Abstract:Mercury pollution in aquatic environment is becoming increasingly serious, which poses a threat to the ecological environment and human health. Traditional methods are costly and time consuming since they require sophisticated instruments and professional operation. Thus simple and on-site detection of mercury (II) ions (Hg2+) is extremely expected. In this study, we constructed a one-step lateral flow strip that can selectively detect Hg2+ in solution based on the thymine-Hg2+-thymidine (T-Hg2+-T) mismatch principle. Detect probe (poly T20) were designed to bind to compete probe (poly A20) which can compete with Hg2+ to bind detect probe, leading to the color change on the test line. Moreover, a home-made bionic E-eye system was developed as a portable in-situ detection platform to efficiently measure the Hg2+ in 10 minutes by direct photographing. Under optimized experimental conditions, the developed lateral flow strip had a detection limit of 2 ppb and could detect up to 400ppb.

Abstract:Mercury is one of the heavy metal ions that are released to the environment through various means including anthropogenic pollution. A very low dose of mercury in environmental systems such as water can lead to debilitating effects on the health of humans and animals, if consumed untreated. To reduce this effect, it is critical to develop a simple, non-expensive, and non-sophisticated detection and monitoring system of mercury. Here, we report a simple colorimetric method based on the peroxidase-like activity of AuNPs/ZnO/Fe 3 O 4 . Different characterization techniques such as scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and surface-enhanced Raman spectroscopy (SERS) were used to investigate the successful formation of these composites. The formation of AuNPs was assisted by ZnO. The low Km value (0.7368 mM) obtained from the kinetic study of the catalytic oxidation of 3, 3′, 5, 5′ − Tetramethylbenzidine (TMB) showed strong affinity of TMB to AgNPs/ZnO/Fe 3 O 4 . The peroxidase-like activity of AgNPs/ZnO/Fe 3 O 4 , was further enhanced in the presence of mercury (II). The absorption intensities of the resulting oxidized TMB (oxTMB) increased linearly in a concentration range of 0.05 – 20 μM. The method also exhibited high selectivity to mercury (II) ions. The limit of detection (LOD) in this linear range was found to be 28.1 nM. The feasibility of the method in determination of mercury (II) ions from real samples was confirmed from high recovery rates in tap water samples. The method shows a great potential for determination of mercury (II) ions in environmental samples.

Abstract:Mercury is a common heavy metal element in natural systems and is highly toxic to the human body. Herein, a novel colorimetric detection of Hg2+ ions is proposed based on the aggregation of gold nanoparticles (AuNPs) induced by carbon quantum dots (CDs) with the assistance of glutathione (GSH). In this sensing system, the AuNP/CDs composite forms through Au-N bonds. Simultaneously, the color of the solution turns from wine red to blue. The well-dispersed AuNPs can be restored after the addition of GSH, because GSH competes with CDs to bind onto the surface of AuNPs and protect AuNPs from aggregation. In the presence of Hg2+ ions, GSH can chelate with Hg2+ to form a complex, which subsequently enables CDs to facilitate the aggregation of the AuNPs again. Therefore, according to the red-to-blue color change, a colorimetric sensor is established for the sensitive and selective detection of Hg2+ with a detection limit of 7.5 nM. Moreover, this sensor is successfully used to detect Hg2+ spiked in environmental water. This very simple and cost-effective strategy will promote the development of a colorimetric sensor for the determination of other metal ions in biological and environmental fields.

