Abstract:Light quality optimization is an efficient method for improving the growth and quality of lettuce in plant factories. In this study, lettuce seedlings were illuminated under different light-emitting diode (LED) lights, namely, red-blue (RB), red-blue-green (RBG), red-blue-purple (RBP), and red-blue-far-red (RBF) LED lights, to investigate the effect of light quality on growth, quality, and nitrogen metabolism. The combination of 75% red and 25% blue light was set as the basic light source, and 20% of green, purple and far-red light were added to basic light source, respectively. All the treatments were set to 200 μmol m –2 s –1 . Results showed that the fresh weight and dry weight of aboveground lettuce under RBG, RBP, and RBF treatments were significantly lower than those under the RB treatment because of the decrease in the effective photon flux density for chlorophyll absorption. The vitamin C content of the lettuce leaves was increased by about 23% with the addition of purple light. For nitrate reduction, the addition of green light significantly increased the nitrite content of the lettuce leaves. It also promoted the reduction from nitrite to ammonium through the activation of the nitrite reductase ( NiR ) expression and enzyme activity. The nitrate and ammonium content decreased with the addition of purple light because of the inhibited NR and NiR expression and enzyme activity. For nitrogen assimilation, individual (e.g., Asp, Glu, and Leu) and total amino acids were induced to increase by adding green, purple, and far-red light. The addition of light was hypothesized to have inhibited protein biosynthesis, thereby causing the accumulation of amino acids. Correlation analysis showed that the relative expression levels between HY5 and NR / NiR presented a significantly negative correlation. Transcription factor HY5 might mediate the regulation of light quality on nitrogen metabolism by inhibiting NR and NiR expressions. It might also exert a negative effect on nitrate reduction. Further studies via genome editing techniques on the identification of HY5 functions for nitrate assimilation will be valuable. Nevertheless, the results of this work enrich the understanding of the effect of light quality on nitrate metabolism at the level of gene expression and enzyme activity.

Comparison on the Difference of Growth and Nutritional Quality for Lettuce (lactuca Sativa L.) Between Aeroponic and Hydroponic Cultivation Systems under Different Nitrogen Levels

Abstract:Summary Soilless culture is considered to be an alternative to soil-based cultivation.It has been developed quickly because of rapid growth on biomass and high quality of crops.Hydroponics and aeroponics are both soilless culture,but with different techniques.Aeroponics is a newly soilless culture technique,in which the roots are suspended mid-air inside a chamber and are intermittently sprayed with a nutrient solution to supply the plants with mineral and water.Unlike hydroponics using water as a growing medium and relying on essential mineral to sustain plant growth,aeroponics is conducted without a growing medium.Thus,there are significant differences of root zone between aeroponic and hydroponic cultivation systems.However,little effort has been found in the literature evaluating the effect of the two soilless techniques on yield and quality of growing vegetable. In this study,experiments were carried out to compare the biomass production and nutritional quality of lettuce between hydroponic and aeroponic cultivation systems under different nitrogen levels(2,8 and 20 mmol/L NO3 --N). Results showed that under the same nitrogen level,higher biomass,root growth,relative growth rate and photosynthetic rate were observed for lettuces grown in aeroponics than in hydroponics.Nitrate and glutathione contents were lower in aeroponics,while the ferric reducing/antioxidant power(FRAP)value and 1,1-diphenyl-2-picrydrazyl(DPPH)radical scavenging rate were comparable to those in hydroponics.There were no differences in contents of soluble protein,soluble sugar and ascorbic acid,but the accumulations of soluble protein,soluble sugar,ascorbic acid and glutathione in aeroponics were markedly higher than those in hydroponics.Furthermore, the contents of nitrate and soluble protein of lettuces increased both in hydroponics and aeroponics,while the concentrations of soluble sugar,ascorbic acid and glutathione,FRAP value and DPPH radical scavenging rate decreased with increasing nitrogen level.Highest biomass,root morphological index,photosynthetic index, accumulations of soluble protein,soluble sugar,ascorbic acid and glutathione were observed under the nitrogen level of 8 mmol/L. In sum,aeroponics is beneficial for higher biomass,antioxidant accumulation and antioxidative activity, especially under the nitrogen level of 8 mmol/L.
Effects of different LED sources on the growth and nitrogen metabolism of lettuce

