Abstract:Harmful cyanobacterial blooms will be more intense and frequent in the future, contaminating surface waters with cyanotoxins and posing a threat to communities heavily reliant on surface water usage for crop irrigation. Constructed wetlands (CWs) are proposed to ensure safe crop irrigation, but more research is needed before implementation. The present study operated 28 mesocosms in continuous mode mimicking horizontal sub-surface flow CWs. Mesocosms were fed with synthetic lake water and spiked periodically with two cyanotoxins, microcystin-LR (MC-LR) and cylindrospermopsin (CYN), at environmentally relevant cyanotoxins concentrations (10 μg L-1). The influence of various design factors, including plant species, porous media, and seasonality, was explored. The mesocosms achieved maximum MC-LR and CYN mass removal rates of 95 % and 98 %, respectively. CYN removal is reported for the first time in CWs mimicking horizontal sub-surface flow CWs. Planted mesocosms consistently outperformed unplanted mesocosms, with Phragmites australis exhibiting superior cyanotoxin mass removal compared to Juncus effusus. Considering evapotranspiration, J. effusus yielded the least cyanotoxin-concentrated effluent due to the lower water losses in comparison with P. australis. Using the P-kC* model, different scaling-up scenarios for future piloting were calculated and discussed. Additionally, bacterial community structure was analyzed through correlation matrices and differential taxa analyses, offering valuable insights into their removal of cyanotoxins. Nevertheless, attempts to validate microcystin-LR biotransformation via the known mlrA gene degradation pathway were unfruitful, indicating alternative enzymatic degradation pathways occurring in such complex CW systems. Further investigation into the precise molecular mechanisms of removal and the identification of transformation products is needed for the comprehensive understanding of cyanotoxin mitigation in CW. This study points towards the feasibility of horizontal sub-surface flow CWs to be employed to control cyanotoxins in irrigation or recreational waters.
Fungal removal of cyanotoxins in constructed wetlands: The forgotten degraders

Abstract:Harmful cyanobacterial blooms have increased globally, releasing hazardous cyanotoxins that threaten the safety of water resources. Constructed wetlands (CWs) are a nature-based and low-cost solution to purify and remove cyanotoxins from water. However, bio-mechanistic understanding of the biotransformation processes expected to drive cyanotoxin removal in such systems is poor, and primarily focused on bacteria. Thus, the present study aimed at exploring the fungal contribution to microcystin-LR and cylindrospermopsin biodegradation in CWs. Based on CW mesocosms, two experimental approaches were taken: a) amplicon sequencing studies were conducted to investigate the involvement of the fungal community; and b) CW fungal isolates were tested for their microcystin-LR and cylindrospermopsin degradation capabilities. The data uncovered effects of seasonality (spring or summer), cyanotoxin exposure, vegetation (unplanted, Juncus effusus or Phragmites australis ) and substratum (sand or gravel) on the fungal community structure. Additionally, the arbuscular mycorrhizal fungus Rhizophagus and the endophyte Myrmecridium showed positive correlations with cyanotoxin removal. Fungal isolates revealed microcystin-LR-removal potentials of approximately 25 % in in vitro biodegradation experiments, while the extracellular chemical fingerprint of the cultures suggested a potential intracellular metabolization. The results from this study may help us understand the fungal contribution to cyanotoxin removal, as well as their ecology in CWs.

Abstract:Increasingly prevalent Microcystis blooms and the propagation of the associated resistance genes represent global environmental problems. Constructed wetlands (CWs) are a cost-effective technology used for wastewater treatment. In this study, the herb Alisma orientale and three industrial byproducts, namely, blast furnace slag, biochar, and sawdust, were selected to construct mini-CW units. Their potential to remediate toxic Microcystis and their influences on the behaviors of antibiotic-resistant genes (ARGs, sul1, sul2, and intl1) were analyzed. Approximately 98.46% of Microcystis cells were removed by the sawdust-based CW in just 2 d, wherein <0.37 μg/L residual microcystin (MC)-LR was detected, with a removal efficiency of >96.47%, which is potentially caused by the higher relative abundance of MC-degrading gene mlrA on the substrate. Lower target ARG accumulations in the sawdust-based CW may be attributed to the lower intl1 relative abundance and microbial function mobile element content, which could influence horizontal gene transfer. In three sequential batches for the treatment of eutrophic lake water, six sawdust-based CW units were assembled into CW microcosms. The efficiency of removal of Microcystis and MC-LR by planted CW microcosms ranged between 92.00% and 95.88% and between 86.48% and 94.82%, respectively; this was significantly (P < 0.05) higher than that by unplanted ones. Less accumulation of target ARGs was also observed in planted CWs. Planting considerably improved nitrogen removal, possibly owing to the enrichment of genes involved in the KEGG nitrogen metabolism pathway in the substrate through metagenomic analysis.