Abstract:Mercury ion (Hg2+) is an extremely hazardous pollutant to humans, soil, and aquatic life. Nanozyme-based sensing approaches are promising for detecting toxic heavy metal ions. However, applying noble metal nanozymes in developing affordable and portable sensors remains largely unexplored. Herein, a gold nanoparticle (AuNP)-based colorimetric sensor was established for the ultra-trace detection of Hg2+ by capitalizing the inherent peroxidase-mimetic features of AuNPs for oxidizing the colorimetric indicator 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). AuNPs of size less than 10 nm were directly prepared in aqueous media using a stimuli-responsive, poly(ethylene glycol methyl ether methacrylate)-b-poly(dimethylaminoethyl methacrylate (p(PEGEMA)-b-p(DMAEMA)) block copolymer, synthesized through RAFT polymerization. The block copolymers efficiently interacted with Au3+ ions toward the formation of stable, monodispersed AuNPs without additional reducing agents or stabilizers. Using TMB, H2O2, and the AuNP colorimetric system, the concentration of Hg2+ in aqueous media was quantitatively and selectively detected over those of other common interfering metal ions. The selective detection of Hg2+ was promoted by the Au-Hg amalgamation process, which is correlated with intensity of the colorimetric response. Colorimetrically, Hg2+ was linearly measured between 10 nM and 3.5 μM and achieved a detection limit of 0.4 nM. Subsequently, an on-field naked-eye sensing strategy was also developed by integrating the colorimetric sensor on a paper analytical device with a detection limit of 3.5 nM. The efficient colorimetric sensing platform for selective and trace detection of Hg2+ is promising for determining mercury contamination in different water and biological samples.

Abstract:Mercury is a toxic heavy metal that has debilitating effects on human health. Anthropogenic and natural processes are responsible for the availability of mercury in the environment that necessitates periodic monitoring. Most of the techniques commonly employed for analysis of mercury, however, are expensive and require highly trained personnel. Here we report on a cheap colorimetric method using gold nanoparticles loaded carbon covered magnetic nanoparticles (Fe 3 O 4 @C@AuNPs) as nanozymes. The particles were characterized by Fourier translation infrared spectroscopy (FT-IR), X-ray powder diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The AuNPs establish specific interactions with mercury (II) and the peroxidase-like activity of Fe 3 O 4 @C@AuNPs using 3, 3, 5, 5′-tetramethylbenzidine (TMB) as a substrate was enhanced on addition of mercury (II). Our results show that the absorption signal of oxidized TMB increased linearly with concentrations of mercury (II) for a concentration range of 0.001–25 μM. The limit of detection (LOD) was found to be 0.0435 μM (43.5 nM). The feasibility of the method in real sample application was tested by analyzing mercury (II) ions in tap water. From our findings, we anticipate that our method can be applied for analysis of mercury (II) in various environmental samples.

Abstract:Abstract A colorimetric and turn-on fluorometric assay with high sensitivity and selectivity is described for the optical detection of mercury (II) ions (Hg 2+ ), based on carbon dots with –SH (SN-CDs) and gold nanoparticles (AuNPs). On addition of Hg 2+ , the color of the system (SN-CDs/AuNPs) changes from red to blue. A new absorption peak appears at 700 nm, and its absorbance increases with the concentration of Hg 2+ , while at 530 nm, the absorbance of AuNPs decreases. Taking the ratio of absorbance at 700 and 530 nm as a signal, a colorimetric method with linear detection range of 0.5–4.0 μ M was established for the determination of Hg 2+ . Meanwhile, citrate ions on the surface of AuNPs can reduce Hg 2+ to Hg 0 , and through the strong affinity of Hg 0 and gold, gold–mercury alloys were formed to occupy the surface of AuNPs, so that the SN-CDs were re-free and the fluorescence of SN-CDs was restored. Consequently, a fluorometric method was founded in the linear detection range from 0.5 to 15.0 μ M of mercury (II). This dual-mode (colorimetric and turn-on fluorometric) method was applied successfully for determination of Hg 2+ in real water samples.

Abstract:A universal strategy for a rapid colorimetric method for Hg 2+ in an aqueous solution is described. The specific binding of Hg 2+ (thymine-Hg 2+ -thymine) with thiolated DNA-functionalized gold nanoparticles (AuNPs) via Au–S bonds increases the spatial hindrance of the AuNP surface, resulting in a weakened catalytic ability of AuNPs to catalyze the reaction between p-nitrophenol and NaBH 4 . Therefore, the color change time (CCT) of the solution from yellow to colorless becomes longer. Based on the kinetic curve of absorbance over time measured by a UV spectrometer, the level of Hg 2+ in aqueous solutions can be easily quantified. A linear relationship between CCT and Hg 2+ concentration was obtained in the 10–600-nM range with a detection limit of 0.20 nM, which is much lower than the limit value (10 nM) defined by the US Environmental Protection Agency for Hg 2+ in drinking water. The excellent sensitivity comes from CCT as the signal output of the probe, rather than the absorbance or wavelength change used in traditional colorimetric probes as the signal output.