Abstract:Using a light-emitting diode (LED) as the light source, the effects of eight different light treatments [white light (control, W), purple light (P), blue light (B), red light (R), green light (G), yellow light (Y), red–blue light in a 9:1 ratio (9R/1B), and red–blue light in a 4:1 ratio (4R/1B)] on the growth, quality and nitrogen metabolism of lettuce were studied. The results showed that compared with the white light, the purple light, blue light, red light, and the red-blue light combination could all increase the biomass of the aboveground part of lettuce to various degrees, while green light and yellow light inhibited lettuce growth. Under blue light, the contents of soluble protein and flavonoid in lettuce were the highest; under red light, the soluble sugar content was the highest, while the contents of soluble protein, free amino acids, and vitamin C (VC) were relatively higher under the 4R/1B light condition. Compared with white light, the sources of purple, blue, and red lights as well as the red–blue light combination all significantly reduced nitrate accumulation in lettuce, and the activities of the nitrogen (N) metabolism-related enzymes such as nitrate reductase, nitrite reductase, glutamine synthetase, glutamate synthase, and glutamate dehydrogenase were increased to varying degrees. In contrast, the contents of nitrate and ammonium N were significantly accumulated in lettuce under green light, and the activities of relative enzymes were significantly reduced. Therefore, the purple light, blue light, and red–blue combination light sources could promote N assimilation and improve the aboveground biomass accumulation in lettuce by improving the activity of the N metabolism-related enzymes in lettuce. Particularly under the 4R/1B light source, the biomass, soluble protein, VC, and total amino acid content were rather high in lettuce, which indicated that the 4R/1B light source could better effectively improve the nutritional quality and promote the growth of lettuce, while yellow light and green light are not suitable to serve as direct sources in a plant factory. These results provide a certain theoretical basis for the regulation of the light environment in cultivation facilities.
Effects of nitrogen application rates on antioxidant contents and antioxidative activities in Chinese cabbage

Abstract:A pot experiment was carried out to study the effects of nitrogen application rates(0,150 and 300 mg N·kg-1 soil) on antioxidant contents and antioxidative activities in two cultivars(Shuzhouqing and Dongfei) of Chinese cabbage(Brassica chinensis L.).Results show that fresh and dry weights of shoots were markedly increased by increasing nitrogen application rate from 0 mg N·kg-1 to 150 mg N·kg-1,and were significantly decreased with further increasing nitrogen application rate.Nitrate contents in leaf and petiole increased with increasing N application rate.Higher nitrate contents were observed in cultivar of Shuzhouqing than that of Dongfei.Highest contents of ascorbic acid,total phenolics,flavonoid,and DPPH(1,1-diphenyl-2-picrylhydrazyl) radical scavenging rate and FRAP(ferric reducing antioxidant power) value were recorded in the control(0 mg N·kg-1).The contents of ascorbic acid,total phenolics and flavonoids and antioxidative activities were markedly decreased by the increases of nitrogen application rate,while GSH was not affected.However,the highest antioxidant accumulation were founded at the application rate of 150 mg N·kg-1 soil.Therefore,the rate of 150 mg N·kg-1 soil might be the optimal nitrogen application rate for production of both high-yield and high-quality vegetable under this experimental condition.

