Abstract:Separation and classification are important operations in particle technology, but they are still limited in terms of suspended particles in the micrometer and nanometer size-range. Electrical fields can be beneficial for sorting such particles according to material properties. A mechanism based on strong and inhomogeneous fields is dielectrophoresis (DEP). It can be used to separate microparticles according to their material properties, such as conductivity and permittivity, by selectively trapping one particle type while the other can pass the separator. Conventional DEP-separators show either a limitation in throughput or frequency bandwidth. A low throughput limits the economical feasibility in many cases. A lower frequency bandwidth limits the variety of materials that can be sorted by DEP. To separate semiconducting particles from a mixture containing particles with higher conductivity according to their material, high frequencies are required. Possible applications are the separation of semiconducting and metallic carbon nanotubes or the separation of carbon-coated lithium iron phosphate particles from graphite in the recycling process of spent lithium-ion batteries. In this publication, we aim to display how to tune the electrical impedance of a high-throughput DEP separator based on custom-designed printed circuit boards to increase its frequency bandwidth. By adding inductors to the electrical circuit, we were able to increase the frequency bandwidth from 500 kHz to over 11 MHz. The experiments in this study act as proof-of-principle. Furthermore, a non-deterministic way to increase the impedance of the setup is shown, yielding a maximum frequency of 39.16 MHz.

Abstract:Dielectrophoresis (DEP) is a selective electrokinetic particle manipulation technology which is applied since almost 100 years and currently finds most application in biomedical research using microfluidic devices operating at moderate to low throughput. This paper reviews DEP separators capable of high‐throughput operation and research addressing separation and analysis of non‐biological particle systems. Apart from discussing particle polarization mechanisms, this review summarizes the early applications of DEP for dielectric sorting of minerals and lists contemporary applications in solid/liquid, liquid/liquid, and solid/air separation, for example DEP filtration or airborne fiber length classification; the review also summarizes developments in DEP fouling suppression, gives a brief overview of electrocoalescence and addresses current problems in high‐throughput DEP separation. We aim to provide inspiration for DEP application schemes outside of the biomedical sector, for example for the recovery of precious metal from scrap or for extraction of metal from low‐grade ore.This article is protected by copyright. Abstract:Dielectrophoresis (DEP), the induced motion of polarizable particles in a non-uniform electric field, has been proven as a perfect candidate to transport, accumulate, separate and characterize micro-/nano-scale bioparticles in microfluidic systems. However, conventional fabrication technologies are complex, time-consuming and relatively expensive, leading to low throughput of the DEP-based systems. In this paper, we report a novel microfluidic alternating current DEP (AC-DEP) chip fabricated via inexpensive screen printing method. The innovation of our work consists in the extreme simplicity of the fabrication procedure, i.e., the main components, including electrodes and channels, were constructed by layer-by-layer screen printing process, which is especially suitable for high-throughput mass production. Carbon paste, instead of metals, was used to print interdigitated electrodes with semi-3D structure which not only reduces dramatically the chip cost but also increases particle trapping efficiency. To test the chip performance, yeast cells, as model cells, were trapped and separated from a mixed suspension with PS microspheres. Our results show that high capture rate and separation efficiency can be achieved under optimized conditions.

Abstract:Dielectrophoretic (DEP) separation has been recognized as a practical tool in the separation of cells and particles for clinical diagnosis, pharmaceutical industry and environmental monitoring. Assembly of particles and cells under DEP force is a common phenomenon and has an influence on their separation but has not been understood fully. Encouraged by these aspects, we developed a microfluidic device with bipolar electrode array to investigate the assembly and separation of particles and cells at a large scale. Firstly, we studied the assembly and evolution mechanisms of particles of one types under AC electric field. Then, we investigated the interaction and assembly of multiple particles with dissimilar properties under DEP force. Depending on the development of microfluidic device, we visualize the assembly process of yeast cells at the electrode rims and of polystyrene particles at the channel centers, and explore influence of pearl chain formation on their separation. With increasing flow velocity from 288 to 720 L/h, purity of PS5 particles surpasses 94.9%. Furthermore, we studied DEP response of Scenedesmus sp. and C. vulgaris, and explored the influence of cell chains on the isolation of C. vulgaris. Purity of Scenedesmus sp. and C. vulgaris witnessed a decrease from 95.7% to 90.8% when flow rate increases from 288 to 864 L/h. Finally, we investigated extension of electric field under chains of Oocystis sp. at the electrode rims by studying chain formation and capture of C. vulgaris, and study its effect on cell chain length, recovered cell purity and cell concentration. When chains of Oocystis sp. were formed, the purity of C. vulgaris kept unchanged and the concentration decreased from 2793 cells/L to 2039 cells/ L. This work demonstrates continuous DEP-baed assembly and separation of particles and cells, which facilitates high-efficiency isolation of targeted cells.

