Abstract:In this study, the stereochemistry, stereoselective fungicidal bioactivity, and antifungal mechanism of chiral triazole fungicide metconazole were investigated. The configurations of metconazole stereoisomers were determined to be (1R, 5R)-metconazole, (1R, 5S)-metconazole, (1S, 5S)-metconazole, and (1S, 5R)-metconazole through using electronic circular dichroism spectroscopy. The bioactivities of four stereoisomers and their stereoisomer mixture toward Fusarium graminearum Schw and Alternaria triticina were found to be in the following order: (1S, 5R)-metconazole > the stereoisomer mixture > (1S, 5S)-metconazole > (1R, 5R)-metconazole > (1R, 5S)-metconazole. In addition, the fungicidal activities of (1S, 5R)-metconazole against two tested pathogens was 13.9–23.4 times higher than those of (1R, 5S)-metconazole. Molecular docking methodology was applied to characterize the docking energy and distances between Cytochrome P450 CYP51B and the metconazole stereoisomers, and (1S, 5R)-metconazole showed the strongest binding energy and the shortest distance binding to CYP51B than the other three stereoisomers. Moreover, enantioselective metabolisms of (1S, 5R)-metconazole and (1R, 5S)-metconazole by Fusarium graminearum Schw were investigated through NMR-based metabolomics. The amounts of alanine, arginine, acetate, ethanol, and dimethylamine produced in the presence of (1R, 5S)-metconazole were significantly higher than corresponding amounts in the presence of (1S, 5R)-metconazole, whereas the amounts of glucose, glycerol, glutamate, methionine, and trimethylamine formed in the presence of (1R, 5S)-metconazole were much less than those in the presence of (1S, 5R)-metconazole. This systematic investigation of metconazole stereoisomers would provide a new perception of metconazole in stereoisomeric level, including bioactivities, metabolic behaviors and antifungal mechanism.

Abstract:Elaborating the environmental behavior of mefentrifluconazole, a novel triazole fungicide, in stereoselective level is of paramount importance for the application of the pesticide in agriculture. In this study, the enantioselective bioactivity, acute toxicity and stereoselective bioaccumulation of mefentrifluconazole in earthworm (Eisertia fetida) were investigated. Bioactivity tests against four pathogens revealed that R-( - )-mefentrifluconazole exhibited approximately 11-113 times higher bioactivity than its S-( + )-mefentrifluconazole. However, the LC50 of S-( + )-, rac- and R-( - )mefentrifluconazole to earthworm was measured to be 4.1, 11.4 and 7.3 mu g/cm(2), respectively, indicating active ingredient R-( - )-mefentrifluconazole is less toxic than its racemate and S-form. Accumulation of mefentrifluconazole in earthworms was non-enantioselective and negatively related to its adsorption onto soils. The concentration of mefentrilluconazole in in situ pore water (C-IPW) and CaCl2 extraction (C-CaCl2) was closely related to its accumulation in earthworms, suggesting that C-IPW and C-CaCl2 could be appropriate indicators for estimation of the bioavailability of mefentrilluconazole in soil. Conclusively, our study provides necessary information for the risk assessment of mefentrifluconazole in agriculture.
Evaluation of chiral triticonazole in three kinds of fruits: enantioseparation, degradation, and dietary risk assessment

Abstract:AbstractThe enantioselective behaviors of chiral pesticides would affect the accuracy of risk assessment. This study evaluated the enantioselectivity of chiral triticonazole (a widely used fungicide) in three kinds of fruits. Firstly, the enantioseparation of triticonazole enantiomers was carried out within 1.2 min utilizing CHIRALPAK OJ-3 column with a mixture of CO2 and methanol (93:7, v/v) using SFC-MS/MS. Secondly, field trials were conducted to clarify the enantioselective degradation and residue of S-( +)-triticonazole and R-(–)-triticonazole in fruits. The initial concentrations of rac-triticonazole were 25.1–93.1 ng/g, and enantioselective degradation was observed in pear, peach, and jujube after 2 h, 10 days, and 3 days, respectively. The degradation of S-( +)-triticonazole was fastest in pear (T1/2, 2.01 days), while the T1/2 of R-(–)-triticonazole was 5.02 days. The residue concentrations of rac-triticonazole were less than the MRL set by EU (10 ng/g) on the 3rd and 21st day in pear and peach, respectively, which were lower than 10 ng/g in jujube on the 30th day (no MRL). Finally, we found that the dietary intake risks of rac-triticonazole in fruits were low for 2–7 age, 20–50 age/female, and 20–50 age/male. The current study could provide complimentary references for the rational usage, MRL formulation, and risk assessment of chiral triticonazole.

