Abstract:It is of vital importance to develop probes to monitor hypochlorous acid (HClO) in biological systems as HClO is associated with many important physiological and pathological processes. Metal nanoclusters (NCs) are promising luminescent nanomaterials for highly reactive oxygen species (hROS) detection on the basis of their strong reaction ability with hROS. However, metal NCs typically can respond to most common hROS and are susceptible to etching by biothiols, hindering their application in the construction of effective HClO probes. Herein, we proposed a strategy to develop a nanoprobe based on Au NCs for highly sensitive and selective detection of HClO. We synthesized luminescent benzyl mercaptan-stabilized Au NCs and encapsulated them with an amphiphilic polymer (DSPE-PEG). After encapsulation, an obvious emission enhancement and good resistance to the etching by biothiols for Au NCs were achieved. More importantly, the DSPE-PEG encapsulated Au NCs can be used as a nanoprobe for detection of HClO with good performance. The luminescence of the Au NCs was effectively and selectively quenched by HClO. A good linear relationship with the concentration of HClO in the range of 5-35 mu M and a limit of detection (LOD) of 1.4 mu M were obtained. Additionally, this nanoprobe was successfully used for bioimaging and monitoring of HClO changes in live cells, suggesting the application potential of the as-prepared amphiphilic polymer-encapsulated Au NCs for further HClO-related biomedical research.

Abstract:It is necessary to develop sensitive and selective probes for real-time in vivo monitoring of hypochlorous acid (HClO) which plays a significant role in physiological and pathological processes. The second near-infrared (NIRII) luminescent silver chalcogenide quantum dots (QDs) have shown great potential in developing activatable nanoprobe for HClO in terms of their outstanding imaging performance in the living organism. However, the limited strategy for the construction of activatable nanoprobes severely restricts their widespread applications. Herein, we proposed a novel strategy for developing an activatable silver chalcogenide QDs nanoprobe for NIR-II fluorescence imaging of HClO in vivo. The nanoprobe was fabricated by mixing an Au-precursor solution with Ag2Te@Ag2S QDs to allow cation exchange and release Ag ions and then reducing the released Ag ions on the QDs surface to form an Ag shell for quenching of the emission of QDs. The Ag shell of QDs was oxidized and etched in the presence of HClO, resulting in the disappearance of their quenching effect on QDs and the activation of the QDs emission. The developed nanoprobe enabled highly sensitive and selective determination of HClO and imaging of HClO in arthritis and peritonitis. This study provides a novel strategy for the construction of activatable nanoprobe based on QDs and a promising tool for NIR-II imaging of HClO in vivo.
One-pot hydrothermal synthesis of dual-emission fluorescent carbon dots for hypochlorous acid detection

Abstract:<p>In this work, we developed a dual-emission fluorescent carbon dots (CDs) using sodium citrate (SC), triethylenetetramine (TETA) and rose bengal (RB) as raw materials. Under the hydrothermal treatment, the carboxyl group in SC can crosslink with the amino group in TETA to form polymer-like intermediate by amide bond network. The RB molecule, a fluorescein derivative, lost halogen atoms under hydrothermal conditions and integrated with the skeleton structure of the network intermediate. Further dehydration and carbonization resulted in the dual-emission fluorescent CDs. Excited by ultraviolet (UV) light, the CDs exhibited blue and green luminescence centered at 440 nm and 525 nm, respectively. Due to the emission centers of these two colors showed different sensitivity to hypochlorous acid (HClO), we have designed a fluorescence assay to achieve quantitative detection of HClO. Upon addition of different concentrations of HClO to CDs solution in phosphate buffer (pH = 6), the blue emission decreased gradually while the green emission showed only a minimal decrease. By calculating the emission intensity ratio, a good linear relationship was achieved in the range of 0–17 μM, and the limit of detection was as low as 0.75 μM. The results reported here demonstrated a promising way to design organic dye modified CDs for sensing applications.</p>

Facile synthesis of long-wavelength emission carbon dots for hypochlorite sensing and intracellular pH imaging

