Abstract:Our previous work has indicated that enzymatic resolution of glycidyl butyrate are strongly affected by many factors, including concentration of substrate, amount of lipase, the temperature, pH, shaking speed and reaction time. In this study, Plackett-Burman design was undertaken to evaluate the effects of the six factors. By regression analysis, concentration of substrate, amount of lipase and the temperature were found to be important for enzymatic resolution of glycidyl butyrate. In the second phase of the optimization process, a response surfacemethodology (RSM) was used to optimize the above critical factors, and to find out the optimal concentration levels and the relationships between these factors. By solving the quadratic regression model equation using appropriate statistic methods, the optimal parameter of the variables were determined as: 0.499 mol/L glycidyl butyrate, 30.23 mg/g lipase and 29.68 degrees C. In the optimum condition, the value of enantiomeric excess(ee%) was 93.28%. Compared to 84.65% which was the maximum ee% under the non-optimized condition, this study has a significant advancement. The experimental data under various conditions have validated the theoretical values.
Enzymatically catalyzed synthesis of low-calorie structured lipid in a solvent-free system: optimization by response surface methodology

Abstract:A kind of low-calorie structured lipid (LCSL) was obtained by interesterification of tributyrin (TB) and methyl stearate (St-ME), catalyzed by a commercially immobilized 1,3-specific lipase, Lipozyme RM IM from Rhizomucor miehei . The condition optimization of the process was conducted by using response surface methodology (RSM). The optimal conditions for highest conversion of St-ME and lowest content LLL-TAG (SSS and SSP; S, stearic acid; P, palmitic acid) were determined to be a reaction time 6.52 h, a substrate molar ratio (St-ME:TB) of 1.77:1, and an enzyme amount of 10.34% at a reaction temperature of 65 °C; under these conditions, the actually measured conversion of St-ME and content of LLL-TAG were 78.47 and 4.89% respectively, in good agreement with predicted values. The target product under optimal conditions after short-range molecular distillation showed solid fat content (SFC) values similar to those of cocoa butter substitutes (CBS), cocoa butter equivalent (CBE), and cocoa butters (CB), indicating its application for inclusion with other fats as cocoa butter substitutes.
Optimization of solvent-free enzymatic esterification in eutectic substrate reaction mixture

Abstract:The Candida rugosa lipase catalyzed esterification of (-)-menthol and lauric acid (LA) was studied in a eutectic mixture formed by both substrates((-)-menthol:LA 3:1, mol/mol). No additional reaction solvent was necessary, since the (-)-menthol:LA deep eutectic solvent (DES) acts as combined reaction medium and substrate pool. Therefore, the esterification is conducted under solvent-free conditions. The thermodynamic water activity (aw) was identified as a key parameter affecting the esterification performance in the (-)-menthol:LA DES. A response surface methodology was applied to optimize the esterification conditions in terms aw, amount of C. rugosa lipase (mCRL) and reaction temperature. Under the optimized reaction conditions (aw = 0.55; mCRL =60 mg; T =45 °C), a conversion of 95 ± 1% LA was achieved (one day), the final (-)-menthyl lauric acid ester concentration reached 1.36 ± 0.04 M (2.25 days). The experimental product formation rate agreed very well with the model prediction.

Abstract:Short-chain alkyl esters and sugar esters are widely used in the food, pharmaceutical and cosmetic industries due to their flavor and emulsifying characteristics, respectively. Both compounds can be synthesized via biocatalysis using lipases. This work aims to compare the performance of commercial lipases covalently attached to dry acrylic beads functionalized with oxirane groups (lipases from Candida antarctica type B-IMMCALB-T2-350, Pseudomonas fluorescens-IMMAPF-T2-150, and Thermomyces lanuginosus-IMMTLL-T2-150) and a home-made biocatalyst (lipase from Pseudomonas fluorescens adsorbed onto silica coated with octyl groups, named PFL-octyl-silica) in the syntheses of short- and long-chain carboxylic acid esters. Esters with flavor properties were synthetized by esterification of acetic and butyl acids with several alcohols (e.g., ethanol, 1-butanol, 1-hexanol, and isoamyl alcohol), and sugar esters were synthetized by esterification of oleic and lauric acids with fructose and lactose. All biocatalysts showed similar performance in the syntheses of short-chain alkyl esters, with conversions ranging from 88.9 to 98.4%. However, in the syntheses of sugar esters the performance of PFL-octyl-silica was almost always lower than the commercial IMMCALB-T2-350, whose conversion was up to 96% in the synthesis of fructose oleate. Both biocatalysts showed high operational stability in organic media, thus having great potential for biotransformations.

