Abstract:In this study, a new cost-effective and environmentally sustainable microalgal production strategy was developed on the basis of the microalgae-crop symbiosis association. This method allowed the simultaneous production of microalgae and crops by using a simple eco-hydroponic system (EHS), with the input only for crop production. In glasshouse experiments, green algae Chlorella infusionum was successfully cultivated in a hydroponic culture system cultivating tomato. The algal and crop biomass productivities in the EHS were 32 +/- 5 g m(-3) d(-1) and 54.24 +/- 1.81 g dm(-3) d(-1) (n = 6 for each), respectively, which was significantly (P < 0.05) greater than the algal biomass productivity (16 +/- 5 g m(-3) d(-1), n = 6) in the algal monoculture and the crop biomass productivity (33.97 +/- 7.58 g dm(-3) d(-1), n = 6) in the crop monoculture without aeration. The enhancement of the biomass productions was mainly attributed to the aeration from algal photosynthesis and the CO2 fertilization from crop root respiration in the EHS. Particularly, the nitrogen and phosphorus utilization efficiencies were high due to co-utilization of the nutrients by microalgae and crop in the EHS. Optimization of Chlorella Culture Conditions with Response Surface Methodology to Increase Biomass

Abstract:Microalgae is gaining popularity as a major ingredient in nutrition supplements. To mass cultivate, it is imperative to improve the biomass yield hence optimization of cultures conditions becomes paramount. In this work, an attempt has been made to optimize the microalgal production using response surface methodology (RSM) and validate further the optimized parameters. The optimum conditions for the cultivation of Chlorella sp. KPU016 under optimized nutrient conditions were pH 8.2, the light intensity of 3100 lx, glycerol 1.44 g.L-1 (under pre-set conditions of 12 h lighting, the temperature at 27±1°C. With these RSM-driven optimum conditions, the yield of microalgal biomass achieved was 282.50 mg.L-1. For larger-scale microalgal harvesting, the validated optimal conditions can be inferred as the best for enhanced microalgal production. The isolate was partially sequenced and submitted to the NCBI database and the GenBank accession number is MZ348364.
Co-cultivation of microalgae and bacteria for optimal bioenergy feedstock production in wastewater by using response surface methodology

Abstract:This work uses response surface methodology (RSM) to study the co-cultivation of symbiotic indigenous wastewater microalgae and bacteria under different conditions (inoculum ratio of bacteria to microalgae, CO2, light intensity, and harvest time) for optimal bioenergy feedstock production. The findings of this study demonstrate that the symbiotic microalgae-bacteria culture not only increases total microalgal biomass and lipid productivity, but also enlarges microalgal cell size and stimulates lipid accumulation. Meanwhile, inoculum ratio of bacteria to microalgae, light intensity, CO2, and harvest time significantly affect biomass and lipid productivity. CO2 concentration and harvest time have significant interactive effect on lipid productivity. The response of microalgal biomass and lipid productivity varies significantly from 2.1 × 105 to 1.9 × 107 cells/mL and 2.8 × 102 to 3.7 × 1012 Total Fluorescent Units/mL respectively. Conditions for optimum biomass and oil accumulation are 100% of inoculation ratio (bacteria/microalgae), 3.6% of CO2 (v/v), 205.8 µmol/m2/s of light intensity, and 10.6 days of harvest time. This work provides a systematic methodology with RSM to explore the benefits of symbiotic microalgae-bacteria culture, and to optimize various cultivation parameters within complex wastewater environments for practical applications of integrated wastewater-microalgae systems for cost-efficient bioenergy production.
Biomass and lipid production by the native green microalgae Chlorella sorokiniana in response to nutrients, light intensity, and carbon dioxide: experimental and modeling approach