Abstract:Cell separation has become @important in biological and medical applications. Dielectrophoresis (DEP) is widely used due to the advantages it offers, such as the lack of a requirement for biological markers and the fact that it involves no damage to cells or particles. This study aimed to report a novel approach combining 3D sidewall electrodes and contraction/expansion (CEA) structures to separate three kinds of particles with different sizes or dielectric properties continuously. The separation was achieved through the interaction between electrophoretic forces and inertia forces. The CEA channel was capable of sorting particles with different sizes due to inertial forces, and also enhanced the nonuniformity of the electric field. The 3D electrodes generated a non-uniform electric field at the same height as the channels, which increased the action range of the DEP force. Finite element simulations using the commercial software, COMSOL Multiphysics 5.4, were performed to determine the flow field distributions, electric field distributions, and particle trajectories. The separation experiments were assessed by separating 4 μm polystyrene (PS) particles from 20 μm PS particles at different flow rates by experiencing positive and negative DEP. Subsequently, the sorting performances of the 4 μm PS particles, 20 μm PS particles, and 4 μm silica particles with different solution conductivities were observed. Both the numerical simulations and the practical particle separation displayed high separating efficiency (separation of 4 μm PS particles, 94.2%; separation of 20 μm PS particles, 92.1%; separation of 4 μm Silica particles, 95.3%). The proposed approach is expected to open a new approach to cell sorting and separating.

Abstract:Dielectrophoresis (DEP) is an advanced microfluidic manipulation technique that is based on the interaction of polarized particles with the spatial gradient of a non-uniform electric field to achieve non-contact and highly selective manipulation of particles. In recent years, DEP has made remarkable progress in the field of microfluidics, and it has gradually transitioned from laboratory-scale research to high-throughput manipulation in practical applications. This paper reviews the recent advances in dielectric manipulation and separation of microparticles and biological cells and discusses in detail the design of chip structures for the two main methods, direct current dielectrophoresis (DC-DEP) and alternating current dielectrophoresis (AC-DEP). The working principles, technical implementation details, and other improved designs of electrode-based and insulator-based chips are summarized. Functional customization of DEP systems with specific capabilities, including separation, capture, purification, aggregation, and assembly of particles and cells, is then performed. The aim of this paper is to provide new ideas for the design of novel DEP micro/nano platforms with the desired high throughput for further development in practical applications.

Abstract:The separation of target microparticles using microfluidic systems owns extensive applications in biomedical, chemical, and materials science fields. Integration of microfluidic sorting systems employing dielectrophoresis (DEP) technology has been widely investigated. However, enhancing separation efficiency, purity, stability, and integration remains a pressing issue. This study proposes a stepwise multi-stage continuous DEP separation microfluidic chip with a microfilter structure. By leveraging a stepwise electrode configuration, a gradient electric field is generated to drive target microparticles along the electric field gradient, thereby enhancing separation efficiency. Innovative integration of a microfilter structure facilitates simultaneous filtration and improves flow field distribution, thus enhancing system stability. Through the synergistic effect of stepwise electrodes and the microfilter structure, superior coupling of electric and flow fields is achieved, consequently improving the sorting purity, separation efficiency, and system stability of the DEP-based microfluidic sorting system. Validation through simulation and separation of polystyrene microspheres demonstrates the excellent particle separation performance of the proposed system. It evidently shows potential for seamless extension to various biological microparticle sorting applications, harboring significant prospects in the biomedical domain field.