Abstract:Hypochlorite (ClO-), a typical reactive oxygen species, plays an irreplaceable roles in various biological processes. In this work, long-wavelength emission carbon dots (LW-CDs) were fabricated through one-step hydrothermal method by using l-cysteine (cys) and neutral red (NR) as precursors for monitoring of hypochlorite and intracellular pH. Characterizations of as-prepared LW-CDs showed that they had excellent water solubility, high optical stability and sensitive response behavior. Fluorescence intensity of LW-CDs decayed in the presence of ClO- linearly from 10 to 162.5 μM (LOD = 1.021 μM) based on static quenching effect with ideal selectivity. Besides, LW-CDs revealed a pH responsive behavior in the pH range of 2.0 to 10.0, exhibited dual good linear relationships in the pH ranges of 4.2-5.8 and 5.8-7.4. The LW-CDs can also be utilized as imaging reagents in Hela living cells owing excellent biocompatibility and low cytotoxicity. These results demonstrated that the as-mentioned LW-CDs are expected to serve as excellent long wavelength emitting nanomaterials for fluorescence sensing and monitoring of cell fluctuations.

Abstract:Hypochlorous acid (HClO) is produced by white blood cells to defend against injury and bacteria. However, as one of the reactive oxygen species, high intracellular HClO concentration could lead to chronic diseases that affect the cardiovascular and nervous systems. To monitor HClO concentrations in bio-samples, the fluorescent probe is preferred to have: a) absorbability in the far-red window with reduced light-toxicity and improved tissue penetration depth, b) ratiometric feature for accurate analysis. In this study, we reported a far-red ratiometric HClO fluorescence probe based on BODIPY chromophore and aldoxime sensing group. Not only the color change of the probe solution can be detected by naked eyes, but also the emission ratios (I645/I670) showed a significant increase upon the introduction of HClO. More importantly, the feasibility of HClO monitoring in bio-samples was demonstrated in vitro using a confocal microscope.
A red-emitting ratiometric fluorescent probe with large Stokes shift and emission peak shift for imaging hypochlorous acid in living cells and zebrafish

Abstract:Hypochlorous acid (HClO) with strong oxidizing property holds an essential role in the resistance against offensive microorganisms in the inherent immune system of living organism. However, an excess of HClO can bring about oxidative stress and cellular damage, which are associated with many diseases. Herein, a novel nopinone-based ratiometric fluorescent probe BIDID (2-(4-(5,5-dimethyl-4,5,6,7-tetr-ahydro-1H-4,6-methanobenzo[d]imidazole-2-yl)benzylidene)-1H-indene-1,3(2H)-dione) was rationally devised to track exogenous and endogenously generated HClO in vitro and vivo. BIDID itself featured an extremely large Stokes shift (241 nm) and exhibited a bright red emission at 641 nm. Upon treating with HClO, the CC bond of BIDID was exclusively broken, thereby resulting in the generation of compound BID (4-(5,5-dimethyl-4,5,6,7-tetrahydro-1H-4,6-methanobenzo[d]imidazole-2-yl)benzalde-hyde) with a remarkably hypochromatic-shifted emission at 501 nm. As expected, this probe was highly responsive to HClO with good selectivity, superior sensitivity, low limit of detection (13.2 nM), rapid response (within 0.5 min), and wide pH range (6–12). What's more, BIDID was applicable for validation of HClO on solid platform and in real water samples. Additionally, the confocal fluorescent images of HClO in vitro and in vivo demonstrated that BIDID can function as a reliable tool for studying HClO-related physiological and pathological processes.

Abstract:Optically monitoring hypochlorous acid (HClO) in living body favors diagnosis and study of inflammatory diseases. However, this has been hampered by limited strategies to develop highly fluorogenic tools in the deep-penetration near-infrared spectrum. Herein, a near-infrared aza-BODIPY-bisferrocene triad Fc(2)-CBDP that unexpectedly achieves an exceptionally sensitive and selective fluorescence turn-on (>220-fold) response toward HClO through single-ferrocene oxidation and boron-alkynyl hydrolysis cascade is reported. Mechanism insight shows that Fc(2)-CBDP features "enhanced charge transfer"-caused quenching due to intramolecular bisferrocene electronic coupling, which is decoupled in the reaction with HClO. The utility of Fc(2)-CBDP for intracellular HClO imaging is evaluated and, more importantly, in vivo high-contrast deep-tissue imaging of lymphatic inflammation and colitis is realized. This work provides new insights into both HClO and ferrocene chemistry, and extends the reach of fluorogenic strategies in the near-infrared biosensing.
A highly selective ratiometric fluorescent probe for the sensitive detection of hypochlorous acid and its bioimaging applications