Abstract:Introduction: Microalgae are photosynthetic cells that can produce third-generation biofuels and other commercial compounds. Microalgal growth is influenced by two main parameters: light intensity and carbon dioxide concentration, which represent the energy and carbon source, respectively. For photosynthesis, the optimum values of abiotic factors vary among species. Methods: In this study, the microalga Chlorella sorokiniana was isolated from a freshwater lake. It was identified using molecular analysis of the ribosomal internal transcribed spacer. A single-factor design of experiments in 250-mL Erlenmeyer flasks was used to evaluate which concentrations of nitrogen and phosphorus increase the production of biomass and lipids. The response surface methodology was used with a 3 2 -factorial design (light intensity and CO 2 were used to evaluate its effect on biomass, lipid production, and specific growth rates, in 200-mL tubular photobioreactors (PBRs)). Results and Discussion: Low levels of light lead to lipid accumulation, while higher levels of light lead to the synthesis of cell biomass. The highest biomass and lipid production were 0.705 ± 0.04 g/L and 55.1% ± 4.1%, respectively. A mathematical model was proposed in order to describe the main phenomena occurring in the culture, such as oxygen and CO 2 mass transfer and the effect of light and nutrients on the growth of microalgae. The main novelties of this work were molecular identification of the strain, optimization of culture conditions for the indigenous microalgae species that were isolated, and formulation of a model that describes the behavior of the culture.

Utilizing genome-scale models to optimize nutrient supply for sustained algal growth and lipid productivity

Abstract:Nutrient availability is critical for growth of algae and other microbes used for generating valuable biochemical products. Determining the optimal levels of nutrient supplies to cultures can eliminate feeding of excess nutrients, lowering production costs and reducing nutrient pollution into the environment. With the advent of omics and bioinformatics methods, it is now possible to construct genome-scale models that accurately describe the metabolism of microorganisms. In this study, a genome-scale model of the green alga Chlorella vulgaris (iCZ946) was applied to predict feeding of multiple nutrients, including nitrate and glucose, under both autotrophic and heterotrophic conditions. The objective function was changed from optimizing growth to instead minimizing nitrate and glucose uptake rates, enabling predictions of feed rates for these nutrients. The metabolic model control (MMC) algorithm was validated for autotrophic growth, saving 18% nitrate while sustaining algal growth. Additionally, we obtained similar growth profiles by simultaneously controlling glucose and nitrate supplies under heterotrophic conditions for both high and low levels of glucose and nitrate. Finally, the nitrate supply was controlled in order to retain protein and chlorophyll synthesis, albeit at a lower rate, under nitrogen-limiting conditions. This model-driven cultivation strategy doubled the total volumetric yield of biomass, increased fatty acid methyl ester (FAME) yield by 61%, and enhanced lutein yield nearly 3 fold compared to nitrogen starvation. This study introduces a control methodology that integrates omics data and genome-scale models in order to optimize nutrient supplies based on the metabolic state of algal cells in different nutrient environments. This approach could transform bioprocessing control into a systems biology-based paradigm suitable for a wide range of species in order to limit nutrient inputs, reduce processing costs, and optimize biomanufacturing for the next generation of desirable biotechnology products.

Abstract:This study evaluated the biofuel production potential of two algal species, Chlorella pyrenoidosa and Scenedesmus abundans , under stress conditions induced by nutrient supplementation or starvation at varying light intensities. Central composite face-centered design response surface methodology (CCFD-RSM) was employed to optimize stress conditions by varying the sodium nitrate (NaNO 3 ), potassium dihydrogen phosphate (KH 2 PO 4 ), dipotassium hydrogen phosphate (K 2 HPO 4 ), cultivation time, and light intensity. The study included both C. pyrenoidosa and S. abundans , which presented increased biomass yields when subjected to nutrient starvation. Under the optimized conditions, the dry biomass yield was 98.26 mg/L for C. pyrenoidosa and 110 mg/L for S. abundans . Lipid yields were approximately 22.47% for C. pyrenoidosa and 29.06% for S. abundans under these optimized growth conditions. The optimized parameters for maximum biomass and lipid production were identified as C. pyrenoidosa , and the optimized conditions required 0.805 g/L NaNO 3 , 0.052 g/L K 2 HPO 4 , 0.099 g/L KH 2 PO 4 , 17 days of culture, and 5168.39 lx of light intensity. For S. abundans , the optimal conditions were 1.065 g/L NaNO 3 , 0.071 g/L K 2 HPO 4 , 0.058 g/L KH 2 PO 4 , 22 days of cultivation, and 2897 lx of light intensity. Overall, both C. pyrenoidosa and S. abundans have emerged as promising candidates for sustainable biodiesel production, highlighting their potential under stress conditions induced by nutrient modulation and variable light intensities.