Abstract:Dielectrophoresis (DEP) has been widely explored to separate cells for various applications. However, existing DEP devices are limited by the high cost associated with the use of noble metal electrodes, the need of high-voltage electric field, and/or discontinuous separation (particularly for devices without metal electrodes). We developed a DEP device with liquid electrodes, which can be used to continuously separate different types of cells or particles based on positive DEP. The device is made of polydimethylsiloxane (PDMS), and ionic liquid is used to form the liquid electrodes, which has the advantages of low cost and easy fabrication. Moreover, the conductivity gradient is utilized to achieve the DEP-based on-chip cell separation. The device was used to separate polystyrene microbeads and PC-3 human prostate cancer cells with 94.7 and 1.2% of the cells and microbeads being deflected, respectively. This device is also capable of separating live and dead PC-3 cancer cells with 89.8 and 13.2% of the live and dead cells being deflected, respectively. Moreover, MDA-MB231 human breast cancer cells could be separated from human adipose-derived stem cells (ADSCs) using this device with high purity(81.8 and 82.5% for the ADSCs and MDA-MB-231 cells, respectively). Our data suggest the great potential of cell separation based on conductivity-induced DEP using affordable microfluidic devices with easy operation.

Abstract:Traditional methods of dielectrophoretic (DEP) separation using planar microelectrodes and recent research about vertical electrodes cannot improve both effectiveness and integration for industry. This paper describes the design and numerical simulation of microdevice using COMSOL, which can separate particles by lateral electrical fields uniformly along the vertical direction in the channel. Particles tracing is fully coupled to electric field and fluidic field, which is a complete design flow of DEP devices. Particle separation is demonstrated for continuous separation in a flowing mixture of 1 μm and 10 μm particles with 100% separation efficiency.

Abstract:Dielectrophoresis is a robust approach for manipulating bioparticles in microfluidic devices. In recent years, many groups have developed dielectrophoresis-based microfluidic systems for separation and concentration of various types of bioparticles, where the gradient of the electric field causes dielectrophoresis force acting on the suspended particles. Enhancing the gradient of the electric field with three-dimensional (3D) electrodes can significantly improve the efficiency of the system. Implementing planar electrodes in a 3D arrangement is a simple option to form a 3D-electrode configuration. This paper reports the development of a novel dielectrophoretic microfluidic system for continuously manipulating microparticles such as polystyrene microbeads and Saccharomyces cerevisiae cells. The fabrication process was relatively simple, cost-effective, and precise. Moreover, the device was tested to find the impact of various parameters on the concentration of polystyrene microbeads and separation of live and dead cells. The optimum working conditions, including flow rate, applied voltage amplitude, and frequency, were obtained accordingly. Furthermore, the experimentally observed trajectories of the particles agreed well with simulated counterparts. The device was able to efficiently and continuously perform with high throughput. Under an optimum condition, an efficiency of approximately 100% was obtained, confirming the capability of the proposed design with four triangular electrodes for continuous focusing and separation of live and dead cells as well as polystyrene particles.

Abstract:Cell counting and sorting is a vital step in the purification process within the area of biomedical research. It has been widely reported and accepted that the use of hydrodynamic focusing in conjunction with the application of a dielectrophoretic (DEP) force allows efficient separation of biological entities such as platelets from red blood cell (RBC) samples due to their size difference. This paper presents computational results of a multiphysics simulation modelling study on evaluating continuous separation of RBCs and platelets in a microfluidic device design with saw-tooth profile electrodes via DEP. The theoretical cell particle trajectory, particle cell counting, and particle separation distance study results reported in this work were predicted using COMSOL v6.0 Multiphysics simulation software. To validate the numerical model used in this work for the reported device design, we first developed a simple y-channel microfluidic device with square "in fluid" electrodes similar to the design reported previously in other works. We then compared the obtained simulation results for the simple y-channel device with the square in fluid electrodes to the reported experimental work done on this simple design which resulted in 98% agreement. The design reported in this work is an improvement over existing designs in that it can perform rapid separation of RBCs (estimated 99% purification) and platelets in a total time of 6-7 s at a minimum voltage setting of 1 V and at a minimum frequency of 1 Hz. The threshold for efficient separation of cells ends at 1000 kHz for a 1 V setting. The saw-tooth electrode profile appears to be an improvement over existing designs in that the sharp corners reduced the required horizontal distance needed for separation to occur and contributed to a non-uniform DEP electric field. The results of this simulation study further suggest that this DEP separation technique may potentially be applied to improve the efficiency of separation processes of biological sample scenarios and simultaneously increase the accuracy of diagnostic processes via cell counting and sorting.
Tunable membrane-less dielectrophoretic microfiltration by crossing interdigitated electrodes

