Abstract:Paraquat is a very water-soluble herbicide widely used in agricultural and nonagricultural lands. The removal of paraquat from polluted water is very essential. Hence, three experiments were arranged separately to investigate the ability of plant extracts include Aloe vera, Portulaca grandiflora, kalanchoe daigremontiana, Sedum rubotinctum, Brassica rapa var. rapa, Helxine soleirolii, Brassica oleracea var. capitata, Crassula ovate, Aptenia cordifolia, Tradescantia albiflora to the removal of paraquat (PQ) from water. In the 1st experiment, High-performance liquid chromatography was used to determine the removal rate of PQ by using plant extracts. In the 2nd and 3rd experiments, PQ solutions were subjected to plant extracts treatments and then spray on the leaves of littleseed canarygrass (Phalaris minor) under greenhouse and field conditions, respectively. In general, the results of these experiments revealed that all plant extracts resulted in higher PQ removal from water compared to control. The maximum removal of PQ was observed at pH 11, 25 °C, 30 Min, and 0.2 mL in the presence of plant extracts. In the presence of P. grandiflora, C. ovate, k. daigremontiana, A. cordifolia, H. soleirolii, Aloe vera, S. rubotinctum, B. oleracea var. capitata, B. rapa var. rapa, and T. albiflora plant extracts, PQ removal from water increased to 68.34%, 65.45%, 63.97%, 59.81%, 59.29%, 55.44%, 52.06%, 50.34%, 48.86%, and 46.44% under normal conditions and 79.71%, 78.61%, 78.00%, 75.83%, 74.66%, 72.12%, 71.57%, 71.02%, 69.35%, and 68.73% under optimum conditions, respectively. Results of the 2nd and 3rd experiments were demonstrated a decrease in PQ performance on littleseed canarygrass control. It can be concluded that plant extracts can reduce the residual of PQ in water bodies.

Abstract:Herbicide contamination in aquatic systems has become a global concern due to their long- term persistence, accumulation and health risks to humans. Paraquat, a widely used and cost-effective nonselective herbicide, is frequently applied in agricultural fields for pest control. Consequently, the removal of paraquat from contaminated water is crucial. This research presents a sustainable and environmentally benign method for paraquat removal from aqueous system by integrating wetland plants (Eichhornia crassipes) with biochar derived from melamine-modified palm kernel shells. The prepared biochar was characterized by using various analytical techniques. The effectiveness of biochar in enhancing phytoremediation was evaluated through a series of experiments, showing significant paraquat removal efficiencies of 99.7, 98.3, and 82.8% at different paraquat concentrations 50, 100, and 150 mg L-1, respectively. Additionally, present study examined the impact of biochar on the growth of E. crassipes, highlighting its potential to reduce the toxic effects of paraquat even present at higher concentrations. The paraquat removal mechanism was elucidated, focusing on the synergistic role of biochar adsorption and phytoremediation capability of E. crassipes. This innovative approach is an effective, feasible, sustainable and eco-friendly technique that can contribute to the development of advanced and affordable water remediation processes for widespread application.
Smart, Degradable, and Eco-Friendly Carboxymethyl Cellulose-Caii Hydrogel-Like Networks Gated MIL-101(FeIII) Nanoherbicides for Paraquat Delivery.

Abstract:Nanopesticides have been selected as one of the top 10 chemical innovations for enhancing the efficacy and safety of agrochemicals. Herein, smart, degradable, and eco-friendly metal-organic framework MIL-101(FeIII) nanoherbicides coated with carboxymethyl cellulose-CaII (CMC-CaII) cross-linking hydrogel-like networks are synthesized via a simple strategy. The coating of the CMC-CaII hydrogel-like gatekeepers is oriented by the coordination unsaturated FeIII clusters on the surfaces of the MIL-101(FeIII) nanocarriers to form a dense film network to prevent paraquat (PQ) leakage. Based on the stimuli factors (acid/basic pH, GSH, phosphates, and EDTA) of physiological and natural environments of target plants, the nanoherbicides are combined with five stimuli-responsive properties to attain the various controlled release of packaged PQ by the disassembly of the gatekeepers and/or the degradation of the MOF skeleton structure. More importantly, based on the stimuli responsive controlled release mechanisms, the eco-friendly nanocarriers are ultimately degraded against bioaccumulation in plants or soil. The coating of natural CMC could promote the spreading of PQ owing to improvement of wettability for aqueous droplets of nanoherbicides on hydrophobic foliage. The PQ trapped in nanocarriers can effectively prevent PQ degradation, which showed that cumulative degradation rate is ca. 2.6 times lower than that of technical PQ under UV irradiation. The prepared nanoherbicides loaded with PQ show good control efficacy against weeds by controlling the release of PQ; good safety on seed germination (germination rate 97.32-99.67 %), seedling emergence (emergence rate 95.53-99.67 %), and are beneficial for the growth of wheat seedling (increase rate of plant height 1.89-6.97 % and 0.54-5.67 % after 7 and 15 days of seedling emergence, respectively) in the greenhouse; good biosafety for honeybees (Apis mellifera L.), which shows that lethal rates were 2.04 and 2.55 times lower than technical PQ for incubation 24 and 48 h, respectively. The nanoherbicides have potential applications in the field for PQ green agriculture.