Abstract:The contamination of drinking water by heavy metals, such as arsenic and mercury, is a global concern. Herein, we have developed a sensitive, cost-effective, and novel 3D-printed microfluidic device featuring two V-channels (Hg-sensor and As-sensor) for naked-eye colorimetric detection of mercury and arsenic in water samples from Tigris River. In Hg-sensor, a rapid and eco-friendly colorimetric sensor of silver nanoparticles (AgNPs) has been prepared using Boswellia sacra extract as a stabilizing and reducing agent to selectively detect mercury ions. Several techniques have been used to characterize the morphology of the prepared nanosensor of AgNPs, as well as the optical properties. AgNPs decorated with Boswellia sacra molecules (AgNPs@BS) showed high sensitivity and selectivity detection of Hg (II) in river water samples, based on AgNPs' aggregation and the converting brown color of AgNPs to colorless. In As-sensor, a colorimetric sensor was used to detect arsenic ions using leucomalachite (LMG) green dye, where arsenic (III) reacts with potassium iodate in an acidic medium to release iodine, which rapidly oxidizes LMG to malachite green (MG). The application of the microfluidic device for analysis at the point of collection was assessed utilizing colorimetric determination of mercury and arsenic, permitting free imaging of both sensors using a smartphone camera. The limit of detection (LOD) and limit of quantification (LOQ) of this method were reported to be 0.089 and 0.297 mg/L for Hg (II), and 0.042 and 0.143 mg/L for As (III), respectively. The calibration curve that ranged from 2 to 20 mg/L for Hg (II) and 1–15 mg/L for As (III) was obtained for the point of collection device, which reveals a good agreement (97%) with Sector Field ICP-MS.

Abstract:Mercury is very harmful to the environment and human health even at low concentration. Methods for sensitive detection of mercury ion (Hg 2 ＋ ) have increasingly been developed over the past decade owing to the rapid development in nanotechnology. However, the limits of detection (LODs) of these methods are mostly not satisfactory enough to meet the demand of monitoring trace amounts of mercury ion. DNA thymine (T bases) can react with the mercury ion to form T-Hg 2 ＋ T structure, and this interaction has been proved to be much more stable than the interaction between thymine and its com-plementary adenine (A bases). Based on this principle, a series of ultra-sensitive DNA-based colorimetric biosensors, mostly using Au nanoparticles (AuNPs) as DNA carriers, have been designed for detection of mercury ion. In this study, we report a new strategy for highly sensitive Hg 2 ＋ detection based on Hg 2 ＋ -induced AuNPs assembly. AuNPs of different sizes (s-AuNPs of 18 nm and c-AuNPs of 52 nm) were modified with oligonucleotides containing a sequence of continuous T ba-ses. In the presence of Hg 2 ＋ , s-AuNPs would be bound to c-AuNPs in the solution owing to oligonucleotide hybridization, forming a core-satellites nanostrucure. This process was accompanied by a color change of the scattering light from green to orange as observed under dark-field microscopy and a corresponding distinct scattering peak shift. The scattering spectra of the AuNPs were obtained using a spectroscopic system which was established autonomously. The scattering peak shift of color-changed spots corresponded with Hg 2 ＋ concentration. It was increased linearly with logarithm of Hg 2 ＋ concentration over a wide range from 1 pmol/L to 1 nmol/L, with the correlation coefficient of 0.983 ( R 2 ＝ 0.983), and the detection limit of Hg 2 ＋ was estimated to be 1 pmol/L. Other metal ions, such as Ag ＋ , K ＋ , Ca 2 ＋ , Mg 2 ＋ , Zn 2 ＋ , Cd 2 ＋ , Fe 2 ＋ , Pb 2 ＋ , Ni 2 ＋ , Mn 2 ＋ , Al 3 ＋ , induced negligible scattering peak shifts for AuNPs under the same conditions, which showed that this strategy exhib-ited excellent selectivity towards Hg 2 ＋ . Moreover, satisfactory results were obtained when the proposed approach was applied to detect Hg 2 ＋ in real samples with recoveries of 98.7% ～ 103.1% and 105.6% ～ 108.2% for river water and tap water, re-spectively.