Abstract:Filtration is a crucial step in the analysis of living microparticles. In particular, the selective microfiltration of phytoplankton by size and shape remains an open problem, even though these criteria are essential for their gender and/or species identification. However, microfiltration devices necessitate physical membranes which complicate their fabrication, reduce the sample flow rate and can cause unwanted particle clogging. Recent advances in microfabrication such as electrode High Precision Capillary Printing allow to rapidly build electrode patterns over wide areas. In this study, we introduce a new concept of membrane-less dielectrophoretic (DEP) microfiltration suitable for large scale microfabrication processes. The proposed design involves two pairs of interdigitated electrodes at the top and the bottom of a microfluidic channel. We use finite-element calculations to analyse how the DEP force field throughout the channel, as well as the resulting trajectories of particles depend on the geometry of the system, on the physical properties of the particles and suspending medium and on the imposed voltage and flow rates. We numerically show that in the negative DEP regime, particles are focused in the channel mid-planes and that virtual pillars array leads either to their trapping at specific stagnation points, or to their focusing along specific lines, depending on their dielectrophoretic mobility. Simulations allow to understand how particles can be captured and to quantify the particle filtration conditions by introducing a critical dielectrophoretic mobility. We further illustrate the principle of membrane-less dielectrophoretic microfiltration using the proposed setup, by considering the separation of a binary mixture of polystyrene particles with different diameters, and validate it experimentally.

Abstract:Dielectrophoresis is an electric field-based technique for moving neutral particles through a fluid. When used for particle separation, dielectrophoresis has many advantages compared to other methods, providing label-free operation with greater control of the separation forces. In this paper, we design, build, and test a low-voltage dielectrophoretic device using a 3D printing approach. This lab-on-a-chip device fits on a microscope glass slide and incorporates microfluidic channels for particle separation. First, we use multiphysics simulations to evaluate the separation efficiency of the prospective device and guide the design process. Second, we fabricate the device in PDMS (polydimethylsiloxane) by using 3D-printed moulds that contain patterns of the channels and electrodes. The imprint of the electrodes is then filled with silver conductive paint, making a 9 pole comb electrode. Lastly, we evaluate the separation efficiency of our device by introducing a mixture of 3 $\mu$m and 10 $\mu$m polystyrene particles and tracking their progression. Our device is able to efficiently separate these particles when the electrodes are energized with $\pm$12 V at 75 kHz. Overall, our method allows the fabrication of cheap and effective dielectrophoretic microfluidic devices using commercial off-the-shelf equipment.

High Throughput Separation of Cells Achieved Through the Particle Characteristic Dielectrophoretic Response and Further Focusing

Abstract:Previous devices to separate cells by the characteristic force they experience due to dielectrophoresis, which depends on the size and electric properties of the particle, were limited by the flow rates and particle concentrations separation could be achieved at. To unlock the potential of Lab-on-a-chip technology to create flexible, efficient and multifunction devices at low cost it is necessary to increase the rates at which separations can be performed. Here we present a device capable of high throughput continuous separation of cells from microparticles and demonstrate separation at 7 uL/min in a microchannel with a high density of cells and microparticles in the sample of 10^8-10^9 cells/mL. This device uses the characteristic response of the particles due to dielectrophoresis to provide the initial separation before crucially focusing the particles into completely separate particle streams allowing the operation at high flow rates and particle concentrations. The device is demonstrated with the separation of polystyrene (PS) beads 8 microns, 25 microns and human dermal microvascular endothelial cell line (HMEC-1 cells) and human liver cell line (HepG2 cells) suspended in a DEP buffer solution. We show that it is possible to achieve not only high flow rate separation but also high sample purity with, after separation and focusing, one output containing 100% cells.

Abstract:Microfluidics is the combination of micro/nano fabrication techniques with fluid flow at microscale to pursue powerful techniques in controlling and manipulating chemical and biological processes. Sorting and separation of bio-particles are highly considered in diagnostics and biological analyses. Dielectrophoresis (DEP) has offered unique advantages for microfluidic devices. In DEP devices, asymmetric pair of planar electrodes could be employed to generate non-uniform electric fields. In DEP applications, facing 3D sidewall electrodes is considered to be one of the key solutions to increase device throughput due to the generated homogeneous electric fields along the height of microchannels. Despite the advantages, fabrication of 3D vertical electrodes requires a considerable challenge. In this study, two alternative fabrication techniques have been proposed for the fabrication of a microfluidic device with 3D sidewall electrodes. In the first method, both the mold and the electrodes are fabricated using high precision machining. In the second method, the mold with tilted sidewalls is fabricated using high precision machining and the electrodes are deposited on the sidewall using sputtering together with a shadow mask fabricated by electric discharge machining. Both fabrication processes are assessed as highly repeatable and robust. Moreover, the two methods are found to be complementary with respect to the channel height. Only the manipulation of particles with negative-DEP is demonstrated in the experiments, and the throughput values up to 105 particles / min is reached in a continuous flow. The experimental results are compared with the simulation results and the limitations on the fabrication techniques are also discussed.