Abstract:In this study, the ZnO/Fe 3 O 4 catalyst was used as an active catalyst for the oxidation of Paraquat (PQ) herbicide in aqueous solution under ultrasonic (US) waves. FTIR, XRD, FE-SEM, and VSM analyses were performed to characterize the synthesized catalyst. Studies on the effect of radical scavengers were also carried out and the amount of organic matter degradation was determined by measuring the TOC. Under the optimized conditions (catalyst concentration = 0.75 g/L, herbicide concentration = 10 ppm, US power = 70w), the degradation and mineralization rates of the herbicide were acquired as 96.1% and 68% within 60 min, respectively. The quenching tests showed that the hydroxyl (oOH) radical was the most effective oxidant agent in the degradation process of the PQ under ZnO/Fe 3 O 4 /US system. The toxicity of treated effluent assayed by Daphnia Magna was decreased from %73.16 in raw samples to %7.2 in the treated samples, during 96 h. Finally, it can be concluded that ZnO/Fe 3 O 4 /US process can be successfully performed as an effective process to herbicides in aqueous solutions, due to the high efficiency and excellent catalytic activity.

Abstract:Paraquat (PQ) is a toxic, organic herbicide for which there is no specific antidote. Although banned in some countries, it is still used as an irreplaceable weed killer in others. The lack of understanding of the precise mechanism of its toxicity has hindered the development of treatments for PQ exposure. While toxicity is thought to be related to PQ-induced oxidative stress, antioxidants are limited in their ability to ameliorate the untoward biological responses to this agent. Summarized in this review are data on the absorption, distribution, metabolism, excretion, and toxicity (ADME/T) of PQ, focusing on the essential roles of individual transporters and enzymes in these processes. Based on these findings, strategies are proposed to design and test specific and effective antidotes for the clinical management of PQ poisoning.

However, despite its benefits in weed management, PQ poses significant risks due to its severe toxicity, which affects multiple organ systems in both humans and animals. Electrochemical sensors for improved detection of paraquat in food samples: A review

Abstract:Paraquat (1,10-dimethyl-4,40-dipyridinium chloride), also known as methyl viologen, is widely used as a quaternary ammonium herbicide (broadleaf weed killer) all over the world owing to its excellent effect in plant cells for crop protection and horticultural use. However, it is dangerous because of its high acute toxicity even at low concentrations. Its detection in the environment is therefore necessary. As a consequence of its widespread usage, it causes genotoxic, teratogenic as well as other environmental and ecological adverse impacts. Exposure to PQ leads to a high mortality rate because no specific drug is effective for treatment. Excessive consumption of PQ can cause cellular damage and necrosis in the brain, heart, lungs, liver, and kidneys. The diversity and sensitivity of the analyses currently required have forced the experimenter to use more advanced and efficient techniques, which can provide qualitative and quantitative results in complex environments. Electrochemical methods generally meet these criteria while offering other advantages to achieve excellent accuracy and fast handling. This paper provides an overview of the determination of PQ using electrochemical methods combined with several modified electrodes in food samples, including milk, apple juice, tomato juice, and potato juice. Emphasis was placed on the most relevant modifiers used to generate high selectivity and sensitivity such as noble metals, metallic nanoparticles, polymers, biomolecules, clay, and apatite minerals. Comprehensively, it is strongly convincing that the synergy between the sensor substrate and the modifier architecture gives the electrodes a high capacity to detect paraquat in complex matrices such as food. In line with the context, information's on the mechanism of electrooxidation or reduction of PQ has been reported with the discussion of some future prospects and some insights. To the best of our knowledge, there is no review article relating the electrochemical determination of paraquat.

