Abstract:BACKGROUNDQuinoa protein is enriched with a wide range of amino acids, including all nine essential amino acids necessary for the human body, and in appropriate proportions. However, as the main ingredient of gluten-free food, it is difficult for quinoa to form a certain network structure for lack of gluten protein. The aim of this work was to enhance the gel properties of quinoa protein. Therefore, the texture characteristics of quinoa protein treated with different ultrasound intensities coupled with transglutaminase (TGase) were investigated. RESULTSThe gel strength of quinoa protein gel increased markedly by 94.12% with 600 W ultrasonic treatment, and the water holding capacity increased from 56.6% to 68.33%. The gel solubility was reduced and free amino content increased the apparent viscosity and the consistency index. Changes in the free sulfhydryl group and hydrophobicity indicated that ultrasound stretched protein molecules and exposed active sites. The enhanced intrinsic fluorescence intensity at 600 W demonstrated that ultrasonic treatment affected the conformation of quinoa protein. New bands emerged in sodium dodecylsulfate-polyacrylamide gel electrophoresis indicating that high-molecular-weight polymers were generated through TGase-mediated isopeptide bonds. Furthermore, scanning electron microscopy showed that the gel network structure of TGase-catalyzed quinoa protein was more uniform and denser, thereby improving the gel quality of quinoa protein. CONCLUSIONThe results suggested that high-intensity ultrasound combined with TGase would be an effective way to develop higher-quality quinoa protein gel. & COPY; 2023 Society of Chemical Industry.

Abstract:This study sought approaches to improve the textural properties of quinoa protein (QP) from the perspective of processing methods. The effect of microwave (MW) heating on the aggregation changes and gel properties of QP was investigated. The QP was treated with water bath and MW at the same heating rate. Ultraviolet absorption spectrum, intrinsic fluorescence spectrum and circular dichroism showed that MW accelerated the denaturation and unfolding of protein structure, thus more hydrophobic residues were exposed and the surface hydrophobicity was enhanced. The significant depression of free sulfhydryl groups and solubility indicated that MW promoted the production of protein aggregates. Larger protein aggregates observed in turbidity, particle size and atomic force microscopy images. MW treatment improved the gel strength and maintained the water holding capacity of QP gel. This study provided a theoretical reference for the application of MW in gluten-free foods.

Abstract:This study aimed to investigate the effects of ultrasound pretreatment on the yield and the physicochemical properties, structural and digestion characterizations of quinoa protein (QP). Results showed that under the conditions of ultrasonic power density of 0.64 W/mL, ultrasonication time of 33 min, and the liquid-solid ratio of 24 mL/g, the highest yield of QP at 68.403 % was obtained, which was significantly higher than that without ultrasound pretreatment at 51.26 ± 1.76 % ( P < 0.05). Ultrasound pretreatment decreased the average particle size and ζ-potential but increased the hydrophobicity of QP ( P < 0.05). However, no significant protein degradation and secondary structure changes of QP by ultrasound pretreatment were observed. In addition, ultrasound pretreatment slightly improved the in vitro digestibility of QP and reduced the dipeptidyl peptidase IV (DPP-IV) inhibitory activity of the hydrolysate of QP by in vitro digestion. Overall, this work demonstrates that ultrasound-assisted extraction is appropriate for improving the extraction efficiency of QP.

Abstract:This study investigated the effects of microwave stabilization treatment on the dynamic changes in volatile and flavor compounds in quinoa with different milling degrees. Microwave treatment effectively suppressed enzymatic activity in quinoa, resulting in reduced levels of undesirable compounds such as hexanal, nonanal, and others associated with beany, yeasty, and pungent aromas. The microwave-treated quinoa exhibited a broader array of volatile flavor compounds compared to the hot air-treated quinoa. Additionally, it promoted the formation of furanones and 2-acetylpyrrole, imparting roasted and toasted notes to microwave-treated quinoa. As storage time increased, the content of bitter, aldehydic, and fatty aldehydes significantly rose, while alcohols decreased. Remarkably, esters, ketones, and acids exhibited a sharp increase. Notably, a rapid surge of acetic acid after 6 weeks indicated complete spoilage, with the untreated group displaying more pronounced changes compared to the microwave-treated group. This research demonstrated the promising application of microwave in quinoa with satisfactory flavor preservation and alleviating the deterioration during storage.
Biotransformation of Quinoa Phenolic Compounds with Monascus Anka to Enhance the Antioxidant Capacity and Digestive Enzyme Inhibitory Activity