Abstract:Elucidating the enantiomeric chemistry and enantioselective fate of the novel chiral triazole fungicide mefentrifluconazole is of vital importance for agroecosystem safety and human health. The absolute configuration of mefentrifluconazole was identified firstly as S-(+)-mefentrifluconazole and R-(−)-mefentrifluconazole on a cellulose tris(3-chloro-4-methylphenylcarbamate) chiral phase. A baseline resolution (<math>Rs,</math> 2.51), favorable retention (<math>RT</math> ≤ 2.24 min), and high sensitivity (LOQ, 0.5 μg/kg) of enantiomer pair were achieved by reversed-phase liquid chromatography tandem mass spectrometry combined with a 3D response surface strategy. Nationwide field trials were undertaken to clarify the enantiomer occurrence, enantioselective dissipation, terminal concentrations, and storage stability of S-mefentrifluconazole and R-mefentrifluconazole in watermelon across China. The original deposition of the sum of enantiomer pair was estimated to be 14.4–163.7 μg/kg, and terminally decreased to < LOQ–59.3 μg/kg 10 days after foliage application. S-mefentrifluconazole preferentially degraded (T1/2, 3.3–6.0 days), resulting in the relative enrichment of R-mefentrifluconazole (T1/2, 3.9–6.6 days) in watermelon. A probabilistic model is recommended for the dietary risk assessment, although both acute (%ARfD, 0.435–22.188%) and chronic (%ADI, 1.697–9.658%) risks are acceptable for associated population. The long-term exposures should be continuously emphasized given the increasing applications and persistent fate of mefentrifluconazole, especially for urban children.

Abstract:Cyproconazole is a representative and widely used triazole fungicide with four stereoisomers, which will bring some risks to non-target organisms. A fast analytical method on supercritical fluid chromatography-tandem mass spectrometry was established in 4 min, and the environmental hazards of chiral cyproconazole were studied in earthworm-soil microcosm, including stereoselective bioaccumulation and dissipation. In the process of bioaccumulation, the concentrations of cyproconazole stereoisomers in earthworms showed a trend of increasing first and then reaching a stable state at 6 mg/kg treatment, which was different from those at 0.6 mg/kg treatment (decease-increase-equilibrium). The concentration order was (2S,3R)- > (2S,3S)- > (2R,3R)- > (2R,3S)-cyproconazole and (2S,3S)- ≈ (2S,3R)- > (2R,3R)- > (2R,3S)-cyproconazole at 6 and 0.6 mg/kg treatments, respectively. The bioaccumulation factor (BAF) values were in the range of 0.018-0.55, showing weakly relative accumulation capacity. The dissipation of cyproconazole stereoisomers in artificial soil accorded with the first-order kinetics equation, and the half-lives were 20.1-23.6 and 7.66-8.28 days at 6 and 0.6 mg/kg treatments, respectively, without stereoselectivity and diastereoselectivity. In earthworms, the dissipation half-lives were 5.81-6.01 days with the preferential dissipation of (2R,3R)-cyproconazole. The study would help with the rational uses and risk assessments of cyproconazole.

