Abstract:Summary The prevailing account of visually controlled routes is that an ant learns views as it follows a route, while guided by other path-setting mechanisms. Once a set of route views is memorised, the insect follows the route by turning and moving forwards when the view on the retina matches a stored view. We have engineered a situation in which this account cannot suffice in order to discover whether there may be additional components to the performance of routes. One-eyed wood ants were trained to navigate a short route in the laboratory guided by a single black, vertical bar placed in the blinded visual field. Ants thus had to turn away from the route to see the bar. They often turned to look at or beyond the bar and then turned to face in the direction of the goal. Tests in which the bar was shifted to be more peripheral or more frontal than in training produced a corresponding change in the ants’ paths, demonstrating that they were guided by the bar, presumably obtaining information during scanning turns towards the bar. Examination of the endpoints of turns away from the bar suggest that ants use the bar for guidance by learning how large a turn-back is needed to face the goal. We suggest that the ants’ zigzag paths are an integral part of visually guided route following. In addition, on some runs in which ants did not take a direct path to the goal, they still turned to face and sometimes approach the goal for a short stretch. This off-route goal facing indicates that they store a vector from start to goal and use path integration to track their position relative to the endpoint of the vector.

Abstract:Many insects rely on path integration to define direct routes back to their nests. When shuttling hundreds of meters back and forth between a profitable foraging site and a nest, navigational errors accumulate unavoidably in this compass- and odometer-based system. In familiar terrain, terrestrial landmarks can be used to compensate for these errors and safely guide the insect back to its nest with pin-point precision. In this study, we investigated the homing strategies employed by Camponotus fulvopilosus ants when repeatedly foraging no more than 1.25 m away from their nest. Our results reveal that the return journeys of the ants, even when setting out from a feeder from which the ants could easily get home using landmark information alone, are initially guided by path integration. After a short run in the direction given by the home vector, the ants then switched strategies and started to steer according to the landmarks surrounding their nest. We conclude that even when foraging in the close vicinity of its nest, an ant still benefits from its path-integrated vector to direct the start of its return journey.

Abstract:Desert ants are expert navigators, foraging over large distances using visually guided routes. Recent models of route following can reproduce aspects of route guidance, yet the underlying motor patterns do not reflect those of foraging ants. Specifically, these models select the direction of movement by rotating to find the most familiar view. Yet scanning patterns are only occasionally observed in ants. We propose a novel route following strategy inspired by klinokinesis. By using familiarity of the view to modulate the magnitude of alternating left and right turns, and the size of forward steps, this strategy is able to continually correct the heading of a simulated ant to maintain its course along a route. Route following by klinokinesis and visual compass are evaluated against real ant routes in a simulation study and on a mobile robot in the real ant habitat. We report that in unfamiliar surroundings the proposed method can also generate ant-like scanning behaviours.
A comparative study of navigation behaviours in ants

Abstract:Ants inhabit a vast range of ecosystems and exhibit wide morphology. They are expert navigators employing a handful of well-understood navigational strategies. However, the specific relationships among ant navigation behaviours, ecology, and morphology remain unclear, highlighting the need for comparative studies across diverse species. Here, we conducted field displacement experiments with 15 ant species across different habitats, assessing the prevalence of path integration, view-based navigation, olfactory trail following, and backtracking. We further tested whether use of particular navigation strategies was correlated with variation in morphological traits that could affect navigation efficiency, namely body size, eye size (view-based, path integration) and scape length (olfactory). There was a negative correlation between path integration and olfaction across different ant species, and no other clear trade-offs were identified between navigational strategies. Olfactory navigation emerged as the most dominant strategy. Path integration was also prevalent but limited to arboreal ants. View-based navigation was observed in both ground-foraging and tree-climbing ant species, and, unexpectedly, backtracking was also widespread. Species with larger eyes and body size showed a stronger preference for view-based navigation. However, no significant relationship was found between eye size or antennal scape length with preference for either path integration or olfaction. These results highlight the diversity and specialization of navigational strategies in ants, which appear to depend on the species' ecological niche and morphological traits. Our study confirmed that path integration performs better in open sky environments, while view-based navigation appears more effective in cluttered habitats. We also showed the importance of plasticity in both foraging strategies and navigational profile at individual and colony levels, demonstrating the adaptability of ants' navigation strategies to their environment.