Intrauterine insemination IUI: the effect of ovarian stimulation and infertility diagnosis on pregnancy outcome.

Abstract:We determined the intrauterine insemination (IUI) pregnancy outcome in the same group of patients when applying different methods of ovulation induction. A group of patients with unexplained (no. 46) and male factor infertility (no. 101) consented to have the following treatment protocol at the American University of Beirut-Infertility Center: IUI to be performed in three natural ovulatory cycles in all patients, then in three clomiphene citrate (CC) stimulated cycles in the remaining non-pregnant patients, and then three cycles with controlled ovarian hyperstimulation (COH) in the remaining group. Of the total 147 patients 130, 138 and 123 underwent 273 natural, 278 CC and 266 COH IUI cycles, respectively. Semen processing for IUI was done by washing the sperm twice and using the swim-up technique. The chi-square test was used for statistical analysis. Pregnancy rate per cycle of IUI with COH (9.8%) was significantly higher than that of IUI in natural cycles (3.3%) but approached significance when compared to IUI with CC cycles (5.4%). Also unexplained infertility cases had a significantly higher pregnancy rate (58.7%) when compared to that of male factor cases (22.8%). IUI still has a place in the treatment of infertility due to selective causes. Combined with COH, IUI gives the best pregnancy rate although its benefit with natural or CC cycles remains obvious.

Abstract:The removal of organic micropollutants (OMPs) has been investigated in constructed wetlands (CWs) operated as bioelectrochemical systems (BES). The operation of CWs as BES (CW-BES), either in the form of microbial fuel cells (MFC) or microbial electrolysis cells (MEC), has only been investigated in recent years. The presented experiment used CW meso-scale systems applying a realistic horizontal flow regime and continuous feeding of real urban wastewater spiked with four OMPs (pharmaceuticals), namely carbamazepine (CBZ), diclofenac (DCF), ibuprofen (IBU) and naproxen (NPX). The study evaluated the removal efficiency of conventional CW systems (CW-control) as well as CW systems operated as closed-circuit MFCs (CW-MFCs) and MECs (CW-MECs). Although a few positive trends were identified for the CW-BES compared to the CW-control (higher average CBZ, DCF and NPX removal by 10-17% in CW-MEC and 5% in CW-MFC), these proved to be not statistically significantly different. Mesoscale experiments with real wastewater could thus not confirm earlier positive effects of CW-BES found under strictly controlled laboratory conditions with synthetic wastewaters.

Abstract:The bacterial community in the sediment of Dianchi Lake,which can biodegrade microcystins,was enriched with the methods of using extracted and purified MCs as the carbon and nitrogen sources from low to high concentrations of MCs in the culture medium for a long time.MCRR and LR with each initial concentration of about 50 mg/L and 30 mg/L were completely biodegraded by this bacterial community in 3 d.It showed that the growth of this bacterial community was promoted however the biodegradation of MCs could not be improved in the presences of other various of carbon and nitrogen containing compounds,which indicated that MCs could be used as both the carbon and the nitrogen sources for the growth of this bacterial community,and the presence of other organic compounds can not promote the biodegradation of MCs in natural water bodies.
Validation of a headspace GC method for the analysis of a pyrolytic product of methamphetamine in urine.

Abstract:A method has been developed and validated using headspace GC-FID for the identification of 1-phenylpropene in urine. This compound is a pyrolytic product of methamphetamine that has been previously proposed as a marker for smoked methamphetamine. The instrumentation used is the same as employed for blood alcohol determination. The extraction-free procedure is rapid, simple, and quantitative using 2-phenylpropene as the internal standard. The method was validated for linearity over a range of 0.1-20 microg/mL with a limit of detection of 0.05 microg/mL, limit of quantification of 0.1 microg/mL, interday accuracy within 3.2 to -5.3%, intraday accuracy better than 7.5%, interday precision of 7.5 to 10.7%, intraday precision of 2 to 8.6%, and recovery above 80%. For the robustness determination in urine, the accuracy of four different sources of urine at the mid control level (1 microg/mL) ranged from 1.6 to 19% error. The % relative standard deviation of the different urine sources ranged from 3.1 to 11%. Urine samples from nine methamphetamine-positive cases investigated by the Office of the Chief Medical Examiner of West Virginia were included in the study. 1-Phenylpropene was found in two methamphetamine-positive cases (0.25 and 0.44 microg/mL).