Abstract:Microalgae are vital sources of high-value compounds, like polyphenols. Boosting polyphenol production requires considering diverse factors and their impacts on synthesis. This study aims to optimise polyphenol yield in microalga Parachlorella kessleri using an artificial neural network coupled with a genetic algorithm (ANN-GA). P. kessleri was grown under mixotrophic conditions and subjected to 13 experimental treatments. Factors studied included glucose, NaNO3, KH2PO4, LED light wavelength, intensity, and photoperiod. Total polyphenol concentration was quantified with Folin-Ciocalteu reagent; the polyphenol profile was obtained via HPLC. A database was constructed for ANN training to predict polyphenol production. Developed models were compared using indicators: coefficient of determination (R2pred), Mean Absolute Error (MAE), and Mean Square Error (MSE). Optimal models were fine-tuned with a genetic algorithm and validated using randomised experiments. The optimal ANN architecture for P. kessleri polyphenol production is: 6 inputs, 4 hidden layers, 6 neurons per layer, one output. This design achieved high prediction accuracy (R2 = 0.93). The genetically algorithm-optimised ANN model has these conditions: 26.60 g/L glucose; 1.42 g/L NaNO3; 0.59 g/L KH2PO4; white light; 1000 lx light intensity; 12:12 photoperiod. In experimental validation, 28 mg gallic acid equivalents per gram dry weight (GAE/g DW) were obtained, surpassing all models. An ANN-GA optimisation procedure was used for the first time for polyphenol production in P. kessleri. The model under mixotrophic conditions showed higher polyphenol production than autotrophic conditions. Polyphenol composition in P. kessleri varied between autotrophic and mixotrophic cultures, indicating inducible polyphenol production. The ANN-GA approach for polyphenol production in P. kessleri proved effective, with potential for maximising other high-value microalgae metabolites.

Abstract:Glycerol is a carbon source that produces good biomass under mixotrophic conditions. Enhancing the composition of culture media in algae biomass production improves growth rates, biomass yield, nutrient utilization efficiency, and overall cost-effectiveness. Among the key nutrients in the medium, nitrogen plays a pivotal role. Urea can be effectively used as a nitrogen source and is considered a low-cost form of nitrogen compared to other sources. Urea metabolism releases some CO2 in photosynthesis, and magnesium plays a major role in urea uptake. Magnesium is another key nutrient that is key in photosynthesis and other metabolic reactions. To maximize glycerol consumption in the mixotrophic system and to obtain high biomass and lipid productions, the variations in MgSO4·7H2O and urea concentrations were evaluated in the growth medium of the microalgae. A response surface methodology (RSM) using a central composite design (CCD) was designed to maximize glycerol consumption at the initial cellular growth rates (up to four days). The magnesium and urea supply varied from 0.3 to 1.7 g L−1. Response surface methodology was utilized to analyze the results, and the highest glycerol consumption rate, 770.2 mg L−1 d−1, was observed when C. vulgaris was grown at 1.7 g L−1 urea, 1.0 g L−1 MgSO4·7H2O. Using the optimal urea and magnesium concentrations with acetate, glucose, and glycerol as carbon sources, the same lipid content (10% average) was achieved on day 4 of mixotrophic C. vulgaris culture. Overall, the results show that mixotrophic growth of C. vulgaris using urea with an optimum magnesium concentration yields large amounts of fatty acids and that the carbon source greatly influences the profile of the fatty acids.