A unified mechanism for innate and learned visual landmark guidance in the insect central complex

Abstract:Insects can navigate efficiently in both novel and familiar environments, and this requires flexiblity in how they are guided by sensory cues. A prominent landmark, for example, can elicit strong innate behaviours (attraction or menotaxis) but can also be used, after learning, as a specific directional cue as part of a navigation memory. However, the mechanisms that allow both pathways to co-exist, interact or override each other are largely unknown. Here we propose a model for the behavioural integration of innate and learned guidance based on the neuroanatomy of the central complex (CX), adapted to control landmark guided behaviours. We consider a reward signal provided either by an innate attraction to landmarks or a long-term visual memory in the mushroom bodies (MB) that modulates the formation of a local vector memory in the CX. Using an operant strategy for a simulated agent exploring a simple world containing a single visual cue, we show how the generated short-term memory can support both innate and learned steering behaviour. In addition, we show how this architecture is consistent with the observed effects of unilateral MB lesions in ants that cause a reversion to innate behaviour. We suggest the formation of a directional memory in the CX can be interpreted as transforming rewarding (positive or negative) sensory signals into a mapping of the environment that describes the geometrical attractiveness (or repulsion). We discuss how this scheme might represent an ideal way to combine multisensory information gathered during the exploration of an environment and support optimal cue integration.

Abstract:Many insects use memories of their visual environment to adaptively drive spatial behaviours. In ants, visual memories are fundamental for navigation, whereby foragers follow long visually guided routes to foraging sites and return to the location of their nest. Whilst we understand the basic visual pathway to the memory centres (Optic Lobes to Mushroom Bodies) involved in the storage of visual information, it is still largely unknown what type of representation of visual scenes underpins view-based navigation in ants. Several experimental studies have shown ants using “higher-order” visual information – that is features extracted across the whole extent of a visual scene – which raises the question as to where these features are computed. One such experimental study showed that ants can use the proportion of a shape experienced left of their visual centre to learn and recapitulate a route, a feature referred to as “fractional position of mass” (FPM). In this work, we use a simple model constrained by the known neuroanatomy and information processing properties of the Mushroom Bodies to explore whether the use of the FPM could be a resulting factor of the bilateral organisation of the insect brain, all the whilst assuming a “retinotopic” view representation. We demonstrate that such bilaterally organised memory models can implicitly encode the FPM learned during training. We find that balancing the “quality” of the memory match across left and right hemispheres allows a trained model to retrieve the FPM defined direction, even when the model is tested with other shapes, as demonstrated by ants. The result is shown to be largely independent of model parameter values, therefore suggesting that some aspects of higher-order processing of a visual scene may be emergent from the structure of the neural circuits, rather than computed in discrete processing modules.

Abstract:Cataglyphis desert ants are famous navigators. Like all central place foragers, they are confronted with the challenge to return home, i.e. relocate an inconspicuous nest entrance in the ground, after their extensive foraging trips. When leaving the underground nest for the first time, desert ants perform a striking behavior, so-called learning walks that are well structured. However, it is still unclear how the ants initially acquire the information needed for sky- and landmark-based navigation, in particular how they calibrate their compass system at the beginning of their foraging careers. Using high-speed video analyses, we show that different Cataglyphis species include different types of characteristic turns in their learning walks. Pirouettes are full or partial rotations (tight turns about the vertical body axis) during which the ants frequently stop and gaze back in the direction of the nest entrance during the longest stopping phases. In contrast, voltes are small walked circles without directed stopping phases. Interestingly, only Cataglyphis ant species living in a cluttered, and therefore visually rich, environment (i.e. C. noda and C. aenescens in southern Greece) perform both voltes and pirouettes. They look back to the nest entrance during pirouettes, most probably to take snapshots of the surroundings. In contrast, C. fortis inhabiting featureless saltpans in Tunisia perform only voltes and do not stop during these turns to gaze back at the nest - even if a set of artificial landmarks surrounds the nest entrance.
Social learning of navigational routes in tandem running acorn ants (Temnothorax nylanderi)

