is known about the interaction between dietary crude protein (CP) level and CP feeding pattern on the rumen microbiome of dairy cows. Here, the objective was to evaluate the influence of CP level (low protein [LP], 13.8%; high protein [HP], 15.5% of dry matter [DM] basis) and CP feeding pattern (OF = oscillating, SF = static) on the composition and diversity of the rumen microbiome of lactating dairy cows using 16S rRNA gene amplicon sequencing. Rumen-cannulated Holstein cows (n = 8) in mid- to late-lactation were assigned to a 2 x 2 factorial experiment consisting of 4 experimental periods of 28 d. Each experimental period had an adaptation phase to the diet (24 d) and a sampling phase (d 25 to 28). The treatments with oscillating feeding alternated diets every 48 h to vary CP above and below the mean CP level (OF-LP = 13.8 ± 1.8%; OF-HP = 15.5 ± 1.8% CP of DM) whereas diets were constant in the static feeding (SF-LP = 13.8%; SF-HP = 15.5% CP of DM). According to the NASEM (2021) model predictions the LP diet supplied 94% of the microbial protein requirement while the HP diet exceeded (104%) requirement. Rumen samples were collected 4 h after feeding through a rumen cannula and amplicon libraries were generated targeting the V4 region of the 16S rRNA gene. The amplicon sequences were analyzed in qiime2. Sequences where trimmed based on their quality and then grouped in ASVs using DADA2. The ASVs were used to analyze the composition and diversity of the rumen microbiome in each treatment. Alpha diversity of the rumen microbiome did not change across treatments (Kruskal Wallis, P > 0.05), but Pielou’s evenness increased by 0.01 units (Kruskal Wallis, P = 0.04). These results support the hypothesis that the rumen microbiome is stable to oscillating CP feeding patterns and compensates for the changes in crude protein content in the diet. Further research is required to understand the dynamics of this compensation and what other metabolic processes in the ruminant contribute to maintaining production under an oscillating diet.

Abstract:The effect of dietary crude protein (CP) level on intake, digestibility, milk production, and nitrogen (N) use efficiency was studied. Twenty-four Holstein-Friesian cows (17 multiparous and seven primiparous) were grouped by parity, days in milk, milk yield, and live weight into six blocks of four, and randomly assigned to four total mixed ration (TMR) treatments, containing 141, 151, 177, or 210 g CP/kg dry matter (DM), over 28 day experimental periods. Apparent total-tract DM and fiber digestibilities and milk fat composition were similar across treatments. Milk protein and urea-N compositions, and urinary and manure N excretion increased linearly, while milk N efficiency (MNE) decreased linearly with increasing CP. DM intake was highest with the 177 diet, while CP intake increased linearly with increasing CP, peaking at 200 g/kg DM. Milk yield increased with CP intake for the three lower CP levels, peaking at 176 g CP/kg DM. The further increase in CP level from 177 to 210 g/kg DM did not result in improved milk yield, but resulted in decreased milk N secretion and increased urinary N excretion. In summary, milk protein composition increased linearly with increasing CP, accompanied by a linear decrease in MNE, resulting in a bell-shaped relationship between milk yield and dietary CP level.