Abstract:Cylindrospermopsin (CYN) is a cyanotoxin found in natural waters, with potential risk to human health through the inhibition of protein synthesis. Despite the implementation of conventional water treatment procedures, complete removal of CYN remains a question due to its heat-stable nature. Hence, contamination of water sources with CYN is a challenge in providing safe drinking water throughout the world. The present study was conducted to test the ability to degrade CYN at 280C and pH 7, of four bacterial strains: Bacillus cereus-Y, Bacillus cereus-S (B. cereus-S), Micrococcus luteus, and Alcaligenes faecalis, which were previously isolated from different water sources as different hydrocarbon degraders. The CYN degradation kinetics of each bacterial species were studied using High Performance Liquid Chromatography. The greatest CYN degradation (28.22 ± 0.24%) was shown by the bacterium B. cereus-S in 5.0 mg/L CYN within 14 days. The CYN degradation by the other strains was lower than 10% under the same conditions. Further studies employing different initial concentrations of CYN revealed that B. cereus-S could degrade lower CYN concentrations at a higher percentage (1.0 mg/L, 2.5 mg/L, and 5.0 mg/L of CYN removal percentages were 36.83 ± 2.43%, 32.25 ± 1.25%, and 24.72 ± 0.40%, respectively, after 14 days of incubation at 280C and pH 7). The maximum average degradation rates were recorded for 1.0 mg/L, 2.5 mg/L, and 5.0 mg/L CYN on the 6th (0.05 ± 0.00 mg/L/day), 8th (0.04 ± 0.01 mg/L/day), and 12th (0.02 ± 0.01 mg/L/day) days of incubation, respectively. The study showed the potentiality of the bacterium B. cereus-S on the application for degrading CYN among the tested bacteria species.

Abstract:Microcystins (MC) produced by species of cyanobacteria including Microcystis, Anabaena, and Aphanizomenon are a group of monocyclic hepatotoxins posing serious threat to public health. Microcystin-LR (MC-LR) is the most toxic and frequently encountered microcystin variant in the environment, and thus removal of this toxin using bacteria was shown to be a reliable, efficient, and cost-effective method that avoids utilization of chemicals that may produce potentially harmful by-products. The aim of this study was to determine whether a novel indigenous bacterial community designated YFMCD1 was effective in destroying MC. In addition, the influence of environmental factors such as temperature, MC concentration, and pH was examined on the effectiveness of YFMCD1 to degrade MC-LR. MC-degradation products were identified by high performance liquid chromatography coupled with an ultra-high resolution LTQ Orbitrap Velos Pro ETD mass spectrometry equipped with electrospray ionization interface (HPLC-ESI-MS). MC-LR underwent maximal degradation at rate of 0.5 mu g/ml/hr with YFMCD1 containing Klebsiella sp. termed YFMCD1-1 or Stenotrophomonas sp. termed YFMCD1-2. Moreover, Adda (3-amino-9-methoxy-2, 6, 8-trimethyl-10-phenyldeca-4, 6-dienoic acid) is a constituent within the MC-LR molecule found to be responsible for biological activity expression and critical for MC-induced toxicity, which is also degraded by YFMCD1. The results showed that YFMCD1 effectively degraded MC-LR. The degradation rate was significantly affected by temperature, pH, and MC-LR concentrations. Data indicate that this bacterial community may prove beneficial in bioremediation of lakes containing MC.

Abstract:Harmful cyanobacterial blooms (HCBs) frequently occur in eutrophic freshwater ecosystems worldwide. Microcystins (MCs) are considered to be the most prominent and toxic metabolites during HCBs. MCs may be harmful to human and animal health through drinking water and recreational water. Biodegradation is eco-friendly, cost-effective and one of the most effective methods to remove MCs. Many novel MC-degrading bacteria and their potential for MCs degradation have been documented. However, it is a challenge to apply the free MC-degrading bacterial cells in natural environments due to the long-term operational instability and difficult recycling. Immobilization is the process of restricting the mobility of bacteria using carriers, which has several advantages as biocatalysts compared to free bacterial cells. Biological water treatment systems with microbial immobilization technology can potentially be utilized to treat MC-polluted wastewater. In this review article, various types of supporting materials and methods for microbial immobilization and the application of bacterial immobilization technology for the treatment of MCs-contaminated water are discussed. This article may further broaden the application of microbial immobilization technology to the bioremediation of MC-polluted environments.