Abstract:Tandem running in ants is a form of social learning that involves an informed leader guiding a naive nestmate to a valuable resource, such as a nest site or a food source. Little is currently known about what tandem followers learn and how socially acquired navigational information affects future trips. While some studies suggest that tandem followers learn the resource position but not the route taken by the tandem pair to reach the resource, more recent evidence contradicts this view. We studied tandem running in foraging acorn ants Temnothorax nylanderi and provide evidence that tandem followers socially learn routes from their leaders and later use these routes when travelling between their nest and a food source. Followers that became tandem leaders themselves then guided their follower along the same routes in 90% of tandem runs, demonstrating that navigational information can spread in a forager population through sequential social learning. Ants increased their travelling speed, but not path straightness over successive trips. We also found that ants needed less time on subsequent trips if they experienced longer-lasting tandem runs, suggesting that longer lasting tandem runs allow followers to learn routes more efficiently. Adding visual cues did not affect most of the quantified variables, and we currently know little about the cues used by T. nylanderi during navigation. We discuss how the visual environment inhabited by different species might affect the importance of route learning during tandem running.

Abstract:Navigation mechanisms include path integration and landmark‐based navigation, and we show the first evidence for molecular pathways required for navigation in desert ants. Transcriptional activity in the central brain but not in the optic lobes might be necessary for the estimation of distance traveled, which is crucial for path integration. Correlation in gene expression and correctness in the optic lobes may indicate the importance of visual processing in route recognition. Navigation is crucial for central‐place foragers to locate food and return to the nest. Cataglyphis ants are renowned for their advanced navigation abilities, relying on landmark cues and path integration. This study aims to uncover the transcriptomic basis of exceptional spatial learning in the central nervous system of Cataglyphis niger. Ants navigated a maze with a food reward, and we examined expression changes linked to correct decisions in subsequent runs. Correct decisions correlated with expression changes in the optic lobes, but not the central brain, showing a downregulation of genes associated with sucrose response and Creb3l1. The latter gene is homologous to Drosophila crebA, which is essential for long‐term memory formation. To understand how ants use distance information during path integration, we analyzed expression shifts associated with the last distance traveled. We uncovered a transcriptomic footprint in the central brain, but not in the optic lobes, with genes enriched for energy consumption and neurological functions, including neuronal projection development, synaptic target inhibition, and recognition processes. This suggests that transcriptional activity in the central brain is necessary for estimating distance traveled, which is crucial for path integration. Our study supports the distinct roles of different brain parts for navigation in Cataglyphis ants.

Abstract:Eukaryotic translation initiation factor 2alpha (eIF-2alpha), a target molecule of the interferon-inducible double-stranded-RNA-dependent protein kinase (PKR), was cleaved in apoptotic Saos-2 cells on treatment with poly(I).poly(C) or tumour necrosis factor alpha. This cleavage occurred with a time course similar to that of poly(ADP-ribose) polymerase, a well-known caspase substrate. In addition, eIF-2alpha was cleaved by recombinant active caspase-3 in vitro. By site-directed mutagenesis, the cleavage site was mapped to an Ala-Glu-Val-Asp(300) downward arrowGly(301) sequence located in the C-terminal portion of eIF-2alpha. PKR phosphorylates eIF-2alpha on Ser(51), resulting in the suppression of protein synthesis. PKR-mediated translational suppression was repressed when the C-terminally cleaved product of eIF-2alpha was overexpressed in Saos-2 cells, even though PKR can phosphorylate this cleaved product. These results suggest that caspase-3 or related protease(s) can modulate the efficiency of protein synthesis by cleaving the alpha subunit of eIF-2, a key component in the initiation of translation.
The Use of Edges in Visual Navigation by the Ant Leptothorax albipennis