Abstract:Light intensity and nitrogen concentration in nutrient solutions are two main factors affecting the growth and nutritional quality of lettuce(Lactuca sativa L.). This study investigated the method of regulating vegetable cultivation through the interactive control of the light environment and solution ratio. A quadratic regression orthogonal model was developed to understand the role of light intensity (43 to 237 µmol m−2s−1) and nitrogen concentration (4.3 to 23.7 mmol L−1) in lettuce growth. Dry weight, vitamin C, and nitrate contents were measured and modelled to understand the coupling effect of light intensity and nitrogen concentration on lettuce. The optimised conditions for lettuce growth were 237 µmol m−2 s−1 of light intensity and 9.2 mmol L−1 of nitrogen concentration, and these conditions were predicted to produce lettuce with an average dry biomass of 3.9 g plant−1, 149.9 µg g−1 of Vitamin C, and 212.1 µg g−1 of nitrate content based on their respective weighting factors (0.50, 0.25, and 0.25). The lettuce yield and quality were verified using the modelled optimised growth conditions and the lettuce grown was indeed more nutritional compared to the commercially available lettuce. The findings are important for mass production of lettuce in plant factories and greenhouses.
Morphology, Photosynthetic Traits, and Nutritional Quality of Lettuce Plants as Affected by Green Light Substituting Proportion of Blue and Red Light

Abstract:Green light, as part of the photosynthetically active radiation, has been proven to have high photosynthetic efficiency once absorbed by plant leaves and can regulate plant physiological activities. However, few studies have investigated the appropriate and efficient way of using the green light for plant production. Thus, the objective of this study was to investigate a moderate amount of green light, partially replacing red and blue light, for plant growth and development. In this experiment, four treatments were set up by adjusting the relative amount of green light as 0 (RB), 30 (G30), 60 (G60), and 90 (G90) μmol m −2 s −1 , respectively, with a total photosynthetic photon flux density of 200 μmol m −2 s −1 and a fixed red-to-blue ratio of 4:1. Lettuce ( Lactuca sativa cv. ‘Tiberius’) plant growth and morphology, stomatal characteristics, light absorptance and transmittance, photosynthetic characteristics, and nutritional quality were investigated. The results showed that: (1) shoot dry weight increased by 16.3 and 24.5% and leaf area increased by 11.9 and 16.2% under G30 and G60, respectively, compared with those under RB. Plant stem length increased linearly with increasing green-to-blue light ratio; (2) light transmittance of lettuce leaf under treatments employing green light was higher than that under RB, especially in the green region; (3) stomatal density increased, whereas stomatal aperture area decreased with the increase in the relative amount of green light; and (4) carbohydrate accumulation increased under G60 and G90. Soluble sugar contents under G60 and G90 increased by 39.4 and 19.4%, respectively. Nitrate contents under G30, G60, and G90 decreased by 26.2, 40.3, and 43.4%, respectively. The above results indicated that 15–30% green light replacing red and blue light effectively increased the yield and nutritional quality of lettuce plants.

Effects of constant versus fluctuating red–blue LED radiation on yield and quality of hydroponic purple-leaf lettuce

Abstract:This study investigated the effects of constant light and alternating relatively high-intensity (500 µmol m−2 s−1) and low-intensity (150 µmol m−2 s−1) red–blue LEDs (4R:1B) on the biomass production and quality of hydroponic purple-leaf lettuce (Lactuca sativa L. cv. 'Zishan') in an environmentally controlled plant factory. Four treatments were set up to separate 1 h of high light into four different alternating frequencies in a 24-h light−dark cycle (16/8 h): one time (A1), three times (A3), six times (A6), and twelve times (A12). In addition, one constant light treatment with the same daily light integral (DLI, 9.8 mol m−2 per day) as other treatments was set as the control (CK, 170 µmol m−2 s−1). The results indicated that A6 significantly reduced shoot fresh weight and increased the root–shoot ratio of lettuce compared with CK, but there was no significant difference among other treatments. Alternating light treatments did not promote the accumulation of soluble sugar, soluble protein, and phenolic substances compared with CK. Meanwhile, A12 significantly promoted the accumulation of total ascorbate (TA) in lettuce leaves compared with other treatments but decreased ascorbate/TA ratio. Above all, under the same DLI condition, alternating high and low light did not have obvious positive effects on biomass production and the accumulation of nutrient substance in lettuce under constant light was better than that under alternating light. Therefore, compared with the fluctuating radiation with the same DLI, constant radiation is a better choice for lettuce production.
Short-Term Nitrate Limitation Prior to Harvest Improves Phenolic Compound Accumulation in Hydroponic-Cultivated Lettuce (lactuca Sativa L.) Without Reducing Shoot Fresh Weight