Abstract:Reducing the dietary crude protein (CP) concentration can decrease the financial cost and lower the environmental impact of milk production. Two studies were conducted to examine the effects of reducing the dietary CP concentration on animal performance, nutrient digestibility, milk fatty acid (FA) profile, and nitrogen use efficiency (NUE; milk N/N intake) in dairy cows fed legume silage-based diets. Thirty-six multiparous Holstein-Friesian dairy cows that were 76 ± 14 (mean ± SD) days in milk and 698 ± 54 kg body weight were used in a 3 × 3 Latin square design in each of 2 studies, with 3 periods of 28 d. In study 1, cows were fed diets based on a 50:50 ratio of red clover to grass silage [dry matter (DM) basis] containing 1 of 3 dietary CP concentrations: high (H) = 175 g of CP/kg of DM; medium (M) = 165 g of CP/kg of DM; or low (L) = 150 g of CP/kg of DM. In study 2, cows were fed 175 g of CP/kg of DM with a 50:50 ratio of alfalfa to corn silage (H50) or 1 of 2 diets containing 150 g of CP/kg of DM with either a 50:50 (L50) or a 60:40 (L60) ratio of alfalfa to corn silage. Cows in both studies were fed a total mixed ration with a forage-to-concentrate ratio of 52:48 (DM basis). All diets were formulated to meet the MP requirements, except L (95% of MP requirements). In study 1, cows fed L ate 1.6 kg of DM/d less than those fed H or M, but milk yield was similar across treatments. Mean milk protein, fat, and lactose concentrations were not affected by diet. However, the apparent total-tract nutrient digestibility was decreased in cows fed L. The NUE was 5.7 percentage units higher in cows fed L than H. Feeding L also decreased milk and plasma urea concentrations by 4.4 mg/dL and 0.78 mmol/L, respectively. We found no effect of dietary treatment on the milk saturated or monounsaturated FA proportion, but the proportion of polyunsaturated FA was increased, and milk odd- and branched-chain FA decreased in cows fed L compared with H. In study 2, DM intake was 2 kg/d lower in cows receiving L50 than H50. Increasing the alfalfa content and feeding a low-CP diet (L60) did not alter DMI but decreased milk yield and milk protein concentration by 2 kg/d and 0.6 g/kg, respectively, compared with H50. Likewise, milk protein and lactose yield were decreased by 0.08 kg/d in cows receiving L60 versus H50. Diet had no effect on apparent nutrient digestibility. Feeding the low-CP diets compared with H50 increased the apparent NUE by approximately 5 percentage units and decreased milk and plasma urea concentrations by 7.2 mg/dL and 1.43 mmol/L, respectively. Dietary treatment did not alter milk FA profile except cis-9,trans-11 conjugated linoleic acid, which was higher in milk from cows receiving L60 compared with H50. We concluded that reducing CP concentration to around 150 g/kg of DM in red clover and grass or alfalfa and corn silage-based diets increases the apparent NUE and has little effect on nutrient digestibility or milk performance in dairy cows.

Abstract:Ornithine decarboxylase antizyme (OAZ) has recently emerged as a potential therapeutic target in various malignant tumors because it plays vital roles in cellular functions including proliferation, differentiation, apoptosis and genomic stability. Therefore, there is a significant interest in discovering its function in chronic myeloid leukemia (CML). Firstly, OAZ1 mRNA was measured by qRT-PCR in 43 cases with CML and 23 controls, and we demonstrated that it is significantly down-regulated in CML patients. To further understand its functions in CML pathogenesis, OAZ1 was overexpressed, and the human leukemia PCR array analysis was used to monitor the expression of key genes commonly involved in leukemia development, classification and therapeutic response. We found several favorable up-regulation factors including CXCL10, DAPK1 and IKZF3. In conclusion, OAZ1 may be a useful therapeutic target in CML due to its potential ability to induce erythroid differentiation and cell apoptosis. These functions were proven to be associated with several gene changes that were directly or indirectly caused by OAZ1. The mechanism of how OAZ1 affects other genes remains to be elucidated.

Abstract:Feeding corn dried distillers grains with solubles (DDGS) in low crude protein (CP) diets could limit N waste in lactating cows. However, it also could possibly reduce metabolizable AA supply, especially Lys, and compromise milk production. Therefore, the objective of this study was to evaluate the effects of feeding supplemental blends of rumen undegradable protein (RUP) and rumen-protected (RP) AA in a low compared with high CP diet containing corn DDGS on milk production and selected measures of N utilization. Six multiparous Holstein cows (619.3 ± 49.8 kg of body weight; 26.8 ± 6.2 d in milk) were subjected to a split-plot, 3 × 3 Latin square design (21-d periods) with dietary CP content [low (14.6%; LP) or high (16.6%; HP)] as the whole-plot factor, and blend of RUP and RP-AA [control (CON), no supplement; blend A (0.11 kg/cow per d); or blend B (0.45 kg/cow per d)] as the sub-plot factor. All diets contained 10% corn DDGS; blends of RUP and RP-AA were top-dressed during morning feeding. There was no dietary CP content × supplemental blend interaction for all measured variables. Nutrient (dry matter, organic matter, neutral detergent fiber, acid detergent fiber, and CP), milk and milk component yields, and feed and apparent N efficiency did not differ for cows fed the low- compared with the high-protein diet. However, apparent total-tract CP digestibility, blood and milk urea-N concentrations, and urinary excretion (g/d) of N and urea-N were lower for cows fed the low-protein compared with the high-protein diet. There was no supplemental blend effect on nutrient intake and apparent total-tract digestibility, and milk and milk component yields. Except for a tendency for total urinary purine derivative excretion and microbial N flow to be lower for cows fed blend B compared with CON but not blend A, there was no supplemental blend effect on measures of N utilization. Both dietary CP content and supplemental blend of RUP and RP-AA had a marginal effect on the plasma free AA profile. In summary, reducing dietary CP content in diets containing corn DDGS had no effect on lactation performance, possibly accounting for the lack of a positive response following the provision of supplemental blends of RUP and RP-AA. However, reducing dietary CP content resulted in a decrease in blood and milk urea-N concentrations, and urinary excretion of N and urea-N, suggestive of an improvement in the efficiency of N use.