Abstract:This research aims to change the phenolic fractions and the bioactivities of quinoa by solid-state fermentation (SSF) with the edible fungus Monascus anka (M. anka). The contents of protein and fat increased in the fermented product, while the carbohydrate decreased. After 6-day fermentation with M. anka, the amount of phenolic compound reached the highest level. The majority of phenolic forms in fermented quinoa were phenolic acids, mainly ferulic acid, protocatechuic acid and p-hydroxybenzoic acid. Ultrahigh-performance liquid chromatography-mass spectrometry (UHPLC-MS) results showed that SSF was an effective method to transform quinoa phenolic compounds. Free fractions could be rapidly absorbed in the small intestine, suggesting that SSF with M. anka was a useful method to enhance bioavailable antioxidants. Antioxidant ability in vitro results showed that phenolic fractions from fermented quinoa were greater than the unfermented quinoa, and a significant cellular antioxidant activity (CAA) increment of 135% was obtained in the free phenolic fraction of fermented quinoa. Moreover, α-amylase and α-glucosidase inhibition activities were enhanced with fermentation. Correlation matrix analysis revealed that most of the free phenolic compounds showed strong positive correlations with antioxidant activities and digestive enzyme activities. Consequently, fermentation with M. anka was a particularly promising method to enhance the bioactivity of quinoa.
Effect of magnetic field pretreatment on germination characteristics, phenolic biosynthesis, and antioxidant capacity of quinoa

Abstract:The development of quinoa-based functional foods with cost-effective methods has gained considerable attention. In this study, the effects of magnetic field pretreatment on the germination characteristics, phenolic synthesis, and antioxidant system of quinoa (Chenopodium quinoa Willd.) were investigated. The results showed that the parameters of magnetic field pretreatment had different effects on the germination properties of five quinoa varieties, in which Sanjiang-1 (SJ-1) was more sensitive to magnetic field pretreatment. The content of total phenolics and phenolic acids in 24-h germinated seeds increased by 20.48% and 26.54%, respectively, under the pretreatment of 10 mT magnetic fields for 10 min compared with the control. This was closely related to the activation of the phenylpropanoid pathway by increasing enzyme activities and gene expression. In addition, magnetic field improved 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) free radicals scavenging capacities and increased peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione peroxidase (GSH-Px) activities. This study suggests that magnetic field pretreatment enhanced gene expression of phenylalanine ammonia lyase (PAL), 4-coumarate-CoA ligase (4CL), chalcone synthase (CHS) and chalcone isomerase (CHI), increased antioxidant enzyme activity and phenolics content. Thereby lead to an increase in the antioxidative capacity of quinoa.

Abstract:Effects of microwave and traditional water bath treatment at different temperatures (70, 80, 90 degrees C) on in vitro digestion rate and antioxidant activity of digestion products of quinoa protein were investigated. The results indicated microwave treatment at 70 degrees C produced the highest quinoa protein digestion rate and the strongest antioxidant activities of its digestion products (P < 0.05), which was further verified by the results of free amino, sulfhydryl group, gel electrophoresis, amino acid profiles and the molecular weight distribution of the digestion products. However, limited exposure of active groups induced by water bath treatment might decrease the susceptibility of digestive enzymes and subsequently lower the digestibility and antioxidant activities of quinoa protein. The results suggested that a moderate microwave treatment could be used as a potential way to enhance the in vitro digestion rate of quinoa protein, as well as increase the antioxidant activities of its digestion products.

Abstract:BACKGROUND This study was performed to evaluate the amplification of HER-2/neu in patients with melanoma. MATERIALS AND METHODS Amplification of HER-2/neu was evaluated in a group of patients with melanoma, referred to two referral centers in Tehran, using immunohistochemistry (IHC) and chromogenic in situ hybridization (CISH) techniques. RESULTS Forty patients with mean age 57.9±19.5 years were enrolled in this study. The most frequent type of melanoma was acral, while lower limbs were the most frequent sites. The amplification of HER2/ neu was negative in 97.5% of patients with IHC and in 100% of patients with CISH technique. Only one case (2.5%) shows weak positive staining (+2) in IHC method. Fifty five percent of melanoma was ulcerative, and the most common stages of tumors were stages 4b and 3b. More than 47% of cases were in Clark level III, while the mean of Breslow thickness was 3.56±2.87 mm. The stage of the case that showed weakly positive staining (2+) in IHC was 4b. CONCLUSIONS The amplification of HER2/neu biomarker was negative in patients with melanoma, using both CISH and IHC techniques.
Effect of Germination on the Nutritional Properties, Phytic Acid Content, and Phytase Activity of Quinoa (Chenopodium quinoa Willd)

