Abstract:The effects of anaerobic treatment on the content of polyphenols, antioxidant capability, tocols, and free amino acids (FAAs) in the germinated brown rice (GBR) were investigated. The values of polyphenols and antioxidant capability significantly increased in treated groups, in which the three phenolic acids, i.e. ferulic, p-coumaric, and sinapic acids, showed the most pronounced increase. No significant change was observed in tocols. Among the evaluated FAAs, only the content of aspartic and glutamic acids in the white, red and black GBRs decreased by 75, 65 and 69%, and by 30, 18 and 39% after anaerobic treatment, respectively. While the contents of serine and threonine remained unchanged, all others significantly increased. In particular, the contents of γ-aminobutyric acid (GABA) increased 4.2-, 9.7- and 4.8-folds in the white, red and black GBRs, respectively. The results indicated that additional anaerobic treatment could be used in the production of GBR as a functional food.
Effect of Germination and Parboiling Treatment on Distribution of Water Molecular, Physicochemical Profiles and Microstructure of Rice

Abstract:The effect of germination and parboiling treatment on distribution of water molecular, physicochemical profiles and microstructure of rice was investigated. The results showed that the water molecular was evenly distributed in rice kernel after germination, but after subjecting to parboiling treatment, mobile water molecular might be confined to small region, which diffused from the outer regions into the rice kernel core. The reason may be that parboiling treatment produced macromolecular matrix where dispersed amylose chains form double-helical associations between starch granules so that water molecular is restrained and not entrapped within the starch crystallites. Meanwhile, the changes of microstructure and distribution of water molecular may be attributed to softness of germinated rice and hardness of parboiling rice. In addition, parboiling treatment had significant (p < 0.05) effect on the physicochemical properties of germinated rice. The viscosity and pasting temperature significantly (p < 0.05) increased. The storage modulus (G′), loss modulus (G′′), tan δ and shear stress had lower value compared to untreated germinated rice. Colour difference significantly (p < 0.05) increased, the microstructure of surface became smoother and compacter.
Accumulation Of Gamma-Aminobutyric Acid In Giant-Embryo Rice Grain In Relation To Glutamate Decarboxylase Activity And Its Gene Expression During Water Soaking

Abstract:Glutamate decarboxylase (GAD) is the rate-limiting enzyme for gamma-aminobutyric acid (GABA.) synthesis. However, the relationship of GABA accumulation, GAD activity, and its gene expression in rice grain during water soaking is unknown yet. We assessed the role of GAD activity and its gene expression to GABA accumulation after the rice grain was soaked in water for 0, 1, 2, 4, 8, 16, and 32 hr. The results showed a continuous accumulation of GABA in steeped embryo, embryoless brown rice, and complete brown rice, respectively. Among them, embryo of brown rice had the maximal accumulation. The activity of GAD was gradually increasing during water soaking, and the enzyme activity was significantly higher in the embryo than in either the embryoless brown rice or whole brown rice. The results further indicated that the activity of GAD was markedly stimulated by 0.5 mmol/L of Ca2+, whereas it was almost completely inhibited by 1 mmol/L of Hg2+. Accordingly, the content of GABA in embryo paralleled the activity of GAD treated with Ca2+ or Hg2+. To measure the expression of three GAD homologous genes in steeped rice grain, a RT-PCR method was developed using the housekeeping gene Actin as the control. Gene expression studies demonstrated differential expression patterns of the three GAD transcripts: OsGAD1, 2 and 3 in brown rice. These results suggested that the accumulation of GABA in rice grain could be attributed to the increase of OsGAD transcripts.
Negative HER2/neu amplification using immunohistochemistry and chromogenic in situ hybridization techniques in skin melanoma cases.