Abstract:Certain navigating insects home in on their goal by moving so that currently viewed images of landmarks fall on the same retinal locations memorized during previous visits. Here we show that ants can use similar retinotopic learning to guide lengthy routes, by memorizing and walking parallel to a distinct visual edge. We induced workers of the ant Leptothorax albipennis to travel parallel to a prominent wall. When the wall’s height was changed, the ants’ paths consistently shifted toward a lowered wall and away from a raised wall, as would be expected if they attempt to keep the wall’s image at a constant retinal position. These path shifts were smaller than would be expected if the wall was the only guide to navigation, suggesting that other cues are also important. Significantly larger shifts were seen when edge guidance was enhanced by using two walls, one on each side of the path.

The new Sheffield risk and benefit tables for the elderly.

Abstract:Several charts or tables are used to guide treatment in primary prevention of cardiovascular disease (CVD). These usually relate to patients up to 75 years of age, leaving older patients without guidance. Most also present this information as risk, leaving patients to estimate the benefit of treatment and decide whether it is worthwhile. We present tables to display both CVD risk and benefit from treatment in the elderly. A systematic review identified CVD risk functions for the elderly. The Dubbo study of older patients' 5-year CVD risk equation was deemed most appropriate, due to the population studied, endpoints observed and risk factors recorded. By dichotomizing most risk factors, we produced a new risk table in the form of the original 'Sheffield table'. Risk is calculated by selecting the appropriate table for gender and the appropriate cell from the rows and columns, representing age and risk factor contributors, respectively. Total cholesterol above a cell value corresponds to a 20 or 40% 10-year CVD risk. A simple risk scoring system was then derived from the Dubbo equation. Calculation of risk score requires knowledge of a patient's simple demographics, systolic blood pressure and total and high-density lipoprotein cholesterol. Positive integers corresponding to level of risk for each contributing factor are then added together to give a final risk score. A Markov chain model was produced based on the Dubbo derived risk and relative risk reductions from published meta-analyses of 3-hydroxy-3-methyl-glutaryl-CoA reductase inhibitors (statins) and anti-hypertensive treatment. Using this model, individual scores were mapped to likely benefit from treatment in terms of disease free years. Our risk table provides a simple means for calculating risk in the elderly, to two major thresholds, while the benefit table explores the concept of presenting benefit of taking CVD-preventing medication.

Abstract:Abstract The behaviours and cognitive mechanisms animals use to orient, navigate, and remember spatial locations exemplify how cognitive abilities have evolved to suit a number of different mobile lifestyles and habitats. While spatial cognition observed in vertebrates has been well characterised in recent decades, of no less interest are the great strides that have also been made in characterizing and understanding the behavioural and cognitive basis of orientation and navigation in invertebrate models and in particular insects. Insects are known to exhibit remarkable spatial cognitive abilities and are able to successfully migrate over long distances or pinpoint known locations relying on multiple navigational strategies similar to those found in vertebrate models—all while operating under the constraint of relatively limited neural architectures. Insect orientation and navigation systems are often tailored to each species’ ecology, yet common mechanistic principles can be observed repeatedly. Of these, reliance on visual cues is observed across a wide number of insect groups. In this review, we characterise some of the behavioural strategies used by insects to solve navigational problems, including orientation over short-distances, migratory heading maintenance over long distances, and homing behaviours to known locations. We describe behavioural research using examples from a few well-studied insect species to illustrate how visual cues are used in navigation and how they interact with non-visual cues and strategies.

Abstract:Decades of research into intelligent animal navigation posits that organisms build and maintain internal spatial representations (or maps) 1 of their environment, that enables the organism to determine and follow task-appropriate paths (Epstein, Patai, Julian, & Spiers, 2017; O'keefe & Nadel, 1978; Tollman, 1948). Hamsters, wolves, chimpanzees, and bats leverage prior exploration to determine and follow shortcuts they may never have taken before (Chapuis & Scardigli, 1993; Harten, Katz, Goldshtein, Handel, & Yovel, 2020; Menzel, 1973; Peters, 1976; Toledo et al., 2020). Even blind mole rats and animals rendered situationally-blind in dark environments demonstrate shortcut behaviors (Avni, Tzvaigrach, & Eilam, 2008; Kimchi, Etienne, & Terkel, 2004; Maaswinkel & Whishaw, 1999). Ants forage for food along meandering paths but take near-optimal return trips (Müller & Wehner, 1988), though there is some controversy about whether insects like ants and bees are capable of forming maps (Cheung et al., 2014; Cruse & Wehner, 2011).
Vervet monkeys use paths consistent with context-specific spatial movement heuristics