Abstract:Contamination of the environment by hazardous metal ions has been a major environmental issue for the past several decades. Among several hazardous metals, mercury ion (Hg(II)) is of particular concern as its compounds are extremely toxic. Hence, developing detection methods for traces of Hg(II) ions in aquatic systems is critical for mercury pollution mitigation. One method that can be used to monitor Hg(II) in aquatic systems is colorimetry-based method which is simple, rapid, and low-cost, yet selective and sensitive. The method can be conducted by applying the Localized Surface Plasmon Resonance phenomenon of metal nanoparticles, such as silver nanoparticles. There are non-Hg(II) ions in the aquatic environment that can interfere the measurements. Thus, a selective method is needed to obtain a valid measurement result. Here, we introduced silver nanoparticles-chitosan (AgNPs-Ch) synthesized by chemical reduction as a selective probe of Hg(II) in an aqueous solution. The AgNPs-Ch was synthesized from silver nitrate at 80°C using trisodium citrate and chitosan as reducing agent and stabilizer, respectively. The synthesized nanoparticles were spherical with an average size below 15.0 nm. Moreover, the AgNPs-Ch was selective for Hg(II) with a linearity of 0.9556 in the concentration range of 1 - 5 ppm and was able to detect the ion down to 1.33 ppm.
Colorimetric and SERS Dual-Mode Sensing of Mercury (II) Based on Controllable Etching of Au@Ag Core/shell Nanoparticles

Abstract:Mercury ion (Hg2+) is a bio-accumulative and toxic heavy metal, which has caused significant damage to our health. It is vital to establish a method for the determination of Hg2+. Here, a new strategy for colorimetric/SERS dual-mode determination of Hg2+ was proposed based on controllable etching Au@Ag NPs. The silver shells could be etched by S2O32- causing the orange colloidal solution to turn into brick red. However, after Hg2+ is added, the etching effect is suppressed due to the reaction between Hg2+ and S2O32- to form an insoluble substance HgS. As a result, the UV-vis absorption at 394 nm and Raman intensity of R6 G increase with a silver shellless etched by S2O32-. About the colorimetric method, the limit of detection was 2 mu M and 0.2 mu M through the naked eye and UV-vis spectroscopy, respectively. Moreover, the SERS method possesses a linear regression for the concentration of Hg2+ (0.1 nM(-1) mu M), and the LOD was 0.1 nM. Furthermore, the sensing strategy has been used to determine Hg2+ in traditional Chinese medicine and real water samples. The selectivity and practicability studies indicate that our method has excellent specificity and sensitivity. Thus, this strategy is suitable for on-site fast detecting of Hg2+.
High-Throughput Color Imaging Hg2+ Sensing via Amalgamation-Mediated Shape Transition of Concave Cube Au Nanoparticles