Abstract:Cyanobacterial blooms are expected to become more frequent and severe in surface water reservoirs due to climate change and ecosystem degradation. It is an emerging challenge that especially countries relying on surface water supplies will face. Nature-based solutions (NBS) like constructed wetlands and biofilters can be used for cyanotoxin remediation. Both technologies are reviewed and critically assessed for different types of water resources. The available information on cyanotoxins (bio)transformation products (TPs) is reviewed to point out the potential research gaps and to disclose the most reliable enzymatic degradation pathways. Knowledge gaps were found, such as information on the performance of the revised NBS in pilot and full scales, the removal processes covering different cyanotoxins (besides the most widely studied microcystin-LR), and the difficulties for real-world implementation of technologies proposed in the literature. Also, most studies focus on bacterial degradation processes while fungi have been completely overlooked. This review also presents an up-to-date overview of the transformation of cyanotoxins, where degradation product data was compiled in a unified library of 22 metabolites for microcystins (MCs), 7 for cylindrospermopsin (CYN) and 10 for nodularin (NOD), most of them reported only in a single study. Major gaps are the lack of environmentally relevant studies with TPs in pilot and full-scale scale treatment systems, information on TP's toxicity, as well as limited knowledge of environmentally relevant degradation pathways. NBS have the potential to mitigate cyanotoxins in recreational and irrigation waters, enabling the water-energy-food nexus and avoiding the degradability of the ecosystems.

Microbe-mediated Simultaneous Nitrogen Reduction and Sulfamethoxazole/n-Acetylsulfamethoxazole Removal in Lab-Scale Constructed Wetlands.

Abstract:Constructed wetlands (CWs) are increasingly used to treat complex pollution such as nitrogen and emerging organic micropollutants from anthropogenic sources. In this study, the denitrification, anaerobic ammonium oxidation, dissimilatory nitrate reduction to ammonium, and nitrous oxide release rates following exposure to the frequently detected sulfonamides sulfamethoxazole (SMX) and its human metabolite, N-acetylsulfamethoxazole (N-SMX), were investigated in lab-scale CWs. Over a period of 190 d, the denitrification rates were noticeably inhibited in the SMX and N-SMX groups at week 5. Subsequently, the denitrification rates recovered, accompanied by an increase in the relevant nitrogen reduction and antibiotic resistance genes (ARGs). The composition of the microbial community also changed during this process. After the denitrification rates recovered, Burkholderia_Paraburkholderia and Gordonia exhibited a significant positive correlation with SMX exposure, which simultaneously reduced nitrate concentrations and degraded antibiotics. Burkholderia_Paraburkholderia is a key carrier of ARGs. Finally, nitrogen reduction (> 90%) and antibiotic removal (> 80%) also recovered in both SMX- and N-SMX-exposed lab-scale CWs during the operation, which revealed the interaction of SMX or N-SMX removal and nitrogen reduction.

Abstract:Microcystin-leucine-arginine (MC-LR), a cyclic potentially carcinogenic hepatotoxin, occurs frequently in aquatic habitats worldwide and seriously threatens ecosystem and public health. Limited effectiveness of physicochemical treatments to remove MC-LR from drinking water has led to a search for alternative cost-effective and environment friendly biodegradation strategies. Obtaining MC-degrading bacteria and understanding their MC-degrading mechanisms are outstanding challenges. Here, a novel indigenous bacterium named Sphingopyxis sp. YF1 with a high efficient capacity for MC-degradation was successfully isolated from eutrophic Lake Taihu. Through integrating mass spectrometer and multi-omics analyses accompanied by functional verification of certain genes and proteins, a complete MC-degradation pathway was firstly identified, in which MC-LR was sequentially degraded into linearized MC-LR, tetrapeptide, Adda, phenylacetic acid, and finally potential product CO2. Some specific proteins such as microcystinase, linearized-microcystinase, tetrapeptidease and PAAase responsible for this pathway were identified. This study pioneeringly demonstrated that MC-LR can be completely degraded through natural remediation processes and revealed a significant potential for MC-LR biodegradation in both natural environment and engineered systems.