Abstract:BACKGROUND This study was performed to evaluate the amplification of HER-2/neu in patients with melanoma. MATERIALS AND METHODS Amplification of HER-2/neu was evaluated in a group of patients with melanoma, referred to two referral centers in Tehran, using immunohistochemistry (IHC) and chromogenic in situ hybridization (CISH) techniques. RESULTS Forty patients with mean age 57.9±19.5 years were enrolled in this study. The most frequent type of melanoma was acral, while lower limbs were the most frequent sites. The amplification of HER2/ neu was negative in 97.5% of patients with IHC and in 100% of patients with CISH technique. Only one case (2.5%) shows weak positive staining (+2) in IHC method. Fifty five percent of melanoma was ulcerative, and the most common stages of tumors were stages 4b and 3b. More than 47% of cases were in Clark level III, while the mean of Breslow thickness was 3.56±2.87 mm. The stage of the case that showed weakly positive staining (2+) in IHC was 4b. CONCLUSIONS The amplification of HER2/neu biomarker was negative in patients with melanoma, using both CISH and IHC techniques.

Abstract:The response surface methodology (RSM) is applied to optimize the process conditions for producing BABA in ger-minated rice. The best conditions to obtain high content of BABA in germinated rice production is: 9.1 h soaking time, ger-mination time 14.7 h, germination temperature of 28.03 ℃, under this condition, the content of GABA 48.1 mg/100 g.

Abstract:Pre-harvest sprouting (PHS) is a constrain problem in hybrid rice production. The present study was conducted to investigate the inhibitory effect of eugenol on seed germination and PHS of hybrid rice variety (Qian You 1). The results showed that seed germination speed and the activities of α-amylase were inhibited by eugenol pre-soaking and these effects enhanced with the increasing of eugenol concentrations; while seedling growth was not negatively affected. In field trials, eugenol application caused a significant decline in PHS as compared with control, whereas no sustained inhibition in post-harvested seed germination was observed. The HPLC analysis indicated that eugenol raised the internal ABA content by 1–4 times more than control, and seeds treated with eugenol had relatively lower OsABA8OH2 and higher transcript levels of OsNCED2 expression during early stages of seed imbibitions. In addition, seed germinated faster after GA 3 application than eugenol alone, and seed endogenous ABA content decreased obviously. It suggested that eugenol strongly delayed seed germination and the PHS in the field, which might be mainly due to the increased ABA contents caused by eugenol. However, the phenomenon of delayed germination and high ABA content caused by eugenol could be effectively recovered by exogenous GA 3 .
Variation in Polyphenols, Tocols, Γ‐aminobutyric Acid, and Antioxidant Properties in Whole Grain Rice (oryza Sativa L.) As Affected by Different Germination Time

Abstract:Dynamic alterations in the polyphenols, tocols, antioxidant activities (AOA), and γ‐aminobutyric acid (GABA) contents of white, red, and black germinated rice upon different germination periods were studied. The results indicated that the contents of the free, bound, and total phenolics and AOA steadily increased during germination, even though the free part of colored germinated brown rice (GBR) showed a notable decline at the earlier stages, but still higher than the white rice. The compositions of the nine phenolic acids and six tocols generally accumulated in a time‐ and variety‐dependent manner during the germination process. However, the contents of p‐coumaric, ferulic, and sinapic acids displayed a pronounced increase with the duration of germination in all cultivars. The GABA in white, red, and black GBR accumulated to the highest content at the end of germination, which was 4.7, 14.2, and 6.7‐fold of their respective nongerminated counterparts. Interestingly, the total anthocyanin content in black GBR displayed a constant drop at earlier stages and then a rise at the later stages. These results suggest that prolonged germination may further increase the functional properties of GBR.

Abstract:The content of γ-aminobutyric acid (γ-GABA), one of the main functional components in rice, changes with the parts and the germination periods of rice. This study aimed to analyze the changes in γ-GABA content in differert parts of rice and brown rice of 26 varieties (containing 12 japonica varieties and 14 indica varieties) that have been recently bred in Yunnan province or Zhejiang province and were grown in Xinping county in Yunnan province during germination. The results showed that Diannong S-1/Dianjing 8, HIPj 1, Wendao1/ IR 36 and Wendao 2/IR 36 had the highest γ-GABA content among the 26 rice varieties; the γ-GABA contents in different parts of rice before germination decreased in the following order: rice germ rice husk brown rice polished rice; the germination activation for 24 - 28 h could significantly promote the accumulation of γ-GABA in rice and brown rice, showing a trend of first increase and then decrease, and the γ-GABA content in brown rice reached maximum at 24 h of germination; the accumulation rate and quantity in different parts of germinated rice and germinated brown rice were different, and the γ-GABA contents in different parts decreased in the following order: rice germ brown rice polished rice rice husk; after germination activation, the γ-GABA accumulation quantity in brown rice was more than that in rice, and the average content of γ-GABA in rice germ of 12 japonica varieties was higher than that of 14 indica varieties.

