Abstract:An aza-BODIPY-CNOH probe attached aldoxime group demonstrated the specific detection for hypochlorous acid by the turn-on red emission signal. NMR and HRMS experiments confirmed that the fluorescence originated from the oxidation degradation of the non-fluorescence, aldoxime-based aza-BODIPY-CNOH probe into the red-fluorescence, nitrile oxide-based aza-BODIPY compound aza-BODIPY-CNO. The aza-BODIPY-CNOH probe showed good biocompatibility and was low toxic to living cells as shown from MTT experiments. Living RAW264.7 cells imaging indicated the aza-BODIPY-CNOH probe had good permeability and either exogenous or endogenous HClO caused the intracellular bright-red fluorescence, showing its potential hypochlorous acid-specific sensing ability in biological systems.

Abstract:It is necessary to develop sensitive and selective probes for real-time in vivo monitoring of hypochlorous acid (HClO) which plays a significant role in physiological and pathological processes. The second near-infrared (NIRII) luminescent silver chalcogenide quantum dots (QDs) have shown great potential in developing activatable nanoprobe for HClO in terms of their outstanding imaging performance in the living organism. However, the limited strategy for the construction of activatable nanoprobes severely restricts their widespread applications. Herein, we proposed a novel strategy for developing an activatable silver chalcogenide QDs nanoprobe for NIR-II fluorescence imaging of HClO in vivo. The nanoprobe was fabricated by mixing an Au-precursor solution with Ag2Te@Ag2S QDs to allow cation exchange and release Ag ions and then reducing the released Ag ions on the QDs surface to form an Ag shell for quenching of the emission of QDs. The Ag shell of QDs was oxidized and etched in the presence of HClO, resulting in the disappearance of their quenching effect on QDs and the activation of the QDs emission. The developed nanoprobe enabled highly sensitive and selective determination of HClO and imaging of HClO in arthritis and peritonitis. This study provides a novel strategy for the construction of activatable nanoprobe based on QDs and a promising tool for NIR-II imaging of HClO in vivo.

Abstract:<p>Accurate detection of HOCl with high sensitivity and high selectivity is essential to elucidate its functions in health or disease states. Herein we propose a new fluorescence HOCl probe (NFL-S) by combining reasonable design strategies and dye screening methods. The probe NFL-S has fast response rate, excellent sensitivity (LOD = 35.0 nM) and outstanding selectivity toward HOCl. It has been successfully applied to detect endogenous HOCl produced by RAW264.7 cells, and further applied to oxidative stress imaging in a mouse arthritis model. Therefore, the probe NFL-S is highly promising to be useful for biological tool to disclose the roles of HOCl in different physiological and pathological contexts.</p>

Abstract:Recent investigations have suggested that abnormally elevated levels of HOCl may be tightly related to the severity of neuroinflammation. Although some successes have been achieved, fluorescent probes with far-red fluorescence emission and capable of detecting HOCl with high specificity in pure aqueous solution are still urgently needed. Herein, a responsive far-red fluorescent probe, DCI-H, has been constructed to monitor HOCl activity in vivo and in vitro. DCI-H could rapidly respond to HOCl within 120 s and had a low detection limit for HOCl of 1.5 nM. Importantly, physiologically common interfering species, except for HOCl, did not cause a change in the fluorescence intensity of DCI-HOCl at 655 nm. The results of confocal imaging demonstrated the ability of DCI-H to visualize endogenous HOCl produced by MPO-catalyzed H2O2/Cl− and LPS stimulation. With the assistance of DCI-H, upregulation of HOCl levels was observed in the mice model of LPS-induced neuroinflammation. Thus, we believed that DCI-H provided a valuable tool for HOCl detection and diagnosis of inflammation-related diseases.

Abstract:The phagocyte's lysosome is the primary site of hypochlorous acid (HOCl) synthesis, and HOCl can be used as a biomarker for osteoarthritis diagnosis and treatment evaluation. Accurate detection of HOCl with high sensitivity and selectivity is required to understand its activities in healthy bio-systems and diseases. By integrating acceptable design principles and dye screening methodologies, we proposed and developed a novel near-infrared fluorescent HOCl sensing probe (FNIR-HOCl). The FNIR-HOCl probe has a quick reaction rate, high sensitivity (LOD = 70 nM), and excellent selectivity toward HOCl over other metal ions and reactive oxygen species. It has been successfully implemented to detect endogenous HOCl produced by RAW264.7 cells, as well as in vivo imaging towards mice with osteoarthritis. As a result, the probe FNIR-HOCl is extremely promising as a biological tool for revealing the roles of HOCl in various physiological and pathological contexts.