DEVELOPMENT OF COST EFFECTIVE AND ECONOMIC METHOD FOR CULTIVATION OF CHLORELLA SINGULARIS USING WASTE CARBON AND NITROGEN SOURCE

Abstract:Single cell organism based oil from oleaginous microorganisms has involved the attention because of its significance in the production of biodiesel as well as reducing pollution and waste. This possibility of algal biomass for utilizing waste carbon and nitrogen content was tested in this study. In this work, Chlorella singularis was cultivated in medium containing waste carbon, nitrogen as a nutrient source and BBM medium as control. The growth kinetics of the algae on these mediums was observed. The microalgae, Chlorella singularis can utilize the nitrogen content present in biogas digester wastewater as a substrate for its growth. Different low cost and easily available carbon and nitrogen based feedstock such as Potato peel, and Pea peel was used in this study. The growth of microalgae was found to follow the Monod growth model satisfactorily. Under the optimum photobioreactor condition, a maximum biomass of 3.75 gm/l in waste carbon and nitrogen medium and 1.5 gm/l in BBM was obtained in fifteen days of batch process. Based on Mond growth kinetics, net specific growth rate and doubling time of Chlorella singularis was found to be 0.054 day-1 and 12.83 hr-1. Monod kinetics, closed photo-biotector cultivation.

Application of ANN-MOGA for nutrient sequestration for wastewater remediation and production of polyunsaturated fatty acid (PUFA) by Chlorella sorokiniana MSP1

Abstract:Chlorella bears excellent potential in removing nutrients from industrial wastewater and lipid production enriched with polyunsaturated fatty acids. However, due to the changing nutrient dynamics of wastewater, growth and metabolic activity of Chlorella are affected. In order to sustain microalgal growth in wastewater with concomitant production of PUFA rich lipids, RSM (Response Surface Methodology) followed by heuristic hybrid computation model ANN-MOGA (Artificial Neural Network- Multi-Objective Genetic Algorithm) were implemented. Preliminary experiments conducted taking one factor at a time and design matrix of RSM with process variables viz. Sodium chloride (1 mM-40 mM), Magnesium sulphate (100 mg-800 mg) and incubation time (4th day to 20th day) were validated by ANN-MOGA. The study reported improved biomass and lipid yield by 54.25% and 12.76%, along with total nitrogen and phosphorus removal by 21.92% and 18.72% respectively using ANN-MOGA. It was evident from FAME results that there was a significantly improved concentration of linoleic acid (19.1%) and γ-linolenic acid (21.1%). Improved PUFA content makes it a potential feedstock with application in cosmeceutical, pharmaceutical and nutraceutical industry. The study further proves that C. sorokiniana MSP1 mediated industrial wastewater treatment with PUFA production is an effective way in providing environmental benefits along with value addition. Moreover, ANN-MOGA is a relevant tool that could control microalgal growth in wastewater.

Abstract:Chlamydomonas reinhardtii has emerged as a powerful green cell factory for metabolic engineering of sustainable products created from the photosynthetic lifestyle of this microalga. Advances in nuclear genome modification and transgene expression are allowing robust engineering strategies to be demonstrated in this host. However, commonly used lab strains are not equipped with features to enable their broader implementation in non-sterile conditions and high-cell density concepts. Here, we used combinatorial chloroplast and nuclear genome engineering to augment the metabolism of the C. reinhardtii strain UVM4 with publicly available genetic tools to enable the use of inorganic phosphite and nitrate as sole sources of phosphorous and nitrogen, respectively. We present recipes to create phosphite-buffered media solutions that enable high cell density algal cultivation. We then combined previously reported engineering strategies to produce the heterologous sesquiterpenoid patchoulol to high titers from our engineered green cell factories and show these products are possible to produce in non-sterile conditions. Our work presents a straightforward means to generate C. reinhardtii strains for broader application in bio-processes for the sustainable generation of products from green microalgae.