Abstract:In China, breeding efforts to create Indica-type rice cultivars with textural and functional properties similar to Japonica-type rice have not been successful. An alternative strategy is to develop a processing method to modify the end-use quality of Indica-type rice. Milled rice samples with low, intermediate and high apparent amylose content (AAC) were treated with Co-60 gamma rays. AAC was impacted only by high doses of gamma rays while gel consistency was significantly increased. Increasing levels of gamma irradiation (GIR) reduced rice flour viscosity and increased the temperature of gelatinization onset. Exposure to gamma rays caused the RVA setback (rice pasting properties) but not the instrumental texture properties of an Indica-type cultivar to become more like that of the Japonica-type rice. GIR of milled rice for human consumption would be limited to between 2 and 4 kGy because of its negative effects at higher doses on rice color and aroma. However, that level will destroy insects and some microorganism and change rice of unacceptable quality for some consumers closer to being acceptable and may also broaden the functionality of rice for industrial uses.

Abstract:This study investigated the effects of ultrasonication treatment on the germination rate of brown rice. Brown rice grains were subjected to ultrasound (40 kHz/30 min) and then incubated for 36 h at 37 °C to germinate the seeds. Ultrasonic treatment increased the germination rate of brown rice by up to ∼28 % at 30 h. Transcriptomic and metabolomic analyses were performed to explore the mechanisms underlying the effect of ultrasonic treatment on the brown rice germination rate. Comparing the treated and control check samples, 867 differentially expressed genes (DEGs) were identified, including 638 upregulated and 229 downregulated), as well as 498 differentially accumulated metabolites (DAMs), including 422 up accumulated and 76 down accumulated. Multi-omics analysis revealed that the germination rate of brown rice was promoted by increased concentrations of low-molecular metabolites (carbohydrates and carbohydrate conjugates, fatty acids, amino acids, peptides, and analogues), and transcription factors (ARR-B, NAC, bHLH and AP2/EREBP families) as well as increased carbon metabolism. These findings provide new insights into the mechanisms of action of ultrasound in improving the brown rice germination rate and candidate DEGs and DAMs responsible for germination have been identified.

Abstract:γ-Aminobutyric acid (GABA) is a four-carbon non-protein amino acid and has been identified to have a function in reducing blood pressure. In order to increase GABA contents in rice grains, we constructed a RNA interference (RNAi) vector suppressing the expression of GABA transaminase 1 gene (OsGABA-T1). GABA transaminase is a key enzyme that converts GABA to succinate semialdehyde (SSA). The RNA interference vector was transformed into the callus ofjaponica cv. Ningjing 1. qRT-PCR analysis showed that OsGABA-T1 transcript abundance in the RNAi transgenic lines was effectively reduced, so did the homolog geneOsGABA-T2. The GABA contents of brown rice in T3 generation of RNAi lines were 13 times higher than these in Ningjing 1, so did in polished rice as well as brown rice and polished rice stored for four months of post-harvest in transgenic rice, while other main amino acid contents did not change much. In summary, this research provides a basis for breeding a rice variety which could contribute to reduce blood pressure due to the enrichment of GABA contents in rice seeds though effectively sup-pressing the expression ofOsGABA-T1 gene by the RNA interference technique.
Optimization of Germination Inhibitors for Controlling Pre-Harvest Sprouting in Hybrid Rice