Abstract:The solvent-free esterification of the free fatty acids (FFAs) obtained by the hydrolysis of castor oil (a non-edible vegetable oil) with 2-ethyl-1-hexanol (a branched fatty alcohol) was catalyzed by different free lipases. Eversa Transform 2.0 (ETL) features surpassed most commercial lipases. Some process parameters were optimized by the Taguchi method (L16′). As a result, a conversion over 95% of the FFAs of castor oil into esters with lubricants properties was achieved under optimized reaction conditions (15 wt% of biocatalyst content, 1:4 molar ratio (FFAs/alcohol), 30 °C, 180 rpm, 96 h). The substrates molar ratio had the highest influence on the dependent variable (conversion at 24 h). FFAs/2-ethyl-1-hexanol esters were characterized regarding the physicochemical and tribological properties. Interestingly, the modification of the FFAs with 2-ethyl-1-hexanol by ETL increased the oxidative stability of the FFAs feedstock from 0.18 h to 16.83 h. The biolubricants presented a lower friction coefficient than the reference commercial mineral lubricant (0.052 ± 0.07 against 0.078 ± 0.04). Under these conditions, ETL catalyzed the oligomerization of ricinoleic acid (a hydroxyl fatty acid) into estolides, reaching a conversion of 25.15% of the initial FFAs (for the first time).

Abstract:Trimethylolpropane fatty acid triester (TFATE) is a promising biolubricant as an environmentally friendly substitute to conventional mineral-based lubricants. However, the issues of extensive energy consumption from vacuum maintenance and feedstock saponification caused by strong alkaline catalysts exist in current TFATEbased biolubricant synthesis. Here, sustainable production of biolubricant was achieved with methyl oleate (MO) from non-edible oil sources as the feedstock and moderate alkaline K2CO3 as the catalyst. Nitrogen gas stripping method instead of vacuum was adopted for methanol removal. The effects of agitation speed, N-2 flow rate, catalyst amount, MO to trimethylolpropane (TMP) molar ratio and temperature on MO conversion and TFATE selectivity were examined. The TFATE selectivity reached 95.6% under the optimized conditions. Leaching tests indicated neglectable K2CO3 dissolution in reaction products. The kinetic parameters for the three consecutive transesterification reactions were calculated. In addition, the synthesized biolubricants exhibit low acid value and volatility, moderate iodine value, superior thermal stability, and good rheological and frictional properties.
Efficient water removal in lipase-catalyzed esterifications using a low-boiling-point azeotrope

Abstract:High conversions in lipase-catalyzed syntheses of esters from free acyl donors and an alcohol requires efficient removal of water preferentially at temperatures compatible to enzyme activity. Using a lipase B from Candida antarctica (CAL-B)-mediated synthesis of sugar fatty-acid esters, we show that a mixture of ethyl methylketone (EMK) and hexane (best ratio: 4:1, vo/vo) allows efficient removal of water generated during esterification. Azeotropic distillation of the solvent mixture (composition: 26% EMK, 55% hexane, 19% water) takes place at 59 degrees C, which closely matches the optimum temperature reported for CAL-B. Water is then removed from the azeotrope by membrane vapor permeation. In case of glucose stearate, 93% yield was achieved after 48 h using an equimolar ratio of glucose and stearic acid. CAL-B could be reused for seven reaction cycles, with 86% residual activity after 14 d total reaction time at 59 degrees C. A decrease in fatty-acid chain length as well as increasing temperatures (75 degrees C) resulted in lower conversions. In addition, immobilization of CAL-B on a magnetic polypropylene carrier (EP 100) facilitated separation of the biocatalyst.