Abstract:yield thousands of single nucleotide polymorphisms (SNPs), which can be used to identify polygenic contributors to disease and ultimately to design individualized prognostic strategies and therapies. Exploiting these, however, will require powerful and automated scoring methods. Current techniques have their roots in allele-specific oligonucleotide hybridization (ASOH)1. In its basic form (i.e., hybridization, stringent washing, and signal detection), ASOH is limited by the difficult challenge of defining discriminatory assay conditions. Therefore, newer methods include additional steps furnishing more robust allele scoring. Such procedures include the ligation chain reaction (ASOH plus selective ligation and amplification)2, mini-sequencing (ASOH plus a single base extension)3, and DNA “chips” (miniaturized ASOH with multiple oligonucleotide probe arrays)4. Alternatively, ASOH with singleor dual-labeled probes has been merged with PCR, as in the 5 ́ exonuclease assay5, and with molecular beacons6. While effective, these methods also entail considerable optimization efforts and/or costly enzymatic or oligonucleotide labeling steps. We discuss here a new SNP scoring method, dynamic allele-specific hybridization (DASH), that does not suffer these drawbacks. The key to DASH is dynamic heating and coincident monitoring of DNA denaturation. No additional enzymes or reaction steps are involved. DASH therefore retains the simplicity of ASOH, but achieves unambiguous discrimination of all SNP variations using standardized reaction conditions. The assay is conducted in a microtiter plate format compatible with automation, and uses convenient fluorescence signal detection.
The Removal Mechanisms of Microcystin-Lr Through Electrolysis Biofilters

Abstract:Microcystin-leucine arginine (MC-LR) is the most common cyanotoxin produced by harmful cyanobacterial blooms with a cyclic structure which is hard to be degraded. In this study novel electrolysis biofilters (EBFs) with two Ti/RuO2-IrO2 anodes and one iron cathode were developed for intensified MC-LR degradation in contaminated water. Test results showed that biochar-EBF could efficiently remove intracellular and extracellular MCLR. The MC-LR concentration decreased to 0.18 +/- 0.06 ug center dot L-1 when the mean influent concentration was 2.51 +/- 0.08 ug center dot L-1 with a removal rate of 92.83 +/- 1.2%. The degradation rate was 0.47 +/- 0.006 ug center dot(m2 center dot h) -1, and the energy consumption was 0.3 kWh center dot m- 3, which meets the World Health Organization standard for drinking water quality. A removal kinetic study found that electrolysis, adsorption and biodegradation were the main roles in the degradation process. The removal curves were more consistent with the pseudo-second-order model, as the R2 were all 0.99. The biofilm bacteria genera were mainly chemoheterotrophic, aerobic chemoheterotrophic, and methylotrophic. These biofilm bacteria genera could metabolize MC-LR and its intermediate products. According to the liquid chromatography-high resolution mass spectrometry (LC-HRMS) analysis of the MC-LR degradation intermediates by biochar-EBF, the degradation pathway of MC-LR first broke the amide bond between Adda and arginine, and then formed a linear MC-LR, which further degraded into a tetrapeptide compound. That compound was then further broken into phenylacetic acid (PAA) and small molecule amino acids. Thus, biochar-EBF enhanced the MC-LR degradation, and the biological toxicity of MC-LR was also reduced.
Naturalization of treated wastewater by a constructed wetland in a water-scarce Mediterranean region

Abstract:The effluents from conventional wastewater treatment plants (WWTP), even if accomplishing quality regulations, substantially differ in their characteristics with those of waters in natural environments. Constructed wetlands (CWs) serve as transitional ecosystems within WWTPs, mitigating these differences and restoring natural features before water is poured into the natural environment. Our study focused on an experimental surface-flow CW naturalizing the WWTP effluent in a semiarid area in Eastern Spain. Despite relatively low pollutant concentrations entering the CW, it effectively further reduced settled organic matter and nitrogen. Dissolved organic matter (DOM) reaching the CW was mainly protein-like, yet optical property changes in the DOM indicated increased humification, aromaticity, and stabilization as it flowed through the CW. Flow cytometry analysis revealed that the CW released less abundant but more active bacterial populations than those received. MiSeq Illumina sequencing highlighted changes in the prokaryotic community composition, with phyla Proteobacteria, Bacteroidetes, Firmicutes, and Actinobacteria dominating the CW outflow. Functional prediction tools (FaproTax and PICRUSt2) demonstrated a shift towards microbial guilds aligned with those of the natural aquatic environments, increased aerobic chemoheterotrophs, photoautotrophs, and metabolic reactions at higher redox potentials. Enhanced capabilities for degrading plant material correlated well with changes in the DOM pool. Our findings emphasize the role of CWs in releasing biochemically stable DOM and functionally suited microbial populations for natural receiving environments. Consequently, we propose CWs as a naturalization nature-based solution (NBS) in water-scarce regions like the Mediterranean, where reclaimed discharged water can significantly contribute to ecosystem's water resources compared to natural flows.