Abstract:Animal foraging routes are analogous to the computationally demanding "traveling salesman problem" (TSP), where individuals must find the shortest path among several locations before returning to the start. Humans approximate solutions to TSPs using simple heuristics or "rules of thumb," but our knowledge of how other animals solve multidestination routing problems is incomplete. Most nonhuman primate species have shown limited ability to route plan. However, captive vervets were shown to solve a TSP for six sites. These results were consistent with either planning three steps ahead or a risk-avoidance strategy. I investigated how wild vervet monkeys (Chlorocebus pygerythrus) solved a path problem with six, equally rewarding food sites; where site arrangement allowed assessment of whether vervets found the shortest route and/or used paths consistent with one of three simple heuristics to navigate. Single vervets took the shortest possible path in fewer than half of the trials, usually in ways consistent with the most efficient heuristic (the convex hull). When in competition, vervets' paths were consistent with different, more efficient heuristics dependent on their dominance rank (a cluster strategy for dominants and the nearest neighbor rule for subordinates). These results suggest that, like humans, vervets may solve multidestination routing problems by applying simple, adaptive, context-specific "rules of thumb." The heuristics that were consistent with vervet paths in this study are the same as some of those asserted to be used by humans. These spatial movement strategies may have common evolutionary roots and be part of a universal mental navigational toolkit. Alternatively, they may have emerged through convergent evolution as the optimal way to solve multidestination routing problems.
Collective search in ants: Movement determines footprints, and footprints influence movement

Abstract:Collectively searching animals might be expected to coordinate with their groupmates to cover ground more evenly or efficiently than uncoordinated groups. Communication can lead to coordination in many ways. Previous work in ants suggests that chemical 'footprints', left behind by individuals as they walk, might serve this function by modulating the movement patterns of following ants. Here, we test this hypothesis by considering the two predictions that, first, ants may turn away from sites with higher footprint concentrations (klinotaxis), or, second, that they may change their turning patterns depending on the presence of footprints (klinokinesis). We tracked 5 whole colonies of Temnothorax rugatulus ants in a large arena over 5h. We approximated the footprint concentration by summing ant visitations for each point in the arena and calculated the speed and local path straightness for each point of the ant trajectories. We counterintuitively find that ants walk slightly faster and straighter in areas with fewer footprints. This is partially explained by the effect that ants who start out from the nest walking straighter move on average further away from the nest, where there are naturally fewer footprints, leading to an apparent relationship between footprint density and straightness However, ants walk slightly faster and straighter off footprints even when controlling for this effect. We tested for klinotaxis by calculating the footprint concentrations perceived by the left and right antennae of ants and found no evidence for a turning-away (nor turning-towards) behavior. Instead, we found noticeable effects of environmental idiosyncrasies on the behavior of ants which are likely to overpower any reactions to pheromones. Our results indicate that search density around an ant colony is affected by several independent processes, including individual differences in movement pattern, local spatial heterogeneities, and ants' reactions to chemical footprints. The multitude of effects illustrates that non-communicative coordination, individual biases and interactions with the environment might have a greater impact on group search efficiency and exploratory movements than pheromone communication.