Abstract:The development of simple techniques for monitoring hypochlorous acid (HOCl) in living cells is urgently needed to disclose the important roles of HOCl in biological systems. Although a number of fluorescent probes have been exploited to detect HOCl, most of them are easily influenced by other competing reactive oxygen species (ROS) and reactive nitrogen species (RNS), and disturbed by many factors including variabilities in excitation, probe distribution and environmental conditions due to the recognition of HOCl based on the fluorescence changes in single wavelength. Herein, in this work, we present a simple fluorescent probe AETU-HOCl containing a new recognition group (2-amino-ethyl)thiourea (AETU) for the ratiometric detection of HOCl in living cells. Probe AETU-HOCl exhibits outstanding selectivity for HOCl over other ROS/RNS at higher concentrations (0.5 mM, 100 equiv.). More importantly, the rapid response (1 min) and high sensitivity (a detection limit of 3.9 nM) allow it to successfully visualize the intracellular HOCl levels by the ratiometric imaging. The present study would offer a simple, ratiometric, rapid, and ultrasensitive assay for the accurate determination of HOCl in the biological systems.
Cationic fluorescent carbon dots with solution ultra-stability and its rapid/on-site sensing application for HClO

Abstract:Carbon dots (CDs) as a remarkable fluorescent nanomaterial have the advantages of easy preparation, good photostability and high sensitivity. However, the poor aqueous solution stability of carbon dots largely limited their practical application due to the characteristic of easily forming precipitation for long time storage. Here, a kind of cationic fluorescent carbon dots CDs-P(Ph) 3 was designed by introducing a cationic compound, (4-carboxybutyl) triphenyl phosphonium bromide, to construct an electrostatic shell outside the dots. Such electrostatic shell could highly improve carbon dots stability in an aqueous solution to make CDs-P(Ph) 3 stable for long-term storage with negligible aggregation. Meanwhile, the sensitivity of CDs-P(Ph) 3 for hypochlorous acid (HClO) was also enhanced on the basis of the electron-withdrawing effect of cationic substituents on the surface of carbon dots. The limit of detection of CDs-P(Ph) 3 for HClO was as low as ∼0.32 μM. Additionally, the fluorescence of CDs-P(Ph) 3 could be rapid quenched by HClO with a quenching efficiency of more than 80% within 30 s. The excellent stability of CDs-P(Ph) 3 in an aqueous solution made it suitable for on-site detecting HClO in real samples, such as tap, well and lake water. Such designed fluorescent nanomaterial would provide a practical application pathway for optical sensing detection in environmental samples.

Abstract:The generation and presence of excessive hypochlorous acid derivative ionic form (ClO−) could cause various diseases, such as arteriosclerosis, DNA damage, and cardiovascular illness. It is a critical need to develop a highly sensitive sensor for reliable detection of ClO− in cells and water-soluble systems. In this work, a hydroxyl group has been introduced into the compound 2-amino-3-(((E)-4-(2-(2-(2-hydroxyethoxy)ethyl)-1,3-dioxo-2,3-dihydro-1H-benzo[de]isoquinolin-6-yl)benzylidene)amino)maleonitrile (NDC) to increase its solubility in water, at the same time, the hydrazone unit was designed as a specific recognition group for the "off–on" fluorescence probe of ClO−. The probe NDC presents high selectivity, sensitivity, anti-interference, and low detection limit (67 nM) for ClO−. The recognition mechanism that ClO− breaks the C=N bond and forms the fluorescent compound 4-(2-(2-(2-hydroxyethoxy)ethyl)-1,3-dioxo-2,3-dihydro-1H-benzo[de]isoquinolin-6-yl)benzaldehyde (ND-3) has been confirmed by time-of-flight mass spectrometry. The probe NDC presents a good performance in the actual test of water samples and can be designed as the test papers for the quick and convenient detection of ClO− range from 0 to 1 μM. Moreover, the practical application was demonstrated by the successful imaging of endogenous and exogenous ClO− in HeLa cells. Our fluorescent biomass-based platform opens vast possibilities for repeatability, sensitivity, and selectivity detection of ClO− in cells and water-soluble systems.