Abstract:To innovate the design of water treatment technology for algal toxin removal, this research investigated the mechanisms of cyanotoxin microcystin-LR (MC-LR) removal by a coupled adsorption-biodegradation. Eight types of woody carbonaceous adsorbents with and without Sphingopyxis sp. m6, a MC-LR degrading bacterium, were tested for MC-LR removal in water. All adsorbents showed good adsorption capability, removing 40 % to almost 100 % of the MC-LR (4.5 mg/L) within 48 h in batch experiments. Adding Sphingopyxis sp. m6 continuously promoted MC-LR biological removal, and successfully broke the barrier of adsorption capacity of tested adsorbents, removing >90 % of the MC-LR in most of the coupled adsorption-biodegradation tests, especially for those adsorbents had low physiochemical adsorption capacity. Variance partitioning analysis indicated that mesopore was the dominant contributor to adsorption capacity of MC-LR in pure adsorption treatments, which acted synergistically with electrical conductivity, polarity and total functional groups on the absorbent. Pore structure was the key factor beneficial for the growth of Sphingopyxis sp. m6 (51% contribution) and subsequent MC-LR biological removal rate (80 % contribution). Overall, pinewood-based carbonaceous adsorbents (especially pinewood activated carbon) exhibited the highest adsorption capacity towards MC-LR and provided the most favorable conditions for biological removal of MC-LR, largely because of their high mesopore volume, total functional groups and electric conductivity. The research outcomes not only deepened the quantitative understanding of mechanisms for MC-LR removal by the coupled process, but also provided theoretical basis for future materials' selection and modification during the practical application of coupled process.

Abstract:The bioaugmentation role of microbes is often impeded by heavy metal (HM) ions in constructed wetlands (CWs). To explore the interaction between microbes and HM ions, two identical CWs: an MCW (with resistant microorganisms) and a CCW (as control) were used in this study. Experiments analyzed static adsorption performance in a synthetic HM solution. The removal performance of Cd2+ and Zn2+ was further investigated in both CWs. The removal efficiencies (REs) of 81.92-99.56% and 74.05-98.79% were achieved for Cd2+ and Zn2+ in the adsorption study, respectively. Significantly higher REs of Cd2+ (99.60%), and Zn2+ (94.41%) were achieved in the MCW. The microbial community analysis revealed that the dominant genera were Serratia and Pseudomonas in the MCW. The subcellular analysis further demonstrated that the HMs bioaccumulated mainly in the cytomembrane and cell wall. These results indicate that CW with resistant microorganisms inoculated was an effective strategy for treating HMs wastewater.

Abstract:Microbial fuel cells implemented in constructed wetlands (CW-MFCs), albeit a relatively new technology still under study, have shown to improve treatment efficiency of urban wastewater. So far the vast majority of CW-MFC systems investigated were designed as lab-scale systems working under rather unrealistic hydraulic conditions using synthetic wastewater. The main objective of this work was to quantify CW-MFCs performance operated under different conditions in a more realistic setup using meso-scale systems with horizontal flow fed with real urban wastewater. Operational conditions tested were organic loading rate (4.9+-1.6, 6.7+-1.4 and 13.6+-3.2 g COD/m2.day) and hydraulic regime (continuous vs intermittent feeding) as well as different electrical connections: CW control (conventional CW without electrodes), open-circuit CW-MFC (external circuit between anode and cathode not connected) and closed-circuit CW-MFC (external circuit connected). Eight horizontal subsurface flow CWs were operated for about four months. Each wetland consisted of a PVC reservoir of 0.193 m2 filled with 4/8 mm granitic riverine gravel. All wetlands had intermediate sampling points for gravel and interstitial liquid sampling. The CW-MFCs were designed as three MFCs incorporated one after the other along the flow path of the CWs. Results showed no significant differences between tested organic loading rates, hydraulic regimes or electrical connections, however, on average, systems operated in closed-circuit CW-MFC mode under continuous flow outperformed the other experimental conditions. Closed-circuit CW-MFC compared to conventional CW control systems showed around 5% and 22% higher COD and ammonium removal, respectively. Correspondingly, overall bacteria activity, as measured by the fluorescein diacetate technique, was higher (4% to 34%) in closed-circuit systems when compared to CW control systems.