Abstract:Paraquat herbicide has served over five decades to control annual and perennial weeds. Despite agricultural benefits, its toxicity to terrestrial and aquatic environments raises serious concerns. Paraquat cannot rapidly degrade in the environment and is adsorbed in clay lattices that require urgent environmental remediation. Advanced oxidation processes (AOPs) and bioaugmentation techniques have been developed for this purpose. Among various techniques, bioremediation is a cost-effective and eco-friendly approach for pesticide-polluted soils. Though several paraquat-degrading microorganisms have been isolated and characterized, studies about degradation pathways, related functional enzymes and genes are indispensable. This review encircles paraquat removal from contaminated environments through adsorption, photocatalyst degradation, AOPs and microbial degradation. To provide in-depth knowledge, the potential role of paraquat degrading microorganisms in contaminated environments is described as well.

A sustainable approach for the removal of chlorpyrifos pesticide from aqueous phase using novel nano magnetized biochar

Abstract:In last two decades, organophosphate pesticides have become a prominent class of toxic water pollutants. Besides, these are not only poisonous for living beings, but their decomposition is also a time-taking process. For the first time, our study reports the application of nano-magnetized and carbonaceous adsorbents, obtained from orange peels (bio-waste), for the removal of chlorpyrifos from water resources. The -nano magnetized orange peels (NMOPs) were synthesized by the co-precipitation method with the use of iron salts. The competency of orange peels for chlorpyrifos adsorption was further enhanced by converting untreated orange peels (UOPs) into carbonaceous orange peels (COPs). The adsorbents were analyzed with FT-IR, SEM, EDX, XRD and VSM. The peak at 540 cm −1 in FTIR spectra corresponded to the Fe-O bond and confirmed the successful doping of iron salts on orange peels. Biosorption studies carried out by varying different parameters showed that these adsorbents are efficient for the uptake of chlorpyrifos. The effects of adsorbent dose, contact time, chlorpyrifos initial concentration, and pH were investigated to assess the adsorbents' efficiency via biosorption studies. It was noted that the adsorbents showed maximum adsorption with adsorbent dose = 0.4 g, contact time = 50 mins, chlorpyrifos concentration = 150 ppm, pH = 4 and temperature = 30 °C. The adsorption maximum values (q max ) were found to be 68, 108 & 100 mg g −1 for the UOPs, COPs and NMOPs, respectively. Kinetic studies indicate that the removal of chlorpyrifos followed the pseudo-second-order model of adsorption. The adsorption results were effectively described by the Langmuir isotherm and Freundlich isotherm models.

Abstract:Paraquat (PQ) is a highly effective herbicide with contact toxicity. PQ mainly accumulates in the lungs after absorption into the blood circulation. The respiratory function failure caused by PQ-induced lung injury, especially the irreversible pulmonary fibrosis in late phase, is the leading cause of death in patients with PQ poisoning. The mechanism of PQ poisoning is still unclear. Now it is speculated that oxidative stress and inflammation injury are the main pathogenic mechanisms, and abnormal gene expression, mitochondrial damage, loss of pulmonary surfactant, cytokine network and unbalanced matrix metalloproteinases/tissue inhibitors may be also involved in the pathogenesis. In addition to reducing poison absorption and increasing its removal, the current clinical treatment is mainly composed of antioxidant and anti-immune response, but has poor therapeutic effects. Although many novel methods of treatment have been proposed, most of them are still in the experimental stage. It is a hot spot to clarify the mechanism of PQ poisoning and to seek safe and effective treatment of pulmonary fibrosis. This article reviews the research progress on pathogenesis and treatment of PQ-induced pulmonary fibrosis.
Specific recognition of cationic paraquat in environmental water and vegetable samples by molecularly imprinted stir-bar sorptive extraction based on monohydroxylcucurbit[7]uril-paraquat inclusion complex