Abstract:Simultaneous enhancement in the productivities of microalgal biomass and lipids that are inversely correlated to each other has been a long-standing challenge before the scientists engaged in algal biodiesel research and innovation. This study, develops an Artificial Neural Network (ANN) model and uses multi-objective optimization approach to determine the process parameters, namely, light intensity, CO 2 %, air flow rate, and C/N ratio to maximize biomass and lipid productivities of microalga, Chlorella sorokiniana, simultaneously. Experimental data based on central composite design (CCD) were used to train a feed-forward multilayer ANN with four critical parameters. The global sensitivity analysis was performed on the trained ANN model using Sobol's method to assess the relative significance of the four process variables. Since the objectives of maximizing biomass and lipid productivities conflict with each other, multi-objective optimization approach was used for deriving the optimal process parameters, experimental validation of which resulted in approximately 3 times increase in biomass productivity and 7 times increase in lipid productivity. On transesterification of the lipid, the biodiesel product with saturated to unsaturated fatty acids ratio of 48:52 conformed very well to the international standards, ASTM D6751 and EN14214, thereby making it an environment friendly green biofuel. Thus, the present study showcases the successful application of machine learning tool in analysing the global sensitivity analysis of process variables, and employ it for multi-objective optimization towards achieving maximum microalgal biomass and lipid productivities concomitantly for potentially sustainable production of biodiesel.

Study on Dynamic Model Construction and Analysis Method of Bacteria-algae Co-culture

Abstract:To investigate the dynamic changes in growth and product synthesis of a mixed culture of Rhodotorula glutinis and Chlorella vulgaris, this study utilized fermentation data to analyze the rules and preliminary results of the microbial-algal symbiotic system. Employing Matlab, the author fitted a mathematical model proposed by predecessors, seeking parameters to establish a dynamic model for microbial fermentation. This model not only characterized parameter significance but also shed light on interactions, energy transfer mechanisms, and natural regulation balance during symbiotic fermentation. Results revealed that, under the influence of algae's photosynthetic carbon fixation, bacteria exhibited secondary growth, coupled with lipid synthesis and cell growth. The mixed culture model demonstrated a significant increase of 25.9% in biomass and 42.1% in oil content (reaching 30.694 g/L and 14.54 g/L, respectively). These findings suggest the model's potential as a theoretical guide for optimizing mixed culture fermentation processes.
Microfluidic Chemostatic Bioreactor for High-Throughput Screening and Sustainable Co-Harvesting of Biomass and Biodiesel in Microalgae

Abstract:As a renewable and sustainable source for energy, environment, and biomedical applications, microalgae and microalgal biodiesel have attracted great attention. However, their applications are confined due to the cost-efficiency of microalgal mass production. One-step strategy and continuous culturing systems could be solutions. However, current studies for optimization throughout microalgae-based biofuel production pipelines are generally derived from the batch culture process. Better tools are needed to study algal growth kinetics in continuous systems. A microfluidic chemostatic bioreactor was presented here, providing low-bioadhesive cultivations for algae in a cooperative environment of gas, nutrition, and temperature (GNT) involved with high throughput. The chip was used to mimic the continuous culture environment of bioreactors. It allowed simultaneously studying of 8 x 8 different chemostatic conditions on algal growth and oil production in parallel on a 7 x 7 cm2 footprint. On-chip experiments of batch and continuous cultures of Chlorella. sp. were performed to study growth and lipid accumulation under different nitrogen concentrations. The results demonstrated that microalgal cultures can be regulated to grow and accumulate lipids concurrently, thus enhancing lipid productivity in one step. The developed on-chip culturing condition screening, which was more suitable for continuous bioreactor, was achieved at a half shorter time, 64-times higher throughput, and less reagent con-sumption. It could be used to establish chemostat cultures in continuous bioreactors which can dramatically accelerate the development of renewable and sustainable algal for CO2 fixation and biosynthesis and related systems for advanced sustainable energy, food, pharmacy, and agriculture with enormous social and ecological benefits.
Optimal growth conditions to enhance Chlorella vulgaris biomass production in indoor phyto tank and quality assessment of feed and culture stock