Patch‐Test Reactions to Thioureas Are Frequently Relevant

Abstract:Background: Thioureas are used as antioxidants in the manufacture of rubber (especially neoprene) and as fixatives in photography and photocopy paper. The number of reported cases of allergic contact dermatitis from thioureas is relatively low, and standard patch test series often do not contain thiourea allergens. Thioureas are included in our institution's standard patch test series. Objective: To examine the occurrence and relevance of patch‐test reactions to thioureas. Methods: We retrospectively analyzed patch‐test data for patients evaluated between January 1999 and June 2001. Patients included in the study underwent testing with a standard screening series of allergens and a standardized patch testing technique. Our standard patch test series includes mixed dialkyl thioureas at a concentration of 1% in petrolatum. Results: Of 1,368 patients tested with our standard series, 33 patients (2.4%) had patch‐test reactions to mixed dialkyl thioureas at day 5. Reactions in 14 (42%) of those patients were of definite clinical relevance, indicating a confirmed exposure to thioureas in many different settings. Conclusion: Patch‐test reactions to thioureas occur as often as do reactions to many other allergens in the standard series and are often relevant. Institutions should consider including mixed dialkyl thioureas in their standard patch test screening series.
The Effects of Feeding Rations That Differ in Neutral Detergent Fiber and Starch Concentration Within a Day on Production, Feeding Behavior, Total-Tract Digestibility, and Plasma Metabolites and Hormones in Dairy Cows.

Abstract:Dairy cows exhibit a daily pattern of feed intake and milk synthesis, and feeding a single total mixed ration over the day may not synchronize ruminal fermentation, nutrient absorption, and milk synthesis. Our hypothesis was that feeding a high-fiber diet during the high-intake period of the day and a low-fiber diet during the low-intake overnight period would stabilize rumen fermentation, nutrient absorption, and the availability of nutrients for milk synthesis. Nine ruminally cannulated cows were used in a 3×3 Latin square design with 23-d periods. Diets were a control [CON; 33.3% neutral detergent fiber (NDF)], a low-fiber diet (LF; 29.6% NDF), and a high-fiber diet (HF; 34.8% NDF). The HF and LF diets were balanced to provide the same nutrient composition as the control diet when cows were fed a 7:3 ratio of HF:LF. Control cows were fed once daily at 0900h. Cows in the high/low treatment (H/L) were fed HF at 70% of daily offering at 0900h and LF at 30% of daily offering at 2200h; cows in the low/high (L/H) treatment were fed LF at 30% of daily offering at 0900h and HF at 70% of daily offering at 1300h. Dry matter intake was decreased 1.9kg/d by H/L compared with CON, but intake did not differ between H/L and L/H. The H/L and L/H treatments resulted in a large amount of feed consumed after each feeding, which shifted the daily patterns of starch and fiber intake. Treatments did not differ in total-tract digestibility. We detected no differences between CON and H/L for milk yield and composition. The H/L treatment unexpectedly tended to reduce milk and reduced milk fat yield compared with L/H, although L/H was expected to result in poorer rumen fermentation and decreased milk fat yield. Treatment did not affect milk fat concentrations of trans-10 C18:1, trans-11 C18:1, or de novo synthesized fatty acids, but H/L increased milk fat concentration of preformed fatty acids compared with CON (39.15 vs. 37.38% of FA, respectively). Treatment had no effect on plasma insulin or glucose at individual time points but did modify the phase and amplitude of the daily rhythms. Daily average plasma nonesterified fatty acids (NEFA) were decreased by H/L compared with CON and L/H, and H/L treatments decreased the amplitude of the daily rhythm of NEFA by 3.4 and 6.7μEq/L compared with CON and L/H, respectively. The amplitude of the daily rhythm of plasma blood urea N was increased by more than 15% by H/L compared with CON and L/H. Feeding multiple rations over the day changed the daily pattern of starch and fiber intake and decreased daily intake without decreasing milk yield or body weight gain over the 23-d observation periods. Feeding a high-fiber diet during the high-intake period of the day and a low-fiber diet during the low-intake overnight period failed to show other benefits, likely because of the change in feeding behavior.
Effect of increasing dietary protein with constant lysine:methionine ratio on production and omasal flow of nonammonia nitrogen in lactating dairy cows