Abstract:Hypochlorous acid (HOCl), one of the physiologically important reactive oxygen species (ROS), is engaged in a diverse variety of biological and pathophysiological functions. Excessive HOCl buildup can cause a variety of illnesses, therefore, the identification of events in real-time of abnormal levels of exogenous and endogenous HOCl in biological settings is extremely important. In the present paper, we reported a new near-infrared emissive fluorescent probe D-HBT-HOCl for the detection and bioimaging of HOCl in live cells. With regard to the construction of D-HBT-HOCl, we combined the dicyanoisophorone motif with a 2-aminobenzenethiol unit, resulting in the near-infrared emission wavelength of 700 nm and an ultra-large Stokes shift of 180 nm, which potentially endowed D-HBT-HOCl with superiority in biological imaging. In addition, the N, N-dimethylthiocarbamate served as an efficient recognition group for HOCl as well as a fluorescence quenching group in the probe. The results of the experiments showed that the newly designed probe exhibited excellent sensing capacity, including outstanding selectivity, a high signal-to-noise ratio, and a rapid response time for the detection of HOCl. What's more, this probe was further effectively leveraged for the imaging of both extrinsic and intrinsic HOCl in live cells, demonstrating its versatility, and possessed the potential to be used to further investigate the physiological and pathological activity of HOCl in biological systems.

Abstract:Hypochlorous acid (HClO) is a bioactive molecule that bears an important function in various physiological and pathological processes such as cell signal transduction, pathogen invasion, and redox balance regulation. The near-infrared (NIR) fluorescent probe with a large Stokes shift for bioimaging of HClO is envisioned to outperform other fluorescence probes with following advantages: greater penetration depth, smaller overlap between emission and absorption spectra, and greater reduction in false results caused by the excitation light and the scattered light, aiming to achieve selective and sensitive bioimaging of HClO in vivo. Herein, a NIR fluorescent probe, TJM, was synthesized by modifying the structure of rhodamine dye to make it water soluble and biocompatible for living cells. Under being excited at 585 nm, TJM exhibits an emission peak at 730 nm with a large Stokes shift of 145 nm. TJM also provides a fast NIR fluorescent response to HClO with excellent selectivity, high sensitivity, and low detection limit (0.11 μM). The fluorescence intensity of TJM is triggered by HClO based on HClO-induced oxidization of dibenzoyl hydrazine to dibenzoyl diimide, allowing imaging of HClO in living cells, zebrafish, and different disease mice models. TJM has been demonstrated a suitable NIR probe for imaging endogenous HClO in biological systems.

Abstract:Detection of hypochlorous acid (HClO) in the living system may help to uncover its essential biological functions. However, current imaging agents suffer from poor water solubility that limit their live-tissue applications. Here, a water-soluble probe (NNH) is devised through innovative hydrazone modification of 1,8-naphthalimide at 3 position. NNH was successfully applied to tracking endogenous HClO in both cultured macrophages and a liver injury model in mice. NNH demonstrated remarkably increased water solubility and multiple desirable features including two-photon absorbance, anti-bleaching capability, rapid cellular uptake, and low cytotoxicity. NNH is the first hydrazone-based probe for real-time imaging of HClO in live tissue.

Abstract:Hypochlorous acid (HClO) is a reactive oxygen species and a relatively strong antibacterial substance in the immune defense system. The normal concentration of HClO in the human body is approximately 200 μM, and its high concentration can cause tissue damage and some diseases. Herein, a chromenoquinoline-based ratiometric fluorescent probe was developed to detect and quantify HClO. The developed Probe 1 exhibited the advantages of large Stokes shift (137 nm), high synthetic yield (84.7 %), simple synthesis method, short response time (<4 min), low detection limit (5.1 nM), and low toxicity. The probe was successfully validated in live cells and zebrafish.

Abstract:Hypochlorous acid (HClO) is produced by white blood cells to defend against injury and bacteria. However, as one of the reactive oxygen species, high intracellular HClO concentration could lead to chronic diseases that affect the cardiovascular and nervous systems. To monitor HClO concentrations in bio-samples, the fluorescent probe is preferred to have: a) absorbability in the far-red window with reduced light-toxicity and improved tissue penetration depth, b) ratiometric feature for accurate analysis. In this study, we reported a far-red ratiometric HClO fluorescence probe based on BODIPY chromophore and aldoxime sensing group. Not only the color change of the probe solution can be detected by naked eyes, but also the emission ratios (I645/I670) showed a significant increase upon the introduction of HClO. More importantly, the feasibility of HClO monitoring in bio-samples was demonstrated in vitro using a confocal microscope.