Abstract:The present study was conducted to screen different growth inhibitors i.e. ethephon (125 mM), methyl jasmonate (1 mM MeJA) and their combinations (125mM ethephon + 1mM MeJA) for minimizing the harmful effects of pre-harvest sprouting during hybrid rice seed production. The combination of both growth inhibitors significantly reduced germination and growth attributes with a more prominent decrease in cultivar Zhu Liang You 06 (ZY) as compared to cultivar Qian You No. 1 (QY). Moreover, the activities of alpha-amylase, peroxidase (POD), superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and malonaldahyde (MDA) contents were significantly increased with the combination of both growth inhibitors as compared to the control. CAT activity was significantly increased with 1mM of MeJA as compared to the control. The combination of both growth inhibitors resulted in the increase of abscisic acid (ABA) contents and the decrease of gibberellins (GA) contents as compared with the control. Gene expression analysis showed that the transcription levels of ABA genes (OsNCED1, OsNCED3 and OsABA80x1) were up-regulated with the combination of both growth inhibitors. While, GA genes (OsGA3ox1, OsGA20ox1 and OsGA2oox2) were down-regulated by seed soaking with both growth inhibitors and their combination.

Abstract:This study aimed to prepare germinated brown rice (GBR) with high ?-aminobutyric acid (GABA) content and short root length. Brown rice (BR) was exposed to 10 mT magnetic field at 25 C for 60 min, followed by incubating in GABA solutions (0-10 mM, 30 C) for 24 h. After treatment, GABA content in GBR significantly increased from 16.43 to 25.64-50.54 mg/100 g, while root length was shorter than 3.0 mm. Although GABAaminotransferase activity was reduced by 4.61%-16.14%, the absorption of exogenous GABA caused by higher electrical conductivity, increased cell membrane permeability and more root hairs was the dominant reason for GABA accumulation. The hyperaccumulation of GABA conversely retarded the positive effect of M on a-amylase activity, starch structure, and anti-oxidase activity, thus maintaining the root length at 2.03-2.45 mm. Overall, this study provided an efficient method for preparing GBR with high GABA content and short root.

Abstract:γ-aminobutyric acid (GABA) plays an important role in anti-anxiety by inhibiting neurotransmitter in the central nervous system (CNS) of mammals, which is generated in the germinating seeds. The key enzymes activity of GABA metabolism pathway and nutrients content in hemp seeds during germination were studied after treated with ultrasound and CaCl2 . The mechanism of exogenous stress on key enzymes in GABA metabolism pathway was investigated by molecular dynamics simulation. The results showed that ultrasonic combined with 1.5 mmol· L−1 CaCl2 significantly increased the activities of glutamate decarboxylase (GAD) and GABA transaminase (GABA-T) in seeds, and promoted the conversion of glutamate to GABA, resulting in the decrease of glutamate content and the accumulation of GABA. Molecular dynamics simulations revealed that Ca2+ environment enhanced the activity of GAD and GABA-T enzymes by altering their secondary structure, exposing their hydrophobic residues. Ultrasound, germination and CaCl2 stress improved the nutritional value of hemp seeds.

Abstract:In order to improve the content of GABA in japonica rice, so as to improve the nutritional composition and enhance the added value of rice, this study explored a combined process of low-pressure steam enrichment of γ-aminobutyric acid (GABA) content in japonica rice and low-pressure superheated steam (LPSS) drying of japonica rice. The low-pressure energy and the energy stored in rice grains were used to promote water absorption and activation of rice germs in a steam environment. Under the action of glutamate decarboxylase, the germ glutamate underwent an enzymatic reaction to generate GABA, which was transferred to and stored in the rice endosperm due to the concentration gradient and water gradient. After germination, LPSS was successively applied to dry the rice grains. The response surface method was used to optimize the parameters of low-pressure wet steam enrichment and LPSS drying of the rice grains. The experimental results showed that when the pressure was 0.026 MPa, the temperature was 60.66°C, the time was 5.01 h, and the flow rate was 0.31 L/h; the GABA content produced by low-pressure wet steam germination of japonica rice peaked at 94.1751 mg/100 g. When the germinated and enriched japonica rice was put through LPSS drying with a pressure of 0.021 MPa, a temperature of 66.55°C, a time of 1.57 h, and a flow rate of 0.28 L/h, the GABA content peaked at 104.161 mg/100 g. The results show that the combined process of low-pressure steam enrichment and drying continuously produced GABA in the soaking enrichment and drying stages, and the content of GABA in the drying stage accounted for 10% of the total content, which effectively increased the content of GABA in japonica rice and shortened the enrichment time. This study provides a technical reference for industrialized production of GABA-rich brown rice, embryonic rice, and polished rice.