Abstract:Eight lactating cows were fed 4 diets in which dietary crude protein (CP) was increased in steps of approximately 2 percentage units from 11 to 17% of DM by replacing high-moisture corn with soybean meal supplemented with rumen-protected Met to maintain a Lys:Met ratio of 3:1 in metabolizable protein. Trial design was a replicated 4 × 4 Latin square; experimental periods lasted 28 d, with data and sample collection being performed during wk 3 and 4 of each period. Digesta samples were collected from the rumen as well as the omasum to measure metabolite concentrations and ruminal outflow of N fractions using infusion of 15N-enriched ammonia to quantify microbial nonammonia N (NAN) and nonmicrobial NAN. Data were analyzed using the MIXED procedure of SAS (SAS Institute Inc.). There were linear increases in the yields of milk and true protein and concentration of milk urea N, and a linear decrease in N efficiency, with increasing dietary CP. Apparent ruminal and total-tract N digestibility increased linearly with increasing dietary CP, but estimated true total-tract N digestibility was not affected. Apparent digestibility of the other macronutrients was not influenced by diet. Ruminal ammonia, total AA and peptides, and branched-chain VFA also increased linearly with dietary CP. The 15N enrichment of liquid- and particle-associated microbes linearly declined with increasing dietary CP due to decreasing 15N enrichment of the ammonia pool. Although no effect of dietary CP on nonmicrobial NAN flow was detected, total NAN flow increased linearly from 525 g/d at 11% CP to 637 g/d at 17% CP due to the linear increase in microbial NAN flow from 406 g/d at 11% CP to 482 g/d at 17% CP. Under the conditions of this study, when dietary CP was increased by adding soybean meal supplemented with rumen-protected Met, improved milk and protein yields were driven not by RUP supply but by increased ruminal outflow of microbial protein.
Effects of rumen-encapsulated methionine and lysine supplementation and low dietary protein on nitrogen efficiency and lactation performance of dairy cows

Abstract:Low crude protein (CP) diets might be fed to dairy cows without affecting productivity if the balance of absorbed AA were improved, which would decrease the environmental effect of dairy farms. The aim of this study was to investigate the effects of supplementing ruminally protected Lys (RPL) and Met (RPM) at 2 levels of dietary CP on nutrient intake, milk production, milk composition, milk N efficiency (MNE), and plasma concentrations of AA in lactating Holstein cows and to evaluate these effects against the predictions of the new NASEM (2021) model. Fifteen multiparous cows were used in a replicated 3 × 3 Latin square design with 21-d periods. The 3 treatments were (1) a high-protein (HP) basal diet containing 16.4% CP (metabolizable protein [MP] balance of -130 g/d; 95% of target values), (2) a medium-protein diet containing 15% CP plus RPL (60 g/cow per day) and RPM (25 g/cow per day; MPLM; MP balance of -314 g/d; 87% of target values), and (3) a low-protein diet containing 13.6% CP plus RPL (60 g/cow per day) and RPM (25 g/cow per day; LPLM; MP balance of -479 g/d; 80% of target values). Dry matter intake was less for cows fed MPLM and LPLM diets compared with those fed the HP diet. Compared with the HP diet, the intake of CP, neutral detergent fiber, acid detergent fiber, and organic matter, but not starch, was lower for cows fed MPLM and LPLM diets. Milk production and composition were not affected by MPLM or LPLM diets relative to the HP diet. Milk urea N concentrations were reduced for the MPLM and LPLM diets compared with the HP diet, indicating that providing a low-protein diet supplemented with rumen-protected AA led to greater N efficiency. There was no significant effect of treatment on plasma AA concentrations except for proline, which significantly increased for the MPLM treatment compared with the other 2 treatments. Overall, the results supported the concept that milk performance might be maintained when feeding lactating dairy cows with low CP diets if the absorbed AA balance is maintained through RPL and RPM feeding. Further investigations are needed to evaluate responses over a longer time period with consideration of all AA rather than on the more aggregated MP and the ratio between Lys and Met.