Abstract:Mercury, as one type of toxic heavy metal, represents a great threat to environmental and biological metabolic systems. Thus, reliable and sensitive quantitative detection of mercury levels is particularly meaningful for environmental protection and human health. We proposed a high-throughput single-particle color imaging strategy under dark-field microscopy (DFM) for mercury ions (Hg2+) detection by using individual concave cube Au nanoparticles as optical probes. In the presence of ascorbic acid (AA), Hg2+ was reduced to Hg which forms Au–Hg amalgamate with Au nanoparticles, altering their localized surface plasmon resonance (LSPR). Transmission electron microscopy (TEM) images demonstrated that the concave cube Au nanoparticles were approaching to sphere upon increasing the concentration of Hg2+. The nanoparticles underwent an obvious color change from red to yellow, green, and finally blue under DFM due to the shape-evolution and LSPR changes. In addition, we demonstrated for the first time that the LSPR of Au–Hg amalgamated below 400 nm. Inspired by the above-mentioned results, single-particle color variations were digitalized by converting the color image into RGB channels to obtain (green+blue)/red intensity ratios [(G+B)/R]. The concentration-dependence change was quantified by statistically analyzing the (G+B)/R ratios of a large number of particles. A linear range from 10 to 2000 nM (R2 = 0.972) and a limit of detection (LOD) of 1.857 nM were acquired. Furthermore, many other metal ions, like Cu2+, Cr3+, etc., did not interfere with Hg2+ detection. More importantly, Hg2+ content in industrial wastewater samples and in the inner regions of human HepG2 cells was determined, showing great potential for developing a single-particle color imaging sensor in complex biological samples using concave cube Au nanoparticles as optical probes.

A novel colorimetric probe for highly selective recognition of Hg2+ ions in aqueous media based on inducing the aggregation of CPB-capped AgNPs: accelerating direct detection for environmental analysis

Abstract:In this study, we have developed a novel and highly selective naked-eye colorimetric probe for the detection of mercury ions (Hg2+) in aqueous solution, using cetylpyridinium bromide (CPB)-capped colloidal silver nanoparticles (AgNPs), based on induced aggregation. The synthesized AgNPs and their interaction with Hg2+ were characterized using UV-Vis absorption spectroscopy, Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS) measurements and zeta potential measurement techniques. The color of the AgNP solution changed from yellowish-brown to colorless within a few minutes following the addition of Hg2+, resulting in a blue shift from 408–354 nm with quenching in the absorption spectra. Under the most favorable conditions, the calibration plot of (A0−A) against concentration of Hg2+ was linear in the range of 0.5–10 μg mL−1 with a correlation coefficient of 0.9931. The concentration of Hg2+ is quantitatively determined using a UV-Vis spectrophotometer with a Limit of Detection (LOD) of 0.80 μg mL−1. In addition, this method shows an excellent selectivity towards Hg2+ over sixteen other interfering cations tested. The accuracy and reliability of the method were further ascertained for the detection of Hg2+ from water bodies via a standard addition method, with percentage recoveries in the range of 99.96–100.13%. The probable mechanism for the observed color changes was also discussed. The method is simple, rapid, specific, and highly selective with good precision.

Abstract:In this study, we present a novel colorimetric method for the precise and sensitive detection of Hg ions utilizing silver nanoparticles (AgNPs) produced and stabilized in an aqueous medium using Poly allyl amine hydrochloride (PAH). Nanoparticles were prepared from silver nitrate and NaBH4 along with CTAB at 26 ± 2 °C with an average size of 30.21 ± 6.5 nm. The prepared AgNPs solution exhibited yellowish brown color with a surface Plasmon peak at 420 nm. By adding Hg ions, the yellow color of the solution was transformed into a colorless suspension, and the color change was proportional to the Hg ion concentration. The presence of other metal ions had no effect on the color (sensitivity); confirming the selectivity of CTAB stabilized PAH capped AgNPs towards Hg ions. The generated probe has a 1 nM low detection limit when measured using a UV–visible spectrophotometer. The proposed approach has the potential to be utilized in real-time environments with greater precession by applying RGB values to make a revolutionary sensor using visible light imaging technique.