Abstract:Background Separation, classification, and focusing of microparticles are essential issues in microfluidic devices that can be implemented in two categories: using labeling and label-free methods. Label-free methods differentiate microparticles based on their inherent properties, including size, density, shape, electrical conductivity/permittivity, and magnetic susceptibility. Dielectrophoresis is an advantageous label-free technique for this objective. Besides, centrifugal microfluidic devices exploit centrifugal forces to move liquid and particles. The simultaneous combination of dielectrophoretic and centrifugal forces exerted on microparticles still needs to be scrutinized more to predict their trajectories in such devices. Results An integrated system utilizing two categories in microfluidics is proposed: dielectrophoretic manipulation of microparticles and centrifugal-driven microfluidics, followed by a numerical analysis. In this regard, we assumed a rectangular microchannel with internal unilateral planar electrodes equipped with three equal-sized outlets placed radially on a centrifugal platform where microparticles flow toward the disc's outer edge. The effect of different coordinate-based parameters, including radial and lateral distances (X and Y offsets)/tilting angles toward the radius direction (α), on the particles' movement was investigated. Additionally, the effect of operational parameters, including applied voltage, the microchannel width, the number of enabled electrodes, the diameter of particles, and the configuration of electrodes, were analyzed, and the distributions of particles toward the outlets were monitored. It was found that enhanced particle focusing becomes possible at lower rotation speeds and higher electric field values. Furthermore, the proposed centrifugal-DEP system's efficiency for classifying red blood cells/platelets and Live/Dead yeast cells systems was evaluated. Significance Our integrated system is introduced as a novel method for focusing and classifying various microparticles with no need for sheath flows, having the ability to conduct particles at desired routes and focusing width. Furthermore, the system effectively separates various bioparticles and offers ease of operation and high-efficiency throughput over conventional dielectrophoretic devices.

Abstract:This paper presents a microfluidic system for manipulation and separation of micron-sized particles based on the combined use of negative dielectrophoresis (DEP) and hydrodynamic forces. A 3-D microelectrode structure (so called paired electrode array) are constructed face to face on the top and bottom sides of the microchannel and driven with high-frequency AC voltage to generate dielectrophoretic gates. Depending on the relative strengths of the two forces, particles such as polystyrene beads or cells carrying by a laminar flow can either penetrate the gate or settle there. This gives rise to certain applications including selectively concentrating particles from the flow, separating particles depending on their sizes or dielectric properties, and automatically positioning particles to selective locations. For this purpose, a microfluidic device consisting of the paired electrode array sitting on the channel has been fabricated using microfabrication techniques. Polystyrene beads were used to study the performance of the device. Experimental results including the concentration and separation of particles are presented.

Abstract:This study explores the frontiers of microparticle manipulation by introducing an actuator platform for the three-dimensional positioning of microparticles using dielectrophoresis (DEP), a technique known for its selectivity and ease of integration with microtechnology. Leveraging advancements in carbon-based devices due to their biocompatibility and electrochemical stability, our work extends the application of DEP from two-dimensional constraints to precise 3D positioning within microvolumes, employing a photolithography-based fabrication process known as Carbon-MEMS technology (C-MEMS). We present the design, finite element simulation, fabrication, and testing of this platform, which utilizes a unique combination of planar and 3D carbon microelectrodes individually addressable on a transparent substrate. This setup enables the application of DEP forces, allowing for high-throughput manipulation of multiple microparticles simultaneously, as well as displacement of individual microparticles in any desired direction. Demonstrated with spherical 1μm and 10μm diameter polystyrene microparticles, this platform features straightforward fabrication and is suitable for batch industrial production. The study concludes with a discussion of the platform's advantages and limitations, marking a significant step toward a valuable tool for studying complex biological systems.