Abstract:A fluorescent probe capable of HClO detection containing a para-DMTC benzyl pyridinium moiety at the meso position of BODIPY as self-immolative portion was developed. The probe exhibited excellent specificity for HOCl over other ROS/RNS. Density functional theory calculation showed that photoinduced electron transfer took place from the electron-rich BODIPY core to the electron-poor 4-DMTC benzyl pyridinium moiety, resulting in the weak fluorescence of the probe. Moreover, the probe could monitor fluctuations of exogenous and endogenous HOCl in HeLa and RAW 264.7 cells. A novel NIR fluorescent probe with fast response and large stokes shift for the detection and imaging of hypochlorous acid in living cells

Abstract:Hypochlorous acid (HOCl), one of the physiologically important reactive oxygen species (ROS), is engaged in a diverse variety of biological and pathophysiological functions. Excessive HOCl buildup can cause a variety of illnesses, therefore, the identification of events in real-time of abnormal levels of exogenous and endogenous HOCl in biological settings is extremely important. In the present paper, we reported a new near-infrared emissive fluorescent probe D-HBT-HOCl for the detection and bioimaging of HOCl in live cells. With regard to the construction of D-HBT-HOCl, we combined the dicyanoisophorone motif with a 2-aminobenzenethiol unit, resulting in the near-infrared emission wavelength of 700 nm and an ultra-large Stokes shift of 180 nm, which potentially endowed D-HBT-HOCl with superiority in biological imaging. In addition, the N, N-dimethylthiocarbamate served as an efficient recognition group for HOCl as well as a fluorescence quenching group in the probe. The results of the experiments showed that the newly designed probe exhibited excellent sensing capacity, including outstanding selectivity, a high signal-to-noise ratio, and a rapid response time for the detection of HOCl. What's more, this probe was further effectively leveraged for the imaging of both extrinsic and intrinsic HOCl in live cells, demonstrating its versatility, and possessed the potential to be used to further investigate the physiological and pathological activity of HOCl in biological systems.

Abstract:Hypochlorite (ClO-), as one of reactive oxygen species (ROS), is closely linked to various illnesses and is essential for the proper functioning of immune system. Hence, monitoring and assessing ClO- levels in organisms are extremely important for the clinical diagnosis of ClO--related disorders. In this study, a novel ClO--selective fluorescent probe, DCP-ClO, was synthesized with dicyanoisophorone-xanthene unit as parent fluorophore, which displayed excellent selectivity towards ClO-, near-infrared emission (755 nm), large Stokes shift (100 nm), real-time response to ClO-, high sensitivity (LOD = 3.95 × 10-8 M), and low cytotoxicity. The recognition mechanism of DCP-ClO towards ClO- was confirmed to be a typical ICT process by HPLC-MS, HR-MS, 1H NMR and theoretical calculations. Meanwhile, DCP-ClO demonstrated remarkable efficacy in monitoring ClO- levels in water samples and eye-catching ability in imaging endogenous/exogenous ClO- in living organisms, which verified its potential as a powerful tool for the recognition of ClO- in complex biological systems.
Preparation of One-Emission Nitrogen-Fluorine-Doped Carbon Quantum Dots and Their Applications in Environmental Water Samples and Living Cells for ClO− Detection and Imaging