Abstract:Long-term exposure to a low nitrogen supply could effectively improve phenolic content in vegetables, but it also greatly decreases the yield. Therefore, it is necessary to develop cost-effective strategies of nitrogen management to overcome these obstacles. Here, we find that decreasing the nitrogen (NO3-) supply from 8.0 to below 2.0 mM during the last 7 days before harvest significantly increases phenolic levels and antioxidant capacity of the soluble fraction in two hydroponic-cultivated lettuces, which is demonstrated by the up-regulation of related genes ( PAL1, CHS, F3H, DFR, F35H, and UFGT) involved in the phenolic synthesis pathway. Importantly, short-term nitrate limitation before harvest does not affect yield production unless nitrogen supply is reduced below 1.0 and 0.5 mM for Ziluoma and Lvluo, respectively. These findings suggest that appropriate short-term nitrate limitation before harvest, considering genotype variation, is feasible for improving the phenolic content and antioxidant capacity of vegetables without reducing their yield.
Effects of Supplemental UV-A LEDs on the Nutritional Quality of Lettuce: Accumulation of Protein and Other Essential Nutrients

Abstract:Light plays an important role in influencing the nutritional quality of food crops, especially with regard to the health-promoting phytochemicals. However, its role in affecting the nutritional quality with regard to the essential nutrients is not well understood. In this study, the effects of preharvest UV-A treatment on the nutritional quality of lettuce (Lactuca sativa, cv. red-leaf ‘New Red Fire’ and green-leaf ‘Two Star’) in relation to the essential nutrients and health-promoting phytochemicals were explored. Lettuce plants were grown in a growth chamber and were subjected to supplemental UV-A LEDs (peak wavelength 375 nm) for a brief period (3–6 days) prior to harvest. UV-A LEDs were equipped with lenses to control the light dispersion. Many growth indices such as shoot fresh mass, leaf area, and leaf number were unaffected by supplemental UV-A in both varieties while shoot dry mass decreased in response to a 6-day UV-A treatment compared to the control. Leaf chlorophyll and carotenoid concentrations increased significantly in green-leaf lettuce after 3 or 6 days of UV-A treatment, but only after 3 days of UV-A treatment in red-leaf lettuce compared to the control. Leaf protein concentration increased significantly in both lettuce varieties along with a number of essential nutrients such as phosphorus, potassium, calcium, manganese, and sulfur in response to supplemental UV-A. Supplemental UV-A increased the accumulation of protein by approximately 48% in green-leaf lettuce and 31% in red-leaf lettuce compared to the control plants. Moreover, in addition to the above essential nutrients, green-leaf lettuce accumulated higher amounts of magnesium, copper, and zinc compared to the control plants, indicating that green-leaf lettuce was more responsive to preharvest supplemental UV-A treatment than red-leaf lettuce. However, the accumulation of total phenolic compounds and flavonoids in both varieties was lower under supplemental UV-A. Furthermore, the use of LED lenses did not have a consistent impact on most of the plant responses studied. Overall, the results indicate that a brief preharvest exposure of both red- and green-leaf lettuce varieties to UV-A increased their nutritional quality by enhancing the accumulation of protein and other major essential nutrients.

Abstract:Abstract Light and nutrient are important factors for vegetable production in plant factory or greenhouse. The total 12 treatments which contained the combination of four light intensity (150, 250, 350 and 450 μmol · m −2 · s −1 ) and three nutrient solution concentration (NSC) (1/4, 1/2, 3/4 strength NSC) were established for investigation of lettuce growth and quality in a growth chamber. The combination of light intensity and NSC exhibited significant effects on photosynthetic pigment, nutritional quality, mineral content and antioxidant capacity. That a higher light intensity were readily accessible to higher chlorophyll a/b showed in lettuce of treatment of 350 μmol · m −2 · s −1 × 3/4NSC and 450 μmol · m −2 · s −1 × 1/4NSC. Lower total N contents, higher content of soluble protein, vitamin C, soluble sugar and free amino acid exhibited in lettuce under treatment of 250 and 350 μmol · m −2 · s −1 × 1/4NSC or 3/4NSC. With increasing NSC and LED irradiance, the content of total P and K in lettuce increased and decreased, respectively. The highest and lowest total Ca content were found in treatment of 150 μmol · m −2 · s −1 × 1/4NSC and 450 μmol · m −2 · s −1 × 1/4NSC, respectively, and higher content of total Mg and Zn was observed under 250 μmol m −2 s −1 × 1/4NSC and 150 μmol · m −2 · s −1 × 3/4NSC, respectively. The antioxidant contents generally decreased with increasing NSC level. The higher antioxidant content and capacity occurred in lettuce of 350 μmol · m −2 · s −1 × 1/4NSC treatment. The interaction of 350 μmol · m −2 · s −1 × 1/4NSC might be the optimal condition for lettuce growth in plant factory.