Abstract:In this study, the synthesis of aroma esters by the direct esterfication of carboxylic acids with aromatic alcohols mediated by lipase B from Candida antarctica encapsulated in a sol–gel matrix in a solvent-free system is presented. Vacuum was used in order to remove the resultant water. The reaction parameters were optimized by factorial design experiments considering four factors (acid excess, temperature, vacuum and time) on two levels. As a result, the conversions were significantly increased (for example, from an isolation yield of 49.4% to 94.3% for cinnamyl butyrate). A semi-preparative experiment was further set up for cinnamyl butyrate preparation. The green chemistry metrics, such as the E -factor of 4.76 and mass intensity of 6.04, demonstrated that the newly developed enzymatic process is suitable for industrial application based on green chemistry principles.

Abstract:The strategy of optimization using sequential factorial design was employed to enhance the tensio-active emulsifying agent produced by Candida lipolytica using soybean oil refinery residue as substrate. A full factorial design was used to evaluate the impact of three fermentation factors-amounts of refinery residue, glutamic acid and yeast extract. This allowed exclusion of the yeast extract. Full factorials designs were then sequentially used to optimize the levels of the residue and glutamic acid. The surface tension value was finally reduced to 25.29 mN/m. The maximum emulsifier activity using different substrates was within 40 h of cultivation. The surface tension of the cell-free broth containing the biosurfactant remained very stable during exposure to a wide range of pH (2-12), temperatures (0-120 degrees C) and salinity (2-10% NaCl). The combination of an industrial waste and a cheap substrate therefore seems to be very promising for the low-cost production of potent biosurfactant.

Abstract:The enzymatic production of biolubricants with desirable properties such as high viscosity index and resistance to oxidation is investigated. These characteristics are ascribed to their branched structure and high molecular weight, as well as, to the absence of unsaturated bonds in the molecule. Solvent-free processes with moderate enzyme concentrations (up to 5% wt.) and reaching conversions of 95% or more, have been conducted. Firstly, up to fifteen branched esters and diesters, all of them saturated, are obtained from a branched acid, 2-methylhexanoic acid, and a variety of linear and branched alcohols and linear diols. Among them, three esters have viscosity indexes close to 200 or higher (1,10-decanediolyl 2-methylhexanoate, 1,12-dodecanediolyl 2-methylhexanoate and 2-octyl-1-dodecanoyl 2-methylhexanoate). For the synthesis of these compounds, optimization of biocatalyst concentration and temperature has been carried out, reaching conversions above 95% within operation times between 2 and 3.5 hours.

Abstract:The optimization of the esterification of the free fatty acids (FFAs) from castor oil with neopentylglycol (NPG) catalyzed by the commercially immobilized lipase from Thermomyces lanuginosus (Lipozyme® TL IM) is herein reported. An 95 ± 3.3% acid consumption was achieved at 55 °C, 6 wt.% biocatalyst content and a substrates molar ratio (acid/alcohol)) of 2:1 after 24 h of reaction in a solvent-free medium. The addition of 1 M heptane seemed to lead to biocatalyst inactivation. The final product was composed of over 85% of diesters of NPG, 5% of monoesters of NPG and 5% estolides (estolide number of 7), together to 2% of unreacted FFAs. This product improved the feedstock viscosity index from 87.15 to 119.21 and the oxidative stability from 0.18 h to 24 h. It also exhibited a lower friction coefficient (0.0518 against 0.0592) and a lower wear scar diameter (117.99 against 136.78) than the commercial lubricant 15W-50. Lipozyme® TL IM retained approximately half of its initial FFAs conversion after eight consecutive reaction cycles of 24 h.

Abstract:Free lipase-mediated alcoholysis for biodiesel production has drawn increasing attention in recent years due to its advantages of lower cost and faster reaction rate compared to immobilized lipase. Ethanol, derived from renewable biomass, has a great potential for biodiesel production. A previous study showed that free lipase NS81006 could effectively catalyze the ethanolysis of triglycerides for biodiesel preparation. Since most crude plant oils always contain an amount of free fatty acids, oleic acid was used as the model substrate for this study on lipase-mediated esterification for biodiesel production. The central composite design of the response surface methodology was adopted for process optimization. A biodiesel yield of over 90 % was achieved under optimal reaction conditions and the repeated use of the free lipase was easily realized through phase separation either by natural gravity force or centrifugation.