Abstract:The aim of this study is to evaluate the effect of desaponification, soaking, germination, and refrigerated storage on the phytase activity, phytic acid content, and nutritional properties of three varieties of quinoa: white, red, and black. Desaponification and soaking reduced the number of minerals and the nutritional content. Germination of the seeds was carried out in the desaponified samples. The nutritional values, phytase activity, and phytic acid content of quinoa were measured after 6 h of soaking and then at 4 and 7 days during germination plus 7 days of refrigerated storage (4 °C). Germination increased the fibre and protein content as well as the iron, zinc, and calcium content. Germination significantly increased the phytase activity in all varieties and decreased the phytic acid content. The phytic acid content decreased during germination from 32 to 74%. Refrigerated storage had no significant effect on most of the factors studied. Germination boosts nutritional content and phytase activity while decreasing phytic acid content. Germination can be a simple method to reduce phytic acid in quinoa and may also improve the nutritional quality of this pseudocereal with the potential for use in functional foods and vegetarian diets.

Abstract:The effects of processing on the content of amino acids and fatty acids and the release of glucose from quinoa grains were evaluated in this paper. The processes included dehulling, boiling, extrusion, heating under pressure, and baking (infrared heating). The retention rate (AR) of essential amino acids and fatty acids of dehulled and boiled quinoa was 100%. The oil content of the extruded quinoa samples of two varieties was 47.71% and 39.75% lower than the corresponding raw quinoa samples. Baking and heating under pressure had different effects on the essential amino acid content, fatty acid content, and hydrolysis rate of quinoa starch. The results indicated the different cooking methods affect the essential amino acid content, fatty acid composition, release of glucose, and nutritional quality of quinoa, and moderate processing should be adopted to fully utilize the essential amino acids, fatty acids, and starch in quinoa.

Abstract:Protein oxidation leads to covalent modification of structure and deterioration of functional properties of quinoa protein. The objective of this study was to investigate the effects of ultrasonic treatment on the functional and physicochemical properties of quinoa protein oxidation aggregates. In this concern, 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) was selected as oxidative modification of quinoa protein. The microstructure of quinoa protein displayed by scanning electron microscope (SEM) indicated that oxidation induced extensive aggregation, leading to carbonylation and degradation of sulfhydryl groups. Aggregation induced by oxidation had a negative effect on the solubility, turbidity, emulsifying stability. However, according to the analysis of physicochemical properties, ultrasonic significantly improved the water solubility of quinoa protein. The quinoa protein treated by ultrasonic for 30 min exhibited the best dispersion stability in water, which corresponded to the highest ζ-potential, smallest particle size and most uniform distribution. Based on the FT-IR, SDS-PAGE and surface hydrophobicity analysis, the increase of α-helix, β-turn and surface hydrophobicity caused by cavitation effect appeared to be the main mechanism of quinoa protein solubilization. In addition, the hydrophobic region of the protein was re-buried by excessive ultrasonic treatment, and the protein molecules were reaggregated by disulfide bonds. Microstructural observations further confirmed that ultrasonic treatment effectively inhibited protein aggregation and improved the functional properties of quinoa protein.

Abstract:The pasting characteristics induced by different milling degrees on the cooking quality of quinoa were investigated. The results showed that significant variations were observed in the basic composition of quinoa with different milling degrees, which indicated the existence of effects due to the degree of milling. The combination of starch and water promoted the pasting of quinoa, and with increasing milling, quinoa textural properties tended to produce softer and more viscous. LF NMR results indicated that more water was present in the dense network formed by the starch gelatinization. Rheological properties further demonstrated that milling degree enhanced the interactions between starch molecules and correspondingly increased the energy storage modulus and loss modulus. The microstructures (SEM and CLSM) confirmed that the physical property changes in quinoa were attributed to a dense network structure caused by starch pasting, which was similar to the results of RVA. The higher the amylose/amylopectin ratio (between 0.113 and 0.167), the more pronounced the characteristic peaks. The different degrees of milling of quinoa changed the quinoa crystallinity. Consequently, the degree of milling improved the cooking characteristics and enhanced the physicochemical properties. These results help the quinoa processing industry to develop healthy quinoa products with moderate milling.