Abstract:Plants grow better when they are supplied with a combination of ammonium (NH4+) and nitrate (NO3−) than when either one is supplied as the sole N (nitrogen) source. However, the effects of N forms on N metabolism and major N assimilation enzymes in different plants, especially vegetables, are largely neglected. This study was conducted on two plants with distinct NH4+ tolerances to compare the responses of two popular leafy vegetables, Korean cabbage (Brassica campestris L.) ‘Ssamchu’ and lettuce (Lactuca sativa L.) ‘Caesar green’, to the N source. To this end, plant growth and quality, photosynthesis, carbohydrate, N contents (in the forms of NO3−, NO2−, NH4+, total protein), and key N assimilation-related enzyme (NR, NIR, GS, GDH) activities were investigated. When plants were subjected to one of three NH4+:NO3− regimes, 0:100, 50:50, or 100:0, lettuce was relatively more tolerant while cabbage was extremely sensitive to high NH4+. Both plants benefited more from being grown with 50:50 NH4+:NO3−, as evidenced by the best growth performance, ameliorated photosynthesis, and enriched carbohydrate (C) stock content. In addition, as compared to cabbage, the GS and GDH activities were reinforced in lettuce in response to an increasing external NH4+ level, resulting in low NH4+ accumulation. Our findings suggested that boosting or maintaining high GS and GDH activities is an important strategy for the ammonium tolerance in vegetables.

Abstract:The nitrogen content of nutrient solution and light intensity are two crucial factors affecting the yield and quality of controlled environment hydroponic vegetables. In this study, Purple Bok Choy (Brassica rapa var. Chinensis) was evaluated in a two-factor experiment with five levels of light intensity and five levels of nitrogen concentration. The plants were harvested on the 35th day after planting and different parameters such as shoot fresh weight, root fresh weight, anthocyanin, and soluble sugar were measured as representatives of yield and quality. This paper investigated the effects of different light/nitrogen combinations on the yield and quality of Purple Bok Choy, and established an optimal model coupled with yield and quality indicators. The result reveals that the light intensity and nitrogen concentration have a strong synergistic interaction on shoot fresh weight, root fresh weight, anthocyanin, and soluble sugar of Purple Bok Choy. A combination of light/nitrogen could be found to achieve the common improvement for the indicators with a similar change trend. This work gives valuable insights into the combinational regulation of nitrogen concentration and light intensity and provides a new idea for the comprehensive improvement of production yield and quality.

Short-Term Alteration of Nitrogen Supply Prior to Harvest Affects Quality in Hydroponic-Cultivated Spinach (spinacia Oleracea)