Abstract:The accurate and sensitive determination of basal hypochlorous acid (HOCl) in the biosystems is critical for unraveling its various physiological and pathological functions. Additionally, recent studies have shown that cancer cells possess higher concentration HOCl than normal cells. However, related studies about the discrimation of tumor cells and tissues from normal cells and tissues by using a simple HOCl probe have seldom been reported. Herein, we presented a simple, water-soluble, two-photon long-wavelength fluorescent probe TPFR for detecting HOCl with outstanding selectivity over other potential interfering substances. The fluorescence titration experiments demonstrated that probe TPFR was capable of quantitatively detecting 0-500 nM HOCl (DL = 0.9 nM). Additionally, the rapid response ( < 1 min) of probe TPFR for HOCl facilitated the detection of HOCl with high reactivity and short-time in the biosystems. The outstanding sensing performances of probe TPFR enabled it to be used to track intracellular basal HOCl and differentiate cancer cells from normal cells. Furthermore, probe TPFR was successfully used to discriminate tumor tissues from normal tissues in mice by two-photon excitation microscopy.

Abstract:The detection of hypochlorous acid (HOCl) in vivo is vitally important because the local concentration of HOCl is highly correlated with some diseases such as atherosclerosis and rheumatoid arthritis. However, in vivo detection of HOCl remains a challenge due to the lack of a suitable probe. We report here a near-infrared (NIR) emissive "turn-on" probe (FDOCl-1) based on a methylene blue derivative, which can quickly detect HOCl via a newly found deformylation mechanism. FDOCl-1 displays remarkable selectivity and sensitivity towards HOCl. The dramatic changes in colour and NIR emission were used to detect HOCl in vitro and in vivo in a mouse arthritis model.

Abstract:Hypochlorous acid (HClO/ClO-) is a key reactive oxidative species (ROS) in the body. The HClO/ClO- concentrations are imbalanced during cancer formation due to the ROS stress response. This paper introduces a novel chitosan-based self-calibration fluorescent nanoprobe (ChCyNil) constructed by molecular assembly for the ratiometric detection of HClO/ClO-. Two chromophores with different fluorescence characteristics and HClO/ClO- sensitivity were labeled on chitosan, and nanoparticles were prepared by a self-assembly strategy for HClO/ClO- detection. ChCyNil exhibits several advantages, such as dual near-infrared emissions at 670 nm and 845 nm, tunable fluorescence intensity, self-calibration fluorescence, and good biocompatibility, improving its accuracy in HClO/ClO- detection. Our study confirmed that ChCyNil exhibits a well-assembled spheroidal nanostructure and good photophysical properties in solution. The fluorescence imaging properties were further proved by detecting endogenous HClO/ClO- produced by LPS/PMA stimuli in cells and zebrafish. In addition, ChCyNil was used to detect the fluorescence behavior of HClO/ClO- in tumors of live mice. The successful design and fabrication of ChCyNil have presented a new strategy for constructing detection tools with improved fluorescence properties for HClO/ClO- in live animals.

Abstract:Overexposure to hypochlorous acid (HOCl) can lead to cellular oxidative stress. In response to the stress, the body activates related factor that upregulates a potent antioxidant enzyme, NAD(P)H:quinone oxidoreductase 1 (NQO1). A method that can simultaneously detect HOCl and NQO1 can help to understand their interactions during homeostasis. However, the methods are still limited due to the complexity of the biological environment. In this work, we present for the first time a dual-ratiometric fluorescent probe (NH) that can distinguish between HOCl and NQO1. Probe NH had ratiometric fluorescence responses by changing from red (λmaxem = 643 nm) to green (λmaxem = 517 nm) in the presence of HOCl and from red to mixed blue (λmaxem = 483 nm)/red in the presence of NQO1 under single-wavelength excitation. This probe had high selectivity and sensitivity towards HOCl and NQO1 in vitro. Moreover, probe NH could specifically image exogenous or endogenous HOCl and NQO1 in living cells through different emission channels. This probe provides a practical tool for studying the correlation of HOCl and NQO1 in biological systems.