Abstract:The enantioselective effects of chiral herbicides on aquatic organisms have received increasing attention. As one kind of freshwater algae responsible for most algal blooms, Microcystis aeruginosa can produce hepatotoxic microcystin and cause serious health concerns for drinking water. Thus, the effects of chiral herbicides on M. aeruginosa are of vital significance but poorly understood, especially as the structures of chiral herbicides become more complex. In this study, the enantioselective effects of four metolachlor enantiomers based on carbon center and axis chirality on M. aeruginosa were investigated for the first time at an enantiomeric level. The results of the investigation into algal growth inhibition, chlorophyll a content, and cell integrity indicated that ( S)-metolachlor [( S)-Met] was significantly more toxic than any other isomer. The toxicity ranking of different enantiomers at the highest concentration (15 mg/L) against M. aeruginosa was ( S)-Met > (α R,1' S)-Met > (α S,1' S)-Met > (α S,1' R)-Met > (α R,1' R)-Met, with (α S,1' S)-Met and (α R,1' S)-Met displaying a synergistic effect. Additionally, the Fe distribution in M. aeruginosa presented distinct enantioselectivity, which may contribute to the enantioselective toxicity of metolachlor. Furthermore, metolachlor upregulated the expression of genes mcyD and mcyH in an enantioselective manner, indicating that this herbicide can potentially promote the synthesis and efflux of microcystin, thus aggravating agricultural water contamination to different extents. Overall, this study will help to understand the ecotoxicity of metolachlor at a deeper level and provide theoretical insights into the enantioselective behaviors of metolachlor.
Enantioselective Toxicity Assessment of Prothioconazole on Earthworms (eisenia Foetida) in Artificial Soil Environments

Abstract:The chiral fungicide prothioconazole (PTZ) is extensively employed in agricultural practices, prompting serious concern due to its environmental impact. PTZ is prone to undergo metabolism, leading to the formation of chiral prothioconazole-desthio (dPTZ) in the environment. However, limited knowledge exists regarding its enantioselective behavior and toxicity towards invertebrate organisms in soil ecosystems. In this study, R-(-)- and S-(+)- PTZ enantiomers were individually synthesized, and their stereoselective toxicity effects on earthworms (E. foetida) were studied in artificial soil under environmentally relevant concentration exposures. The results showed a significant accumulation of dPTZ in earthworms, surpassing the levels of PTZ. Moreover, the concentration of S-(-)- dPTZ in earthworms was notably higher than that of R-(+)- dPTZ after exposure, reaching peak levels on day 14. Concurrently, oxidative stress induced by S-(+)- PTZ enantiomers in earthworms exhibited a substantial increase compared to R-(-)- enantiomers on day 14, indicating a higher ecological risk associated with the former in non-target organisms. Transcriptome analysis unveiled distinct impacts on earthworm physiology. S-(+)-PTZ exposure significantly affected energy metabolism, immune responses and digestive systems. In contrast, R-(-)-PTZ exposure influenced the synthesis of carbohydrates, proteins, and lipids. These insights contribute to understanding the complex interactions between PTZ enantiomers and soil-dwelling organisms, providing a scientific foundation for advancing the application of high efficiency, low toxicity PTZ monomer pesticides.

Abstract:Continued application of new chiral fungicide mefentrifluconazole (MFZ) increases its risk to soil ecosystem. However, the toxicity of MFZ enantiomers to soil fauna and whether stereoselectivity exists remains poorly elucidated. Based on multilevel toxicity endpoints and transcriptomics, we investigated the negative effects of racemic, R-(-)-, and S-(+)-MFZ on Eisenia fetida . After exposure to S-(+) configuration at 4 mg/kg for 28 day, its reactive oxygen species levels were elevated by 15.4% compared to R-(-) configuration, inducing enantiospecific oxidative stress and transcriptional aberrations. The S-(+) isomer induced more severe cell membrane damage and apoptosis than the R-(-) isomer, and notably, the selectivity of apoptosis is probably dominated by the mitochondrial pathway. Mechanistically, differential mitochondrial stress lies in: S-(+) isomer specifically up-regulated mitochondrial cellular component compared to R-(-) isomer and identified more serious mitochondrial fission. Furthermore, S-(+) conformation down-regulated biological processes associated with ATP synthesis and metabolism, with specific inhibition of mitochondrial respiratory electron transport chain complex I and IV activity resulting in more severe electron flow disturbances. These ultimately mediated enantioselective ontogenetic process disorders, which were supported at phenotypic (weight loss), genetic, and protein (reverse modulate TCTP and Sox2 expression) levels. Our findings offer an important reference for elucidating the enantioselective toxicological mechanism of MFZ in soil fauna.