Abstract:Past studies have shown that isoacids (ISO) improve dairy cow performance, with effects varying based on dietary forage levels, leading us to speculate that ISO supplementation may also differentially affect enteric methane (CH4) emissions depending on dietary forage levels. Therefore, our primary objective was to examine the effects of ISO supplementation on enteric CH4 emissions in lactating dairy cows fed 2 forage NDF levels (FL), along with monitoring feed particle sorting and chewing behaviors to assess any potential interactions. Sixty-four (64) mid-lactation Holstein cows were utilized in a 10-wk long randomized complete block design trial. Parity, DIM, and prior milk yield (MY) for multiparous cows or genetic merit for primiparous cows were used as blocking factors. Cows were randomly assigned to 1 of 4 diets (n = 16 per diet) with a 2 × 2 factorial arrangement of treatment combinations, including 2 FL, 17 (LF) and 21% forage NDF (HF), without or with ISO supplementation (7.85 mmol/kg DM for isobutyrate and 3.44 mmol/kg DM for 2-methylbutyrate, respectively). Enteric CH4 and chewing activity (rumination and eating time) were measured using the GreenFeed system and sensor-based ear tag system, respectively. The particle size of each diet and ort from individual cows was measured using the Penn State Particle Separator, and a sorting index was calculated. A sorting index of 100 indicates no sorting, while values above or below 100 indicate sorting for or against, respectively. Data were analyzed using a mixed model including FL, ISO, and FL × ISO as fixed effects and block as a random effect (lme4 in R). Our result shows that ISO increased sorting index for long particle in LF (96.1 vs. 109; P < 0.01) but decreased it in HF (100.8 vs. 92.5; P = 0.04). In contrast, ISO did not affect the physically effective particle sorting index (P = 0.51) or intake (P = 0.27) regardless of FL. In alignment with the long particle sorting index, ISO decreased eating and chewing time in the HF but increased them in the LF diet (P < 0.01). In contrast, rumination time was comparable between FL (P = 0.70) and ISO levels (P = 0.19). In the LF diet, ISO supplementation reduced daily CH4 production (g/d) by 9% and intensity (g/kg of MY) by 18% (P < 0.01). In the HF diet, ISO supplementation led to a 10% increase in daily CH4 (P < 0.01) but did not change CH4 intensity (P = 0.17; g/kg of MY) due to improved milk production. Overall, ISO altered feed sorting, feeding behaviors and enteric CH4 emissions depending on FL.
Electrophysiology of renal capillary membranes: gel concept applied and Starling model challenged.

Abstract:In the classical Starling model the hydrostatic pressure in the pores is generally lower than that in capillary plasma, a phenomenon that necessitates the assumption of a rigid porous membrane. In flexible gel membranes, the capillary pressure is suggested to be balanced by a gel swelling pressure generated by negative fixed charges. Regarding the fluid transfer, the transmembranous electrical potential gradient will generate a net driving electroosmotic force. This force will be numerically similar to the net driving Starling force in small pores, but distinctly different in large pores. From previous data on the hydrostatic and colloid osmotic forces, the fixed charge density at the two interfaces of 1) the glomerular and 2) the peritubular capillary membrane were calculated and used to predict the flux of a series of charged protein probes. The close fit to the experimental data in both the capillary beds is in line with the gel concept presented. The gel concept (but hardly a rigid membrane) explains the ability of capillary membranes to alter their permeability in response to external forces. Gel membranes can furthermore be predicted to have a self-rinsing ability, as entrapped proteins will increase the local fixed charge density, leading to fluid entry into the region between the particle and the pore rim, which by consequent widening of the channel will facilitate extrusion of trapped proteins.
Levels and degradability of crude protein in digestive metabolism and performance of dairy cows

Abstract:Two experiments were conducted to evaluate the effect of the level and degradability of crude protein (CP) on the digestive metabolism and productive performance of dairy cows. In both experiments, 15 Holstein cows with 585 ± 40 kg of body weight were distributed in a Latin square design with five contemporary squares, three periods of 21 days and three treatments. In experiment 1, treatments consisted of three CP levels (130, 160 or 180 g CP/kg DM), while in experiment 2, the treatments consisted of three levels of rumen degradable protein (RDP; 80, 100 or 120 g RDP/kg DM) in diets with average of 163 g CP/kg DM. Variables evaluated in both experiments were dry matter intake (DMI), total apparent digestibility, milk yield (MY) and composition, ruminal fermentation and N balance. In experiment 1, the increase of CP from 130 to 180 linearly increased the organic matter, CP, neutral detergent fiber (NDF) and acid detergent fiber (ADF) intake (kg) and the apparent total digestibility coefficient of DM and CP. In addition, a linear increase of MY, fat corrected milk (FCM) and daily production of fat, protein, lactose, casein and total solids was observed. A linear increase in ruminal ammoniacal nitrogen (NH3-N) concentration and nitrogen excretion in milk, feces and urine was also observed. However, there was no observed effect on SCFA concentration. In experiment 2, the increase of the RDP from 80 to 120 increased the DMI, MY, FCM, milk protein content and digestibility coefficient of the NDF, ADF and ethereal extract. Additionally, there was an increase in NH3-N concentration and milk nitrogen excretion. The studies indicated that the increase of CP content up to 100 g RDP/kg DM increased the DMI and the productive performance of the cows, but also increased urine N. Thus, it is desirable that the increase of the CP through the increase of the RDP is carried out up to 100 g of RDP/kg DM, since there is elimination of nitrogen, decrease of milk yield and decrease of propionic acid in values above that level.
Effect of Varying Dietary Ratios of Alfalfa Silage to Corn Silage on Production and Nitrogen Utilization in Lactating Dairy Cows