Abstract:BACKGROUND:Quality-associated problems, such as excessive in planta accumulation of oxalate, often arise in soillessly cultivated spinach (Spinacia oleracea). Maintaining a higher level of ammonium (NH₄⁺) compared to nitrate (NO₃⁻) during the growth period can effectively decrease the oxalate content in hydroponically cultivated vegetables. However, long-term exposure to high concentrations of NH₄⁺ induces toxicity in plants, and thus decreases the biomass production. Short-term application of NH₄⁺ before harvesting in soilless cultivation may provide an alternative strategy to decrease oxalate accumulation in spinach, and minimise the yield reduction caused by NH₄⁺ toxicity.RESULT:The plants were pre-cultured in 8 mmol L⁻¹ NO₃⁻ nutrient solution. Next, 6 days before harvest, the plants were transferred to a nutrient solution containing 4 mmol L⁻¹ NO₃⁻ and 4 mmol L⁻¹ NH₄⁺. This new mix clearly reduced oxalate accumulation, increased levels of several antioxidant compounds, and enhanced antioxidant capacity in the edible parts of spinach plants, but it did not affect biomass production. However, when the 8 mmol L⁻¹ NO₃⁻ was shifted to either nitrogen-free, 4 mmol L⁻¹ NH₄⁺ or 8 mmol L⁻¹ NH₄⁺ treatments, although some of the quality indexes were improved, yields were significantly reduced.CONCLUSIONS:Short-term alteration of nitrogen supply prior to harvest significantly affects quality and biomass of spinach plants, and we strongly recommend to simultaneously use NO₃⁻ and NH₄⁺ in hydroponic cultivation, which improves vegetable quality without decreasing biomass production.
High-Proportion Blue Light Irradiation at the End-of-Production Stage Promotes the Biosynthesis and Recycling of Ascorbate in Lettuce

Abstract:Ascorbate (AsA), an essential antioxidant for both plants and the human body, plays a vital role in maintaining proper functionality. Light plays an important role in metabolism of AsA in horticultural plants. Our previous research has revealed that subjecting lettuce to high light irradiation (HLI) (500 μmol·m−2·s−1) at the end-of-production (EOP) stage effectively enhances AsA levels, while the optimal light quality for AsA accumulation is still unknown. In this study, four combinations of red (R) and blue (B) light spectra with the ratio of 1:1 (1R1B), 2:1 (2R1B), 3:1 (3R1B), and 4:1 (4R1B) were applied to investigate the biosynthesis and recycling of AsA in lettuce. The results demonstrated that the AsA/total-AsA content in lettuce leaves was notably augmented upon exposure to 1R1B and 2R1B. Interestingly, AsA levels across all treatments increased rapidly at the early stage (2–8 h) of irradiation, while they increased slowly at the late stage (8–16 h). The activity of L-galactono-1,4-lactone dehydrogenase was augmented under 1R1B treatment, which is pivotal to AsA production. Additionally, the activities of enzymes key to AsA cycling were enhanced by 1R1B and 2R1B treatments, including ascorbate peroxidase, dehydroascorbate reductase, and monodehydroascorbate reductase. Notably, hydrogen peroxide and malondialdehyde accumulation increased dramatically following 16 h of 1R1B and 2R1B treatments. In addition, although soluble sugar and starch contents were enhanced by EOP-HLI, this effect was comparatively subdued under the 1R1B treatment. Overall, these results indicated that AsA accumulation was improved by irradiation with a blue light proportion of over 50% in lettuce, aligning with the heightened activities of key enzymes responsible for AsA synthesis, as well as the accrual of hydrogen peroxide. The effective strategy holds the potential to enhance the nutritional quality of lettuce while bolstering its antioxidant defenses.

Light Quality Affected the Growth and Root Organic Carbon and Autotoxin Secretions of Hydroponic Lettuce

Abstract:Light is a crucial environmental signal and photosynthetic energy for plant growth, development, and primary and secondary metabolism. To explore the effects of light quality on the growth and root exudates of hydroponic lettuce (Lactuca sativa L.), white LED (W, control) and four the mixtures of red (R) and blue (B) LED with different R/B light intensity ratios (R/B = 2, 2R1B; R/B = 3, 3R1B; R/B = 4, 4R1B; and R/B = 8, 8R1B) were designed. The results showed that the biomass of lettuce under 8R1B and W treatments was higher than that under other light quality treatments. The photosynthetic rate (Pn) under red and blue light was significantly higher than that of white light. Total root length, root surface area, and root volume were the highest under 8R1B. 4R1B treatment significant increased root activity by 68.6% compared with W. In addition, total organic carbon (TOC) content, TOC content/shoot dry weight, TOC content/root dry weight, and TOC content/root surface area were the highest under 4R1B. Moreover, 8R1B treatment reduced the concentration of benzoic acid and salicylic acid, and the secretion ability of benzoic acid and salicylic acid by per unit root surface area and accumulation by per unit shoot dry weight. In addition, 2R1B and 3R1B reduced the secretion ability of gallic acid and tannic acid by per unit root surface area and accumulation by per unit shoot dry weight. In conclusion, this study showed that the secretion of autotoxins could be reduced through the mediation of red and blue light composition of LEDs in a plant factory. In terms of autotoxin secretion reduction efficiency and yield performance of lettuce, 8R1B light regime is recommended for practical use.
Effects of supplementing green light to red and blue light on the growth and yield of lettuce in plant factories