Dynamics of 5 fungicides residue in grape brewing fermentation process

Abstract:The residue of five fungicides (myclobutanil, famoxadone, mandipropamid, propiconazole, difenoconazole) in homemade wine fermentation process was analyzed by high performance liquid chromatography-mass spectrometry/mass spectrometry (HPLC-MS/MS).The effect of adding yeast on fungicides residue was also studied.It was shown that the average recoveries of the determination method ranged in 75.4％-112.5％.Five fungicides showed different degrees of degradation in the first fermentation stage, yet myclobutanil and mandipropamid showed no significant degradation during the fermentation process with yeast addition.The five fungicides residue showed a balanced state during the secondary fermentation stage.Adding yeast during fermentation did not significantly affect the degradation tendency.The residue of five fungicides in grape brewing process was not only related to the brewing process, but also related to the physicochemical properties of the fungicides.
Stereoselective bioactivity, toxicity and degradation of novel fungicide sedaxane with four enantiomers under rice-wheat rotation mode

Abstract:Sedaxane was a novel chiral fungicide that contains four enantiomers. Unfortunately, the stereoselective bioactivity, toxicity and degradation of sedaxane have not been clarified. In this study, we identified the absolute configuration of the four sedaxane enantiomers at first time. The stereoselective bioactivity toward three wheat and rice pathogens, stereoselective acute toxicity to aquatic organisms (Selenastrum capricornutum and Daphnia magna), and stereoselective degradation of sedaxane were studied. The 1 S,2S-(+)-sedaxane possessed 5.4-7.3 times greater bioactivity than 1 R,2R-(-)-sedaxane to Rhizoctonia solani and Rhizoctonia cerealis. Contrarily, the 1 R,2S-(+)-sedaxane had 4.2 times greater activity than 1 S,2S-(+)-sedaxane against Fusarium graminearum. The 1 R,2R-(-)-sedaxane had 2.8 times greater toxicity than 1 S,2S-(+)-sedaxane to S. capricornutum. The chiral determination method used ultrahigh-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The recovery of sedaxane stereoisomers ranged from 83.1 % to 98.2 %, with RSDs (Relative standard deviations) of 1.2 %- 8.4 %. The trans-sedaxane existed stereoselective degradation phenomenon in the rice-wheat rotation mode, and 1 S,2S-(+)-sedaxane was preferentially degraded. Our results would provide scientific importance and practical guidance to the safety evaluation of chiral pesticides.
Metolachlor Stereoisomers: Enantioseparation, Identification and Chiral Stability

Abstract:Metolachlor is a chiral herbicide consisting of four stereoisomers, which is typically used as a racemic mixture or is enriched with the herbicidally active 1'S-isomers. Because studies on the enantioselective behavior of phyto-biochemical processes and the environmental fate of metolachlor have become significant, a practical method for analyzing and separating metolachlor stereoisomers must be developed. In the present study, the enantiomeric separation of metolachlor was achieved using OD-H, AS-H, OJ-H and AY-H chiral columns. The effects of different organic modifiers in an n-hexane-based mobile phase were investigated, and various temperatures and flow rates, which may influence metolachlor separation, were also explored. The optimal resolution was obtained using an AY-H column with n-hexane/EtOH (96/4) as the mobile phase at a rate and temperature of 0.6mLmin(-1) and 25°C, respectively. The absolute configuration of the four stereoisomers was identified as αSS, αRS, αSR, αRR using computed and experimentally measured ECD and VCD spectra. Thermal interconversion and solvent stability experiments were also performed. Pure metolachlor stereoisomers in different organic solvents and water at 4°C or 30°C were stable. These results were used to establish a sound method for analyzing, preparing, characterizing, and preserving individual metolachlor stereoisomers in most natural environments.
Dissipation behaviour, residue distribution and dietary risk assessment of tetraconazole and kresoxim-methyl in greenhouse strawberry via RRLC-QqQ-MS/MS technique