Abstract:Mercury ions (Hg 2+ ) are widely found in the environment; it is considered a major pollutant. Therefore, the rapid and reliable detection of Hg 2+ is of great technical interest. In this study, a highly fluorescent, sensitive, and selective fluorometric assay for detecting Hg 2+ ions was developed using vancomycin functionalized and polyethyleneimine stabilized gold nanoparticles (PEI-f-AuNPs@Van). The as-made gold nanoparticles were highly fluorescent, with excitation and emission maxima occurring at 320 and 418 nm, respectively. The size of nanoparticles was ∼7 nm; a zeta potential of ∼38.8 mV was determined. The XRD analysis confirmed that the nanoparticles possessed crystalline structure with face centerd cubic symmetry. Using the PEI-f-AuNP@Van probe, the detection limit of Hg 2+ ion was achieved up to 0.988 nM (within a linear range) by calculating the KSV. However, the detection limit in a natural environmental sample was shown to be 12.5 nM. Furthermore, the selectivity tests confirmed that the designed probe was highly selective to mercury (II) cations among tested other divalent cations. Owing to its sensitivity and selectivity, this approach for Hg 2+ ions detection can be utilized for the analysis of real water samples.

Abstract:A very sensitive and selective colorimetric biosensor for the measurement of mercury ion (Hg 2+ ) in environmental samples has been developed using functionalized gold nanoparticles with bromelain enzyme (brn-AuNPs). This work has shown that Hg 2+ measurement based on spectrophotometer and digital image analysis is a very innovative and successful method for providing an effective preliminary system and has promise for the future of water quality biomonitoring. Response Surface Methodology (RSM), a Box–Behnken design-based technique, was used to identify the optimum levels of functionalization of bromelain to AuNPs. The created model's validity was confirmed, and statistical analysis revealed that the ideal functionalize conditions were 1 mM of AuNPs, functionalize with 0.59 mM bromelain concentration on 14 °C temperature and 72 h incubation time. The lowest colorimetric detection concentration (LOD) of brn-AuNPs of Hg 2+ was 0.0092 ppm and 0.011 ppm for spectrophotometer and digital image analysis. As shown, digital image analysis had advantages based on the LOD result comparable to UV–VIS spectrophotometer. The practical application of the brn-AuNPs sensing was proven with mercury determination in water samples. The present study developed a robust sensor, which successfully implemented in a compact portable sensor kit, turning this sensor into a very potent tool for the development water quality biomonitoring system of Hg 2+ application.

Abstract:In this study, a gold nanoparticles colorimetric probe (AuNPs) with direct response to mercury ions (Hg 2+ ) were developed using treated N-methylpyrrolidone (NMP) and chloroauric acid (HAuCl 4 ) as precursors. NMP showed good reducibility after high temperature hydrolysis and could be used as reducing and stabilizing agent to synthesize AuNPs. The prepared AuNPs have obvious characteristic absorption peaks and appear wine-red. At the same time, it was found that the presence of Hg 2+ can cause the aggregation of AuNPs, increased the absorbance at 700 nm, and changed the color of the solution into blue-gray. This method is capable of sensitive and specific determination of Hg 2+ ranging from 1 to 30 μM, with the limit of detection (LOD) at 0.3 μM. The method showed good specificity for the determination of Hg 2+ and has the potential to be applied to Hg 2+ detection in sewage samples in the environment.

Abstract:Abstract Heavy metal ion pollution is always a serious problem worldwide. Therefore, monitoring heavy metal ions in environmental water is a crucial and difficult step to ensure the safety of people and the environment. A mercury ion (Hg 2+ ) fluorescence probe with excellent sensitivity and selectivity is described here. The functionalized graphitic carbon nitride nanosheets (T/G-C 3 N 4 ) fluorescence probe was fabricated using melamine as a precursor by the pyrolysis technique, followed by a rapid KOH heat treatment method for 2 min. The chemical structure and morphology of the T/G-C 3 N 4 probe were characterized using multiple analytical techniques including UV–Vis, SEM, XPS, XRD, and fluorometer spectroscopy. Geometry optimization of T/G-C 3 N 4 as a modified probe was performed to assess its stability and interaction ability with Hg(II) via using the density function approach. The T/G-C 3 N 4 probe showed a linear response based on quenching over the range 0–1.25 × 10 3 nM Hg(II); the detection limit was 27 nM. The remarkable sensitivity of T/G-C 3 N 4 towards the Hg 2+ ions was explained by the intense coordination and fast chelation kinetics of Hg 2+ with the NH 2 , CN, C=N, and OH groups of T/G-C 3 N 4 nanoprobe. The T/G-C 3 N 4 probe demonstrates exceptional selectivity for Hg 2+ ions among other metal ions including (Na + , Ag + , Mg 2+ , Fe 2+ , Fe 3+ , Co 2+ , Ni 2+ , Cd 2+ , K + , Ca 2+ , Cu 2+ , Pb 2+ , Mn 2+ and Hg 2+ ) and over a broad pH range (6–10), together with remarkable long-term fluorescence stability in water (> 30 days) and minimal toxicity. T/G-C 3 N 4 was used to detect and quantify Hg 2+ ions in tuna and mackerel fish and the results compared to ICP-AES. The results obtained offer a new simple and green technique for the design of multifunctional fluorescent probe appropriate for environmental applications. Graphical Abstract