Abstract:A novel HOCl‐activated NIR‐IIb emissive lanthanide nanoprobe is designed by integrating NaLuF4Yb/Er@NaLuF4 downshift nanoparticles (DSNPs) and HOCl recognition/NIR‐IIb emissive modulation unit of M2‐xS (M = Cu, Co, Pb) nanodots for real‐time monitoring of HOCl levels via competitive absorption induced fluorescence quenching mechanism. Hypochlorous acid (HOCl), as an indispensable signaling molecule in organisms, is one of the key members of reactive oxygen species (ROS). However, in vivo, real‐time dynamic near‐infrared fluorescence imaging of HOCl levels in the 1400‐1700 nm sub‐window (NIR‐IIb) remains a major challenge due to the lack of suitable detection methods. Herein, a general design of HOCl‐responsive NIR‐IIb fluorescence nanoprobe is proposed by integrating NaLuF4Yb/Er@NaLuF4 downshift nanoparticles (DSNPs) and HOCl recognition/NIR‐IIb emissive modulation unit of M2‐xS (M = Cu, Co, Pb) nanodots for real‐time monitoring of HOCl levels. The fluorescence modulation unit of M2‐xS nanodots presents remarkably enhanced absorption than Yb sensitizer at 980 nm and greatly inhibits the NIR‐IIb fluorescence emission via competitive absorption mechanism. While, the M2‐xS nanodots are easily degraded after triggering by HOCl, resulting in HOCl responsive turn‐on (≈ten folds) NIR‐IIb emission at 1532 nm. More importantly, in vivo highly precise and specific monitoring of inflammatory with abnormal HOCl expression is successfully achieved. Thus, the explored competitive absorption mediated quenching‐activation mechanism provides a new general strategy of designing HOCl‐responsive NIR‐IIb fluorescence nanoprobe for highly specific and sensitive HOCl detection.

Abstract:Based on the important role of hypochlorous acid (HOCl) in the immune system and numerous physiological processes, the detection of intracellular basal HOCl is of significant interest. In this work, we present a simple thiocarbamate-protected fluorescein fluorescent probe, TCFL, for imaging basal HOCl in live cells. Surprisingly, probe TCFL could determine HOCl quantitatively in a large concentration range with a detection limit of 0.65 nM. In addition, probe TCFL showed excellent specificity for HOCl in the presence of other higher concentration analytes (1 mM). Moreover, probe TCFL exhibited a rapid response (within 3 s) to HOCl and thus could provide a tool for real-time monitoring of HOCl. Importantly, probe TCFL with outstanding response features could be applied for monitoring basal HOCl in live cells.

Abstract:Development of exquisitely selective and sensitive HClO/ClO– sensor in living system is of the utmost importance. To achieve near-infrared (NIR)-responsive detection of HClO/ClO–, a new nanoprobe (csUCNP-Cy820) is composed of the ClO–-sensitive Cy820 (energy acceptor), and NaLuF4:20%Yb,1%Tm@NaLuF4 core@shell upconversion nanoparticles (csUCNP, energy donor) capable of emitting NIR upconversion luminescence (UCL) of Tm3+ (800 nm). Through the mechanism of Förster resonance energy transfer (FRET), the UCL emission can be recovered in the presence of HClO/ClO–. The csUCNP-Cy820 nanoprobe is effectively adapted as a precise ClO– detection sensor with a low limit of detection (LoD) of 58 nmol/L in vitro. Moreover, owing to excitation and emission wavelengths both falling within the NIR region, the nanoprobe facilitates high quality imaging in mice models of peritonitis and arthritis, thereby enabling deeper penetration depth for imaging detection in vivo.

Abstract:The development of simple techniques for monitoring hypochlorous acid (HOCl) in living cells is urgently needed to disclose the important roles of HOCl in biological systems. Although a number of fluorescent probes have been exploited to detect HOCl, most of them are easily influenced by other competing reactive oxygen species (ROS) and reactive nitrogen species (RNS), and disturbed by many factors including variabilities in excitation, probe distribution and environmental conditions due to the recognition of HOCl based on the fluorescence changes in single wavelength. Herein, in this work, we present a simple fluorescent probe AETU-HOCl containing a new recognition group (2-amino-ethyl)thiourea (AETU) for the ratiometric detection of HOCl in living cells. Probe AETU-HOCl exhibits outstanding selectivity for HOCl over other ROS/RNS at higher concentrations (0.5 mM, 100 equiv.). More importantly, the rapid response (1 min) and high sensitivity (a detection limit of 3.9 nM) allow it to successfully visualize the intracellular HOCl levels by the ratiometric imaging. The present study would offer a simple, ratiometric, rapid, and ultrasensitive assay for the accurate determination of HOCl in the biological systems.