Abstract:Hypochlorite (ClO−) has received extensive attention owing to its significant roles in the immune defense and pathogenesis of numerous diseases. However, excessive or misplaced production of ClO− may pose certain diseases. Thus, to determine its biological functions in depth, ClO− should be tested in biosystems. In this study, a facile, one-pot synthesis of nitrogen-fluorine-doped carbon quantum dots (N, F-CDs) was developed using ammonium citrate tribasic, L-alanine, and ammonium fluoride as raw materials under hydrothermal conditions. The prepared N, F-CDs demonstrate not only strong blue fluorescence emission with a high fluorescence quantum yield (26.3%) but also a small particle size of approximately 2.9 nm, as well as excellent water solubility and biocompatibility. Meanwhile, the as-prepared N, F-CDs exhibit good performance in the highly selective and sensitive detection of ClO−. Thus, a wide concentration response range of 0–600 μM with a low limit of detection (0.75 μM) was favorably obtained for the N, F-CDs. Based on the excellent fluorescence stability, excellent water solubility, and low cell toxicity, the practicality and viability of the fluorescent composites were also successfully verified via detecting ClO− in water samples and living RAW 264.7 cells. The proposed probe is expected to provide a new approach for detecting ClO− in other organelles.

Abstract:Hypochlorous acid (HClO) is a bioactive molecule that bears an important function in various physiological and pathological processes such as cell signal transduction, pathogen invasion, and redox balance regulation. The near-infrared (NIR) fluorescent probe with a large Stokes shift for bioimaging of HClO is envisioned to outperform other fluorescence probes with following advantages: greater penetration depth, smaller overlap between emission and absorption spectra, and greater reduction in false results caused by the excitation light and the scattered light, aiming to achieve selective and sensitive bioimaging of HClO in vivo. Herein, a NIR fluorescent probe, TJM, was synthesized by modifying the structure of rhodamine dye to make it water soluble and biocompatible for living cells. Under being excited at 585 nm, TJM exhibits an emission peak at 730 nm with a large Stokes shift of 145 nm. TJM also provides a fast NIR fluorescent response to HClO with excellent selectivity, high sensitivity, and low detection limit (0.11 μM). The fluorescence intensity of TJM is triggered by HClO based on HClO-induced oxidization of dibenzoyl hydrazine to dibenzoyl diimide, allowing imaging of HClO in living cells, zebrafish, and different disease mice models. TJM has been demonstrated a suitable NIR probe for imaging endogenous HClO in biological systems.

Abstract:As a vital reactive oxygen species (ROS), endogenous hypochlorous acid (HOCl) deeply engaged in masses of pathophysiological processes owing to its high reactivity, thus the detection of HOCl is awfully valuable for elucidating HOCl-related pathophysiology mechanism. Despite the extensive development of outstanding HOCl fluorescent probes so far, very few of them could be applied for monitoring HOCl in the pure water system. Herein, we present a novel rhodol-based fluorescent probe 6 for the unambiguous detection of HOCl in pure water, which was effectively designed and synthesized by using the spirocyclic hydrazide group as the HOCl sensitive site and introducing two hydroxyethyl groups for obtaining high hydrophilicity into the rhodol skeleton, a unique rhodamine structure. Compared to the control compound 5 without hydrophilic groups only capable to be used in the aqueous system containing organic solvent, 6 achieves the detection of HOCl not only in the above regular system but also in pure water, indicating that the introduction of hydrophilic hydroxyethyl group is an effective and reasonable approach to achieve the broader-spectrum detection of HOCl. More importantly, different with 5, highly water-soluble 6 regardless of dissolving in the regular system or pure water system can achieve the visual monitoring of HOCl with prominent sensitivity and high spatiotemporal resolution in living HepG2 cells and zebrafish, confirming the robust imaging ability of our probe 6 to HOCl in complex biosystem.

Abstract:Hypochlorous acid (HOCl) is crucial in various physiological processes, and monitoring its level in organisms is essential for understanding its biological functions and pathological effects. In this study, we developed a novel fluorescent probe based on dicyclohexylphosphine for rapid and sensitive detection of HOCl in an aqueous solution. The probe exhibits a significant fluorescence turn-on response to HOCl through the specific oxidative reaction between dicyclohexylphosphine and HOCl. It possesses high selectivity, instantaneous response (approximately 1 s), and a low detection limit of 15.3 nM. Moreover, biological imaging results demonstrated that the probe could be used for real-time fluorescence imaging of HOCl in living cells and zebrafish. The development of this probe may provide new insights into designing phosphine-based fluorescent probes for detecting oxidative species.