Abstract:20% commercial suspension emulsion (SE) of (8% tetraconazole + 12% kresoxim-methyl), as a pre-registered product in China, was firstly investigated under Chinese greenhouse-field conditions. A MWCNTs-based QuEChERS method for simultaneous determination of tetraconazole and kresoxim-methyl in strawberry was developed and validated via RRLC-QqQ-MS/MS. On basis of this method, the dissipation behaviours, residue distributions and dietary risk probability of these fungicides in strawberry were further investigated for food safety. The dissipations of tetraconazole and kresoxim-methyl followed first-order kinetics with the half-lives of 8.0-18.2 days. The highest residues (HRs) of these fungicides in the supervised trials at the pre-harvest interval (PHI, 3 days) were below 0.8970mgkg-1. The total national estimated daily intake (NEDI) of tetraconazole and kresoxim-methy in strawberry at the PHI 3day was 0.2784mg and 0.4031mg, respectively, based on Chinese dietary pattern and terminal residue distributions under good agricultural practices (GAP) conditions. The risk quotients (RQs) of tetraconazole and kresoxim-methy at PHI 3 days were below 82.7% and 1.6%, respectively, showing that the evaluated strawberry exhibited an acceptably low dietary risk to consumers. The current study could not only guide reasonable usage of the formulation, but also facilitate the setting of maximum residue limits (MRLs) of tetraconazole in strawberry.

Abstract:The chiral pesticide famoxadone is mainly applied to control fungal diseases on fruiting vegetables. The fungicidal activity, ecotoxicological effects, and degradation behavior of famoxadone enantiomers are less well known. In this study, a systemic assessment of the stereoselectivity of famoxadone was performed in cucurbits and soil. Famoxadone enantiomers presented distinct inhibitory activities among different fungal species. The bioactivities of R-(−)-famoxadone were 2.7–178 times higher than S-(+)-famoxadone toward five phytopathogens. Based on the obtained LC50 values, famoxadone was super toxic to Eisenia foetida (E. foetida). Moreover, the acute toxicity of R-(−)-famoxadone presented 167 times greater to E. foetida than that of S-(+)-famoxadone, indicating that R-(−)-famoxadone showed higher bioactivity toward target organisms and non-target organisms than S-(+)-famoxadone. In addition, a simple high-performance liquid chromatography (HPLC) method was established to determine the stereoselective degradation of famoxadone in two species of cucurbits (cucumber and chieh-qua) and in field soil. The half-life values of famoxadone degradation were from 5.4 to 14.1 days, indicating that famoxadone was easily degraded. Additionally, no stereoselective degradation was found in cucurbits and soil. The results may provide promising implications for comprehensive environmental and ecological risk assessments of famoxadone.

Abstract:The increasing application of chiral pesticides has enhanced interest in their enantioselectivity. However, little relevant information is currently available for enantioselective activity of chiral plant growth regulators. In an attempt to screen active enantiomers of uniconazole, this work investigated enantiomeric separation and the enantioselective effect of uniconazole on the growth of rice seedlings and cyanobacteria. Baseline resolution of uniconazole enantiomers was achieved on a Chiralpak AD column by chiral high-performance liquid chromatography (HPLC). The relationship among circular dichroism (CD), optical rotation (OR), and absolute configuration was successfully established by coupling of CD and OR detection. The t test at the 95% level of confidence indicated significant differences between the enantiomers in their retardant activity toward growth of rice seedlings and stimulation effect on growth of cyanobacteria, the natural biofertilizers in rice paddy fields. The S-(+)-enantiomer was more active than the R-(-)-enantiomer in retarding growth of rice seedlings and stimulating growth of Microcystis aeruginosa. This special enantiomeric selectivity was further elucidated by probing the binding mode of enantiomers to gibberellin (GA) 20-oxidase by molecular docking. The S-(+)-enantiomer was found to bind tightly with GA 20-oxidase. The results suggested that the S-(+)-enantiomer instead of a racemate of uniconazole should be used to improve rice seedling quality.