Abstract:Navigating animals must integrate a diverse array of sensory cues into a single locomotor decision. Insects perform intricate navigational feats using a brain region termed the central complex in which an animal's heading direction is transformed through several layers of circuitry to elicit goal-directed locomotion. These transformations occur mostly in the fan-shaped body (FB), a major locus of multi-sensory integration in the central complex. Key aspects of these sensorimotor computations have been extensively characterized by functional studies, leveraging the genetic tools available in the fruit fly. However, our understanding of how neuronal activity in the FB dictates locomotor behaviors during navigation remains enigmatic. Here, we manipulate the activity of two key neuronal populations that input into the FB—the PFN and PFN neurons—used to encode the direction of two complex navigational cues: wind plumes and optic flow, respectively. We find that flies presented with unidirectional optic flow steer along curved walking trajectories, but silencing PFN neurons abolishes this curvature. We next use optogenetic activation to introduce a fictive heading signal in the PFNs to establish the causal relationship between their activity and steering behavior. Our studies reveal that the central complex guides locomotion by summing the PFN-borne directional signals and shifting movement trajectories left or right accordingly. Based on these results, we propose a model of central complex-mediated locomotion wherein the fly achieves fine-grained control of sensory-guided steering by continuously integrating its heading and goal directions over time.

Transient impairment of renal function is a common complication of contrast studies, particularly in patients with surgical disease such as carcinoma of the kidney. When radical nephrectomy follows arteriographic studies by a day or two, renal failure may become manifest in the postoperative interval, complicating convalescence from major surgical intervention. Many believed that the degree of renal failure observed after arteriography was related to the total volume of contrast material employed in a given study. The authors did not observe such a correlation, but they do point out that multiple injections of contrast material apparently, irrespective of volume, will be more likely to predispose impairment of renal function. Abstract:Many ant species are able to respond to dramatic changes in local conditions by relocating the entire colony to a new location. While we know that careful learning walks enables the homing behavior of foraging ants to their original nest, we do not know whether additional learning is required to navigate to the new nest location. To answer this question, we investigated the nest relocation behavior of a colony of Australian desert ants Melophorus bagoti that relocated their nest in response to heavy rainfall in the semi-desert terrain of Alice Springs. We identified five types of behavior: exploration between nests (Old-to-New nest and New-to-Old nest), transport from Old to New nest, and re-learning walks at Old and New nests. Initially, the workers performed relearning walks at the Old nest and exploratory walks between the Old and New nests. Once they completed the exploratory walks, the workers transported resources and brood to the new nest. Finally, we observed the workers performing relearning walks at the New nest. While the relearning walks at the Old nest were slow and appear to enable exploratory walks to the New nest, the relearning walks at the new nest were faster and appeared to enable homing from foraging trips. These observations shed insight on how learning helps these ants to respond to sudden changes in their environment.

Abstract:Cockroaches are scavengers that forage through dark, maze-like environments. Like other foraging animals, for instance rats, they must continually asses their situation to keep track of targets and negotiate barriers. While navigating a complex environment, all animals need to integrate sensory information in order to produce appropriate motor commands. The integrated sensory cues can be used to provide the animal with an environmental and contextual reference frame for the behavior. To successfully reach a goal location, navigational cues continuously derived from sensory inputs have to be utilized in the spatial guidance of motor commands. The sensory processes, contextual and spatial mechanisms, and motor outputs contributing to navigation have been heavily studied in rats. In contrast, many insect studies focused on the sensory and/or motor components of navigation, and our knowledge of the abstract representation of environmental context and spatial information in the insect brain is relatively limited. Recent reports from several laboratories have explored the role of the central complex (CX), a sensorimotor region of the insect brain, in navigational processes by recording the activity of CX neurons in freely-moving insects and in more constrained, experimenter-controlled situations. The results of these studies indicate that the CX participates in processing the temporal and spatial components of sensory cues, and utilizes these cues in creating an internal representation of orientation and context, while also directing motor control. Although these studies led to a better understanding of the CX's role in insect navigation, there are still major voids in the literature regarding the underlying mechanisms and brain regions involved in spatial navigation. The main goal of this review is to place the above listed findings in the wider context of animal navigation by providing an overview of the neural mechanisms of navigation in rats and summarizing and comparing our current knowledge on the CX's role in insect navigation to these processes. By doing so, we aimed to highlight some of the missing puzzle pieces in insect navigation and provide a different perspective for future directions.