Abstract:Molecularly imprinted stir-bar coatings were created based on a hydroxylcucurbit[7]uril-paraquat inclusion complex. The inclusion complex that contained paraquat (PQ) as a template and monohydroxylcucurbit[7]uril ((OH)Q[7]) as a monomer was preassembled mainly through cavity inclusion interaction of (OH)Q[7] to form a one-dimensional self-assembly structure. The inclusion complex was anchored chemically on the surface of a glass stir bar with hydroxy-terminated poly(dimethylsiloxane) by the sol-gel technique to obtain a molecularly imprinted polymer-coated stir bar (MIP-SB). The molecularly imprinted coating showed specific adsorption for cationic PQ in aqueous media. Other quaternary amine compounds with a similar structure that coexisted in the solution, such as ethyl-viologen, diquat, and difenzoquat, were almost not extracted by the prepared MIP-SB. The sorptive capacity of the MIP-SB for PQ was nearly four times that of the non-imprinted stir bar (NIP-SB). The recognition mechanism indicated that the selectivity and extraction capacity resulted mainly from the imprinted cavity in the polymer that was formed by a one-dimensional assembly structure consisting of the (OH)Q[7]-PQ inclusion complex. The imprinted cavity was complementary to the PQ in shape, size, and functionality. A method to determine PQ in environmental water and vegetable samples was developed by combining MIP-SB sorptive extraction with HPLC-UV. The linear range was from 100 to 10,000 ng L-1 with a 8.2 ng L-1 detection limit for water samples and 0.02-0.85 mg kg-1 with a 0.005 mg kg-1 detection limit for vegetable samples. The limit of detection for both samples was lower than the EU-established maximum residual levels and that of other previously reported methods. The average recoveries were 70.0-96.1% with a relative standard deviation ≤ 7.6%, which showed the successful application in real sample analysis. Molecularly imprinted stir-bar coatings were created based on a hydroxylcucurbit[7]uril-paraquat (PQ) inclusion complex, which showed a specific recognition toward cationic PQ. A method to determine PQ in environmental water and vegetable samples was established by combining MIP-SB sorptive extraction with HPLC-UV.
A novel 2D g-C3N4 material applied for Paraquat adsorbing and detoxifying in vitro and in vivo

Abstract:In the environmental safety area, the widespread use of the herbicide Paraquat (PQ) poses a great threat to hydrobionts and mammals. Due to the lack of specific antidote, it may lead to irreversible pulmonary fibrosis with a mortality rate of 60%. Therefore, it is necessary to develop an effective and specific PQ antidote. The g-C3N4 (HPCN) with excellent surface physicochemical properties was prepared by a two-step calcination method using urea and dicyandiamide as raw materials, showing a significant photocatalyst against environmental PQ pollution. The SEM results showed that HPCN possesses a porous layered structure. X-ray diffraction and infrared spectroscopy indicated that the conjugated aromatic rings were orderly stacked, forming a 2D layered structure of g-C3N4. The HPCN had a larger specific surface area (56.84 m2 g-1) and pore volume (0.2718 cm3 g-1), which enhanced its adsorption capacity and photocatalytic activity. HPCN exhibited an effective adsorption rate of 38.25% for PQ in water under light. Compared with the PQ group (54.8%), the cell viability of the HPCN group (91.4%) significantly increased by 36.6%, and the SEM observation revealed the restoration of normal cell morphology. The HPCN effectively reduced PQ content in zebrafish and mice in vivo, resulting in an approximately 70% increase in survival rate. The UV-Vis results indicated that the adsorption rate of HPCN for PQ in zebrafish was 43.5%. The enhanced catalytic performance of HPCN provides a promising solution for the detoxification of PQ and of other environmental pollutants.
Dienediamine: A safe surrogate for the herbicide paraquat

Abstract:Paraquat (PQ) has been used as an herbicide worldwide because of its potent activity against weeds. However, it is highly toxic to humans. The very high fatality of PQ poisoning is due to its inherent toxicity and the lack of any effective treatment. Consequently, developing a non-toxic herbicide with comparable efficacy to PQ will contribute to global food security and help prevent PQ-related fatalities. Herein, we report a new herbicide called dienediamine, which was discovered from how to intervene the redox cycle of PQ, an inherent toxicity nature. Dienediamine, the "reduced" form of PQ with no function as an electron transfer agent, was shown to be non-toxic through comprehensive in vivo and in vitro experiments at molar concentrations equivalent to PQ's absolute lethal dose. Remarkably, dienediamine can undergo conversion to PQ under natural sunlight and ambient air conditions, exhibiting herbicidal activities that are comparable to those of PQ. The conversion of dienediamine to PQ, which is toxic to chloroplasts, is the key mechanism underlying its potent herbicidal activity. Our study discovers that dienediamine is a safe and superior alternative to PQ, possessing significant potential for application in sustainable agriculture globally.
Research progress of combination therapy of blood purification in the treatment of paraquat poisoning

Abstract:Paraquat is a kind of herbicide effective in weeding, characterized by low cost and environmentally friendly property, which is widely used in agricultural weeding. However, the high toxicity of paraquat can cause damage to the lungs, liver, kidney, and other organs after oral intake. Worse still the patient will develop pulmonary fibrosis and will die of respiratory failure. At present, there is no specific detoxification drug for patients poisoned by paraquat. Based on the current therapeutic schedule, blood purification is considered to be the most effective method for the removal of paraquat-related toxins. However, there is a lot of controversy over the combined method. This paper expounds on the poisoning mechanism of paraquat and the advantages and disadvantages of different blood purification methods, aiming to find a specific scheme that can be applied to the clinic and improve the organ function and prognosis of patients.