Abstract:Comment on the editorial by Edwards et al Smoking restrictions have been progressively introduced in enclosed public spaces in a growing number of nations as the evidence of the harms of passive smoking accumulates and public preference for smoke free air grows. Bans on smoking on public transport and airlines, and in cinemas, theatres, and elevators have long been taken for granted in many nations. Indoor workplaces are also increasingly becoming smoke free by virtue of either explicit legislation or management policy. Yet throughout the world, hospitality venues—particularly pubs and bars—have staunchly resisted this momentum. The editorial by Edwards et al in this issue1 describes a depressingly typical “Groundhog Day” scenario to anyone monitoring global developments in this area. The situation today is an object lesson in “half-pregnant” policy and in how public health law should not be made. Those least exposed—the relatively transient patrons of restaurants, public transport, cinemas, and shopping malls—have often full legal protection from passive smoking. Yet those most exposed—bar and casino staff—are the least protected. In effect, government inaction says to them “we’ll protect citizens from passive smoking even when their exposure is brief. But notwithstanding your unparalleled high exposure, we’ll offer you no protection”. It is unimaginable that workplace health protection could proceed in such a fashion in relation to toxic fumes, noise, or asbestos. Abstract:In this work, the enantioselective bioaccumulation and dissipation of uniconazole enantiomers in earthworm-soil microcosm were studied. A fast enantioseparation method of uniconazole through supercritical fluid chromatography-tandem mass spectrometry (SFC-MS/MS) was established. The CHIRALCEL OZ-3 column and a mixture of CO2 and methanol (80:20, v/v) were used within 1.0 min to separate uniconazole enantiomers. The recoveries of uniconazole enantiomers in earthworm and soil samples ranged from 83.3 to 113%, and the intra-day and inter-day relative standard deviation values were lower than 11%. In earthworms, the bioaccumulation concentrations of uniconazole enantiomers increased with time and reached the maximum on the 7th day and then decreased. The elimination of uniconazole enantiomers in earthworms followed the first-order kinetics equation, and the elimination half-lives were approximately 7 days. In artificial soil, the dissipation of uniconazole enantiomers was slow, and the dissipation half-lives were both 25.7 days. No enantioselectivity occurred in the earthworm-soil microcosm. These results may reduce the uncertainty of environmental risk assessment for uniconazole.

Abstract:Difenoconazole is a commonly used triazole fungicide that consists of four stereoisomers [(2S,4S)-, (2S,4R)-, (2R,4R)-, and (2R,4S)-isomers] with different bioactivity. For example, the toxicity of the (2R,4S)-isomer to fish is approximately seven times higher than that of the (2S,4S)-isomer. However, the stereoselective toxic effects of difenoconazole stereoisomers on mammals have received little attention. In the present study, adult male mice were orally treated with a mixture of the four stereoisomers or each stereoisomer individually (0, 30, or 100 mg/kg/d) by gavage for 28 days. Pathological staining of the liver sections showed that the (2R,4R)-isomer caused lipid droplet accumulation. The mixture or each individual stereoisomers decreased the levels of amino acids and acyl-carnitine in serum. Moreover, the (2S,4R)-, (2R,4R)-, and (2R,4S)-isomers affected intestinal permeability, causing decreases in mucus secretion and tight junction protein expression in colon. Analysis of the gut microbiota composition showed that the stereoisomers caused decreases of OTU numbers and observed species at different levels. Interestingly, difenoconazole and its four stereoisomers reduced the relative abundance of Bacteroidetes at the phylum level and some short-chain fatty acid (SCFA)-producing bacteria. Taking the findings together, 2R-difenoconazole with strong bioactivity against pathogenic fungi also had significant effects in mammals, disrupting hepatic lipid metabolism, intestinal permeability, and gut microbiota. It is concluded that the health risks of the four difenoconazole stereoisomers to mammals should not be overlooked.

Abstract:Over the past few decades, pesticides have been used in large quantities, and they pose potential risks to organisms across various environments. Reducing the use of pesticides and their environmental risks has been an active research focus and difficult issue worldwide. As a class of pesticides with special structures, chiral pesticides generally exhibit enantioselectivity differences in biological activity, ecotoxicity, and environmental behavior. At present, replacing the racemates of chiral pesticides by identifying and developing their individual enantiomers with high efficiency and environmentally friendly characteristics is an effective strategy to reduce the use of pesticides and their environmental risks. In this study, we review the stereoselective behaviors of chiral pesticide, including their environmental behavior, stereoselective biological activity, and ecotoxicity. In addition, we emphasize that the systematic evaluation of chiral pesticides at the enantiomeric level is a promising novel strategy for developing highly effective and less harmful pesticides, which will provide important data support and an empirical basis for reducing pesticide application.