Abstract:Molecular docking is an important computational analysis widely used to predict the interaction of enzymes with several starting materials for developing new valuable products from several starting materials, including oils and fats. In the present study, molecular docking was used as an efficient in silico screening tool to select biocatalysts with the highest catalytic performance in butyl esters production in a solvent-free system, an eco-friendly approach, via direct esterification of free fatty acids from Licuri oil with butanol. For such purpose, three commercial lipase preparations were used to perform molecular docking studies such as Burkholderia cepacia (BCL), Porcine pancreatic (PPL), and Candida rugosa (CRL). Concurrently, the results obtained in BCL and CRL are the most efficient in the esterification process due to their higher preference for catalyzing the esterification of lauric acid, the main fatty acid found in the licuri oil composition. Meanwhile, PPL was the least efficient because it preferentially interacts with minor fatty acids. Molecular docking with the experimental results indicated the better performance in the synthesis of esters was BCL. In conclusion, experimental results analysis shows higher enzymatic productivity in esterification reactions of 1294.83 μmol/h.mg, while the CRL and PPL demonstrated the lowest performance (189.87 μmol / h.mg and 23.96 μmol / h.mg, respectively). Thus, molecular docking and experimental results indicate that BCL is a more efficient lipase to produce fatty acids and esters from licuri oil with a high content of lauric acid. In addition, this study also demonstrates the application of molecular docking as an important tool for lipase screening to achieve more sustainable production of butyl esters with a view synthesis of biolubricants.

Abstract:This study reports the synthesis of a hybrid sol-gel material, based on organically modified silanes (ORMOSILs) with epoxy functional groups, and its application in the stabilization of lipase type B from Candida antarctica (CalB) through sol-gel entrapment. The key immobilization parameters in the sol-gel entrapment of lipase using epoxysilanes were optimized by the design of numerous experiments, demonstrating that glycidoxypropyl-trimethoxysilane can allow the formation of a matrix with excellent properties in view of the biocatalytic esterifications catalyzed by this lipase, at an enzyme loading of 25 g/mol of silane. The characterization of the immobilized biocatalyst and the correlation of its catalytic efficiency with the morphological and physicochemical properties of the sol-gel matrix was accomplished through scanning electron microscopy (SEM), fluorescence microscopy (FM), as well as thermogravimetric and differential thermal analysis (TGA/DTA). The operational and thermal stability of lipase were increased as a result of immobilization, with the entrapped lipase retaining 99% activity after 10 successive reaction cycles in the batch solventless synthesis of n-amyl caproate. A possible correlation of optimal productivity and yield was attempted for this immobilized lipase via the continuous flow synthesis of n-amyl caproate in a solventless system. The robustness and excellent biocatalytic efficiency of the optimized biocatalyst provide a promising solution for the synthesis of food-grade flavor esters, even at larger scales.

Abstract:Using lipases to catalyze the synthesis of the most differentiated type of compounds remains one of the major challenges among scientists. Seeking more economic and advantageous catalysts is a current goal of green chemistry. In this work, we demonstrate the potential of a chemically modified form of lipase from Thermomyces lanuginosus (cmLTL) for the synthesis of both hydrophobic (heptyl heptanoate, heptyl octanoate, heptyl decanoate, decyl heptanoate, decyl octanoate and decyl decanoate) and amphiphilic (2‐(2‐ethoxyethoxy)ethyl oleate and 2‐(2‐ethoxyethoxy)ethyl linoleate) esters, in bulk. The results were compared with its native (LTL) and immobilized (imLTL) forms. The data revealed that LTL showed poor activity for all reactions performed with n‐heptane (h 60%), with exception of the short ester, heptyl heptanoate. cmLTL was the only form of LTL capable of producing hydrophobic and amphiphilic esters, without compromising the yield when the reactions were performed under solvent‐free conditions (>50%). Molecular modeling showed that the active pocket of cmLTL is able to deeply internalize transcutol, with stronger interactions, justifying the outstanding results obtained. Furthermore, owing to the possibility of cmLTL filtration, the reusability of the catalyst is ensured for at least 6 cycles, without compromising the reaction yields.