Abstract:In recent years, organic dyes and herbicides have become a major source of water pollution threatening human health and the environment. In this study, acid-alkali-activated lightweight expanded clay aggregate (LECA) was used for the binary adsorption of methylene blue (MB) dye and paraquat (PQ) herbicide. The effects of various parameters, including initial dye and paraquat concentration, adsorbent dosage, contact time, solution pH, and temperature, were studied and optimized to maximize single and binary mixture adsorption. In the binary system, MB's maximum adsorption capacity was 37.03 mg/g, which was higher than in the single system (31.74 mg/g). In contrast, PQ adsorption capacity in the binary system was significantly reduced (21.23 to 10.09 mg/g). As a result, MB inhibited PQ uptake in the binary system significantly. Adsorption kinetics and equilibrium isotherms were performed for both single and binary mixtures. Experimental data were simulated using the Langmuir, Freundlich, and Temkin isotherm models in the single system and competitive Langmuir in the binary system. Kinetic study revealed that dye and paraquat herbicide adsorption process followed the pseudo-second-order model. Moreover, to evaluate the reuse performance of the adsorbents, the adsorption-desorption process of MB and PQ was repeated five times. The study proved that activated LECA can simultaneously remove dyes and herbicides from aqueous solutions as a sustainable and reusable adsorbent.

Abstract:Paraquat (PQ) has high acute toxicity, even at low concentrations. For most people, the main pathway of exposure to PQ is through the diet. Therefore, the development of simple and efficient methods for PQ testing is critical for ensuring food safety. In this study, a new electrochemical detection strategy for paraquat is proposed based on the specific binding of PQ to its nucleic acid aptamer. Firstly, the Zn/Fe bimetallic ZIF derived nanoporous carbon (Zn/Fe-ZIF-NPC) and nickel hexacyanoferrate nanoparticles (NiHCF-NPs) were sequentially modified onto the glassy carbon electrode (GCE). NiHCF-NPs served as the signal probes, while Zn/Fe-ZIF-NPC facilitated electron transfer and effectively enhanced the sensing signal of NiHCF-NPs. Au nanoparticles (AuNPs) were then electrodeposited on the NiHCF-NPs/Zn/Fe-ZIF-NPC/GCE and then the thiolated aptamer was assembled on the AuNPs/NiHCF-NPs/Zn/Fe-ZIF-NPC/GCE via Au-S bonding. When incubated with PQ, the formation of PQ-aptamer complexes delayed the interfacial electron transport reaction of NiHCF-NPs, which caused a decrease in the current signals. As a result, simple and highly sensitive detection of PQ can be readily achieved by detecting the signal changes. A linear range was obtained from 0.001 to 100 mg/L with a detection limit as low as 0.34 μg/L. Due to the recognition specificity of the aptamer to its target molecule, the proposed method has excellent anti-interference ability. The prepared electrochemical aptasensor was successfully used for PQ assay in lettuce, cabbage and agriculture irrigation water samples with recoveries ranging from 96.20% to 104.02%, demonstrating the validity and practicality of the proposed method for PQ detection in real samples.

Abstract:Paraquat (PQ) and diquat (DQ) are quaternary ammonium herbicides which have been used worldwide for controlling the growth of weeds on land and in water. However, PQ and DQ are well known to be toxic. PQ is especially toxic to humans. Moreover, there is no specific antidote for PQ poisoning. The main treatment for PQ poisoning is hemoperfusion to reduce the PQ concentration in blood. Therefore, it is essential to be able to detect PQ and DQ concentrations in biological samples. This critical review summarizes the articles published from 2010 to 2022 and can help researchers to understand the development of the sample treatment and analytical methods for the determination of PQ and DQ in various types of biological samples. The sample preparation includes liquid–liquid extraction, solid-phase extraction based on different novel materials, microextration methods, and other methods. Analytical methods for quantifying PQ and DQ, such as different chromatography and spectroscopy methods, electrochemical methods, and immunological methods, are illustrated and compared. We focus on the latest advances in PQ and DQ treatment and the application of new technologies for these analyses. In our opinion, tandem mass spectrometry is a good choice for the determination of PQ and DQ, due to its high sensitivity, high selectivity, and high accuracy. As far as we are concerned, the best LOD of 4 pg/mL for PQ in serum can be obtained.