Abstract:Twenty-eight (8 ruminally cannulated) lactating, multiparous Holstein cows were blocked by DIM and randomly assigned to 7 replicated 4 x 4 Latin squares (28-d periods) to investigate the effects of different dietary ratios of alfalfa silage (AS) to corn silage (CS) on production, N utilization, apparent digestibility, and ruminal metabolism. The 4 diets contained (dry matter basis): A) 51% AS, 43% rolled high-moisture shelled corn (HMSC), and 3% solvent soybean meal (SSBM); B) 37% AS, 13% CS, 39% HMSC, and 7% SSBM; C) 24% AS, 27% CS, 35% HMSC, and 12% SSBM; and D) 10% AS, 40% CS, 31% HMSC, and 16% SSBM. Dietary crude protein contents were 17.2, 16.9, 16.6, and 16.2% for diets A, B, C, and D. All 4 diets were high in energy, averaging 49% nonfiber carbohydrates and 24% neutral detergent fiber. Intake of dry matter, yield of milk, 3.5% fat-corrected milk and fat, milk fat content, and apparent digestibility of neutral detergent fiber and acid detergent fiber all decreased linearly when CS replaced AS. Effects on fiber digestion and milk fat may have been due to increasing fluctuation in ruminal pH and time the pH remained < 6.0 when CS replaced AS. Milk protein content increased linearly with increasing CS, but there were no differences in protein yield. There were linear increases in apparent N efficiency and decreases in N excreted in urine and feces when CS replaced AS. Production was depressed on the diet highest in CS. Quadratic analysis indicated that milk and protein yields were maximal at dietary AS:CS ratios of, respectively, 37:13 and 31:19. No diet minimized N excretion without negatively affecting production. Diet C, with an AS:CS ratio of 24:27, was the best compromise between improved N efficiency and sustained production. Because CS is complementary with AS, it is recommended that CS be fed in AS-based diets to maintain milk yield while improving N utilization.

The aim of this study was to evaluate dietary protein utilization at different crude protein (CP) levels and to predict nitrogen content in faeces on the basis of nutritional parameters and milk urea nitrogen content (MUN, mg dL -1 ). The study was conducted on three groups (A, B, C) of lactating dairy cows (8 cows per group, including Latvian Brown and Holstein Black and White breeds) from 10 to 30 days in milk. Total mixed rations containing different levels of CP (approximately 18.0%, 17.5% and 17.0% for A, B and C, respectively) were fed. The amount of feed consumed by each cow was measured and feed samples collected during the trial. Milk yield (kg d-1 -1 ) and faeces amount were recorded, and samples were collected at day 21 of the study for further analysis. Feed samples were analysed for CP, net energy for lactation (NEL, MJ kg -1 ) and other parameters. Milk samples were analysed for fat (%), total protein (%), casein (%) and urea content (mg dL -1 ). The statistical investigation was conducted using ANOVA, and correlation and regression analyses. The results showed that milk yield, fat, total protein, casein, urea, and MUN were not significantly different among groups being not affected by the dietary CP levels. The correlation between faecal nitrogen content and CP content in feed was moderately positive and statistically significant (r=0.44, P=0.03), while the correlation between faecal nitrogen content and MUN was moderately negative and showed tendency towards significance (r=-0.39, P=0.06). The regression analysis showed that feed CP explained approximately 20% of faeces nitrogen content. Effects of Dietary Protein Level on Growth Performance and Nitrogen Excretion of Dairy Heifers