Abstract:Red and blue light-emitting diodes (LEDs) are widely used as light sources in plant factories. Plants grown under red and blue light only produces purplish-gray light environment where plant leaves look purplish-gray. Under that light condition, it makes difficulty to check health status of plants specially insect and disease infected leaves by human naked eye. But the leaf color turns green when inspected under full spectrum of light environment. In this regard, the use of green light to red and blue create a white light environment which is congenial for the grower. Likewise, CO2 assimilation of green light is usually lower compared to red and blue light because of its lower absorptance under low PPFD. But at higher PPFD (≥500 μmol m−2 s−1), green light shows higher CO2 assimilation rate compared to red and blue light through uniform distribution of full spectrum (red, green and blue) of light into the plant canopy and the lower leaves. In this study, optimal intensity of green light supplementation to the red and blue LEDs was evaluated for better growth, yield, and quality of lettuce. In experiment I and II, green light was added with red and blue light where total photosynthetic photon flux density (PPFD) was increased for the additional green light. In experiment III, 0, 10, 20, 30, 40, 50, 60, 70, 80 μmol m−2 s−1 green (G) light was supplemented to 235 μmol m−2 s−1 red (R) and 59 μmol m−2 s−1 blue (B) light maintaining 294 μmol m−2 s−1 total PPFD. Lettuce plants were cultivated hydroponically in three-step vertical grow beds using half-strength of Enshi nutrient solution. The temperature was maintained at 20 ± 2 °C at day and night and the day length was 16 h. In experiment I and II, the fresh mass of lettuce grown under the combination of R, B and G LEDs was found higher than those cultivated under R and B LEDs only. In experiment III, lettuce plants produced high fresh masses when 30 μmol m−2 s−1 of G light was supplemented with R and B lights maintaining the ratio 211:30:53 (R:G:B). By supplementing excessive G light (≥50 μmol m−2 s−1) with R and B lights, lettuce shoot fresh mass declined. The various combinations of irradiation of R, G and B LED did not significantly affect the contents of ascorbic acid and mineral nutrients. It is recommended to supplement 30 μmol m−2 s−1 of green light to red and blue light for a higher yield of lettuce. Therefore, the suitable LED (R:G:B) combination would be 72% of red, 10% of green and 18% of blue for lettuce cultivation following recycled hydroponics in plant factories.

Abstract:Spectral quality of radiation has a major impact on the growth, development and nutritional quality of crops. The effect of supplemental radiation (blue, red and far-red) on the growth and nutritional quality with regard to health-promoting phytochemical and micronutrient composition of two lettuce ( Lactuca sativa ) varieties (red leaf “New Red Fire” and green leaf “Two Star”) was studied. Supplemental radiation was provided by blue (450 nm), red (660 nm) or far-red (730 nm) LEDs against a background of white light (fluorescent lighting, PAR; 270 μ mol/m2/s) in a growth chamber study. All the supplemental radiation treatments increased dry shoot biomass in both varieties. However, supplemental far-red radiation increased both fresh and dry shoot biomass in both varieties. In addition, supplemental far-red radiation produced distinct morphological characteristics in lettuce plants. It produced the largest shoot biomass, bigger and taller plants, fewer leaves but with larger leaf area compared to the control, similar to the shade avoidance response. With regard to the accumulation of phytochemicals, supplemental blue radiation enhanced the total phenolic compound concentration in both varieties. In addition, supplemental blue radiation sharply increased the accumulation of several phenolic compounds in green leaf lettuce including chlorogenic acid, chicoric acid, rutin, kaempferol, luteolin and apigenin. For example, the leaf concentration of rutin in green leaf lettuce increased by 20-fold under supplemental blue radiation. Similarly, supplemental red radiation increased the concentration of many of these phenolic compounds in red leaf lettuce. However, supplemental far-red radiation had an inhibitory effect on the accumulation of chlorogenic acid, chicoric acid, rutin and kaempferol in red leaf lettuce. While supplemental radiation did not affect the accumulation of most of the micronutrients, it had a negative impact on the accumulation of some micronutrients, the response being variety dependent. The results show that supplementing white light with specific spectral quality has a major impact on the biomass accumulation, morphology and on the accumulation of many health-promoting phytochemicals and micronutrients in lettuce. While it had a large positive effect in enhancing the accumulation of several phytochemicals, it also suppressed the accumulation of some micronutrients.
Pre-harvest Short-Term Continuous LED Lighting Improves the Nutritional Quality and Flavor of Hydroponic Purple-Leaf Lettuce