Treatment of Paraquat Poisoning in Man: Methods to Prevent Absorption

Abstract:Theoretically, absorption of an ingested dose of paraquat may be reduced by (1) gastric lavage, (2) induced emesis, (3) whole-gut lavage or (4) by the oral administration of adsorbent substances. 1 Animal experiments suggest that paraquat is absorbed poorly from the stomach and absorbed incompletely (< 5%) from the small intestine over a 1-6-h period. Although gastric lavage would therefore seem a logical way to ameliorate the toxicity of an ingested dose of paraquat, peak plasma concentrations are attained rapidly and evidence for the efficacy of gastric lavage in man is poor. 2 In 1977, a potent emetic (PP796) was added to liquid and solid formulations of paraquat because experiments in primates had demonstrated a fivefold reduction in toxicity. In man, ingestion of formulations containing an emetic is more likely to cause spontaneous vomiting within 30 min than non-emetic preparations. However, definite evidence of benefit, as judged by improved patient prognosis, has yet to be established. 3 Gut lavage has been shown to remove only a small proportion of an ingested dose of paraquat. At the flow rates employed in man (75 ml/min), approximately 0.5-1.0 litres of lavage fluid/h may be absorbed across the intestinal wall. Since there is a theoretical risk of increasing paraquat absorption, the use of whole-gut lavage cannot be recommended. 4 Bipyridilium herbicides are adsorbed by soil and clay minerals, and montmorillonite in particular has been shown to be a strong binding agent in vitro. Accordingly, the use of Fuller's Earth (calcium montmorillonite) and Bentonite (sodium montmorillonite) for the treatment of poisoning has been investigated in animal models. Both agents reduce plasma paraquat concentrations and mortality in animals when administered after an oral dose of paraquat. Recently, other adsorbent materials have been found to have high maximum adsorption capacities for paraquat. In particular, activated charcoals and cation-exchange resins have attracted interest. Unfortunately, as yet, there is no evidence that the use of adsorbents in man is of therapeutic value.

Abstract:An in vivo microcapillary sampling (MCS) method coupled with matrix-assisted laser desorption/ionization fourier transform ion cyclotron resonance mass spectrometry (MALDI-FTICR-MS) analysis was utilized to monitor the real-time bipyridine quaternary ammonium herbicides concentrations and assess their uptake and elimination behaviors in living cabbage plants noninvasively. Under optimized conditions, the proposed method for paraquat (PQ) and diquat (DQ) determination showed wide linear ranges (7.81-500 μg/kg), low limits of detection (0.1-0.9 μg/kg), and good reproducibility. In vivo tracking results demonstrated that different absorption behaviors between PQ and DQ existed in living vegetables and DQ was more easily absorbed. Through decay kinetics model fitting, herbicide half-lives were 1.32 and 1.86 days for PQ and DQ, respectively. To summarize, in vivo MCS method provides valuable information on herbicide risks for agricultural production, which is suitable for temporal, spatial, and longitudinal studies in the same living system and multicompartmental studies in the same organism.

Abstract:Abstract Paraquat (PQ) is a widely used fast-acting pyridine herbicide. Accidental ingestion or self-administration via various routes can cause severe organ damage. Currently, no effective antidote is available commercially, and the mortality rate of poisoned patients is exceptionally high. Here, the efficacy of anthrahydroquinone-2-6-disulfonate (AH 2 QDS) was observed in treating PQ poisoning by constructing in vivo and ex vivo models. We then explored the detoxification mechanism of AH 2 QDS. We demonstrated that, in a rat model, the PQ concentration in the PQ + AH 2 QDS group significantly decreased compared to the PQ only group. Additionally, AH 2 QDS protected the mitochondria of rats and A549 cells and decreased oxidative stress damage, thus improving animal survival and cell viability. Finally, the differentially expressed genes were analysed in the PQ + AH 2 QDS group and the PQ group by NextGen sequencing, and we verified that Nrf2’s expression in the PQ + AH 2 QDS group was significantly higher than that in the PQ group. Our work identified that AH 2 QDS can detoxify PQ by reducing PQ uptake and protecting mitochondria while enhancing the body's antioxidant activity.