Abstract:Mercury (Hg) contamination of the major environmental matrices, arising from human indiscriminate pursuit of industrialization without recourse to environment beneficiation is rife and worrisome. Amongst all the different forms of Hg, huge interest has been devoted to the inorganic Hg2+ determination, especially in water. Herein, a creatinine (CRN) conjugated on 3,3′-dithiodipropionic acid di(N-hydroxysuccinimide ester) (DTSP) functionalized gold nanoparticles (Au@DTSP/CRN)-based colorimetric assay was developed for Hg2+ detection in water samples. The DTSP was used to functionalize gold nanoparticles through the Au–S interaction, so as to impart a significantly high stability on the fabricated Au@DTSP, while also conferring coordination capacity on the nanoparticles. The Au@DTSP can bind to the amino group of CRN through its N-hydroxysuccinimide ester moiety, with the formation of an amide bond. The generated optical probe Au@DTSP/CRN aggregated in the presence of Hg2+, with a distinct change of solution colors from ruby red, through purple and to blue-grey within 10 min. The detection limit of 28.5 nM was achieved with linear calibration curves within 0.10–0.35 and 0.35–4.00 μM, while using the absorbance ratio (A680/A521) as the analytical response. Recoveries of 99.0–103.2% were obtained and the relative standard deviations were less than 5.5%. The Au@DTSP/CRN nanoprobe was applied for quantitative determination of Hg2+ in real water samples without significant interference from other metal ions, which is a positive indication of its utilitarian potentiality for Hg2+ profiling in aqueous environment.

Abstract:We report a highly sensitive electrochemical sensor for the detection of Hg(2+) ions in aqueous solution by using a thymine (T)-rich, mercury-specific oligonucleotide (MSO) probe and gold nanoparticles (Au NPs)-based signal amplification. The MSO probe contains seven thymine bases at both ends and a "mute" spacer in the middle, which, in the presence of Hg(2+), forms a hairpin structure via the Hg(2+)-mediated coordination of T-Hg(2+)-T base pairs. The thiolated MSO probe is immobilized on Au electrodes to capture free Hg(2+) in aqueous media, and the MSO-bound Hg(2+) can be electrochemically reduced to Hg(+), which provides a readout signal for quantitative detection of Hg(2+). This direct immobilization strategy leads to a detection limit of 1 microM. In order to improve the sensitivity, MSO probe-modified Au NPs are employed to amplify the electrochemical signals. Au NPs are comodified with the MSO probe and a linking probe that is complementary to a capture DNA probe immobilized on gold electrodes. We demonstrated that this Au NPs-based sensing strategy brings about an amplification factor of more than 3 orders of magnitude, leading to a limit of detection of 0.5 nM (100 ppt), which satisfactorily meets the sensitivity requirement of U.S. Environmental Protection Agency (EPA). This Au NPs-based Hg(2+) sensor also exhibits excellent selectivity over a spectrum of interference metal ions. Considering the high sensitivity and selectivity of this sensor, as well as the cost-effective and portable features of electrochemical techniques, we expect this Au NPs amplified electrochemical sensor will be a promising candidate for field detection of environmentally toxic mercury.