Abstract:Commercial microalgae cultivation is a dynamic field with ongoing efforts to improve efficiency, reduce costs, and explore new applications. We conducted a study to examine how different light exposure periods affect Chlorella vulgaris's growth. We employed a Phyto tank batch system of approximately 3.5 L with LED light control, controlled airflow, and sterilized bags, maintained at 22.0 ± 2.0 °C indoors. Various methods, including spectrophotometry, and cell counter were employed to monitor Chlorella vulgaris growth under different light exposure cycles. Additionally, quality analysis as feed source was employed by proximate, amino acid, beta-glucan, and microbial content analysis. The results revealed significant variations in C. vulgaris biomass production based on light exposure duration. Notably, the 16:8-h light-dark photoperiod exhibited the highest biomass concentration, reaching 6.48 × 107 ± 0.50 cells/mL with an optical density (OD) of 1.165 absorbance at 682 nm. The 12:12-h light-dark photoperiod produced the second-highest biomass concentration, with 2.305 × 106 ± 0.60 cells/mL at an OD of 0.489. Proximate analysis of dry algae powder revealed low lipid content (0.48 %), high protein content (37.61 %), variable ash concentration (average 10.75 %), and a significant carbohydrate fraction (51.16 %) during extended daylight and shorter dark periods. Amino acid analysis identified nine essential amino acids, with glutamic acid being the most abundant (17.7 %) and methionine the least (0.4 %). Furthermore, quality analysis and microbiological assays demonstrated that the C. vulgaris biomass is well-suited for fish and livestock use as a feed source and possibility as human nutraceuticals. These findings can be considered more environmentally friendly and ethically sound due to the absence of genetic modification.

Abstract:The wide metabolic diversity of microalgae, their fast growth rates, and low-cost production make these organisms highly promising resources for a variety of biotechnological applications, addressing critical needs in industry, agriculture, and medicine. The use of microalgae in consortia with bacteria is proving valuable in several areas of biotechnology, including the treatment of various types of wastewater, the production of biofertilizers, and the extraction of various products from their biomass. The monoculture of the microalga Chlamydomonas has been a prominent research model for many years and has been extensively used in the study of photosynthesis, sulphur and phosphorus metabolism, nitrogen metabolism, respiration, and flagellar synthesis, among others. Recent research has increasingly recognised the potential of Chlamydomonas-bacteria consortia as a biotechnological tool for various applications. The detoxification of wastewater using Chlamydomonas and its bacterial consortia offers significant potential for sustainable reduction of contaminants, while facilitating resource recovery and the valorisation of microalgal biomass. The use of Chlamydomonas and its bacterial consortia as biofertilizers can offer several benefits, such as increasing crop yields, protecting crops, maintaining soil fertility and stability, contributing to CO2 mitigation, and contributing to sustainable agricultural practises. Chlamydomonas-bacterial consortia play an important role in the production of high-value products, particularly in the production of biofuels and the enhancement of H2 production. This review aims to provide a comprehensive understanding of the potential of Chlamydomonas monoculture and its bacterial consortia to identify current applications and to propose new research and development directions to maximise their potential.
Medium Optimization for the Production of Anti-Cyanobacterial Substances by Streptomyces Sp. HJC-D1 Using Response Surface Methodology