Abstract:Quinoa (Chenopodium quinoa Willd) sprouts are rich in bioactive compounds that offer numerous health benefits. However, limited research exists on their cultivation, nutritional value, and processing potential. This study compared the nutritional composition and antioxidant activity of quinoa sprouts from different varieties at various time points. Results showed a general increase in most nutrients over time. At the 24 h mark, JQ-W3 exhibited a 17.77% increase in leucine, 1.68 times higher than in eggs, along with a 6.11-fold elevation in GABA content. JQ-B1 exhibited the preeminent antioxidant potency composite (APC) score. Saponins, known for their bitter taste, decreased at 12 h but returned to original levels by 24 h. Based on nutritional components and saponin content, 24 h sprouted black quinoa JQ-B1 and white quinoa JQ-W3 were selected, providing a basis for quinoa sprout development in the food industry. These findings contribute to the understanding and utilization of quinoa sprouts.
Impact of Sprouting Process on the Protein Quality of Yellow and Red Quinoa (Chenopodium quinoa)

Abstract:The demand for plant-based proteins has increased remarkably over the last decade. Expanding the availability and variety of plant-based protein options has shown positive potential. This study aimed to investigate the qualitative and quantitative changes in amino acids of yellow and red quinoa seeds (YQ and RQ) during a 9-day germination period. The results showed that the germination process led to an increase in the total amino acids by 7.43% and 14.36% in the YQ and RQ, respectively. Both varieties exhibited significant (p < 0.05) increases in non-essential and essential amino acids, including lysine, phenylalanine, threonine, and tyrosine. The content of non-essential amino acids nearly reached the standard values found in chicken eggs. These results were likely attributed to the impact of the germination process in increasing enzymes activity and decreasing anti-nutrient content (e.g., saponins). A linear relationship between increased seeds' hydration and decreased saponins content was observed, indicating the effect of water absorption in changing the chemical composition of the plant. Both sprouts showed positive germination progression; however, the sprouted RQ showed a higher germination rate than the YQ (57.67% vs. 43.33%, respectively). Overall, this study demonstrates that germination is a promising technique for enhancing the nutritional value of quinoa seeds, delivering sprouted quinoa seeds as a highly recommended source of high-protein grains with notable functional properties.

Abstract:The health-promoting activities of polyphenols and their metabolites originating from germinated quinoa (GQ) are closely related to their digestive behavior, absorption, and colonic fermentation; however, limited knowledge regarding these properties hinder further development. The aim of this study was to provide metabolomic insights into the profile, bioaccessibility, and transepithelial transport of polyphenols from germinated quinoa during in vitro gastrointestinal digestion and Caco-2 cell transport, whilst also investigating the changes in the major polyphenol metabolites and the effects of prebiotics during colonic fermentation. It was found that germination treatment increased the polyphenol content of quinoa by 21.91%. Compared with RQ group, 23 phenolic differential metabolites were upregulated and 47 phenolic differential metabolites were downregulated in GQ group. Compared with RQ group after simulated digestion, 7 kinds of phenolic differential metabolites were upregulated and 17 kinds of phenolic differential metabolites were downregulated in GQ group. Compared with RQ group after cell transport, 7 kinds of phenolic differential metabolites were upregulated and 9 kinds of phenolic differential metabolites were downregulated in GQ group. In addition, GQ improved the bioaccessibilities and transport rates of various polyphenol metabolites. During colonic fermentation, GQ group can also increase the content of SCFAs, reduce pH value, and adjust gut microbial populations by increasing the abundance of Actinobacteria, Bacteroidetes, Verrucomicrobiota, and Spirochaeota at the phylum level, as well as Bifidobacterium, Megamonas, Bifidobacterium, Brevundimonas, and Bacteroides at the genus level. Furthermore, the GQ have significantly inhibited the activity of α-amylase and α-glucosidase. Based on these results, it was possible to elucidate the underlying mechanisms of polyphenol metabolism in GQ and highlight its beneficial effects on the gut microbiota.
Appropriate Microwave Improved the Texture Properties of Quinoa Due to Starch Gelatinization from the Destructed Cyptomere Structure

Abstract:Texture characteristics of quinoa under microwave (MW) irradiation were studied from the perspective of starch gelatinization. MW reduced the light transmittance and increased the hardness and stickiness of quinoa. Microstructure showed that MW dispersed the vesicular structure of starch aggregates into separate starch particles, resulting in the full hydration of starch and water molecules to form denser network structures. The value of peak viscosity and setback decreased in RVA after MW treatment, but the gelatinization temperature remained stable. DSC further proved that moderate MW treatment could reduce the gelatinization enthalpy of starch and made quinoa accessible to gelatinize. However, XRD showed that the crystal structure of starch was preserved, but the crystallinity increased. Finally, low field NMR showed that moderate MW stimulated the full hydration of starch to form denser network structures, while excessive MW treatment made starch partially gelatinize and form rigid structure, resulting in negative texture properties.