Abstract:Objective Protein supplementation is costly and can result in excess nitrogen (N) excretion. The objective of this study was to evaluate the effects of feeding different levels of dietary protein on average daily gain, body size, rumen fermentation, and nitrogen excretion of 8 to 10 month-old Holstein heifers. Methods Thirty-six Holstein heifers were divided into 12 blocks according to age (273±6.2 d) and were randomly assigned to diets containing a low (10.2% dry matter [DM]), medium (11.9% DM), or high (13.5% DM) level of dietary crude protein (CP). All diets contained approximately 70% roughage and 30% concentrate with similar dietary metabolizable energy (ME) content (2.47 Mcal/kg). Results Dry matter intake did not differ among the treatments, and average daily gain increased with the increasing dietary protein, 0.79, 0.95, 0.97 kg/d for low, medium, and high group, respectively. Body height increased linearly with increasing dietary CP but no other significant differences in body dimensions were found among the treatments. The increased ratios of dietary CP improved the rate of rear teat length growth remarkably (p<0.05). There was no difference in rumen pH or ruminal major volatile fatty acid (acetate, propionate, and butyrate) concentration among the 3 diets, but rumen ammonia-N concentration increased with the higher dietary CP (p<0.05). Increasing N intake led to increased total N excretion; urinary N excretion was significantly increased (p<0.05) but fecal N excretion was similar among the treatments. Conclusion These data suggest that the diet containing 11.9% CP (ME 2.47 Mcal/kg) could meet the maintenance and growth requirements of 9 to 11 month-old Holstein heifers gaining approximately 0.9 kg/d.
Effect of nitrate supplementation, dietary protein supply, and genetic yield index on performance, methane emission, and nitrogen efficiency in dairy cows

Abstract:The objective was to investigate the effect of nonprotein nitrogen source, dietary protein supply, and genetic yield index on methane emission, N metabolism, and ruminal fermentation in dairy cows. Forty-eight Danish Holstein dairy cows (24 primiparous cows and 24 multiparous cows) were used in a 6 × 4 incomplete Latin square design with 4 periods of 21-d duration. Cows were fed ad libitum with the following 6 experimental diets: diets with low, medium, or high rumen degradable protein (RDP):rumen undegradable protein (RUP) ratio (manipulated by changing the proportion of corn meal, corn gluten meal, and corn gluten feed) combined with either urea or nitrate (10 g NO3-/kg of dry matter) as nonprotein nitrogen source. Samples of ruminal fluid and feces were collected from multiparous cows, and total-tract nutrient digestibility was estimated using TiO2 as flow marker. Milk samples were collected from all 48 cows. Gas emission (CH4, CO2, and H2) was measured by 4 GreenFeed units. We observed no significant interaction between dietary RDP:RUP ratio and nitrate supplementation, and between nitrate supplementation and genetic yield index on CH4 emission (production, yield, intensity). As dietary RDP:RUP ratio increased, intake of crude protein, RDP, and neutral detergent fiber and total-tract digestibility of crude protein linearly increased, and RUP intake linearly decreased. Yield of milk, energy-corrected milk, and milk protein and lactose linearly decreased, whereas milk fat and milk urea nitrogen concentrations linearly increased as dietary RDP:RUP ratio increased. The increase in dietary RDP:RUP ratio resulted in a linear increase in the excretion of total purine derivatives and N in urine, but a linear decrease in N efficiency (milk N in % of N intake). Nitrate supplementation reduced dry matter intake (DMI) and increased total-tract organic matter digestibility compared with urea supplementation. Nitrate supplementation resulted in a greater reduction in DMI and daily CH4 production and a greater increase in daily H2 production in multiparous cows compared with primiparous cows. Nitrate supplementation also showed a greater reduction in milk protein and lactose yield in multiparous cows than in primiparous cows. Milk protein and lactose concentrations were lower for cows receiving nitrate diets compared with cows receiving urea diets. Nitrate supplementation reduced urinary purine derivatives excretion from the rumen, whereas N efficiency tended to increase. Nitrate supplementation reduced proportion of acetate and propionate in ruminal volatile fatty acids. In conclusion, no interaction was observed between dietary RDP:RUP ratio and nitrate supplementation, and no interaction between nitrate supplementation and genetic yield index on CH4 emission (production, yield, intensity) was noted. Nitrate supplementation resulted in a greater reduction in DMI and CH4 production, and a greater increase in H2 production in multiparous cows than in primiparous cows. As the dietary RDP:RUP ratio increased, CH4 emission was unaffected and RDP intake increased, but RUP intake and milk yield decreased. Genetic yield index did not affect CH4 production, yield, or intensity.
Dietary CP and digestion kinetics influence BW loss, litter weight gain, and reproduction by affecting postprandial amino acid metabolism in lactating sows