Abstract:Enhancing nutritional and flavor quality has always been a pursuit of growers in the hydroponic production of lettuce in plant factories with artificial lighting (PFALs). While continuous lighting (CL) is a unique lighting mode that promotes plant yield and quality by eliminating the in-darkness period and prolonging the photosynthetic duration of plants, the specific method of CL that takes into account energy conservation is still unclear. In this study, the purple-leaf lettuce variety, 'Zixiangye', was used to investigate the effects of continuous LED lighting with different intensities and durations before harvest on the plant growth and quality of hydroponic lettuce. The results show that pre-harvest short-term CL with LED exerted a positive influence on the dry matter, as well as the nutritional and flavor quality of hydroponic purple-leaf lettuce. CL with increased light intensities (50,100,150 μmol m−2 s−1) and durations (2 days and 4 days) improved the levels of carotenoids, ascorbic acid (AsA), and anthocyanin, accompanied by a decrease in the content of nitrate, where CL of 150 μmol m−2 s−1 and 2 days is a better supplemental light condition in the study. Furthermore, 150 μmol m−2 s−1 CL gradually improved the nutritional and flavor quality of lettuce within 48 h, by promoting the accumulation of soluble sugar content and decreasing the content of lettuce bitterness, lactucin, and lactucopicrin. In summary, the utilization of pre-harvest short-term continuous LED lighting could improve the nutritional and flavor quality of hydroponic purple-leaf lettuce in PFALs. Our study proposes a light-environment strategy that uses pre-harvest short-term CL to enhance the nutritional quality and flavor of lettuce, which provides theoretical and technical guidance for the energy-efficient production of lettuce in PFALs.
Evaluation of the Effects of Nitrogen, Phosphorus, and Potassium Applications on the Growth, Yield, and Quality of Lettuce (Lactuca sativa L.)

Abstract:Lettuce (Lactuca sativa L.), a popular edible vegetable, is highly responsive to mineral fertilizers. A two-year field trial was carried out to determine how nitrogen (N), phosphorous (P), and potassium (K) application ratios affected lettuce growth, yield, and quality. Within a certain range of nutrient application rate, the plant height, stem diameter, yield, and the concentrations of vitamin C and soluble sugar were higher than those without fertilization. Compared with no fertilization condition, the yield increased approximately 13.56–22.03%, 15.15–42.42% and 7.14–10.32% under increased N, P, and K application conditions, respectively. Furthermore, substantial quadratic parabola relationships were observed between lettuce yield and fertilizer application amounts as well as most quality traits. The nitrate concentration was proportional to the N fertilization rate, while the concentrations of soluble sugar and vitamin C positively correlated with N and P application ratios, respectively. The lettuce leaves had the largest N (60.59%) and K (52.25%) accumulations, whereas the lettuce stem had the highest P (46.33%) accumulation. Above all, the optimal N, P, and K application amounts for lettuce were 315 kg N ha−1, 210 kg P2O5 ha−1, and 285 kg K2O ha−1, respectively.