Abstract:Cultivating salt-tolerant crops is a feasible way to effectively utilize saline-alkali land and solve the problem of underutilization of saline soils. Quinoa, a protein-comprehensive cereal in the plant kingdom, is an exceptional crop in terms of salt stress tolerance level. It seems an excellent model for the exploration of salt-tolerance mechanisms and cultivation of salt-tolerant germplasms. In this study, the seeds and seedlings of the quinoa cul-tivar Shelly were treated with different concentrations of NaCl solution. The physiological, biochemical charac-teristics and agronomic traits were investigated, and the response patterns of three salt stress-responsive genes (SSRGs) in quinoa were determined by real-time PCR. The optimum level of stress tolerance of quinoa cultivar Shelly was found in the range of 250-350 mM concentration of NaCl. Salt stress significantly induced expression of superoxide dismutase (SOD), peroxidase (POD), and particularly betaine aldehyde dehydrogenase (BADH). BADH was discovered to be more sensitive to salt stress and played an important role in the salt stress tolerance of quinoa seedlings, particularly at high NaCl concentrations, as it displayed upregulation until 24 h under 100 mM salt treatment. Moreover, it showed upregulation until 12 h under 250 mM salt stress. Taken together, these results suggest that BADH played an essential role in the salt-tolerance mechanism of quinoa. Based on the expression level and prompt response induced by NaCl, we suggest that the BADH can be considered as a mole-cular marker for screening salt-tolerant quinoa germplasm at the early stages of crop development. Salt treatment at different plant ontogeny or at different concentrations had a significant impact on quinoa growth. Therefore, an appropriate treatment approach needs to be chosen rationally in the process of screening salt-tolerant quinoa germplasm, which is useful to the utilization of saline soils. Our study provides a fundamental information to deepen knowledge of the salt tolerance mechanism of quinoa for the development of salt-tolerant germplasm in crop breeding programs.
Effect of Storage Conditions on the Textural Properties and in Vitro Digestibility of Wheat Bread Containing Whole Quinoa Flour

Abstract:This research aimed to clarify the evolution of textural characteristics and in vitro digestibility under different storage conditions. Quickly increasing hardness and decreasing resilience was observed in quinoa bread after 5d cold (4 degrees C) storage. However, when stored at-18 degrees C for 5d, bread crumb still maintains a dense alveographic with 52.33% of cell density and 33.76% area fractions. In addition, the starch digestion rate decreased with increasing retrogradation degree. Scanning electron microscope (SEM) characterizations illustrated that the chunky matrix gradually disappeared with digestion. However, quinoa starch (QS) granules were the most difficult to digest owing to the relative intactness and wrapping in the protein-starch bulk. Meanwhile, X-ray diffraction (XRD) showed that the crystal structure of starch remained 21.09% stored in frozen conditions, and wheat type-A (WA) starch was mainly involved in gelatinization. The results of water migration proved that water participated in the retrogradation of starch and affected the quality of quinoa bread. Overall, bread stored in frozen conditions can maintain good texture characteristics and slow starch digestion rate. This study indicates that quinoa flour fortified bread has the potential to be developed as staple food with both low glycemic index values and storage stability.

Abstract:Quinoa okara could be used as an innovative additive for application in the emerging plant‐based food market. Ultrasound technology allows adding value to a by‐product with potential applications as a food emulsifier. Summary High‐intensity ultrasound (HIUS) treatment is a physical method utilised to modify and improve some technologically important compounds (such as protein, fibres) that serves as an environment‐friendly process. In this regard, the effect of HIUS on the composition and emulsifying properties of quinoa okara fractions was analysed. Dispersions (1:10) of ground quinoa grains alkalinised to pH 9.0 were treated by high‐intensity ultrasound. Quinoa okara and sonicated quinoa okara (SQOK) exhibited a similar extraction yield (60%), although SQOK showed a three‐fold increase in protein content. Scanning electron microscopy showed that HIUS reduced the particle size of okara molecular structures. Analysis of O/W emulsions formulated with the okara samples demonstrated that the SQOK had a smaller droplet size and increased the stability of the system against the destabilisation processes of creaming and coalescence. The quinoa okara optimised with the application of the clean ultrasound technology allows adding value to a by‐product with potential applications as a food emulsifier. Quinoa okara could be used as an innovative additive for application in the emerging plant‐based food market.