Abstract:To avoid a high body protein mobilization in modern lean sows during lactation, an adequate dietary amino acid (AA) supply and an efficient AA utilization are crucial. This study evaluated the effects of dietary CP and in vitro protein digestion kinetics on changes in sow body condition, litter weight gain, milk composition, blood metabolites, protein utilization efficiency and subsequent reproductive performance. We hypothesized that a slower digestion of dietary protein would improve AA availability and utilization. In total, 110 multiparous sows were fed one of four lactation diets in a 2 × 2 factorial design, with two CP concentrations: 140 g/kg vs 180 g/kg, and two protein digestion kinetics, expressed as a percentage of slow protein (in vitro degradation between 30 and 240 min): 8 vs 16% of total protein. Feeding sows the high CP diets reduced sow weight loss (Δ = 7.6 kg, P < 0.01), estimated body fat loss (Δ = 2.6 kg, P = 0.02), and estimated body protein loss (Δ = 1.0 kg, P = 0.08), but only at a high percentage of slow protein. A higher percentage of slow protein increased litter weight gain throughout lactation (Δ = 2.6 kg, P = 0.04) regardless of CP concentrations, whereas a higher CP only increased litter weight gain during week 3 of lactation (Δ = 1.2 kg, P = 0.01). On Day 15 postfarrowing, serial blood samples were taken from a subsample of sows fed with the high CP diets. In these sows, a high percentage of slow protein resulted in higher plasma AA concentrations at 150 and 180 min after feeding (Δ = 0.89, P = 0.02, Δ = 0.78, P = 0.03, mmol/L, respectively) and lower increases in urea at 90 and 120 min after feeding (Δ = 0.67, P = 0.04, Δ = 0.70, P = 0.03, mmol/L, respectively). The higher dietary CP concentration increased total nitrogen loss to the environment (Δ = 604 g, P < 0.01) with a reduction of protein efficiency (Δ = 14.8%, P < 0.01). In the next farrowing, a higher percentage of slow protein increased subsequent liveborn litter size (Δ = 0.7, P < 0.05). In conclusion, feeding sows with a high dietary CP concentration alleviated maternal weight loss during lactation when the dietary protein digestion rate was slower, but lowered protein efficiency. A slower protein digestion improved litter weight gain, possibly by reducing AA oxidation and improving plasma AA availability, thus, improving protein efficiency.
Metabolic and endocrine responses to short-term nutrient imbalances in the feed ration of mid-lactation dairy cows

Abstract:Short-term imbalances of dietary nutrients occur during natural fluctuations in roughage quality (e.g. on pasture) or temporal shortages of supplementary feed components. In contrast to a deficiency, macronutrients (i.e. carbohydrates, proteins, lipids) beyond the adequate supply with other nutrients may, for instance, alter milk composition, increase BW or result in a greater excretion of nitrogen. Especially dairy cows with a moderate performance, in mid- or late lactation, or in extensive farming systems may be exposed to imbalanced rations. A better understanding of metabolic and endocrine responses depending on macronutrient supply may help to precisely feed dairy cows. The present study investigated short-term metabolic and endocrine responses to different levels of concentrates formulated to particularly provide one major macronutrient source (carbohydrates, proteins or lipids). Based on parity number, lactational stage, milk yield and BW, nine mid-lactating cows (211 ± 19 days in milk) were grouped into three blocks of three animals each. Concentrates (aminogenic: rich in CP and nitrogen sources; glucogenic: high content of carbohydrates and glucogenic precursors; lipogenic: high lipid content) were fed in addition to hay in a factorial arrangement at increasing levels from 2.5 to 7.5 kg/d during 9 d. Milk yield, BW and feed intake were recorded daily. Blood and milk were sampled every 3 d at the end of each concentrate level. Milk fat, protein, lactose and urea contents were determined. In blood, concentrations of various metabolites, endocrine factors and enzyme activities (e.g. glucose, non-esterified fatty acids (NEFAs), β-hydroxybutyrate, urea, cholesterol, triglycerides, insulin, glucagon, aspartate aminotransferase (ASAT), gamma-glutamyltransferase (GGT) and glutamate dehydrogenase activity (GLDH)) were measured. Milk yield, milk composition and BW were not affected by type and level of concentrates. Feed intake increased in cows with greater amounts of the aminogenic and lipogenic concentrate compared with the glucogenic concentrate. Milk and plasma urea concentrations were elevated in the aminogenic and to a lesser extent in the lipogenic treatment compared with the glucogenic treatment. Glucose concentrations in plasma were not affected by treatments, whereas insulin and glucagon increased, and NEFA concentrations decreased only in cows fed 7.5 kg/d aminogenic concentrate compared with the glucogenic and lipogenic treatment. Activities of ASAT, GGT and GLDH as well as the total antioxidant capacity were not affected by diets. In conclusion, immediate metabolic and endocrine responses were observed due to the short-term dietary changes. Particularly, a surplus of nitrogen supply via the aminogenic diet affected metabolic responses and stimulated insulin and glucagon secretion.