Abstract:Abstract Germinated brown rice (GBR) is a popular functional food containing considerable amounts of beneficial nutrients and bioactive compounds. Here, autoclaving at 115°C for 20 min was employed to process GBR (AGBR) to evaluate the effect of autoclaving on the nutritional and health function of GBR in microstructure, taste value, aroma, as well as the physiological ingredients. The results showed that autoclaving treatment influenced the starch gelatinization and aroma to improve the taste of cooked AGBR. Autoclaving treatment significantly increased the gamma‐aminobutyric acid (GABA) and ferulic acid levels of AGBR (p < .05). In addition, consuming AGBR for 1 month significantly decreased the fasting plasma glucose (FPG), 0.5, 1, and 2 hr postprandial plasma glucose (PPG), triglyceride (TG), total cholesterol (TC), high‐density lipoprotein cholesterol (HDL‐c), and low‐density lipoprotein cholesterol (LDL‐c) in metabolic syndrome (MS) patients (p < .05). Therefore, autoclaving treatment, as a promising processing strategy, may both improve the sensory attributes and the nutrition of GBR.
Effects of ~(60)Co Gamma-Irradiation as Pretreatment on Quality of Rice

Abstract:A new technology of pretreating late short-grain rice by gamma-irradiation and then drying the rice with hot-air was introduced in this work.It was found by microstructure observation that the starch granules in the irradiated rice endosperm were tore up more with increasing the irradiation dose.The milling and eating quality of the irradiated rice were studied.Results show that the irradiation pretreatment leads no decrease to rice milling quality;on the contrary,it leads the improvement of rice eating quality evidently.Further more,for the eating quality improvement,the technics of irradiation before drying is more effective than the technics of irradiation after drying.A quadratic rotation-orthogonal composite experimental design,with three variables(irradiation dose,drying temperature,and initial moisture content,at five levels) and three response functions(apparent amylose content,gel consistency,and gelatinization temperature) were employed to study the effects of the three variables on the response functions.The three response functions significantly correlate with these variables by second order polynomials consisting of linear,quadratic and interaction terms.The optimum levels of the three variables for obtaining optimum magnitude(maximum or minimum) of the three response functions are obtained.The response surfaces analysis,with X and Y axes representing two of the most influential variables while keeping another variable constant at their optimum point,are presented to aid in visualizing the effect of the variables.

Abstract:Rice is a staple food for more than half of the world's population and it is regarded as a high glycemic index (GI) food. Breeders developed high amylose rice having low digestibility, but it also has inferior palatability. This study used high-amylose rice (HAR) produced by gamma irradiation and compared the digestion and physicochemical properties related to palatability with those of low-amylose rice (LAR). Pre-soaking and different-pH treatments were adopted to find a way to enhance the palatability of HAR while maintaining its low digestibility. After pre-soaking, HAR had a higher water uptake ratio (4.68 vs. 4.11 g g(-1)), proportion of leached starch (16.6 vs. 12.6%) and adhesiveness (77 vs. 39), but lower setback (0.092 vs. 0.107 Pa s), hardness (10.1 vs. 12.6 kg) and resilience (0.20 vs. 0.25). The results showed that pre-soaking was able to enhance the quality of the cooked rice mainly by modifying the starch amorphous region while maintaining the low digestibility of HAR. Pre-soaking can be adopted as a practical and effective household cooking method to prepare rice with relatively low digestibility and good palatability.