Abstract:Production of esters, organic compounds with emollient, emulsifying, and aromatic properties is growing due to high demand and their applications in the cosmetics industry. The synthesis of these compounds involves the reaction between a carboxylic acid and an alcohol, which can be catalyzed by lipases. In this context, the objective of this study was to produce mycelium-bound lipases of the fungus Rhizopus oryzae through submerged cultivation, with the aim of applying them in the synthesis of ethylene glycol oleate, ethylene glycol stearate, and isoamyl palmitate esters, which are relevant to the cosmetic industry. The influence of solvent usage (heptane), biocatalyst mass, and acid/alcohol molar ratio was evaluated in the esterification reactions. In the synthesis of the three tested esters, and in the presence of heptane solvent, an equimolar ratio (1:1) of each acid and alcohol with a concentration of 0.01 mol L-1 was utilized. The reactions were performed in agitated Erlenmeyer flasks at 40 °C with an agitation speed of 200 rpm. The lipolytic biomass of Rhizopus oryzae showed better performance in the synthesis of isoamyl palmitate, achieving a maximum conversion of 83% in 24 h of reaction using 0.30 g of lipolytic biomass and the solvent heptane. In addition, it was observed that higher concentrations of catalyst biomass (0.45 g) and lower molar ratios (1:1.5) of acid to alcohol resulted in a higher conversion of isoamyl palmitate ester, reaching conversions of 48.59 and 83.11%, respectively.

Abstract:Rhamnolipids are becoming an important class of glycolipid biosurfactants. Herein, we describe for the first time the enzymatic synthesis of rhamnose fatty acid esters by the transesterification of rhamnose with fatty acid vinyl esters, using lipase from Pseudomonas stutzeri as a biocatalyst. The use of this lipase allows excellent catalytic activity in the synthesis of 4-O-acylrhamnose (99% conversion and full regioselectivity) after 3 h of reaction using tetrahydrofuran (THF) as the reaction media and an excess of vinyl laurate as the acyl donor. The role of reaction conditions, such as temperature, the substrates molar ratio, organic reaction medium and acyl donor chain-length, was studied. Optimum conditions were found using 35 °C, a molar ratio of 1:3 (rhamnose:acyldonor), solvents with a low logP value, and fatty acids with chain lengths from C4 to C18 as acyl donors. In hydrophilic solvents such as THF and acetone, conversions of up to 99–92% were achieved after 3 h of reaction. In a more sustainable solvent such as 2-methyl-THF (2-MeTHF), high conversions were also obtained (86%). Short and medium chain acyl donors (C4–C10) allowed maximum conversions after 3 h, and long chain acyl donors (C12–C18) required longer reactions (5 h) to get 99% conversions. Furthermore, scaled up reactions are feasible without losing catalytic action and regioselectivity. In order to explain enzyme regioselectivity and its ability to accommodate ester chains of different lengths, homology modelling, docking studies and molecular dynamic simulations were performed to explain the behaviour observed.

Abstract:Pentyl valerate was synthesized biocatalytically using Candida rugosa lipase (CRL) immobilized in microemulsion based organogels (MBGs). The optimum conditions were found to be pH 7.0, temperature of 37°C, ratio of concentration of water to surfactant (Wo) of 60, and the surfactant sodium bis-2-(ethylhexyl)sulfosuccinate (AOT) for MBG preparation. Although kinetic studies revealed that the enzyme in free form had high affinity towards substrates ( K m = 23.2 mM for pentanol and 76.92 mM for valeric acid) whereas, after immobilization, the K m values increased considerably (74.07 mM for pentanol and 83.3 mM for valeric acid) resulting in a slower reaction rate, the maximum conversion was much higher in case of immobilized enzyme (~99%) as compared to free enzyme (~19%). Simultaneous effects of important parameters were studied using response surface methodology (RSM) conjugated with Box-Behnken design (BBD) with five variables (process parameters), namely, enzyme concentration, initial water content (Wo), solvent used for MBG preparation, substrate ratio and time, and response as the final product formation, that is, pentyl valerate (%). The MBGs were reused for 10 consecutive cycles for ester synthesis. Efficacy of AOT/isooctane as dehydrating agent for extracting excess water from MBGs was found to exert a positive effect on the esterification reaction.