Abstract:Bioremediation using isolated anti-cyanobacterial microorganism has been widely applied in harmful algal blooms (HABs) control. In order to improve the secretion of activated anti-cyanobacterial substances, and lower the cost, a sequential optimization of the culture medium based on statistical design was employed for enhancing the anti-cyanobacterial substances production and chlorophyll a (Chl a) removal by Streptomyces sp. HJC-D1 in the paper. Sucrose and KNO3 were selected as the most suitable carbon and nitrogen sources based on the one-at-a-time strategy method, and sucrose, KNO3 and initial pH were found as major factors that affected the anti-cyanobacterial ability of the isolated stain via the Plackett–Burman design. Based on the response surface and canonical analysis, the optimum condition of culture medium was obtained at 22.73 g l-1 of sucrose, 0.96 g l-1 of KNO3, and initial pH 8.82, and the Chl a removal efficiency by strain HJC-D1 increased from 63 ± 2 % to 78 ± 2 % on the optimum conditions.
Powerful Tools for Productivity Improvements in Microalgal Production

Abstract:Technology for microalgae cultivation has potential to maximize biomass generation to production process and economic viability of microalgal production. Powerful tools that increase the production and value of microalgae are therefore of great significance. In this review, valid approaches involved in strain screening, microalgal cultivation, cell harvesting and disruption are discussed with regards to productivity improvements and energy efficiency. Particularly, current research in kinetic models, metabolic flux analysis and genetic tools are considered. Kinetic models of nutrient, light, temperature and cell quota to increase the biomass concentration and targeted products are discussed. Metabolic flux analysis, especially GC–MS-based 13C-metabolic flux analysis is discussed to describe how the central carbon flux to targeted products can be precisely and rapidly engineered. This approach is exemplified by Chromochloris zofingiensis under batch and fed-batch cultures. Molecular approaches including gene silencing methods and CRISPR–Cas9 are also discussed as these provide powerful tools for enhanced production of biomass and associated lipids and carotenoids. Finally, the huge potential of combined applications of the described tools are discussed. A better understanding of cell behavior in response to these tools will provide the means to improve microalgal production at a competitive cost.

Abstract:A comprehensive numerical model system for the bioreactor design, structure and cultivation condition optimization, scale-up, and carbon fixation performance evaluation was proposed. Three basic models of computational fluid dynamics, particle and mass transfer, and two improved models of algae growth and light accumulation were integrated innovatively. The algae growth model directly predicted the algal production performance of the bioreactor, and the error was less than 15% compared with the experimental results. The light accumulation model was proposed to simulate the spatial distribution of light utilization with the changes of algal concentration and light intensity. The model system was applied to scale up the flat plate photobioreactor with inclined baffles, and the results showed that the photobioreactor with 9 cm light path had a maximum ideal areal productivity (505.94 g/m2) and better performance. Based on the light accumulation model, the strategy of changing the surface light intensity during the cultivation was proposed to maintain the light utilization and increase the production. Under the optimized surface light intensity, the maximum algal concentration and the average carbon fixation rate of the bioreactor were 43.41 and 248.35% higher than those of the constant light intensity. The proposed model system is a reliable and efficient experimental alternative tool for algal production evaluation and carbon fixation application of the bioreactor.

Abstract:The high cost of oleaginous microalgae harvesting is a major factor limiting the industrialization of microalgae biodiesel. In order to obtain an efficient, environmentally friendly and cost-effective method for harvesting microalgae, this study used Citrobacter freundii (No. W4) and Mucor circinelloides for the harvest of oil-bearing microalgae Chlorella pyrenoidosa. The effects of flocculation conditions, including the initial pH, bacterial dosage, fungal dosage and glucose dosage were investigated, respectively. According to the results of response surface optimization, when the initial pH was 7.0, bacteria: microalgae ratio was 1.6:1, microalgae: fungi ratio was 333:1, the glucose concentration was 1.47 g/L, the flocculation efficiency of the actual sewage cultured C. pyrenoidosa attained 97.45%. The results showed that bacteria and fungi had a good potential for the harvest of microalgae.
